Abstract:Article Details: Received: 2020-10-20 | Accepted: 2020-11-27 | Available online: 2021-01-31 https://doi.org/10.15414/afz.2021.24.mi-prap.1-4 Abstract The aim of the study was to observe the salivary concentrations of cortisol in horses during daily routine over the course of four weeks lasting experiment in three different stages of day to monitor the changing values of cortisol in horse saliva during the day. Saliva was obtained from 12 Slovak warmblood horses – 1 stallion, 6 mares and 5 geldings. In the experiment we focused purely on changes of salivary cortisol concentrations over the course of the daily routine.The results shown us multiple significant changes between individual sample collections and thus, we can state that according to our results the highest concentration of cortisol in horse saliva is in the morning and it decreases throughout the day with lowest measured concentrations being in the final sample collection of the day (at 22:00). These changes had no visible effect on horses´ organism throughout entirety of the experiment and were caused due to horses daily rhythm. From stable to stable: how does stress influence the first line of immune defense in sport horses

Abstract:Stressful factors, including physical exercise, can influence the body’s defense mechanisms and can be involved in the etiology and evolution of systemic diseases. The aim of this study was to investigate the influence of various stressors to which sport horses are commonly subjected on the innate immune system. For this, four experimental protocols were designed including different animals, namely: (a) resting horses subjected to everyday medium level exercise (n = 18, average age = 7 years, range: 6 to 11 years), (b) medium course transportation, where the distance of one-way trips represented 270 km (n = 8, average age = 7 years, range: 6 to 9 years), (c) a long course, with a 561 km one-way distance (n = 5, average age = 8.2, range: 7 to 11 years) and (d) pre- and post-competition investigations (n = 9, average age = 7 years, range: 6 to 11). Blood was sampled on heparine (50 UI/ml) before and after the workout or transportation. The phagocytic activity of leukocytes was measured by a carbon particle inclusion test, along with total immunoglobulin/opsonin levels (Ig) (24% Zn sulphate precipitation test) and circulating immune complexes (4.2% polyethylene glycol precipitation test). The results were statistically interpreted by use of Micrososft Excel program (Student’s t test). The various types of stress differently influenced the equine athletes. While a moderate daily exercise did not substantially influence the humoral and cellular effectors of innate immunity, transportation exerted stronger beneficial effects based on the distance to the competition site. As a fast acting antimicrobial mechanism, the phagocytosis underwent the most pronounced changes, positively influenced mainly by the short course transportation. While the transportation stress enhanced phagocytosis, the competition decreased the potential of the cells to react, without significantly influencing their numbers. The experiment suggested that management measures, such as age-tailored exercise and maternal care and diet could help in alleviating stress from stable to stable, during the competition season, thus improving animal welfare and allowing a better health forecast.

Abstract:Immunoglobulin A (IgA), which is heavily glycosylated, interacts with a variety of receptors, e.g. the asialoglycoprotein receptor (ASGP‐R), which binds terminal galactose residues, and the Fcα receptor (FcαRI). It has thus been proposed that elevated serum levels of IgA in primary Sjögren's syndrome (pSS) are caused by its defective clearance. To test this hypothesis, we developed a method (based on sialyl transferases eluted from a hepatoma cell line) to increase the amount of sialic acid (SA) on IgA, and used a battery of IgA1‐ and IgA2‐specific glycosidases to reduce this amount. Binding of IgA1 and IgA2 to ASGP‐R and FcαRI was found to be sugar dependent because oversialylated IgA bound less than native or desialylated IgA. However, individual sugars did not play a direct role in this binding. Given that IgA are oversialylated in pSS, defective clearance of IgA may indeed be ascribed to an excess of SA in IgA1 and IgA2.

Abstract:The study assessed the impact of adverse conditions of feeding and keeping horses on the state of their non-specific resistance. Morphological indicators of blood (hemoglobin content, number of erythrocytes and leukocytes), as well as indicators of humoral (BASC, LASC) and cellular link of innate immunity (phagocytic activity of leukocytes) were used. During the experiment, the horses had a satisfactory general clinical condition, and the main physiological indicators (body temperature, arterial pulse frequency, number of respiratory movements) were within the age norm. It is important to note that there was no significant difference between these parameters in animals at different time periods, which indicated a stable physiological state of the horses at the beginning and at the end of the experiment. The results of the study showed that long-term stress can cause changes in the hematological and biochemical parameters of horses. For example, the level of hemoglobin in clinically healthy horses was 136±2.2 g/l, but after exposure to stress factors it decreased to 110±1.8 g/l (р<0.001). At the stage of adaptation, the hemoglobin level increased to 124±2.6 g/l (р<0.01) and almost reached the values of clinically healthy horses (132±3.2 g/l) at rest. The number of erythrocytes in clinically healthy horses was 8.8±0.1 T/l. Under stress, this indicator increased to 9.8±0.2 T/l (р<0.01), but decreased to 9.6±0.2 T/l (р<0.05) at the stage of adaptation and reached the lowest level at rest — 8.6±0.1 T/l. A decrease in the number of leukocytes under the influence of stress has been confirmed by our research. In clinically healthy horses, the number of leukocytes was 6.7±0.3 G/l, while under stress this indicator decreased to 5.3±0.2 g/l (p<0.001). At the stage of adaptation, the number of leukocytes increased to 5.5±0.3 g/l (р<0.01) and was 6.2±0.3 G/l at rest. Stress also affected the indicators of non-specific resistance. In clinically healthy horses, the level of BASC was 85±1.8%, but under stress this indicator decreased to 70±2.1% (р<0.001). At the stage of adaptation, BASK increased to 75±1.1% (р<0.01), and at rest it was 82±1.7%. The level of LASK in clinically healthy horses was 35±0.2%, but under stress this indicator decreased to 28±0.4% (р<0.001). At the stage of adaptation, LASK increased to 30±0.2% (p<0.001) and reached 36±0.5% at rest. The phagocytic activity of leukocytes in clinically healthy horses was 60±1.5%, but under stress this indicator decreased up to 50±1.1% (р<0.001). At the stage of adaptation, phagocytic activity increased to 55±0.5% (р<0.01) and was 63±0.9% (р<0.05) at rest. Thus, the results of our study confirm that stress negatively affects the hematological and biochemical parameters of horses, as well as their non-specific resistance.

Abstract:Many horses exhibit stereotypies, especially when living in human controlled environments that may prevent horses from satisfying natural needs in terms of feeding, drinking, moving, and socializing. In human medicine, obsessive compulsive disorder and other severe psychiatric disturbances are associated with stereotypic behaviors; salivary biomarkers evaluation is considered a reliable tool for diagnosis of common mental health disorders because saliva collection easy to obtain and noninvasive. In this study, we hypothesized that salivary cortisol concentrations, in addition to alpha-amylase (sAA) and butyrylcholinesterase (BChE) activities, are considered stress biomarkers that may be influenced in horses trained for racing competition with stereotypic behaviors. Saliva at rest condition was obtained from ten non-stereotypic Thoroughbreds horses involved in high-level competition; eleven Thoroughbreds high-level competition horses showing stereotypic behaviors, and five Thoroughbreds leisure non-competition horses. Cortisol was found to be higher in high-level competition non-stereotypic horses and sAA was significantly higher in non-stereotypic leisure horses when compared to horses involved in competition, while BChE did not change between groups. These results may represent the basis for further behavioural evaluation to elucidate how stereotypic horses and horses involved in competition overcome stressful situations.

Physiological and Psychological Responses to a Maximal Swimming Exercise Test in Adolescent Elite Athletes

Abstract:Background: Continuously rising performances in elite adolescent athletes requires increasing training loads. This training overload without professional monitoring, could lead to overtraining in these adolescents. Methods: 31 elite adolescent athletes (boys: n = 19, 16 yrs; girls: n = 12, 15 yrs) participated in a field-test which contained a unified warm-up and a 200 m maximal freestyle swimming test. Saliva samples for testosterone (T) in boys, estradiol (E) in girls and cortisol (C) in both genders were collected pre-, post- and 30 min post-exercise. Lactate levels were obtained pre- and post-exercise. Brunel Mood Scale, Perceived Stress Scale and psychosomatic symptoms questionnaires were filled out post-exercise. Results: Lactate levels differed between genders (boys: pre: 1.01 ± 0.26; post: 8.19 ± 3.24; girls: pre: 0.74 ± 0.23; post: 5.83 ± 2.48 mmol/L). C levels increased significantly in boys: pre- vs. post- (p = 0.009), pre- vs. 30 min post-exercise (p = 0.003). The T level (p = 0.0164) and T/C ratio (p = 0.0004) decreased after field test which draws attention to the possibility of overtraining. Maximal and resting heart rates did not differ between genders; however, heart rate recovery did (boys: 29.22 ± 7.4; girls: 40.58 ± 14.50 beats/min; p = 0.008). Conclusions: Our models can be used to explain the hormonal ratio changes (37.5-89.8%). Based on the results this method can induce hormonal response in elite adolescent athletes and can be used to notice irregularities with repeated measurements.
How training loads in the preparation and competitive period affect the biochemical indicators of training stress in youth soccer players?

Abstract:Background Physical fitness optimization and injury risk-reducing require extensive monitoring of training loads and athletes’ fatigue status. This study aimed to investigate the effect of a 6-month training program on the training-related stress indicators (creatine kinase – CK; cortisol – COR; serotonin – SER; brain-derived neurotrophic factor – BDNF) in youth soccer players. Methods Eighteen players (17.8 ± 0.9 years old, body height 181.6 ± 6.9 cm, training experience 9.7 ± 1.7 years) were blood-tested four times: at the start of the preparation period (T0), immediately following the preparation period (T1), mid-competitive period (T2), and at the end of the competitive period (T3). CK activity as well as concentrations of serum COR, SER and BDNF were determined. Training loads were recorded using a session rating of perceived exertion (sRPE). Results Statistical analyzes revealed significant effects for all biochemical parameters in relation to their time measurements (T0, T1, T2, T3). The statistical analyzes of sRPE and differences of biochemical parameters in their subsequent measurements (T0–T1, T1–T2, T2–T3) also demonstrated significant effects observed for all variables: sRPE (H KW = 13.189 (df = 2); p = 0.00), COR (H KW = 9.261 (df = 2); p = 0.01), CK (H KW = 12.492 (df = 2); p = 0.00), SER (H KW = 7.781 (df = 2); p = 0.02) and BDNF (H KW = 15.160 (df = 2); p < 0.001). Discussion In conclusion, it should be stated that the most demanding training loads applied in the preparation period (highest sRPE values) resulted in a significant increase in all analyzed biochemical training stress indicators. The reduction in the training loads during a competitive period and the addition of recovery training sessions resulted in a systematic decrease in the values of the measured biochemical indicators. The results of the study showed that both subjective and objective markers, including training loads, are useful in monitoring training stress in youth soccer players.

Do You Think I Am Living Well? A Four-Season Hair Cortisol Analysis on Leisure Horses in Different Housing and Management Conditions

Abstract:The satisfaction of leisure horses’ behavioral needs has begun to be considered a priority, linked to the awareness that horses kept in single boxes may be deprived of social contact and the possibility to perform natural behaviors. Several factors may influence horses’ quality of life also in the paddock, and there are very few data on the effects of those variables on leisure horses’ chronic stress, measurable in terms of activation of the hypothalamic–pituitary–adrenocortical axis. Therefore, managerial choices faced by owners and stables managers are only based on experience, common sense, and anecdotal beliefs. This study assessed and compared the chronic stress levels in leisure horses hosted in structures in the same geographic and climatic area with different daily routines to verify which management strategy could be the one that better contributes to achieving the welfare of horses. Forty-seven horses were divided into three groups homogeneous in terms of sex and age: Mixed management group (n = 12), Paddock group (n = 19), and Natural management group (n = 16). The hair cortisol concentration, a reliable marker of long-term stress, was analyzed in all the horses the same day at four time points of the year. In addition to management strategies, the influences of other variables (sex, age, coat color, and season) were evaluated. Independently from the management strategies, significantly higher hair cortisol values were detected in the autumn and summer, as well as in individuals older than 15 years. No significant differences were highlighted between the sexes or the coat colors. The comparison of the different management strategies showed that, in the summer, autumn, and winter, the hair cortisol levels were significantly lower in the Mixed management group horses than the Paddock group, highlighting that those subjects had better homeostasis. The Natural management group horses’ hair cortisol levels were intermediate between the other two groups of horses in all the seasons. Spending the night in the stables would seem to positively impact the well-being of the horses. These findings, if confirmed by further studies, may be helpful in enhancing horse welfare and assisting in managerial choice decision-making.

Comparison between Match and Training Session on Biomarker Responses in Handball Players

Abstract:A variety of loads are placed upon an athlete in team sports (e.g., training, match, or competitions). However, the volume of the training load plays an important role in match success. Therefore, the aim of this study was to compare the differences in biomarker dynamics during the match and during training, and to evaluate if such training represents a good stimulus for an athlete to adapt to match stress. Ten male handball players (average age of 24.1 ± 3.17 years, body height of 1.88 ± 0.64 m, and body mass of 94.6 ± 9.6 kg) took part in this study. Their saliva cortisol, testosterone, and alpha-amylase were sampled during the match and training (sessions of 90 min duration, respectively). The results showed that cortisol had higher values after the match (0.65 μg/dL) than after training (0.32 μg/dL) (p = 0.05; ES = 0.39). Testosterone concentrations had a steeper increase during a match (65%) than after training (37%). Alpha-amylase levels did not differ significantly between the match and training (p = 0.77; ES = −0.06). Overall, the results showed that the environment of a match was more stressful for the athletes; therefore, a match provoked a stronger endocrine response in the studied markers. Therefore, we concluded that a match seemed to be a stronger trigger for all of the measured biomarker responses.
Peripheral Modulators of the Central Fatigue Development and Their Relationship with Athletic Performance in Jumper Horses

Abstract:The current study aimed to investigate whether peripheral modulators of serotoninergic function and neurohumoral factors’ changes in athletic horses during an official jumping competition, and to evaluate their relationship with the physical performance of competing horses. From 7 Italian Saddle mares (6–9 years; mean body weight 440 ± 15 kg), performing the same standardized warm-up and jumping course during an official class, heart rate (HR) was monitored throughout the competition. Rectal temperature (RT) measurement, blood lactate and glucose concentration, serum tryptophan, leucine, valine, the tryptophan/branched-chain amino-acids ratio (Try/BCAAs), dopamine, prolactin, and non-esterified fatty acids (NEFAs) were assessed before the exercise event (T0), at the end of the competition stage (5 min ± 10 s following the cessation of the exercise, TPOST5), and 30 min after the end of competition (TPOST30). Highest HR values were recorded during the course and at the outbound (p < 0.0001); blood lactate concentration and RT increased after exercise with respect to the rest condition (p < 0.0001). Lower leucine and valine levels (p < 0.01), and higher tryptophan, Try/BCAAs ratio, and NEFAs values were found at TPOST5 and TPOST30 with respect to T0 (p < 0.0001). A higher prolactin concentration was found at TPOST5 and TPOST30 compared to T0 (p < 0.0001), whereas dopamine showed decreased values after exercise compared to rest (p < 0.0001). Statistically significant correlations among the peripheral indices of serotoninergic function, neurohumoral factors, and athletic performance parameters were found throughout the monitoring period. The findings provide indirect evidence that the serotoninergic system may be involved in fatigue during jumper exercise under a stressful situation, such as competition, in which, in addition to physical effort, athletic horses exhibit more passive behavior.

Hormone Concentrations and Semen Parameters in Criollo Breed Stallions Under Training

Abstract:The stress associated with training may reduce reproductive efficiency in Criollo stallions. The objective of this study was to compare semen quality and hormone concentrations in Criollo stallions under training or under regular field conditions. Criollo breed stallions (n = 18) were evaluated during the spring. The exercise group (n = 9) performed 1 hour of exercise per day and participated in competitions during the experimental period. The control group (n = 9) neither performed exercise nor participated in competitions. Serum and semen samples were obtained every 15 days (two separate ejaculates an hour apart). Sperm motility, velocity, and morphology were evaluated with a phase-contrast microscope and concentration by a hemocytometer. Diff-Quik stain was used to identify polymorphonuclear cells, and the degree of chromatin condensation was evaluated with the toluidine blue stain. The sperm survival test was performed at a room temperature of 22°C. Semen evaluation was performed in raw samples and in samples diluted in a skim milk and glucose-based extender. Cortisol, testosterone, and estradiol were measured using radioimmunoassay. There was no effect of exercise on testosterone and estradiol concentrations (P = .28 and P = .97, respectively). However, in the exercise group, cortisol concentration was higher after exercise (P = .004). There was an effect of exercise on the following semen parameters: gel-free volume (P < .001), sperm motility (P < .0001), total number of sperm (P = .0001), normal sperm morphology (P < .0001), and total number of morphologically normal and motile sperm (P < .001). No effect of exercise was found in the following semen parameters: color, pH, and sperm concentration. This study showed that exercise had a negative impact on seminal quality; nevertheless, semen parameters were within the normal ranges established for the equine species.
Blood-Based Markers for Skeletal and Cardiac Muscle Function in Eventing Horses before and after Cross-Country Rides and How They Are Influenced by Plasma Volume Shift

Abstract:Horses competing in cross-country tests are subjected to high physical demands. Within the scope of this prospective longitudinal study, blood values of 20 elite eventing horses were examined before and after two- to four-star cross-country rides. The aim was to find out whether blood-based markers for skeletal muscle and cardiac muscle function change after cross-country exercise. Parameters that provide information about fluid balance, muscle enzymes, metabolites and cardiac muscle-specific markers were investigated. We developed an approach to eliminate the concentration changes caused by reduced plasma volume. Parameters were measured pre, 10 and 30 min post exercise and the next morning and were evaluated using a mixed model. Thirty minutes after exercise, most parameter concentrations changed in an exercise-dependent manner. The next morning, most exercise-related markers recovered rapidly, while creatine kinase (CK) (26% increase; p = 0.008) and lactate dehydrogenase (LDH) (15% increase; p < 0.001) showed a declining but sustained increase. Cardiac troponin I (cTnI) increased above the reference range in 40 of the 55 rides (73%) and in 18 of 20 horses in the morning after exercise.

The aim of our study was to determine the effect of indirect factors, such as year of start, sex, breed, number of starts, age and proportion of genes of Slovak Warmblood horse breed. For determination of indirect factors on performance level, results from test efficiency and final placement in showjumping category 4, 5 and 6 years old young horses were used. 540 horses of Slovak Warmblood breed were analysed between 2015 - 2019 and 1155 data samples were collected. Based on the results of the linear model, parameters – the effect of gender and the influence of the gene proportion of Slovak Warmblood horse had statistically significant influence (p<0.05). High significant effect (p<0.001) on performance of Slovak Warmblood horse was observed in parameters – year of start, age during start and breed. Abstract:A variety of stressful situations commonly occur on dairy farms which can impair the well-being of the animals. The aim of this study was to analyse the concentration of cortisol in the saliva of dairy goats and on the basis thereof to determine the degree of stress experienced by them in relation to selected situations on farms. The following situations were selected as stressful: first visit to the milking parlour; weaning off; loading and transport; deworming; and the disruption of social hierarchy. We examined 344 samples from 100 animals using cotton swabs for the saliva collection. Commercially available ELISA kits (Cortisol EIA Kit, BosterBio, California, USA) which can detect cortisol in the saliva of all animal species, were used for the analysis. During the first visit to the milking parlour, weaning off, deworming and disruption of social hierarchy there was a significant ( P < 0.05) increase in cortisol concentrations compared to the basal values. For loading and transport there was a highly significant ( P < 0.01) increase in cortisol concentrations compared to the basal values. Although these situations are inevitable on farms, efforts should be made to eliminate them as much as possible due to the stress the animals experience in them.

Abstract:In many countries, horses remain involved in traditional equestrian events such as those celebrated in Menorca (Balearic Islands, Spain) every year since at least the 14th century. The present study aimed to evaluate the variations in salivary cortisol concentrations to estimate the physiological stress response in horses at the Menorca patronal festivals. Two different editions (years 2016 and 2018) of the festivals in honor of the Virgin of Grace in Maó (Menorca, Spain) were studied. Nineteen and seventeen Pure Breed Menorca stallions were included in the study, respectively. The stallions were aged between seven and twelve years. During celebrations, samples were collected before the start of the festivals between 8–9 a.m. and during the festivals at 8–9 p.m. On the second day of celebrations, the samples were collected at 8–9 a.m. and 3–4 p.m. Finally, on the day after the festivals, one sample was collected at 8–9 p.m. Additionally, a control group was sampled at 8–9 a.m., 3–4 p.m., and 8–9 p.m. Salivary cortisol concentrations were assessed by using a commercial enzyme immunoassay kit specially validated to quantify salivary cortisol in horses. Salivary cortisol concentrations did not show significant differences between sampling hours in the control group (p > 0.05). All the samples collected during festivals were significantly higher than samples of the control group (p 0.05). Hence, the present study describes that the participation of the horses in these particular acts generate an acute and transitory stress response. Overall, the current work provides a reasonable basis for future research on the stress physiology and well-being of horses participating in traditional celebrations or similar events.

Abstract:Horse welfare is the product of multiple factors, including behavioral and physiological adjustments to cope with stressful situation regarding environment and housing condition. Collectively, it is supposed that a horse kept in the wild has a lower level of stress than other housing system, and the aim of the present study was to investigate the level of stress in domestic horses reared in the wild and then moved to human controlled housing, through saliva analysis. Twelve clinically healthy Catria (Italian local breed) mares, usually reared in the wild, were moved into collective paddocks for a folkloric event. Saliva samples were obtained before and after the change of housing condition to evaluate stress biomarkers including salivary cortisol, salivary alpha-amylase, and butyrylcholinesterase (BChol). The mares were also scored using the Welfare Aggregation and Guidance (WAG) Tool to highlight the presence of abnormal behaviors. Despite the absence of differences in behavioral scores between wild and paddocks, salivary cortisol and BChol were found to be higher in the wild and lower when mares were moved to paddocks. The highest concentrations in stress biomarkers like salivary cortisol and BChol in the wild was unexpected, but the need for managing hierarchical relationships, and the exposure to feral animals, predators, and weather changes, might explain these findings. The overall results of the present study may provide further knowledge toward stress response in domesticated horses living in the wild moved to human controlled housing system.

Abstract:The chronic exposure to regular exercise training seems to increase resistance to oxidative stress and improves the antioxidant defense system. The purpose of the present study was to investigate the effect of an exercise test of moderate intensity on oxidative stress biomarkers, antioxidant enzymes activity, and osmotic resistance of erythrocytes in well-trained equine athletes. Eighteen middle-aged horses of Ukrainian warmblood (8.3 ± 1.6 years) and Holsteiner (7.4 ± 1.9 years) breeds were used in this study. All horses have been in regular training for several years. The exercise test induced a significant increase of erythrocyte values, hemoglobin concentration, and hematocrit in horses of both breeds. Regular training induces activation the antioxidant enzymes and thereby can reduce oxidative stress in athletic horses. Our results suggest that the exercise test in horses of both breeds attenuates oxidative stress and accompanied with a significant decrease of lipid peroxidation and oxidatively modified proteins in erythrocytes after exercise. The findings of the present study demonstrated the elevated level of erythrocytes' catalase and glutathione reductase in Ukrainian warmblood horses, as well as decreased level of superoxide dismutase and glutathione reductase in Holsteiner horses reporting changes in levels of exercise-induced oxidative stress biomarkers in horses of both breeds. Statistically significant differences in the percentage of hemolyzed erythrocytes between pre-exercise and postexercise tests were observed and thereby signifying an oxidative stress-dependent impairment of erythrocyte stability. Our data suggest that oxidative stress and enzymatic antioxidant defense biomarkers can be used for the monitoring of fitness level, health benefits, and performance of equine athletes.
