Abstract:Amounts of cost-effective biological observing and detecting instruments are needed in biology and medical applications. In these demands, a low-cost, portable microscope with light-weight and tele-communication, is very attractive in the resource-limit area. In this manuscript, a portable singlet phase contrast microscope based on smart-phones are proposed. In the optics hardware, an external singlet lens is designed and closely attached before the rear cameras of a smart-phone, which constructs the imaging part of a simplified microscope. And a circular oblique illumination is adopted. In the computational imaging part, the in-focus images recorded by the smart-phone are virtually enhanced and style-transferred to phase contrast images by deep learning methods. Through pairs of experimental image data, we train the deep learning network and demonstrate the effectiveness of this virtual phase contrast imaging approach using multiple samples, e.g., unstained/transparent tumor tissue pathological slide, and H&E (Hematoxylin and Eosin) stained tumor tissue pathological slide. Our results provide a powerful example of low-cost portable phase contrast microscope with high-speed tele-communication abilities in the 5G + mobile internet era.

Abstract:Methods for the detection, enumeration, and typing of cells are important in many areas of research and healthcare. In this context, flow cytometers are a widely used research and clinical tool but are also an example of a large and expensive instrument that is limited to specialized laboratories. Smartphones have been shown to have excellent potential to serve as portable and lower-cost platforms for analyses that would normally be done in a laboratory. Here, we developed a prototype smartphone-based flow cytometer (FC). This compact 3D-printed device incorporated a laser diode and a microfluidic flow cell and used the built-in camera of a smartphone to track immunofluorescently labeled cells in suspension and measure their color. This capability was enabled by high-brightness supra-nanoparticle assemblies of colloidal semiconductor quantum dots (SiO2@QDs) as well as a support vector machine (SVM) classification algorithm. The smartphone-based FC device detected and enumerated target cells against a background of other cells, simultaneously and selectively counted two different cell types in a mixture, and used multiple colors of SiO2@QD-antibody conjugates to screen for and identify a particular cell type. The potential limits of multicolor detection are discussed alongside ideas for further development. Our results suggest that innovations in materials and engineering should enable eventual smartphone-based FC assays for clinical applications.
Ultra High-Throughput Multiparametric Imaging Flow Cytometry: Towards Diffraction-Limited Sub-Cellular Detection

Abstract:ABSTRACT Flow cytometry is widely recognized as the gold-standard technique for the analysis and enumeration of heterogeneous cellular populations and has become an indispensable tool in diagnostics, 1 rare-cell detection 2 and single-cell proteomics. 3 Although contemporary flow cytometers are able to analyse many thousands of cells per second, with classification based on scattering or fluorescence criteria, the vast majority require unacceptably large sample volumes, and do not allow the acquisition of spatial information. Herein, we report a sheathless, microfluidic imaging flow cytometer that incorporates stroboscopic illumination for blur-free fluorescence and brightfield detection at analytical throughputs in excess of 60,000 cells/s and 400,000 cells per second respectively. Our imaging platform is capable of multi-parametric fluorescence quantification and subcellular (co-)localization analysis of cellular structures down to 500 nm with microscopy image quality. We demonstrate the efficacy of our approach by performing challenging high-throughput localization analysis of cytoplasmic RNA granules in yeast and human cells. Results suggest significant utility of the imaging flow cytometer in the screening of rare events at the subcellular level for diagnostic applications.

Abstract:Increasing demand for understanding the vast heterogeneity of cellular phenotypes has driven the development of imaging flow cytometry (IFC), that combines features of flow cytometry with fluorescence and bright field microscopy. IFC combines the throughput and statistical advantage of flow cytometry with the ability to discretely measure events based on a real or computational image, as well as conventional flow cytometry metrics. A limitation of existing IFC systems is that, regardless of detection methodology, only two-dimensional (2D) cell images are obtained. Without tomographic three-dimensional (3D) resolution the projection problem remains: collapsing 3D information onto a 2D image, limiting the reliability of spot counting or co-localization crucial to cell phenotyping. Here we present a solution to the projection problem: three-dimensional imaging flow cytometry (3D-IFC), a high-throughput 3D cell imager based on optical sectioning microscopy. We combine orthogonal light-sheet scanning illumination with our previous spatiotemporal transformation detection to produce 3D cell image reconstruction from a cameraless single-pixel photodetector readout. We further demonstrate this capability by co-capturing 3D fluorescence and label-free side-scattering images of single cells in flow at a velocity of 0.2 m s-1, corresponding to a throughput of approximately 500 cells per second with 60,000 voxels (resized subsequently to 106 voxels) for each cell image at a resolution of less than 1 micron in X, Y, and Z dimensions. Improved high-throughput imaging tools are needed to phenotype-genotype recognized heterogeneity in the fields of immunology, oncology, cell- and gene- therapy, and drug discovery.

Abstract:High-throughput single-cell heterogeneity imaging and analysis is essential for understanding complex biological systems and for advancing personalized precision disease diagnosis and treatment. Here, we present a miniaturized lensless chemiluminescence chip for high-throughput single-cell functional imaging with subcellular resolution. With the sensitive chemiluminescence sensing and wide field of view of contact lensless imaging, we demonstrated the chemiluminescent imaging of over 1000 single cells, and their membrane glycoprotein and the high-throughput single-cell heterogeneity of membrane protein imaging were examined for precision analysis. Furthermore, the functional adhesion and heterogeneity of single live cells were imaged and explored. This miniaturized lensless on-chip CL-CMOS imaging platform enables high-throughput single-cell imaging and analysis with high sensitivity and subcellular resolution, providing new techniques for the cellular study of biological heterogeneity and has potential application in precision disease diagnosis and treatment at the point-of-care settings.

Abstract:Integration micro lens arrays (MLAs) into a microfluidic chip has presented outstanding performances in flow cytometry field due to capability of eliminating trade-off between sensitivity and field of view (FOV), allowing high throughput multiplex analysis, and enhancing signal-to-noise ratio (SNR). However, the practices are still remained inside high-end laboratory environment, restricting MLAs to be utilized in point-of-care (POC) based flow cytometry applications. A portable system for droplet flow cytometry was successfully developed, which consists of an optofluidic chip integrated with MLAs and a smartphone application (APP). To cater to the demand for point-of-care testing, a straightforward optical setup that employs inclined LED illumination and micro lens pairs was used. This configuration effectively amplifies fluorescence signals, allowing them to be captured and analyzed solely using a smartphone. Through a series of repeated experiments, clear and consistent results were obtained that highlight the system's capabilities: (1) the smartphone APP installed in the system enabled both quantitative and qualitative analyses of flowing fluorescence droplets. These analyses could be performed in real time, providing immediate detection results, (2) by incorporating MLAs into the detection process, a notable enhancement in detection sensitivity was achieved when compared to cases without MLAs. Specifically, in experiments involving droplet concentrations ranging from 10 μm to 25 μm, resolvability showed an increase, the sensitivity increased by up to 142%. and the SNR exhibited an 11 dB improvement, (3) the customized APP exhibited comparable droplet counting results to a commonly utilized object tracking program. The measurement differences between the two methods ranged from 1.08% to 4.14%, reaffirming the accuracy of our self-developed APP. In conclusion, this portable system, coupled with the smartphone APP and MLAs-based optical configuration, demonstrated the ability to perform real-time, on-site droplet flow cytometry analysis with competitive detection performance.

Abstract:A smartphone-based analysis system is favored for point-of-care testing applications. The present work proposes a novel micro-spectroscopy/imaging system comprising a portable spectrometer as an optical sensor and a compact homemade microscope to acquire the image and spectra of micron-scale regions. Protein concentration quantification based on the bicinchoninic acid method was demonstrated with the proposed micro-spectroscopy/imaging system to analyse the spectrometer signals. Morphologies of onion endothelial and human breast cancer cells, used as biological sample models, were characterized to demonstrate the microscopic imaging capacity of the device. The ability to simultaneously obtain morphological and spectral information using the proposed portable device was demonstrated by examining the 10 mu m sub-pixels of a smartphone screen. These results highlight the potential for adopting a smartphone-based micro-spectroscopy/imaging system for point-of-care testing.

Abstract:We propose a light sheet based imaging flow cytometry technique for simultaneous counting and imaging of cells on a microfluidic platform. Light sheet covers the entire microfluidic channel and thus omits the necessity of flow focusing and point scanning based technology. Another advantage lies in the orthogonal detection geometry that totally cuts-off the incident light, thereby substantially reducing the background in the detection. Compared to the existing state-of-art techniques the proposed technique shows marked improvement. Using fluorescently-coated Saccharomyces cerevisiae cells we have recorded cell counting with throughput as high as 2,090 cells/min in the low flow rate regime and were able to image the individual cells on-the-go. Overall, the proposed system is cost-effective and simple in channel geometry with the advantage of efficient counting in operational regime of low laminar flow. This technique may advance the emerging field of microfluidic based cytometry for applications in nanomedicine and point of care diagnostics.

Abstract:Although single-cell intrinsic structural and electrical parameters (e.g. D (c) of cell diameter, D (n) of nuclear diameter, sigma (cy) of cytoplasmic conductivity and C (sm) of specific membrane capacitance) are promising for cell-type classification, they cannot be obtained simultaneously due to structural limitations of previously reported flow cytometry. This paper presented a microfluidic flow cytometry made of a double T-type constriction channel plus a predefined fluorescence detection domain, capable of high-throughput characterizing single-cell D (c), D (n), sigma (cy) and C (sm) leveraging a home-developed impedance-fluorescence model. As a demonstration, the microfluidic platform quantified D (c), D (n), sigma (cy) and C (sm) from similar to 10 000 individual cells of three well-established tumor cell lines of A549, SW620 and HeLa where successful rates of cell-type classification were estimated as 54.5 +/- 1.3% (D (c)), 68.9 +/- 6.8% (D (c) + D (n)) and 84.8 +/- 4.4% (D (c), D (n), sigma (cy) + C (sm)) based on neural pattern recognition. Then D (c), D (n), sigma (cy) and C (sm) derived from similar to 10 000 single cells of K562 vs Jurkat of leukemia and SACC-LM vs CAL 27 of oral tumor were quantified and compared, where successful rates of cell-type classification were estimated as 87.3% (K562 vs Jurkat) and 79.5% (SACC-LM vs CAL 27), respectively. In summary, the microfluidic platform reported in this study could quantify single-cell intrinsic structural and electrical parameters simultaneously, leading to significant increases in successful rates of cell-type classification.
A high-throughput all-optical laser-scanning imaging flow cytometer with biomolecular specificity and subcellular resolution

Abstract:Image-based cellular assay advances approaches to dissect complex cellular characteristics through direct visualization of cellular functional structures. However, available technologies face a common challenge, especially when it comes to the unmet need for unraveling population heterogeneity at single-cell precision: higher imaging resolution (and thus content) comes at the expense of lower throughput, or vice versa. To overcome this challenge, a new type of imaging flow cytometer based upon an all-optical ultrafast laser-scanning imaging technique, called free-space angular-chirp-enhanced delay (FACED) is reported. It enables an imaging throughput (>20 000 cells s-1 ) 1 to 2 orders of magnitude higher than the camera-based imaging flow cytometers. It also has 2 critical advantages over optical time-stretch imaging flow cytometry, which achieves a similar throughput: (1) it is widely compatible to the repertoire of biochemical contrast agents, favoring biomolecular-specific cellular assay and (2) it enables high-throughput visualization of functional morphology of individual cells with subcellular resolution. These capabilities enable multiparametric single-cell image analysis which reveals cellular heterogeneity, for example, in the cell-death processes demonstrated in this work-the information generally masked in non-imaging flow cytometry. Therefore, this platform empowers not only efficient large-scale single-cell measurements, but also detailed mechanistic analysis of complex cellular processes.

Abstract:In this paper, we present a microfluidic flow cytometer for simultaneous imaging and dielectric characterization of individual biological cells within a flow. Utilizing a combination of dielectrophoresis (DEP) and high-speed imaging, this system offers a dual-modality approach to analyze both cell morphology and dielectric properties, enhancing the ability to analyze, characterize, and discriminate cells in a heterogeneous population. A high-speed camera is used to capture images of and track multiple cells in real-time as they flow through a microfluidic channel. A wide channel is used, enabling analysis of many cells in parallel. A coplanar electrode array perpendicular to cell flow is incorporated at the bottom of the channel to perform dielectrophoresis-based dielectric characterization. A frequency-dependent voltage applied to the array produces a non-uniform electric field, translating cells to higher or lower velocity depending on their dielectric polarizability. In this paper, we demonstrate how cell size, obtained by optical imaging, and DEP response, obtained by particle tracking, can be used to discriminate viable and non-viable Chinese hamster ovary cells in a heterogeneous cell culture. Multiphysics electrostatic-fluid dynamics simulation is used to develop a relationship between cell incoming velocity, differential velocity, size, and the cell's polarizability, which can subsequently be used to evaluate its physiological state. Measurement of a mixture of polystyrene microspheres is used to evaluate the accuracy of the cytometer.

Abstract:We present a novel, low-cost, portable smartphone-based fluorescence microscope capable of directly detecting single molecules without signal amplification. The setup leverages the image sensors and data handling capacity of mass-produced smartphones, making it adaptable to any smartphone and capable of detecting single molecules across the visible spectral range. We showcase this capability through single-molecule measurements on DNA origami models and super-resolution microscopy of biological cells by single-molecule localization microscopy. This development paves the way for biotechnology innovations making use of massively distributed or personalized assays with single-molecule sensitivity with the potential to revolutionize digital bioassays, point-of-care diagnostics, field expeditions, STEM outreach, and life science education.

High‐Throughput Parallel Optofluidic 3D‐Imaging Flow Cytometry

Abstract:3D light‐sheet microscopy is a powerful tool to obtain content‐rich information of cell populations. However, the limited frame rate of high‐quantum‐efficiency cameras hinders its application in high‐throughput flow cytometry. Herein, a high‐throughput 3D‐imaging flow cytometry technique based on light‐sheet microscopy that can screen thousands of cells per second is introduced. This method enables fast, parallel optofluidic scanning of cells by performing 1D acoustofluidic focusing on multiple cells under wide‐field light‐sheet microscopy with a single objective lens. Multicolor 3D‐imaging flow cytometry with cell‐line samples at a record detection throughput of over 2000 cells s−1 is demonstrated. Furthermore, an unprecedentedly large‐scale 3D‐morphology‐based flow cytometric analysis at an order of 105 cells is demonstrated. Results show that the system is able to capture subcellular structures even at high throughputs and obtain cellular information that is overlooked under 2D‐imaging cytometry. High‐throughput 3D‐imaging flow cytometry is realized by parallel light‐sheet imaging of cells flowed with 1D acoustofluidic focusing. With a maximum detection throughput of over 2000 cells s−1, the technology enables large‐scale (>105 cells) 3D‐morphology‐based analysis for practical 3D‐imaging flow cytometry.

Abstract:Abstract Imaging flow cytometry (IFC) combines flow cytometry and fluorescence microscopy to enable high-throughput, multiparametric single-cell analysis with rich spatial details. However, current IFC techniques remain limited in their ability to reveal subcellular information with a high 3D resolution, throughput, sensitivity, and instrumental simplicity. In this study, we introduce a light-field flow cytometer (LFC), an IFC system capable of high-content, single-shot, and multi-color acquisition of up to 5,750 cells per second with a near-diffraction-limited resolution of 400-600 nm in all three dimensions. The LFC system integrates optical, microfluidic, and computational strategies to facilitate the volumetric visualization of various 3D subcellular characteristics through convenient access to commonly used epi-fluorescence platforms. We demonstrate the effectiveness of LFC in assaying, analyzing, and enumerating intricate subcellular morphology, function, and heterogeneity using various phantoms and biological specimens. The advancement offered by the LFC system presents a promising methodological pathway for broad cell biological and translational discoveries, with the potential for widespread adoption in biomedical research.

Abstract:Smartphone technology has been recently applied for biomedical image acquisition and data analysis due to its high-quality imaging capability, and flexibility to customize multi-purpose apps. In this work, we developed and characterized a smartphone-microfluidic fluorescence imaging system for studying the physiology of pancreatic islets. We further evaluated the system capability by performing real-time fluorescence imaging on mouse islets labeled with either chemical fluorescence dyes or genetically encoded fluorescent protein indicators (GEFPIs). Our results showed that the system was capable of analyzing key beta-cell insulin stimulator-release coupling factors in response to various stimuli with high-resolution dynamics. Furthermore, the integration of a microfluidics allowed high-resolution detection of insulin secretion at single islet level. When compared to conventional fluorescence microscopes and macro islet perifusion apparatus, the system has the advantages of low cost, portable, and easy to operate. With all of these features, we envision that this smartphone-microfluidic fluorescence imaging system can be applied to study islet physiology and clinical applications.

Abstract:Flow cytometry is among the most powerful tools for single-cell analysis, while the high cost and mechanical complexity of the commercial instrumentation limit the applications in personalized single-cell analysis. For this issue, we hereby construct an open and low-cost flow cytometer. It is highly compact to integrate the functions of (1) single cell aligning by a lab-made modularized 3D hydrodynamic focusing device, and (2) fluorescence detection of the single cells by a confocal laser-induced fluorescence (LIF) detector. The ceiling cost of the entire hardware for the LIF detection unit and 3D focusing device is 400 respectively. A sheath flow velocity of 150 μL/min produces a focused sample stream of 17.6 μm × 14.6 μm at sample flow of 2 μL/min, based on the LIF response frequency and the laser beam spot diameter. The assay performance of the flow cytometer was evaluated by characterizing fluorescent microparticles and acridine orange (AO) stained HepG2 cells, producing throughputs of 40.5/s and 6.2/s respectively. Favorable assay precision and accuracy were demonstrated by the agreement of frequency histogram with imaging analysis, and good Gaussian-like distributions of fluorescent microparticles and AO-stained HepG2 cells. Abstract:There are numerous diagnostic and therapeutic applications for the detection and enumeration of specific cell types. Flow cytometry is the gold standard technique for this purpose but is poorly suited to point-of-need assays. The ideal platform for these assays would combine the immunocytochemical capabilities of flow cytometry with low-cost, portable instrumentation, and a simple and rapid assay workflow. Here, we present a smartphone-based imaging platform (SIP) in tandem with magnetic-fluorescent suprananoparticle assemblies as a step toward these ideal criteria. The assemblies (MNP@QD) are magnetic iron oxide nanoparticles surrounded by a dense corona of many brightly luminescent semiconductor quantum dots (QDs), where both the assemblies and their immunoconjugates are prepared by self-assembly. As proof of concept, we show that the MNP@QD and SIP pairing is able to selectively isolate, fluorescently immunolabel, and count breast cancer cells that are positive for human epidermal growth factor receptor 2 (HER2). These results are an important foundation for future point-of-need diagnostics capable of multiplexed isolation, counting, and immunoprofiling of cells on a smartphone, enabled by the highly advantageous optical properties of QDs.

Abstract:The study presents a computational hyperspectral microflow cytometry capable of detecting the full fluorescence emission spectrum from individual flowing cells within the 450 to 650 nm spectral range, achieving a high spectral resolution of ≈3 nm in a compact form‐factor. Miniaturized flow cytometry has significant potential for portable applications, such as cell‐based diagnostics and the monitoring of therapeutic cell manufacturing, however, the performance of current techniques is often limited by the inability to resolve spectrally‐overlapping fluorescence labels. Here, the study presents a computational hyperspectral microflow cytometer (CHC) that enables accurate discrimination of spectrally‐overlapping fluorophores labeling single cells. CHC employs a dispersive optical element and an optimization algorithm to detect the full fluorescence emission spectrum from flowing cells, with a high spectral resolution of ≈3 nm in the range from 450 to 650 nm. CHC also includes a dedicated microfluidic device that ensures in‐focus imaging through viscoelastic sheathless focusing, thereby enhancing the accuracy and reliability of microflow cytometry analysis. The potential of CHC for analyzing T lymphocyte subpopulations and monitoring changes in cell composition during T cell expansion is demonstrated. Overall, CHC represents a major breakthrough in microflow cytometry and can facilitate its use for immune cell monitoring.
