Abstract:Background: White rot fungi produce various reactive oxygen species and ligninolytic enzymes for lignocellulose deconstruction. However, their interactions during the deconstruction of lignocellulosic structural barriers for efficient enzymatic saccharification remain unclear. Results: Herein, the extracellular enzyme activities and secretomic analysis revealed the sequential expression of hydroxyl radical (⋅OH) and manganese peroxidases (MnPs) for lignocellulose deconstruction by the white rot fungus Irpex lacteus. Subsequently, in vitro functional studies found that ⋅OH possessed the ability to disrupt the smooth surface structure of corn stover, resulting in increased enzymatic saccharification and cellulose accessibility. Purified recombinant MnPs from I. lacteus were able to cleave the β-O-4 bond in phenolic and non-phenolic lignin model dimers without the help of any mediators. Furthermore, the sequential pretreatment of corn stover with ⋅OH and MnP exhibited significant synergistic effects, increasing enzymatic saccharification and cellulose accessibility by 2.9-fold and 1.8-fold, respectively. Conclusions: These results proved for the first time the synergistic effects of ⋅OH and MnP pretreatment in improving the enzymatic saccharification and cellulose accessibility of corn stover. These findings also demonstrated the potential application of ⋅OH and MnP pretreatment for the efficient enzymatic saccharification of corn stover.

Abstract:Lytic polysaccharide monooxygenase (LPMO) is known as an oxidatively cleaving enzyme in recalcitrant poly-saccharide deconstruction. Herein, we report a novel AA10 LPMO derived from Bacillus subtilis (BsLPMO10A). A substrate specificity study revealed that the enzyme exhibited an extensive active-substrate spectrum, particu-larly for polysaccharides linked via 8-1,4 glycosidic bonds, such as 8-(Man1-* 4Man), 8-(Glc1-* 4Glc) and 8-(Xyl1-* 4Xyl). HPAEC-PAD and MALDI-TOF-MS analyses indicated that BsLPMO10A dominantly liberated native oligosaccharides with a degree of polymerization (DP) of 3-6 and C1-oxidized oligosaccharides ranging from DP3ox to DP6ox from mixed linkage glucans and beechwood xylan. Due to its synergistic action with a variety of glycoside hydrolases, including glucanase IDSGLUC5-38, xylanase TfXYN11-1, cellulase IDSGLUC5-11 and chitinase BtCHI18-1, BsLPMO10A dramatically accelerated glucan, xylan, cellulose and chitin saccharifi-cation. After co-reaction for 72 h, the reducing sugars in Icelandic moss lichenan, beechwood xylan, phosphoric acid swollen cellulose and chitin yielded 3176 +/- 97, 7436 +/- 165, 649 +/- 44, and 2604 +/- 130 mu mol/L, which were 1.47-, 1.56-, 1.44-and 1.25-fold higher than those in the GHs alone groups, respectively (P < 0.001). In addition, the synergy of BsLPMO10A and GHs was further validated by the degradation of natural feedstuffs, the co-operation of BsLPMO10A and GHs released 3266 +/- 182 and 1725 +/- 107 mu mol/L of reducing sugars from Oryza sativa L. and Arachis hypogaea L. straws, respectively, which were significantly higher than those produced by GHs alone (P < 0.001). Furthermore, BsLPMO10A also accelerated the liberation of reducing sugars from Celluclast (R) 1.5 L, a commercial cellulase cocktail, on filter paper, A. hypogaea L. and O. sativa L. straws by 49.58 % (P < 0.05), 72.19 % (P < 0.001) and 54.36 % (P < 0.05), respectively. This work has characterized BsLPMO10A with a broad active-substrate scope, providing a promising candidate for lignocellulosic biomass biorefinery.

Abstract:Sugarcane bagasse (SCB), a widely available lignocellulosic by-product, could be potentially used in macrofungal bioconversion for high-value bioactive compounds. For macrofungus with poor lignin degradation capacity, its recalcitrant lignin structure blocked the release of fermentable sugars for cell growth. Herein, the main objective was to construct an effective delignification pretreatment strategy for SCB biotransformation and synthesis of bioactive substances by Agaricus sinodeliciosus var. Chaidam. Different pretreatments including chemical (alkali and Fenton oxidation) and ultrasound methods were used to intensify the delignification process. Physico-chemical characteristics of native and pretreated SCB were examined via XRD, XPS, FT-IR and C-13 ssNMR to characterize crystallinity index, surface lignin and chemical structure. Lignocellulose-degrading enzymes were also used to assess the ability of the macrofungus to degrade SCB. By comparing morphological structure, chemical composition and fermentation characteristics, a suitable multiple sequential pretreatment method was finally identified. Our findings revealed that Fenton-ultrasound-alkali pre-treatment has an optimum delignification effect on SCB with never-uncovered excellent ability to detoxify harmful substances produced by Fenton oxidation. Under this pretreatment condition, the cellulose content was increased by 106.45%, whereas hemicelluloses and lignin were decreased by 48.34% and 74.15%, respectively. The macrofungal growth, intracellular polysaccharides, total phenols and terpenoids formation were 2.4, 1.46, 1.36 and 1.66 times higher than those of the untreated, respectively. Fermentation kinetic results showed outstanding fit and prediction of macrofungal growth and enzymatic activity secretion on above pretreated SCB. This study shed light on developing a novel pretreatment strategy for low-cost SCB lignocellulose transformation into high-value products.

Abstract:Even though the discovery of lytic polysaccharide monooxygenases (LPMOs) has fundamentally shifted our understanding of biomass degradation, most of the current studies focused on their roles in carbohydrate oxidation. However, no study demonstrated if LPMO could directly participate to the process of lignin degradation in lignin-degrading microbes. This study showed that LPMO could synergize with lignin-degrading enzymes for efficient lignin degradation in white-rot fungi. The transcriptomics analysis of fungi Irpex lacteus and Dichomitus squalens during their lignocellulosic biomass degradation processes surprisingly highlighted that LPMOs co-regulated with lignin-degrading enzymes, indicating their more versatile roles in the redox network. Biochemical analysis further confirmed that the purified LPMO from I. lacteus CD2 could use diverse electron donors to produce H2O2, drive Fenton reaction, and synergize with manganese peroxidase for lignin oxidation. The results thus indicated that LPMO might uniquely leverage the redox network toward dynamic and efficient degradation of different cell wall components.

Abstract:Recently low-molecular lignin was reported to activate lytic polysaccharide monooxygenases (LPMOs) to oxidize cellulose. However, whether lignin formed in cell wall can play the role as electron donor for LPMOs is still largely unknown due to the complex ultrastructure of lignocellulosic biomass. In this study, we presented a new strategy to elucidate in-situ lignin function in LPMOs reaction. A lignocellulosic mimicking model was used as substrate, which was equipped with a polysaccharide template of self-assembled bacterial cellulose film and synthesized lignin. Remarkably, it has been demonstrated that lignin polymer deposited on cellulose can reduce LPMOs in-situ for cellulose oxidation and then boost cellulose hydrolysis, and the cellulose conversion ratio of the mimicked lignocellulosic film was increased by 26.0%. More importantly, lignin in-situ might exceed the well-known reductant of ascorbic acid to drive LPMOs for cellulase enzymatic hydrolysis with equivalent cellulose oxidation efficiency and extremely lower H2O2 generation, avoiding the inactivation of enzymes. The maximum H2O2 yield from lignin-driven LPMO reaction was 75.8% lower than that from ascorbic acid-driven reaction. Therefore, by using the lignocellulosic mimicking model, we have elucidated the function of in-situ lignin in boosting enzymatic hydrolysis. Such understanding could significantly promote current utilization of LPMOs in lignocellulosic biorefinery.

Abstract:Lytic polysaccharide monooxygenases (LPMOs) can improve the effectiveness with which agricultural waste is utilized. This study described the potent AA9 family protein Ms LPMO3, derived from Morchella sextelata . It exhibited strong binding to phosphoric acid swollen cellulose (PASC), and had the considerable binding ability to Cu 2+ with a K d value of 2.70 μM by isothermal titration calorimetry (ITC). Ms LPMO3 could also act on PASC at the C1 carbon via MALDI-TOF-MS results. Moreover, Ms LPMO3 could boost the hydrolysis efficiency of corncob and wheat bran in combination with glycoside hydrolases. Ms LPMO3 also exhibited strong oxidizing ability for 2,6-dimethoxyphenol (2,6-DMP), achieving the best V max value of 443.36 U·g -1 for pH 7.4 with a H 2 O 2 concentration of 300 μM. The structure of Ms LPMO3 was obtained using AlphaFold2, and the molecular docking results elucidated the specific interactions and key residues involved in the recognition process between Ms LPMO3 and cellulose. Altogether, this study expands the knowledge of AA9 family proteins in cellulose degradation, providing valuable insights into the mechanisms of synergistic degradation of lignocellulose with cellulases.

Abstract:Although lytic polysaccharide monooxygenase (LPMO) genes are highly abundant in the genomes of ascomycetes and basidiomycetes, their biological roles in single species for lignocellulose degradation remain unclear. Herein, transcriptomic analysis of the efficient biomass degrader Myceliophthora thermophila grown on corn straw revealed a high number and transcript abundance of LPMOs and glycoside hydrolases (GHs) in response to lignocellulose degradation. To gain a deeper understanding of the roles of LPMOs, phenotypic characterization of single gene deletion mutants and catalytic properties of recombinant LPMOs were valuated. Single gene deletion mutants of these highly expressed AA9 LPMOs exhibited no significant reduction in growth rate on corn straw compared to the wild-type strain. Functional characterization of these AA9 LPMOs showed significant differences in oxidative regioselectivity, catalytic activity, and substrate specificity. Notably, MtLPMO9G exhibited dual catalytic activities, degrading cellulose and xylan through oxidation and hydrolysis reactions, respectively. Moreover, synergistic actions between AA9 LPMOs and GHs from various families on enzymatic saccharification of corn straw with alkaline pretreatment showed that MtLPMO9G and MtLPMO9H increased the catalytic efficiency of cellulolytic GHs by 28 to 242%. These findings demonstrate great application potential of MtLPMO9G and MtLPMO9H in designing LPMO-containing cellulase cocktails for efficient enzymatic saccharification of lignocellulosic biomass.

Abstract:A carbon-based material was synthesized using potato peels (BPP) and banana pseudo-stems (BPS), both of which were modified with manganese to produce BPP-Mn and BPS-Mn, respectively. These materials were assessed for their ability to activate peroxymonosulfate (PMS) in the presence of MnCO3 to degrade acetaminophen (ACE), an emerging water contaminant. The materials underwent characterization using spectroscopic, textural, and electrochemical techniques. Manganese served a dual function: enhancing adsorption properties and facilitating the breaking of peroxide bonds. Additionally, carbonate ions played a structural role in the materials, transforming into CO2 at high temperatures and thereby increasing material porosity, which improved adsorption capabilities. This presents a notable advantage for materials that have not undergone de-lignification. Among the materials tested, BPS exhibited the highest efficiency in the carbocatalytic degradation of ACE, achieving a synergy index of 1.31 within just 5 min, with 42% ACE degradation in BPS compared to BPS-Mn, which achieved 100% ACE removal through adsorption. Reactive oxygen species such as sulfate, hydroxyl, and superoxide anion radicals were identified as the primary contributors to pollutant degradation. In contrast, no degradation was observed for BPP and BPP-Mn, which is likely linked to the lower lignin content in their precursor material. This work addressed the challenge of revalorizing lignocellulosic waste by highlighting its potential as an oxidant for emerging pollutants. Furthermore, the study demonstrated the coexistence of various reactive oxygen species, confirming the capacity of carbon-based matrices to activate PMS.

Abstract:Background: The discovery of lignin as activator for the redox enzyme lytic polysaccharide monooxygenases (LPMOs) for the oxidation of cell-wall polysaccharides opens a new scenario for investigation of the interplay between different lignocellulose-degrading enzymes. The lignin-active enzymes in one hand, and the carbohydrate active in the other, are linked through a variety of electrons carrier molecules either derived from lignin or enzymatically transferred. Likewise, in nature, many lignocellulose-degrading organisms are expressing those enzymes simultaneously, and we wanted to test if a major commercial available lignin oxidase enzyme, i.e., laccase could benefit and synergize the activity of the LPMOs by depolymerizing the insoluble lignin. Results: In this work, two fungal laccases together with a mediator (ABTS) were used to isolate low-molecular-weight lignin from lignocellulosic biomass. The isolated lignins were used as electron donors for activation of LPMOs. A direct correlation between the low-molecular-weight lignin isolated with laccases and an increased activity of a cellulolytic cocktail containing LPMO was found when pure cellulose was hydrolyzed. We then tried to implement existing commercial cellulases cocktail with laccase enzymes, but under the conditions tested, the co-incubation of laccases with LPMOs showed a substrate competition towards oxygen inhibiting the LPMO. In addition, we found that laccase treatment may cause other modifications to pure cellulose, rendering the material more recalcitrant for enzymatic saccharification. Conclusions: Laccase-mediated system was able to depolymerize lignin from pre-treated and native sugarcane bagasse and wheat straw, and the released phenolic molecules were able to donate electrons to LPMO enzymes boosting the overall enzymatic hydrolysis of cellulose. Likewise, other poly-phenol oxidase, we might have just started showing possible pros or cons in applying several oxidase enzymes for a simultaneous degradation of cellulose and lignin, and we found that the competition towards oxygen and their different consumption rates must be taken into account for any possible co-application.

Abstract:Abstract Background Biochemical conversion of lignocellulosic biomass to simple sugars at commercial scale is hampered by the high cost of saccharifying enzymes. Lytic polysaccharide monooxygenases (LPMOs) may hold the key to overcome economic barriers. Recent studies have shown that controlled activation of LPMOs by a continuous H 2 O 2 supply can boost saccharification yields, while overdosing H 2 O 2 may lead to enzyme inactivation and reduce overall sugar yields. While following LPMO action by ex situ analysis of LPMO products confirms enzyme inactivation, currently no preventive measures are available to intervene before complete inactivation. Results Here, we carried out enzymatic saccharification of the model cellulose Avicel with an LPMO-containing enzyme preparation (Cellic CTec3) and H 2 O 2 feed at 1 L bioreactor scale and followed the oxidation–reduction potential and H 2 O 2 concentration in situ with corresponding electrode probes. The rate of oxidation of the reductant as well as the estimation of the amount of H 2 O 2 consumed by LPMOs indicate that, in addition to oxidative depolymerization of cellulose, LPMOs consume H 2 O 2 in a futile non-catalytic cycle, and that inactivation of LPMOs happens gradually and starts long before the accumulation of LPMO-generated oxidative products comes to a halt. Conclusion Our results indicate that, in this model system, the collapse of the LPMO-catalyzed reaction may be predicted by the rate of oxidation of the reductant, the accumulation of H 2 O 2 in the reactor or, indirectly, by a clear increase in the oxidation–reduction potential. Being able to monitor the state of the LPMO activity in situ may help maximizing the benefit of LPMO action during saccharification. Overcoming enzyme inactivation could allow improving overall saccharification yields beyond the state of the art while lowering LPMO and, potentially, cellulase loads, both of which would have beneficial consequences on process economics.

Abstract:Rice husk is an abundant agricultural waste generated from rice production, but its application is limited. Considering its complex components, the rice husk was hydrolyzed by different enzymes to enhance its saccharification. In this study, saccharification of the rice husk by cellulase, xylosidase, and xylanase was first investigated. The synergistic effect of LPMO on the above hydrolases and different enzyme combinations in the saccharification process was then explored. Thereafter, the formulation of the enzyme cocktail and the degradation conditions were optimized to obtain the highest saccharification efficiency. The results showed that the optimum enzyme cocktail consists of Celluclast 1.5 L (83.3 mg/g substrate), the key enzymes in the saccharification process, worked with Bp Xyl (20 mg/g substrate), Bp Xyn11 (24 mg/g substrate), and R17L/N25G (4 mg/g substrate). The highest reducing sugar concentration (1.19 mg/mL) was obtained at pH 6.0 and 60°C for 24 h. Fourier transform infrared spectroscopy and scanning electron microscopy were employed to characterize the structural changes in the rice husk after degradation. The results showed that the key chemical bonds in cellulose and hemicellulose were broken. This study illuminated the concept of saccharifying lignocellulose from rice husk using LPMO synergistically assisted combined-hydrolase including cellulase, xylosidase, and xylanase, and provided a theoretical basis for lignocellulose biodegradation.

Abstract:Hydrolyzing lignocellulose, the most abundant biomass raw material in nature, into monosaccharides by enzymes and then transforming the sugars into chemicals and fuels by biological or chemical methods is one of the most important parts of biorefining. In lignocellulosic biomass, cellulose is often tightly wrapped by lignin along with hemicelluloses. Although some pretreatments can partially remove lignin, residual lignin in pretreated substrates still physically blocks the accessibility of cellulose, non-productively adsorbs cellulases, and sterically hinders the enzymatic hydrolysis of cellulose. So, lignin is generally considered to be an inhibitor to the enzymatic hydrolysis of lignocellulosic substrates. However, new research shows that under the right conditions, lignin does not inhibit the enzymatic hydrolysis of lignocellulose, and can even increase the final sugar yield. For example, recently, some novel lignin-target pretreatments have been developed, they can reduce or overcome the inhibitory effect of lignin. And researchers also find that lignin can be functionalized and used as an activator/promoter of enzymatic hydrolysis of lignocellulose, their effectiveness is comparable to common high-efficiency surfactants. What's more, different lignin-based carriers were also synthesized to carry and recycle cellulases during the hydrolysis, they can obviously reduce the load and cost of cellulases. These works indicate that lignin can play a positive role in the enzymatic hydrolysis of lignocellulose. In order for readers to dialectically view lignin and understand its multiplicity, this review is to report the progress in the studies of how researchers tame the "lignin dog" to facilitate enzymatic hydrolysis of lignocellulose.

Abstract:This study describes a unique bi‐catalytic enzyme that combines an oxidative activity (LPMO) acting on cellulose and a hydrolase activity (Man5) acting on mannan thus facilitating the degradation of complex, co‐polymeric plant cell walls. The enzyme works by removing the most abundant cellulose‐covering softwood hemicellulose, mannan, which then allows oxidative cleavage of the underlying crystalline cellulose, which again promotes further degradation by classical cellulases. Because of the association with other complex polysaccharides, extracting and utilizing cellulose from lignocellulosic materials requires the combined action of a broad range of carbohydrate‐active enzymes, including multiple glycoside hydrolases (GHs) and lytic polysaccharide monooxygenases (LPMOs). The interplay between these enzymes and the way in which Nature orchestrates their co‐existence and combined action are topics of great scientific and industrial interest. To gain more insight into these issues, we have studied the lignocellulose‐degrading abilities of an enzyme from Caldibacillus cellulovorans (CcLPMO10‐Man5), comprising an LPMO domain, a GH5 mannanase domain and two family 3 carbohydrate‐binding modules (CBM3). Using a natural softwood substrate, we show that this enzyme promotes cellulase activity, i.e., saccharification of cellulose, both by removing mannan covering the cellulose and by oxidatively breaking up the cellulose structure. Synergy with CcLPMO10‐Man5 was most pronounced for two tested cellobiohydrolases, whereas effects were smaller for a tested endoglucanase, which is in line with the notion that cellobiohydrolases and LPMOs attack the same crystalline regions of the cellulose, whereas endoglucanases attack semi‐crystalline and amorphous regions. Importantly, the LPMO domain of CcLPMO10‐Man5 is incapable of accessing the softwood cellulose in absence of the mannanase domain. Considering that LPMOs not bound to a substrate are sensitive to autocatalytic inactivation, this intramolecular synergy provides a perfect rationale for the evolution of modular enzymes such as CcLPMO10‐Man5. The intramolecular coupling of the LPMO with a mannanase and two CBMs ensures that the LPMO is directed to areas where mannans are removed and cellulose thus becomes available.

Abstract:Conversion of lignocellulosic biomass to various renewable fuels, chemicals, materials, and electricity has attracted more and more interest in recent decades to decrease the dependence on fossil resources and reduce net CO2 emissions. Herein, we have developed an integrated process of biomass pretreatment for enzymatic hydrolysis of cellulose coupled with electricity generation with polyoxometalates (POMs) and ferric ion as electron mediators and proton carriers. The pretreatment is a charging process, and the re-oxidation of the POMs is effectively a discharging process. The pretreated substrate showed a good enzymatic digestibility (approximate to 80% cellulose conversion) within a short incubation period (<24h). Fe3+ was screened as a cheap, effective liquid mediator to transfer electrons to air, which is the terminal electron acceptor in the discharging process. The highest output power densities of 10.8 and 12.4mWcm(-2) were obtained for discharging of reduced H3PMo12O40 and H4PMo11VO40, respectively. This power density is 5000-6000 times higher than that of phenol-fueled microbial fuel cells, and 10times higher than that of a recently reported direct biomass fuel cell with an air cathode covered with Pt catalyst. Moreover, this work also provides a conceptual combination of a direct biomass fuel cell and a redox flow cell, which can be flexibly switched between electricity generation from biomass and stationary energy storage.
Enhanced lignin biodegradation by consortium of white rot fungi: microbial synergistic effects and product mapping

Abstract:Abstract Background As one of the major components of lignocellulosic biomass, lignin has been considered as the most abundant renewable aromatic feedstock in the world. Comparing with thermal or catalytic strategies for lignin degradation, biological conversion is a promising approach featuring with mild conditions and diversity, and has received great attention nowadays. Results In this study, a consortium of white rot fungi composed of Lenzites betulina and Trametes versicolor was employed to enhance the ligninolytic enzyme activity of laccase (Lac) and manganese peroxidase (MnP) under microbial synergism. The maximum enzymatic activity of Lac and MnP was individually 18.06 U mL −1 and 13.58 U mL −1 along with a lignin degradation rate of 50% (wt/wt), which were achieved from batch cultivation of the consortium. The activities of Lac and MnP obtained from the consortium were both improved more than 40%, as compared with monocultures of L. betulina or T. versicolor under the same culture condition. The enhanced biodegradation performance was in accordance with the results observed from scanning electron microscope (SEM) of lignin samples before and after biodegradation, and secondary-ion mass spectrometry (SIMS). Finally, the analysis of heteronuclear single quantum coherence (HSQC) NMR and gas chromatography–mass spectrometry (GC–MS) provided a comprehensive product mapping of the lignin biodegradation, suggesting that the lignin has undergone depolymerization of the macromolecules, side-chain cleavage, and aromatic ring-opening reactions. Conclusions Our results revealed a considerable escalation on the enzymatic activity obtained in a short period from the cultivation of the L. betulina or T. versicolor due to the enhanced microbial synergistic effects, providing a potential bioconversion route for lignin utilization.

Abstract:Along with long-term evolution, the plant cell wall generates lignocellulose and other anti-degradation barriers to confront hydrolysis by fungi. Lytic polysaccharide monooxygenase (LPMO) is a newly defined oxidase in lignocellulosic degradation systems that significantly fuels hydrolysis. LPMO accepts electrons from wide sources, such as cellobiose dehydrogenase (CDH), glucose-methanol-choline (GMC) oxidoreductases, and small phenols. In addition, the extracellular cometabolic network formed by cosubstrates improves the degradation efficiency, forming a stable and efficient lignocellulose degradation system. In recent years, using structural proteomics to explore the internal structure and the complex redox system of LPMOs has become a research hotspot. In this review, the diversity of LPMOs, catalytic domains, carbohydrate binding modules, direct electron transfer with CDH, cosubstrates, and degradation networks of LPMOs are explored, which can provide a systematic reference for the application of lignocellulosic degradation systems in industrial approaches.
