Abstract:Sleep plays a crucial role in memory stabilization. In the dorsal hippocampus (dHPC), fast oscillations (sharp-wave ripples) during non-REM sleep consolidate spatial memories through place cell reactivation. The hippocampal formation is heterogeneous; the dHPC processes mostly contextual information, while the ventral part (vHPC) encodes valence and anxiety. We propose that dorsal-ventral communication during sleep is involved in the consolidation of emotional memories, potentially integrating or segregating contextual and valence information. To test this, we recorded the electrophysiological activity in the dHPC and vHPC in rats during reward and aversion-motivated exploration of a linear track and subsequent sleep. We detected dorsal and ventral SWRs during NREM sleep as well as neuronal assemblies during each exploration type and identified dorsal, ventral, and joint (dorso-ventral) assemblies. We found that dorso-ventral hippocampal coordination involves coordinated ripples and bursts of ripples. Aversive joint assemblies exhibited higher reactivation during non-REM sleep than the joint assemblies from the reward condition. Interestingly, joint aversive assemblies reactivated selectively during dorso-ventral coordinated ripples. Furthermore, their reactivation strength positively correlated with their strength of activation during the task. These results suggest the existence of dorsal-ventral hippocampal functional communication capable of sustaining the selective reactivation of experiences based on their emotional valence.

Abstract:Research has shown that sleep is beneficial for the long-term retention of memories. According to theories of memory consolidation, memories are gradually reorganized, becoming supported by widespread, distributed cortical networks, particularly during postencoding periods of sleep. However, the effects of sleep on the organization of memories in the hippocampus itself remains less clear. In a 3-d study, participants encoded separate lists of word–image pairs differing in their opportunity for sleep-dependent consolidation. Pairs were initially studied either before or after an overnight sleep period, and were then restudied in a functional magnetic resonance imaging (fMRI) scan session. We used multivariate pattern similarity analyses to examine fine-grained effects of consolidation on memory representations in the hippocampus. We provide evidence for a dissociation along the long axis of the hippocampus that emerges with consolidation, such that representational patterns for object–word memories initially formed prior to sleep become differentiated in anterior hippocampus and more similar, or overlapping, in posterior hippocampus. Differentiation in anterior hippocampal representations correlated with subsequent behavioral performance. Furthermore, representational overlap in posterior hippocampus correlated with the duration of intervening slow wave sleep. Together, these results demonstrate that sleep-dependent consolidation promotes the reorganization of memory traces along the long axis of the hippocampus.

Abstract:There is a long-standing division in memory research between hippocampus-dependent memory and non-hippocampus-dependent memory, as only the latter can be acquired and retrieved in the absence of normal hippocampal function1,2. Consolidation of hippocampus-dependent memory, in particular, is strongly supported by sleep3–5. Here we show that the formation of long-term representations in a rat model of non-hippocampus-dependent memory depends not only on sleep but also on activation of a hippocampus-dependent mechanism during sleep. Rats encoded non-hippocampus-dependent (novel-object recognition6–8) and hippocampus-dependent (object–place recognition) memories before a two-hour period of sleep or wakefulness. Memory was tested either immediately thereafter or remotely (after one or three weeks). Whereas object–place recognition memory was stronger for rats that had slept after encoding (rather than being awake) at both immediate and remote testing, novel-object recognition memory profited from sleep only three weeks after encoding, at which point it was preserved in rats that had slept after encoding but not in those that had been awake. Notably, inactivation of the hippocampus during post-encoding sleep by intrahippocampal injection of muscimol abolished the sleep-induced enhancement of remote novel-object recognition memory. By contrast, muscimol injection before remote retrieval or memory encoding had no effect on test performance, confirming that the encoding and retrieval of novel-object recognition memory are hippocampus-independent. Remote novel-object recognition memory was associated with spindle activity during post-encoding slow-wave sleep, consistent with the view that neuronal memory replay during slow-wave sleep contributes to long-term memory formation. Our results indicate that the hippocampus has an important role in long-term consolidation during sleep even for memories that have previously been considered hippocampus-independent.

Abstract:Long-term memories are formed by creating stable memory representations via memory consolidation, which mainly occurs during sleep following the encoding of labile memories in the hippocampus during waking. The entorhinal cortex (EC) has intricate connections with the hippocampus, but its role in memory consolidation is largely unknown. Using cell-type- and input-specific in vivo neural activity recordings, here we show that the temporoammonic pathway neurons in the EC, which directly innervate the output area of the hippocampus, exhibit potent oscillatory activities during anesthesia and sleep. Using in vivo individual and populational neuronal activity recordings, we demonstrate that a subpopulation of the temporoammonic pathway neurons, which we termed sleep cells, generate delta oscillations via hyperpolarization-activated cyclic-nucleotide-gated channels during sleep. The blockade of these oscillations significantly impaired the consolidation of hippocampus-dependent memory. Together, our findings uncover a key driver of delta oscillations and memory consolidation that are found in the EC.

Abstract:Neuronal ensembles are crucial for episodic memory and spatial mapping. Sleep, particularly non-REM (NREM), is vital for memory consolidation, as it triggers plasticity mechanisms through brain oscillations that reactivate neuronal ensembles. Here, we assessed their role in consolidating hippocampal spatial representations during sleep. We recorded hippocampus activity in rats performing a spatial object-place recognition (OPR) memory task, during encoding and retrieval periods, separated by intervening sleep. Successful OPR retrieval correlated with NREM duration, during which cortical oscillations decreased in power and density as well as neuronal spiking, suggesting global downregulation of network excitability. However, neurons encoding specific spatial locations (i.e., place cells) or objects during OPR showed stronger synchrony with brain oscillations compared to non-encoding neurons, and the stability of spatial representations decreased proportionally with NREM duration. Our findings suggest that NREM sleep may promote flexible remapping in hippocampal ensembles, potentially aiding memory consolidation and adaptation to novel spatial contexts.

Abstract:Sleep is crucial for healthy cognition, including memory. The two main phases of sleep, REM (rapid eye movement) and non-REM sleep, are associated with characteristic electrophysiological patterns that are recorded using surface and intracranial electrodes. These patterns include sharp-wave ripples, cortical slow oscillations, delta waves, and spindles during non-REM sleep and theta oscillations during REM sleep. They reflect the precisely timed activity of underlying neural circuits. Here, we review how these electrical signatures have been guiding our understanding of the circuits and processes sustaining memory consolidation during sleep, focusing on hippocampal theta oscillations and sharp-wave ripples and how they coordinate with cortical patterns. Finally, we highlight how these brain patterns could also sustain sleep-dependent homeostatic processes and evoke several potential future directions for research on the memory function of sleep.

Abstract:Abstract Introduction Memory consolidation is facilitated by trace reactivation during non-rapid eye movement slow wave sleep (SWS), involving reinstatement of a hippocampal index driving integration of activity patterns stored in cortical modules. Slow oscillation (SO) and sleep spindle (SP) coupling reflects this process. Here, we used a novel graph theory approach to examine whether network modularity (Q) and the relative import of the hippocampus and amygdala in network function, measured by eigenvector centrality (EC) and betweenness centrality (BC), are associated with SWS expression, SO-SP coupling, and emotional memory consolidation. Methods Thirty-six cognitively intact older adults (μ=72.9±5.6 years, 23 Females) completed structural and resting-state functional magnetic resonance imaging (MRI) at 3T, an overnight polysomnography with electroencephalography, and, prior to and following sleep, performed the Emotional Mnemonic Discrimination Task, with the Lure Discrimination Index quantifying memory. MRI data were preprocessed with the CONN toolbox. Network nodes were defined using the Brainnetome atlas. Graph metrics were computed using the Brain Connectivity Toolbox. In a subset, p-welch derived SO power (N=18) and mean vector length computed phase-amplitude SO-SP coupling (N=16) were derived. Results Q was associated with diminished emotional memory retention (B=-1.394, p=0.042) and greater SWS percentage (B=48.476, p=0.023), which was also associated with emotional memory retention (B=0.013, p=0.020). An inconsistent mediation effect of SWS in the Q-emotional memory retention relationship was found (95% CIs: 0.015, 0.345). Hippocampal EC (B=36.845, p=0.008) and amygdala EC (B=47.277, p=0.022) were associated with emotional memory retention. Hippocampal BC was associated with immediate test performance (B=19.859, p=0.027), whereas amygdala BC was associated with delayed test (B=1.823, SE=0.745, p=0.020) and emotional memory retention (B=1.699, p=0.047). Hippocampal EC (B=0.475, p=0.047) and BC (B=0.426, p=0.086) were associated with frontal SO expression, which was associated with delayed test performance (B=0.492, p=0.038). Hippocampal EC (B=0.649, p=0.006) and amygdala BC (B=0.545, p=0.029) were associated with frontal SO-SP coupling. Conclusion These findings suggest that greater network integration supports SWS-dependent emotional memory consolidation during sleep. A more topologically influential hippocampus aids in memory acquisition and retention, whereas a more influential amygdala supports emotional memory consolidation. Abstract:Abstract Introduction Rapid eye movement (REM) sleep has been implicated in the consolidation of emotional memories. Our recent work found a candidate system for REM-related memory consolidation. We showed that during REM sleep, the frontal cortices are dominated by theta (4–8 Hz) oscillations and bursts of beta (15–35 Hz) activity. Studies suggest that rhythmic interactions between the frontal cortices and limbic structures, in particular the amygdala, play a critical role in the consolidation of emotional memories. However, the mechanisms responsible for memory consolidation during these rhythmic interactions during REM sleep remain unknown. Methods We used biophysically based neural models to build a large-scale network model of the prefrontal cortex (PFC) and amygdala (AMY) and incorporated synaptic plasticity mechanisms, such as spike-timing dependent plasticity (STDP), into the connections between these two regions. Norepinephrine (NE) and serotonin (SE) levels were manipulated to mimic the different physiological conditions during the awake state and REM sleep. Results We were able to reproduce the oscillatory dynamics observed in experimental studies and identify cell-type specific synaptic changes caused by STDP. During the awake state, PFC connections to all cell types of the AMY become strengthened when PFC neurons provide theta frequency inputs, with the connections strengthening to a greater extent when inputs are in burst mode rather than single spike mode. When the PFC provides beta inputs, we see the exact opposite relationship: synaptic strengths become weaker when inputs are in burst mode rather than single spike mode. During REM sleep conditions, the connections to all principal cell types of the AMY become strengthened, with synaptic connections to some subtypes of pyramidal cells becoming stronger than others. Surprisingly, however, the synaptic connections to the interneurons become weaker in response to theta frequency inputs. Conclusion Using our large-scale network model, we show how the levels of the neurotransmitters NE and SE during the awake state and REM sleep affect oscillatory dynamics and in turn influence the strengthening or weakening of connections related to emotional memories. Abstract:Sleep plays an important role in memory consolidation through the facilitation of neuronal plasticity; however, how sleep accomplishes this remains to be completely understood. It has previously been demonstrated that neural oscillations are an intrinsic mechanism by which the brain precisely controls neural ensembles. Inter-regional synchronization of these oscillations is also known to facilitate long-range communication and long-term potentiation (LTP). In the present study, we investigated how the characteristic rhythms found in local field potentials (LFPs) during non-REM and REM sleep play a role in emotional memory consolidation. Chronically implanted bipolar electrodes in the lateral amygdala (LA), dorsal and ventral hippocampus (DH, VH), and the infra-limbic (IL), and pre-limbic (PL) prefrontal cortex were used to record LFPs across sleep-wake activity following each day of a Pavlovian cued fear conditioning paradigm. This resulted in three principle findings: (1) theta rhythms during REM sleep are highly synchronized between regions; (2) the extent of inter-regional synchronization during REM and non-REM sleep is altered by FC and EX; (3) the mean phase difference of synchronization between the LA and VH during REM sleep predicts changes in freezing after cued fear extinction. These results both oppose a currently proposed model of sleep-dependent memory consolidation and provide a novel finding which suggests that the role of REM sleep theta rhythms in memory consolidation may rely more on the relative phase-shift between neural oscillations, rather than the extent of phase synchronization.
[Shock wave therapy for recalcitrant plantar fasciitis with heel spur: a prospective randomized placebo-controlled double-blind study].

Abstract:AIM Efficacy of low-energy shock wave therapy for recalcitrant plantar fasciitis. METHOD 32 patients were randomly assigned into real and placebo ESWT groups, treatment comprised 1000 impulses of 0,08 mJ/mm 2 at 14 kV (OssaTron OSA 120, HMT AG, Switzerland) in 12 cases repeated after six weeks or placebo (energy-absorbing foil). Follow-up evaluation (19, 32 and 48 wks.) included specific questionnaire, clinical-functional examination and measurement of plantar pressure while walking (Emed AT-4. pedograph, Novel GmbH, Munich). Examiner and patients were blinded. RESULTS 88 % of the treatment group were pain free or had good results. None of the placebo group were pain free, 33.3 % had good results (Roles and Maudsley Score). The treatment group showed significantly better outcome for morning and resting pain, pressure stamp-tolerance and walking ability. Pedography did not show a clear correlation between pain relief and load pattern. CONCLUSION The results of this study corroborate the value of ESWT for recalcitrant plantar fascitis. As a non invasive technique with low side effects, it can complement the row of conservative treatments.
Hippocampus-retrosplenial cortex interaction is increased during phasic REM and contributes to memory consolidation

Abstract:Abstract Hippocampal (HPC) theta oscillation during post-training rapid eye movement (REM) sleep supports spatial learning. Theta also modulates neuronal and oscillatory activity in the retrosplenial cortex (RSC) during REM sleep. To investigate the relevance of theta-driven interaction between these two regions to memory consolidation, we computed the Granger causality within theta range on electrophysiological data recorded in freely behaving rats during REM sleep, both before and after contextual fear conditioning. We found a training-induced modulation of causality between HPC and RSC that was correlated with memory retrieval 24 h later. Retrieval was proportional to the change in the relative influence RSC exerted upon HPC theta oscillation. Importantly, causality peaked during theta acceleration, in synchrony with phasic REM sleep. Altogether, these results support a role for phasic REM sleep in hippocampo-cortical memory consolidation and suggest that causality modulation between RSC and HPC during REM sleep plays a functional role in that phenomenon.

Abstract:Abstract How do we build up our knowledge of the world over time? Many theories of memory formation and consolidation have posited that the hippocampus stores new information, then “teaches” this information to neocortex over time, especially during sleep. But it is unclear, mechanistically, how this actually works — how are these systems able to interact during periods with virtually no environmental input to accomplish useful learning and shifts in representation? We provide a framework for thinking about this question, with neural network model simulations serving as demonstrations. The model contains hippocampus and neocortical areas, which replay memories and interact with one another completely autonomously during simulated sleep. Oscillations are leveraged to support error-driven learning that leads to useful changes in memory representation and behavior. The model has a non-Rapid Eye Movement (NREM) sleep stage, where dynamics between hippocampus and neocortex are tightly coupled, with hippocampus helping neocortex to reinstate high-fidelity versions of new attractors, and a REM sleep stage, where neocortex is able to more freely explore existing attractors. We find that alternating between NREM and REM sleep stages, which alternately focuses the model’s replay on recent and remote information, facilitates graceful continual learning. We thus provide an account of how the hippocampus and neocortex can interact without any external input during sleep to drive useful new cortical learning and to protect old knowledge as new information is integrated.
Coordinated Interactions between the Hippocampus and Retrosplenial Cortex in Spatial Memory

Abstract:While a hippocampal-cortical dialogue is generally thought to mediate memory consolidation, which is crucial for engram function, how it works remains largely unknown. Here, we examined the interplay of neural signals from the retrosplenial cortex (RSC), a neocortical region, and from the hippocampus in memory consolidation by simultaneously recording sharp-wave ripples (SWRs) of dorsal hippocampal CA1 and neural signals of RSC in free-moving mice during the delayed spatial alternation task (DSAT) and subsequent sleep. Hippocampal-RSC coordination during SWRs was identified in nonrapid eye movement (NREM) sleep, reflecting neural reactivation of decision-making in the task, as shown by a peak reactivation strength within SWRs. Using modified generalized linear models (GLMs), we traced information flow through the RSC-CA1-RSC circuit around SWRs during sleep following DSAT. Our findings show that after spatial training, RSC excitatory neurons typically increase CA1 activity prior to hippocampal SWRs, potentially initiating hippocampal memory replay, while inhibitory neurons are activated by hippocampal outputs in post-SWRs. We further identified certain excitatory neurons in the RSC that encoded spatial information related to the DSAT. These neurons, classified as splitters and location-related cells, showed varied responses to hippocampal SWRs. Overall, our study highlights the complex dynamics between the RSC and hippocampal CA1 region during SWRs in NREM sleep, underscoring their critical interplay in spatial memory consolidation.
Effects of smokeless tobacco on the periodontal, mucosal and caries status of adolescent males.

Abstract:The use of smokeless tobacco (ST) products, such as snuff and chewing tobacco, was investigated in a school population of 565 males with a mean age of 13.8 years. All students completed a tobacco usage questionnaire and were given intraoral examinations by a team of dentists to determine the possible relationship of ST usage to the presence of gingivitis, gingival recession, mucosal pathology and caries. The overall prevalence of ST usage was 13.3%, which was much higher than the 1.4% prevalence of cigarette smoking. Pairwise chi square analysis revealed that there was no relationship between ST usage and the prevalence of gingivitis, but that the prevalence of gingival recession was significantly elevated in ST users, P less than 0.001. The odds of having gingival recession were 9 times greater in the students using smokeless tobacco as compared to healthy, non-user cohorts. ST usage did not affect the prevalence of gingival recession in students without gingivitis. The frequency of occurrence of soft tissue pathology was significantly elevated, about 6-fold in students who used smokeless tobacco, as compared to non-users, P less than 0.01. This was principally due to the increased prevalence of white mucosal lesions in smokeless tobacco users. However, there was no attributable risk for mucosal pathology in ST users who were free of gingivitis. Similarly, the use of smokeless tobacco was associated with a 1.6-fold elevation in mean DMF in students with gingivitis, P less than 0.001, but this increase in caries experience was not seen in ST users who were free of gingivitis. In summary, in students with clean mouths that were free of gingivitis, the use of smokeless tobacco was not associated with a change in the prevalence of gingival recession, mucosal pathology, or in the mean DMF score. In contrast, smokeless tobacco usage was a significant risk factor in individuals with co-existing gingivitis, associated with a marked increase in the prevalence of gingival recession, mucosal pathology and caries experience.

Repeated neuronal replay of representations originating from the hippocampus during slow-wave sleep leads to a gradual transformation and integration of representations in neocortical networks. We highlight three features of this process: (i) hippocampal replay that, by capturing episodic memory aspects, drives consolidation of both hippocampus-dependent and non-hippocampus-dependent memory; (ii) brain oscillations hallmarking slow-wave and rapid-eye movement sleep that provide mechanisms for regulating both information flow across distant brain networks and local synaptic plasticity; and (iii) qualitative transformations of memories during systems consolidation resulting in abstracted, gist-like representations.

Abstract:Learning processes continue well beyond the time when the information to be learned is presented. Some offline gains are rapid, whereas others require longer periods of consolidation, including sleep. These sleep-associated gains are heavily dependent on the hippocampus. Although the hippocampus has traditionally been assumed to support only the formation of declarative memories (e.g., facts, events), recent research has shown that it is also involved in the creation of procedural memories, such as motor skill acquisition (Albouy et al., 2013). For instance, in the motor sequence task (MST), which requires participants to learn finger-tapping sequences by typing them, performance normally improves over sleep (Albouy et al., 2013; Schapiro et al., 2019), but such consolidation gains were absent in patients with a hippocampal lesion, suggesting that the hippocampus is necessary for overnight gains in the MST. However, the patients were able to learn the MST and exhibited gains in performance during training, suggesting that the hippocampus was not necessary for gains during training itself (Schapiro et al., 2019). A recent large-scale study showed that performance in the MST improved after very short breaks lasting just a few seconds in between training trials (Bönstrup et al., 2020). This suggested that some form of consolidation occurs during these during short periods of wakefulness. Importantly, the extent of this improvement was tightly linked with hippocampal activity during quiet rest: In a magnetoencephalography (MEG) study by Buch et al. (2021), neural activity elicited during training was replayed in the hippocampus during rest periods and the extent of replay correlated with performance improvements. Similarly, in a functional magnetic resonance imaging (fMRI) study by Jacobacci et al. (2020), the amount of hippocampal activity during wakeful rest correlated with performance improvements. Synaptic Homeostasis and Restructuring across the Sleep-Wake Cycle

Abstract:Sleep is critical for hippocampus-dependent memory consolidation. However, the underlying mechanisms of synaptic plasticity are poorly understood. The central controversy is on whether long-term potentiation (LTP) takes a role during sleep and which would be its specific effect on memory. To address this question, we used immunohistochemistry to measure phosphorylation of Ca2+/calmodulin-dependent protein kinase II (pCaMKIIα) in the rat hippocampus immediately after specific sleep-wake states were interrupted. Control animals not exposed to novel objects during waking (WK) showed stable pCaMKIIα levels across the sleep-wake cycle, but animals exposed to novel objects showed a decrease during subsequent slow-wave sleep (SWS) followed by a rebound during rapid-eye-movement sleep (REM). The levels of pCaMKIIα during REM were proportional to cortical spindles near SWS/REM transitions. Based on these results, we modeled sleep-dependent LTP on a network of fully connected excitatory neurons fed with spikes recorded from the rat hippocampus across WK, SWS and REM. Sleep without LTP orderly rescaled synaptic weights to a narrow range of intermediate values. In contrast, LTP triggered near the SWS/REM transition led to marked swaps in synaptic weight ranking. To better understand the interaction between rescaling and restructuring during sleep, we implemented synaptic homeostasis and embossing in a detailed hippocampal-cortical model with both excitatory and inhibitory neurons. Synaptic homeostasis was implemented by weakening potentiation and strengthening depression, while synaptic embossing was simulated by evoking LTP on selected synapses. We observed that synaptic homeostasis facilitates controlled synaptic restructuring. The results imply a mechanism for a cognitive synergy between SWS and REM, and suggest that LTP at the SWS/REM transition critically influences the effect of sleep: Its lack determines synaptic homeostasis, its presence causes synaptic restructuring.

Abstract:Post-learning sleep is essential for hippocampal memory processing, including contextual fear memory consolidation. We labeled context-encoding engram neurons in the hippocampal dentate gyrus (DG) and assessed reactivation of these neurons after fear learning. Post-learning sleep deprivation (SD) selectively disrupted reactivation of inferior blade DG engram neurons, linked to SD-induced suppression of neuronal activity in the inferior, but not superior DG blade. Subregion-specific spatial profiling of transcripts revealed that transcriptomic responses to SD differed greatly between hippocampal CA1, CA3, and DG inferior blade, superior blade, and hilus. Activity-driven transcripts, and those associated with cytoskeletal remodeling, were selectively suppressed in the inferior blade. Critically, learning-driven transcriptomic changes differed dramatically between the DG blades and were absent from all other regions. Together, these data suggest that the DG is critical for sleep-dependent memory consolidation, and that the effects of sleep loss on the hippocampus are highly subregion-specific.
