Abstract:Changes in the starch digestibility and the properties of the starches isolated from waxy, low-, and high-amylose rice kernels subjected to heat-moisture treatment (HMT) at 100 degrees C for 16 h at a moisture content of 25% were investigated. The results showed that amylose content is important in determining pasting properties for both HMTrice starches and in-kernel HMTrice starches. Amylose leaching, swelling power, and solubility index decreased in hydrothermal treatment of both isolated rice starches and rice kernels. The gelatinization temperatures of both hydrothermal-treated samples were affected by different treatments to different extents, and the changes were more pronounced for starches isolated from the treated rice kernels. The relative crystallinity increased in the starch from treated waxy and low-amylose rice kernels but decreased in the starch from treated high-amylose rice kernels. Starch digestibility study suggested that thermal treatment of rice kernels was more effective in producing low-glycemic-index starch-based foods than the same treatment of isolated starch. The study showed that HMT on rice kernels had significant impacts of physicochemical properties and starch digestibility of waxy, low-, and high-amylose rice starches to various extents.

Abstract:BACKGROUND Currently there is much interest in the application of physical modification techniques such as heat-moisture treatment (HMT). The effects of HMT on normal and waxy rice starches, subject to different levels of moisture content and duration of heating, were investigated. RESULTS Water solubility index (determined at 90 °C) decreased after HMT for normal and waxy rice starches, while swelling power (determined at 90 °C) showed inconsistent results (decrease for normal type, increase for waxy type) after HMT. Values in pasting parameters of normal and waxy rice starch increased initially, but the extent of increase slowed down with moisture content and length of treatment increasing. HMT decreased gelatinization temperatures with 4 h and 8 h treatment, but when length of treatment was prolonged to 16 h gelatinization temperature increased. Degree of crystallinity decreased for all treatments, and decreased much more at higher levels of moisture content. CONCLUSION Variations in levels of moisture content and duration of heating had significant effects on physicochemical and crystalline properties to different extents.

Abstract:Serum designated as IS obtained from a young healthy infertile woman induced a head-to-head agglutination of ejaculated boar sperm. The immunoglobulin G (IgG) prepared from IS localized to the acrosomal region of the sperm head obtained from the corpus and cauda epididymis as determined by an indirect immunofluorescent method. The IgG interacted with a boar sperm protein with an estimated molecular weight of 45-kDa, determined by sodium dodecyl sulfate-polyacrylamide gel electrophoretic (SDS-PAGE) immunoblotting technique. However, the IgG did not interact with proteins extracted from sperm obtained from the testis and caput epididymis or from non-gonadal tissues including liver, kidney, spleen, muscle and serum. The IgG interacted with additional proteins of about 75- and 38-kDa present in the corpus and cauda epididymal fluids but not those in the caput epididymal fluid. The staining intensity of the 75-kDa band was reduced and that of the 38-kDa was nullified with ejaculated seminal plasma proteins. The interacting proteins were adsorbed when chromatographed on Concanavalin A Sepharose column, suggesting that they are glycoproteins.

Abstract:Thermal treatment has been used to modify starch properties, but few studies pay attention to the change in highland barley starch (HBS) under the whole kernel treatment. The different effect of superheated steam (SS) on the HBS properties from other traditional thermal treatment is still unclear, not mention to the exactly structural change of HBS. This study explored the different effects of SS, steam and roasting treatments of HB kernel on the multi-scale structure, rheological and digestive properties of HBS. Thermal treatments led to the aggregation of HBS granules, the rough surface, and the absence of Maltese cross, as well as the increase in short-chain length ratio and amylose content. Compared with steam and roasting treatments, SS treatment increased the average molecular weight of HBS at 140 °C while reduced it at 180 °C due to starch interactions and depolymerization effect. Multi-scale structure analysis revealed that SS-180 showed the most significant disruption of HBS crystalline structure, the disorder of short-range structure, the decrease in double helix content and the disruption of lamellar structure. These structural changes of HBS resulted in the enhanced thermal stability, and decreased values of G′ and G″ representing the weaker gel structures. Meanwhile, much more resistant starch was converted from rapid digestible starch and slowly digestible starch in SS-treated HB than other thermal-treated HB. In particularly, the most change in multi-scale structure of HBS caused by SS-180 was responsible for its best thermal stability, weakest gel structures and slowest starch digestive rate.

Abstract:To investigate the characteristics of chemically modified wheat starch, such as oxidized (OWS), cross-linked (CLWS) and dual-modified wheat starches, cross-linked oxidized (COWS) and oxidized cross-linked (OCWS) wheat starches were obtained by 12% hydrogen peroxide (H2O2) and 2% sodium phytate treatments, respectively. After modifications of wheat starch by cross-linking and oxidization, it was determined that native wheat starch was effectively modified with respect to the structure and physicochemical characteristics, as detected by Fourier transform infrared spectroscopy (FT-IR), differential scanning calorimetry (DSC), Rapid Visco Analyzer (RVA), scanning electron microscopy (SEM) and X-ray diffraction (XRD). DSC analysis demonstrated that peak temperature (Tp) 64.41°C of the COWS was the highest. The RVA viscosity indexes of CLWS were noticeably increased compared with the other four starches, of which three oxidized starch samples showed lower RVA profiles because of oxidation. The XRD result indicated that cross-linked phosphates from sodium phytate were primarily located in the amorphous regions of starch granules. After modifications by different treatments, the starch samples exhibited different morphological characteristics, including A-type and B-type wheat starch granules, which showed much closer contact with each other by the cross-linking reaction under SEM observations. The functional characteristics, including solubility, swelling power, light transmittance and freeze-thaw stability (FTS), of all five treated and untreated wheat starches demonstrated that COWS had the highest solubility at 0.57 and that CLWS showed the best swelling power at 12.63 (g/g). The paste clarity of COWS was improved to 47.72% higher than that of native starch (7.54%), and the water loss of COWS (21.62%) was the lowest, which is beneficial to the production of quick-frozen food. Our results showed that dual-modified starch by using sodium phytate and hydrogen peroxide had significantly altered structural and functional properties. The present study provides fundamental information of dual-modified wheat starch for its potential industrial application.

Abstract:Microwave treatment is an environmentally friendly method for modification of high-amylose maize starch (HAMS). Here, the effects of short-time (≤120 s) microwave treatment on the structure and pasting of two types of HAMSs, Gelose 50 (HAMSI) and Gelose 80 (HAMSII), with apparent amylose content (AAC) of 45 % and 58 %, respectively, was studied using a multiscale approach including X-ray scattering, surface structures, particle size distribution, molecular size distributions and high temperature/pressure Rapid Visco Analysis (RVA)-4800 pasting. As compared to starch with no amylose (waxy maize starch, WMS) and 25 % amylose content (normal maize starch, NMS), HAMSI underwent similar structural and pasting changes as WMS and NMS upon microwave treatment, and it might primarily be attributed to the amylopectin fraction that was affected by cleavage of the connector chains between double helices and backbone chains, which decreased the crystallinity and thickness of the crystalline lamellae. However, the multi-scale structure of HAMSII was almost unaffected by this treatment. The pasting properties of fully gelatinized HAMSI starch showed a decrease in RVA-4800 peak and final viscosities after microwave treatment. In contrast, for HAMSII starch, the microwave treatment led to an increase in these viscosities. The combined results highlight the influence of varying AAC on the effects of microwave-mediated modification, leading to diverse alterations in the structure and functionality of starches.
Appropriate Microwave Improved the Texture Properties of Quinoa Due to Starch Gelatinization from the Destructed Cyptomere Structure

Abstract:Texture characteristics of quinoa under microwave (MW) irradiation were studied from the perspective of starch gelatinization. MW reduced the light transmittance and increased the hardness and stickiness of quinoa. Microstructure showed that MW dispersed the vesicular structure of starch aggregates into separate starch particles, resulting in the full hydration of starch and water molecules to form denser network structures. The value of peak viscosity and setback decreased in RVA after MW treatment, but the gelatinization temperature remained stable. DSC further proved that moderate MW treatment could reduce the gelatinization enthalpy of starch and made quinoa accessible to gelatinize. However, XRD showed that the crystal structure of starch was preserved, but the crystallinity increased. Finally, low field NMR showed that moderate MW stimulated the full hydration of starch to form denser network structures, while excessive MW treatment made starch partially gelatinize and form rigid structure, resulting in negative texture properties.

Abstract:To investigate the effect of heat-moisture treatment (HMT) on the physiochemical properties and in vitro di-gestibility of corn starch, the pasting behavior, viscoelasticity, thermal properties, long/short range structure, morphology and in vitro digestion of corn starch treated with different HMT conditions (HMT-20, 25, 30, 35 and 40 %) were characterized. Results indicated that after HMT, the pasting and disintegration behaviors of corn starch were affected and correlated with the moisture content. The dynamic viscoelasticity of corn starch was changed, and when glassy conditions were reached, the elastic properties decreased with increasing moisture while the viscous properties increased, especially for the HMT-40 %. The thermal stability of starch was improved by HMT, although the enthalpy of pasting (& UDelta;H) was reduced. Additionally, the HMT processing also promoted the conversion of RDS to SDS and/or RS (SDS and RS increased to 39.80 % and 31.68 % for HMT-40 %, respectively), which might attribute to the rearrangement of free starch molecules. The present work provides a potential approach to make functional starches with healthy properties.

Abstract:Non-thermal physical treatments has obvious advantages in regulating the structure and properties of starch compared with chemical treatment. Hance, this article summarized and compared the effects of three kinds of non-thermal physical treatments including grinding and ball milling, high hydrostatic pressure and ultrasonic on the structure, properties and chemical activity of starches from different plants. The potential applications of non-thermal physical modified starch were introduced. And strategies to solve the problems in the current research were put forward. It is found that although starch has a dense structure, the starch granules could be deformed under three kinds of non-thermal physical treatments, which could damage the granule morphology, microstructure, and crystal structure of starch, reduce particle size, increase solubility and swelling power, and promote starch gelatinization. Three kinds of non-thermal physical treated starch could be used as flocculant thickener, starch based edible films and fat substitutes. Non-thermal physical treatments caused the structure of starch to undergo three stages, which were similar to mechanochemical effects. When starch was in the stress stage and the transition stage from aggregation to agglomeration, its active sites significantly increase and move inward, ultimately leading to a significant increase in the chemical activity of starch.

Abstract:In this study, the differences in the modification effects and related mechanisms of different times (20 and 40 min) of autoclaved heat (AH) treatment and different doses (2 and 4 kGy) of electron beam irradiation (EBI) in different sequences of effects on acorn starch were investigated. The results showed that both AH and EBI reduced the amylose content (22.70 to 19.59%) and enthalpy (10.28 to 1.84%) of starch but increased the resistant starch content (53.69 to 64.11%). AH treatment made the crystalline regions of the residual starch granules denser, which was resistant to the action of amylase enzymes. EBI degraded the long chain of starch, which increased the solubility. Notably, EBI pretreatment improves the reactive sites by inducing depolymerization and disorder in starch internal structure, thus increasing the modification extent of AH-modified starch, forming starch with lower viscosity, better hydration, and digestibility resistance, therefore being used as an ingredient for functional foods.

Abstract:This study investigated the effect of liquid nitrogen ball-milled mechanochemical treatment on multiscale structure and physicochemical properties of starches with typically selected A (rice starch, ReS), B (potato starch, PtS) and C (pea starch, PeS) crystal types. The morphology of starch samples changed from integral granules to irregular fragments, and the interaction between the exposure OH bonds led to a serious agglomeration. As the treatment times extended, the crystalline structure of starch samples was gradually destroyed, and the excessive treatment approached amorphization. Moreover, the thermal stability of starch samples showed the downward tendency; and with amorphization increased, the swelling power (SP), solubility (S), water absorption capacity (WAC), oil absorption capacity (OAC) and hydrolysis rate of starch samples gradually increased. The obtained results provided a theoretical foundation for broadening the application range of ball-milled starches with different crystal types.
Impacts of Hydrothermal Treatments on the Morphology, Structural Characteristics, and In Vitro Digestibility of Water Caltrop Starch

Abstract:The influence of hydrothermal treatments on the structural properties and digestibility of water caltrop starch was investigated. Scanning electron microscopy (SEM) showed some small dents on the surface of starch granules for samples treated with heat moisture treatment (HMT), but not for samples treated with annealing (ANN) which generally showed smoother surfaces. The gelatinization temperature of starch was generally increased by hydrothermal treatments, accompanied by a trend of decreasing breakdown viscosity. These results implied the improvement of thermal and shearing stability, particularly for HMT in comparison to ANN. After being cooked, the native and ANN-modified water caltrop starch granules were essentially burst or destroyed. On the other hand, the margin of starch granules modified by HMT and dual hydrothermal treatments remained clear with some channels inside the starch granules. X-ray diffraction revealed that the crystalline pattern of water caltrop starch changed from the CA-type to the A-type and the relative crystallinity reduced with increasing moisture levels of HMT. Results of ANN-modified water caltrop starch were mostly similar to those of the native one. Moreover, water caltrop starch modified with HMT20 and dual modification contained a pronouncedly higher resistant starch content. These results suggested that HMT, ANN, and dual modification effectively modified the functional properties of water caltrop starch.

Abstract:To tailor the properties and enhance the applicability of starch, various ways of starch modification have been practiced. Among them, physical modification methods (micronization, nonthermal plasma, high-pressure, ultrasonication, pulsed electric field, and γ-irradiation) are highly potential for starch modification considering its safety, environmentally friendliness, and cost-effectiveness, without generating chemical wastes. Thus, this article provides an overview of the recent advances in nonthermal physical modification of starch and summarizes the resulting changes in the multi-level structures and physicochemical properties. While the effect of these techniques highly depends on starch type and treatment condition, they generally lead to the destruction of starch granules, the degradation of molecules, decreases in crystallinity, gelatinization temperatures, and viscosity, increases in solubility and swelling power, and an increase or decrease in digestibility, to different extents. The advantages and shortcomings of these techniques in starch processing are compared, and the knowledge gap in this area is commented on.

Abstract:Extrusion can modify the structure and physical properties of starch, while the extent of improved extrusion cooking technology (IECT) affects the starch with high moisture content and different crystal types remaining unclear. Therefore, the influence of IECT at different screw speeds on the structure, physical properties and in vitro digestibility of corn (A-type), potato (B-type) and pea (C-type) starches with high moisture content (42 %) was explored. Results indicated that IECT treatment caused similar variations on structure, physical properties, and in vitro digestibility of the 3 types of starches. The contents of slowly digestible starch (SDS) and resistant starch (RS) decreased by IECT treatment, accompanied by a reduction of crystallinity, enthalpy of gelatinization, gelatinization temperature and viscosity, while the content of rapidly digestible starch (RDS) and the ratio of bound water increased. And the changes in in vitro digestibility of starch were closely related to the damage to starch structure caused by IECT. Furthermore, most of starch granules were in the agglomeration stage by appropriate IECT treatment, which induced the exposure of a great quantity of enzyme binding sites to enhance the in vitro digestibility.

Abstract:The effects of microwave (MW) treatments for 30 and 90 s or cold plasma (CP) treatments for 2, 6, and 10 min on structural, physicochemical, and digestive properties of rice starch were investigated and compared with the effects of combined MW and CP treatments. Dual modification of starch using combined MW and CP treatments significantly changed starch granules' morphology, and starch showed lower crystallinity, swelling power, and peak viscosity than rice starch with a single modification. However, no significant changes were found in rice starch's crystalline structure and FT-IR spectra pattern after single or combined treatments. Effects of these treatments on the amylopectin branch chain length distribution and molecular weight of rice starch varied considerably depending on treatment type and treatment duration. Moreover, the dually modified rice starch contained more great content of resistant starch. Thus, the synergistic effects between MW and CP had more profound modification than the single treatment. Also, the dual treatment can be used to produce modified starch for industrial applications.
The Combined Effects of Extrusion and Recrystallization Treatments on the Structural and Physicochemical Properties and Digestibility of Corn and Potato Starch

Abstract:This study aimed to compare the effects of extrusion and recrystallization treatments on the structural and physicochemical properties and digestibility of corn (CS) and potato starch (PS). CS and PS were conditioned at 40 % and 60 % moisture content and extruded at 40 degrees C-120 degrees C. The extrudates were stored for 1, 7, and 14 days at 4 degrees C for recrystallization. The structural properties of extruded CS (ECS) and extruded PS (EPS), as well as the crystallization and thermal properties, water solubility, water absorption, and in vitro digestibility of recrystallized ECS (RECS) and recrystallized EPS (REPS), were evaluated. Compared with native starch, both ECS and EPS exhibited a significant decrease in their measured molecular weight. The chain length distribution did not change significantly, but the amylose content increased remarkably. Both RECS and REPS with 60 % moisture content presented a typical B-type pattern, and RECS exhibited V-type characteristic peaks. The melting signals of recrystallized amylopectin (46-64 degrees C) and amylose (136-158 degrees C) of RECS were detected. In vitro digestion experiments showed that RECS had lower digestibility than REPS. The highest slowly digestible starch and resistant starch contents of RECS were approximately 3.5 and 6.5 times that of unextruded CS, respectively.
Supramolecular structure and pasting/digestion behaviors of rice starches following concurrent microwave and heat moisture treatment

Abstract:From a supramolecular structure viewpoint, we concern how concurrent microwave and heat moisture treatment (M/HMT) tailors the pasting and digestion features of indica rice starch (IRS) and waxy rice starch (WRS), using combined analytical methods such as scanning electron microscopy and small-angle X-ray scattering. The results confirm that, for the starches (especially IRS), M/HMT led to a concave granule surface, weakened alignment of helices in lamellae, loosened packing of chains in amorphous regions, and somewhat disrupted crystallites. Consequently, the susceptibility of starch structures to hydrothermal effects was insignificantly altered, as reflected by unchanged pasting temperature; the enzyme absorption/hydrolysis could be properly accelerated, as affirmed by a modestly increased digestion rate and slightly reduced enzymatic resistance. Also, compared to WRS, IRS had more amylose with relatively long branches, which probably contributed to starch chain interactions during M/HMT. This tended to reduce the paste viscosity related to the swelling degree of starch granules, and increase the paste stabilities during heating and cooling related to the robustness of swelled granules and the reorganization of glucan chains.
Influence of enzymatic modification on the basis of improved extrusion cooking technology (IECT) on the structure and properties of corn starch

Abstract:Enzymatic modification can directly affect the structure and properties of starch, but generally causes high energy consumption in drying process. Improved extrusion cooking technology (IECT) itself is a starch modification technology. In this work, a co-extrusion method of starch with 42 % moisture and enzyme was adopted to reveal the effects of different enzyme dosages on the structure and properties of corn starch. After enzyme treatment on the basis of IECT, starch granules were broken into fragments without the occurrence of clear Maltese cross. R 1047/1022 and R 995/1022 values, peak intensity of Raman spectra and gelatinization temperature decreased, and the full width at half maximum at 480 cm −1 of Raman spectra raised. Moreover, the bound water proportion decreased from 87.44 % to 85.84 % ∼ 78.67 %, and the maximum light transmittance and dextrose equivalent values increased to 34.13 % and 26.14, respectively. The solubility of starch granules was all above 60 %. Findings supported that the mechanochemical effect of IECT on starch was conducive to the enzymatic modification.

Abstract:Resistant starch (RS) type 2-high-amylose corn starch (HACS) was subjected to simultaneous hydrothermal (25% moisture content, 90 °C for 12 h) and microwave (35% moisture content, 40 W/g microwaving for 4 min) treatment and zein (at a zein to treated starch ratio of 1:5, 50 °C for 1 h) to improve its resistance to enzymolysis. Scanning electron microscopy (SEM) highlighted the aggregation and adhesion of the composite. The average particle size of the composite (27.65 μm) was exceeded that of both the HACS (12.52 μm) and the hydrothermal and microwave treated HACS (hydro-micro-HACS) (12.68 μm). The X-ray diffraction results revealed that the hydro-micro-HACS and composite remained B-type, while their crystallinity significantly decreased to 16.98% and 12.11%, respectively. The viscosity of the hydro-micro-HACS and composite at 50 °C was 25.41% and 35.36% lower than that of HACS. The differential scanning calorimetry (DSC) results demonstrated that the composite displayed a new endothermic peak at 95.79 °C, while the weight loss rate and decomposition temperature were 7.61% and 2.39% lower than HACS, respectively. The RS content in HACS, the hydro-micro-HACS, and composite was 47.12%, 57.28%, and 62.74%, respectively. In conclusion, hydrothermal and microwave treatment combined with zein provide an efficient physical strategy to enhance the RS type 2-HACS.
