Abstract:To elucidate the roles of microorganisms in the nitrogen geo-chemical cycle, the distributions of nitrogen-utilising bacteria were studied in a small eutrophic lake. Ammonifiers, nitrogen fixers and nitrate reducers were more concentrated at the severely polluted Xili sub-area than at the Centre of Outer (CO), whereas the contrary was true for autolithotrophic nitrifiers. Molecular identification of predominant ammonifiers and nitrogen fixers showed that two of the ammonifiers were gamma-Proteobacteria, two were beta-Proteobacteria, one was CFB group and the other three were low G+C% Gram positive Bacillus. Among four nitrogen fixers, one belonged to alpha-Proteobacteria, and the other three were gamma-Proteobacteria.

Abstract:Anaerobic ammonium oxidation (anammox) is recognized as an important process for nitrogen cycling, yet little is known about its role in the subsurface biosphere. In this study, we investigated the presence, abundance, and role of anammox bacteria in upland soil cores from Tianjin, China (20 m depth) and Basel, Switzerland (10 m depth), using isotope-tracing techniques, (q)PCR assays, and 16 S rRNA & hzsB gene clone libraries, along with nutrient profiles of soil core samples. Anammox in the phreatic (water-saturated) zone contributed to 37.5–67.6% of the N-loss (up to 0.675 gN m−2d−1), with anammox activities of 0.005–0.74 nmolN g−1soil h−1, which were even higher than the denitrification rates. By contrast, no significant anammox was measured in the vadose zone. Higher anammox bacterial cell densities were observed (0.75–1.4 × 107copies g−1soil) in the phreatic zone, where ammonia-oxidizing bacteria (AOB) maybe the major source of nitrite for anammox bacteria. The anammox bacterial cells in soils of the vadose zone were all <103copies g−1soil. We suggest that the subsurface provides a favorable niche for anammox bacteria whose contribution to N cycling and groundwater nitrate removal seems considerably larger than previously known.

Abstract:Nitrate contamination in groundwater originates mainly from excessive use of fertilizers and uncontrolled discharge to land of incompletely-treated wastewater associated with agricultural activities. A systematic field investigation was carried out in a sub-catchment of Dianchi Lake, Kunming, Yunnan, China, into the hydrological, biological and geological processes of nitrogen transport and transformation in the aeration zone and aquifer system. In situ experiments showed that the quantity of NO3-N recharged into groundwater was related to fertilization. Nitrification and denitrification behaved quite differently but were affected by moisture content and Eh value. The vertical infiltration rate was controlled by the groundwater table and hydraulic conductivity of the soil. The existence of a zero-flux plane reflected the dynamics of water fluxes in the soil profile and Eh was measured in the aeration zone. In response to these factors, the nitrification rate was greatest in the top soil and reduced with the depth of soil; it was 6.53 mg/(kg.h) in the vegetated plot and 0.2-0.3 mg/(kg.h) in the unvegetated one. The denitrification rate in the unvegetated plot was 6.36 mg/(kg.h), and it was 2.79 mg/(kg.h) in the vegetated one.
Elucidating biogeochemical characterization of nitrogen in the vadose zone integrating geochemistry, microorganism, and numerical simulation

Abstract:A thorough comprehension of nitrogen biogeochemical processes in the vadose zone is crucial for the effective prevention and remediation of soil-groundwater system contamination. Despite the growing research on this subject, the full scope of nitrogen biogeochemical characterization in different geological environments remains poorly understood. This study addresses this knowledge gap by integrating geochemical, microbiological and numerical simulation approaches to gain a deeper insight into nitrogen biogeochemistry in agriculture. Our findings indicate the biogeochemical behavior of nitrogen in the vadose zone is mediated by microorganisms, driven by hydraulics, influenced by geological conditions and environmental factors. Along the groundwater flow, NH4+-N was found to be heavily accumulated in the topsoil of 0-40 cm, while NO3--N was transported and driven by hydrodynamics from both vertical and horizontal directions. Microbial diversity, species composition and functional microorganisms were significantly influenced by soil depth, rather than geomorphological types. Oxidation-reduction potential (ORP), total organic carbon (TOC), soil moisture (MOI), bicarbonate (HCO3-), and ferrous (Fe2+) were identified as the principal environmental factors that regulate nitrogen metabolism and the dominant biochemical processes, encompassing nitrogen fixation, nitrification, and denitrification. Driven by hydrodynamics, NH4+-N, NO2--N and NO3--N tend to form distinct biochemical reaction zones in the vertical vadose zone. These areas are dynamic and subject to geomorphologies. It should be noted that NO3--N can migrate towards groundwater from the clayey sand in the Alluvial Plain, which presents a potential risk of groundwater contamination. The fissure structure of loess may serve as the major transport pathway for groundwater nitrogen contamination in the Loess Tableland. This finding highlights the importance of integrating microbiology, geochemistry and hydraulics to elucidate the biogeochemical processes of nitrogen in the vadose zone with a dynamic mindset.

Abstract:The ecological environment of the ecotone of agriculture–animal husbandry in North China is fragile and the groundwater resources are in short supply, in particular, nitrogen contamination is serious. So, the nitrogen source, distribution and transformation in groundwater need to be investigated. A case study was conducted in Tuoketuo County, Inner Mongolia. Hydro-geological, hydro-geochemical, and quality monitoring of groundwater were investigated in the study area. Altogether, 66 groundwater samples were taken with an equal number from both the unconfined and confined aquifers in the study area. The results show that the ammonium compound (NH4+) was the most prevalent species of nitrogen contamination. Both the irrigations of farmland with contaminated surface water and the excessive usage of chemical fertilizers were probably two major non-point sources of nitrogen contamination. The large-scale dairy farm effluents also play an important role in nitrogen contamination in local areas of an unconfined aquifer. The nitrogen contamination in the confined aquifer was mainly due to the leakage recharge of the contaminated unconfined water. The mean concentrations of NO3−, NO2− and NH4+ in confined aquifer were significantly lower and the areas exceeding the water quality standard were similar but smaller than those in an unconfined aquifer. The spatial distributions, migration and transformation of ammonium (NH4+), nitrate (NO3−) and nitrite (NO2−) in the investigation area were closely related to the characteristics of the groundwater flow system and the hydro-geochemical environment. In the groundwater recharge area, nitrification plays a leading role, with NO3− as the major contaminant. Along the groundwater discharge zone, the denitrification, anammox and dissimilatory nitrate reduction to ammonium (NDRA) processes may occur, causing NH4+ to become the main contaminant. The Cl– contamination was also severe both in unconfined and confined aquifers. High concentrations of NO3− and NH4+ were mainly observed in the groundwater of Ca·Mg–Cl and Na–Cl types. The background type of water for unconfined and confined aquifers was mainly Ca–HCO3. This study will provide helpful information to identify the most contaminated locations in the study area for environmental management in future and provides a reference for preserving the environment of ecologically fragile areas.

Abstract:Nitrate is a significant constituent of the total nitrogen pool in shallow aquifers and poses an escalating threat to groundwater resources, making it crucial to comprehend the source, conversion, and elimination of nitrogen using appropriate techniques. Although dual-isotope dynamics in nitrate have been widely used, uncertainties remain regarding the asynchronously temporal changes in δ18O-NO3- and δ15N-NO3- observed in hypoxic aquifers. This study aimed to investigate changes in nitrogen sources and transformations using temporal changes in field-based NO3- isotopic composition, hydro-chemical variables, and environmental DNA profiling, as the groundwater table varied. The results showed that the larger enrichment in δ18O-NO3- (+13‰) compared with δ15N-NO3- (−2‰) on average during groundwater table rise was due to a combination of factors, including high 18O-based atmospheric N deposition, canopies nitrification, and soil nitrification transported vertically by rainfalls, and 18O-enriched O2 produced through microbial and root respiration within denitrification. The strong association between functional gene abundance and nitrogen-related indicators suggests that anammox was actively processed with nitrification but in small bacterial population during groundwater table rise. Furthermore, bacterial species associated with nitrogen-associated gradients provided insight into subsurface nitrogen transformation, with Burkholderiaceae species and Pseudorhodobacter potentially serving as bioindicators of denitrification, while Candidatus Nitrotogn represents soil nitrification. Fluctuating groundwater tables can cause shifts in hydro-chemical and isotopic composition, which in turn can indicate changes in nitrogen sources and transformations. These changes can be used to improve input sources for mixture models and aid in microbial remediation of nitrate.

In-situ Nitrogen Fate in the Vadose Zone of Different Soil Types and Its Implications for Groundwater Quality in the Huaihe River Basin, China

Abstract:This paper focused on nitrate fate in the vadose zone (VZ) and its implications for groundwater vulnerability under different soil types in the agricultural area of Huaihe River Basin, China. Isotopic compositions of nitrate (δ15N and δ18O) along with NO3− and Cl− concentrations were determined in the VZ-shallow groundwater continuum beneath silty-loam and silty-clay-loam, which are distinctive in texture and organic carbon (OC). In the soil zone (< 1 m in depth), measured δ18O–NO3− suggested the ubiquitous of nitrification regardless of soil types. In the subsoil zone (> 1 m in depth), however, the concurrent enrichment of δ15N–NO3− and δ18O–NO3− indicated the occurrence of denitrification, which showed a dependence on subsoil properties. Specifically, during wheat and maize land uses, denitrification removed as much as 76 %–88 % of the total nitrate where the subsoil was dominated by stratified OC-rich silty-clay-loam. In contrast, only 0 %–28 % of the nitrate was degraded via denitrification where the subsoil was composed of uniform, OC-depleted silty-loam. Furthermore, inactive denitrification and higher permeability in the silty-loam VZ implied higher groundwater vulnerability. This observation was consistent with the fact that groundwater NO3−–N concentration beneath silty-loam (11.24 mg L−1) was over two times higher than that of the silty-clay-loam (5.32 mg L−1), where stricter fertilization management and conservation strategies should be applied to protect groundwater quality.
Nitrogen cycling and microbial cooperation in the terrestrial subsurface

Abstract:The nitrogen cycle plays a major role in aquatic nitrogen transformations, including in the terrestrial subsurface. However, the variety of transformations remains understudied. To determine how nitrogen cycling microorganisms respond to different aquifer chemistries, we sampled groundwater with varying nutrient and oxygen contents. Genes and transcripts involved in major nitrogen-cycling pathways were quantified from 55 and 26 sites, respectively, and metagenomes and metatranscriptomes were analyzed from a subset of oxic and dysoxic sites (0.3-1.1 mg/L bulk dissolved oxygen). Nitrogen-cycling mechanisms (e.g. ammonia oxidation, denitrification, dissimilatory nitrate reduction to ammonium) were prevalent and highly redundant, regardless of site-specific physicochemistry or nitrate availability, and present in 40% of reconstructed genomes, suggesting that nitrogen cycling is a core function of aquifer communities. Transcriptional activity for nitrification, denitrification, nitrite-dependent anaerobic methane oxidation and anaerobic ammonia oxidation (anammox) occurred simultaneously in oxic and dysoxic groundwater, indicating the availability of oxic-anoxic interfaces. Concurrent activity by these microorganisms indicates potential synergisms through metabolite exchange across these interfaces (e.g. nitrite and oxygen). Fragmented denitrification pathway encoding and transcription was widespread among groundwater bacteria, although a considerable proportion of associated transcriptional activity was driven by complete denitrifiers, especially under dysoxic conditions. Despite large differences in transcription, the capacity for the final steps of denitrification was largely invariant to aquifer conditions, and most genes and transcripts encoding N2O reductases were the atypical Sec-dependant type, suggesting energy-efficiency prioritization. Results provide insights into the capacity for cooperative relationships in groundwater communities, and the richness and complexity of metabolic mechanisms leading to the loss of fixed nitrogen.

Abstract:Nitrogen source identification is critical to effectively govern nitrogen contamination, as excessive nitrogen content in groundwater has become a crucial global environmental problem. The traditional stable isotope approach (δ15N-NO3-, δ18O-NO3-, δ15N-NH4+) has been widely used to identify nitrogen sources by comparing the isotopic signals between groundwater samples and the possible nitrogen source. However, nitrogen source apportionment based only on that method would be inaccurate because isotopic signals overlap between the nitrogen sources and nitrogen that has undergone biochemical reactions. To solve the shortcomings of the traditional stable isotope approach, the isotope ratios of N2O, the SP-δ18O model and the traditional stable isotope approach were combined to depict the nitrogen transformation processes and reconstruct the "initial" isotopic compositions of nitrogen sources. A total of 58 groundwater samples were collected in the Dongguan Basin and were analysed during June-July 2018. NO3--N was the dominant species of dissolved inorganic nitrogen in most of the groundwater samples. In the groundwater flow direction, NO3- contents decreased, while the δ15N-NO3- and δ18O-NO3- values increased. The positive relationship between δ15N-NO3- and δ18O-NO3- and the calculation results of the SP-δ18O model indicated that both denitrification and nitrification processes occurred in the groundwater, and denitrification acted as the dominant nitrogen transformation process. The initial isotope ratio of nitrogen obtained from the combined use of the calculation results of the SP-δ18O model and δ15N-NO3- (or δ15N-NH4+) value suggested that manure and septic wastes were the primary sources of NO3- and NH4+ in the groundwater. The redox-sensitive components and the calculation results of the SP-δ18O model depicted the reductive level of NO3- in the groundwater flow direction: NO3- reduction did not occur in the upstream area, NO3- reduction was strong in the intermediate transition zone, and NO3- contents were completely consumed and CH4 occurred in the downstream region. This study demonstrated that the combination of the traditional stable isotope approach and the isotope ratios of N2O are useful tools for revealing the source and transformation of nitrogen in groundwater. The method used in this study provides new insight for nitrogen source identification.

Abstract:Anaerobic ammonium oxidation (anammox) process plays a significant role in the marine nitrogen cycle. However, the quantitative importance of this process in nitrogen removal in wetland systems, particularly in natural freshwater wetlands, is still not determined. In the present study, we provided the evidence of the distribution and activity of anammox bacteria in a natural freshwater wetland, located in southeastern China, by using 15 N stable isotope measurements, quantitative PCR assays and 16S rRNA gene clone library analysis. The potential anammox rates measured in this wetland system ranged between 2.5 and 25.5 nmol N 2 g −1 soil day −1 , and up to 20 % soil dinitrogen gas production could be attributed to the anammox process. Phylogenetic analysis of 16S rRNA genes showed that anammox bacteria related to Candidatus Brocadia, Candidatus Kuenenia, Candidatus Anammoxoglobus and two novel anammox clusters coexisted in the collected soil cores, with Candidatus Brocadia and Candidatus Kuenenia being the dominant anammox genera. Quantitative PCR of hydrazine synthase genes showed that the abundance of anammox bacteria varied from 2.3 × 10 5 to 2.2 × 10 6 copies g −1 soil in the examined soil cores. Correlation analyses suggested that the soil ammonium concentration had significant influence on the activity of anammox bacteria. On the basis of 15 N tracing technology, it is estimated that a total loss of 31.1 g N m −2 per year could be linked the anammox process in the examined wetland.
Hydrochemical and microbial community characteristics and the sources of inorganic nitrogen in groundwater from different aquifers in Zhanjiang, Guangdong Province, China

Abstract:Groundwater from different aquifers in the Zhanjiang area suffers from different degrees of nitrogen pollution, which poses a serious threat to the health of urban and rural residents as well as the surrounding aquatic ecological environment. However, neither the water chemistry and microbial community characteristics in different aquifer media nor the sources of inorganic nitrogen pollution have been extensively studied. This study integrated water quality parameters, dual isotopes (δ15N-NO3- and δ18O-NO3-), and 16S rRNA data to clarify the hydrochemical and microbial characteristics of loose rock pore water (LRPW), layered bedrock fissure water (LBFW), and volcanic rock pore fissure water (VRPFW) in the Zhanjiang area and to determine inorganic nitrogen pollution and sources. The results show that the hydrochemistry of groundwater in different aquifers is complex and diverse, which is mainly affected by rock weathering and atmospheric precipitation, and the cation exchange is strong. High NO3- concentration reduces the richness of the microbial community (VRPFW). There are a large number of bacteria related to nitrogen (N) cycle in groundwater and nitrification dominated the N transformation. A quarter of the samples exceeded the relevant inorganic nitrogen index limits specified in the drinking water standard for China. The NO3- content is highest in VRPFW and the NH4+ content is highest in shallow loose rock pore water (SLRPW). In general, NO3-/Cl-, dual isotope (δ15N-NO3- and δ18O-NO3-) data and MixSIAR quantitative results indicate manure and sewage (M&S) and soil organic nitrogen (SON) are the main sources of NO3-. In LRPW, as the depth increases, the contribution rate of M&S gradually decreases, and the contribution rate of SON gradually increases. The results of uncertainty analysis show that the UI90 values of SON and M&S are higher. This study provides a scientific basis for local relevant departments to address inorganic nitrogen pollution in groundwater.

Abstract:Biological nitrogen fixation (BNF) is a crucial process that provides bioavailable nitrogen and supports primary production in freshwater lake ecosystems. However, the characteristics of diazotrophic community and nitrogenase activity in freshwater lake sediments remain poorly understood. Here, we investigated the diazotrophic communities and nitrogenase activities in the sediments of three large river-connected freshwater lakes in eastern China using 15N-isotope tracing and nifH sequencing. The sediments in these lakes contained diverse nitrogenase genes that were phylogenetically grouped into Clusters I and III. The diazotrophic communities in the sediments were dominated by stochastic processes in Hongze Lake and Taihu Lake, which had heterogeneous habitats and shallower water depths, while in Poyang Lake, which had deeper water and a shorter hydraulic retention time, the assembly of the diazotrophic community in the sediments was dominated by homogeneous selection processes. Temperature and water depth were also found the key environmental factors affecting the sediment diazotrophic communities. Sediment nitrogenase activities varied in the three lakes and within distinct regions of an individual lake, ranging from 0 to 14.58 nmol/(kg·hr). Nitrogenase activity was significantly correlated with ferric iron, total phosphorus, and organic matter contents. Our results suggested that freshwater lake sediment contain high diversity of nitrogen-fixing microorganisms with potential metabolic diversity, and the community assembly patterns and nitrogenase activities varied with the lake habitat.

Abstract:Nitrate and ammonia overload in groundwater can lead to eutrophication of surface water in areas where surface water is recharged by groundwater. However, this process remained elusive due to the complicated groundwater N cycling, which is governed by the co-evolution of hydrogeochemical conditions and N-cycling microbial communities. Herein, this process was studied along a generalized groundwater flowpath in Ganjing Delta, Poyang Lake area, China. From groundwater recharge to the discharge area near the lake, oxidation-reduction potential (ORP), NO 3 –N, and NO 2 –N decreased progressively, while NH 3 –N, total organic carbon (TOC), Fe 2+ , sulfide, and TOC/NO 3 − ratio accumulated in the lakeside samples. The anthropogenic influences such as sewage and agricultural activities drove the nitrate distribution, as observed by Cl − vs. NO 3 − /Cl − ratio and isotopic composition of nitrate. The hydrogeochemical evolution was intimately coupled with the changes in microbial communities. Variations in microbial community structures was significantly explained by Fe 2+ , NH 3 –N, and sulfide, while TOC/NO 3 − controlled the distribution of predicted N cycling gene. The absence of NH 3 –N in groundwater upstream was accompanied by the enrichment in Acinetobacter capable of nitrification and aerobic denitrification. These two processes were also supported by Ca 2+ + Mg 2+ vs. HCO 3 − ratio and isotopic composition of NO 3 − . The DNRA process downstream was revealed by both the presence of DNRA-capable microbes such as Arthrobacter and the isotopic composition of NH 4 + in environments with high concentrations of NH 3 –N, TOC/NO 3 − , Fe 2+ , and sulfide. This coupled evolution of N cycling and microbial community sheds new light on the N biogeochemical cycle in areas where surface water is recharged by groundwater.

Abstract:The exogenous input of nitrate in groundwater may change microbial community, and in turn, the change of microbial community affects the biogeochemical transformation of nitrate. However, how this mutual interaction varies along the groundwater flowpath remains poorly understood. Herein, we investigated the co-variation of hydrochemical characteristics and microbial community compositions in nitrate-contaminated groundwater along a generalized flowpath in Linzhou-Anyang area, southern North China Plain. The groundwater-rock interactions and human activities collectively controlled the transition patterns of hydrogeochemical conditions. Along the groundwater flowpath, the nitrate, ammonia, major ions, total organic carbon (TOC), C/N, and relative abundance of Proteobacteria and Epsilonbacteraeota subsequently increased, while nitrite, oxidation reduction potential (ORP), and overall microbial diversity decreased. Both microbial community composition and microbial co-occurrence network were primarily governed by the levels of TOC, nitrate, and nitrite, a sum of which explained 48.4% variance of community compositions, greater than the effect of water-rock interaction (basically ion contents, 12.9%) and ORP (4%), suggesting the dominant role of exogenous nitrate and organic carbon inputs in shaping microbial community. Accordingly, the variations of keystone bacterial lineages, e.g., Vogesella sp., Acinetobacter sp., Rhodoferax sp., Pseudomonas sp., and Flavobacterium sp., and archaeal lineage Candidatus Nitrosoarchaeum along the groundwater flowpath were putatively involved in the dissimilatory nitrate reduction, indicating the potential role of heterotrophic nitrate reducers in nitrate transformation. Our findings suggest that microbial community distribution along groundwater flowpath is intimately related to exogenous nitrate and organic carbon inputs, which likely outweigh the effect of groundwater circulation or water-rock interaction. This study could help us to better understand the inter-relationship between hydrochemical environments and microbial communities in nitrate-polluted groundwater.

Abstract:Nitrogen pollution exceeding the standard because of intensive farming and cropping systems has been a widespread problem in Northeast China. This study investigated the characteristics of functional microorganisms in groundwater in the Bang River farming area. Metagenomic sequencing was used to analyze microbial community structures and Canoco was applied to reveal the response relationship between the microbial community and water environmental factors and to identify changes in the microbial population in response to the addition of electronic donors NH4+-N, NO2−-N, and NO3−-N. The results showed that the dominant microorganisms in groundwater belong to the genera Exiguobacterium, Citrobacter, Acinetobacter, and Pseudomonas, which accounted for more than 40% of the total microbes in the study area. When combined with the results of a water chemical factor test, the dominant bacteria were found to be correlated with Fe2+, Mn2+, NH4+, NO3−, NO2−, HCO3−, DOC, and pH in the water. However, the microbial population changed after the addition of the electron donor, with the genera Pseudomonas, Serratia, Enterobacter, Azomonas, and Ewingella accounting for 97.06% of the total sequences. Indigenous nitrogen-degrading bacteria suitable for low temperature, low oxygen, and oligotrophic groundwater were screened out. The total removal efficiency of NH4+-N, NO2−-N, and NO3−-N in 120 h was 90.83%, 75.04%, and 73.35%, respectively. According to the experimental results, the degradation reaction kinetics followed a pseudo-second-order equation. The results presented herein provide an important scientific basis for the microbial remediation of groundwater contaminated by ammonia.

Meta-analysis of diazotrophic signatures across terrestrial ecosystems at the continental scale

Abstract:Biological nitrogen fixation performed by diazotrophs forms a cornerstone of Earth's terrestrial ecosystem productivity. However, the composition, diversity and distribution of soil diazotrophs are poorly understood across different soil ecosystems. Furthermore, the biological potential of the key diazotroph species in relation to key environmental parameters is unknown. To address this, we used meta-analysis approach to merge together 39 independent diazotroph amplicon sequencing (nifH gene) datasets consisting of 1988 independent soil samples. We then employed multiple statistical analyses and machine-learning approaches to compare diazotroph community differences and indicator species between terrestrial ecosystems on a global scale. The distribution, composition and structure of diazotroph communities varied across seven different terrestrial ecosystems, with community composition exhibiting an especially clear effect. The Cyanobacteria were the most abundant taxa in crust ecosystems (accounting for ~45% of diazotrophs), while other terrestrial ecosystems were dominated by Proteobacteria, including Alpha-, Beta- and Gamma-Proteobacteria (accounting for ~70% of diazotrophs). Farmland ecosystems harboured the highest and crust ecosystems the lowest alpha and phylogenetic diversities. Azospirillum zeae, Skermanella aerolata and four Bradyrhizobium species were identified as key indicator species of potential diazotroph activity. Overall, diazotroph abundances and distribution were affected by multiple environmental parameters, including soil pH, nitrogen, organic carbon, C:N ratio and annual mean precipitation and temperature. Together, our findings suggest that based on the relative abundance and diversity of nifH marker gene, diazotrophs have adapted to a range of environmental niches globally.

Abstract:PurposeBiological nitrogen fixation (BNF) plays an important role in nitrogen cycling by transferring atmospheric dinitrogen to the soil. BNF is performed by symbiotic and asymbiotic nitrogen-fixing microorganisms. However, the abundance, activity, and community structure of diazotrophs under different nitrogen fertilizer application rates and how root exudates influence diazotrophs remain unclear.Materials and methods15N-N2 and 13C-CO2 labeling, DNA-based stable isotope probing (SIP), and molecular biological techniques were used in this study. The abundance, activity, and structure of symbiotic and asymbiotic diazotrophs under different nitrogen fertilizer applications in paddy soil were investigated.Results and discussionWe found that the nitrogen fixation capacity in milk vetch (Astragalus sinicus L.) and nifH gene abundance in the root nodules were significantly higher in the low-nitrogen treatment than in the control (zero) and high-nitrogen treatments. Nitrogen-fixing bacteria were abundant in the soils with a high biodiversity. Soil nifH gene sequences were dominated by α-, β-, and δ-proteobacteria, as well as by Cyanobacteria. The relative abundance of α-proteobacteria was lower, and the relative abundance of Cyanobacteria was higher under high nitrogen. Incubation of soil with 13CO2 and subsequent DNA-SIP analysis demonstrated that OTU65 from α-proteobacteria was relatively more abundant in heavy fractions of the 13C-labeled soils than that in the unlabeled soils, indicating that α-proteobacteria may prefer rhizodeposition carbon to other organic carbon. However, OTU24 and OTU73 from δ-proteobacteria had relatively high abundances in light fractions both in the 13C-labeled and unlabeled samples, indicating that δ-proteobacteria may prefer other soil organic carbon to rhizodeposition carbon.ConclusionsThe results suggested that soil N availability and rhizodeposition strongly modified the microbial communities of nitrogen-fixing bacteria. Moderate nitrogen application increased the symbiotic biological N fixing activity in the Astragalus sinicus L. rhizosphere. The BNF activity in the legume-rhizobia system is regulated by the exchange of organic C and N nutrient between the host plant and N-fixing bacteria.
