Abstract:CBT cell line developed from a warm-water fish Colossoma Brachypomum,was used to evaluate the effects of cold stress on fish cells. Cell viability was measured by Thiazolyl blue (MTT) method and cellular ultrastructure was examined under transmission electron microscope. Externalization of phosphatidylserine(PS) and mitochondrial membrane potential(MMP) were monitored with confocal laser scanning microscope. Results showed that cell viability and MMP decreased in a time-dependent manner, and the early events of apoptosis such as chromatin condensation, margination and PS externalization occurred, when the cells were exposed to 10 degrees C. It suggested that low temperature (10 degrees C) could induce apoptosis in Colossoma Brachypomum CBT cells.
Transcriptome reveals the important role of metabolic imbalances, immune disorders and apoptosis in the treatment of Procambarus clarkii at super high temperature

Abstract:Temperature is an important environmental factor in the living environment of crustaceans. Changes in temperature can affect their normal growth and metabolism and even cause bacterial disease. Currently, the potential anti-reverse molecular reaction mechanism of crustaceans during high-temperature conditions has not yet been fully understood. Therefore, in this study, we characterised the transcriptome of Procambarus clarkii using RNA sequencing and performed a comparison between super-high-temperature treated samples and controls. After assembly and annotation, 81,097 unigenes with an average length of 069 bp and 358 differentially expressed genes (DEGs) were identified. Among these DEGs, 264 were differentially upregulated and 94 were differentially downregulated. To obtain comprehensive gene function information, we queried seven databases, namely, Nr, Nt, Pfam, KOG, Swiss-Prot, KEGG, and GO to annotate gene functions. Transcriptome analysis revealed that the identified DEGs have significant effects on immune-related pathways, including lysosomal and phagosomal pathways, and that super-high-temperature conditions can cause disease in P. clarkii. Some significantly downregulated genes are involved in oxidative phosphorylation and the PPAR signalling pathway; this suggests a metabolic imbalance in P. clarkia during extreme temperature conditions. In addition, elevated temperature changed the expression patterns of key apoptosis genes XIAP, CASP2, CASP2, CASP8, and CYTC, thereby confirming that high-temperature conditions caused immune disorders, metabolic imbalance, and, finally, triggered apoptosis. Our results provide a useful foundation for understanding the molecular mechanisms underlying the responses of P. clarkii during high-temperature conditions.
Physiological and Histological Changes in Crayfish Procambarus Clarkii after Acute Temperature Stress

Abstract:Temperature stress affect a stress response of aquatic animals including physiological metabolism, immune defense and other metabolic processes, damaged their immune response even cause dead. In this study, the physiological and histological changes of Procambarus clarkii under the high temperature (30 degrees C) and the low temperature (10 degrees C) stress were investigated. With the increase of temperature, the pyruvate kinase activity in the hepatopancreas and gill tissues of crayfish was gradually increased, and the fatty acid synthase activity at 10 degrees C and 30 degrees C were significantly lower than the control group of 25 degrees C. Furthermore, lysozyme, catalase and superoxide dismutase in 10 degrees C and 30 degrees C were significantly lower than that of 25 degrees C. ELISA revealed that HSP90 activities was enhanced with the increasing temperature in hepatopancreas after 36h of treatment. HE sections showed that high temperature stress or hypothermia caused deformation of R cells (storage cells), rupture of B cells (secretory cells), and partial rupture of the basement membrane (BM) in hepatopancreas, as well as the deformation of epithelial cells and respiratory epithelial cells of BM in gill tissues. While the temperature stress (30 degrees C and 10 degrees C treatment) caused the deformation of epithelial cells and respiratory epithelial cells, making rupture of the BM, comparing with the group at 25 degrees C. The study provides the data about the response of crayfish to temperature stress at the molecular level, and benefit to develop the strategy to defend the damage in the crayfish aquaculture caused by the temperature neither in the different regions nor in the different season.
Effects of temperature and salinity on tissue structure, antioxidant capacity and heat shock protein gene expression of Penaeus monodon

Abstract:Abstract In order to explore the influence of temperature and salinity on stress response of P. monodon , four experimental groups were set, respectively were normal temperature and normal salt (Control group), normal temperature and low salt(Group A), high temperature and low salt(Group B), high temperature and normal salt(Group C). The stress experiment was conducted for 96 hours, and the activities of acid phosphatase(ACP), alkaline phosphatase(AKP), total superoxide dismutase (T-SOD) and total antioxidant capacity (T-AOC) in hepatopancreas, the changes of tissue structure and the expression of related heat shock protein genes were measured．The results showed that the branchial structures of the three stress groups were found to be damaged to varying degrees. In Group A, AKP and ACP levels increased initially and then decreased, while T-SOD levels decreased initially and then steadily increased. In Group B and Group C, the four enzymes showed similar trends.There was no significant difference in the expression level of Hsps in the low salt group. The expression levels of Hsp-10, Hsp-60 and Hsp-90 genes showed a similar trend, and the maximum expression levels of the four genes were all at 96h. The expression levels of Hsps in muscle tissue were similar to those in hepatopancreas tissue. However, the highest expression levels of Hsp-70 and Hsp-90 in high temperature and low salt group were at 24h. Through this work on P. monodon stress responses to high temperature and low salinity, we have gained a deeper understanding of the physiological responses to environmental stressors, which will be useful in the development of protective measures for the shrimps under aquaculture conditions that replicate these stressors.
Studies on Cold-Induced Apoptosis in Colossoma Brachypomum Cells

Abstract:To observe the causes of hypothermia(10 ℃) leading to cell death in Colossoma Brachypomum CBT and probe the potential mechanism involved.Cell viability was monitored by Cell Counting Kit-8.The production of reactive oxygen species(ROS) in cells was determined by H_2DCFDA.Apoptosis was analyzed by hypodiploid apex and Tunel.Agarose gel electrophoresis was used to detect fragmentation.Hoechst 33258 dying was applied to observe cold-induced morphological changes.The release of dehydrogenase(LDH) was determined by colorimetric assay.The results show that exposure of CBT cells to the low temperature(10 ℃) caused a substantial decrease in cell viability,an increase in production of ROS in cells and a concurrent release of LDH(P0.01).When CBT cells were treated with low temperature for 3days,flow cytometry revealed the hypodiploid apex and the apoptotic rate was shown to be 13%.Then if the cell temperature was brought-back to 32℃ which lasted for more than 12 h,a DNA ladder pattern representing fragmentation was observed.Tunel detection revealed that after 3 day,4 day and 5 day cold incubation(10 ℃) followed by 12 h rewarming(32 ℃),the apoptotic rates of CBT cells were 23.49%,27.72% and 35.10% respectively.After the CBT cells were incubated at the low temperature for 5 days followed by 12 h rewarming,typical apoptotic bodies began to appear.Therefore low temperature of 10 ℃ could induce early apoptosis in CBT cell and additional exposure to temperature of 32 ℃ could promote CBT cell to late apoptosis.ROS might play a key role in cold-induced apoptosis of CBT cell.
Characterization of Biological Pathways Regulating Acute Cold Resistance of Zebrafish

Abstract:Low temperature stress represents a major threat to the lives of both farmed and wild fish species. However, biological pathways determining the development of cold resistance in fish remain largely unknown. Zebrafish larvae at 96 hpf were exposed to lethal cold stress (10 °C) for different time periods to evaluate the adverse effects at organism, tissue and cell levels. Time series RNA sequencing (RNA-seq) experiments were performed to delineate the transcriptomic landscape of zebrafish larvae under cold stress and during the subsequent rewarming phase. The genes regulated by cold stress were characterized by progressively enhanced or decreased expression, whereas the genes associated with rewarming were characterized by rapid upregulation upon return to normal temperature (28 °C). Genes such as trib3, dusp5 and otud1 were identified as the representative molecular markers of cold-induced damages through network analysis. Biological pathways involved in cold stress responses were mined from the transcriptomic data and their functions in regulating cold resistance were validated using specific inhibitors. The autophagy, FoxO and MAPK (mitogen-activated protein kinase) signaling pathways were revealed to be survival pathways for enhancing cold resistance, while apoptosis and necroptosis were the death pathways responsible for cold-induced mortality. Functional mechanisms of the survival-enhancing factors Foxo1, ERK (extracellular signal-regulated kinase) and p38 MAPK were further characterized by inhibiting their activities upon cold stress and analyzing gene expression though RNA-seq. These factors were demonstrated to determine the cold resistance of zebrafish through regulating apoptosis and p53 signaling pathway. These findings have provided novel insights into the stress responses elicited by lethal cold and shed new light on the molecular mechanisms underlying cold resistance of fish.

Loss of AML 1 / Runx 1 accelerates the development of MLL-ENL leukemia through down-regulation of p 19 ARF *

Abstract:Dysfunction of AML1/Runx1, a transcription factor, plays a crucial role in the development of many types of leukemia. Additional events are often required for AML1 dysfunction to induce full-blown leukemia; however, a mechanistic basis of their cooperation is still elusive. Here, we investigated the effect of AML1 deficiency on the development of MLL-ENL leukemia in mice. Aml1 excised bone marrow cells lead to MLL-ENL leukemia with shorter duration than Aml1 intact cells in vivo. Although the number of MLL-ENL leukemia-initiating cells is not affected by loss of AML1, the proliferation of leukemic cells is enhanced in Aml1excised MLL-ENL leukemic mice. We found that the enhanced proliferation is the result of repression of p19ARF that is directly regulated by AML1 in MLL-ENL leukemic cells. We also found that downregulation of p19ARF induces the accelerated onset of MLL-ENL leukemia, suggesting that p19ARF is a major target of AML1 in MLL-ENL leukemia. Molecular Mechanism Based on Histopathology, Antioxidant System and Transcriptomic Profiles in Heat Stress Response in the Gills of Japanese Flounder

Abstract:As an economically important flatfish in Asia, Japanese flounder is threatened by continuously rising temperatures due to global warming. To understand the molecular responses of this species to temperature stress, adult Japanese flounder individuals were treated with two kinds of heat stress-a gradual temperature rise (GTR) and an abrupt temperature rise (ATR)-in aquaria under experimental conditions. Changes in histopathology, programmed cell death levels and the oxidative stress status of gills were investigated. Histopathology showed that the damage caused by ATR stress was more serious. TUNEL signals confirmed this result, showing more programmed cell death in the ATR group. In addition, reactive oxygen species (ROS) levels and the 8-O-hDG contents of both the GTR and ATR groups increased significantly, and the total superoxide dismutase (T-SOD) activities and total antioxidant capacity (T-AOC) levels decreased in the two stressed groups, which showed damage to antioxidant systems. Meanwhile, RNA-seq was utilized to illustrate the molecular mechanisms underyling gill damage. Compared to the control group of 18 °C, 507 differentially expressed genes (DEGs) were screened in the GTR group; 341 were up-regulated and 166 were down-regulated, and pathway enrichment analysis indicated that they were involved in regulation and adaptation, including chaperone and folding catalyst pathways, the mitogen-activated protein kinase signaling (MAPK) pathway and DNA replication protein pathways. After ATR stress, 1070 DEGs were identified, 627 were up-regulated and 423 were down-regulated, and most DEGs were involved in chaperone and folding catalyst and DNA-related pathways, such as DNA replication proteins and nucleotide excision repair. The annotation of DEGs showed the great importance of heat shock proteins (HSPs) in protecting Japanese flounder from heat stress injury; 12 hsp genes were found after GTR, while 5 hsp genes were found after ATR. In summary, our study records gill dysfunction after heat stress, with different response patterns observed in the two experimental designs; chaperones were activated to defend heat stress after GTR, while replication was almost abandoned due to the severe damage consequent on ATR stress.
Effects of low-temperature stress on intestinal structure, enzyme activities and metabolomic analysis of juvenile golden pompano (Trachinotus ovatus)

Abstract:Water temperature plays a crucial role in the growth, survival, and reproduction of fish species, as they make up the majority of aquatic fauna. In this study, the effects of low temperature were studied on the functional state of juvenile golden pompano ( Trachinotus ovatus ) under low-temperature stress. The study was conducted at 28°C in the control group and 18°C in the cold group for 14 d to determine the intestinal tissue, digestive and antioxidant enzyme activities, and metabolites of juvenile fish. The results showed that: (1) the swelling degree of the muscle layer deepened and was congested with a longer low-temperature stress period. The folds were sparse, from slight swelling to shedding and deformation. The intestinal mucosa was necrotic and had vacuoles, and the number gradually increased. Serious erosion of the villi occurred. (2) The specific activities of digestive enzymes showed a downward trend. (3) The intestinal superoxide dismutase (SOD) activity, malondialdehyde (MDA) content, and glutathione peroxidase (GSH-Px) activity showed an upward trend. The intestinal catalase (CAT) activity showed a downward trend. (4) Compared with the control group, there were 28 metabolites in the cold group showing significant differences, among which Z, 11Z, 14Z-eicosatrienoic acid, stearic acid, and adrenic acid showed an upward trend. In contrast, spermidine and uracil showed a downward trend. Among the enriched metabolic pathways, the main differential pathways were unsaturated fatty acid biosynthesis, fatty acid biosynthesis, linoleic acid metabolism, pyrimidine metabolism, and β-alanine metabolism. According to metabolomic analysis, under low-temperature stress, the fish body improved the synthesis of unsaturated fatty acids and saturated fatty acids to adapt to a low-temperature environment and consumed spermidine to improve its immune ability to clear the peroxide generated by the synthesis of unsaturated fatty acids in the body so that the cells were protected from oxidative damage. After 14 days, low-temperature stress affected metabolites and enzyme activity indices in juvenile golden pompano. Low-temperature stress causes changes in intestinal antioxidants and digestive enzymes and damage intestinal tissues. As a result of this exploration of how low temperatures affect the juvenile golden pompano, the foundation is laid for future studies, such as the molecular mechanisms of low-temperature adaptation in fish species.

Application of transcriptome analysis to investigate the effects of long-term low temperature stress on liver function in the tiger puffer (Takifugu rubripes)

Abstract:The tiger puffer ( Takifugu rubripes ) is an important economic fish species in northern China. However, it is a warm-temperature species, and low winter temperatures can result in high mortality in aquaculture. Understanding the mechanisms of cold resistance in tiger puffers will thus provide critical information to help cope with winter cold. In this study, we performed transcriptome analysis of livers from puffer fish kept at different temperatures (18°C, 13°C, and 8°C) to identify the key pathways and genes involved in the response to low-temperature stress. We also detected serum levels of proteases, arginine, and proline to obtain further information on the response to cold adaption. Totals of 51, 942, and 195 differentially expressed genes were identified in the 18°C vs 13°C, 18°C vs 8°C, and 13°C vs 8°C groups, respectively. Pathway analysis showed that significantly enriched pathways were mainly related to digestion, metabolism, and environmental adaptation. Most genes in the pathways related to digestion and metabolism were down-regulated, while most genes in the pathways related to environmental adaptation were up-regulated. Serum levels of proteases were significantly lower in the low-temperature groups (13°C and 8°C) compared with the control group (18°C), while arginine and proline levels were significantly higher in the 8°C group compared with the other two groups. These results suggest that low temperature caused digestive and metabolic disorders, as well as adaptive changes to low temperature in tiger puffers. On this premise, we found that some up-regulated genes in the pancreatic secretion pathway, arginine and proline metabolism pathway, and circadian rhythm pathway played important roles in the survival, growth, and development of tiger puffers under low-temperature stress. The accumulation of arginine and proline can maintain metabolism and circulation and resist cold stress. The circadian rhythm is closely related to digestion and metabolism, which is an adaptive change and plays a positive role in the resistance to low temperature. The results of this study provide new insights and a theoretical basis for the study of cold tolerance in tiger puffers.

Abstract:<h3 class="Heading">Background</h3><p class="Para"><em class="EmphasisTypeItalic">T. fasciatus</em> (<em class="EmphasisTypeItalic">Takifugu fasciatus</em>) faces the same problem as most warm water fish: the water temperature falls far below the optimal growth temperature in winter, causing a massive death of <em class="EmphasisTypeItalic">T. fasciatus</em> and large economic losses. Understanding of the cold-tolerance mechanisms of this species is still limited. Integrated application of multi-omics research can provide a wealth of information to help us improve our understanding of low-temperature tolerance in fish.</p><h3 class="Heading">Results</h3><p class="Para">To gain a comprehensive and unbiased molecular understanding of cold-tolerance in <em class="EmphasisTypeItalic">T. fasciatus</em>, we characterized mRNA-seq and metabolomics of <em class="EmphasisTypeItalic">T. fasciatus</em> livers using Illumina HiSeq 2500 and UHPLC-Q-TOF MS. We identified 2544 up-regulated and 2622 down-regulated genes in the liver of <em class="EmphasisTypeItalic">T. fasciatus</em>. A total of 40 differential metabolites were identified, including 9 down-regulated and 31 up-regulated metabolites. In combination with previous studies on proteomics, we have established an mRNA-protein-metabolite interaction network. There are 17 DEMs (differentially-expressed metabolites) and 14 DEGs-DEPs (differentially co-expressed genes and proteins) in the interaction network that are mainly involved in fatty acids metabolism, membrane transport, signal transduction, and DNA damage and defense. We then validated a number of genes in the interaction network by qRT-PCR. Additionally, a number of SNPs (single nucleotide polymorphisms) were revealed through the transcriptome data. These results provide key information for further understanding of the molecular mechanisms of <em class="EmphasisTypeItalic">T. fasciatus</em> under cold stress.</p><h3 class="Heading">Conclusion</h3><p class="Para">The data generated by integrated application of multi-omics can facilitate our understanding of the molecular mechanisms of fish response to low temperature stress. We have not only identified potential genes and SNPs involved in cold tolerance, but also show that some nutrient metabolites may be added to the diet to help the overwintering of <em class="EmphasisTypeItalic">T. fasciatus</em>.</p>

Abstract:The increasing frequency of high-temperature extremes threatens largemouth bass Micropterus salmoides , a significant fish for freshwater ecosystems and aquaculture. Our previous studies at the transcript level suggested that heat stress induces hepatic apoptosis in largemouth bass. In the current study, we sought to validate these findings and further investigate the role of the c-Jun N-terminal kinase (JNK)/P53 signaling in hepatic apoptosis under heat stress. First, heat treatments were conducted in vivo and in vitro under different temperatures: 28 °C, 32 °C, and 37 °C. In primary hepatocytes subjected to heat treatment, cell viability was evaluated via the Cell Counting Kit-8, while mitochondrial membrane potential and nuclear morphology were assessed through JC-1 and Hoechst 33258 staining, respectively. We observed reductions in both cell viability and mitochondrial membrane potential (ΔΨm), along with alterations in nuclear morphology, in primary hepatocytes exposed to heat stress at temperatures of 32 °C and 37 °C. Quantitative real-time PCR revealed significant alterations in the expression profiles of intrinsic apoptosis-related genes within liver tissues under heat stress. Immunohistochemistry analysis revealed that JNK1 signaling increased as the temperature increased, JNK2 expression increased only at 37 °C, and JNK3 expression did not change with temperature. We speculate that JNK1 and JNK2 have pro- and anti-apoptotic effects, respectively. Western blot analysis conducted on cultured hepatocytes further validated these findings. JNK inhibition reduced hepatocyte apoptosis, improved nuclear morphology, and maintained ΔΨm even after 37 °C treatment. These results not only confirm that heat stress led to intrinsic apoptosis of hepatocytes but also indicated that JNK1 could mediate P53 expression and activate caspase-dependent intrinsic apoptosis in largemouth bass hepatocytes under such conditions. This study illuminates the physiological responses of largemouth bass to acute heat stress, offering valuable insights into the potential impacts of climate change on freshwater fishes and the sustainability of aquaculture.

Abstract:Hypoxia is a frequent challenge to aquatic vertebrates as compared with that for their terrestrial counterparts. All vertebrates respond to hypoxia in a similar, but not identical manner, indicating that these responses appeared early in the evolution of vertebrates. The aim of this study is to find out the effects of hypoxia on apoptosis in the central nervous system (CNS) of sturgeon, an archaic fish. With the regional specialization of the CNS, we hypothesize that if cell death does occur, the response will vary between regions, i.e., some CNS areas will be more susceptible to hypoxia than the others would. Sturgeons (Acipenser shrenckii) were subjected to hypoxia by exposure to either air or hypoxic water. After 6- or 30-h recovery they were sacrificed and the following regions of the CNS: retina, olfactory lobe, optic tectum, pituitary, cerebellum, pons/medulla, and spinal cord were examined by the terminal transferase mediated dUTP nick end labeling technique and for the cleaved fragment of activated caspase-3 by Western blotting. In hypoxia-treated sturgeons, the retina, optic tectum, pituitary, and spinal cord were found to have significantly more apoptotic cells than did untreated sturgeons at both 6 and 30 h after the hypoxic insults, indicating prolonged damage. Apoptosis was confirmed by Western blotting of the cleaved fragment of activated caspase-3. Olfactory lobe, cerebellum, and pons/medulla had relatively few apoptotic cells. The CNS of sturgeon showed a differential pattern of apoptosis in response to hypoxia.
Comprehensive analysis of histophysiology, transcriptome and metabolome tolerance mechanisms in black porgy (Acanthopagrus schlegelii) under low temperature stress

Abstract:Low temperature stress has adverse effects on fish growth and reproduction, causing huge economic losses to the aquaculture industry. Especially, black porgy (Acanthopagrus schlegelii) farming industry in north of Yangtze River has been severely affected by low temperature for a long time. To explore the tolerance mechanism of black porgy to low temperature stress, the experiment was designed. The liver and gill tissues of black porgy were taken from the water temperature point of 15 °C (control group named as CG), 3.8 °C (cold sensitive group named as CS) and 2.8 °C (cold tolerant group named as CT) with a cooling rate of 3 °C/d from 15 °C for histophysiology, transcriptomics and metabolomics analysis. After cold stress, the histological results showed that the nucleus of the black porgy liver tissue appeared swelling, the cell arrangement was disordered; meanwhile the gill lamellae were twisted and broken, the epidermis was detached and aneurysm appeared. In addition, the expression of antioxidant, glucose metabolism and immune-related enzymes in the liver and gill of black porgy also changed significantly after low temperature stress. By analyzing the transcriptome and metabolome dates of black porgy liver, 3474 differentially expressed genes (DEGs) and 689 differentially expressed metabolites (DEMs) involved in low temperature stress were identified, respectively. The results of the transcriptome and metabolome combined analysis showed that individuals in the CS group mainly supplied energy to the body through lipid metabolism and amino acid metabolism, and meanwhile the apoptosis pathway was activated. While, individuals in the CT group mainly through glucose metabolism and steroid hormone biosynthesis to supply energy for the body. The validation results of qPCR on eight functional genes further demonstrated the reliability of RNA-Seq data. In summary, the results provide molecular information about adaptation to climate change and genetic selection of black porgy.

Abstract:<p>In aquatic ecosystems, the temperature of the water is an important ecological factor that modulates aquatic organisms' metabolism, growth, development, and reproduction. In this study, the morphological, transcriptomic, and metabolomic analyses of response of <em>Marsupeneus japonicus</em> to acute cold stress was investigated. The results revealed that low temperature caused profound morphological damage to the hepatopancreas. Transcriptomic responses suggested that energy and primary metabolism, cytoskeleton structure, and apoptosis signaling were altered. The metabolic responses to cold stress included changes of multiple amino acids and unsaturated fatty acids. Combined transcriptomic and metabolomic data indicated that energy metabolism pathways were downregulated in the hepatopancreas under cold stress. However, <em>M. japonicus</em> increased ATP and unsaturated fatty acids production to ameliorate. Moreover, cold stress caused significant attenuation of macrophage apoptosis. This study provides key information to increase our understanding of low-temperature tolerance in shrimp.</p>

Abstract:Postmortem alteration by apoptosis has significant effects on flesh quality. Currently, the information necessary to understand the apoptotic behavior and the molecular mechanisms during postmortem alteration in fish muscle is still lacking. Activation of apoptosis and the cytokines involved in regulating apoptosis in fish muscle were evaluated during postmortem condition at 4 °C for 5 days in terms of apoptotic morphology changes, nucleus DNA fragmentation, caspases activation and related gene expressions. The triggering apoptotic mechanisms associated with multiple cytokines transcriptional levels showed that the up-regulated pro-apoptotic mediators [IFN-γ2, TNF-α, IL-6, IL-1β, IL-17D, IL-12p35 and IL-10 (except IL-15)] and the down-regulated anti-apoptotic mediators of [IL-8 and IL-11 (except TGF-β and IL-4)] both regulated apoptosis at early stage, which were regulated by NF-κB and TOR, respectively. Results suggested that transcriptional regulation of multiple cytokines produce a positive outcome on triggering apoptosis.
Acute cold stress leads to zebrafish ovarian dysfunction by regulating miRNA and mRNA

Abstract:Temperature is a critical factor that regulates the reproduction processes in teleost. However, the gonadal response mechanism to cold stress in fish remains largely unknown. In the present study, female zebrafish were exposed to different extents of low temperatures at 18 °C and 10 °C for 48 h. The ovarian histology was remarkably damaged after cold stress exposure. Integrated analysis of miRNA-mRNA was used to investigate the ovarian response to acute cold stress. A large number of mRNAs and miRNAs were altered by cold stress, which are involved in extensive biological processes. It is indicated that the signal transduction of MAPK and Calcium signaling pathway is highly engaged in zebrafish ovary to adapt to cold stress. The immune system was dysregulated by cold stress while the ovarian autophagy was activated. Remarkably increased gene number related to reproductive functions was identified in the cold stress at 10 °C compared to the control. The cold stress-induced dysregulated reproductive genes include star, hsd3b1, hsd17b1, inha, insl3, amh, nanos1 and foxl2. Combined with the dysregulated insulin, IGF and progesterone signaling, it is suggested that cold stress affects ovarian function in multiple aspects, including oocyte meiosis, folliculogenesis, final maturation and ovarian maintenance. On the other hand, the ovarian miRNA-mRNA regulatory network response to cold stress was also constructed. Overall, our result revealed the ovarian response to cold stress in zebrafish and provided insight into the fish adaptation mechanism to acute temperature change.

Abstract:Global warming has led to an increase in the frequency of cold extremes, causing significant economic losses in aquaculture, particularly in subtropical regions. Milkfish ( Chanos chanos ) holds significant importance in aquaculture within subtropical Asian regions. Despite milkfish's ability to tolerate varying salinity levels, frequent cold snaps associated with extreme weather events have caused substantial mortality. Understanding the molecular and cellular mechanisms underlying cold stress-induced cell death is crucial for developing effective strategies to mitigate such losses. Given the pivotal role of the liver in fish physiology, we established a primary milkfish hepatocyte culture demonstrating robust proliferation and expressing a unique marker leptin A . The molecular characterization of cold-treated hepatocytes at 18 °C showed that the mRNA levels of superoxide dismutase ( sod1 ) and catalase ( cat ), responsible for neutralizing reactive oxygen species (ROS), were downregulated in freshwater (FW) conditions, while cat expression was upregulated in seawater (SW) conditions. This differential modulation of ROS signaling implies distinct responses in FW and SW, leading to higher ROS levels and increased cell death in FW condition compared to those in SW. Transcriptomic analysis of liver tissues from milkfish reared in FW or SW, with or without cold stress, revealed distinct gene expression patterns. Although cold stress affected a common set of genes in both FW and SW conditions, SW-specific cold responsive genes are associated with metabolic pathways while FW-specific genes were linked to cell proliferation pathways. Notably, gene set enrichment analysis highlighted the downregulation of cytochrome-related genes, a major source of ROS production, in response to cold stress in SW. In summary, our study unveils an insightful mechanism whereby the salinity of SW counteracts cold stress-induced ROS signaling, emphasizing the feasibility and practicality of preconditioning fish in SW as a preventive measure against cold stress-induced mortality.

Abstract:Teleost fish have evolved various adaptations that allow them to tolerate cold water conditions. However, the underlying mechanism of this adaptation is poorly understood in Tibetan Plateau fish. RNA-seq combined with liquid chromatography‒mass spectrometry (LC‒MS/MS) metabolomics was used to investigate the physiological responses of a Tibetan Plateau-specific teleost, Gymnocypris przewalskii , under cold conditions. The 8-month G. przewalskii juvenile fish were exposed to cold (4 °C, cold acclimation, CA) and warm (17 °C, normal temperature, NT) temperature water for 15 days. Then, the transcript profiles of eight tissues, including the brain, gill, heart, intestine, hepatopancreas, kidney, muscle, and skin, were evaluated by transcriptome sequencing. The metabolites of the intestine, hepatopancreas, and muscle were identified by LC‒MS/MS. A total of 5,745 differentially expressed genes (DEGs) were obtained in the CA group. The key DEGs were annotated using Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analysis. The DEGs from the eight tissues were significantly enriched in spliceosome pathways, indicating that activated alternative splicing is a critical biological process that occurs in the tissues to help fish cope with cold stress. Additionally, 82, 97, and 66 differentially expressed metabolites were identified in the intestine, hepatopancreas, and muscle, respectively. Glutathione metabolism was the only overlapping significant pathway between the transcriptome and metabolome analyses in these three tissues, indicating that an activated antioxidative process was triggered during cold stress. In combination with the multitissue transcriptome and metabolome, we established a physiology-gene‒metabolite interaction network related to energy metabolism during cold stress and found that gluconeogenesis and long-chain fatty acid metabolism played critical roles in glucose homeostasis and energy supply.

Abstract:High temperatures and low oxygen in aquatic environments, such as intensive aquaculture or in natural watersheds, inevitably cause stress in fish. Fish are exposed to high temperatures during the summer, which exacerbates hypoxia. Hypoxia (1.2 +/- 0.2 mg/L) under 20 degrees C (20 HG) and 26 degrees C (26 HG) was simulated to induce stress in largemouth bass (Micropterus salmoides). Related enzymes and genes involved in antioxidant, immune, and apoptotic responses were selected to explore the interactive effects of temperature and hypoxia on largemouth bass. The results showed that malondialdehyde (MDA) levels in plasma, gill, and liver increased in the 26 HG (p < 0.05). Liver superoxide dismutase (SOD) activity increased in the 26 HG. Peak SOD (SOD1, SOD2, SOD3a, and SOD3b), CAT, and GSH-Px mRNA levels in the gill and liver were observed at 12-24 h of stress. The levels of gill and liver total antioxidant capacity, catalase (CAT), glutathione peroxidase (GSH-Px) activities and other enzyme activities and genes in the 26 HG were higher than those in the 20 HG (p < 0.05). The gill and liver add phosphatase and alkaline phosphatase activities increased with time in the 26 HG (p < 0.05), while gill and liver lysozyme activities in the 26 HG were lower than those in the 20 HG (p < 0.05). Tumor necrosis factor-alpha mRNA level was upregulated in the gill and downregulated in the liver at 24 h in the 26 HG. Interleukin (IL)-1 beta and IL-8 mRNA levels were upregulated in the gill and liver in the 26 HG at 24 h, whereas IL-15 mRNA level was downregulated in the 26 HG at 12 h. Transforming growth factor-beta 1 mRNA level was upregulated in the gill in the 20 HG at 24 h, but downregulated in gill and liver in the 26 HG at 24 h. Similarly, IL-10, Hepcidin-1, and Hepcidin-2 showed lower expression levels in the 26 HG. Gill and liver caspase-3 activities were higher in the 26 HG (p < 0.05), and gill caspase-3 activity was higher than that in the liver. The mRNA levels of proapoptotic genes (caspase-3, caspase-8, and caspase-9) were higher in the 26 HG. The present study demonstrates the interactive effects of temperature and hypoxia on stress in largemouth bass gill and liver. These results will be helpful to understand the mechanisms of stress induced by temperature and hypoxia in fish and provide a theoretical basis for aquaculture management.
