Abstract:A platinum(IV) prodrug, cis,cis,trans-[Pt(NH3)2Cl2(biotin)(L)] (1), derived from cisplatin, where HL is the PEGylated red-light active boron-dipyrromethene (BODIPY) ligand, was synthesized, characterized and its photocytotoxicity evaluated. The complex showed a near-IR absorption band at 653 nm (ε ∼9.19 × 104 M-1 cm-1) in dimethyl sulfoxide and Dulbecco's phosphate-buffered saline (1 : 1 v/v) at pH 7.2. When excited at 630 nm, it showed an emission band at 677 nm in DMSO with a fluorescence quantum yield of 0.13. The 1,3-diphenylisobenzofuran titration experiment gave a singlet oxygen quantum yield (ΦΔ) of ∼0.32. A mechanistic DNA photocleavage study revealed singlet oxygen as the reactive oxygen species (ROS). The complex with biotin and PEGylated-distyryl-BODIPY showed significantly higher cellular uptake in A549 cancer cells as compared to non-cancerous Beas-2B cells from flow cytometry, indicating selectivity towards cancer cells. A dichlorodihydrofluorescein diacetate assay showed cellular ROS generation. Confocal images revealed predominant internalization in the mitochondria. The prodrug showed remarkable photodynamic therapy (PDT) activity in cancerous A549 and multidrug-resistant MDA-MB-231 cells with a high photocytotoxicity index value (half-maximal inhibitory concentration (IC50): 0.61-1.54 μM in red light), while being non-toxic in the dark. The chemo-PDT activity was significantly less in non-tumorigenic lung epithelial cells (Beas-2B). The prodrug effectively triggered cellular apoptosis, which was confirmed by the Annexin V-FITC/propidium iodide assay, and the alteration of the mitochondrial membrane potential was substantiated by the JC-1 dye assay. The β-tubulin immunofluorescence assay confirmed that incubating the cells with a light-treated complex resulted in the rapture of the cytoskeletal structure and the formation of apoptotic bodies. The results demonstrate that the prodrug triggered apoptosis via DNA damage, a reduction in mitochondrial function and disruption of the cytoskeletal framework.

Abstract:Organelle and nucleus dual-targeted anticancer drugs are being increasingly used for efficient cancer therapy as they can attack the double vital sites of tumor cells. In this work, we synthesized and characterized two new porphyrin compounds Pt-Por-RB and Me-Por-RB. The spectral titration results suggest that both Pt-Por-RB and Me-Por-RB bind to DNA efficiently in an intercalation binding mode. Upon irradiation, Pt-Por-RB with low dark-cytotoxicity can rapidly generate singlet oxygen to damage the tumor cells through the process of photodynamic therapy. Compared with Me-Por-RB, Pt-Por-RB was not only internalized in the organelles, but also in the nuclei of HeLa cells, probably due to the presence of platinum complexes, as analyzed using the confocal laser scanning microscope. Thus, with the combination of organelle and nucleus dual-targeting property and high efficiency of singlet oxygen generation, Pt-Por-RB showed a significant therapeutic activity against tumor cells.

Abstract:From the experiment of the Magnus method using an isolated hind gut of the crayfish , the antagonism between acetylcholin (ACh) and 4-aminobutyric acid (GABA) was tested , according to the technique of the drug-action curve analysis . From these results, it is suggested that the hind gut of the crayfish may have two receptor systems R1 and R2, to ACh, and that the antagonism between ACh and GABA includes the two non-competi tive types to R1 or R2, respectively. BODIPY-Tagged Platinum(II) Curcumin Complexes for Endoplasmic Reticulum-Targeted Red Light PDT

Abstract:[Pt(RB)(Cur)]NO3 (RBC), [Pt(IRB)(Cur)]NO3 (IRBC), and [Pt(L)(Cur)]NO3 (PBC), where HCur is curcumin, L is 1-benzyl-2-(2-pyridyl)benzimidazole, and RB and IRB are red-light-active non-iodo and diiodo-BODIPY tagged to L, respectively, were synthesized and characterized, and their anticancer activities were studied (BODIPY, boron-dipyrromethene). RBC and IRBC displayed BODIPY-centered absorption bands within 615-635 nm along with the respective curcumin bands at 445 and 492 nm in 10% dimethyl sulfoxide (DMSO)-Dulbecco's phosphate-buffered saline (DPBS). Emission bands were observed at 723 and 845 nm for RBC and IRBC, respectively, in 10% DMSO-DPBS. RBC (ΦΔ, 0.27) and IRBC (ΦΔ, 0.40) generated singlet oxygen in red light (λ = 642 nm) as evidenced from 1,3-diphenylisobenzofuran (DPBF) titrations. The formation of 1O2 from BODIPY and HO• from the curcumin was evidenced from the mechanistic pUC19 DNA photocleavage studies. The BODIPY complexes showed photocytotoxicity in A549, HeLa, and MDA-MB-231 cells while being less toxic in the dark [IC50: 1.3-6.9 μM, red light; 7.2-12.8 μM, 400-700 nm visible light]. The emissive RBC displayed localization in the endoplasmic reticulum (ER). Apoptotic cell death was evidenced from the Annexin-V/fluorescein isothiocyanate (FITC)/propidium iodide (PI) assay and green fluorescence in red light in the Fluo-4 AM assay due to ER stress, and mitochondrial dysfunction was evidenced from the 5,5,6,6'-tetrachloro-1,1',3,3'-tetraethylbenzimidazolylcarbocyanine iodide (JC-1) assay in A549 cells.
Synthesis, Reactivity Studies and Cytotoxicity of Two Iodidoplatinum(II) Complexes. Does Photoactivation Work?

Abstract:Trans platinum complexes have been the landmark in unconventional drugs prompting the development of innovative structures that might exhibit chemical and biological profiles different to cisplatin. Iodido complexes made a new turning point in the platinum drug design field since their cytotoxicity was reevaluated and reported. In this new study, we have synthesized and evaluated diodido complexes bearing aliphatic amines and pyridines in trans configuration. X-ray diffraction support the structural characterization. Their cytotoxicity has been evaluated in tumor cell lines such as SAOS-2, A375, T-47D and HCT116. Moreover, we report their solution behavior and reactivity with biological models. UVA irradiation induces an increase in their reactivity towards model nucleobase 5´-GMP in early stages, and promotes the release of the pyridine ligand (spectator ligand) at longer reaction times. Density Functional calculations have been performed and the results are compared with our previous studies with other iodido derivatives.
Cisplatin-appended BODIPY for near infrared II fluorescent and photoacoustic imaging-guided synergistic phototherapy and chemotherapy of cancer

Abstract:Combining phototherapy and chemotherapy has been considered a promising modality for cancer therapy due to their synergistic effect. Herein, we developed three D-π-A-structured boron dipyrromethenes (BODIPYs) (named as B-B, B-C, and B-C-Pt). Due to their enlarged π-conjugated structure and high intramolecular charge transfer effect, the synthesized BODIPYs had photothermal conversion capability, and their absorption and fluorescence spectra were red-shifted. The cisplatin-appended BODIPY (B-C-Pt) exhibited good singlet oxygen (1O2) generation ability and near infrared (NIR) absorption and fluorescence (λAbs = 748 nm, λEm = 947 nm). After being encapsulated by distearoyl phosphoethanolamine polyethyleneglycol 2000 (DSPE-PEG-2000), which could inhibit the H-aggregation of B-C-Pt, the absorption and fluorescence of the obtained B-C-Pt nanoparticles (NPs) were red-shifted to 762 and 985 nm, respectively. The 1O2 quantum yield and photothermal conversion efficiency of the B-C-Pt NPs were 4.0% and 40.6%, respectively. Moreover, B-C-Pt NPs had chemotherapeutic efficacy due to the presence of cisplatin. In vitro and in vivo studies further demonstrated that B-C-Pt NPs had synergistic therapeutic efficacy. Together, B-C-Pt NPs could be employed in NIR II fluorescent and photoacoustic imaging-guided synergistic phototherapy and chemotherapy for cancer treatment.

Abstract:Photodynamic therapy (PDT) has attracted extensive attention in cancer treatment because of its minimum trauma, less side effects, and so on. Photosensitizers, as one of the core elements of PDT, usually have to face problems such as poor water solubility and light stability, lack of targeting, and other problems, which seriously affect the therapeutic effect. In this work, two BODIPY (boron-dipyrromethene)-based monofunctional Pt (II) complexes, 1a and 2a, were designed and synthesized, and their PDT effect was studied. The Pt atom improved the singlet oxygen quantum yield (0.19 for 1a and 0.14 for 2a, respectively), which effectively improves the efficiency of PDT. MTT assay confirmed that the short time photo-irradiation distinctly promoted antitumor cytotoxicity of Pt (II) compounds against different cell lines. For 1a under irradiation, the IC50 value of cancer cell lines were 13.1 μM for HeLa cells and 7.6 μM for MCF-7 cells, while those of normal cell lines were 32.4 μM for HBL-100 cells and 48.6 μM for L02 cells. The results demonstrated that 1a showed specific phototoxicity to cancer cells. This specific selectivity could be attributed to the synergistic effect of increased cellular uptake (determined by ICP-MS) and higher ROS generation (detected by Cell ROX Deep Red) in cancer cells after irradiation. This study laid the foundation for the future design and synthesis of effective PDT photosensitizers.

Abstract:Photodynamic therapy (PDT) agents induce photodamage only where photosensitizers localize in tumor cells. Since apoptosis is a major pathway of PDT-mediated cell death in vivo , the therapeutic efficacy of PDT drugs against tumor cells may be improved by regulating intrinsic apoptosis. Mitochondria are the major intracellular organelles that regulate apoptosis, thus triphenylphosphonium, a well-known mitochondria-targeting molecule, was appended to a photoactive platinum diimine complex. The ability of the triphenylphosphonium-appended complex to generate singlet oxygen was assessed by fluorescence spectroscopy using 1,3-diphenylisobenzofuran as a probe. The photocytotoxicity towards HeLa cells (human cervical cancer cell line) or HL-7702 cells (human liver cell line) was assessed by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assays. The platinum complex has moderate capability to generate singlet oxygen and it has more potent photocytotoxicity than its analogue Pt-CA. Both platinum diimine complexes studied are largely nontoxic to human liver cells either in the dark or after irradiation.

Remarkable anticancer activity of ferrocenyl-terpyridine platinum(ii) complexes in visible light with low dark toxicity

Abstract:Ferrocenyl platinum(ii) complexes (), viz. [Pt(Fc-tpy)Cl]Cl (), [Pt(Fc-tpy)(NPC)]Cl (, HNPC = N-propargyl carbazole) and [Pt(Fc-bpa)Cl]Cl (), were prepared, characterized and their anti-proliferative properties in visible light in human keratinocyte (HaCaT) cell lines have been studied. [Pt(Ph-tpy)Cl]Cl () was prepared and used as a control. Complexes and , structurally characterized by X-ray crystallography, show distorted square-planar geometry for the platinum(ii) centre. Complexes and having the Fc-tpy ligand showed an intense absorption band at ∼590 nm. The ferrocenyl complexes are redox active showing the Fc(+)-Fc couple near 0.6 V vs. SCE in DMF-0.1 M tetrabutylammonium perchlorate (TBAP). Complexes showed external binding to calf thymus DNA. Both and showed remarkable photocytotoxicity in HaCaT cell lines giving respective IC50 values of 9.8 and 12.0 μM in visible light of 400-700 nm with low dark toxicity (IC50 >60 μM). Fluorescent imaging studies showed the spread of the complexes throughout the cell localising both in cytoplasm and the nucleus. The ferrocenyl complexes triggered apoptosis on light exposure as evidenced from the Annexin V-FITC/PI and DNA ladder formation assays. Spectral studies revealed the formation of ferrocenium ions upon photo-irradiation generating cytotoxic hydroxyl radicals via a Fenton type mechanism. The results are rationalized from a TDDFT study that shows involvement of ferrocene and the platinum coordinated terpyridine moiety as respective HOMO and LUMO.
Synthesis and Biological Activity of Binuclear Platinum Complexes Containing Two Monofunctional Cis-[Pt(Nh3)(2)Cl](+) Units Bridged by 4,4 '-Dipyridyl Selenides or Sulfides

Abstract:The synthesis of four binuclear platinum complexes of general formula ¿cis-[Pt(NH3)2Cl]2(L)¿ (NO3)2 [L is 4,4'-dipyridyl sulfide (compound I), 4,4'-dipyridyl selenide (compound II), bis(3-methyl-4-pyridyl) sulfide (compound III) or bis(3-methyl-4-pyridyl) selenide (compound IV)] has been achieved. These compounds have been characterized by elemental analysis, IR, 1H-NMR, 13C-NMR and 195Pt-NMR spectroscopy. The above dinuclear platinum complexes have been assayed for antitumor activity in vitro against the cisplatin-sensitive L1210 (the mice leukemia) and cisplatin-insensitive HCT8 (the human coloadenocarcinoma) cell lines. The compounds show IC50 values comparable to or higher than cisplatin against L1210 cell line; however, they have lower IC50 values than cisplatin against the cisplatin-insensitive HCT8 cell line. The complexes containing S exhibit a cytotoxicity against the two cancer cell lines superior to the Se analogues. DNA binding studies indicate the compound I possibly interacts with DNA nonintercalatively. The mode of DNA binding of the dinuclear Pt(II) complexes bridged by 4,4'-dipyridyl selenides or sulfides may be different to that of the aliphatic diaminebridged dinuclear Pt(II) complexes reported previously.
Synthesis and Photoactivity of a Pt(II) Complex Based on an O-Nitrobenzyl-derived Ligand

Abstract:Development of photoactivatable platinum complexes was an attractive approach to improve the anticancer behavior of Pt-based agents. In this work, a new photolabile Pt(II) complex, NMPt, was prepared using a photosensitive compound, 2-hydroxy-2-(2-nitrophenyl)acetic acid (HNPAC). X-ray crystallographic data disclosed that Pt(II) center of NMPt adopts a square-planar geometry similar to that of nedaplatin. In NMPt molecule, the Pt(II) center is coordinated by two oxygen atoms of the hydroxyl and carboxyl groups in HNPAC ligand and two ammine ligands. The quick conversion of NMPt (100μM) in PBS buffer upon irradiation at 365nm was observed, displaying a distinct absorption bathochromic shift from 265 to 300nm. The 1H NMR determination also confirmed the UV irradiation-induced conversion of NMPt complex. The CD spectroscopic determination revealed that NMPt is able to distort the conformation of CT-DNA only upon UV-irradiation. In addition, agarose gel analysis of plasmid pBR 322 DNA demonstrated that NMPt induces plasmid DNA nicking upon UV-irradiation with a distinct dose- and time-dependence. Preliminary results showed that the cytotoxicity of NMPt at lower concentration (5–20μM) against human breast adenocarcinoma (MCF-7) cells is comparable to that of nedaplatin upon UV irradiation, while its cytotoxicity is very low without UV irradiation. Therefore, UV irradiation-induced DNA distortion and damage might be correlated to the photo-induced cytotoxicity enhancement for NMPt. This study provides an interesting approach for the exploration of photoactivatable Pt(II) complex as anticancer agent, although the photo-induced anticancer activity of NMPt remains to be improved.
Developing photo-activable ruthenium (II) complexes for PDT: Synthesis, characterization, photophysical and biological studies

Abstract:The easy tunable photophysical properties of Ru(II) polypyridyl complexes, together with their water solubility, photochemical stability and high biocompatibility make them suitable as agents for photodynamic therapy (PDT). The development of complexes containing new ligands is a step forward in the improvement and application of these compounds as photosensitizer and photocytotoxic agents. We report the synthesis of a set of Ru(II)-polypyridyl complexes with different electronic properties and delocalized π systems: one homoleptic complex [Ru II (dpbpy) 3 ](PF 6 ) 2 ( 1 ), and three heteroleptic complexes [Ru II (dpbpy) 2 (phen)](PF 6 ) 2 ( 2 ), [Ru II (dpbpy)(phen) 2 ](PF 6 ) 2 ( 3 ) and [Ru II (dpbpy) 2 (CN–Me)](PF 6 ) 2 ( 4 ). All of them contain 4,4′-diphenyl-2,2′-bipyridine ( dpbpy ) and 1,10-phenantroline ( phen ) or N-methyl-N′-2-pyridilimidazolium (CN–Me) ligands. The complexes have been characterized by spectroscopic, structural and electrochemical methods. UV–vis absorption in solution show the red shift for the metal to ligand charge transfer (MLCT) transitions after changing phen by dpbpy and carbene ligands. The photoluminescence emission spectra of 1 – 4 supported by theoretical calculations using time-dependent density-functional theory (TDDFT) suggest that the lowest energy excited state is mainly 3 MLCT. Complex 2 exhibits the highest phosphorescence emission with a quantum yield (Φ P ) in CH 2 Cl 2 of 44.1 %, followed by complex 1 (Φ P = 40.0 %), whereas complex 4 shows the lowest quantum efficiency (Φ P = 16.0 %), what suggest that the introduction of the CN–Me carbene ligand produces a significant quenching of the phosphorescence. Phosphorescence triplet state lifetimes between 0.96 and 1.74 μs were shown for 1 – 4 in degassed CH 2 Cl 2 . All complexes are proven to be efficient singlet oxygen photosensitizers. The trend in the redox potentials becomes more positive by increasing the number of phen ligands in the complexes, in full agreement with the HOMO's energy level and the blue shift of the MLCT transitions in the absorption spectra. Complexes internalization was analysed in tumorigenic mammary epithelial SKBR-3 cells, being complexes 1 and 2 the most well internalized. The effect of the photodynamic treatment using light-activated complexes 1 and 2 for 10 min demonstrated to increase cell death, being the homoleptic complex 1 an outstanding candidate as potential theranostic agent for bioimaging and PDT.

Abstract:Real-time biodistribution monitoring and enhancing the therapeutic efficacy of platinum(II)-based anticancer drugs are urgently required to elevate their clinical performance. Herein, a tetraphenylethene derivative (TP) with aggregation-induced emission (AIE) properties and an iodine atom are selected as ligands to endow platinum (II) complex TP-Pt-I with real-time in vivo self-tracking ability by fluorescence (FL) and computerized tomography (CT) imaging, and improved anticancer efficacy by the combination of chemotherapy and photodynamic therapy. Especially, benefiting from the formation of a donor-acceptor-donor structure between the AIE photosensitizer TP and Pt-I moiety, the heavy atom effects of Pt and I, and the presence of I, TP-Pt-I displayed red-shifted absorption and emission wavelengths, enhanced ROS generation efficiency, and improved CT imaging capacity compared with the pristine TP and the control agent TP-Pt-Cl. As a result, the enhanced intratumoral accumulation of TP-Pt-I loaded nanoparticles is readily revealed by dual-modal FL and CT imaging with high contrast. Meanwhile, the TP-Pt-I nanoparticles show significantly improved tumor growth-inhibiting effects on an MCF-7 xenograft murine model by combining the chemotherapeutic effects of platinum(II) and the photodynamic effects of TP. This self-tracking therapeutic complex thus provides a new strategy for improving the therapeutic outcomes of platinum(II)-based anticancer drugs.

Abstract:The novel hetero-dinuclear complex trans , trans , trans -[Pt IV (py) 2 (N 3 ) 2 (OH)(μ-OOCCH 2 CH 2 CONHCH 2 -bpyMe)Ir III (ppy) 2 ]Cl ( Pt-Ir ), exhibits charge transfer between the acceptor photochemotherapeutic Pt(IV) ( Pt-OH ) and donor photodynamic Ir(III) ( Ir-NH 2 ) fragments. It is stable in the dark, but undergoes photodecomposition more rapidly than the Pt(IV) parent complex ( Pt-OH ) to generate Pt(II) species, an azidyl radical and 1 O 2 . The Ir(III) * excited state, formed after irradiation, can oxidise NADH to NAD⋅ radicals and NAD + . Pt-Ir is highly photocytotoxic towards cancer cells with a high photocytotoxicity index upon irradiation with blue light (465 nm, 4.8 mW/cm 2 ), even with short light-exposure times (10–60 min). In contrast, the mononuclear Pt-OH and Ir-NH 2 subunits and their simple mixture are much less potent. Cellular Pt accumulation was higher for Pt-Ir compared to Pt-OH . Irradiation of Pt-Ir in cancer cells damages nuclei and releases chromosomes. Synchrotron-XRF revealed ca . 4× higher levels of intracellular platinum compared to iridium in Pt - Ir treated cells under dark conditions. Luminescent Pt-Ir distributes over the whole cell and generates ROS and 1 O 2 within 1 h of irradiation. Iridium localises strongly in small compartments, suggestive of complex cleavage and excretion via recycling vesicles (e.g. lysosomes). The combination of PDT and PACT motifs in one molecule, provides Pt-Ir with a novel strategy for multimodal phototherapy.

Abstract:Here, we report a photodynamic therapy (PDT) photosensitizer of N^(∧)C^(∧)N-coordinated Pt(II) complexes: [Pt(L)(solv)]^(+) (HL = 1,3-(2-dipyridyl)benzene) and [Pt(L)]^(+)@HSA, which is the Pt(II) complex encapsulated in human serum albumin (HSA). Abstract:Light induced release of cisplatin from Pt(IV) prodrugs is a promising tool for precise spatiotemporal control over the antiproliferative activity of Pt-based chemotherapeutic drugs. A combination of light-controlled chemotherapy (PACT) and photodynamic therapy (PDT) in one molecule has the potential to overcome crucial drawbacks of both Pt-based chemotherapy and PDT via synergetic effect. Herein we report green-light activated Pt(IV) prodrugs GreenPt with BODIPY-based photosentitizer in axial position with incredible high light response and singlet oxygen generation ability. GreenPt demonstrated the ability to release cisplatin under low-dose green light irradiation up to 1 J/cm2. The investigation of the photoreduction mechanism of GreenPt prodrug using DFT modeling and ΔG0 PET estimation revealed that the anion-radical formation and substituent photoinduced electron transfer from the triplet excited state of the BODIPY axial ligand to the Pt(IV) center is the key step in the light-induced release of cisplatin. Green-light activated BODIPY-based photosentitizers 5 and 8 demonstrated outstanding photosensitizing properties with extraordinary phototoxicity index (PI) >1300. GreenPt prodrug demonstrated gradual intracellular accumulation and light-induced phototoxicity with PI > 100, thus demonstrating dual action through light-controlled release of both cisplatin and a potent BODIPY-based photosensitizer.

Abstract:A convenient synthesis of a BODIPY (1,3,5,7-tetramethyl-8-(4-pyridyl)-4,4'-difluoroboradiazaindacene) labeled platinum compound (BODIPY-Pt) was developed by direct conjugation of cisplatin with the pyridine group of BODIPY. The membrane permeability and selective uptake of BODIPY-Pt in the mitochondria was studied using confocal laser scanning microscopy (CLSM). The fluorescent BODIPY-Pt conjugate showed high cellular proliferation inhibition against human cervical carcinoma (HeLa) and human breast cancer (MCF-7) cells, with half maximal inhibitory concentrations (IC50) of 27.37 and 12.14 μM, respectively. This work highlights the potential of using BODIPY labeled Pt compounds to realize the visualization of Pt distribution in living cells.

Abstract:Triple-negative breast cancer (TNBC) is one of the most malignant breast tumors for its poor prognosis and high tumor recurrence. It is urgent to develop new strategy or effective agents to overcome resistance and improve therapeutic effectiveness. Boron-dipyrromethene (BODIPY) based photosensitizers possess exciting photophysical features suitable for theranostic applications, namely, photodynamic therapy (PDT). We have designed a luminescent monofunctional platinum(II) complex with BODIPY derivative, namely I2BC-Pt, as novel high PDT agent against triple negative breast cancer (TNBC). The di-iodinated BODIPY complex I2BC-Pt showed excellent PDT effect against TNBC cells in green light (520 nm) giving IC50 values of 0.11-0.13 μM in MDA-MB-231 and MDA-MB-468 cells. I2BC-Pt predominately aggregated in the mitochondria of MDA-MB-231 cells and emitted green fluorescence. Besides, the anticancer mechanism studies demonstrated that I2BC-Pt could help DNA repair through attenuating RAD51, FoxM1 and BRCA1/2, and induce p53-mediated apoptosis of TNBC cells. Taken together, I2BC-Pt could be potentially developed as a BODIPY-based photosensitizers for TNBC therapy.
