Strong Local Coordination Structure Effects on Subnanometer PtOx Clusters over CeO2 Nanowires Probed by Low-Temperature CO Oxidation

Abstract:A fundamental understanding of the structural effects of supported metal catalysts at the molecular level is extremely important for developing high-performance catalysts that are widely used in industry, which is still a longstanding attractive but challenging topic in multidisciplinary fields. In this work, we report the strong effects of local coordination structures on the catalytic activity of subnanometric PtOx clusters over CeO2 nanowires in low-temperature CO oxidation as a probe reaction. Atoms and subnanometric clusters of Pt were deposited to form the coordination structure of PtOx on the well-defined CeO2 nanowires with mainly exposed (110) facets. The reactivity of active sites and the variation of the local coordination structures of the PtOx sites were deeply investigated with in situ spectroscopic experiments, assisted by density functional theory simulations. According to our observation, although the highly dispersed Pt sites at the subnanometric scale could provide increased accessible sites, some of the Pt sites did not show high activity for CO oxidation due to the increased surrounding oxygen that seemed to overstabilize the Pt atoms. An increased proportion of both adsorbed CO intermediates on oxidized Pt sites and the interfacial lattice oxygen of PtOx clusters tended to become inactive on samples with a high coordination number of oxygen bonded to Pt sites (CN(Pt-O)), leading to the loss of effective active sites and a decrease in the catalytic activity. A relatively small CN(Pt-O) value in the subnanometric PtOx/CeO2 NWs, which was found to be the appropriate structure for their catalytic performance, remarkably increased the activity by about 1/2 order of magnitude. We believe our investigation on the interfacial coordination structure effects of subnanometric PtOx clusters dispersed on CeO2 nanowires can provide some new basic chemical insights into the metal-support interfacial interactions of Pt/CeO2 catalysts for understanding their catalytic performance in some relevant reactions.

Abstract:Nanozymes have shown great clinical application potential in the treatment of tumors with reactive oxygen species (ROS). However, conventional nanozyme technologies face challenges such as low catalytic activity and unknown therapeutic mechanisms for tumor therapy. Herein, we designed platinum (Pt)/nanoceria (CeO2) hybrid nanozymes using Pt single-atom and cluster engineering on the CeO2 surface, named Pt/CeO2-NR and Pt/CeO2-R respectively. The results showed that Pt/CeO2-R had higher enzyme-like activity than Pt/CeO2-NR because the Pt clusters (Pt0 and Pt2+) stabilized the oxygen vacancies better than the single Pt atoms (Pt2+). Further studies showed that Pt/CeO2-R exhibited selective toxicity to tumor cells, induced tumor cell apoptosis, and inhibited cell migration. In addition, we studied the antitumor mechanism of Pt/CeO2-R with stable isotopic labeling by amino acids in cell culture (SILAC)-based quantitative proteomics. Mechanistically, large amounts of the ROS produced by Pt/CeO2-R affected both the mitochondria and the endoplasmic reticulum and ultimately induced tumor cell apoptosis. Additionally, Pt/CeO2-R inhibited tumor cell migration by downregulating the proteins on the cell membrane surface. This study provided an approach for optimizing the enzyme-like activities of CeO2 through the efficient use of platinum engineering, and explored the mechanisms by which Pt/CeO2-R killed tumor cells and inhibited cell migration during the treatment of tumors.

Abstract:As a class of nanomaterials with natural enzyme-like characteristics, nanozymes have shown their great potential in various applications. Reducible metal oxides featured with defect structures, and single-atom catalysts with isolated metal sites are regarded as two of the most promising nanozymes. However, the strategies to construct highly performed nanozymes by combining these advantages are rarely reported. Herein, we report the coordination-unsaturated single-atomic Cu species supported on sintered CeO 2 , which combines the advantages of defect engineering and single-atom catalysis, exhibiting a largely enhanced peroxidase (POD)-like activity. The high-temperature calcination induces the transformation of inert Cu 1 O 4 species into coordination-unsaturated Cu 1 O 3 sites. This novel Cu 1 O 3 active sites with an unsaturated coordination work as a new type of defect sites to greatly activate the isolated Cu atoms and accelerate the dissociation of H 2 O 2 to form hydroxyl radicals (·OH). The obtained nanozyme with a high POD-like activity possesses low cytotoxicity, showing potential applications for the tumor inhibition in vitro and in vivo .

Unraveling Ros Conversion Through Enhanced Enzyme‐Like Activity with Copper‐Doped Cerium Oxide for Tumor Nanocatalytic Therapy

Abstract:A high degree of surface oxygen mobility and a low level of oxygen vacancy formation energy is exposed over the CeO2/Cu‐W surface, which is employed as a mimetic enzyme of superoxide dismutase (SOD) to convert O2∙‐ →H2O2 and peroxidase (POD) to realize H2O2→∙OH for oncotherapy. Nanozyme catalytic therapy for cancer treatments has become one of the heated topics, and the therapeutic efficacy is highly correlated with their catalytic efficiency. In this work, three copper‐doped CeO2 supports with various structures as well as crystal facets are developed to realize dual enzyme‐mimic catalytic activities, that is superoxide dismutase (SOD) to reduce superoxide radicals to H2O2 and peroxidase (POD) to transform H2O2 to ∙OH. The wire‐shaped CeO2/Cu‐W has the richest surface oxygen vacancies, and a low level of oxygen vacancy (Vo) formation energy, which allows for the elimination of intracellular reactive oxygen spieces (ROS) and continuous transformation to ∙OH with cascade reaction. Moreover, the wire‐shaped CeO2/Cu‐W displays the highest toxic ∙OH production capacity in an acidic intracellular environment, inducing breast cancer cell death and pro‐apoptotic autophagy. Therefore, wire‐shaped CeO2/Cu nanoparticles as an artificial enzyme system can have great potential in the intervention of intracellular ROS in cancer cells, achieving efficacious nanocatalytic therapy.

Abstract:Noble metals are downsized to nano-/subnanoscale to improve their catalytic activity and atom-economy. However, the stabilities in chemical state and catalytic performance of these nanocatalysts often suffer during harsh conditions. For Pt nanoparticles (NPs) supported on CeO2, activated oxygen diffused from the support over-stabilizes the active sites of Pt, degrading its performance at mild temperature. In this work, Pt nanocatalysts with unique structure of triple-junction are synthesized by selectively growing Pt NPs on the carbon-CeO2 interface. Impressively, the Pt NPs exhibit much enhanced catalytic stability and high activity for CO oxidation at mild temperature. The enhancement is attributed to electron donation from graphitized carbon and the confinement effect from the high-density nanopores of the CeO2 support. The triple-junction of Pt-C-CeO2, combining the merits of CeO2 for activating O2 and electron donating capability of carbon, provides new inspiration to the fabrication of high-performance nanocatalysts.
Oxygen vacancy-engineered cerium oxide mediated by copper-platinum exhibit enhanced SOD/CAT-mimicking activities to regulate the microenvironment for osteoarthritis therapy

Abstract:Cerium oxide (CeO 2 ) nanospheres have limited enzymatic activity that hinders further application in catalytic therapy, but they have an "oxidation switch" to enhance their catalytic activity by increasing oxygen vacancies. In this study, according to the defect-engineering strategy, we developed PtCuO X /CeO 2-X nanozymes as highly efficient SOD/CAT mimics by introducing bimetallic copper (Cu) and platinum (Pt) into CeO 2 nanospheres to enhance the oxygen vacancies, in an attempt to combine near-infrared (NIR) irradiation to regulate microenvironment for osteoarthritis (OA) therapy. As expected, the Cu and Pt increased the Ce 3+ /Ce 4+ ratio of CeO 2 to significantly enhance the oxygen vacancies, and simultaneously CeO 2 (111) facilitated the uniform dispersion of Cu and Pt. The strong metal-carrier interaction synergy endowed the PtCuO X /CeO 2-X nanozymes with highly efficient SOD/CAT-like activity by the decreased formation energy of oxygen vacancy, promoted electron transfer, the increased adsorption energy of intermediates, and the decreased reaction activation energy. Besides, the nanozymes have excellent photothermal conversion efficiency (55.41%). Further, the PtCuO X /CeO 2-X antioxidant system effectively scavenged intracellular ROS and RNS, protected mitochondrial function, and inhibited the inflammatory factors, thus reducing chondrocyte apoptosis. In vivo , experiments demonstrated the biosafety of PtCuO X /CeO 2-X and its potent effect on OA suppression. In particular, NIR radiation further enhanced the effects. Mechanistically, PtCuO X /CeO 2-X nanozymes reduced ras-related C3 botulinum toxin substrate 1 (Rac-1) and p -p65 protein expression, as well as ROS levels to remodel the inflammatory microenvironment by inhibiting the ROS/Rac-1/nuclear factor kappa-B (NF-κB) signaling pathway. This study introduces new clinical concepts and perspectives that can be applied to inflammatory diseases.

Abstract:There is considerable interest in the pH-dependent, switchable, biocatalytic properties of cerium oxide (CeO2) nanoparticles in biomedicine, where these materials exhibit beneficial antioxidant activity against reactive oxygen species (ROS) at a basic physiological pH but cytotoxic prooxidant activity in an acidic cancer cell pH microenvironment. While the general characteristics of the role of oxygen vacancies are known, the mechanism of their action at the atomic scale under different pH conditions has yet to be elucidated. The present work applies density functional theory (DFT) calculations to interpret, at the atomic scale, the pH-induced behavior of the stable {111} surface of CeO2 containing oxygen vacancies. Analysis of the surface-adsorbed media species reveals the critical role of pH on the interaction between ROS (•O2- and H2O2) and the defective CeO2 {111} surface. Under basic conditions, the superoxide dismutase (SOD) and catalase (CAT) biomimetic reactions can be performed cyclically, scavenging and decomposing ROS to harmless products, making CeO2 an excellent antioxidant. However, under acidic conditions, the CAT biomimetic reaction is hindered owing to the limited reversibility of Ce3+ ↔ Ce4+ and formation ↔ annihilation of oxygen vacancies. A Fenton biomimetic reaction (H2O2 + Ce3+ → Ce4+ + OH- + •OH) is predicted to occur simultaneously with the SOD and CAT biomimetic reactions, resulting in the formation of hydroxyl radicals, making CeO2 a cytotoxic prooxidant.
In-situ Formed Stable Pt Nanoclusters on Ceria-Zirconia Solid Solutions Induced by Hydrothermal Aging for Efficient Low-Temperature CO Oxidation

Abstract:Pt nanoclusters (Pt-C) on the surface of ceria-zirconia solid solutions exhibit superior low-temperature CO oxidation activity compared to Pt single atoms (Pt-1). However, the redispersion behavior of the Pt-C to Pt-1 lowers the efficiency of CO Oxidation. In this work, the stable surface hydroxyl groups were constructed by high-temperature hydrothermal aging treatment. Specifically, the Pt-1 catalyst (Pt/CZOe-SA) prepared by atom trapping was subjected to high-temperature hydrothermal treatment to obtain the stable Pt-C catalyst (Pt/CZOe-HT) with in-situ formed Pt-C. The experimental results showed that Pt-C with 2-3 nm size could achieve excellent low-temperature activity and thermal stability. In addition, the ab initio molecular dynamics and density functional theory calculations revealed the specific evolution process of dynamic dispersion for Pt-C with or without hydroxyl groups from the femtosecond scale, suggesting that the hydroxyl groups limited the dispersion of Pt-C in the form of "energy fence". The hydrothermal treatment not only introduced high-temperature stable hydroxyl groups to improve the stability of the in-situ formed Pt-C, but also optimized the ratio of Pt-1 and Pt-C, and then enhanced the low-temperature activity by the synergistic effect between Pt-1 and Pt-C. The present work can provide theoretical guidance for developing high-performance and high-stability Pt-based catalysts.
Nanozyme with Robust Catalase Activity by Multiple Mechanisms and Its Application for Hypoxic Tumor Treatment

Abstract:Utilizing catalase-mimicking nanozymes to produce O2 is an effective method to overcome tumor hypoxia. However, it is challenging to fabricate nanozymes with ultrahigh catalytic activity. Palladium nanosheet (Pd NS), a photothermal agent for photothermal therapy (PTT), has superior catalase-mimicking activity. Here, titanium dioxide (TiO2) is used to modify Pd NS (denoted Pd@TiO2) by a simple one-step method to improve its catalytic activity about 8 times. The enhancement mechanism's fundamental insights are discussed through experiments and density functional theory calculations. Next, zinc phthalocyanine is loaded on Pd@TiO2 to form a nanomotor (denoted PTZCs) with the synergistic activities of photodynamic therapy and PTT. PTZCs inherit the catalase activity of Pd@TiO2 to facilitate the decomposition of endogenous H2O2 to O2, which can relieve tumor hypoxia and propel PTZC migration to expand the reach of PTZCs, further enhancing its synergistic treatment outcome both in vitro and in vivo. It is proposed that this work can provide a simple and effective strategy for catalytic activity enhancement and bring a critical new perspective to studying and guiding the nanozyme design.

A Bimetallic Nanocatalyst for Light-Free Oxygen Sensitization Therapy

Abstract:Singlet oxygen (O-1(2)) is extensively employed by reactive oxygen species (ROS)-based cancer therapies, such as photodynamic therapy (PDT) and chemodynamic therapy (CDT). However, the dependences of PDT on light and CDT on complex chemodynamic reactions greatly limit their O-1(2)-generating efficiencies. Here, we exploit strain and electronic effects to fabricate a bimetallic nanocatalyst by coating a gold nanorod (AuNR) with similar to 2 platinum(Pt) atomic layers (AuPt0.09), which efficiently generates O-1(2) from ground- state oxygen (O-3(2)) by electron-transfer-mediated spin reduction. Density functional theory reveals strain, and electronic effects promote the adsorption of O-3(2) onto AuPt0.09, which dramatically enhances O-1(2) generation and imparts AuPt0.09, the highest catalytic constant ever reported for 3,3',5,5'-tetramethylbenzidine oxidation to the best of our knowledge. With a pH- dependent catalytic activity, AuPt0.09 realizes acidity-dependent antitumor effects both in vitro and in vivo, a proof-of-concept demonstration of autocatalytic bimetallic nanocatalyst for light-free oxygen sensitization therapy, which may open a new avenue for O-1(2)-centered therapeutics.

Abstract:Pyroelectric nanostructures could effectively generate temperature‐mediated reactive oxygen species (ROS) through the pyroelectric effect, providing promise for treating hypoxic tumors, and therefore, the synergistic application of photothermal therapy (PTT) and pyroelectric dynamic therapy (PEDT) presents an intriguing approach for cancer therapy. However, this method still faces challenges in improving pyroelectric catalysis and achieving precise tumor localization. In this study, a nano‐heterojunction based on CeO2‐BaTiO3 nanorods (IR1061@PCBNR) is reported, which exhibits highly effective pyroelectric catalysis for simultaneous tumor‐targeted dynamic therapy and gentle photothermal therapy through the utilization of the rich oxygen vacancies. The oxygen vacancies create active sites that facilitate the migration of pyroelectrically‐induced charge carriers, improving charge separation and ROS generation. IR1061@PCBNR also demonstrates high tumor penetration while minimizing damage to normal cells. This precise nanomedicine strategy holds great potential for advancing dynamic cancer therapies by overcoming the limitations of conventional approaches. This article is protected by copyright. Tumor microenvironment-activated single-atom platinum nanozyme with H2O2 self-supplement and O2-evolving for tumor-specific cascade catalysis chemodynamic and chemoradiotherapy

Abstract:Nanozyme-based tumor collaborative catalytic therapy has attracted a great deal of attention in recent years. However, their cooperative outcome remains a great challenge due to the unique characteristics of tumor microenvironment (TME), such as insufficient endogenous hydrogen peroxide (H2O2) level, hypoxia, and overexpressed intracellular glutathione (GSH). Methods: Herein, a TME-activated atomic-level engineered PtN4C single-atom nanozyme (PtN4C-SAzyme) is fabricated to induce the "butterfly effect" of reactive oxygen species (ROS) through facilitating intracellular H2O2 cycle accumulation and GSH deprivation as well as X-ray deposition for ROS-involving CDT and O2-dependent chemoradiotherapy. Results: In the paradigm, the SAzyme could boost substantial ∙OH generation by their admirable peroxidase-like activity as well as X-ray deposition capacity. Simultaneously, O2 self-sufficiency, GSH elimination and elevated Pt2+ release can be achieved through the self-cyclic valence alteration of Pt (IV) and Pt (II) for alleviating tumor hypoxia, overwhelming the anti-oxidation defense effect and overcoming drug-resistance. More importantly, the PtN4C-SAzyme could also convert O2·- into H2O2 by their superior superoxide dismutase-like activity and achieve the sustainable replenishment of endogenous H2O2, and H2O2 can further react with the PtN4C-SAzyme for realizing the cyclic accumulation of ∙OH and O2 at tumor site, thereby generating a "key" to unlock the multi enzymes-like properties of SAzymes for tumor-specific self-reinforcing CDT and chemoradiotherapy. Conclusions: This work not only provides a promising TME-activated SAzyme-based paradigm with H2O2 self-supplement and O2-evolving capacity for intensive CDT and chemoradiotherapy but also opens new horizons for the construction and tumor catalytic therapy of other SAzymes.
CeO2 and Glucose Oxidase Co-Enriched Ti3C2Tx MXene for Hyperthermia-Augmented Nanocatalytic Cancer Therapy

Abstract:Foreseen as foundational in forthcoming oncology interventions are multimodal therapeutic systems. Nevertheless, the tumor microenvironment (TME), marked by heightened glucose levels, hypoxia, and scant concentrations of endogenous hydrogen peroxide could potentially impair their effectiveness. In this research, two-dimensional (2D) Ti3C2 MXene nanosheets are engineered with CeO2 nanozymes and glucose oxidase (GOD), optimizing them for TME, specifically targeting cancer therapy. Following our therapeutic design, CeO2 nanozymes, embodying both peroxidase-like and catalase-like characteristics, enable transformation of H2O2 into hydroxyl radicals for catalytic therapy while also producing oxygen to mitigate hypoxia. Concurrently, GOD metabolizes glucose, thereby augmenting H2O2 levels and disrupting the intracellular energy supply. When subjected to a near-infrared laser, 2D Ti3C2 MXene accomplishes photothermal therapy (PTT) and photodynamic therapy (PDT), additionally amplifying cascade catalytic treatment via thermal enhancement. Empirical evidence demonstrates robust tumor suppression both in vitro and in vivo by the CeO2/Ti3C2-PEG-GOD nanocomposite. Consequently, this integrated approach, which combines PTT/PDT and enzymatic catalysis, could offer a valuable blueprint for the development of advanced oncology therapies.

Abstract:Nanostructured cerium-based catalytic materials have attracted much attention in the field of redox catalysis, due to the excellent active sites exposed on the nanocrystal surface, and herein, the oxygen vacancy (OV) and the crystal facet effect of typical model nanostructured ceria (Nano-CeO2) were focused and investigated for the catalytic aerobic cyclohexane oxidation. Specifically for the CeO2 nanocubes with a {100} facet, the CeO2 nanopolyhedrons with a dominant {111} facet, and the CeO2 nanorods with a dominant {110} facet, the catalytic roles of the OV and acid sites toward cyclohexane oxidation were systematically investigated. Surface oxygen vacancy (Sur-OV) and intrinsic oxygen vacancy (Int-OV), and particularly the active Sur-OV and active Int-OV performing catalytic activity under a working state specific for cyclohexane oxidation, were accurately recognized and distinguished. Furthermore, the activation ability of the OV sites for activating molecular oxygen into active oxygen species (OActive) was also revealed, and the acid site properties, which include the acidity and the types of acid sites, were probed and quantified. The Nano-CeO2 with an exposed {100} facet possesses the most active Sur-OV, determining the catalytic activity, and has the strongest activation ability, contributing to a high reaction activity under extremely inert conditions. The Nano-CeO2 with a {110} dominant facet with the most active Int-OV and abundant acid sites could accelerate further oxidation to adipic acid; the effect of the two key sites was described particularly by the α factor (= Total Acidity × Amount of Active Int-OV). Finally, the property–performance relationship toward cyclohexane oxidation was speculated based on the Nano-CeO2 catalytic system.

Abstract:Nanocatalytic medicine was attracting considerable attention, owing to the specific treatment by the intrinsic chemical reactions within tumors. However, the insufficient hydrogen peroxide (H 2 O 2 ) and unsuitable pH value limited its application. Herein, a tumor microenvironment (TME) responsive multifunctional nanoprobe with self-accelerating cascade catalysis (MPB@CeO 2 -GOx) was developed for enhanced catalytic therapy, in which ultrasmall cerium oxide (CeO 2 ) and glucose oxidase (GOx) were modified on mesoporous Prussian blue (MPB). The developed MPB@CeO 2 @-GOx nanoprobes exhibited the dual-nanozyme activity of catalase (CAT) and peroxidase (POD), by which the O 2 and hydroxyl radical (·OH) were generated by Ce 4+ and Ce 3+ . Moreover, the sufficient O 2 promoted the glucose consumption of GOx, and the generated H 2 O 2 and gluconic acid further enhanced the catalytic therapy by reducing pH value, by which ·OH yield increased by 26.21%. In addition, the photothermal effect of MPB also significantly improved the POD activity of CeO 2 . Compared with MPB@CeO 2 , the cell survival reduced from 20.8% to 3.8%, and the tumor growth was completely inhibited. Therefore, by self-supplying O 2 /H 2 O 2 and reducing pH value, the developed MPB@CeO 2 -GOx nanoprobes with self-accelerating cascade reactions achieved PTT and enhanced catalytic therapy against breast cancer.

In Vivo Regenerable Cerium Oxide Nanozyme-Loaded pH/H 2 O 2 -Responsive Nanovesicle for Tumor-Targeted Photothermal and Photodynamic Therapies

Abstract:Photodynamic therapy (PDT) and photothermal therapies (PTTs) are both promising strategies for effective tumor therapy. However, the absence of O2 at tumor sites hinders the sustained response of photosensitizers. Here, we develop a recycled cerium oxide (CeO2) catalase nanozyme-loaded hyaluronic acid nanovesicle to address the hypoxic tumor microenvironments and targeted delivery of the photosensitizers [indocyanine green (ICG)] to tumors. A polysaccharide complex effectively modifies the surface of a polyethylenimine phenylboronic acid nanostructure to achieve the CeO2 nanozyme-loading nanovesicles that exhibit both tumor-targeted enhancement and an improved hypoxic microenvironment. Also, the hydrogen peroxide responsiveness and acid-sensitive cleavage of phenylboronic acid specifically disintegrate the ICG/nanozyme coloaded nanovesicles in the tumor microenvironment. The in vitro synergistic tests and tumor suppression rate tests indicated that the cerium oxide nanozyme significantly improves the outcomes of PDT via cerium-element valence state recycling and hypoxia improvement, thus enhancing the tumor suppression efficiency. Abstract:ROS-based tumor therapy based on nanocatalytic medicine has recently been proposed for its tumor-specificity. However, a safe and highly efficient strategy towards getting high enough ROS to kill the hypoxic cancer cells is still a great challenge. Herein, we report a simple pH/H2O2-activatable, O2-evolving, and ROS regulating doxorubicin (DOX) and indocyanine green (ICG) co-loading PEGylated polyaniline (PANI) coated CeOx@polyacrylic acid (PAA) nanoclusters for highly selective and optimized cancer combination treatment. It can selectively and greatly enhance intracellular O2 and ROS levels in tumor region, which depends on two-step catalytic properties of nanoceria (Ce4+/Ce3+ = 3.46, neutral surface charge, mostly localize into the cytoplasm, pH 7.4–6.5, catalase-like catalytic agents convert to Ce4+/Ce3+ = 0.58, negative surface charge, mostly localize into the lysosomes, pH 5–4, oxidase-like catalytic agents, triggered by near infrared (NIR) laser irradiation). Furthermore, the protective effect of polyethylene glycol (PEG), PANI, and PAA ensure that the nanoceria can only play the role of catalase under the irradiation of NIR light arrived at the tumor area. Moreover, loading of nanoceria and ICG onto PANI greatly enhanced photo thermal effect of nanoparticles (NPs), which is useful for killing cancer cells by relieving hypoxia and promoting cross-membrane drug delivery to further enhance photodynamic therapy and chemotherapy efficiency. The chemo-photo combination therapies fantastically inhibited tumor growth and prevented tumor recurrence in vivo, suggesting a smart nanotheranostic system to achieve more precise and effective therapies in O2-deprived tumor tissue.

Abstract:Cerium oxide (CeO2-x) performs well in photothermal and catalytic properties due to its abundance of oxygen vacancies. Based on this, we designed a thermosensitive therapeutic nanoplatform to achieve continuous circular drug release in tumor. It can solve the limitation caused by insufficient substrate in the process of tumor treatment. Briefly, CeO2-x and camptothecin (CPT) were wrapped in an agarose hydrogel, which could be melted by the photothermal effect of CeO2-x. At the same time, the local temperature increase provided photothermal treatment, which could induce the apoptosis of tumor cell. After that, CPT was released to damage the DNA in tumor cells to realize chemical treatment. In addition, CPT could active nicotinamide adenine dinucleotide oxidase to react with O2 to increase the intracellular H2O2. After that, the exposed CeO2-x could catalyze H2O2 to generate cytotoxic reactive oxygen species for chemodynamic therapy. More importantly, CeO2-x could catalyze H2O2 to produce O2, which could combine with the catalytic action of CPT to construct a substrate self-cycling nanoenzyme system. Overall, this self-cycling nanoplatform released hypoxia in the tumor microenvironment and built a multimode tumor treatment, which achieved an ideal antitumor affect.

Abstract:Nanozymes have obvious advantages in improving the efficiency of cancer treatment. However, due to the lack of tissue specificity, low catalytic efficiency, and so on, their clinical applications are limited. Herein, the nanoplatform CeO2@ICG@GOx@HA (CIGH) with self‐accelerated cascade reactions is constructed. The as‐prepared nanozyme shows the superior oxidase (OXD)‐like, superoxide dismutase (SOD)‐like, catalase (CAT)‐like, and peroxidase (POD)‐like activities. At the same time, under 808 nm near‐infrared (NIR) irradiation, the photodynamic and photothermal capabilities are also significantly enhanced due to the presence of indocyanine green (ICG). We demonstrate that the nanozyme CIGH can efficiently accumulate in the tumor and exhibit amplified cascade antitumor effects with negligible systemic toxicity through the combination of photodynamic therapy (PDT), photothermal therapy (PTT), chemodynamic therapy (CDT) and starvation therapy. The nanozyme prepared in this study provides a promising candidate for catalytic nanomedicines for efficient tumor therapy.

Abstract:CeO2 nanozymes have garnered significant attention in chemodynamic therapy due to their peroxidase-like activity and ability to deplete glutathione. However, their catalytic efficiency is constrained by the low conversion rate of Ce3+/Ce4+. To overcome this limitation, the electron transfer was accelerated by introducing the transition metal Mn atom as a valence electron donor through lattice charge transfer. In detail, we introduced Ce1-xMnxO2 into the poly-L-lactic acid (PLLA) scaffold fabricated by selective laser sintering. The conversion from Ce3+ to Ce4+ promoted the decomposition of H2O2 within the tumor microenvironment to generate hydroxyl radicals with high oxidative activity, consequently inducing oxidative stress. The conversion from Ce4+ to Ce3+ depleted intracellular antioxidant glutathione, disrupting redox balance in tumor cells. This continuous redox cycle ultimately triggered apoptosis in tumor cells. Using a first-principles Hubbard-corrected approximate density-functional method, the analysis of the electron band structure revealed the presence of donor energy levels within the bandgap of Ce1-xMnxO2, enabling electron transfer channels around 0.645 eV. Electrochemical experiments confirmed that Ce0.8Mn0.2O2 significantly reduced the activation overpotential from 0.907 V to 0.646 V, enhancing its redox capacity. As a result, the scaffold exhibited a threefold increase in tumor-killing rate. These findings highlight the immense potential of CeO2 nanozymes in enhancing chemodynamic therapy for effective tumor treatment.
