Abstract:The objective of this study was to evaluate the potential benefits of feeding spray-dried porcine plasma (SDPP) to pigs infected with African swine fever virus (ASFV). Two groups of twelve weaned pigs each were fed with CONVENTIONAL or 8% SDPP enriched diets. Two pigs (trojans)/group) were injected intramuscularly with the pandemic ASFV (Georgia 2007/01) and comingled with the rest of the pigs (1:5 trojan:naïve ratio) to simulate a natural route of transmission. Trojans developed ASF and died within the first week after inoculation, but contact pigs did not develop ASF, viremia, or seroconversion. Therefore, three more trojans per group were introduced to optimize the ASFV transmission (1:2 trojan:naïve ratio). Blood, nasal, and rectal swabs were weekly harvested, and at end of the study ASFV-target organs collected. After the second exposure, rectal temperature of conventionally fed contact pigs increased >40.5 °C while fever was delayed in the SDPP contact pigs. Additionally, PCR Ct values in blood, secretions, and tissue samples were significantly lower (p < 0.05) for CONVENTIONAL compared to SDPP contact pigs. Under these study conditions, contact exposed pigs fed SDPP had delayed ASFV transmission and reduced virus load, likely by enhanced specific T-cell priming after the first ASFV-exposure.
Infrequent Transmission of HIV-1 Drug-Resistant Variants

Abstract:Transmission of drug-resistant variants is influenced by several factors, including the prevalence of drug resistance in the population of HIV-1-infected patients, HIV-1 RNA levels and transmission by recently infected patients. In order to evaluate the impact of these factors on the transmission of drug-resistant variants, we have defined the population of potential transmitters and compared their resistance profiles to those of newly infected patients. Sequencing of pol gene was performed in 220 recently infected patients and in 373 chronically infected patients with HIV-1 RNA >1000 copies/ml. Minimal and maximal drug-resistance profiles of potential transmitters were estimated by weighting resistance profiles of chronically infected patients with estimates of the Swiss HIV-1-infected population, the prevalence of exposure to antiviral drugs and the proportion of infections attributed to primary HIV infections. The drug-resistance prevalence in recently infected patients was 10.5% (one class drug resistance: 9.1%; two classes: 1.4%; three classes: 0%). Phylogenetic analysis revealed significant clustering for 30% of recent infections. The drug-resistance prevalence in chronically infected patients was 72.4% (one class: 29%; two classes: 27.6%; three classes: 15.8%). After adjustment, the risk of transmission relative to wild-type was reduced both for one class drug resistance (minimal and maximal estimates: odds ratio: 0.39, P<0.001; and odds ratio: 0.55, P=0.011, respectively), and for two to three class drug resistance (odds ratios: 0.05 and 0.07, respectively, P<0.001). Neither sexual behaviour nor HIV-1 RNA levels explained the low transmission of drug-resistant variants. These data suggest that drug-resistant variants and in particular multidrug-resistant variants have a substantially reduced transmission capacity.

Abstract:African swine fever virus (ASFV) is known to be very stable and can remain infectious over long periods of time especially at low temperatures and within different matrices, particularly those containing animal-derived organic material. However, there are some gaps in our knowledge pertaining to the survivability and infectivity of ASFV in groundwater. This study aims to determine the stability and infectivity of the cell culture-adapted ASFV strain BA71V by plaque assay after incubation of the virus within river water samples at three different environmentally relevant temperatures (4 °C, 15 °C, and 21 °C) over the course of 42 days. The results from this study indicate that ASFV can remain stable and infectious when maintained at 4 °C in river water for more than 42 days, but as incubation temperatures are increased, the stability is reduced, and the virus is no longer able to form plaques after 28 days and 14 days, respectively, when stored at 15 °C and 21 °C. Characterizing the survivability of ASFV in groundwater can allow us to develop more appropriate inactivation and disinfection methods to support disease control and mitigate ASFV outbreaks.
Inactivation rate of African swine fever virus by a formaldehyde-based product

Abstract:African swine fever (ASF) is an important transboundary animal disease with a high mortality rate. The high African swine fever virus (ASFV) titers in the excretion of infected wild boar possibly contaminate the feed ingredient. Once contaminated, it could support persistent residual titer of the ASFV. The chemical inactivation of imported feed ingredients is a precautionary risk management measure to restrict the import risk of ASFV through international trade. The log ASFV titers were linearly reduced as a function of the inactivation time after exposure to 0.03%, 0.05%, 0.1%, and 0.2% formaldehyde-based product (FBP). A four-log reduction of ASFV titer was achieved after exposure to 0.2% FBP and 0.03%-0.1% FBP for 30-min and 60-min inactivation times, respectively. The decimal reduction time or D value is defined as the time required to inactivate the virus titer by 1 log. The ASFV inactivation rate from the independent experiment of FBP concentration was converted to a D value. The observed mean D0.2%, D0.1%, D0.05%, and D0.03% of FBP were 13.4, 44.9, 45.0, and 45.3 min per log reduction of ASFV, respectively. The interpretation of D0.2% of FBP is that the ASFV titer is inactivated by 1 log after being exposed to 0.2% FBP for every 13.4 min. A more effective chemical has a lower D value because of a shorter inactivation time required to achieve the same 1-log reduction. In addition, the hypothetical inactivation time by any chemical additive is scenario-specific and is calculated by the product of D value (at a certain concentration) and log titers of residual ASFV. This study introduces the concept and application of the D value to compare the virucidal activity of chemicals and to determine the hypothetical inactivation time of chemicals depending on the chemical concentration including the virus titer in the feed.

Abstract:Interleukin 1 (IL-1) has been implicated in the production of characteristic changes in psoriatic lesions such as epidermal hyperproliferation and accumulation of polymorphonuclear leukocytes within the epidermis. Because lobenzarit disodium, an immunomodulator, inhibits the production of IL-1 in peripheral and peritoneal macrophages, we studied the clinical effects of this compound on psoriatic lesions. A 12-week course of oral lobenzarit disodium treatment did not induce any remarkable changes in psoriasis lesions. This does not support the major pathogenic role of IL-1 in the development of psoriasis.

Abstract:This study was based on similar physicochemical characteristics of pseudorabies virus (PRV) and African swine fever virus (ASFV). A cellular model for evaluation of disinfectants was established with PRV as an alternative marker strain. In the present study, we evaluated the disinfection performance of commonly used commercialized disinfectants on PRV to provide a reference for the selection of good ASFV disinfectants. In addition, the disinfection (anti-virus) performances for four disinfectants were investigated based on the minimum effective concentration, onset time, action time, and operating temperature. Our results demonstrated that glutaraldehyde decamethylammonium bromide solution, peracetic acid solution, sodium dichloroisocyanurate, and povidone-iodine solution effectively inactivated PRV at concentrations 0.1, 0.5, 0.5, and 2.5 g/L on different time points 30, 5, 10, and 10 min, respectively. Specifically, peracetic acid exhibits optimized overall performance. Glutaraldehyde decamethylammonium bromide is cost effective but requires a long action time and the disinfectant activity is severely affected by low temperatures. Furthermore, povidone-iodine rapidly inactivates the virus and is not affected by environmental temperature, but its application is limited by a poor dilution ratio such as for local disinfection of the skin. This study provides a reference for the selection of disinfectants for ASFV.

Evaluation of Extended Storage of Swine Complete Feed for Inactivation of Viral Contamination and Effect on Nutritional, Microbiological, and Toxicological Profiles

Abstract:The extended storage of feed ingredients has been suggested as a method to mitigate the risk of pathogen transmission through contaminated ingredients. To validate the approach of extended storage of complete swine feed for the inactivation of swine viruses, an experiment was conducted wherein swine feed was inoculated with 10 mL of 1 × 105 TCID50/mL of porcine reproductive and respiratory syndrome virus (PRRSV), porcine epidemic diarrhea virus (PEDV), and Senecavirus A (SVA) and stored for 58 d at 23.9 °C. Measures of feed quality were also evaluated at the initiation and conclusion of the storage period including screening for mycotoxins, characterization of select microbiological measures, and stability of phytase and dietary vitamins. Storing feed for 58 d under either ambient or anaerobic and temperature-controlled storage conditions did not result in substantial concerns related to microbiological profiles. Upon exposure to the feed following 58 d of storage in a swine bioassay, previously confirmed naïve pigs showed no signs of PEDV or SVA replication as detected by the PCR screening of oral fluids and serum antibody screening. Infection with SVA was documented in the positive control room through diagnostic testing through the State of Minnesota. For PRRSV, the positive control room demonstrated infection. For rooms consuming inoculated feed stored for 58 d, there was no evidence of PRRSV infection with the exception of unintentional aerosol transmission via a documented biocontainment breach. In summary, storing feed for 58 d at anaerobic and temperature-controlled environmental conditions of 23.9 °C validates that the extended storage of complete swine feed can be a method to reduce risks associated with pathogen transmission through feed while having minimal effects on measures of nutritional quality.

Abstract:African swine fever (ASF) is an economically important disease due to high morbidity and mortality rates and the ability to affect all ages and breeds of pigs. Biosecurity measures to prevent the spread of the causative agent, African swine fever virus (ASFV), include prescriptive cleaning and disinfection procedures. The aim of this study was to establish the biocidal effects of twenty-four commercially available disinfectants including oxidizing agents, acids, aldehydes, formic acids, phenol, and mixed-class chemistries against ASFV. The products were prepared according to the manufacturer's instructions and a suspension assay was performed with ASFV strain, BA71V using Vero cells (African green monkey cells) to test efficacy in reducing ASFV infection of cells. Generally, disinfectants containing formic acid and phenolic compounds, as well as oxidizing agents reduced viral titers of ASFV by over 4 log10 at temperatures ranging from 4 °C to 20 °C. Hydrogen peroxide, aldehyde, and quaternary ammonium compounds containing disinfectants were cytotoxic, limiting the detection of viral infectivity reductions to less than 4 log10. These preliminary results can be used to target research on disinfectants which contain active ingredients with known efficacy against ASFV under conditions recommended for the country where their use will be applied.

Abstract:African swine fever (ASF) is a disease that is a growing threat to the global swine industry. Regulations and restrictions are placed on swine movement to limit the spread of the virus. However, these are costly and time-consuming. Therefore, this study aimed to determine if high-pressure processing (HPP) sanitization techniques would be effective against the ASF virus. Here, it was hypothesized that HPP could inactivate or reduce ASF virus infectivity in tissue homogenates. To test this hypothesis, 30 aliquots of each homogenate (spleen, kidney, loin) were challenge-infected with the Turin/83 strain of ASF, at a 10 7.20 median hemadsorption dose (HAD)50/mL. Subsequently, eight aliquots of each homogenate were treated with 600 millipascal (600 MPa) HPP for 3, 5, and 7 min. Six untreated aliquots were used as the controls. Virological results showed a reduction in the viral titer of more than 7-log. These results support the validity of the study hypothesis since HPP treatment was effective in inactivating ASFV in artificially prepared samples. Overall, this study suggests the need for further investigation of other ASFV-contaminated meat products.

Abstract:African swine fever virus (ASFV) causes a fatal infectious disease affecting domestic pigs and wild boars. ASFV is highly stable and easily transmitted by consumption of contaminated swine feed and pork products. Heat treatment of feed ingredients is a means to minimize the risk of contamination through swine feed consumption. The objectives of this study were to determine the thermal inactivation of ASFV in non-animal and animal origin feed ingredients. The rate of thermal inactivation is represented by decimal reduction time ( D T ) or time required to reduce ASFV per 1 log at temperature T . The mean D 60 , D 70 , D 80 and D 85 of meat and bone meal (MBM), soybean meal (SBM), and maize grain (MZ) are in the ranges 5.11–6.78, 2.19–3.01, 0.99–2.02, and 0.16–0.99 min, respectively. D T is used to compare the heat resistance of ASFV in the feed ingredient matrices. The mean D T of ASFV in MBM, SBM and MZ was not statistically significant, and the heat resistance of ASFV in MBM, SBM, and MZ was not different at 60, 70, 80, or 85 °C. The multiple D T was used to develop a D T model to predict D T at various inactivation temperatures. The D T models for MBM, SBM, and MZ are log D T = − + 2.69, log D T = − + 2.55, and log D T = − + 4.01. To expand and ease the field applications, a spreadsheet predicting the D T and the inactivation time (with 95% confidence interval) from these D T models is available to download.

Abstract:Abstract Spray dried plasma (SDP) is a functional protein source obtained from blood of healthy animals, approved by the veterinary authorities from animals declared to be fit for slaughter for human consumption. Blood of these animals is collected at the slaughterhouse, treated with an anticoagulant, chilled and transported to industrial facilities in which blood is centrifuged to separate the red blood cells from the plasma fraction. Plasma is then concentrated, and spray dried at high temperatures (80 °C throughout its substance) to convert it in a powder. Such method preserves the biological activity of its proteins, mainly albumins and globulins. SDP is mainly used in pig feed diets to significantly improve daily gain, feed intake, production efficiency, and to reduce post-weaning lag caused by the appearance of post-weaning diarrhea. Although SDP is considered a safe product and its manufacturing process consists of several biosafety steps, the security of the SDP is often questioned due to its nature as raw blood by-product, especially when emergent or re-emergent pathogens appear. This review provides an evaluation and validation of the different safety steps present in the manufacturing process of SDP, with special focus on a new redundant pathogen inactivation step, the UV-C irradiation, that may be implemented in the manufacturing process of the SDP. Overall results showed that the manufacturing process of SDP is safe and the UV-C radiation was effective in inactivating a wide range of bacteria and viruses spiked and naturally present in commercially collected liquid animal plasma and it can be implemented as a redundant biosafety step in the manufacturing process of the SDP.

Nerve Management and Chronic Pain After Open Inguinal Hernia Repair: A Prospective Two Phase Study

Abstract:Introduction:This prospective cohort study involved 781 elective primary inguinal hernia operations performed on 736 patients at the Hernia Centre of Reinbek Hospital from April 2000 to April 2002. Material and Methods:Small hernias were fixed by the Shouldice repair, and large defects by the Lichtenstein repair with conventional polypropylene mesh. Pain was assessed before the operation, on day 0, 1, 2, 7, 6 months and 5 years after the operation by the visual analogue scale (VAS). The follow-up was 90.1% after 6 months and 82.6% after 5 years. Chronic pain (CP) was assessed in relation to preoperative pain, nerve anatomy and intraoperative nerve management. The anatomy of the iliohypogastric nerve (IHN), ilioinguinal nerve (IIN), and genital branch of the genitofemoral nerve (GB) before and after surgery was recorded in every operation. Results:The preoperative pain rate was 41.0%. The CP and sensory disorder rate after 6 months were 16.4% and 15.9, respectively. The only independent significant parameters for CP after 6 months were preoperative pain (P < 0.002) and sensory disorder of the groin after 6 months (P < 0.0001).After 5 years 16.1% of patients reported pain and 20.3% sensory disorder of the groin. Independent significant predictors of CP were: Preoperative pain (P < 0.024), IIN neurolysis in Lichtenstein repair (=IIN mobilization from its natural bed and nerve preservation; P < 0.002), CP after 6 months (P < 0.006) and a sensory disorder after 5 years (P < 0.0001). Eleven of 12 patients with relevant CP (VAS > 3) after 5 years had had a Lichtenstein repair with IIN neurolysis.The CP population of preoperative pain free patients changed with time: 65% of the patients with CP after 6 months were pain free after 5 years, and 69% of the patients with CP after 5 years were asymptomatic after 6 months. Conclusion:Mesh contact with a nerve removed from its natural bed may cause chronic long-term pain. The combination of IIN neurolysis and the Lichtenstein repair should be avoided.
Stability of African Swine Fever Virus in Carcasses of Domestic Pigs and Wild Boar Experimentally Infected with the ASFV “Estonia 2014” Isolate

Abstract:Europe is currently experiencing a long-lasting African swine fever (ASF) epidemic, both in domestic pigs and wild boar. There is great concern that carcasses of infected wild boar may act as long-term virus reservoirs in the environment. We evaluated the tenacity of ASF virus (ASFV) in tissues and body fluids from experimentally infected domestic pigs and wild boar, which were stored on different matrices and at different temperatures. Samples were analysed at regular intervals for viral genome and infectious virus. ASFV was most stable in spleen or muscles stored at −20 °C and in blood stored at 4 °C. In bones stored at −20 °C, infectious virus was detected for up to three months, and at 4 °C for up to one month, while at room temperature (RT), no infectious virus could be recovered after one week. Skin stored at −20 °C, 4 °C and RT remained infectious for up to three, six and three months, respectively. In urine and faeces, no infectious virus was recovered after one week, irrespective of the matrix. In conclusion, tissues and organs from decomposing carcasses that persist in the environment for a long time can be a source of infection for several months, especially at low temperatures.

Abstract:African swine fever is an important viral disease of domestic pigs and wild boars. Inactivation of the African swine fever virus (ASFV) contaminating equipment and premises is an effective biosecurity risk management measure to prevent the introduction of ASFV through contaminated fomites and the environment. Even though some chemical agents' virucidal activity against ASFV was previously demonstrated, compound disinfectants are still limited. For the fact that the ASFV titer in some excretions from pigs was far >4 log. Even after the application of disinfectants as instructed by the manufacturers, the excessive ASFV load in the excretions from pigs could contaminate fomites or the environment. To solve this dilemma, the concept of contact time of disinfectant for 1–log reduction was introduced, as the decimal reduction time or D value. Therefore, the objective of this study was to assess the inactivation rate of four compound disinfectants against ASFV as the D value. Four compound disinfectants (iodine and phenol, quaternary ammonium and alcohol, and two quaternary ammonium and glutaraldehyde compounds) currently used in the veterinary field were evaluated in a suspension test at 25 °C in the absence of interfering substances. Primary porcine alveolar macrophages were used to determine the titer of ASFV genotype II (strain VNUA/HY-ASF1/Vietnam/2019). The virucidal activity of four compound disinfectants achieved 4–log ASFV reduction. The mean D 0.5%, D 0.25%, and D 0.125% values of all disinfectants inactivating ASFV were 1.62–3.60, 2.60–3.8, and 2.89–3.90 min, respectively. The D value of disinfectants allows users to manipulate the appropriate concentration and/or contact time of the disinfectant, depending on the magnitude of ASFV contamination, to completely inactivate the ASFV titer on the fomites to minimise a potential biosecurity risk.

Abstract:Understanding African swine fever virus (ASFV) transmission is essential for strategies to minimize virus spread during an outbreak. ASFV can survive for extended time periods in animal products, carcasses, and the environment. While the ASFV genome was found in environments around infected farms, data on the virus survival in soil are scarce. We investigated different soil matrices spiked with ASFV-positive blood from infected wild boar to see if ASFV can remain infectious in the soil beneath infected carcasses. As expected, ASFV genome detection was possible over the entire sampling period. Soil pH, structure, and ambient temperature played a role in the stability of infectious ASFV. Infectious ASFV was demonstrated in specimens originating from sterile sand for at least three weeks, from beach sand for up to two weeks, from yard soil for one week, and from swamp soil for three days. The virus was not recovered from two acidic forest soils. All risk mitigation experiments with citric acid or calcium hydroxide resulted in complete inactivation. In conclusion, the stability of infectious ASFV is very low in acidic forest soils but rather high in sandy soils. However, given the high variability, treatment of carcass collection points with disinfectants should be considered.

The disease is known for almost 100 years, an effective vaccine or treatment is still unavailable, only proper biosecurity measures, including disinfection, are being applied, in order to prevent disease outbreaks. Eight active substances, i.e., formaldehyde, sodium hypochlorite, caustic soda, glutaraldehyde, phenol, benzalkonium chloride, potassium peroxymonosulfate and acetic acid, were tested, in order to confirm their effectiveness against African swine fever virus (ASFV). This specific selection was done based on the World Organisation for Animal Health (OIE)’s recommendation and previous disinfectant studies on surfaces. The result of our study shows that most of them inactivate the virus, in recommended concentrations. In order to reduce the cytotoxicity of the four substances, Microspin S-400 HR columns were applied, therefore making it possible to demonstrate four logarithms virus titer reduction. Sodium hypochlorite, glutaraldehyde, caustic soda and potassium peroxymonosulfate showed the best ASFV inactivation rates, achieving titer reductions over 5 logs. Despite microfiltration, the virucidal activity of formaldehyde was not assessable, due to its high cytotoxicity. Our results showed that cleaning is particularly important, because removal of the soiling provides improved effectiveness of the tested chemical compounds.

Abstract:In recent years, African swine fever (ASF) has been the main threat to the development of the pig industry not only in Russia, but also in many countries of the world for many years. Due to the lack of effective and safe means of specific prevention of infection, disinfection is one of the ways to combat and prevent the spread of the ASF pathogen in the system of antiepizootic measures. The most important task of modern disinfection is the development and creation of disinfectants with a wide range of bactericidal and viricidal effects. The composition of such despreprates includes several active substances that, due to interaction with each other, enhance the disinfecting ability of the drug.The aim of the work is to study in vitro the viricidal activity of the composite disinfectants "Dezon Vetklin" and "Dezon Vet" against a highly virulent ASF pathogen. The objects of research are domestic modern composite disinfectants "Dezon Vetklin" and "Dezon Vet". The tests of the drugs were carried out on the basis of the Federal State Budgetary Institution "Federal Center for Animal Health Protection" (FGBI "VNIIZH", Vladimir). As a test culture, a primary culture of pig spleen cells sensitive to ASF virus (CCSS) was used with the addition of Igla growth culture medium containing 20% fetal serum of cattle and 0.1% (v/v) pig erythrocytes. As a protein load, inactivated pig blood serum obtained from animals from ASF-safe farms of the Russian Federation was used. For experimental infection of CCSS, a highly virulent hemadsorbent ASF virus of genotype II strain ASF/Arm/07 (reference) was used in a titer not lower than 6.0 lg GAdE50/cm3. For experimental testing of disinfectants, metal, wooden, concrete and plastic test surfaces with a size of 10x10 cm were used. According to the results of the studies, viricidal activity against the ASF pathogen of two tested disinfectants was established at a concentration of 5.0% with a minimum exposure of 10 minutes each. The tested disinfectants can be recommended for preventive disinfection at veterinary surveillance facilities in ASF.
Live vaccine preserved at room temperature: Preparation and characterization of a freeze-dried classical swine fever virus vaccine

Abstract:The heat-stable live-attenuated classical swine fever virus (CSFV) vaccine is an urgent need in many countries of Asia, Europe and Latin America. In this study, the thermostability of lyophilized live-attenuated CSFV vaccine formulations were investigated using accelerated stability at 37 °C for 10 days. The freeze-dried heat-stable formulation ST16, containing excipient combinations of trehalose, glycine, thiourea and phosphate buffer shows the superior thermostability. Moreover, the lyophilized vaccine with formula ST16 kept loss of viral activity less than 0.5 log10 during 24 months at storage temperatures of 2–8 °C. In thermal study, ST16 stabilized the vaccine within 1.0 log10 loss after storage at up to 25 °C for 6 months and room temperature for 7 months. Even under the harshest storage conditions of 37 °C for 25 days and 45 °C for 2 weeks, the virus titer dropped less than 1.0 log10 using ST16. Besides, it is notable that ST16 excluded gelatin and exogenous proteins, which might cause allergic reactions, thus avoiding immune side effects. The vaccine formulated ST16 proved to be safe and effective when immunized to piglets in vivo. The characteristics of dried vaccines were analyzed by X-ray powder diffraction, residual water measurements, differential scanning calorimetry and it was found that vaccine antigen were preserved in an amorphous matrix with high glass transition temperature above 60 °C and low residual water content below 2%, which made the vaccine more stable during storage.

Abstract:African swine fever (ASF) causes fatal disease in pigs and is an escalating threat to the global swine industry. ASF has re-emerged from Africa as a transcontinental epidemic spreading through the Caucasus into Europe, Russia, China, numerous Asian countries, and the Caribbean. ASF virus (ASFV) is a U.S. select agent requiring handling in high-containment biosafety level 3 (BSL-3) laboratories for pathogen work. Formalin-fixation eliminates infectivity and preserves the genome, providing noninfectious specimens for BSL-2 work. Recovery of DNA from formalin-fixed, paraffin-embedded tissue (FFPET) is challenging and cumbersome. A reliable and easy-to-perform method for DNA recovery from FFPET would facilitate surveillance. To meet this objective, we developed a high-throughput protocol for the recovery of ASFV DNA from FFPET. Deparaffinization, tissue lysis, and reversal of cross-linking were performed in a single tube, followed by DNA purification via automated magnetic bead extraction. Quantitative PCR (qPCR) detection was used to determine the copy number of the B646L gene that encodes for the ASFV p72 protein in tissues (5 pigs, 4 tissues) from pigs with lesions consistent with acute ASF. Copy numbers obtained from FFPET were within one log of copy numbers obtained from fresh tissue, thus enabling ASF qPCR surveillance from formalin-inactivated and preserved tissues at BSL-2 at diagnostic sensitivity similar to fresh tissues tested at BSL-3.
