Abstract:Sappanwood is a resourceful, flowering tree belongs to the family Leguminosae with its origin mainly reported from Southeast Asia. It has been used for ages to cure human ailments including heart diseases, jaundice, respiratory problems, coughs, wounds, and allergies. The different parts of sappanwood including wood, heartwood, seeds, leaves, and bark are reported with bioactive constituents, which deliver essential assistance to the human body. Beyond contribution to health status, they also have encouraging benefits for food industries. The brazilin extracted from the heartwood part of sappanwood has been used as a food preservative and as an ingredient for functional foods. The studies prescribed in the given manuscript defined the role of sappanwood and its parts (wood, bark, seeds, leaves, and heartwood) in nutritional, food, and pharmacological area. Recent studies have shown several promising activities, particularly relaxant, digestive, antioxidant, antimicrobial, antispasmodic, anticancer, and antagonistic effects. The popularity of sappanwood because of its medicinal and industrial importance specifies comprehensive references for its nutritional benefits. The existent work stated the recent research conveyed till date for valuation of nutritional eminence, phytochemicals constituents, industrial applications, and pharmacological significance of sappanwood.

Abstract:Cardiovascular diseases are the leading cause of death worldwide. The long-term aim of cardiovascular disease therapy is to reduce the mortality rate and decelerate the progression of cardiovascular organ damage. Current therapies focus on recovering heart function and reducing risk factors such as hyperglycemia and dyslipidemia. However, oxidative stress and inflammation are important causes of further damage to cardiovascular organs. Caesalpinia sappan Linn. (Fabaceae), a flowering tree native to tropical Asia, has antioxidant and anti-inflammatory properties. It is used as a natural dye to color food and beverages and as a traditional treatment for diarrhea, diabetes, and blood stasis. The phytochemical compounds in C. sappan, mainly the homoisoflavonoids brazilin, sappanone A, protosappanin, and hematoxylin, can potentially be used to protect cardiovascular organs. This review aims to provide updates on recent developments in research on C. sappan in relation to treatment of cardiovascular diseases. Many studies have reported protective effects of the plant's bioactive compounds that reduce cardiac damage and enhance vasorelaxation. For example, brazilin and sappanone A have an impact on molecular and cellular changes in cardiovascular disease pathogenesis, mainly by modulating oxidative, inflammatory, and apoptotic signaling pathways. Therefore, bioactive compounds of C. sappan have the potential to be developed as therapeutic agents to combat cardiovascular diseases like myocardial infarction and vascular disease. This review could help further the understanding of the possible modulatory role of the compounds in cardiovascular diseases, thereby facilitating future studies.

Abstract:Caesalpinia sappan L. or known as sappan wood is a plant that had been used in traditional food, beverages and traditional medicine. It contains many active phytochemicals compound such as brazilin, sappanchalcone, xanthone, coumarin and flavones. Extraction is the common method to get the active compounds in plants and the use of new method of extraction is very recommended. The research was aimed to analyze the effect of different solvent (ethanol, water and 2-propanol) to the characteristic of sappan wood extract obtained from ultrasound assisted extraction (UAE) process. Approximately 10 g of sappan wood was mixed with 100 ml solvent and extracted using probe type of ultrasound extractor for 15 mins and 500C. Extracts were analyzed for their yield, color, phytochemical compound, total phenolic content (TPC) and antioxidant activity. Ethanol gives the highest yield (6.125%) with the color tends to be between ruby red and traffic red. This result is in accordance with the calculation of Hansen solubility parameter (HSP). All of the three extracts contained saponin, tannin, flavonoid and triterpenoid. Ethanol also gives the highest TPC (254.14 mg GAE/100 g) and lowest IC50 (3.19). It can be concluded that ethanol is better solvent to extract phytochemical component in sappan wood.

Abstract:To study the chemical constituents of Sappan Lignum.Chemical constituents were isolated by method of solvent extraction,repeated chromatography with silica gel,Sephadex LH-20,and ODS.The structures were elucidated based on spectroscopic data.Fourteen compounds were isolated and their structures were identified as brazilin (1),sappanone B (2),(E)-3-(3,4-dihydroxybenzylidene)-7-hydroxychroman-4-one (3),3-deoxysappanone B (4),brazilide A (5),euxanthone (6),quercetin (7),rhamnetin (8),sappanchalcone (9),3-deoxysappanchalcone (10),butein (11),2,4,5-trihydroxybenzaldehyde (12),3,8,9-trihydroxy6H-benzo[c]chromen-6-one (13) and ~-sitosterol (14).Compounds 12 and 13 were two new natural compounds,and the 13C NMR data of compound 13 were reported for the first time.Compound 6 was the first xanthone isolated from the genus Caesalpinia.
Contribution of extractives to the bark color of Caesalpinia sappan

Abstract:The purpose of this study was to investigate the contribution of Caesalpinia sappan (sappan wood) bark extractives by analyzing color change of the bark after extraction and the color of the extracts with several color measurement methods. Successive extraction was performed with n-hexane, ethyl acetate, methanol, and hot water. Color change of the bark was measured using CIELab color system and the extracts were analyzed with Ultraviolet-Visible Spectrophotometer, total phenolic content (TPC), and Gas Chromatography Mass Spectrometry (GC-MS). The results showed that the highest change on the bark color after methanol extraction with the Δa* and Δb* values of -2,53 ± 0,60 and -3,64 ± 1,20 respectively. Also, methanol extract showed the highest total phenolic content (860,24 ± 30,19 mg GAE/g). In addition, the Ultraviolet-Visible is analysis showed a peak at 478 nm in the hot-water soluble extract and two peaks in the methanol soluble extract at 396 nm and 478 nm. Hydroquinone was detected as one of the major compounds by Gas Chromatography Mass Spectrometry in the methanol soluble extract. It was suggested that the color of Caesalpinia sappan (sappan wood) bark as well as the deep red coloration of its extract might be contributed by multiple phenolic compounds contained in the methanol extract with hydroquinone as its precursor. Therefore, it is also a potential source for coloring matter.

Abstract:This is a review on the chemical constituents, pharmacology, toxicology and biosynthesis of Sphaerophysa. Eight compounds have been isolated from it, which include flavonoids, coumarins, alkaloids and sterol. The pharmacological experiments have concluded that the total flavonoidal glycosides of Swainsonia salsula Taub. had the effect of anti-hypertension and the swainsonine was an inhibitor of lysosomal alpha-mannosidase. The biosynthesis of Swainsonine in fungus had been proposed and its precursor was L-lysine.

This study provide biological function prediction and mapping through in silico approach. The compounds from Caesalpinia sappan were listed and downloaded from Knapsack webserver. PubChem NCBI was used to retrieve the canonical smiles of Caesalpinia sappan compounds. Biological activity of Caesalpinia were predicted by Online PASS Two Way drug website and performed in heatmap. Structure activity relationship (SAR) of quercetin-3,7-Di-O-Methyl Ether 3'-O-Methylbrazilin, 2'-Methoxyisoliquiritigenin, Isoliquiritigenin 2'-Methy Ether and Quercetin-3′,4′-Di-O-Methyl Ether revealed antioxidant, nitric oxide scavenger, antimycobacterial, antibacterial, antiinflammation, and act as lipoxygenase inhibitor agent. Furthermore, Quercetin-3′,4′-Di-O-Methyl Ether (17) proved higher antivirus activity than other compounds. 2'-Methoxyisoliquiritigenin (20) and Isoliquiritigenin 2'-Methy Ether (27) performed higher neuroprotective effect than others. Molecular docking and dynamis are required for further investigation.
Antibacterial Activity against Foodborne Pathogens and Inhibitory Effect on Anti-Inflammatory Mediators' Production of Brazilin-Enriched Extract from Caesalpinia sappan Linn

Abstract:Caesalpinia sappan L. heartwood was collected from Mae Chaem District, Chiang Mai Province, Thailand. Crude extracts were prepared by Soxhlet’s extraction using 50, 60, and 70% of ethanol (EtOH) at 50, 60, and 70 °C, and the brazilin content was measured using reversed-phase high performance liquid chromatography (RP-HPLC). The antibacterial activity against foodborne pathogens and anti-inflammatory aspects were investigated. C. sappan, prepared from 70% EtOH at 70 °C (E70T70), significantly (p < 0.05) exhibited the highest amount of brazilin (7.90 ± 0.50% w/w). All extracts were investigated for anti-inflammatory activity through an inhibition effect on nitric oxide (NO) and inducible nitric oxide synthase (iNOS) production in RAW264.7 mouse macrophage cells. The inhibitory effect on cyclooxygenase-2 (COX-2) production in HT-29 and HCT116 was also studied. All the extracts inhibited NO, iNOS, and COX-2 production induced by combined lipopolysaccharide and interferon-γ, especially E70T70, indicating the highest inhibition effect among other extracts. Additionally, E70T70 was selected to determine the antibacterial activity against foodborne pathogens, including Staphylococcus aureus, Escherichia coli, Salmonella enteritidis, and Vibrio parahaemolyticus. The result showed that 200 µg/mL extract reduced all test pathogens 100% at 24 h. These results suggested the potential of using C. sappan L. extract as a natural preservative in food and a natural active pharmaceutical ingredient.

Abstract:Objective To study the chemical constituents from the heartwood of Caesalpinia sappan.Methods The chemical constituents were isolated and purified by column chromatography on silica gel,Sephadex LH-20,and semi-preparative HPLC.The structures of compounds were identified by physicochemical properties,spectral analysis,and co-TLC with reference substances.Results Eleven compounds were isolated from the petroleum ether fraction of 95% ethanol extract and were identified as 1-hydroxy-7-methoxy xanthone(1),2-hydroxyxanthone(2),1,5-dihydroxyxanthone(3),1,7-dihydroxyxanthone(4),chrysophanol(5),friedelin(6),ursolic acid(7),β-sitosterol palmitate(8),ergosta-4,6,8(14),22(23)-tetraen-3-one(9),stigmasterol(10),and β-sitosterol(11).Conclusion Except compounds 10 and 11,others are isolated from this plant for the first time.Compounds 1—5 are firstly isolated from the genus Caesalpinia L.and are also compounds with xanthone and anthraquinone skeletons firstly isolated from the plants of Caesalpinia L.

Abstract:Antioxidant activities of the 95% ethanol extract from Caesalpinia sappan heartwood (ECS), protosappanin A, protosappanin B, and brazilein were studied in vitro. The inhibition of the formation of malondialdehyde (MDA) and the scavenging of superoxide anions, hydrogen peroxide, and hydroxyl radicals were assayed. The experimental results show that all four substances had antioxidant activity in vitro but their capabilities differed for the different indicators. ECS, protosappanin A, and protosappanin B show more inhibition of MDA and scavenging of hydrogen peroxide, while brazilein shows more scavenging of hydroxyl radicals. All the samples show little scavenging of superoxide anions.

Their structures were identified by spectroscopic analysis methods, such as 1D and 2D NMR, along with the high resolution mass spectral data. Abstract:Saraca asoca is a traditional medicinal plant whose all plant parts are exceptionally effective in terms of antimicrobial, anti-inflammatory, antioxidant, anti-carcinogenic, free radical scavenging,anti-arthritic, and hypolipidemic properties. As cardio-vascular problems occur for many reasons, antioxidants with free radical scavenging properties of plants and herbs are highly effective in treating cardio-related disorders. Though Saraca asoca has been preferred as a tonic and medicinal supplement for women's health, because of the huge variety of bioactive compounds, Saraca asoca needs to be explored for its cardio-protective properties. This review aims to summarize the in vivo and in vitro studies done on Saraca asoca along with the exploration of bioactive compounds in various parts of the plant which will display its cardio-protective potential with its rich bioactive compounds as ACE inhibitors. All relevant information on Saraca asoca in treating and preventing cardio-related disorders has been collected from electronic databases including PubMed, Google Scholar, Web of Science, and Science Direct. Various parts of Saraca asoca were studied to assess its pharmacological and cardioprotective properties. The bioactive compounds of Saraca asoca have been assessed to explore its role as anti-hypertensive, antioxidant, ACE inhibitors, and cardio-protective with the help of in-vivo, in-vitro studies and other research studies. This thorough review focuses on the potent natural bioactive compounds in various parts of Saraca asoca exhibiting its potential as a cardioprotective agent while incorporating historical, chemical, and therapeutic views.

Abstract:Medicinal plants are natural sources of bioactive phytochemical constituents that can be utilized to treat a variety of ailments due to the marked physiological effects they have on the human body. India, with its diverse ethnic groups and unique biodiversity, has a centuries-old ethnobotanical history for health promotion and treatment of illness. Among the medicinal herbs, the plant Cardiospermum halicacabum belonging to the family Sapindaceae is a climber with inflated fruits and seeds with a white heart-shaped pattern. Alcohols, phenols, alkynes, flavonoids, alkanes, and aliphatic esters are the major phytochemical components present in this plant. The phytochemical examination revealed that the herb contains terpenoids, flavonoids, tannins, proteins, saponin, glycosides, carbohydrates, volatile esters, and fatty acids. Cardiospermum halicacabum has been demonstrated to exhibit anti-oxidant, anti-viral, anti-ulcer, anti-diabetic, anti-convulsant, antipyretic, anxiolytic, anti-cancer, anti-bacterial, anti-arthritic, anti-fungal, anti-parasitic and fertility activities. The decoction of this plant is used in the treatment of dysentery, diarrhea, cold, asthma and pertussis. Various traditional uses of the plant include its usage as a refrigerant, emetic, stomachic, diuretic, laxative, wound healing and in treating earache. This review summarizes the wide range of phytochemical constituents, pharmacological activities along with microscopical, macroscopical characteristics and traditional uses of Cardiospermum halicacabum.

Abstract:Background: Iron overload and inflammation are severe conditions that can lead to various chronic diseases. However, the current iron chelator drugs have their limitations. The phytochemical compounds from herbals, such as brazilin, the major active compound in Caesalpinia sappan Linn., have significant therapeutic potential in various chronic diseases. Our study was designed to examine the effect of brazilin on iron chelating properties, antioxidant activity in hepatocytes, and anti-inflammatory potential in macrophages. Methods: This study focused on the isolation, purification, and evaluation of brazilin, the principal bioactive constituent found in C. sappan wood. Brazilin was extracted via methanol maceration followed by column chromatography purification. The purified compound was characterized using high-performance liquid chromatography (HPLC), nuclear magnetic resonance (NMR) spectroscopy, and mass spectrometry (MS). The antioxidant potential of brazilin was assessed by in vitro assays, including 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azinobis-(3-ethylbenzthiazolin-6-sulfonic acid (ABTS), and ferric-reducing antioxidant power (FRAP). Furthermore, its cellular antioxidant activity was evaluated using hydrogen peroxide-induced oxidative stress in the hepatocellular carcinoma cell line (Huh-7). The iron-chelating capacity of brazilin was determined spectrophotometrically, and Job's plot method was used to elucidated the stoichiometry of the iron-brazilin complex formation. The anti-inflammatory properties of brazilin were investigated in lipopolysaccharide (LPS)-stimulated macrophages (RAW 264.7). Nitric oxide (NO) inhibition was quantified using the Griess reagent, while the expression levels of pro-inflammatory cytokines, interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α), were evaluated by RT-qPCR. Results: The results demonstrated that brazilin exhibited potent antioxidant activity in vitro and hepatocytes in a concentration-dependent manner. It also showed anti-inflammatory activity, in which NO production was significantly reduced and IL-6 and TNF-α expression in LPS-induced macrophages were repressed. Furthermore, it can bind ferric and ferrous ions. Brazilin acts as a bidentate iron chelator that forms a complex with iron in a 2:1 ratio, and two water molecules are used as additional chelators in this complex. Conclusions: Our findings have significant implications. Brazilin can potentially alleviate the harmful effects of iron-induced oxidative stress and inflammatory disorders.

Abstract:Background Semecarpus anacardium (SCA) fruits are found in India's sub-Himalayan, tropical, and central regions and have been utilized for centuries in traditional Indian medicine to treat various ailments. In recent times, a growing body of research has emerged indicating that the extracts and active components found in SCA fruits possess qualities that can potentially inhibit the development of cancer and inflammatory markers. Purpose This study aims to provide a comprehensive review of the existing literature on the pharmacological mechanisms underlying the effects of extracts and phytochemicals of SCA fruits in cellular, animal models, and clinical trials of cancer and inflammatory diseases. Methods A comprehensive literature search was conducted utilizing several databases, including PubMed, Scopus, Google Scholar, preprint platforms, and the Cochrane Database of Systematic Reviews using the keywords " Semecarpus anacardium ", "Anti-inflammatory," and "cancer". The collection of articles started with establishing the database and continued until April 2024. Results Out of 1130 retrieved database records, 316 pertained to systematic reviews. The remaining 814 records focused on examining the anticancer and anti-inflammatory properties of SCA fruits. In the course of these investigations, the four primary cancer types linked to SCA fruits are identified as lung cancer, hepatocellular carcinoma, breast cancer, and blood cancer. Conclusion The findings will provide more support for investigating SCA fruits in cancer treatment and will furnish thorough reference data and recommendations for future studies on this botanical medication.

Brazilin: Biological activities and therapeutic potential in chronic degenerative diseases and cancer

Abstract:Caesalpinia sappan and Haematoxylum brasiletto belong to the Fabaceae family, predominantly distributed in Southeast Asia and America. The isoflavonoid brazilin has been identified from the bark and heartwood of these plants. This review summarizes the studies describing the biological activities of these plants and brazilin. Mainly, brazilin protects cells from oxidative stress, shows anti-inflammatory and antibacterial properties, and hypoglycemic effect. In addition, it has a biological impact on various pathologies such as Alzheimer's disease, Parkinson's disease, fibrillogenesis, and osteoarthritis. Interestingly, most of the antecedents are related to the anticancer effect of brazilin. In several cancers such as osteosarcoma, neuroblastoma, multiple myeloma, glioblastoma, bladder, melanoma, breast, tongue, colon, cervical, head, and neck squamous cell carcinoma, brazilin induces autophagy by increasing the levels of the LC3-II protein. Furthermore, it inhibits cell proliferation and induces apoptosis through increased expression of Bcl-2, Bcl-XL, p21, p27, activation of caspase-3 and -7, and the cleavage of PARP and inhibiting the expression of Bax. In addition, it blocks the expression of JNK and regulates the nuclear translocation of Nrf2. Together, these data positions brazilin as a compound of natural origin with multiple bioactivities and therapeutic potential in various chronic degenerative diseases and cancer.

Abstract:Methanol and formate oxidation supported the assimilation of [14C]acetate by cell suspensions of Methylococcus capsulatus; oxidation of other primary alcohols, except ethanol, did not. The extent of [1-14C]acetate assimilation supported by methanol oxidation was decreased in the presence of primary alcohols, except ethanol. Potassium cyanide (0.33 mM) completely inhibited the oxidation of formate and its stimulation of [1-14C]acetate assimilation. The amount of [1-14C]acetate assimilation supported by methanol oxidation was significantly inhibited by cyanide.

Abstract:Sarcandra glabra (Thunb.) Nakai is a perennial evergreen herb categorised within the Sarcandra Gardner genus under the Chloranthaceae family. Indigenous to tropical and subtropical regions of East Asia and India, this species is extensively distributed across China, particularly in the southern regions (Sichuan, Yunnan, and Jiangxi). In addition to its high ornamental value, S. glabra has a rich history of use in traditional Chinese medicine, evident through its empirical prescriptions for various ailments like pneumonia, dysentery, fractures, bruises, numbness, amenorrhea, rheumatism, and other diseases. Besides, modern pharmacological studies have revealed various biological activities, such as antitumour, anti-bacterial, anti-viral anti-inflammatory and immunomodulatory effects. The diverse chemical constituents of S. glabra have fascinated natural product researchers since the 1900s. To date, over 400 compounds including terpenoids, coumarins, lignans, flavonoids, sterols, anthraquinones, organic acids, and organic esters have been isolated and characterised, some featuring unprecedented structures. This review comprehensively examines the current understanding of S. glabra's phytochemistry and pharmacology, with emphasis on the chemistry and biosynthesis of its unique chemotaxonomic marker, the lindenane-type sesquiterpenoids.
The Ethanolic Extract of Caesalpinia Sappan Heartwood Inhibits Cerebral Ischemia/Reperfusion Injury in a Rat Model Through a Multi-Targeted Pharmacological Mechanism

Abstract:Background:Caesalpinia sappan L. (C. sappan) is a traditional Chinese medicinal plant. The dried heartwood of C. sappan (also known as Sappan wood) has been widely used for the folkloric medical treatment of ischemic cerebral stroke in China. However, the detailed underlying pharmacological mechanism still remains largely unexplored.Methods: In this study, a middle cerebral artery occlusion (MCAO) rat model was employed to elucidate the mechanism of the anti-cerebral ischemic effects of C. sappan ethanolic extract (CEE). Moreover, systemic multi-target identification coupled with gene ontology biological process (GO BP) and reactome pathway analysis was used to investigate the potential neuroprotective mechanism. Furthermore, the presumed mechanism was confirmed through biological analysis by determining the effects of CEE on the identified signaling pathways in PC12 cells model-induced by oxygen-glucose deprivation/reperfusion (OGD/R).Results: Our study demonstrates that CEE (both through in vivo administration at a dosage of 300 mg/kg and through in vitro incubation at a dosage of 2.4 μg/mL) is a neuroprotective agent that can effectively inhibit neuronal damage, promote synaptic generation, and suppress the activation of neutrophils, microglia, and astrocytes. Moreover, the neuroprotective mechanism of CEE is mediated via regulating 150 potential target proteins, which are associated with 6 biological processes and 10 pathways, including JAK-STAT, HSP90 and DNA damage/telomere stress.Conclusion: CEE can exert neuroprotective effect through multi-target pharmacological mechanisms to prevent ischemia/reperfusion-induced cerebral injury.
