Abstract:Background: Mastitis in dairy cattle is a highly prevalent infectious disease, caused by various pathogens, mainly Staphylococcu aureus and Escherichia Coli, considerable economic loss worldwide. Objectives: The aim of this study was to evaluate the in vitro activity of Herbal plants used against S. aureus and E. coli bacteria which are the causative agents of mastitis. Methods: Therefore, in this study we investigate the antimicrobial effect of plant to evaluate the in vitro antibacterial activity of squaw mint (Mentha pulegium L., Lamiaceae family), catnip (Nepeta cataria L., Lamiaceae), lemon balm (Melissa officinalis L., Lamiaceae), for mastitis treatment. Solutions prepared in fixed oils, against S. aureus and E. coli bacteria which are the main agents of mastitis. Isolation and antibiotic susceptibility analyses of milk samples taken from 100 subclinical mastitis dairy cows were performed. The antibacterial properties of the solutions were analysed by a disk diffusion method. Results: In the bacterial isolation, S. aureus was determined 97.7% and E. coli 53.5% positive of cows with mastitis. Antibacterial susceptibility test of the Lemon balm extract and essential oil showed maximum zone of inhibition against S. aureus 30 µl (23 mm), followed by 20 µl (19 mm), E. coli (19 mm) and 10 µl (5-7 mm), of the same extract against the Gram-positive bacteria. The ethanol extracts show the similar activity against the Gram-negative bacteria at 30, 20, and 10 µl (18-20 mm). Followed by S. aureus, when the zone areas for the susceptible solutions (Lemon balm, and essential oil) and the control group were compared, determined that there was little difference between for S. aureus and E. coli. Conclusions: This study hence indicated that in vitro cultured plantlets of lemon balm and peppermint oil can be used as the alternative method for production of mastitis and cheap source its precursor with antimicrobial activities.
Evaluation of a Postmilking Teat Disinfectant Containing a Phenolic Combination for the Prevention of Mastitis in Lactating Dairy Cows

Abstract:A trial was conducted for 12 months in a herd of 120 Holstein cows in order to determine the efficacy of a teat disinfectant, which contained a phenolic combination, for the prevention of bovine intramammary infections during lactation. Postmilking teat disinfection was compared to a negative control using a split-udder experimental design. The percentage of quarters newly infected by mastitis pathogens was 45% lower in mammary glands with teats that had been dipped in the experimental teat disinfectant after milking than it was in undipped controls. New infections caused by Streptococcus uberis, Staphylococcus aureus, coagulase-negative Staphylococcus species, and Corynebacterium bovis were significantly lower in mammary glands with teats that had been dipped in the experimental teat disinfectant than in undipped controls. No statistical differences in the incidence of clinical mastitis between treatment groups were observed. No irritation or chapping of teats dipped in the experimental teat disinfectant were observed. The results of this study suggest that the experimental teat disinfectant containing a phenolic combination is an effective postmilking teat disinfectant for use in the prevention of new intramammary infections by both contagious and environmental mastitis pathogens.

Abstract:One hundred and nine Holstein cows were divided at random into two groups: a trial group and a control group. Teat dipping with Dianxiaoling (containing 0.5% available iodine) after milking and dry cow therapy with TA-125 (mainly containing benzathine penicillin and cloxacillin) at drying off were applied in the trial group of 53 cows. The other 56 cows in the control group were not dipped, but their quarters were infused following the last milking with solutions of penicillin salt and dihydrostreptomycin. After a 22-month experiment, the incidence of subclinically mastitic quarters in the trial group was reduced about 50% in comparison with that in the control group (P < 0.01). The incidences of clinical mastitis and of blind teats were also significantly reduced.
Chemical Composition, Antioxidant and Antibacterial Activity of Two Different Essential Oils Against Mastitis Associated Pathogens

Abstract:Abstract Mastitis is one of the most common and costly diseases affecting dairy cows worldwide. Since antibiotic resistance has become a global threat to both animal and human health, it is becoming more urgent to continuously search for new therapeutical alternatives for the control and treatment of bovine mastitis. Hence, our research aimed to test the therapeutic use of two essential oils (EOs) based on their chemical composition, antibacterial and antioxidant potential. The present study was conducted by collecting milk samples from the cows diagnosed with clinical or subclinical mastitis with the aim of isolating and identifying bacterial strains. The antioxidant potential of essential oils of Menthae piperitae (MP) and Melissa officinalis (MO) was evaluated in several in vitro assays. In the MP EO, a total of 38 compounds were identified, with menthol as the dominant compound, whereas in MO EO 51 compounds were identified. Furthermore, the values of minimal inhibitory concentrations (MICs) and minimal bactericidal concentrations (MBCs) have been used to quantitatively measure the antibacterial activity of each essential oil. In accordance with which, MP EO samples exhibited a higher degree of antibacterial activity than MO EO. Thus, EOs have been shown to be promising alternatives to antibiotics because of their availability, biodegradability, and lower risk of side effects as compared with conventional, antimicrobial treatment. Nevertheless, further clinical studies are needed to test the potential role of EOs in treating mastitis in dairy cows.

Abstract:The aim of this investigation was to study in vitro antibacterial activity of essential oils (EOs) ( Citrus bergamia Risso & Poiteau, Melaleuca alternifolia and Chenopodium botrys) on pathogenic strains isolated in milk samples from mastitic sheep. The in vitro antibacterial activity showed that EOs induced higher bacterial inhibition against pathogens isolated in mastitic milk (Staphylococcus xylosus , Staphylococcus epidermidis, Streptococcus salivarius, Enterococcus faecium, Streptococcus agalactiae and Pseudomonas aeruginosa) than common antimicrobial agents used in veterinary medicine, with Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) values significantly lower, excepted for P. Aeruginosa . The antibacterial activity of EOs has been attributed to the chemical composition of each phyto-complexes, constituted by monoterpenes, sesquiterpenes and their associated alcohols, identified by GC-MS analysis, with a synergic action in the bacterial cells. The results obtained confirmed in vitro antimicrobial activity of these EOs and their potential use in the treatment of sheep mastitis infection.

Abstract:Both Gram-negative and Gram-positive bacteria have recently developed antibiotic resistance to treatments for bovine mastitis, creating a serious concern for public and animal health. The objective of this study was to analyse in vitro microbicidal activity of tea tree oil, thymol and carvacrol (composed of oregano and thyme essential oils) on bacteria isolated from clinical mastitis. Field isolates and ATCC strains of the Staphylococcus spp, Streptococcus spp, Escherichia coli, Klebsiella pneumoniae, and Candida albicans genera were analysed. The agar diffusion technique was used to test bactericidal susceptibility and plate microdilution was utilized to determine the minimum inhibitory, bactericidal, and fractional inhibitory concentrations. Thymol alone and the combinations of thymol-carvacrol and thymol-TTO obtained the highest inhibition diameters for Gram-negative bacteria, while for Gram-positive bacteria and C. albicans , thymol and the combination thymol-carvacrol obtained the highest indices. TTO, thymol, and carvacrol had MIC values of 1.56–25 mg/ml, 0.05–0.4 mg/ml, and 0.02–0.2 mg/ml, respectively. CMB results for the Gram-negative and gram-positive groups were 0.39–0.78 mg/ml, and for C. albicans , 0.78–1.56 mg/ml. Results for the fractional inhibitory concentrations show that the TTO+thymol and thymol+carvacrol combinations had additive activity against groups of Gram-negative bacteria and C. albicans . These natural components, evaluated individually and in combinations, have an effectiveness above 70%.

Abstract:During the current investigation, eight essential oils (EOs) were tested for their antimicrobial activity against six species, belonging to the genus of staphylococcus, multi-resistant to antibiotics (S. epidermidis, S. cohni, S. wareneri, S. scuiri, S. chromogenes, S. pasteuri), three methicillin-resistant Staphylococcus aureus strains (MRSA) and two strains of Escherichia coli, producing extended-spectrum β-lactamase (ESBL) responsible for bovine mastitis. Our results indicated that the antimicrobial activities of eight EOs varied significantly among the types of EOs and bacterial species. Thymus capitatus and Trachyspermum ammi EOs display important antibacterial activity against all tested strains, with the inhibition zone diameters situated between 20 and 45 mm, while EOs of Artemisia absinthium, Eucalyptus globulus, Eucalyptus camaldulensis, Myrtus communis and Mentha pulegium exerted an intermediate activity. For Cymbopogon citratus, this effect depends on bacteria species. In fact, an important effect was observed against S. warneri, S. epidermidis, S. cohenii, S. pasteuri and MRSA (EC 39+) strains. In addition, the important lytic effect was observed against MRSA strains, showing that Gram-positive bacteria were more sensitive to T. capitatus EO than Gram-negative ones. Concerning the characterization of the mode action of T. capitatus, experiments of kill-time, bacteriolytic, loss of salt tolerance and loss of cytoplasmic material showed that the used EO was able to destroy cell walls and membranes followed by the loss of vital intracellular materials. In addition, it inhibits the normal synthesis of DNA, causing the bacterial death of E. coli and MRSA strains. This study shows the potential of using of EOs, particularly T. capitaus, to inhibit the growth of Gram-positive and Gram-negative bacteria multi-resistant to antibiotics causing bovine mastitis.

Abstract:In the present investigation, we determined the in vitro antimicrobial activity of eight essential oils (EOs) and three medium-chain fatty acids (MCFAs) alone and in combination against Staphylococcus aureus ATCC 700698, Klebsiella pneumoniae ATCC 700603, and E. coli FcW5. The interactions between EOs and MCFAs were determined by fractional inhibitory concentration indices. Moreover, mode of action of selected bioactive components was studied by changes in bacterial surface charge, morphology, and membrane integrity assays. Among EOs, carvacrol (CAR), trans-cinnamaldehyde (TC), and thymol (TM) showed strong antimicrobial activity. In combination study, CAR+OA (octanoic acid), CAR+DA (decanoic acid), and TM+OA were observed as the most significant (P≤0.05) which were also confirmed through time-kill plots. Based on these results, CAR+OA were found to be most efficacious in terms of killing time (P≤0.05). Changes in the surface charge, morphology, and membrane integrity upon the combined treatment of CAR+OA were also observed, which ultimately leads to cell death. Results suggest that CAR+OA when used in combination offer a significant (P≤0.05) additive antimicrobial activity against the selected pathogenic bacteria. Therefore, these natural bioactive molecules could be interesting alternatives to conventional therapy for the control of mastitis caused by multi-drug-resistant pathogens in bovine animals to ensure the milk safety.

Abstract:Nisin is an antimicrobial polypeptide produced by Lactococcus lactis and is believed nontoxic to humans. The objective of this study was to evaluate a nisin-based formulation for the treatment of bovine clinical mastitis in lactating dairy cattle. A total of 92 cows with 107 clinically mastitic quarters were randomly assigned to nisin- (48 cows with 51 quarters) and gentamicin (GM)-treated (44 cows with 56 quarters) groups. In the nisin-treated group, cows received an intramammary infusion of nisin at a dose of 2,500,000 IU; in the GM-treated group, intramammary infusion of GM was administered at a dose of 0.8 g. Results indicated that nisin offered a clinical cure rate similar to GM (90.2 vs. 91.1%) and no difference in bacteriological cure rate than GM-treated group (60.8 vs. 44.6%, respectively). Proportion of the quarters with milk somatic cell counts <500,000 cells/mL was not different in the nisin-treated group (50.0 and 47.8%) compared with the GM-treated group (33.3 and 37.3%) 1 and 2 wk after treatment. Of 17 Staphylococcus aureus isolates, 82.5% were resistant to penicillin, and 35.3% to GM, but none of them to nisin. Nisin therapy eliminated 54.5% (6 of 11) of S. aureus IMI, whereas GM eliminated 33.3% (2 of 6). Nisin in milk (4.5 +/- 0.8 IU/mL) was detected only at 12 h following intramammary infusion, which was much lower than the upper limit (500 mg/mL) allowed as preservative in milk by the China authority. Because of its efficacy in the treatment of bovine clinical mastitis, especially resistant Staph. aureus-caused IMI, as well as its safety in humans, nisin deserves further study to clarify its effects on mastitis caused by different mastitis pathogens on a larger scale.
Distribution of Major Mastitis-Causing Bacteria and Common Antimicrobial Drugs in Milk.

Abstract:This study was designed to investigate variations in major constituents of milk during milking in dairy cows,and in vitro distribution of major mastitis-causing bacteria and common antimicrobial drugs in milk.Results indicated that the concentration of fat gradually increased with the progress of milking,and was significantly higher in the milk collected during the last period than those collected during other period of milking;the concentration of milk protein and lactose changed little.Staphylococcus aureus and Escherichia coli were found largely in upper portion of the milk,and the colony forming units(CFU)/mL of both S.aureus and E.coli were significantly higher than those in middle and lower portions(P0.01).Antibacterial drug benzylpenicillin sodium,gentamycin sulfate,ciprofloxacin lactate,ceftiofur sodium,lincomycin hydrochloride,erythromycin lactobionate,tetracycline hydrochloride,ampicillin sodium were found to have significantly higher concentration in lower portion of milk than in middle and upper portions.Nisin Z was distributed more evenly but a little higher concentration in the upper portion.It is suggested that bacteria getting entrance into teat canal may float up through attaching to fat globes and invade the deep tissues.Bacteria and antibacterial drugs may not contact each other sufficiently due to their contra-direct movements,resulting in poor response of mastitic cases to antibacterial therapy.

Abstract:Mastitis is an inflammatory reaction of the udder caused by microorganisms, chemical or mechanical injuries. Bacterial mastitis, caused by several types of bacteria, is divided into two main groups: contagious mastitis caused by Staphylococcus aureus, Streptococcus agalactiae, Streptococcus dysgalactiae, Mycoplasma bovis and mastitis caused by the ubiquitous bacteria Escherichia coli, Pseudomonas aeruginosa or Proteus spp. Currently, instead of using antibiotics, new strategies are being sought to reduce this clinical health problem. The aim of this study is to determine the sensitivity of bacterial strains causing mastitis to antibiotics and essential oils of oregano (Origanum compactum, Origanum majorana) and thyme (Thymus serpyllum). Mammary gland secretion samples, obtained by mechanical and manual methods, from dairy cows from the Kalesija and Gradačac areas were used. A total of 200 samples from dairy cows in different periods of lactation and age were examined. Mastitis control included determining somatic cells count in milk with the California mastitis test, or counting somatic cells on the Fossomatik apparatus, after which all samples with an increased number of somatic cells were microbiologically tested. Based on the obtained results, it can be concluded that there are justified reasons why the use of essential oils should be considered as an alternative to the use of antibiotics in the therapy of animals suffering from mastitis.

Abstract:The essential oils of Lippia citriodora(Ort.) and Lippia origanoides(Kunth) have shown antimicrobial activity associated with mastitis. The objective of this study was to evaluate its ecotoxic effect withthe Artemia salinabioassay and the prevention of mastitis through an in vivotest in cattle (n=20) with a product based on these oils using a conventional product as a control. Contact hypersensitivity, the effect on somatic cells, and residuality in milk samples were evaluated. The results of the Artemia salinabioassay were 10.05 and 19.36 (μg/mL) respectively. No negative effects or contact hypersensitivity were observed, and no residual metabolites were found in post-test milk. The somatic cell count showed 75% effectiveness in the prevention of mastitis with essential oils compared to 62.5% with the conventional product. The evaluated formulation could be used in the prevention of bovine mastitis safely, further investigation is required.

Abstract:Staphylococcus aureus, particularly multi-drug resistant strains, presents significant challenges in dairy farming due to its role in causing bovine mastitis, which leads to substantial economic losses and limited treatment options. Seeking alternative therapies, we investigated the potential of a topical formulation derived from the medicinal herb Salvia officinalis to combat S. aureus growth and biofilms associated with bovine mastitis. Through systematic extraction in different solvents and fractionation by column chromatography, we isolated and identified three key multicyclic terpenoids-ferruginol, sugiol, and sclareol-exhibiting significant antimicrobial activity. The formulation effectively inhibited biofilm formation, with minimum inhibitory concentration (MIC) values ranging from 0.09 to 0.74 mg ml-1 against clinical S. aureus strains, comparable to or lower than those of the pure compounds. Moreover, it displayed robust anti-adhesive properties, reducing biofilm formation by 20%-79% at subinhibitory concentrations. Furthermore, the formulation successfully disrupted pre-existing biofilms, achieving reductions ranging from 30% to 82%. Cytotoxicity assays confirmed the safety of the formulation on mammary epithelial cells, with cell viability maintained at 100% at MIC. Our findings underscore the therapeutic potential of Sa. officinalis-derived compounds in managing bovine mastitis caused by S. aureus, emphasizing their antimicrobial efficacy and safety profile.
Evaluation of the effectiveness of some essential oils against zoonotic methicillin-resistant Staphylococcus aureus isolated from dairy products and humans

Abstract:Objective: Staphylococcus aureus (S. aureus) is a zooanthroponotic, nosocomial, and community-associated pathogen that threatens livestock management and even public health. The goal of this investigation was to clarify the role of S. aureus in zoonotic illnesses. Besides that, a novel trial was conducted in the current Egyptian study using oil extracts such as cactus oil, tea oil, geranium oil, and thyme oil to demonstrate the susceptibility of methicillin-resistant S. aureus (MRSA) isolates to these organic oils in response to the alarming global concern regarding the decreased susceptibility of S. aureus to known antibiotics, which exacerbates control and treatment protocols. Materials and methods: A total of 110 samples (45 raw cattle milk samples, 35 Karish cheese samples, and 30 human sputum samples) were collected. The bacterium was identified via traditional culturing methods, Gram staining, and the application of several biochemical tests. After that, various kinds of known commercial antibiotics were used to detect the antimicrobial susceptibility (AMS) of the obtained isolates. Furthermore, conventional polymerase chain reaction (PCR) testing was performed to identify S. aureus (nuc gene) and MRSA (mecA gene), with further application of multiplex PCR for screening of all the obtained isolates for vancomycin resistance via targeting vanA, vanB, and vanC genes. Finally, the agar gel diffusion method was performed to assess the antibacterial activity of four plant extracts (cactus oil, tea oil, geranium oil, and thyme oil) against the obtained MRSA. Results: The culturing method revealed S. aureus positivity in raw cattle milk (13.33%), in Karish cheese (28.57%), and in human samples (20%). The obtained isolates showed mainly resistance to amoxicillin-clavulanic and ampicillin antibiotics, while the dairy samples showed further resistance against ceptaxime and an intermediate reaction against erythromycin. On the molecular side, PCR positivity was present in human samples (10%), raw cow milk (13.33%), and Karish cheese (14.29%). Nine of the fourteen PCR isolates were methicillin-resistant S. aureus (MRSA) isolates. Comparing the four oil extracts against the acquired MRSA isolates, cactus oil extract proved to be the most effective. Conclusion: The study's results are highly promising as they support the notion that certain essential oils possess strong antimicrobial properties against zoonotic S. aureus, thereby reducing the excessive use of antibiotics in veterinary and medical settings.
Susceptibility patterns of Escherichia coli and streptococcal isolates from bovine mastitis cases to antibiotics and selected South African plant extracts with known antibacterial activities

Abstract:Bovine mastitis, an inflammatory condition affecting the mammary glands of dairy cattle, poses a significant economic burden on the dairy industry. Staphylococcus species, Streptococcus species and Escherichia coli are common pathogens. Managing mastitis typically involves antibiotics, but antimicrobial resistance (AMR) is a growing concern, promoting further research exploring alternative treatments including plant extracts. This study aimed to determine the antibiotic resistance of E. coli and streptococcal isolates from mastitis cases, and to investigate four indigenous South African plants for their selective antibacterial activities against these pathogens. The antimicrobial susceptibility of the bacterial strains was determined using a standard disc diffusion method. Minimum inhibitory concentration (MIC) values of acetone and ethanol extracts of Searsia lancea, Indigofera frutescens, Erythrina caffra and Antidesma venosum were determined against the bacterial isolates using a serial microdilution assay, and cytotoxicity was also investigated. The results showed that 82.14% of the clinical isolates tested were resistant to at least one antimicrobial agent used, and 52.17% of the antibiotic resistant isolates were multidrug resistant. All plant extracts had antibacterial activity against all the bacterial isolates, but S. lancea demonstrated higher efficacy compared to other plants. The MIC values ranged from 0.01 to 2.50 mg/mL, with the lowest range obtained with the acetone extract of S. lancea (0.01 to 0.57 mg/mL). Furthermore, the extracts were relatively non-cytotoxic to bovine dermis and Vero cells, with the highest mean selectivity index value of 25.70 recorded with S. lancea . This study highlights the growing concern of AMR in livestock management, and demonstrates the promising therapeutic potential of the selected plant species, particularly S. lancea , in treating bovine mastitis. Further exploration of S. lancea is recommended to develop novel alternative or complementary formulations for mastitis management.

Assessment of the effects of sugar cane plantation burning on daily counts of inhalation therapy.

Abstract:This study was designed to evaluate the association between sugar cane plantation burning and hospital visits in Araraquara in the state of São Paulo, Brazil. From June 1 to August 31, 1995, the daily number of visits of patients who needed inhalation therapy in one of the main hospitals of the city was recorded and used as health impairment estimation. Sedimentation of particle mass (the amount of particles deposited on four containers filled with water) was measured daily. The association between the weight of the sediment and the number of visits was evaluated by means of Poisson regression models controlled for seasonality, temperature, day of the week, and rain. We found a significant and dose-dependent relationship between the number of visits and the amount of sediment. The relative risk of visit associated with an increase of 10 mg in the sediment weight was 1.09 (1-1.19), and the relative risk of an inhalation therapy was 1.20 (1.03-1.39) on the most polluted days (fourth quartile of sediment mass). These results indicate that sugar cane burning may cause deleterious health effects in the exposed population.

Abstract:Bovine subclinical mastitis is an inflammation of the mammary gland caused by bacterial intramammary infection, accounting for large economic losses. Treatment of subclinical mastitis is not suggested for lactating cows due to the risk of milk contamination. The objectives of this study were to evaluate an antimicrobial peptide, nisin, in the treatment of subclinical mastitis in lactating cows. A total of 90 lactating Holstein cows with subclinical mastitis were randomly divided into nisin-treated (n = 46) and control (n = 44) groups. In the nisin-treated group, cows received an intramammary infusion of nisin at a dose of 2,500,000 IU once daily for 3 days while the control cows received no treatment. Milk samples were collected from the affected mammary quarters before treatment and 1 and 2 weeks after treatment for analyses of bacteria, somatic cells, and N-acetyl-beta-D-glucosaminidase (NAGase). Results indicated that nisin therapy had bacteriological cure rates of 90.1% for Streptococcus agalactiae (10 of 11), 50% for Staphylococcus aureus (7 of 14), 58.8% for coagulase-negative staphylococci (7 of 17), and 65.2% for all cases (30 of 46). Meanwhile, only 15.9% (7 of 44) of untreated cows spontaneously recovered. NAGase activity in milk samples and the number of mammary quarters with a milk somatic cell count of > or =500,000/ml were significantly decreased after nisin treatment while no significant changes took place in the control group. Because of its therapeutic effects on bovine subclinical mastitis, as well as its safety in humans, nisin deserves further study to clarify its effects on mastitis caused by different pathogens.
