Abstract:Several classes of new insecticides with novel mode of action,such as emamectin benzoate,abamectin,indoxacarb,chlorfenapy,methoxyfenozide,JS118 and tebufenozide were developed or introduced to control the beet armyworm,spodoptera exigua(Hubner)in China.Resistance of this pest to many conventional insecticides has been reported in different countries and areas,but there is few evidence of resistance to these new insecticides in beet armyworm as yet.In order to make good use of these new insecticides and implement effective resistance management strategy,susceptibility to several classes of new insecticides was determined using leaf-dipping method in field populations of Spodoptera exigua from six areas of Jiangsu and Henan Province,in China.The results showed that in all six field populations tested with abamectin,three populations from Fengxian and Fu'ning counties,Jiangsu province and Henan province were susceptible or had decreased susceptibility(2.5~4.4-fold),one population from Dafeng county,Jiangsu province developed low level resistance(5.2-fold),and two populations from Jiangning and Sheyang counties,Jiangsu province first developed moderate levels of resistance(10.4~14.2-fold).In all four populations tested,three populations from Henan province and Jiangning,Fu'ning and Fengxian of Jiangsu province were susceptible(2.3~2.9-fold)to indoxacarb except that one population from Fengxian of Jiangsu province had low level of resistance(6.8-fold)to indoxacarb.Four populations from Henan province and Jiangning,Fu'ning and Sheyang counties of Jiangsu province were susceptible to emamectin benzoate(RR0.6~2.0-fold).In case of chlorfenapy,all four populations tested were susceptible(1.5~2.7-fold).Similarly,all populations tested were susceptible to methoxyfenozide(0.4~1.7-fold)and JS118(0.3~1.3-fold).In all six populations tested with tebufenozide,four populations from Dafeng,Jiangning and Fengxian of Jiangsu province and Henan province were susceptible(1.6~2.5-fold),and two populations from Fu'ning and Sheyang,Jiangsu province had decreased susceptibility(3.2-fold).These results provide a guideline for developing a more effective resistance management program for beet armyworm.
Using Field-Evolved Resistance to Cry1F Maize in a Lepidopteran Pest to Demonstrate No Adverse Effects of Cry1F on One of Its Major Predators

Abstract:Spodoptera frugiperda (JE Smith) represents the first documented case of field-evolved resistance to a genetically engineered crop expressing an insecticidal protein from Bacillus thuringiensis (Bt). In this case it was Cry1F-expressing maize (Mycogen 2A517). The ladybird beetle, Coleomegilla maculata , is a common and abundant predator that suppresses pest populations in maize and many other cropping systems. Its larvae and adults are polyphagous, feeding on aphids, thrips, lepidopteran eggs and larvae, as well as plant tissues. Thus, C. maculata may be exposed to Bt proteins expressed in genetically engineered crops by several pathways. Using Cry1F-resistant S. frugiperda larvae as prey, we evaluated the potential impact of Cry1F-expressing maize on several fitness parameters of C. maculata over two generations. Using Cry1F resistant prey removed any potential prey-mediated effects. Duration of larval and pupal stages, adult weight and female fecundity of C. maculata were not different when they were fed resistant S. frugiperda larvae reared on either Bt or control maize leaves during both generations. ELISA and insect-sensitive bioassays showed C. maculata were exposed to bioactive Cry1F protein. The insecticidal protein had no effect on C. maculata larvae, even though larvae contained 20–32 ng of Cry1F/g by fresh weight. Over all, our results demonstrated that the Cry1F protein did not affect important fitness parameters of one of S. frugiperda ’s major predators and that Cry1F protein did not accumulate but was strongly diluted when transferred during trophic interactions.
Eliminating host-mediated effects demonstrates Bt maize producing Cry1F has no adverse effects on the parasitoid Cotesia marginiventris

Abstract:The fall armyworm, Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae), is an important pest of maize in the United States and many tropical areas in the western hemisphere. In 2001, Herculex I ® (Cry1F) maize was commercially planted in the United States to control Lepidoptera, including S. frugiperda . In 2006, a population of S. frugiperda was discovered in Puerto Rico that had evolved resistance to Cry1F maize in the field, making it the first well-documented case of an insect with field resistance to a plant producing protein from Bacillus thuringiensis (Bt). Using this resistant population, we conducted tri-trophic studies with a natural enemy of S. frugiperda . By using resistant S. frugiperda , we were able to overcome possible prey-mediated effects and avoid concerns about potential differences in laboratory- or field-derived Bt resistance. We used the Cry1F-resistant S. frugiperda to evaluate effects of Cry1F on Cotesia marginiventris (Cresson) (Hymenoptera: Braconidae), a larval endoparasitoid of S. frugiperda, over five generations. Our results clearly demonstrate that Cry1F maize does not affect development, parasitism, survivorship, sex ratio, longevity or fecundity of C. marginiventris when they parasitize Cry1F maize-fed S. frugiperda. Furthermore, the level of Cry1F protein in the leaves was strongly diluted when transferred from Bt maize to S. frugiperda and was not detected in larvae, cocoons or adults of C. marginiventris. Our results refute previous reports of C. marginiventris being harmed by Bt proteins and suggest that such results were caused by prey-mediated effects due to using Bt-susceptible lepidopteran hosts.
Using Resistant Prey Demonstrates That Bt Plants Producing Cry1Ac, Cry2Ab, and Cry1F Have No Negative Effects on Geocoris Punctipes and Orius Insidiosus.

Abstract:Geocoris punctipes (Say) and Orius insidiosus (Say) are generalist predators found in a wide range of crops, including cotton (Gossypium hirsutum L.) and maize (Zea mays L.), where they provide important biological control services by feeding on an array of pests, including eggs and small larvae of caterpillars. A high percentage of cotton and maize in the United States and several other countries are transgenic cultivars that produce one or more of the insecticidal Cry proteins of Bacillus thuringiensis Berliner (Bt). Here we quantify effects of three Cry proteins on the life history of these predators over two generations when they are exposed to these Cry proteins indirectly through their prey. To eliminate the confounding prey quality effects that can be introduced by Bt-susceptible prey, we used Cry1Ac/Cry2Ab-resistant Trichoplusia ni (Hübner) and Cry1 F-resistant Spodoptera frugiperda (J.E. Smith) in a series of tri-trophic studies. Survival, development, adult mass, fecundity, and fertility were similar when predators consumed larvae feeding on Cry1Ac/Cry2Ab cotton or Cry1 F maize compared with prey feeding on isogenic or near-isogenic cotton or maize. Repeated exposure of the same initial cohort over a second generation also resulted in no differences in life-history traits when feeding on non-Bt- or Bt-fed prey. Enzyme-linked immunosorbent assay showed that predators were exposed to Bt Cry proteins from their prey and that these proteins became increasingly diluted as they moved up the food chain. Results show a clear lack of effect of three common and widespread Cry proteins on these two important predator species. The use of resistant insects to eliminate prey quality effects provides a robust and meaningful assessment of exposure and hazard.
Resistance monitoring and mechanism in the fall armyworm Spodoptera frugiperda (Lepidoptera: Noctuidae) for chlorantraniliprole from Sichuan Province, China

Abstract:The fall armyworm, Spodoptera frugiperda (Noctuidae: Lepidoptera), is a wide-reaching notorious insect pest of important cereal crops, which has developed resistance to multiple classes of insecticides. It invaded the Sichuan Province of China in 2019. In this study, we performed resistance monitoring of insecticides for 11 field-collected populations from Sichuan, and all the populations were susceptible to emamectin benzoate and chlorpyrifos. The variations in resistance level to indoxacarb (resistance ratio (RR), 9.23–45.53-fold), spinetoram (RR, 4.32–18.05-fold), and chlorantraniliprole (RR, 2.02–10.39-fold) were observed among these populations. To investigate the resistance mechanism of chlorantraniliprole, synergism tests were performed and showed that piperonyl butoxide had a slight synergistic effect on chlorantraniliprole for the QJ-20 population (1.43-fold) in moderate resistance (RR, 10.39-fold) compared with the treatment group without synergist. Furthermore, the expression scanning for resistance-related genes showed that five P450 genes ( CYP6AE43, CYP321A8, CYP305A1, CYP49A1 , and CYP306A1 ) and the ryanodine receptor gene ( Ryr , chlorantraniliprole target) were overexpressed in the QJ-20 population. These results indicated that the fall armyworm in Sichuan has exhibited diverse susceptibilities to several classes of insecticides, and the overexpression of Ryr and several P450 genes may contribute to the development of resistance in S. frugiperda to chlorantraniliprole.

Abstract:The fall armyworm (FAW), Spodoptera frugiperda (J.E. Smith), is one of the most important insect pests affecting corn crops worldwide. Although planting transgenic corn expressing Bacillus thuringiensis (Bt) toxins has been approved as being effective against FAW, its populations' resistance to Bt crops has emerged in different locations around the world. Therefore, it is important to understand the interaction between different Bt proteins, thereby delaying the development of resistance. In this study, we performed diet-overlay bioassays to evaluate the toxicity of Cry1Ab, Cry1Ac, Cry1B, Cry1Ca, Cry1F, Cry2Aa, Cry2Ab, Vip3Aa11, Vip3Aa19, and Vip3Aa20, as well as the interaction between Cry1Ab-, Cry1F-, Cry2Ab-, and Vip3Aa-class proteins against FAW. According to our results, the LC50 values of Bt proteins varied from 12.62 ng/cm2 to >9000 ng/cm2 (protein/diet), among which the Vip3Aa class had the best insecticidal effect. The combination of Cry1Ab and Vip3Aa11 exhibited additive effects at a 5:1 ratio. Cry1F and Vip3Aa11 combinations exhibited additive effects at 1:1, 1:2, and 5:1 ratios. The combination of Cry1F and Vip3Aa19 showed an antagonistic effect when the ratio was 1:1 and an additive effect when the ratio was 1:2, 2:1, 1:5, and 5:1. Additionally, the combinations of Cry1F and Vip3Aa20 showed antagonistic effects at 1:2 and 5:1 ratios and additive effects at 1:1 and 2:1 ratios. In addition to the above combinations, which had additive or antagonistic effects, other combinations exhibited synergistic effects, with variations in synergistic factors (SFs). These results can be applied to the establishment of new pyramided transgenic crops with suitable candidates, providing a basis for FAW control and resistance management strategies.

Abstract:Despite the successful application of toxins from Bacillus thuringiensis as biological control agents against pests, pests are showing resistance against an increasing number of Bacillus thuringiensis toxins due to evolution; thus, new toxins with higher toxicity and broad-spectrum activity against insects are being increasingly identified. To find new toxins, whole genome sequencing of the novel B. thuringiensis strain Bt S3076-1 was performed, and ten predicted toxic genes were identified in this study, including six cry genes, two tpp genes, one cyt gene and one vip gene, among which six were novel toxins. Subsequently, SDS‒PAGE analysis showed that the major proteins at the spore maturation stage were approximately 120 kDa, 70 kDa, 67 kDa, 60 kDa and 40 kDa, while active proteins after trypsin digestion (approximately 70 kDa and 40 kDa) exhibited LC 50 values of 149.64 μg/g and 441.47 μg/g against Spodoptera frugiperda and Helicoverpa armigera larvae, respectively. Furthermore, pathological observation results showed that the peritrophic membrane of Spodoptera frugiperda and Helicoverpa armigera larvae was degraded. These findings will provide an experimental reference for further research on the insecticidal activity, toxicity spectrum and synergism of these toxins in Bt S3076-1.

Abstract:Yellow Peach Moth (YPM), Conogethes punctiferalis (Guenée), is one of the most destructive maize pests in the Huang-Huai-Hai summer maize region of China. Transgenic Bacillus thuringiensis (Bt) maize provides an effective means to control this insect pest in field trials. However, the establishment of Bt resistance to target pests is endangering the continued success of Bt crops. To use Bt maize against YPM, the baseline susceptibility of the local populations in the targeted areas needs to be verified. Diet-overlay bioassay results showed that all the fourteen YPM populations in China are highly susceptible to Cry1Ab. The LC50 values ranged from 0.35 to 2.38 ng/cm2 over the two years of the collection, and the difference between the most susceptible and most tolerant populations was sevenfold. The upper limit of the LC99 estimates of six pooled populations produced >99% larval mortality for representative eight populations collected in 2020 and was designated as diagnostic concentrations for monitoring susceptibility in YPM populations in China. Hence, we evaluated the laboratory selection of resistance in YPM to Cry1Ab using the diet-overlay bioassay method. Although the resistant ratio was generally low, YPM potentially could evolve resistance to Cry1Ab. The potential developmentof resistance by target pests points out the necessity to implement resistance management strategies for delaying the establishment of pest resistance to Bt crops.

Comparative toxicity and enzymatic detoxification responses in Spodoptera frugiperda (Lepidoptera: Noctuidae) to two insecticides

Abstract:The fall armyworm (FAW), Spodoptera frugiperda Smith (Lepidoptera: Noctuidae), poses a significant threat to food security, necessitating effective management strategies. While chemical control remains a primary approach, understanding the toxicity and detoxification mechanisms of different insecticides is crucial. In this study, we conducted leaf-dipping bioassays to assess the toxicity of quinalphos and beta-cypermethrin·emamectin benzoate (β-cyp·EMB) on S. frugiperda larvae. Additionally, we assessed the response of alterations in CarE, GST, MFO, and AChE activities to sublethal concentrations of these insecticides over various treatment durations. Results indicated that β-cyp·EMB exhibited higher toxicity than quinalphos in S. frugiperda. Interestingly, the highest activities of GST, CarE, MFO, and AChE were observed at 6 h exposure to LC10 and LC25 of β-cyp·EMB, surpassing equivalent sublethal concentrations of quinalphos. Subsequently, GST and CarE activities exposure to β-cyp·EMB steadily decreased, while MFO and AChE activities exposure to both insecticides was initially decreased then increased. Conversely, two sublethal concentrations of quinalphos notably enhanced GST activity across all exposure durations, with significantly higher than β-cyp·EMB at 12-48 h. Similarly, CarE activity was also increased at various durations. Our research has exhibited significant alterations in enzyme activities exposure to both concentration and duration. Furthermore, Pearson correlation analysis showed significant correlations among these enzyme activities at different treatment durations. These findings contribute to a better understanding of detoxification mechanisms across different insecticides, providing valuable insights for the rational management of S. frugiperda populations.
Laboratory Evaluation of Selected Botanicals and Insecticides against Invasive Spodoptera frugiperda (Lepidoptera: Noctuidae)

Abstract:Background Maize is an economical crop of China, and its production has been severely affected by the invasive Spodoptera frugiperda in recent year. Application of synthetic pesticides are one of the most effective practices against FAW as an emergency control. This pest causes serious damage to the maize crop worldwide in recent decade, especially in China. Methods To find an alternative to synthetic insecticides there were total 16 different chemicals were used including ten botanical insecticides comprising of seven botanicals, azadirachtin, pyrethrin, nicotine, osthole, rotenone, Celastrus angulatus , matrine, and three insect growth regulators, diflubenzuron, lufenuron and buprofezin. Six synthetic insecticides, including emamectin benzoate, indoxacarb, imidacloprid and thiamethoxan, chlorantraniliprole, chlorfenapyr, were evaluated against 2nd instar of S. frugiperda larvae using leaf-dip method. Results: The results revealed that osthole, azadirachtin, buprofezin and pyrethrin were showed the significant larval mortality of 98.0, 96.7, 94.0 and 90.7%, in 120 h observation and exhibiting minimum LC 50 (39.04, 35.58, 61.45 and 48.46 mg/L, respectively) and LT 50 (48.91, 68.85, 58.67 and 58.57 hours, respectively) values. Among tested synthetic insecticides, emamectin benzoate, chlorantraniliprole and chlorfenapyr were showed significant higher mortality against larvae of S. frugiperda (99.3, 96.0 and 89.3%, respectively) in 72 h observation by exhibiting minimum LC 50 (0.26, 0.39 and 0.72 mg/L, respectively) and and LT 50 (10.18, 10.57 and 13.42 hours, respectively) values. More study is needed to test the laboratory findings in the field, although the efficient biorational pesticides might be utilized as part of integrated pest management against S. frugiperda . Conclusion The effective chemicals could be used in the management for S. frugiperda . The highest discriminating concentrations of tested botanical insecticides, insect growth regulators and insecticides caused significant mortality of S . frugiperda larvae.

Exposure to Cry1 Toxins Increases Long Flight Tendency in Susceptible but Not in Cry1F-Resistant Female Spodoptera frugiperda (Lepidoptera: Noctuidae)

Abstract:The fall armyworm (JE Smith) (Spodoptera frugiperda) is a polyphagous pest targeted by selected Cry and Vip3A insecticidal proteins from the bacterium Bacillus thuringiensis (Bt) that are produced in transgenic Bt corn and cotton. Available evidence suggests that sublethal larval exposure to Cry1Ac increases flight activity in adult Spodoptera spp. However, it is not known whether this effect is also observed in survivors from generally lethal exposure to Cry1Ac. Moreover, while multiple cases of field-evolved resistance to Bt proteins have been described in the native range of S. frugiperda, the effect of resistance on flight behavior has not been examined. Long-distance migratory flight capacity of S. frugiperda is of concern given its ongoing global spread and the possibility that migrants may be carrying resistance alleles against pesticides and Bt crops. In this study, we used rotational flight mills to test the effects of generally lethal exposure to Cry1Ac in susceptible and sublethal exposure in Cry1F-resistant S. frugiperda strains. The results detected altered pupal weight after larval feeding on diet containing Cry proteins, which only translated in significantly increased tendency for longer flights in female moths from the susceptible strain. This information has relevant implications when considering current models and assumptions for resistance management of Bt crops.
Sublethal effects of Bt toxin and chlorpyrifos on various Spodoptera exigua populations

Abstract:Abstract Bt cotton (Cry1Ac) has been commercially grown in China since 1997, saving China's cotton production from attack by Bt‐target pests and also tremendously reducing pesticide usage. In recent years, however, Bt cotton, with 4.2 million ha of cultivation, has suffered from a secondary target pest, S podoptera exigua ( H übner) ( L epidoptera: N octuidae). In C hina, growers have even had to re‐adopt conventional pesticides to control the pest, and this practice has already caused serious pesticide residue. In order to clarify the sublethal effects of chemical pesticide, the responses of a Bt‐susceptible and a Bt‐tolerant (Bt10) S . exigua strain to three treatment combinations were examined, including Bt toxin, sublethal chlorpyrifos, and Bt + sublethal chlorpyrifos. The susceptible and the Bt10 strain responded differently to dual pressure. Bt toxin + sublethal chlorpyrifos treatment lowered larval mortality and stimulated population increase of the susceptible S . exigua , whereas it delayed growth and development of the Bt10 strain. Under dual pressure, although larvae of the Bt10 strain developed faster than larvae of the susceptible strain, the Bt10 population experienced higher larval mortality, prolonged pupal duration, decreased pupal weight, decreased emergence rate, and shortened adult longevity. Compared with the susceptible strain, the Bt10 strain was deleteriously affected by sublethal chlorpyrifos. The Bt‐tolerant/resistant S . exigua population was more vulnerable to chemical pesticides like chlorpyrifos regardless of whether it was exposed to Bt toxin or not. Our study provides a reference for increasing the efficacy of control of S . exigua in Bt‐cotton planting areas.

A novel Cry1A resistance allele of fall armyworm in the new invaded region

Abstract:The fall armyworm, Spodoptera frugiperda , is a devastating pest in its native range Western Hemisphere and has become a major invasive pest around the globe. Transgenic crops producing Bt toxins have been widely used to control S. frugiperda . However, the evolution of resistance threatens the sustainability of Bt crops. Field-evolved S. frugiperda resistance to Bt crops was observed in America, whereas, no case of field-resistance was reported in its newly invaded East Hemisphere. Here we investigated the molecular mechanism of a Cry1Ab-resistant LZ-R strain of S. frugiperda , which selected 27-generations using Cry1Ab after being collected in corn fields from China. Complementation tests between LZ-R strain and SfABCC2-KO strain, which have been knockout SfABCC2 gene and confer 174-fold resistance to Cry1Ab, showed a similar level of resistance in the F 1 -progeny as their parent stains, indicating that a common locus of SfABCC2 mutation in LZ-R stain. Sequencing of the full length of SfABCC2 cDNA from LZ-R strain, we characterize a novel mutation allele of SfABCC2 . Cross-resistance results showed that Cry1Ab-resistance strain also confers >260-fold resistance to Cry1F, with no cross-resistance to Vip3A. These results provided evidence of a novel SfABCC2 mutation allele in the newly invaded East Hemisphere of S. frugiperda .

Dextropropoxyphene induced hepatotoxicity mimicking biliary tract disease r biopsy

Abstract:Three patients are described with recurrent jaundice, upper abdominal pain -and rigors attributable to dextropropoxyphene hepatotoxicity. The diagnosis was established in each patient by rechallenge; post challenge hepatic histology is reported in two. Twelve previous patients with probable dextropropoxyphene hepatic toxicity have been described and are reviewed. In 10 of the 15 patients, a clinical diagnosis of gall stone disease was made. Liver function tests are usually hepatitic shortly after challenge, but more cholestatic after a few days. No fatalities have been described, but as dextropropoxyphene is widely available in many different analgesic preparations possible toxicity should be considered in patients with relapsing jaundice mimicking biliary disease, in whom gall stones have been excluded.

Abstract:Helicoverpa armigera (Hubner) (Lepidoptera: Noctuidae) is an important lepidopteran pest of cotton, Gossypium hirsttttun. L. in Asia. Transgenic cotton expressing the cry1Ac gene from Bacillus thuringiensis (Bt) has been widely planted for control of this pest. For managing the potential risk from resistance evolution in this pest, a new transgenic Bt cotton containing cry1Ac and cry2Ab genes (gene pyramided strategy) was designed as a successor of cry1Ac cotton. This article reports on levels of Cry2Ab tolerance in H. armigera populations from Cry1Ac cotton planting region in China based on bioassays of F-1 and F-2 offspring of isofemale lines. In total, 572 isofemale families of H. armigera from Xiajin County of Shandong Province (an intensive Bt cotton-planting area) and 124 families from And County of Hebei Province [a multiple-crop system, including corn (Zea mays L.), soybean (Gycine max (L.) Merr., peanut (Arachis spp.), and Bt cotton] were screened with both Cry1Ac- and Cry2Ab-containing diets in 2008. The bioassays results indicated that relative average development rates (RADR) of F-1 full-sib families from field-collected female moths on Cry1Ac- and Cry2Ab-containing diet were positively correlated. The same correlation was found in the F2 generation, indicating cross-tolerance between Cry1Ac and Cry2Ab in field populations of H. armigera in Yellow River cotton-fanning region of China. This cross-tolerance must be considered in evaluating the utility of pyramiding Bt genes in cotton for delaying evolution of resistance.

Abstract:The susceptibility of the neonates of rice lepidopterans,striped stem borer Chilo suppressalis(Walker),pink stem borer Sesamia inferens(Walker) and leaffolder,Cnaphalocrocis medinalis(Guenée) to four Cry proteins,i.e.Cry1Ab,Cry1Ac,Cry1C,and Cry2A,from Bacillus thuringiensis were bioassayed using diet-feeding method and leaf-immersion method.The LC50 values of Cry1Ab,Cry1Ac,Cry1C,and Cry2A to C.suppressalis were 1.739 ng/mg,2.645 ng/mg,2.794 ng/mg,and 15.965 ng/mg,respectively;and those to S.inferens were 2.455 ng/mg,10.046 ng/mg,11.803 ng/mg,and 35.359 ng/mg,respectively;and to C.medinalis,4.000μg/ml,1.611μg/ml,69.031μg/ml,and 12.708μg/ml,respectively.Finally,the difference in susceptibility of rice lepidopterans to Cry proteins and its application to transgenic insect-resistant crops were also discussed.
Sphingomonas bacteria could serve as an early bioindicator for the development of chlorantraniliprole resistance in Spodoptera frugiperda

Abstract:The fall armyworm (Spodoptera frugiperda) was found to have invaded China in December 2018, and in just one year, crops in 26 provinces were heavily affected. Currently, the most effective method for emergency control of fulminant pests is to use of chemical pesticides. Recently, most fall armyworm populations in China were begining to exhibite low level resistance to chlorantraniliprole. At present, it is not possible to sensitively reflect the low level resistance of S. frugiperda by detecting target mutation and detoxification enzyme activity. In this study we found that 12 successive generations of screening with chlorantraniliprole caused S. frugiperda to develop low level resistance to this insecticide, and this phenotype was not attribute to genetic mutations in S. frugiperda, but rather to a marked increase in the relative amount of the symbiotic bacteria Sphingomonas. Using FISH and qPCR assays, we determined the amount of Sphingomonas in the gut of S. frugiperda and found Sphingomonas accumulation to be highest in the 3rd-instar larvae. Additionally, Sphingomonas was observed to provide a protective effect to against chlorantraniliprole stress to S. frugiperda. With the increase of the resistance to chlorantraniliprole, the abundance of bacteria also increased, we propose Sphingomonas monitoring could be adapted into an early warning index for the development of chlorantraniliprole resistance in S. frugiperda populations, such that timely measures can be taken to delay or prevent the widespread propagation of resistance to this highly useful agricultural chemical in S. frugiperda field populations.
Control Efficacy of the Bt Maize Event DBN3601T Expressing Cry1Ab and Vip3Aa Proteins against Beet Armyworm, Spodoptera exigua (Hübner), in China

Abstract:The beet armyworm, Spodoptera exigua (Hübner), is a major pest of maize, cotton, soybean, and many other crops globally. Despite the widespread deployment of Bt transgenic maize for pest control worldwide, the efficacy of Bt lepidopteran-resistant transgenic maize in managing S. exigua remains rarely studied. In this study, we quantified the expression level of pyramided Cry1Ab and Vip3Aa toxins in Bt maize (event DBN3601T) and evaluated their control efficiency against S. exigua under both laboratory and field conditions. The enzyme-linked immunosorbent assay (ELISA) results showed that the expression levels of Cry1Ab and Vip3Aa proteins in DBN3601T maize tissues followed a decreasing order as follows: V5-leaf > V8-leaf > VT-tassel > R2-kernel > R1-silk. Diet-overlay assay results showed that the LC50 values of Cry1Ab and Vip3Aa proteins against S. exigua larvae were 11.66 ng/cm2 and 27.74 ng/cm2, respectively, with corresponding GIC50 values at 1.59 ng/cm2 and 7.93 ng/cm2. Bioassay using various tissues of the DBN3601T maize indicated that after 7 days of infestation, mortality rates of neonates and third-instar larvae ranged from 86% to 100% and 58% to 100%, respectively. Mortality was highest on V5 and V8 leaves, followed by R2-kernel, VT-tassel, and R1-silk. Field trials demonstrated that DBN3601T maize exhibited significantly lower larval density, damage rate, and leaf damage score compared to non-Bt maize. Field cage trial showed that the control efficacy of DBN3601T maize at the vegetative stage could reach 98%. These findings provide a theoretical basis for utilizing Bt transgenic maize to enhance the sustainable management of S. exigua in Asia.

Abstract:Insect resistance evolution poses a significant threat to the advantages of biopesticides and transgenic crops utilizing insecticidal Cry-toxins from Bacillus thuringiensis (Bt). However, there is limited research on the relationship between transcriptional regulation of specific toxin receptors in lepidopteran insects and their resistance to Bt toxins. Here, we report the positive regulatory role of the SfGATAe transcription factor on the expression of the ABCC2 gene in Spodoptera frugiperda. DNA regions in the SfABCC2 promoter that are vital for regulation by SfGATAe, utilizing DAP-seq technology and promoter deletion mapping. Through yeast one-hybrid assays, DNA pull-down experiments, and site-directed mutagenesis, we confirmed that the transcription factor SfGATAe regulates the core control site PBS2 in the ABCC2 target gene. Tissue-specific expression analysis has revealed that SfGATAe is involved in the regulation and expression of midgut cells in the fall armyworm. Silencing SfGATAe in fall armyworm larvae resulted in reduced expression of SfABCC2 and decreased sensitivity to Cry1Ac toxin. Overall, this study elucidated the regulatory mechanism of the transcription factor SfGATAe on the expression of the toxin receptor gene SfABCC2 and this transcriptional control mechanism impacts the resistance of the fall armyworm to Bt toxins.

Abstract:Concerns about resistance development to conventional insecticides in diamondback moth (DBM) Plutella xylostella (L.), the most destructive pest of Brassica vegetables, have stimulated interest in alternative pest management strategies. The toxicity of Bacillus thuringiensis subsp. aizawai (Bt GO33A) combined with chlorantraniliprole (Chl) has not been documented. Here, we examined single and combined toxicity of chlorantraniliprole and Bt to assess the levels of resistance in four DBM strains. Additionally, enzyme activities were tested in field-original highly resistant (FOH-DBM), Bt-resistant (Bt-DBM), chlorantraniliprole-resistant (CL-DBM), and Bt + chlorantraniliprole-resistant (BtC-DBM) strains. The Bt product had the highest toxicity to all four DBM strains followed by the mixture of insecticides (Bt + Chl) and chlorantraniliprole. Synergism between Bt and chlorantraniliprole was observed; the combination of Bt + (Bt + Chl) (1:1, LC50:LC50) was the most toxic, showing a synergistic effect against all four DBM strains with a poison ratio of 1.35, 1.29, 1.27, and 1.25. Glutathione S-transferase (GST) and carboxyl-esterase (CarE) activities showed positive correlations with chlorantraniliprole resistance, but no correlation was observed with resistance to Bt and Bt + Chl insecticides. Expression of genes coding for PxGST, CarE, AChE, and MFO using qRT-PCR showed that the PxGST and MFO were significantly overexpressed in Bt-DBM. However, AChE and CarE showed no difference in the four DBM strains. Mixtures of Bt with chlorantraniliprole exhibited synergistic effects and may aid the design of new combinations of pesticides to delay resistance in DBM strains substantially.
