Abstract:Heavy metal contamination poses a significant threat to food security, requiring the development of plant protection measures. Silicon (Si) deposition on rice roots can inhibit the uptake and translocation of heavy metal ions. However, the slow process of Si polymerization limits its utilization by many plants. This study explored the use of cation-induced silicic acid polymerization to rapidly form extracellular Si nanocoatings on plant roots using ten cationic polymeric materials. Performance, safety, and cost-effectiveness were comprehensively evaluated in enhancing rice's resistance to heavy metals, particularly trivalent chromium. Guar hydroxypropyl trimonium chloride (GHPT) showed the most promising results among the materials studied. A treatment procedure involving immersion in GHPT and silicic acid was found to be effective when repeated twice for 20 min each time. This method effectively doubled Si concentration in rice roots and reduced shoot chromium concentration by 67%. Furthermore, this approach was successfully applied to pakchoi, enabling Si utilization in this non-Si-accumulating plant. The Si concentration in pakchoi roots increased to levels similar to rice (1577.5 mu g/g), and shoot chromium concentration decreased by 75.0%, accompanied by a 66.2% reduction in chromium translocation factor. These findings provide a safe, economical and practical solution to reduce heavy metal stress.

Abstract:In the course of past two decade anthropogenic activities have reinforced, begetting soil and water defilement. A plethora of heavy metals alters and limits plant growth and yield, with opposing effect on agricultural productivity. Silicon often perceived as plant alimentary 'nonentity'. A suite of determinants associated with silicon have been lately discerned, concerning plant physiology, chemistry, gene regulation/expression and interaction with different organisms. Exogenous supplementation of silicon renders resistance against heavy-metal stress. Predominantly, plants having significant amount of silicon in root and shoot thus are barely prone to pest onset and manifest greater endurance against abiotic stresses including heavy-metal toxicity. Silicon-mediated stress management involves abatement of metal ions within soil, co-precipitation of metal ions, gene modulation associated with metal transport, chelation, activation of antioxidants (enzymatic and non-enzymatic), metal ion compartmentation and structural metamorphosis in plants. Silicon supplementation also stimulates expression of stress-resistant genes under heavy-metal toxicity to provide plant tolerance under stress conditions. Ergo, to boost metal tolerance within crops, immanent genetic potential for silicon assimilation should be enhanced. Current study, addresses the potential role and mechanistic interpretation of silicon induced mitigation of heavy-metal stress in plants.

Metagenomic and biochemical analyses reveal the potential of silicon to alleviate arsenic toxicity in rice ( Oryza sativa L.)

Abstract:Arsenic (As) pollution in agricultural systems poses a serious threat to crop productivity and food safety. Silicon (Si) has been reported to mitigate toxic effects of heavy metals in plants. However, the mechanisms behind Si-mediated alleviation of As toxicity in rice ( Oryza sativa L.) remain poorly understood. Here, we performed metagenomic and biochemical analyses to investigate the potential of Si in alleviating As toxicity to rice plants. As exposure reduced plant growth, chlorophyll contents, antioxidant enzyme levels and soil enzymes activity, while increasing reactive oxygen species (ROS) activity and inducing alterations in the rhizosphere microbiome of rice seedlings. Silicon amendments enhanced rice growth (24%), chlorophyll a (25%), and chlorophyll b (26.7%), indicating enhanced photosynthetic capacity. Si amendments also led to the upregulation of antioxidant enzymes viz., superoxide dismutase (15.4%), and peroxidase (15.6%), resulting in reduced ROS activity and oxidative stress compared to the As-treated control. Furthermore, Si treatment reduced uptake and translocation of As in rice plants, as evidenced by the analysis of elemental contents. Microscopic examination of leaf and root ultrastructure showed that Si mitigated As-induced cellular damage and maintained normal morphology. Metagenomic analysis of the rice rhizosphere microbiome revealed that Si application modulated composition and diversity of microbial communities e.g., Proteobacteria, Actinobacteria, and Firmicutes. Additionally, Si amendments upregulated the relative expression levels of OsGSH, OsPCs, OsNIP1;1 and OsNIP3;3 genes, while the expression levels of the OsLis1 and OsLis2 genes were significantly downregulated compared with As-treated rice plants. Overall, these findings contribute to our understanding of Si-mediated plant resilience to As stress and offer potential strategies for sustainable agriculture in As-contaminated regions.

Abstract:Over the past few decades, heavy metal contamination in soil and water has increased due to anthropogenic activities. The higher exposure of crop plants to heavy metal stress reduces growth and yield, and affect the sustainability of agricultural production. In this regard, the use of silicon (Si) supplementation offers a promising prospect since numerous studies have reported the beneficial role of Si in mitigating stresses imposed by biotic as well as abiotic factors including heavy metal stress. The fundamental mechanisms involved in the Si-mediated heavy metal stress tolerance include reduction of metal ions in soil substrate, co-precipitation of toxic metals, metal-transport related gene regulation, chelation, stimulation of antioxidants, compartmentation of metal ions, and structural alterations in plants. Exogenous application of Si has been well documented to increase heavy metal tolerance in numerous plant species. The beneficial effects of Si are particularly evident in plants able to accumulate high levels of Si. Consequently, to enhance metal tolerance in plants, the inherent genetic potential for Si uptake should be improved. In the present review, we have discussed the potential role and mechanisms involved in the Si-mediated alleviation of metal toxicity as well as different approaches for enhancing Si-derived benefits in crop plants.

Abstract:As agroecology deteriorates, agricultural production is threatened by the combined stressors of exposure to environmental pollutants and pathogenic microbes. Proper agronomic practices for crop growth management and fertilization require understanding plant tolerance strategies. Both rice blast and heavy metals substantially impair rice crops, while silicon (Si) is an effective amendment to alleviate the combined stressors. Herein, this study was conducted to investigate the rice physiology and pathology perspective on the mechanism of Si alleviation against both lead (Pb) toxicity and Magnaporthe oryzae infection, utilizing pot experiments with inoculation of the virulent Magnaporthe oryzae strain. Exogenous Si reduced the phyto-availability and plant absorption of Pb, resulting in a 73.5% reduction in exchangeable Pb concentration in soil and a 40.23% reduction in rice plants. Furthermore, Si addition boosted the plant antioxidant system by increasing the activities of related enzymes, as the activities of catalase, superoxide dismutase, and polyphenol oxidase were significantly improved while the activity of peroxidase in rice panicles decreased. As a result, an improvement in dry matter quantity by 19.19% was observed compared to treatments without Si application, and the panicle blast severity (PBS) was reduced by 0.4-37.52%. Notwithstanding the interaction between the combined stressors, this study revealed that the speciation of Pb formation in the rhizosphere was the primary contributor to the alleviation of abiotic stresses, whereas the regulation of oxidative stress by enzymatic antioxidants played a dominant role in alleviating Magnaporthe oryzae colonization and impairments. The regulation process may reveal the mechanism of siliceous fertilizer functioning in the paddy system. Thereby the role of exogenous Si in anti-fungal, heavy metal toxicology, and plant physiology needs further study to fully elucidate the role of Si amendment, which is proposed to be considered from the perspective of soil chemistry and plant physiology.

Abstract:Trivalent chromium [Cr(III)] is a threat to the environment and crop production. Silicon (Si) has been shown to be effective in mitigating Cr(III) toxicity in rice. However, the mechanisms by which Si reduces Cr(III) uptake in rice are unclear. Herein, we hypothesized that the ability of Si to obstruct Cr(III) diffusion via apoplastic bypass is related to silicic acid polymerization, which may be affected by Cr(III) in rice roots. To test this hypothesis, we employed hydroponics experiments on rice (Oryza sativa L.) and utilized apoplastic bypass tracer techniques, as well as model simulations, to investigate 1) the effect of Si on Cr(III) toxicity and its obstruction capacity via apoplastic bypass, 2) the effect of Cr(III) on silicic acid polymerization, and 3) the relationship between the degree of silicic acid polymerization and its Cr(III) obstruction capacity. We found that Si reversed the damage caused by Cr(III) stress in rice. Si exerted an obstruction effect in the apoplast, significantly decreasing the share of Cr(III) uptake via the apoplastic bypass from 18% to 11%. Moreover, Cr(III) reduced silica particles' radii and increased Si concentration in roots. Modeling revealed that a 5-fold reduction in their radii decreased the diffusion of Cr(III) in apoplast by approximately 17%. We revealed that Cr(III) promoted silicic acid polymeri-zation, resulting in the formation of a higher number of Si particles with a smaller radius in roots, which in turn increased the ability of Si to obstruct Cr(III) diffusion. This negative feedback regulatory mechanism is novel and crucially important for maintaining homeostasis in rice, unveiling the unique role of Si under Cr(III) ion stress and providing a theoretical basis for promoting the use of Si fertilizer in the field.
Silicon Enhances Abundances of Reducing Microbes in Rhizoplane and Decreases Arsenite Uptake by Rice (oryza Sativa L.).

Abstract:Although silicon (Si) transporters-mediated uptake of arsenic (As) by rice roots is well-documented, how Si influences As behaviors in rhizosphere and rhizoplane before As entry into roots is still unclear. Here we used three rice genotypes to explore the effect of silicic acid on the root uptake of As as impacted by chemical and microbial changes in bulk soil, rhizosphere, rhizoplane and endosphere. The results show that exogenous Si decreased root arsenite [As(III)] absorption, which was attributed to Si-mediated alteration of traits in chemical plaque and microbial films on the rhizoplane. The pH, Eh, As and Fe in the porewater were not influenced by Si. However, Si enhanced the concentrations of As(III) (16-49%) and Fe (15-80%) in the rhizoplane while decreasing As(III) concentrations in the roots (19-39%) and grains (22-29%). The diversities and richness of microbes in soils and plants were not affected by Si. The microbial connections were negatively influenced by Si in bulk and rhizosphere soils, but positively impacted in rhizoplane and endosphere. Both the abundance of reducing microbes, Anaeromyxobacter and Geobacteraceae, and the level of As(III) and Fe in rhizoplane were significantly increased by the addition of Si, thereby restraining As(III) from uptake into roots. This study provides new insights into the microbial mechanisms of Si-mediated As uptake by rice.

Abstract:Arsenic, a toxic element has been infiltrating the food chain, raising the food safety and security issues. Due to phytotoxicity, mobility and availability of specific transporters for arsenic translocation, it is considered as serious agronomic threat to rice crop. To limit the detrimental effect of this arsenic stress, soil amendment with beneficial substances such as silicon can be helpful in acquiring arsenic tolerance by activating defense mechanism and subsequently improve seedlings vigor. However, its exact mechanism through which it enhances arsenic tolerance is not fully understood. The present study aims to evaluate the potential of exogenous application of silicon to alleviate toxic effects of arsenic stress and the associated morphological, biochemical and physiological mechanism in rice seedlings. In this study, rice seedlings of non-aromatic variety (IR-6) and aromatic variety (Super basmati) were grown in hydroponic system for 28 days. For arsenic stress, 25 μM and 50 μM sodium arsenite were applied along with 1 mM silicic acid (Treatment groups: As0, As25, As50, As25 + Si and As50 + Si). Results indicated that silicon application significantly decreased oxidative stress markers including hydrogen peroxide levels by 34.78%, lipid peroxidation by 20.06% consequently, decrease in relative membrane permeability by 37.79% at 25 μM arsenic stress in Super basmati as compared to IR-6. Furthermore, silicon application under 25 μM arsenic stress also activated enzymatic antioxidant defense system by elevating the activity of superoxide dismutase by 68.89%, guaiacol peroxidase by 53.59% and ascorbate peroxidase by 66.77% in Super basmati except catalase. Additionally, SEM images showed the anatomical adaptation as cuticle thickenings along with deposition of silica in silicon-treated plants. Silicon deposition was also evident by EDX spectra while arsenic was not detected in silicon supplemented rice seedlings under arsenic stress. Hence, these findings suggested that silicon application hinders arsenic translocation and arsenic toxicity. This study would serve as a step towards developing the approach for the rice grains with reduces arsenic content. This mitigation strategy will be helpful in environmental sustainability along with ensuring food security.

Abstract:The soil bioavailability of phosphorus (P) is often low because of its poor solubility, strong sorption and slow diffusion in most soils; however, stress due to excess soil P can occur in greenhouse production systems subjected to high levels of P fertilizer. Silicon (Si) is a beneficial element that can alleviate multiple biotic and abiotic stresses. Although numerous studies have investigated the effects of Si on P nutrition, a comprehensive review has not been published. Accordingly, here we review: (1) the Si uptake, transport and accumulation in various plant species; (2) the roles of phosphate transporters in P acquisition, mobilization, re-utilization and homeostasis; (3) the beneficial role of Si in improving P nutrition under P deficiency; and (4) the regulatory function of Si in decreasing P uptake under excess P. The results of the reviewed studies suggest the important role of Si in mediating P imbalance in plants. We also present a schematic model to explain underlying mechanisms responsible for the beneficial impact of Si on plant adaption to P-imbalance stress. Finally, we highlight the importance of future investigations aimed at revealing the role of Si in regulating P imbalance in plants, both at deeper molecular and broader field levels.
Silicon mediated redox homeostasis in the root-apex transition zone of rice plays a key role in aluminum tolerance

Abstract:The supply of silicon (Si) mitigates the aluminum (Al) toxicity on plant root growth, while the underlying mechanism remains unknown. Transition zone (TZ) emerges as the Al-toxicity target of plant root apex. The objective of the study was to evaluate the effect of Si on redox homeostasis in root-apex TZ of rice seedlings under Al stress. Si alleviated Al toxicity as revealed by promotion of root elongation and less Al accumulation. In Si-deprived plants, treatment with Al altered the normal distribution of superoxide anion (O2·-) and hydrogen peroxide (H2O2) in root tip. Al induced a significant increase of reactive oxygen species (ROS) in root-apex TZ, resulting in the peroxidation of membrane lipid and loss of plasma membrane integrity in root-apex TZ. However, Si greatly increased the activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) and enzymes involved in ascorbate-glutathione (AsA-GSH) cycle in root-apex TZ under Al stress, and enhanced AsA and GSH contents, which reduced ROS and callose contents, thereby reducing malondialdehyde (MDA) content and Evans blue uptake. These results allow to precise the changes of ROS in root-apex TZ after exposure to Al, and the positive role of Si in maintaining redox balance in root-apex TZ.

Abstract:Silicon (Si) is an essential nutrient for rice, but its effects on the yield and quality of rice under heavy metal stress remain uncertain. In this study, two typical paddy soils (acidic and calcareous purple soils) in the western region of Chongqing were selected for field plot experiment, with the purpose of understanding the effects of Si implementation methods on grain yields and cadmium (Cd) uptake, transport, and accumulation in the grain of a hybrid rice (Oryza sativa L, Changliangyou 772). Four treatments were set for the purposes including soil-based Si application, foliar spray of Si alone, foliar spray of selenium (Se)–containing Si fertilizer, and a control without Si application, respectively. The results indicated that the Si applications reduced Cd contents in brown rice by 11.45~51.85% in the slightly Cd-contaminated acidic purple soil (pH = 4.77, soil total Cd content 0.413 mg kg−1) and 26.93~43.77% in the purple calcareous paddy soil (pH = 7.77) with similar Cd-polluting levels. It is worth noting that the Cd content of conventional fertilized rice exceeds the Chinese National Food Safety Standard limit (0.2 mg kg−1, GB2762-2017) in the slightly Cd-contaminated acidic purple soil, and foliar spray treatments showed most effective effects that meets the safety threshold standard. Soil-based Si application reduced Cd accumulation in rice grains mainly by inhibiting the translocation of Cd from stem to the rice grain or root to stem, while foliar sprays of Si mainly by inhibiting the translocation of Cd from stem to brown rice. Si applications increased the rice yield by 17.15 to 25.45% in calcareous paddy soil with foliar spray being the best, while no significant yield increase was found in acidic paddy soil. Si and Se-containing Si fertilizer improved the nutritional quality of rice grain as indicated by the increases of Se, Si, and protein contents and the significant decreases of Cd contents in the rice grains. The comprehensive effects in improving the rice quality follow the order of foliar spray of Se-containing Si fertilizer > foliar spray of Si alone > soil-based Si application. Thus, foliar spray Si-containing fertilizer could be helpful in increasing rice yield while reducing the Cd uptake in rice grains, which might be a feasible approach in controlling Cd entry into the human body via crops.

Abstract:Exogenous Si mitigates the mobility and bioavailability of Cd in the soil, thereby alleviating its phytotoxicity. This study focused on specific Si-induced immobilisation effects within the rhizosphere (S1), near-rhizosphere (S2), and far-rhizosphere (S3) zones. Based on the rhizobox experiment, we found that applying Si significantly elevated soil pH, and the variation amplitudes in the S3 soil exceeded those in the S1 and S2 soils. Si-induced changes in the rhizosphere also included enhanced dissolved organic carbon and diminished soil Eh, particularly in the Si400 treatment. Meanwhile, the introduction of Si greatly enhanced the Fe 2+ and Mn 2+ concentrations in the S1 soil, but reduced them in the S2 soil. The rhizosphere effect of Si which enriched Fe 2+ and Mn 2+ subsequently promoted the formation of Fe and Mn oxides/hydro-oxides near the rice roots. Consequently, the addition of Si significantly reduced the available Cd concentrations in S1, surpassing the reductions in S2 and S3. Moreover, Si-treated rice exhibited increased Fe plaque generation and fixation on soil Cd, resulting in decreased Cd concentrations in rice tissues, accompanied by reduced Cd translocation from roots to shoots and shoots to grains. Structural equation modelling further highlighted that Si is essential in Cd availability in S1 and Fe plaque development, ultimately mitigating Cd accumulation in rice. Si-treated rice also exhibited higher biomass and grain yield than those of control groups. These findings provide valuable insights into Si-based strategies for addressing the Cd contamination of agricultural soils.

Abstract:Soil contamination by heavy metals has become a serious threat to global food security. The application of silicon (Si)-based materials is a simple and economical method for producing safe crops in contaminated soil. However, the impact of silicon on the heavy-metal concentration in plant roots, which are the first line in the chain of heavy-metal entering plants and causing stress and the main site of heavy-metal deposition in plants, remains puzzling. We proposed a process-based model (adsorption-diffusion model) to explain the results of a collection of 28 experiments on alleviating toxic metal stress in plants by Si. Then we evaluated the applicability of the model in Si-mitigated trivalent chromium (Cr[III]) stress in rice, taking into account variations in experimental conditions such as Cr(III) concentration, stress duration, and Si concentration. It was found that the adsorption-diffusion model fitted the experimental data well (R 2 > 0.9). We also verified the binding interaction between Si and Cr in the cell wall using SEM-EDS and XPS. In addition, we designed a simplified biomimetic device that simulated the Si in cell wall to analyze the dual-action switch of Si from increasing Cr(III) adsorption to blocking Cr(III) diffusion. We found that the adsorption of Cr(III) by Si decreased from 58% to 7% as the total amount of Cr(III) increased, and finally the diffusion blocking effect of Si dominated. This study deepens our understanding of the role of Si in mitigating toxic metal stress in plants and is instructive for the research and use of Si-based materials to improve food security.

Abstract:The interaction of silicon and soil microorganisms stimulates crop enhancement to ensure sustainable agriculture. Silicon may potentially increase nutrient availability in rhizosphere with improved plants' growth, development as it does not produce phytotoxicity. The rhizospheric microbiome accommodates a variety of microbial species that live in a small area of soil directly associated with the hidden half plants' system. Plant growth-promoting rhizobacteria (PGPR) play a major role in plant development in response to adverse climatic conditions. PGPRs may enhance the growth, quality, productivity in variety of crops, and mitigate abiotic stresses by reprogramming stress-induced physiological variations in plants via different mechanisms, such as synthesis of indole-3-acetic acid, 1-aminocyclopropane-1-carboxylate deaminase, exopolysaccharides, volatile organic compounds, atmospheric nitrogen fixation, and phosphate solubilization. Our article eye upon interactions of silicon and plant microbes which seems to be an opportunity for sustainable agriculture for series of crops and cropping systems in years to come, essential to safeguard the food security for masses.

Abstract:Abiotic and biotic stresses are the major factors limiting plant growth worldwide. Plants exposed to abiotic and biotic stresses often cause reduction in plant biomass as well as crop yield, resulting in plant biomass carbon loss. As a beneficial and quasi-essential element, silicon accumulation in rhizosphere and plants can alleviate the unfavorable effects of the major forms of abiotic and biotic stress through several resistance mechanisms and thus increases plant biomass accumulation and crop yield. The beneficial effects of silicon on plant growth and crop yield have been widely reviewed over the last years. However, carbon accumulation of silicon-associated plant biomass under abiotic and biotic stresses has not yet been systematically addressed. This review article focuses on both the main mechanisms of silicon-mediated alleviation of abiotic and biotic stresses and their effects on plant biomass carbon accumulation in terrestrial ecosystems. The major points are the following: (1) the recovery of plant biomass via silicon mediation usually exhibits a bell-shaped response curve to abiotic stress severity and an S-shaped response curve to biotic stress severity; (2) although carbon concentration of plant biomass decreases with silicon accumulation, more than 96% of the recovered plant biomass contributes to plant biomass carbon accumulation; (3) silicon-mediated recovery generally increases plant biomass carbon by 35% and crop yield by 24%. In conclusion, silicon can improve plant growth and enhance plant biomass carbon accumulation under abiotic and biotic stresses in terrestrial ecosystems.

Abstract:Silicon (Si) has been well-known to enhance plant resistance to heavy-metal stress. However, the mechanisms by which silicon mitigates heavy-metal stress in plants are not clear. In particular, information regarding the role of Si in mediating resistance to heavy-metal stress at a single cell level is still lacking. Here, we developed a hierarchical system comprising the plant, protoplast, and suspension cell subsystems to investigate the mechanisms by which silicon helps to alleviate the toxic effects of trivalent chromium [Cr(III)] in rice. Our results showed that in whole-plant subsystem silicon reduced shoot Cr(III) concentration, effectively alleviating Cr(III) stress in seedlings and causing changes in antioxidant enzyme activities similar to those observed at lower Cr(III) concentrations without silicon added. However, in protoplast subsystem lacking the cell wall, no silicon deposition occurred, leading to insignificant changes in cell survival or antioxidation processes under Cr(III) stress. Conversely, in suspension cell subsystem, silicon supplementation substantially improved cell survival and changes in antioxidant enzyme activities under Cr(III) stress. This is due to the fact that >95% of silicon was on the cell wall, reducing Cr(III) concentration in cells by 7.7%–10.4%. Collectively, the results suggested that the silicon deposited on the cell wall hindered Cr(III) bio-uptake, which consequently delayed Cr(III)-induced changes in antioxidant enzyme activities. This research emphasizes the significance of cell walls in Si-alleviated heavy-metal stress and deepens our understanding of silicon functioning in plants. Furthermore, the hierarchical system has great potential for application in studying the functioning of other elements in plant cell walls.

Abstract:Chromium (Cr) phytotoxicity affirmed the need of mitigation strategies to remediate polluted soils and restricts its accumulation in the food chains. Salicylic acid (SA) and silicon (Si) play pivotal roles in stimulating the plant performance and stress resilience. So far, their interactive effects against Cr-phytotoxicities are less known. Thus, we evaluated the beneficial roles of alone or/and combine applications of SA and Si in mitigating the toxic effects of Cr in the leaves and roots of rice (Oryza sativa) seedlings. Results indicated that SA (10 μM) and/or Si (5 μM) markedly retrieved the Cr (100 μM) induced toxicities by minimizing the Cr-accretion in both leaves and roots, enhancing the performance of light harvesting pigments (total chlorophylls and carotenoids), water retention and accumulation of osmolytes (water-soluble protein and total soluble sugars) and ultimately improved the growth and biomass. Additionally, SA and/or Si maintained the ionic balance by enhancing the nutrients transport, upregulated the ascorbate-glutathione (AsA-GSH) cycle enzymes, minimized the extra accumulation of reactive oxygen species (ROS) (H2O2 and O2•‒), malondialdehyde (MDA), recovered the membrane stability and damages in cellular ultrastructure in Cr-stressed rice plants. Overall findings suggested that SA underpins Si in mitigating the Cr-induced phytotoxicities on the above-reported parameters and combined applications of SA and Si were more effective than alone treatments. The uptake or cellular accumulation of Cr, osmoprotectants level and antioxidant defense system against oxidative stress can be considered as key toxicity biomarkers for the safe cultivation of rice in Cr-contaminated soils.

Abstract:The objective of this study is to elucidate the roles of silicon (Si) in enhancing tolerance to excess zinc (Zn) in two contrasting rice (Oryza sativa L.) cultivars: i.e. cv. TY-167 (Zn-resistant) and cv. FYY-326 (Zn-sensitive). Root morphology, antioxidant defense reactions and lipid peroxidation, and histochemical staining were examined in rice plants grown in the nutrient solutions with normal (0.15 μM) and high (2 mM) Zn supply, without or with 1.5 mM Si. Significant inhibitory effects of high Zn treatment on plant growth were observed. Total root length (TRL), total root surface area (TRSA) and total root tip amount (TRTA) of both cultivars were decreased significantly in plants treated with high Zn, whereas these root parameters were significantly increased when Zn-stressed plants were supplied with 1.5 mM Si. Supply of Si also significantly decreased Zn concentration in shoots of both cultivars, indicating lower root-to-shoot translocation of Zn. Moreover, superoxide dismutase (SOD), catalase (CAT), and asorbate peroxidase (APX) activities were increased, whereas malondialdehyde (MDA) and hydrogen peroxide (H2O2) concentrations were decreased in Si-supplied plants of both Zn-sensitive and Zn-resistant rice cultivars exposed to Zn stress. These alleviative effects of Si, further confirmed by the histochemical staining methods, were more prominent in the Zn-resistant cultivar than in the Zn-sensitive one. Taken together, all these results suggest that Si-mediated alleviation of Zn toxicity is mainly attributed to Si-mediated antioxidant defense capacity and membrane integrity. The possible role of Si in reduction of root-to-shoot translocation of Zn can also be considered.
