Abstract:Understanding cell–nanoparticle interactions is critical to developing effective nanosized drug delivery systems. Nanoparticles have already advanced the treatment of several challenging conditions including cancer and human immunodeficiency virus (HIV), yet still hold the potential to improve drug delivery to elusive target sites. Even though most nanoparticles will encounter blood at a certain stage of their transport through the body, the interactions between nanoparticles and blood cells is still poorly understood and the importance of evaluating nanoparticle hemocompatibility is vastly understated. In contrast to most review articles that look at the interference of nanoparticles with the intricate coagulation cascade, this review will explore nanoparticle hemocompatibility from a cellular angle. The most important functions of the three cellular components of blood, namely erythrocytes, platelets and leukocytes, in hemostasis are highlighted. The potential deleterious effects that nanoparticles can have on these cells are discussed and insight is provided into some of the complex mechanisms involved in nanoparticle–blood cell interactions. Throughout the review, emphasis is placed on the importance of undertaking thorough, all-inclusive hemocompatibility studies on newly engineered nanoparticles to facilitate their translation into clinical application.

Abstract:The ability to manipulate magnetic nanoparticles with external magnetic fields and their compatibility with biological systems make them versatile tools in the field of nanomedicine. Recently, the integration of various nanotechnologies with biomedical science, pharmacology, and clinical practice has led to the emergence of the discipline of nanomedicine. Owing to the special qualities of nanoparticles and related nanostructures, their uses in controlled drug and gene delivery, imaging, medical diagnostics, monitoring therapeutic outcomes, and supporting medical interventions offer a fresh approach to difficult problems in difficult areas like the treatment of cancer or crippling neurological diseases. The potential for multi-functionality and advanced targeting tactics in nanoparticle products exists. It may maximize the effectiveness of current anticancer drugs by enhancing the pharmacodynamic and pharmacokinetic characteristics of conventional therapies. These nanometer-sized substances’ distinctive electrical, magnetic, and optical characteristics have opened up a wide range of biological uses. As they may be used in healthcare situations due to their bioactivity, iron-oxide-based magnetic nanoparticles, in particular, have been shown to be incredibly useful deep-tissue scanning tools. In addition to having a broader operating temperature range, smaller size, reduced toxicity, easier processing, and less cost of production, newer nanoparticles (MNPs) also offer other benefits. MNPs offer a lot of promise for use in clinical settings because of a variety of exceptional and distinctive chemical and biological features. Modern targeting techniques and nanoparticles studied in clinical trials are included in this review. It highlights the difficulties in applying nanomedicine items and transferring them from the laboratory to the clinical environment. It also addresses topics of nanoparticle design that might create new clinical applications for nanomedicine items. Magnetic nanoparticles used in nanomedicine offer several novel and promising features that make them valuable tools for various applications. When utilized in nanomedicine, magnetic nanoparticles have a number of exciting new properties that make them useful instruments for a range of uses. Drug delivery, hyperthermia therapy, magnetic resonance imaging contrast agents, diagnostic imaging and monitoring, theranostic applications, biocompatibility and biodegradability, remote control and manipulation, and responsive nanoparticles are the main factors that add to their novelty. In general, the amalgamation of nanoscale characteristics and magnetic properties presents a multitude of opportunities for inventive medical applications, offering focused, effective, and least intrusive approaches to diagnosis and treatment. The sector is still investigating novel ways to increase the safety and efficacy of magnetic nanoparticles in nanomedicine. The purpose of this article is to provide basic details about magnetic nanoparticles and the characteristics of these particles in biomedical applications. The features of these nanoparticles in medication delivery and their numerous uses have received extra focus in the study. It seeks to summarize current advancements in MNPs for medical applications and examine the possibilities of MNPs in tumor therapeutic applications, in addition to future study opportunities.

Abstract:Magnetic nanoparticles (MNPs) are gaining much attention due to their applications in specialized fields, including diagnosis, bioimaging, drug delivery, and cell therapy. In recent years, the field of stem cell and regenerative medicine comprising tissue engineering and drug delivery has been powered by nanoparticles. Particularly, mesenchymal stem cells (MSCs) are characterized with adequate immunomodulatory potential coupled with an excellent regenerative ability that contributes to improved therapy of various diseases. For clinical applications of MSCs, tracking and labeling are crucial to determine cell homing and distribution. Therefore, due to the high potential of MNPs in stem cell therapy, this review comprehensively uncovers the advancements in the respective field. Also, it discusses the usage of surface-functionalized materials in improving the physicochemical properties of MNPs, making them suitable candidates for several biomedical applications. Moreover, we have discussed that these nanoparticles within applied magnetic field and electromagnetic field have been considered multifunctional agents for labeling, stimulating, tracking, and targeting stem cells in vitro and in vivo. Additionally, the discussion comprises the progress and challenges associated with MNPs used as effective therapeutic, delivery, and diagnostic agents to treat cancer, cardiovascular, neurodegenerative, and bone-related disorders. Finally, the review discussed that the magnetic nanoparticles have also been widely used for targeted cancer cell therapy due to mechanical force for killing cancer cells under a magnetic field. It is concluded that the mechanical force produced by MNPs in a low-frequency vibrating magnetic field is the most promising and safe option for the destruction of tumor cells. In short, this review summarized the role of magnetic nanomaterials for effective, safe, and efficient nanomaterial-cell-based therapies.

Abstract:This review outlines diverse magnetic nanoparticle (MNP) designs, materials, morphologies, and characterization methods. It assesses various MNP synthesis techniques, their pros and cons, and explores inorganic and organic surface coatings. Biomedical MNP applications are covered, spanning hyperthermia, MRI, drug delivery, biosensors, and magnetic particle imaging. Future MNP structure trends and novel biomedical prospects are also discussed. Nowadays, magnetic nanoparticles (MNPs) are applied in numerous fields, especially in biomedical applications. Since biofluidic samples and biological tissues are nonmagnetic, negligible background signals can interfere with the magnetic signals from MNPs in magnetic biosensing and imaging applications. In addition, the MNPs can be remotely controlled by magnetic fields, which make it possible for magnetic separation and targeted drug delivery. Furthermore, due to the unique dynamic magnetizations of MNPs when subjected to alternating magnetic fields, MNPs are also proposed as a key tool in cancer treatment, an example is magnetic hyperthermia therapy. Due to their distinct surface chemistry, good biocompatibility, and inducible magnetic moments, the material and morphological structure design of MNPs has attracted enormous interest from a variety of scientific domains. Herein, a thorough review of the chemical synthesis strategies of MNPs, the methodologies to modify the MNPs surface for better biocompatibility, the physicochemical characterization techniques for MNPs, as well as some representative applications of MNPs in disease diagnosis and treatment are provided. Further portions of the review go into the diagnostic and therapeutic uses of composite MNPs with core/shell structures as well as a deeper analysis of MNP properties to learn about potential biomedical applications.

Abstract:In this article, the recent applications of different types of magnetic nanoparticles such as α-Fe2O3 (hematite), γ-Fe2O3 (maghemite), Fe3O4 (magnetite), hexagonal (MFe12O19), garnet (M3Fe5O12) and spinel (MFe2O4), where M represents one or more bivalent transition metals (Mn, Fe, Co, Ni, Ba, Sr, Cu, and Zn), and different materials for coating the surface of magnetic nanoparticles like poly lactic acid (PLA), doxorubicin hydrophobic (DOX-HCL), paclitaxel (PTX), EPPT-FITC, oleic acid, tannin, 3-Aminopropyltriethoxysilane (APTES), multi-wall carbon nanotubes (CNTs), polyethylenimine (PEI) and polyarabic acid in drug delivery, biomedicine and treatment of cancer, specially chemotherapy, are reviewed. MNPs possess large surface area to volume ratios because of their nano-size, low surface charge at physiological pH and they aggregate easily in solution due to their essential magnetic nature. These materials are widely used in biology and medicine in many cases. One targeted delivery technique that has gained prominence in recent years is the use of magnetic nanoparticles. In these systems, therapeutic compounds are attached to biocompatible magnetic nanoparticles and magnetic fields generated outside the body are focused on specific targets in vivo. The fields capture the particle complex, resulting in enhanced delivery to the target site. Also, the application of brand new supermagnetic nanoparticles, like Ba,SrFe12O19, is considered and studied in this paper.

Eryptosis Indices As A Novel Predictive Parameter for Biocompatibility of Fe3o4 Magnetic Nanoparticles on Erythrocytes

Abstract:Fe3O4 magnetic nanoparticles (Fe3O4-MNPs) have been widely used in clinical diagnosis. Hemocompatibility of the nanoparticles is usually evaluated by hemolysis. However, hemolysis assessment does not measure the dysfunctional erythrocytes with pathological changes on the unbroken cellular membrane. The aim of this study is to evaluate the use of suicidal death of erythrocytes (i.e. eryptosis indices) as a novel predictive and prognostic parameter, and to determine the impact of Fe3O4-MNPs on cellular membrane structure and the rheology properties of blood in circulation. Our results showed that phosphatidylserine externalization assessment was significantly more sensitive than classical hemolysis testing in evaluating hemocompatibility. Although no remarkable changes of histopathology, hematology and serum biochemistry indices were observed in vivo, Fe3O4-MNPs significantly affected hemorheology indices including erythrocyte deformation index, erythrocyte rigidity index, red blood cell aggregation index, and erythrocyte electrophoresis time, which are related to the mechanical properties of the erythrocytes. Oxidative stress induced calcium influx played a critical role in the eryptotic activity of Fe3O4-MNPs. This study demonstrated that Fe3O4-MNPs cause eryptosis and changes in flow properties of blood, suggesting that phosphatidylserine externalization can serve as a predictive parameter for hemocompatibility assay.

Abstract:Metallic nanoparticles (MNPs) have garnered significant attention due to their ability to improve the therapeutic index of medications by reducing multidrug resistance and effectively delivering therapeutic agents through active targeting. In addition to drug delivery, MNPs have several medical applications, including in vitro and in vivo diagnostics, and they improve the biocompatibility of materials and nutraceuticals. MNPs have several advantages in drug delivery systems and genetic manipulation, such as improved stability and half-life in circulation, passive or active targeting into the desired target selective tissue, and gene manipulation by delivering genetic materials. The main goal of this review is to provide current information on the present issues and prospects of MNPs in drug and gene delivery systems. The current study focused on MNP preparation methods and their characterization by different techniques, their applications to targeted delivery, non-viral vectors in genetic manipulation, and challenges in clinical trial translation.

Abstract:It is generally recognized that nanoparticles possess unique physicochemical properties that are largely different from those of conventional materials, specifically the electromagnetic properties of magnetic nanoparticles (MNPs). These properties have attracted many researchers to launch investigations into their potential biomedical applications, which have been reviewed in this article. First, common types of MNPs were briefly introduced. Then, the biomedical applications of MNPs were reviewed in seven parts: magnetic resonance imaging (MRI), cancer therapy, the delivery of drugs and genes, bone and dental repair, tissue engineering, biosensors, and in other aspects, which indicated that MNPs possess great potentials for many kinds of biomedical applications due to their unique properties. Although lots of achievements have been obtained, there is still a lot of work to do. New synthesis techniques and methods are still needed to develop the MNPs with satisfactory biocompatibility. More effective methods need to be exploited to prepare MNPs-based composites with fine microstructures and high biomedical performances. Other promising research points include the development of more appropriate techniques of experiments both in vitro and in vivo to detect and analyze the biocompatibility and cytotoxicity of MNPs and understand the possible influencing mechanism of the two properties. More comprehensive investigations into the diagnostic and therapeutic applications of composites containing MNPs with "core-shell" structure and deeper understanding and further study into the properties of MNPs to reveal their new biomedical applications, are also described in the conclusion and perspectives part.

Abstract:The noteworthy intensification in the development of nanotechnology has led to the development of various types of nanoparticles. The diverse applications of these nanoparticles make them desirable candidate for areas such as drug delivery, coasmetics, medicine, electronics, and contrast agents for magnetic resonance imaging (MRI) and so on. Iron oxide magnetic nanoparticles are a branch of nanoparticles which is specifically being considered as a contrast agent for MRI as well as targeted drug delivery vehicles, angiogenic therapy and chemotherapy as small size gives them advantage to travel intravascular or intracavity actively for drug delivery. Besides the mentioned advantages, the toxicity of the iron oxide magnetic nanoparticles is still less explored. For in vivo applications magnetic nanoparticles should be nontoxic and compatible with the body fluids. These particles tend to degrade in the body hence there is a need to understand the toxicity of the particles as whole and degraded products interacting within the body. Some nanoparticles have demonstrated toxic effects such inflammation, ulceration, and decreases in growth rate, decline in viability and triggering of neurobehavioral alterations in plants and cell lines as well as in animal models. The cause of nanoparticles’ toxicity is attributed to their specific characteristics of great surface to volume ratio, chemical composition, size, and dosage, retention in body, immunogenicity, organ specific toxicity, breakdown and elimination from the body. In the current review paper, we aim to sum up the current knowledge on the toxic effects of different magnetic nanoparticles on cell lines, marine organisms and rodents. We believe that the comprehensive data can provide significant study parameters and recent developments in the field. Thereafter, collecting profound knowledge on the background of the subject matter, will contribute to drive research in this field in a new sustainable direction.

Abstract:Nanotechnology mainly deals with the production and application of compounds with dimensions in nanoscale. Given their dimensions, these materials have considerable surface/volume ratios, and hence, specific characteristics. Nowadays, environmentally friendly procedures are being proposed for fabrication of Fe nanoparticles because a large amount of poisonous chemicals and unfavorable conditions are needed to prepare them. This work includes an inclusive overview on the economical and green procedures for the preparation of such nanoparticles (flower, fruits, tea, carbohydrates, and leaves). Pure and bimetallic iron nanoparticles, for instance, offer a high bandwidth and excitation binding energy and are applicable in different areas ranging from antibacterial, anticancer, and bioimaging agents to drug delivery systems. Preparation of nano-sized particles, such as those of Fe, requires the application of high quantities of toxic materials and harsh conditions, and naturally, there is a tendency to develop more facile and even green pathways (Sultana, Journal of Materials Science & Technology, 2013, 29, 795-800; Bushra et al., Journal of hazardous materials, 2014, 264, 481-489; Khan et al., Ind. Eng. Chem. Res., 2015, 54, 76-82). This article tends to provide an overview on the reports describing green and biological methods for the synthesis of Fe nanoparticles. The present review mainly highlights selenium nanoparticles in the biomedical domain. Specifically, this review will present detailed information on drug delivery, bioimaging, antibacterial, and anticancer activity. It will also focus on procedures for their green synthesis methods and properties that make them potential candidates for various biomedical applications. Finally, we provide a detailed future outlook.

Abstract:Magnetic nanoparticles (MNPs) represent a subclass within the overall category of nanomaterials and are widely used in many applications, particularly in the biomedical sciences such as targeted delivery of drugs or genes, in magnetic resonance imaging, and in hyperthermia (treating tumors with heat). Although the potential benefits of MNPs are considerable, there is a distinct need to identify any potential toxicity associated with these MNPs. The potential of MNPs in drug delivery stems from the intrinsic properties of the magnetic core combined with their drug loading capability and the biomedical properties of MNPs generated by different surface coatings. These surface modifications alter the particokinetics and toxicity of MNPs by changing protein–MNP or cell–MNP interactions. This review contains current advances in MNPs for drug delivery and their possible organ toxicities associated with disturbance in body iron homeostasis. The importance of protein–MNP interactions and various safety considerations relating to MNP exposure are also addressed.

Abstract:INTRODUCTION: In the field of drug delivery, controlling the release of therapeutic substances at localized targets has become a primary focus of medical research, especially in the field of cancer treatment. Magnetic nanoparticles are one of the most promising drug carriers thanks to their biocompatibility and (super)paramagnetic properties. These properties allow for the combination between imaging modalities and specific release of drugs at target sites using either local stimulus (i.e. pH, conjugation of biomarkers, …) or external stimulus (i.e. external magnetic field).AREAS COVERED: This review provides an update on recent advances with the development of targeted drug delivery systems based on magnetic nanoparticles (MNPs). This overview focuses on active targeting strategies and systems combining both imaging and therapeutic modalities (i.e. theranostics). If most of the examples concern the particular case of cancer therapy, the possibility of using MNPs for other medical applications is also discussed.EXPERT OPINION: The development of clinically relevant drug delivery systems based on magnetic nanoparticles is driven by advantages stemming from their remarkable properties (i.e. easy preparation, facile chemical functionalization, biocompatibility, low toxicity, and superior magnetic responsiveness). This literature review shows that drug carriers based on magnetic nanoparticles can be efficiently used for the controlled release of drug at targeted locations mediated by various stimuli. Advances in the field should lead to the implementation of such systems into clinical trials, especially systems enabling drug tracking in the body.

Abstract:The remarkable multimodal functionalities of magnetic nanoparticles, conferred by their size and morphology, are very important in resolving challenges slowing the progression of nanobiotechnology. The rapid and revolutionary expansion of magnetic nanoparticles in nanobiotechnology, especially in nanomedicine and therapeutics, demands an overview of the current state of the art for synthesizing and characterizing magnetic nanoparticles. In this review, we explain the synthesis routes for tailoring the size, morphology, composition, and magnetic properties of the magnetic nanoparticles. The pros and cons of the most popularly used characterization techniques for determining the aforementioned parameters, with particular focus on nanomedicine and biosensing applications, are discussed. Moreover, we provide numerous biomedical applications and highlight their challenges and requirements that must be met using the magnetic nanoparticles to achieve the most effective outcomes. Finally, we conclude this review by providing an insight towards resolving the persisting challenges and the future directions. This review should be an excellent source of information for beginners in this field who are looking for a groundbreaking start but they have been overwhelmed by the volume of literature.

Abstract:With unique physical properties, chemical properties, and biological effects, magnetic nanomaterials are important functional materials in many fields. In the past decades, iron based magnetic nanomaterials have attracted much attention in the biomedicine field due to their superior magnetic properties and great potential in biomedical applications. In particular, magnetic iron oxide nanoparticles (MIONPs) have been playing a crucial role in the biomedicine field because of their diagnostic and therapeutic functions. Meanwhile, MIONPs are benign, low toxic, biocompatible, and biodegradable, so they are the only inorganic magnetic nanomaterials approved by the U.S. Food and Drug Administration (FDA) for clinical use at present. In this review, we mainly introduce the progress in the preparation of iron based magnetic nanomaterials for biomedical applications, including pure iron nanoparticles, iron-based alloy nanoparticles, and MIONPs, with a focus on MIONPs. Also, we summarize the preparation methods of MIONPs and point out the importance of their developments.

MNPs exhibit great potential as nanomaterials in various biomedical applications, including disease detection and cancer therapy. Typically, MNPs consist of a magnetic core surrounded by surface modification coatings, such as inorganic materials, organic molecules, and polymers, forming a nucleoshell structure that mitigates nanoparticle agglomeration and enhances targeting capabilities. Consequently, MNPs exhibit magnetic responsiveness in vivo for transportation and therapeutic effects, such as enhancing medical imaging resolution and localized heating at the site of injury. MNPs are utilized for specimen purification through targeted binding and magnetic separation in vitro, thereby optimizing efficiency and expediting the process. This review delves into the distinctive functional characteristics of MNPs as well as the diverse bioactive molecules employed in their surface coatings and their corresponding functionalities. Additionally, the advancement of MNPs in various applications is outlined. Additionally, we discuss the advancements of magnetic nanoparticles in medical imaging, disease treatment, and in vitro assays, and we anticipate the future development prospects and obstacles in this field. The objective is to furnish readers with a thorough comprehension of the recent practical utilization of MNPs in biomedical disciplines.

Abstract:Cancer, being one of the deadliest diseases, poses significant challenges despite the existence of traditional treatment approaches. This has led to a growing demand for innovative pharmaceutical agents that specifically target cancer cells for effective treatment. In recent years, the use of metal nanoparticles (NPs) as a promising alternative to conventional therapies has gained prominence in cancer research. Metal NPs exhibit unique properties that hold tremendous potential for various applications in cancer treatment. Studies have demonstrated that certain metals possess inherent or acquired anticancer capabilities through their surfaces. These properties make metal NPs an attractive focus for therapeutic development. In this review, we will investigate the applicability of several distinct classes of metal NPs for tumor targeting in cancer treatment. These classes may include gold, silver, iron oxide, and other metals with unique properties that can be exploited for therapeutic purposes. Additionally, we will provide a comprehensive summary of the risk factors associated with the therapeutic application of metal NPs. Understanding and addressing these factors will be crucial for successful clinical translation and to mitigate any potential challenges or failures in the translation of metal NP-based therapies. By exploring the therapeutic potential of metal NPs and identifying the associated risk factors, this review aims to contribute to the advancement of cancer treatment strategies. The anticipated outcome of this review is to provide valuable insights and pave the way for the advancement of effective and targeted therapies utilizing metal NPs specifically for cancer patients.

Abstract:Due to their good magnetic properties, excellent biocompatibility, and low price, magnetic iron oxide nanoparticles (IONPs) are the most commonly used magnetic nanomaterials and have been extensively explored in biomedical applications. Although magnetic IONPs can be used for a variety of applications in biomedicine, most practical applications require IONP-based platforms that can perform several tasks in parallel. Thus, appropriate engineering and integration of magnetic IONPs with different classes of organic and inorganic materials can produce multifunctional nanoplatforms that can perform several functions simultaneously, allowing their application in a broad spectrum of biomedical fields. This review article summarizes the fabrication of current composite nanoplatforms based on integration of magnetic IONPs with organic dyes, biomolecules (e.g., lipids, DNAs, aptamers, and antibodies), quantum dots, noble metal NPs, and stimuli-responsive polymers. We also highlight the recent technological advances achieved from such integrated multifunctional platforms and their potential use in biomedical applications, including dual-mode imaging for biomolecule detection, targeted drug delivery, photodynamic therapy, chemotherapy, and magnetic hyperthermia therapy.

Abstract:The recent decade has witnessed a remarkable surge in the field of nanoparticles, from their synthesis, characterization, and functionalization to diverse applications. At the nanoscale, these particles exhibit distinct physicochemical properties compared to their bulk counterparts, enabling a multitude of applications spanning energy, catalysis, environmental remediation, biomedicine, and beyond. This review focuses on specific nanoparticle categories, including magnetic, gold, silver, and quantum dots (QDs), as well as hybrid variants, specifically tailored for biomedical applications. A comprehensive review and comparison of prevalent chemical, physical, and biological synthesis methods are presented. To enhance biocompatibility and colloidal stability, and facilitate surface modification and cargo/agent loading, nanoparticle surfaces are coated with different synthetic polymers and very recently, cell membrane coatings. The utilization of polymer‐ or cell membrane‐coated nanoparticles opens a wide variety of biomedical applications such as magnetic resonance imaging (MRI), hyperthermia, photothermia, sample enrichment, bioassays, drug delivery, etc. With this review, our goal is to provide a comprehensive toolbox of insights into polymer or cell membrane‐coated nanoparticles and their biomedical applications, while also addressing the challenges involved in translating such nanoparticles from laboratory benchtops to in vitro and in vivo applications. Abstract:Magnetic iron oxide nanoparticles (MNPs) have been developed and applied for a broad range of biomedical applications, such as diagnostic imaging, magnetic fluid hyperthermia, targeted drug delivery, gene therapy and tissue repair. As one key element, reproducible synthesis routes of MNPs are capable of controlling and adjusting structure, size, shape and magnetic properties are mandatory. In this review, we discuss advanced methods for engineering and utilizing MNPs, such as continuous synthesis approaches using microtechnologies and the biosynthesis of magnetosomes, biotechnological synthesized iron oxide nanoparticles from bacteria. We compare the technologies and resulting MNPs with conventional synthetic routes. Prominent biomedical applications of the MNPs such as diagnostic imaging, magnetic fluid hyperthermia, targeted drug delivery and magnetic actuation in micro/nanorobots will be presented.

Abstract:Magnetic nanoparticles (NPs) are emerging as an important class of biomedical functional nanomaterials in areas such as hyperthermia, drug release, tissue engineering, theranostic, and lab-on-a-chip, due to their exclusive chemical and physical properties. Although some works can be found reviewing the main application of magnetic NPs in the area of biomedical engineering, recent and intense progress on magnetic nanoparticle research, from synthesis to surface functionalization strategies, demands for a work that includes, summarizes, and debates current directions and ongoing advancements in this research field. Thus, the present work addresses the structure, synthesis, properties, and the incorporation of magnetic NPs in nanocomposites, highlighting the most relevant effects of the synthesis on the magnetic and structural properties of the magnetic NPs and how these effects limit their utilization in the biomedical area. Furthermore, this review next focuses on the application of magnetic NPs on the biomedical field. Finally, a discussion of the main challenges and an outlook of the future developments in the use of magnetic NPs for advanced biomedical applications are critically provided.
