Abstract:Ethanol, potassium metabisulfite, formaldehyde and hydrogen peroxide were tested for tumor-promoting activity in a two-stage stomach carcinogenesis experiment. Male outbred Wistar rats were given N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in the drinking water (100 mg/liter) and a diet supplemented with 10% sodium chloride for 8 weeks. Thereafter, they were maintained on drinking water containing either 10% ethanol, 1% potassium metabisulfite, 0.5% formalin (formaldehyde) or 1% hydrogen peroxide for 32 weeks and then sacrificed for necropsy and histological examination. In the pylorus of the glandular stomach, potassium metabisulfite and formaldehyde significantly increased the incidence of adenocarcinoma after initiation with MNNG and sodium chloride. Hydrogen peroxide did not enhance the tumor yield, and ethanol showed a tendency to decrease neoplastic development. In the forestomach the incidence of squamous cell papilloma was significantly increased in the groups given hydrogen peroxide or formaldehyde, irrespective of prior initiation. Duodenal adenocarcinoma was induced by the initiation alone (10%) and the incidence was not affected by the subsequent treatments. The results indicate that potassium metabisulfite and formaldehyde both exert tumor-promoting activity in the rat glandular stomach.
New records of pathogenic bacteria in different species of fleas collected from domestic and peridomestic animals in Spain. A potential zoonotic threat?

Abstract:Climate change is causing many vectors of infectious diseases to expand their geographic distribution as well as the pathogens they transmit are also conditioned by temperature for their multiplication. Within this context, it is worth highlighting the significant role that fleas can play as vectors of important pathogenic bacteria. For this purpose, our efforts focused on detecting and identifying a total of 9 bacterial genera (Rickettsia sp.; Bartonella sp.; Yersinia sp.; Wolbachia sp., Mycobacterium sp., Leishmania sp., Borrelia sp., Francisella sp. and Coxiella sp.) within fleas isolated from domestic and peridomestic animals in the southwestern region of Spain (Andalusia). Over a 19-months period, we obtained flea samples from dogs, cats and hedgehogs. A total of 812 fleas was collected for this study. Five different species were morphologically identified, including C. felis, C. canis, S. cuniculi, P. irritans, and A. erinacei. Wolbachia sp. was detected in all five species identified in our study which a total prevalence of 86%. Within Rickettsia genus, two different species, R. felis and R. asembonensis were mainly identified in C. felis and A. erinacei, respectively. On the other hand, our results revealed a total of 131 fleas testing positive for the presence of Bartonella sp., representing a prevalence rate of 16% for this genus identifying two species B. henselae and B. clarridgeiae. Lastly, both Y. pestis and L. infantum were detected in DNA of P. irritans and C. felis, respectively isolated from dogs. With these data we update the list of bacterial zoonotic agents found in fleas in Spain, emphasizing the need to continue conducting future experimental studies to assess and confirm the potential vectorial role of certain synanthropic fleas.

Abstract:Background: Fleas are ectoparasitic insects with holometabolous development. It has a hematophagous habit with mouthparts adapted to sting and suck its hosts. There are about 3000 species in the world, ∼61 in Brazil, and 19 in Rio Grande do Sul state. The objective of the research is to catalog the diversity of fleas recorded in the state, their respective hosts, and endosymbionts. Materials and Methods: To this end, a search was carried out in the scientific literature, from articles, books, to abstracts submitted to congresses. Results: The 19 species of fleas occurring in Rio Grande do Sul are divided into 7 families and 10 genera. These ectoparasites, in addition to being found in the environment, were associated with 10 different families of hosts in Rio Grande do Sul, and on the endosymbiont, agents found associated with fleas, there were 7 different species. The main agents researched in the state are Rickettsia spp. and Bartonella spp. The relationships between parasites, hosts, environment, and etiological agents present different scenarios, whether anthropized or conserved, but unknown. Sometimes, this overlap, a factor that aggravates the possibility of spillovers, either from cosmopolitan fleas in these conserved areas, or from their endosymbionts. Conclusion: Thus, it is important to characterize the environment so that the complexities of each location are known for the adoption of environmental and public health policies in each case. The challenges are extensive, but necessary in view of the One Health perspective.

Abstract:Vector-borne diseases account for nearly 20% of all globally recognised infectious diseases. Within the spectrum of flea-borne pathogens, Bartonella and Rickettsia bacteria are prominent, contributing to the emergence and resurgence of diseases on a global scale. This study investigates the presence of species of Bartonella and Rickettsia harboured by fleas collected from wild rodents in northwestern Argentina (NWA). A total of 28 fleas from three genera and seven species were assessed. DNA of Bartonella and Rickettsia spp. was found in 12 fleas (42.8%). Phylogenetic analysis of concatenated sequences of gltA and rpoB genes showed the presence of Bartonella quintana in eight fleas of two species, Craneopsylla minerva minerva and Polygenis acodontis . Phylogenetic analysis of concatenated sequences of gltA , ompA and ompB genes identified Rickettsia felis in ten fleas of five species, C. m. minerva , P. acodontis , Polygenis bohlsi bohlsi , Polygenis byturus and Tiamastus palpalis . These bacterial species mark the first report in all flea species studied. This study represents the first survey of flea-borne bacteria for NWA. The results provide information to address strategies for the control and prevention of bartonellosis and rickettsiosis that could have an impact on public health in one of the geographical areas of Argentina with the highest incidence of infections transmitted to humans by ectoparasites.

Abstract:Fleas (Siphonaptera) are ectoparasitic hematophagous insects responsible for causing bites and itchy skin conditions in both humans and animals. Furthermore, they can act as vectors of different pathogens of a wide variety of diseases worldwide, including bartonellosis, rickettsiosis, and bubonic plague. Accurate identification of fleas is necessary for the study of their epidemiology, prevention, and control. In addition to traditional morphological classification approaches and molecular biology techniques, geometric morphometrics is increasingly proving to be a useful complementary tool for discriminating between Siphonaptera taxa. With the objective of determining the capacity of this technique to identify and differentiate synanthropic fleas, a principal component analysis was carried out on populations of Ctenocephalides felis, Pulex irritans, and Archaeopsylla erinacei collected in distinct regions of Andalusia (Spain). The analysis carried out on 81 male and female specimens revealed factorial maps that allowed the differentiation of the populations under study, with only partial overlaps that did not prevent their correct identification. Global size differences were also detected, with a slightly larger size in P. irritans males and a bigger size in A. erinacei females. Therefore, the present study emphasizes the role of geometric morphometrics as a useful complementary technique in taxonomic studies of arthropods, especially in the case of flea specimens lacking representative morphological features.

Abstract:Fleas, one of the most significant ectoparasites, play a crucial role as vectors in spreading zoonotic diseases globally. The Qinghai Province, as part of the Qinghai-Tibet Plateau, is one of the provinces in China with the largest number of flea species. In this study, we characterized the microbial communities of eighty-five adult fleas, belonging to nineteen species within four families (Ceratophyllidae, Ctenophthalmidae, Leptopsyllidae, and Pulicidae). We identified a total of 1162 unique operational taxonomic units at the genus level, with flea-borne pathogens such as Wolbachia , Bartonella , Rickettsia being the members of top abundant taxa. Except for comparison between Ctenophthalmidae and Leptopsyllidae families, the analyses of both alpha- and beta- diversity indicators suggested that bacterial diversity varied among flea families. This could be attributed to flea phylogeny, which also influenced by their geographical sites and animal hosts. Results of Linear discriminant analysis effect size (LEfSe) indicated that 29 genera in Ceratophylloidea, 11 genera in Ctenophthalmidae, 15 genera in Leptopsyllidae, and 22 genera in Pulicidae were significantly responsible for explaining the differences among the four flea families (linear discriminant analysis score > 2, P < 0.05). Phylogenetic Investigation of Communities by Reconstruction of Unobserved States (PICRUSt2) analyses showed that the functional pathways varied significantly across flea families, which was supported by the significant correlation between the functional pathways and the microbial communities.

Composition and diversity of Culicoides biting midges (Diptera: Ceratopogonidae) in rural and suburban environments of Algeria

Abstract:Culicoides biting midges are well known biological vectors of several arboviruses causing more than 100 veterinary and medical diseases worldwide. In Algeria, bluetongue virus, which is transmitted by Culicoides midges, is responsible for one of the most critical insect-borne diseases of sheep. For example, this disease caused clinically severe morbidity of about 2,661 confirmed cases out of 21,175 susceptible sheep in Algeria. This study compared the abundance, richness, and diversity of Culicoides species in rural and suburban environments of semi-arid regions in North Africa. It examined the potential influence of the bio-climatic factors on the variation of population sizes and the elevation distribution of biting midges. Specimen collection was carried out from June to September during two successive years (2018 and 2019) using CDC light traps installed at 15 sites in different environments. Culicoides specimens were dissected, slide-mounted, and morphologically identified using the interactive identification key IIKC and various standard morphological criteria. A total of 1,046 Culicoides specimens (1,024 females and 22 males) were trapped and classified into 22 species, belonged to nine distinct subgenera. Two new species records for Algeria and even North Africa are reported: Culicoides albicans (Winnertz, 1852) and Culicoides nubeculosus (Meigen, 1830). Culicoides newsteadi Austen, 1921 (51.6%) was the dominant species, and it was followed by Culicoides punctatus (Meigen, 1804) (16.3%) and Culicoides odiatus Austen, 1921 (11.5%). These three species, comprising 80% of the collected Culicoides, were the most abundant both outside and inside livestock stables in rural and suburban environments. Species diversity was similar in the two settings, with a slight increase in suburban environments. None of the Culicoides species encountered correlated significantly with the climatic factors (mean temperature, precipitation, and relative humidity). Elevation was the most determinant environmental parameter that affected the abundance and distribution of Culicoides midges in the semi-arid and sub-humid areas studied. The maximum distribution of Culicoides species was detected at mid elevations (400‒800 m). Using a modeling approach, we explored for the first time the variation of composition and diversity in Culicoides communities within different climatic regions, environments and livestock settings in Algeria. This survey deepens our understanding of the relationships among environmental factors, abundance, diversity, and geographic distribution of Culicoides. This is a crucial step to assess the epidemiological situation of the diseases transmitted by these biting midges and to allow mitigation of the associated risks.
A new canid host for subcutaneous ticks: A report from Algeria in an African golden wolf (Canis lupaster)

Abstract:Background: Ticks are hematophagous ectoparasites that feed on the skin surface of their host. Subcutaneous localizations of ticks have predominantly been associated with red foxes, documenting new cases still brings new and potentially valuable data for tackling this unusual situation. The objective of this study is to report the presence of subcutaneous ticks in a new host and on a new continent. Methods: Between February 2022 and September 2023, 47 road-killed animal carcasses belonging mostly to carnivorous species were subjected to a comprehensive parasitological necropsy. Upon detection of subcutaneous tick, the protuberance was excised, dissected, and preserved in labeled vials containing ethanol. Identification of the subcutaneous tick was carried out at the species level. Results: A single nodule of subcutaneous ticks was found in an African golden wolf (Canis lupaster) collected from Northern Algeria; no other cases of unusual tick locations were observed. Based on morphological criteria the tick was identified as Ixodes ricinus / I. inopinatus. Conclusion: This study represents the first documentation of subcutaneous tick in an African golden wolf (Canis lupaster), thereby broadening the host spectrum for this atypical location. Furthermore, it underscores North Africa as a new geographical area where this phenomenon occurs.
On-host flea phenology and flea-borne pathogen surveillance among mammalian wildlife of the pineywoods of East Texas

Abstract:Flea-borne diseases are endemic in Texas, U.S.A., with an increasing incidence of flea-borne typhus and cat scratch disease. Knowledge of flea natural history could provide information to protect public health, yet many knowledge gaps remain outside of plague-endemic regions. Our objective was to characterize seasonal activity patterns of fleas on common mammalian wildlife species and test fleas and wildlife for Rickettsia and Bartonella pathogens. We performed one year of monthly trapping for rodents and medium-sized mammals in a national forest with high recreational use and urban encroachment in East Texas. From 90 mammal captures representing seven species, 101 fleas were collected representing Polygenis spp., Ctenocephalides felis, and Orchopeas species. Virginia opossums (Didelphis virginianus) hosted 99% of the collected fleas (100 fleas) and a single flea was on an eastern woodrat (Neotoma floridana). Flea infestation prevalence of opossums was 79% (23/29). Mean flea abundance was 4.39 fleas, with intensity peaking in spring. One cat flea removed from an opossum was positive for Bartonella henselae. Furthermore, we identified tissue or blood of four raccoons (Procyon lotor) and one golden mouse (Ochrotomys nuttalli) positive for Rickettsia amblyommatis. These findings provide an ecological basis for the maintenance of vectors and pathogens from sylvatic settings.

Abstract:Human papillomaviruses cause the most common sexually transmitted infection in the world and are responsible for nearly all cases of cervical cancer. Genital human papillomavirus infection can be divided into low-risk infections (causing genital warts) and high-risk infections (causing cervical intraepithelial neoplasia, and cervical and other cancers). Exposure to human papilloma- virus typically produces a sexually transmitted infection that may progress to a clinically apparent process, such as genital warts and cervical intraepithelial neoplasia lesions of the lower genital tract. Although most human papillomavirus infections resolve spontaneously within two years, some high-risk infections persist and are considered cancer precursors. Risk factors for persistent infection include multiple sex partners, sex at an early age, history of sexually transmitted infections, and smoking. Condom use is only partially protective against human papillomavirus infection. The two human papillomavirus vaccines are most effective if given to girls before the onset of sexual activity.
Specificity of fleas associated with opossums in a landscape gradient in the Paranaense Rainforest Ecoregion

Abstract:The degree of host specificity of fleas varies from highly specific (monoxenous) to opportunistic (polyxenous). Specific parasite-host associations can be observed among some flea families and tribes and mammalian orders, such as fleas of the tribe Tritopsyllini (Ctenophthalmidae, Doratopsyllinae) and opossums (Didelphimorphia). Specimens of the family Didelphidae are common hosts of fleas of the genus Adoratopsylla (Ewing, 1925). In northern Argentina, Adoratopsylla (Adoratopsylla) antiquorum antiquorum (Rothschild, 1904) and Adoratopsylla (Tritopsylla) intermedia intermedia (Wagner, 1901) were recorded preferentially parasitizing opossums. In order to study parasite/host relationships, fleas were collected from opossums captured in different environments in the Paranaense Rainforest ecoregion, northern Misiones province between 2016 and 2018. A total of 287 fleas were collected from 110 opossums. The fleas were identified as Pulicidae: Ctenocephalides felis felis (Bouché, 1835); Rhopalopsyllidae: Polygenis (Polygenis) rimatus (Jordan, 1932), Polygenis (Polygenis) roberti roberti (Rothschild, 1905); Ctenophthalmidae: A. (T.) i. intermedia, A. (A.) a. antiquorum, Adoratopsylla (Adoratopsylla) antiquorum ronnai (Guimarães, 1954). We report for the first time in Argentina a male specimen of A. (A.) a. ronnai collected on Didelphis albiventris (Lund, 1840) (Didelphidae), and male and female specimens of P. (P.) r. roberti collected on Didelphis aurita (Wied-Neuwied, 1826), and we describe the expansion of the geographic and host distribution of fleas to the Paranaense Rainforest ecoregion. Our records reinforce the specific association between Adoratopsylla and Polygenis fleas and didelphid opossums, mentioned in the literature for the Atlantic Forest ecoregion in Brazil. Furthermore, we observed that flea communities in opossums change in a landscape gradient, with invasive cat fleas and euryxenous fleas common in anthropized areas, and endemic fleas common in natural areas. Our results underline the importance for public health and veterinary medicine the analysis of flea circulation between wild and urban environments due to the risk of pathogen transmission.
Detection of Rickettsia raoultii in Vermipsylla alakurt-Like Fleas of Sheep in Northwestern China

Abstract:Introduction: To date, a total of 2574 validated flea species have been discovered. Vermipsyllidae is a family of fleas that comprises at least eight species. Vermipsylla is a genus of the family Vermipsyllidae within the order Siphonaptera of fleas. Here a novel Vermipsylla species was described, and rickettsial agent was also detected in it. Methods: A total of 128 fleas were collected directly from 260 pastured sheep in China. Of these, eight representative fleas (four males and four females) were identified by key morphological features. Meanwhile, 120 flea DNAs, including six flea samples for molecular taxonomy, were subjected to Rickettsia spp. DNA detection. The molecular identity of fleas was determined by amplification and sequenmce analysis of four genetic markers (the 28S rDNA genes, the 18S rDNA genes, the mitochondrial cytochrome c oxidase subunit I and subunit II). In addition, five Rickettsia-specific gene fragments were used to identify the species of the rickettsial agents. The amplified products were sequenced and phylogenetically analyzed. Results: The morphological characteristics of the flea species identified in this study were similar to Vermipsylla alakurt, but presented difference in hair number of the metepimeron, the third tergum, the genitals and the tibiae of hind leg. The 18S rDNA, 28S rDNA and COII genetic markers from fleas showed the highest identity to those of V. alakurt, shared 98.45% (954/969), 95.81% (892/931) and 85.86% (571/665) similarities, respectively. However, the COI sequence showed the highest identity to that of Dorcadia ioffi with 88.48% (576/651) similarity. Rickettsia raoutii tested positive in 14.17% (17/120) flea DNA samples. Conclusion: Our study reports the detection of R. raoultii in V. alakurt-like fleas infesting sheep in China.
Diversity of biting midges, mosquitoes and sand flies at four dog shelters in rural and peri-urban areas of Central Morocco

Abstract:Blood-feeding arthropods are involved in the transmission of several pathogens that have a major impact on public health. Entomological investigations highlighted the composition, abundance, and diversity of flying hematophagous arthropods at four dog shelters located in central Morocco during an eight-month study, with the aim of discussing their vectorial roles and assessing the risk of these shelters as foci for zoonotic diseases. Monitoring of the arthropod fauna for 64 catch nights resulted in the collection of 2,321 biting midges (Ceratopogonidae), 570 mosquitoes (Culicidae), and 475 sand flies (Psychodidae). Fourteen Culicoides species were recorded and dominant species were Culicoides imicola (55.96%), C. paolae (16.07%), C. circumscriptus (10.29%), and C. newsteadi (5.77%). Three mosquito species were collected, including Culex pipiens s.l. (96.84%), Culiseta longiareolata (2.80%), and Cx. perexiguus (0.36%). Ten sand fly species were collected, including seven Phlebotomus species (62.70%) and three Sergentomyia species (37.30%); Sergentomyia minuta was the most dominant species (34.31%), followed by Phlebotomus sergenti (32.42%), typical Ph. perniciosus (8.63%), Ph. alexandri (6.94%), and Ph. riouxi (6.52%). The coexistence of several vectors in these study areas indicates the potential circulation of a wide range of pathogens, including zoonotic ones, thus requiring the implementation of surveillance and control programs to prevent the emergence and spread of disease outbreaks. Les arthropodes hématophages sont impliqués dans la transmission de plusieurs agents pathogènes ayant un impact majeur sur la santé publique. Des investigations entomologiques ont mis en évidence la composition, l'abondance et la diversité des arthropodes hématophages volants dans quatre chenils situés au centre du Maroc au cours d'une étude de huit mois, dans le but de discuter de leurs rôles vectoriels et d'évaluer le risque de ces chenils comme foyers de maladies zoonotiques. Le suivi de la faune des arthropodes au cours de 64 nuits de capture a permis la collecte de 2 321 culicoïdes (Ceratopogonidae), 570 moustiques (Culicidae) et 475 phlébotomes (Psychodidae). Quatorze espèces de Culicoides ont été recensées et les espèces dominantes étaient Culicoides imicola (55,96 %), C. paolae (16,07 %), C. circumscriptus (10,29 %) et C. newsteadi (5,77 %). Trois espèces de moustiques ont été collectées, dont Culex pipiens s.l. (96,84 %), Culiseta longiareolata (2,80 %) et Cx. perexiguus (0,36 %). Dix espèces de phlébotomes ont été collectées, dont sept espèces de Phlebotomus (62,70 %) et trois espèces de Sergentomyia (37,30 %); Sergentomyia minuta était l'espèce dominante (34,31 %), suivie de Phlebotomus sergenti (32,42 %), Ph. perniciosus typique (8,63 %), Ph. alexandri (6,94 %) et Ph. riouxi (6,52 %). La coexistence de plusieurs vecteurs dans ces zones d'étude indique la circulation potentielle d'un large éventail d'agents pathogènes, y compris zoonotiques, ce qui nécessite la mise en œuvre de programmes de surveillance et de contrôle pour prévenir l'émergence et la propagation d'épidémies.

Abstract:This four-year study (from April 2013 to May 2017) focuses on the inventory and updating of the geographical distribution of some endangered wild mammals in various biotopes in western Algeria. Our work has been started over the years (2013-2017) ie four years of investigation based on direct and indirect observation as well as information collected from a questionnaire and surveys conducted with indigenous populations, hunters and foresters. Our investigations carried out on this mammalian fauna, allowed us to count 14 species of carnivorous mammals, the order of Carnivores contains: the Canids where a wide distribution has been reported on the whole territory to study. The latter are represented by Canis aureus or North African wolf Canis lupus, Vulpes rupellii and Vulpes vulpes. Felids; are poorly represented in the study area. Indeed, the 03 identified species in this group are all taxa listed by subsequent studies as threatened in the region. These species are Felis libyca, Felis margarita and Felis caracal. The Hyaenidae; very low and almost nonexistent in the desert areas. This group is represented by Hyaena hyaena. Mustelids; the 05 species Mustelanivalis, Ictonyx libyca, Mellivora capensis, Lutralutra and Mustela putorius furo showed a very small geographical distribution. The viverrids represented by Genetta genetta are present throughout the area but in small numbers. Finally, the Herpestidae; represented by Herpestes ichneumon are present with numbers that place the species in danger and risk of extinction.
Taxonomy and phylogeny of rodents parasitic fleas in southeastern China with description of a new subspecies of Ctenophthalmus breviprojiciens

Abstract:Accurate differentiation and identification of flea species are essential for both basic and applied research on fleas, as well as for the diagnosis of flea-borne diseases. However, distinguishing between flea species can be challenging, especially among those with minimal morphological differences. Therefore, some scholars have suggested the necessity of comprehensive revisions to the classification of fleas, incorporating morphological, molecular, and phylogenetic data. In this study, we focused on classifying the rodents' parasitic fleas in southeastern China and provided molecular and phylogenetic data. We also described a new subspecies Ctenophthalmus breviprojiciens fujiansis n. ssp. A total of 392 fleas were collected from 8 species of rodents in 10 counties. Morphologically, they belonged to 10 species, 9 genera and 5 families. Barcode identification based on COI gene and phylogenetic analysis based on five genetic markers ( 18S rDNA , 28S rDNA , EF-1a , COI , COII ) revealed that the molecular and morphological identification of Xenopsylla cheopis , Aviostivalius klossi bispiniformis , Leptopsylla segnis , Monopsyllus anisus and Ctenocephalides felis felis were consistent. The taxonomic status of Neopsylla specialis minpiensis and Peromyscopsylla himalaica sinica as subspecies is questionable due to significant intraspecific genetic distance, and further morphological and molecular data are required to determine if they should be elevated to species level. The molecular identification of C. breviprojiciens n. ssp., N. dispar fukienensis , and Nosopsyllus nicanus could not be completed at this time due to a lack of sequences for related species in existing GenBank databases. Additionally, phylogenetic relationships of 31 species from 9 genera and 5 families of Siphonaptera were inferred based on five molecular markers ( 18S rDNA , 28S rDNA , EF-1a , COI and COII ) using Maximum Likelihood analyses. The analyses revealed that various taxa of Siphonaptera are monophyletic at the subspecies, species, and genus levels. However, at the family level, Leptopsyllidae, Ceratophyllidae, Pulicidae, and Pygiopsyllidae are all monophyletic, while Ctenophthalmidae is paraphyletic. we support the view of some authors that revising the catchall group Ctenophthalmidae and elevating each of its constituent subfamilies to family status.
