Abstract:Pheromone binding proteins(PBPs)play an important role in recognition,transmission and processing of insect pheromones.In this study,a cDNA named Ac-ASP1 encoding a PBP was cloned(from the Chinese native honeybee,Apis cerana cerana.The predicted protein has six conservative cysteines in mature peptide,a typical symbol of odorant binding proteins(OBPs).The sequence had been submitted into GenBank and registered is the accession no.DQ449670.The profile of spatio-temporal expression of Ac-ASP1 was identified by real-time PCR.The results of absolute quantitation showed that Ac-ASP1 was expressed at a higher level in antenna(2.07×106 copy number/μg),but at a less level in other tissues including head,thorax,abdomen,wings and legs(102 copy number/μg);while the results of relative quantitation revealed that Ac-ASP1 was expressed in two high abundance periods:from larva to the 6th day of adult and around the 21st day of adult.These results may provide the reference to ascertain the function of Ac-ASP1 involved in signal recognition and transmission of queen pheromone.

Abstract:The spatio-temporal expressed profiles of two kinds of chemical communication-related protein genes, the odorant-binding protein of Ac-ASP2 and chemosensory protein of Ac-ASP3, were identified by real-time polymerase chain reaction (PCR). Results obtained using the 2 -D DCt method showed that Ac-ASP2 was a gene coding antenna-specific protein that did not express in larvae and pupae, but had discontinuous high abundance periods at 1, 9, 15, 27 and 30 days. The expressing abundance at such periods was at least ten times higher than that at other periods. From the distribution ofAc-ASP3 mRNA observed in different tissues, the transcript levels seemed to be higher in the wings, abdomen and thorax (of order � 10 6 ), and lower in the legs, antennae and head (of order � 10 5 ). From highest to lowest, the original copy number was found in the various body parts in the following order: wings, abdomen, thorax, legs, antenna, and head. The results suggest that Ac-ASP3 has an intimate relation with the chemosensory behaviour of wings and abdomen in Apis cerana cerana.

Abstract:The spatio-temporal expressed profiles of two kinds of chemical communication related to genes of Apis cerana cerana proteins,odorant-binding protein of Ac-ASP2 and chemosensory protein of Ac-ASP3,were identified by Real-time PCR.Using the method of 2-ΔΔCt,the results with the method of 2-ΔΔCt showed that Ac-ASP2 was a gene coding antenna specific protein,which did not express in larva and pupa,but had discontinuous high abundance periods: 1,9,15,27 and 30 d,the expressing abundance at such pe-riods was tenfold higher than that at other periods.From the distribution of Ac-ASP3 mRNA in different issues,the transcript amounts seemed to be higher in wings,abdomen and thorax(about 106 order of magnitude),and lower in legs,antennae and head(about 105 order of magnitude),the original copy number was: wings >abdomen >thorax >legs >antennae >head.The results above showed that Ac-ASP3 had intimate relation with chemosensory behavior of wings and abdomen in Apis cerana cerana.
Identification of the Sex Pheromone of the Asparagus Moth, Parahypopta Caestrum (Lepidoptera, Cossidae)

Abstract:Chemical, electrophysiological, and field trapping experiments were carried out to identify the female-produced sex pheromone of the asparagus moth, Parahypopta caestrum , a very serious pests of asparagus cultivations in southern Europe. Gas chromatography coupled with mass spectrometry and electroantennogram detection (GC-MS-EAD) analysis of hexane and solid-phase microextraction (SPME) extracts of sex pheromone glands of calling females consistently detected four compounds eliciting EAG responses in male moth antennae. According to their GC retention times, mass spectra, and comparative EAG analyses with reference standards, these EAD-active compounds were identified as ( Z )-9-tetradecenol ( Z 9-14:OH), ( Z )-5-tetradecenyl acetate ( Z 5-14:Ac), ( Z )-7-tetradecenyl acetate ( Z 7-14:Ac), and ( Z )-9-tetradecenyl acetate ( Z 9-14:Ac), respectively. In the SPME extracts from the head-space of individual abdominal tips, Z 9-14:Ac, Z 5-14:Ac, Z 7-14:Ac, and Z 9:14 OH were detected in the ratio of 82:9:5:4. In EAG dose-response experiments, Z 9-14:Ac was the strongest antennal stimulant at different doses tested. In field trapping experiments, Z 9-14:Ac, Z 7-14:Ac, and Z 5-14:Ac proven to be essential for male attraction and a their 85:5:10 blend loaded onto green rubber septum dispensers was significantly more effective than single-, two-, and any other three-component blend of these compounds. The addition of Z 9-14:OH to the optimal blend resulted in a significant reduction of male catches. The attractive blend here identified allowed for an effective and accurate monitoring of P. caestrum flight activity in southern Italy.

Abstract:The spotted asparagus beetle, Crioceris duodecimpunctata (L.) is an invasive host-specific pest of asparagus cultivations. To contribute to the understanding of the role of plant volatiles in host-finding by this species, behavioural and electrophysiological tests were carried out. Y-tube olfactometer bioassays, testing intact or mechanically-damaged cladophylls vs. clean air, revealed sexually-dimorphic responses with males being the only sex attracted to both plant materials. Electroantennographic (EAG) assays showed that antennae of both sexes can perceive a wide range of asparagus volatiles. Male and female EAG profiles were almost similar and (Z)-3-hexen-1-ol was by far the most EAG-active compound. (E)-2-hexenal, (±)-linalool, and 3-heptanone elicited the strongest EAG amplitude within the corresponding chemical groups. Eight of the most EAG-active compounds elicited dose-dependent responses indicating the sensitivity of male and female olfactory systems to changes in stimulus concentration. In a Y-tube olfactometer bioassay, (Z)-3-hexen-1-ol at the doses of 1, 10, and 50 μg did not elicit female attraction whereas a significant attraction at the 10 μg dose and a repellent effect at the 50 μg dose was induced in males. Sexual dimorphism of male behavioural response to host plant volatiles is discussed. This study provides a basis for future investigations that could contribute to the development of semiochemical-based monitoring and management strategies for this pest.

Abstract:Pheromones are a kind of signal produced by an animal that evoke innate responses in conspecifics. In moth, pheromone components can be detected by specialized olfactory receptor neurons (OSNs) housed in long sensilla trichoids on the male antennae. The pheromone receptors (PRs) located in the dendrite membrane of OSNs are responsible for pheromone sensing in most Lepidopteran insects. The potato tuber moth Phthorimaea operculella is a destructive pest of Solanaceae crops. Although sex attractant is widely used in fields to monitor the population of P. operculella, no study has been reported on the mechanism the male moth of P. operculella uses to recognize sex pheromone components. In the present study, we cloned two pheromone receptor genes PopeOR1 and PopeOR3 in P. operculella. The transcripts of them were highly accumulated in the antennae of male adults. Functional analysis using the heterologous expression system of Xenopus oocyte demonstrated that these two PR proteins both responded to (E, Z)-4,7–13: OAc and (E, Z, Z)-4,7,10–13: OAc, the key sex pheromone components of P. operculella, whilst they responded differentially to these two ligands. Our findings for the first time characterized the function of pheromone receptors in gelechiid moth and could promote the olfactory based pest management of P. operculella in the field.

Abstract:Abstract The white-spotted flower chafer (WSFC), Protaetia brevit arsis Lewis, is native to East Asia. Although their larvae are considered a potential resource insect for degrading plant residues, producing protein fodder, and processing to traditional medicine, adult WSFCs inflict damage to dozens of fruit and economic crops. The control of the WSFC still relies heavily on pesticides and the inefficient manual extraction of adults. Here, we report the identification and evaluation of the aggregation pheromone of WSFCs. From the headspace volatiles emitted from WSFC adults, anisole, 4-methylanisole, 2-heptanone and 2-nonanone were identified as WSFC-specific components. However, only anisole and 4-methylanisole elicited positive dose–response relationship in electroantennography tests, and only 4-methylanisole significantly attracted WSFCs of both sexes in olfactometer bioassays and field experiments. These results concluded that 4-methylanisole is the aggregation pheromone of WSFCs. Furthermore, we developed polyethylene vials as long-term dispensers of 4-methylanisole to attract and kill WSFCs. The polyethylene vial lures could effectively attracted WSFCs for more than four weeks. Pheromone-based lures can be developed as an environmentally friendly protocol for monitoring and controlling WSFC adults.

Pheromone Communication in Anastrepha obliqua (Diptera: Tephritidae): A Comparison of the Volatiles and Salivary Gland Extracts of Two Wild Populations

Abstract:The West Indian fruit fly Anastrepha obliqua Macquart (Diptera: Tephritidae) is one of the major pests on mango (Mangifera indica L.; Sapindales: Anacardiaceae) and starfruit (Averrhoa carambola L.; Oxalidales: Oxalidaceae) crop plantations in Brazil. Pheromone communication inter alia plays an important role in fruit-fly courtship behavior. In order to highlight the site of pheromone synthesis, we identified and compared the volatiles from the aeration extracts of calling males with the volatiles produced by their salivary glands in 2 wild populations of A. obliqua collected from mangoes and starfruits. In addition, we performed a series of bioassays to compare the biological significance of both extracts. In total, 36 volatile compounds were identified, with 8 of them being shared by the 2 populations and the 2 extract types. Linalool and -copaene were exclusively found in the aeration extract while ethyl heptanoate, methyl octanoate, and 1-nonanol were detected only in the salivary-gland extracts. The chemical profiles of the volatiles from the aeration extracts and from the salivary-gland extracts differed significantly between the 2 populations as well as the chemical profiles of both extracts within each population. The quantities of the 8 shared compounds generated a variability of more than 60% in the mango population and 80% in the starfruit population. The similarities observed between the chemical profiles of the aeration extracts and the salivary-gland extracts suggest that the latter could be the storage site and probably also the production site of some pheromone components in this fruit-fly species. This hypothesis is supported by the comparable biological activities of both extracts in terms of their attractiveness for conspecific females.

Abstract:This research delves into plant–pollinator relationships within the Mediterranean region, focusing on two synchronous and sympatric asparagus species: A. acutifolius and A. albus. For the first time, the floral scents of the genus Asparagus are reported. We investigate the volatile organic compounds (VOCs) present in their floral scents and their impact on pollinator attraction. Captured flower-emitted VOCs underwent solid-phase microextraction of headspace (SPME-HS) and gas chromatography and mass spectrometry (GC-MS) analysis. The investigation confirms distinctive aroma profiles for each species. A. albus predominantly emits benzene derivatives and sesquiterpenes, while A. acutifolius is characterized by carotenoid derivatives, monoterpenes, and sesquiterpenes. The only shared compounds between the two species are the sesquiterpenes (Z,E)-α-farnesene and (E,E)-α-farnesene. A positive correlation links peak floral aroma intensity (benzenoids in A. albus and ionones in A. acutifolius) with a higher pollinator visit frequency, emphasizing the critical role of intense floral scents in pollinator attraction. The study of reproductive aspects reveals almost complete gynodioecy in A. acutifolius, influencing unique dynamics for the two species. These adaptations hold significant importance within the Mediterranean ecosystem, particularly during the late dry summer period, when a limited number of plant species vie for a shared primary pollinator.

Abstract:Abstract The iconic European stag beetle ( Lucanus cervus ) (Coleoptera: Lucanidae) is one of the largest terrestrial beetles in Europe. Due to decreasing population numbers, thought to be a consequence of habitat loss, this beetle has become a near-threatened species across much of Europe, and a reliable monitoring system is required to measure its future population trends. As part of a programme aimed at conserving UK populations, we have investigated the chemical ecology of the beetle, with a view to developing an efficient semiochemical-based monitoring system. Such a scheme will be beneficial not only in the UK but across the European range of the species, where the beetle is of conservation concern. Here, we report on a surprising discovery of a male-produced pheromone, which provokes initial sexual receptivity in females, and which has not been previously identified in the animal kingdom. Furthermore, we assign sex pheromone function to a previously described female-specific compound.

Ionospheric effects of whistler waves from rocket‐triggered lightning

Abstract:Lightning‐induced electron precipitation (LEP) is one of the primary mechanisms for energetic electron loss from Earth's radiation belts. While previous works have emphasized lightning location and the return stroke peak current in quantifying lightning's role in radiation belt electron loss, the spectrum of the lightning return stroke has received far less attention. Rocket‐triggered lightning experiments performed at the International Center for Lightning Research and Testing (ICLRT) at Camp Blanding, Florida, provide a means to directly measure the spectral content of individual lightning return strokes. Using an integrated set of numerical models and directly observed rocket‐triggered lightning channel‐base currents we calculate the latitudinal dependence of the precipitation signature. Model results indicate that rocket‐triggered lightning may produce detectable LEP events and that return strokes with higher ELF (3 Hz–3 kHz) content cause proportionally more ionospheric ionization and precipitate more electrons at higher latitudes than return strokes with proportionally higher VLF (3 kHz–30 kHz) content. The predicted spatio‐temporal signature of the induced electron precipitation is highly dependent upon the return stroke spectral content. As a result, we postulate that rocket‐triggered lightning experiments enable us to the estimate the spectral profile of energetic electrons precipitated from the Earth's radiation belts.

Abstract:Phorbol esters, which activate protein kinase C (PKC), enhance synaptic transmission in the CA1 subfield of hippocampus, both in situ and in vitro. The increase in synaptic transmission could be the consequence of enhanced Ca influx into nerve terminals, and perhaps a more general increase in voltage‐dependent Ca currents. The effects of phorbol 12, 13‐diacetate (PDAc) on the high‐voltage activated (HVA) Ca currents, as well as spontaneous transient currents were therefore investigated by intracellular recording in hippocampal slices. PDAc selectively augmented, by 45% ± 10%, the early peak of the HVA Ca current (but not its sustained component), and also spontaneous inhibitory postsynaptic currents. The inactive phorbol ester, 4 α‐PDAc, had no comparable effects. The actions of PDAc were reversible on prolonged washing, and they were antagonized by the PKC inhibitors (1‐(5‐isoquinolinesulfonyl)‐2‐methyl) piperazine (H‐7) and monosialoganglioside (GM1). In addition, GM1, which also activates the Ca/calmodulin‐dependent kinase, enhanced spontaneous excitatory postsynaptic currents, while inhibiting the IPSCs. It is concluded that activation of PKC increases HVA (probably N‐type) Ca current and facilitates ongoing GABAergic IPSCs.
A plant volatile alters the perception of sex pheromone blend ratios in a moth

Abstract:Mate finding in most moths is based on male perception of a female-emitted pheromone whose species specificity resides in component chemistry and proportions. Components are individually detected by specialized olfactory receptor neurons (ORNs) projecting into the macroglomerular complex (MGC) of the male brain. We asked how robust ratio recognition is when challenged by a plant volatile background. To test this, we investigated the perception of the pheromone blend in <i>Agrotis ipsilon,</i> a moth species whose females produce a blend of Z7-dodecenyl acetate (Z7-12:Ac), Z9-tetradecenyl acetate (Z9-14:Ac), and Z11-hexadecenyl acetate in a 4:1:4 ratio optimally attractive for males. First, we recorded the responses of specialist ORNs for Z7 and Z9 and showed that heptanal, a flower volatile, activated Z7 but not Z9 neurons. Then, we recorded intracellularly the responses of MGC neurons to various ratios and showed that heptanal altered ratio responses of pheromone-sensitive neurons. Finally, we analyzed the behavior of males in a wind tunnel and showed that their innate preference for the 4:1:4 blend was shifted in the presence of heptanal. Pheromone ratio recognition may thus be altered by background odorants. Therefore, the olfactory environment might be a selective force for the evolution of pheromone communication systems.
A predicted sex pheromone receptor of codling moth Cydia pomonella detects the plant volatile pear ester

Abstract:Plant volatiles mediate host discrimination and host finding in phytophagous insects. Understanding how insects recognize these signals is a current challenge in chemical ecology research. Pear ester, ethyl (E,Z)-2,4-decadienoate, is a powerful, bisexual attractant of codling moth Cydia pomonella (Lepidoptera, Tortricidae) and strongly synergizes the male response to female-produced sex pheromone. We show here that the codling moth odorant receptor (OR) CpomOR3 is dedicated to detecting this plant volatile. Heterologous expression of CpomOR3 in Drosophila T1 trichoid and ab3A basiconic sensilla, followed by a screening with codling moth pheromone compounds and known plant volatile attractants, confirms that CpomOR3 binds to pear ester. Although CpomOR3 does not respond to any of the pheromone components tested, a phylogenetic analysis of lepidopteran chemosensory receptor genes reveals a close relationship of CpomOR3 with pheromone receptors (PRs) in moths. This corroborates the interaction of ecological and social chemosensory cues during premating communication. The finding that a plant volatile compound, pear ester, is a specific ligand for a PR-like lepidopteran receptor adds to our understanding of insect-plant interactions and emphasizes the interaction of natural and sexual selection during the phylogenetic divergence of insect herbivores.

Abstract:The lacewing Chrysopa phyllochroma Waesmael is a polyphagous predator of many pests. Releasing lacewings is an important component of biological control programs, but it is difficult to establish populations on field crops. Electrophysiological and behavioral responses to 10 common plant volatiles were recorded to screen for lacewing-attracting compounds. Electroantennographic assays indicated that all of the tested compounds elicited responses from C. phyllochroma. Three green-leaf volatiles-(E)-2-hexenal, (Z)-3-hexenyl acetate, and (Z)-3-hexenol-produced the strongest responses. Weaker responses were observed to six terpenes-ocimene, linalool, (3E)-4,8-dimethyl-1,3,7-nonatriene, (E,E)-α-farnesene, limonene, and nerolidol-and to methyl salicylate. Using a Y-tube olfactometer, the behavioral assays of the eight most active compounds demonstrated that four-(Z)-3-hexenyl acetate, (Z)-3-hexenol, (3E)-4,8-dimethyl-1,3,7-nonatriene, and linalool-were significant attractants for C. phyllochroma at specific concentrations. Three common plant volatile compounds-(Z)-3-hexenyl acetate, (3E)-4,8-dimethyl-1,3,7-nonatriene, and linalool-were also found to significantly enhance female ovipositing, resulting in a concentration of eggs. These observations are important for lacewing release as a pest control measure because they suggest means for retaining individuals and establishing populations using common plant volatiles.

Abstract:Highly developed chemoreception allows insects to detect foods, find mates, and escape natural enemies. We described the structures and distributions of antennal chemosensilla in Adelphocoris fasciaticollis Reuter by scanning electron microscopy and transmission electron microscopy. Seven major types of antennal sensilla were identified in adults of both sexes. Types 1 and 2 are sensilla chaetica and have thick cuticular walls with conspicuous grooves at their surfaces. Types 3 and 4 are multiporous sensilla trichodea with 1–3 dendrites located at the sensillum lymph, indicating a putative olfactory function. Types 5 and 6 are typical sensilla basiconica but share different characteristics in both external morphology and internal ultrastructure, and may be involved in the perception of host-associated odorants. The last sensilla were Böhm bristles. In addition to the morphological characterization, electrophysiological responses of antennal chemosensilla to 51 semiochemicals were investigated based on electroantennogram (EAG) recordings. Results revealed that different chemical stimuli elicited significantly different dose-dependent EAG responses, in which potential sex pheromone components and green leaf volatiles showed relatively higher EAG responses, but neither monoterpenes nor sesquiterpenes can elicit favorable EAG values. The results provided direct morphological and electrophysiological evidence that the adult antennae of A. fasciaticollis could function in searching for mates and host plants.

Abstract:Agriotes ustulatus is an economically important click beetle in Europe. A female-produced pheromone, (E,E)-farnesyl acetate, has been identified and is used for monitoring and detecting males. More recently, a floral lure targeting females with modest, but significant, activity has been described. Based on preliminary data, we hypothesized, that similar to the effects on the congeneric A. brevis, addition of the pheromone to the floral lure should improve female A. ustulatus catches. Also, as click beetles have been reported to respond to white light, we studied possible interactions between visual and chemical cues. In field trials, the addition of the synthetic pheromone to the floral lure resulted in a dramatic increase in the number of females trapped, whereas male catches remained unaffected and equal to those in traps baited with pheromone only. A white visual cue did not influence trap catches. Maximum catches of both sexes of A. ustulatus can be achieved using the pheromone and the floral lure inside the same trap. Furthermore, the compounds can be formulated in a single polyethylene bag dispenser, making handling of the trap easier. Due to a much larger proportion of females in the catch, this improved trap may be a promising tool for semiochemical-based, environmentally sound agricultural practice against this important pest.
Comparative transcriptome analysis of pheromone biosynthesis-related gene expressions in Plutella xylostella (L.)

Abstract:The pheromone biosynthesis in Plutella xylostella is more active in the scotophase than that of the photophase, indicating that there may be changes of gene expression in the pheromone gland according to the photoperiod. To identify genes contributing to change in pheromone production, we analyzed transcriptomes of pheromone glands from both decapitated females (PG-minus) in the photophase and normal ones (PG-plus) in the scotophase. Deep sequencing for mRNAs in the pheromone gland yielded approximately 7.5Gb and 10,303 transcripts showing positive FPKM value were analyzed. Differentially expressed gene analysis revealed that up- and down-regulated transcripts were 310 and 326 in the PG-plus transcriptome, respectively. Genes putatively involved in the pheromone biosynthesis pathway were identified such as acetyl-CoA carboxylase, acetyl-CoA dehydrogenase, fatty acid synthase (FAS), desaturases (Δ9 and Δ 11) and fatty acid reductases of pheromone gland (pgFAR), alcohol oxidase, aldehyde oxidase and aldehyde reductase, etc. Quantitative RT-PCR revealed that expressions of FAS, Δ11 desaturase and pgFAR were significantly higher in PG-plus, suggesting that they may have crucial roles in sex pheromone biosynthesis of P. xylostella.

Abstract:Volatile sex pheromones are vital for sexual communication between males and females. Females of the American cockroach, Periplaneta americana , produce and emit two sex pheromone components, periplanone-A (PA) and periplanone-B (PB). Although PB is the major sex attractant and can attract males, how it interacts with PA in regulating sexual behaviors is still unknown. In this study, we found that in male cockroaches, PA counteracted PB attraction. We identified two odorant receptors (ORs), OR53 and OR100, as PB/PA and PA receptors, respectively. OR53 and OR100 were predominantly expressed in the antennae of sexually mature males, and their expression levels were regulated by the sex differentiation pathway and nutrition-responsive signals. Cellular localization of OR53 and OR100 in male antennae further revealed that two types of sensilla coordinate a complex two-pheromone-two-receptor pathway in regulating cockroach sexual behaviors. These findings indicate distinct functions of the two sex pheromone components, identify their receptors and possible regulatory mechanisms underlying the male-specific and age-dependent sexual behaviors, and can guide novel strategies for pest management.

Abstract:Sex pheromones are pivotal for insect reproduction. However, the mechanism of sex pheromone communication remains enigmatic in hymenopteran parasitoids. Here we have identified the sex pheromone and elucidated the olfactory basis of sex pheromone communication in Campoletis chlorideae (Ichneumonidae), a solitary larval endoparasitoid of over 30 lepidopteran pests. Using coupled gas chromatography-electroantennogram detection, we identified two female-derived pheromone components, tetradecanal (14:Ald) and 2-heptadecanone (2-Hep) (1:4.6), eliciting strong antennal responses from males but weak responses from females. We observed that males but not females were attracted to both single components and the blend. The hexane-washed female cadavers failed to arouse males, and replenishing 14:Ald and 2-Hep could partially restore the sexual attraction of males. We further expressed six C. chlorideae male-biased odorant receptors in Drosophila T1 neurons and found that CchlOR18 and CchlOR47 were selectively tuned to 14:Ald and 2-Hep, respectively. To verify the biological significance of this data, we knocked down CchlOR18 and CchlOR47 individually or together in vivo and show that the attraction of C. chlorideae to their respective ligands was abolished. Moreover, the parasitoids defective in either of the receptors were less likely to court and copulate. Finally, we show that the sex pheromone and ( Z )-jasmone, a potent female attractant, can synergistically affect behaviors of virgin males and virgin females and ultimately increase the parasitic efficiency of C. chlorideae . Our study provides new insights into the molecular mechanism of sex pheromone communication in C. chlorideae that may permit manipulation of parasitoid behavior for pest control.
