Abstract:Crassulacean acid metabolism (CAM) evolved in arid environments as a water-saving alternative to C3 photosynthesis. There is great interest in engineering more drought-resistant crops by introducing CAM into C3 plants. However, it is unknown whether full CAM or alternative water-saving modes would be more productive in the environments typically experienced by C3 crops. To study the effect of temperature and relative humidity on plant metabolism in the context of water saving, we coupled a time-resolved diel (based on a 24-h day-night cycle) model of leaf metabolism to an environment-dependent gas-exchange model. This combined model allowed us to study the emergence of CAM as a trade-off between leaf productivity and water saving. We show that vacuolar storage capacity in the leaf is a major determinant of the extent of CAM. Moreover, our model identified an alternative CAM cycle involving mitochondrial isocitrate dehydrogenase as a potential contributor to initial carbon fixation at night. Simulations across a range of environmental conditions show that the water-saving potential of CAM strongly depends on the daytime weather conditions and that the additional water-saving effect of carbon fixation by isocitrate dehydrogenase can reach 11% total water saving for the conditions tested.
A case of giant malignant phaeochromocytoma.

Abstract:Malignant phaeochromocytoma is defined as the presence of tumour deposits at sites that are normally devoid of chromaffin cells. We report on a 63-year-old man who had a giant malignant phaeochromocytoma of the right adrenal gland that encased the inferior vena cava. The urinary excretion rates of catecholamines and their metabolites were normal, except for normetanephrine, which was excreted at a higher rate than normal. The tumour was surgically unresectable by laparotomy. Postoperatively, the patient was given a 4-month trial of subcutaneous octreotide and intravenous meta-iodobenzylguanidine I 131. Occult lung secondary tumours were first detected by meta-iodobenzylguanidine scintigraphy after 2 years, and the patient died of bone and lung metastases 1 year later. Because phaeochromocytoma is rare, local experience in managing this disease is limited. This report alerts physicians of the methods of diagnosing and managing surgically unresectable malignant phaeochromocytoma.

Abstract:Crassulacean acid metabolism (CAM) is an adaptation to environments where water availability is seasonal or extremely low. It serves to ensure survival and/ or maintain productivity in these adverse environments. CAM has repeatedly evolved although it requires a large and complex set of enzymes and transporters and regulatory processes to control metabolite flux and pools. To test potential regulatory levels, we analyze the CAM plant Kalanchoë laxiflora embedded in the context of available CAM genome and transcriptome sequences. We show that CAM associated transcripts and proteins do not show a binary on/off pattern between day and night in K. laxiflora. Instead, we observe that many CAM plants display shared amino acid changes compared to C3 plants especially in starch metabolism. Phosphoproteomics identifies phosphoproteome changes in K. laxiflora between day and night. Taken together, the analyses demonstrate the CAM photosynthesis is regulated at the levels of transcripts and proteins.

Abstract:Crassulacean acid metabolism (CAM) is found in a wide variety of vascular plant species, mainly those inhabiting water-limited environments. Identifying and characterizing diverse CAM species enhances our understanding of the physiological, ecological, and evolutionary significance of CAM photosynthesis. In this study, we examined the effect of CO2 elimination on chlorophyll fluorescence-based photosynthetic parameters in two constitutive CAM Kalanchoe species and six orchids. In CAM-performing Kalanchoe species, the effective quantum yield of photosystem II showed no change in response to CO2 elimination during the daytime but decreased with CO2 elimination at dusk. We applied this method to reveal the photosynthetic mode of epiphytic orchids and found that Gastrochilus japonicus, Oberonia japonica, and Bulbophyllum inconspicuum, but not Bulbophyllum drymoglossum, are constitutive CAM. Although B. drymoglossum had relatively high malate content in leaves, they did not depend on it to perform photosynthesis even under water deficient or high light conditions. Anatomical comparisons revealed a notable difference in the leaf structure between B. drymoglossum and B. inconspicuum; B. drymoglossum leaves possess the large water storage tissue internally, unlike B. inconspicuum leaves, which develop pseudobulbs. Our data propose a novel approach to identify and characterize CAM plants without labor-intensive experimental procedures.

Abstract:Direct and non‐destructive measurements of plant‐water relations of plants exhibiting the Crassulacean acid metabolism (CAM) photosynthetic pathway are seldom addressed, with most findings inferred from gas exchange measurements. The main focus of this paper was to study how the water exchange between the chlorenchyma and the hydrenchyma depends on and follows the CAM photosynthetic diel pattern using non‐invasive and continuous methods. Gas exchange and leaf patch clamp pressure probe (LPCP) measurements were performed on Aloe vera (L.) Burm f., a CAM species, and compared to measurements on banana (Musa acuminata Colla), a C3 species. The LPCP output pressure, Pp, of Aloe vera plants follows its diel CAM photosynthetic cycle, reversed to that observed in banana and other C3 species. The four phases of CAM photosynthesis can also be identified in the diel LPCP output pressure, Pp, cycle. The Pp values in Aloe vera are determined by the hydrenchyma turgor pressure, with both parameters being reversely related. A non‐invasive and continuous assessment of the water exchange between the chlorenchyma and the hydrenchyma in CAM plants, namely, by following the changes in the hydrenchyma turgor pressure, is presented. However, showing once more how the LPCP output pressure, Pp, depends on the leaf structure, such an approach can be used to study plant‐water relations in other CAM species with a leaf structure similar to Aloe vera, with the hydrenchyma composing most of the leaf volume.

Abstract:Abstract Photosynthesis is co-limited by multiple factors depending on the plant and its environment. These include biochemical rate limitations, internal and external water potentials, temperature, irradiance, and carbon dioxide (CO2). Amphis- tomatous leaves have stomata on both abaxial and adaxial leaf surfaces. This feature is considered an adaptation to alleviate CO2 diffusion limitations in pro- ductive environments as the diffusion path length from stomate to chloroplast is effectively halved in amphistomatous leaves. Plants may also reduce CO2 limitations through other aspects of optimal stomatal anatomy: stomatal den- sity, distribution, patterning, and size. A number of studies have demonstrated that stomata are overdispersed compared to a random distribution on a sin- gle leaf surface; however, despite their prevelance in nature and near ubiquity among crop species, much less is known about stomatal anatomy in amphis- tomatous leaves, especially the coordination between leaf surfaces. Here we use novel spatial statistics based on simulations and photosynthesis modeling to test hypotheses about how amphistomatous plants may optimize CO2 diffusion in the model angiosperm Arabidopsis thaliana grown in different light environ- ments. We find that 1) stomata are overdispersed, but not ideally dispersed, on both leaf surfaces across all light treatments; 2) the patterning of stomata on abaxial and adaxial leaf surfaces is independent; and 3) the theoretical im- provements to photosynthesis from abaxial-adaxial stomatal coordination are miniscule (≪ 1%) across the range of feasible parameter space. However, we also find that 4) stomatal size is correlated with the mesophyll volume that it supplies with CO2, suggesting that plants may optimize CO2 diffusion limita- tions through alternative pathways other than ideal, uniform stomatal spacing. We discuss the developmental, physical, and evolutionary constraits which may prohibit plants from reaching this theoretical adaptive peak of uniform stomatal spacing and inter-surface stomatal coordination. These findings contribute to our understanding of variation in the anatomy of amphistomatous leaves.

Abstract:Five decades ago, the first report of a shift from C 3 to CAM (crassulacean acid metabolism) photosynthesis following the imposition of stress was published in this journal. The annual, Mesembryanthemum crystallinum (Aizoaceae), was shown to be a C 3 plant when grown under non-saline conditions, and a CAM plant when exposed to high soil salinity. This observation of environmentally triggered CAM eventually led to the introduction of the term facultative CAM, which categorises CAM that is induced or upregulated in response to water-deficit stress and is lost or downregulated when the stress is removed. Reversibility of C 3 –to–CAM shifts distinguishes stress-driven facultative-CAM responses from purely ontogenetic increases of CAM activity. We briefly review how the understanding of facultative CAM has developed, evaluate the current state of knowledge, and highlight questions of continuing interest. We demonstrate that the long-lived leaves of a perennial facultative-CAM arborescent species, Clusia pratensis , can repeatedly switch between C 3 and CAM in response to multiple wet–dry–wet cycles. Undoubtedly, this is a dedicated response to environment, independent of ontogeny. We highlight the potential for engineering facultative CAM into C 3 crops to provide a flexible capacity for drought tolerance.

Abstract:Summary The effect of administration of ethionine upon fatty acid synthesis in a system from the livers of female and male rats has been investigated. Three hours after administration of ethionine, fatty acid synthesis in the livers of both sexes was reduced to about 50% of the control. At 5 hours, male rats recovered from the initial inhibition to 77% of the control, whereas the synthesis in females was further reduced to 24% It was found that the ethionine effect was not merely due to a deficiency in the supply, or in availability of ATP, NADPH, or CoA. Nor was the impairment due to an accumulation of ethionine itself or S-adenosylethio-nine. The possibility is considered that the inhibition of fatty acid synthesis at 5 hours in the liver of female rats may result from the inhibition of the synthesis of acetyl-CoA carboxylase, which is found in the supernatant fraction, in addition to a partial impairment of microsomal function. The cause of the inhibition observed at 3 hours for both sexes remains to be determined.
Metabolic modelling identifies mitochondrial Pi uptake and pyruvate efflux as key aspects of daytime metabolism and proton homeostasis in crassulacean acid metabolism leaves

Abstract:Crassulacean acid metabolism (CAM) leaves are characterized by nocturnal acidification and diurnal deacidification processes related with the timed actions of phosphoenolpyruvate carboxylase and Rubisco, respectively. How CAM leaves manage cytosolic proton homeostasis, particularly when facing massive diurnal proton effluxes from the vacuole, remains unclear. A 12-phase flux balance analysis (FBA) model was constructed for a mature malic enzyme-type CAM mesophyll cell in order to predict diel kinetics of intracellular proton fluxes. The charge- and proton-balanced FBA model identified the mitochondrial phosphate carrier (PiC, Pi/H+ symport), which provides Pi to the matrix to sustain ATP biosynthesis, as a major consumer of cytosolic protons during daytime (> 50%). The delivery of Pi to the mitochondrion, co-transported with protons, is required for oxidative phosphorylation and allows sufficient ATP to be synthesized to meet the high energy demand during CAM Phase III. Additionally, the model predicts that mitochondrial pyruvate originating from decarboxylation of malate is exclusively exported to the cytosol, probably via a pyruvate channel mechanism, to fuel gluconeogenesis. In this biochemical cycle, glyceraldehyde 3-phosphate dehydrogenase (GAPDH) acts as another important cytosolic proton consumer. Overall, our findings emphasize the importance of mitochondria in CAM and uncover a hitherto unappreciated role in metabolic proton homeostasis.
High intraspecific ability to adjust both carbon uptake and allocation under light and nutrient reduction in Halimium halimifolium L.

Abstract:The allocation of recently assimilated carbon (C) by plants depends on developmental stage and on environmental factors, but the underlying mechanisms are still a matter of debate. In the present study, we investigated the regulation of C uptake and allocation and their adjustments during plant growth. We induced different allocation strategies in the Mediterranean shrub Halimium halimifolium L. by a reduction of light (Low L treatment) and nutrient availability (Low N treatment) and analyzed allocation parameters as well as morphological and physiological traits for 15 months. Further, we conducted a (13)CO2 pulse-labeling and followed the way of recently assimilated carbon to eight different tissue classes and respiration for 13 days. The plant responses were remarkably distinct in our study, with mainly morphological/physiological adaptions in case of light reduction and adjustment of C allocation in case of nutrient reduction. The transport of recently assimilated C to the root system was enhanced in amount (c. 200%) and velocity under nutrient limited conditions compared to control plants. Despite the 57% light reduction the total biomass production was not affected in the Low L treatment. The plants probably compensated light reduction by an improvement of their ability to fix C. Thus, our results support the concept that photosynthesis is, at least in a medium term perspective, influenced by the C demand of the plant and not exclusively by environmental factors. Finally, our results indicate that growing heterotrophic tissues strongly reduce the C reflux from storage and structural C pools and therefore enhance the fraction of recent assimilates allocated to respiration. We propose that this interruption of the C reflux from storage and structural C pools could be a regulation mechanism for C translocation in plants.

Abstract:Crassulacean acid metabolism (CAM) photosynthesis has independently evolved many times in arid-adapted plant lineages. (Montiaceae), a desert mass-blooming annual, can upregulate CAM facultatively upon stress such as drought. Few studies, however, consider life history stages when measuring CAM activity or its facultative onset. To test the effect of drought and flowering on photosynthetic activity, we assayed Cistanthe individuals in fully-watered and drought conditions, as well as fully-watered individuals at pre-flowering and flowering life stages. We assembled and annotated a chromosome-scale genome of and compared it with the genome of and analyzed differential gene expression. Results show significantly upregulated CAM in drought conditions as compared to fully-watered conditions; furthermore, flowering individuals showed slightly higher CAM activity as compared to pre-flowering plants, even when fully-watered. Differential gene expression analyses provide preliminary support for the possible co-regulation of CAM expression and reproduction. We emphasize the potentially missed significance of life history in the CAM literature, and consider how the CAM biochemical module could become co-opted into other plant behaviors and responses, such as the shift to reproduction or flowering in annuals.

Abstract:Carbon concentrating mechanisms (CCMs) have evolved numerous times in photosynthetic organisms. They elevate the concentration of CO2 around the carbon-fixing enzyme rubisco, thereby increasing CO2 assimilatory flux and reducing photorespiration. Biophysical CCMs, like the pyrenoid-based CCM of Chlamydomonas reinhardtii or carboxysome systems of cyanobacteria, are common in aquatic photosynthetic microbes, but in land plants appear only among the hornworts. To predict the likely efficiency of biophysical CCMs in C3 plants, we used spatially resolved reaction-diffusion models to predict rubisco saturation and light use efficiency. We found that the energy efficiency of adding individual CCM components to a C3 land plant is highly dependent on the permeability of lipid membranes to CO2, with values in the range reported in the literature that are higher than used in previous modeling studies resulting in low light use efficiency. Adding a complete pyrenoid-based CCM into the leaf cells of a C3 land plant was predicted to boost net CO2 fixation, but at higher energetic costs than those incurred by photorespiratory losses without a CCM. Two notable exceptions were when substomatal CO2 levels are as low as those found in land plants that already employ biochemical CCMs and when gas exchange is limited, such as with hornworts, making the use of a biophysical CCM necessary to achieve net positive CO2 fixation under atmospheric CO2 levels. This provides an explanation for the uniqueness of hornworts' CCM among land plants and evolution of pyrenoids multiple times.

Abstract:CAM (Crassulacean Acid Metabolism) plants reduce the water loss through transpiration in arid environments ( ), using an alternative pathway of carbon assimilation. To ensure food security, engineering CAM into C plants can be achieved by inverting the stomatal rhythm and the timing of major CO uptake from day to night. Identification of the metabolic enzymes and intra-cellular transporters, present in both C and CAM but having different differential temporal activities throughout the diel cycle ( ; ) and quantitative estimations of the flux distributions along the biochemical trajectory of C -to-CAM transition may help us to achieve the goal. Here, we simulate a constraint-based combined metabolic model of guard cell (GC) and mesophyll cell (MC), linking temporal fluctuations of temperature (T) and relative humidity (RH) throughout the diurnal cycle with osmolyte accumulation dependent stomatal opening, CO uptake and transpirational water loss. Starting with C metabolism, gradual increase in water-use efficiency (WUE) captures several known and new differential activities of metabolic enzymes, transporters, sugar-malate cycle etc. in GC and MC during C -to-CAM transition.

Apollo stent for symptomatic atherosclerotic intracranial stenosis: study results.

Abstract:BACKGROUND AND PURPOSE A recent trial shows an 8.3 per 100-patient-years' ischemic stroke rate in the territory of the intracranial stenotic artery, despite aspirin treatment. Our aim was to prospectively study the feasibility and outcome of a new intracranial balloon-expandable Apollo stent for symptomatic atherosclerotic intracranial stenosis (SAIS). MATERIALS AND METHODS Forty-six patients (41 men and 5 women; median, 54 years of age) with forty-eight >or=50% SAISs were enrolled. Procedural feasibility was evaluated by stent success (residual stenosis <or=30%) and procedural time. The primary end point was ischemic stroke in the target-lesion artery territory, including any stroke and death within 30 days. RESULTS Forty-four lesions (91.7%) obtained stent success within a median procedural time of 50.6 minutes. Severe tortuosity correlated with stent failure. Three patients (6.5%, 3/46) had minor strokes within 30 days. All patients were available for follow-up (46 had 30-day follow-up, 45 had 6-month follow-up, 44 had 12- and 18-month follow-up, and 24 had follow-up of >or=24 months), which varied from 1 month to 30.7 months (median, 23.9 months). After 30 days, 1 patient (2.2%, 1/46) developed minor stroke in the target-lesion artery territory at 6.7 months. Primary end point rate was 4.3 per 100 patient years. Angiographic follow-up was performed in 25 patients. Seven restenoses (28%, 7/25) were detected, 1 of which was symptomatic. CONCLUSION Angioplasty with the Apollo stent for symptomatic atherosclerotic intracranial stenosis is feasible. Severe tortuosity is an independent predictor of stent failure. Our clinical outcome seems to compare favorably with results of aspirin therapy, but the restenotic rate was high.

Abstract:Crassulacean acid metabolism (CAM) and the capacity to store large quantities of water are thought to confer high water use efficiency (WUE) and survival of succulent plants in warm desert environments. Yet the highly variable precipitation, temperature and humidity conditions in these environments likely have unique impacts on underlying processes regulating photosynthetic gas exchange and WUE, limiting our ability to predict growth and survival responses of desert CAM plants to climate change. We monitored net CO2 assimilation (Anet), stomatal conductance (gs), and transpiration (E) rates periodically over 2 years in a natural population of the giant columnar cactus Carnegiea gigantea (saguaro) near Tucson, Arizona USA to investigate environmental and physiological controls over carbon gain and water loss in this ecologically important plant. We hypothesized that seasonal changes in daily integrated water use efficiency (WUEday) in this constitutive CAM species would be driven largely by stomatal regulation of nighttime transpiration and CO2 uptake responding to shifts in nighttime air temperature and humidity. The lowest WUEday occurred during time periods with extreme high and low air vapor pressure deficit (Da). The diurnal with the highest Da had low WUEday due to minimal net carbon gain across the 24 h period. Low WUEday was also observed under conditions of low Da; however, it was due to significant transpiration losses. Gas exchange measurements on potted saguaro plants exposed to experimental changes in Da confirmed the relationship between Da and gs. Our results suggest that climatic changes involving shifts in air temperature and humidity will have large impacts on the water and carbon economy of the giant saguaro and potentially other succulent CAM plants of warm desert environments.

Abstract:Stomata of most plants close to preserve water when the demand for CO2 by photosynthesis is reduced. Stomatal responses are slow compared with photosynthesis, and this kinetic difference erodes assimilation and water-use efficiency under fluctuating light. Despite a deep knowledge of guard cells that regulate the stoma, efforts to enhance stomatal kinetics are limited by our understanding of its control by foliar CO2. Guided by mechanistic modelling that incorporates foliar CO2 diffusion and mesophyll photosynthesis, here we uncover a central role for endomembrane Ca2+ stores in guard cell responsiveness to fluctuating light and CO2. Modelling predicted and experiments demonstrated a delay in Ca2+ cycling that was enhanced by endomembrane Ca2+-ATPase mutants, altering stomatal conductance and reducing assimilation and water-use efficiency. Our findings illustrate the power of modelling to bridge the gap from the guard cell to whole-plant photosynthesis, and they demonstrate an unforeseen latency, or 'carbon memory', of guard cells that affects stomatal dynamics, photosynthesis and water-use efficiency.

Abstract:Crassulacean Acid Metabolism (CAM) is an adaptation that temporally separates carbon uptake at night from photosynthesis during the day. CAM has evolved repeatedly across vascular plants, perhaps, in part, because it does not involve any novel gene functions and instead relies on regulatory modification of existing C3 pathways. The co-option of established processes and corresponding alteration of metabolic flux is likely to impact other primary metabolic pathways such as nitrogen metabolism and photorespiration. However, the majority of CAM research to date has focused on a handful of genes in the core CAM pathway. Understanding the complex downstream effects associated with the emergence of CAM is essential to explaining its convergent evolution as well as efforts to engineer carbon concentrating mechanisms in crops. In this study, we integrate genomic, transcriptomic, proteomic, and metabolomic data to compare primary metabolism between the CAM species Yucca aloifolia and closely related C3 species, Y. filamentosa. Despite substantial differences in metabolite abundance across multiple pathways, including citrate and nitrogen metabolism, we observe relatively few gene expression differences outside of the core CAM pathway. Additionally, we observe minimal correlation between protein abundance and mRNA expression, suggesting significant post-transcriptional regulation in CAM species. Our findings provide insights into the metabolic shifts associated with CAM evolution and highlight the complexity of its regulation at multiple biological levels.

Partitioning of mesophyll conductance for CO2 into intercellular and cellular components using carbon isotope composition of cuticles from opposite leaf sides

Abstract:We suggest a new technique for estimating the relative drawdown of CO2 concentration (c) in the intercellular air space (IAS) across hypostomatous leaves (expressed as the ratio cd/cb, where the indexes d and b denote the adaxial and abaxial edges, respectively, of IAS), based on the carbon isotope composition (δ13C) of leaf cuticular membranes (CMs), cuticular waxes (WXs) or epicuticular waxes (EWXs) isolated from opposite leaf sides. The relative drawdown in the intracellular liquid phase (i.e., the ratio cc/cbd, where cc and cbd stand for mean CO2 concentrations in chloroplasts and in the IAS), the fraction of intercellular resistance in the total mesophyll resistance (rIAS/rm), leaf thickness, and leaf mass per area (LMA) were also assessed. We show in a conceptual model that the upper (adaxial) side of a hypostomatous leaf should be enriched in 13C compared to the lower (abaxial) side. CM, WX, and/or EWX isolated from 40 hypostomatous C3 species were 13C depleted relative to bulk leaf tissue by 2.01-2.85‰. The difference in δ13C between the abaxial and adaxial leaf sides (δ13CAB - 13CAD, Δb-d), ranged from - 2.22 to + 0.71‰ (- 0.09 ± 0.54‰, mean ± SD) in CM and from - 7.95 to 0.89‰ (- 1.17 ± 1.40‰) in WX. In contrast, two tested amphistomatous species showed no significant Δb-d difference in WX. Δb-d correlated negatively with LMA and leaf thickness of hypostomatous leaves, which indicates that the mesophyll air space imposes a non-negligible resistance to CO2 diffusion. δ13C of EWX and 30-C aldehyde in WX reveal a stronger CO2 drawdown than bulk WX or CM. Mean values of cd/cb and cc/cbd were 0.90 ± 0.12 and 0.66 ± 0.11, respectively, across 14 investigated species in which wax was isolated and analyzed. The diffusion resistance of IAS contributed 20 ± 14% to total mesophyll resistance and reflects species-specific and environmentally-induced differences in leaf functional anatomy.

Abstract:Stomatal pores in plant leaves mediate CO uptake for photosynthesis and water loss via transpiration. Altered stomatal density can affect plant photosynthetic capacity, water use efficiency, and growth, potentially providing either benefits or drawbacks depending on the environment. Here, we explore, at different air humidity regimes, gas exchange, stomatal anatomy, and growth of Arabidopsis lines designed to combine increased stomatal density ( , ) with high stomatal sensitivity ( , ). We show that the stomatal density and sensitivity traits combine as expected: higher stomatal density increases stomatal conductance, whereas the effect is smaller in the high stomatal sensitivity mutant backgrounds than in the double mutant. Growth under low air humidity increases plant stomatal ratio with relatively more stomata allocated to the adaxial epidermis. Low relative air humidity and high stomatal density both independently impair plant growth. Higher evaporative demand did not punish increased stomatal density, nor did inherently low stomatal conductance provide any protection against low relative humidity. We propose that the detrimental effects of high stomatal density on plant growth at a young age are related with the cost of producing stomata; future experiments need to test if high stomatal densities might pay off in later life stages.
