Abstract:Zein-based nanoparticles loaded with bioactive compounds have positive prospects in the food industry, but an important limiting factor for development is colloidal instability. Currently, extensive researches are focused on solving the instability of zein nanoparticles, but since the beginning of the studies, there has not been a summary of the factors affecting the stability of zein-based nanoparticles. In the present work, the factors were reviewed comprehensively from the perspective of carrier construction and application evaluation. The former mainly includes type, quantity, and characteristics of biopolymer, the mass ratio of biopolymer/bioactive compound to zein, blending sequence of biopolymer, and location of encapsulated bioactive compounds. The latter mainly includes pH, heating, ionic strength, storage, freeze-drying, and gastrointestinal digestion. The former is the prerequisite for the success of the latter. The challenge is that stability research is limited to the laboratory level, and it is difficult to ensure that the stability results are suitable for commercial food matrices due to their complexity. At the laboratory level, the future trends are the influence of external energy and the cross-complexity and uniformity of stability research. The review is expected to provide systematic understanding and guidance for the development of zein-based nanoparticles stability.

Surface coating of zein nanoparticles to improve the application of bioactive compounds: A review

Abstract:Background As an excellent delivery material for bioactive compounds, zein nanoparticles are limited by colloidal instability. Recently, the development of their surface coating has greatly promoted the performance of the zein-based delivery system. In the future, surface-coated zein nanoparticles delivering bioactive compounds will have broader applications in the food industry. Scope and approach This review focuses on introducing different methods and materials of surface-coated zein nanoparticles and their encapsulated bioactive compounds, including the assessments for the improved application of these compounds. Additionally, the application in food-related fields is summarized. Finally, some challenges regarding the surface coating of zein nanoparticles and development trends are presented. Key findings and conclusions Currently, methods for surface coating of zein nanoparticles include antisolvent precipitation, antisolvent co-precipitation, layer-by-layer, solvent evaporation, and pH-driven. The materials are mainly proteins, polysaccharides, surfactants, and their composites. It can not only deliver a single bioactive compound but also achieve co-delivery. The evaluation of the improved compound application includes physical and chemical stability, controlled release/bioaccessibility, and biological activity. Moreover, surface-coated zein nanoparticles can be applied to Pickering emulsions and edible films. There are also some challenges in surface coating development. The future will be to improve the application of zein carriers in commercial products, and laboratory-level researches including new system development, smart co-delivery design, and bioavailability mechanism. In conclusion, surface coating of zein nanoparticles have a bright prospect to improve the application of bioactive compounds in the food industry.

Abstract:Encapsulation is one of the most convenient ways to increase the solubility of hydrophobic compounds and enhance the protection of sensitive compounds. The development of natural and edible encapsulation systems is a challenge for the food industry. This study explored two kinds of nanoparticles (NPs) made by zein and gum arabic (GA) and investigated the protective and controlled release effect of the formed NPs on rutin. Single-layer NPs (ZG) formed with a zein and GA mixture and double-layer NPs (ZWG) with one inner layer of zein and one outer layer of GA were prepared with a series of mass ratios of zein to GA. The particle size, polydispersity index, and zeta potential were obtained, and the structure and morphology of the NPs were observed using scanning electron microscopy. As shown by the differential scanning calorimeter and thermogravimetric analysis, the heat resistance of ZG is higher than the heat resistance of ZWG. ZG showed slower release of rutin in the simulated gastric and small intestinal solutions than ZWG. These results will help in understanding the different behaviors of protection and controlled release of layered NPs made by mixtures of biopolymers, which can be directly applied to the design of delivery systems of sensitive bioactive compounds in the food industry.
Preparation, characterization and in vitro release of zein-pectin capsules for target delivery.

Abstract:Targeted drug delivery has been the interest of many researchers to improve drug efficiency and reduce side effects. Because of the potential toxicity and contamination problems of the current gelatin capsules, plant derived materials are selected for the developments of capsules for drug delivery systems. The objective of this work is to develop a target drug delivery system using zein and pectin. Different ratios of zein and pectin were used to achieve target delivery in stomach and small intestine. Zein-pectin capsule for colon-specific delivery was also developed. In vitro performance of zein-pectin capsules was examined and their structural morphology was characterized using scanning electron microscopy (SEM). Chemical interactions between zein and pectin were analyzed using Fourier transform infrared spectroscopy equipped with attenuated total reflectance (FTIR-ATR). Zein and pectin formed complex by hydrogen bonding. The swelling behavior of pectin was suppressed by zein in the zein-pectin interacted complex. By adjusting the ratio of zein to pectin, the drug release from the capsule in simulated gastric solution for 2 hours can be controlled in the range of 0% to 38%. Zein-pectin capsule for colon-specific delivery had no release in gastric and intestinal solutions while gradual release from zein-pectin capsule was observed in colonic solution, finally reaching about 80% release. Zein-pectin capsule has a potential in developing targeted drug delivery system.

Abstract:ABSTR ACT Owing to the high desorption energy of colloidal particles, Pickering emulsions can effectively protect the encapsulated bioactives. In this study, zein and peach gum polysaccharide (PGP) based biopolymeric nanoparticles were fabricated to stabilize the Pickering emulsion and deliver curcumin. Effects of the PGP addition order during and after anti-solvent precipitation process (zein-PGP nanoparticles or zein/PGP nanoparticles) on the properties of nanoparticles were discussed. The zein-PGP nanoparticles were spherical with mean size of 172.8 nm, while zein/ PGP nanoparticles were rod-like with mean size of 207.5 nm. Both zein-PGP and zein/PGP nanoparticles were negatively charged with the wettability of 93.5 degrees and 90.3 degrees, respectively. Compared to the zein NPs, the zein-PGP and zein/PGP nanoparticles stabilized emulsions had smaller droplet size, better storage and environmental stabilities. The release rates of curcumin in zein-PGP nanoparticles and zein/PGP nanoparticles stabilized emulsions were 23.4% and 26.7% respectively in simulated gastric fluid. The curcumin release rates in zein-PGP nanoparticles and zein/PGP nanoparticles stabilized emulsions were 32.5% and 24.0% respectively in simulated intestinal fluid. These findings might promote an effective delivery system for bioactive food ingredients using zein and PGP based complex nanoparticles.

Abstract:This current research explored the application of cold plasma (CP) treatment to modify zein-alginate oligosaccharide (zein-AOS) composites in an ethanol-water solution. Anti-solvent method was used to prepare zein-AOS nanoparticles (NPs), and the objective was to investigate the mechanism by which CP promotes interaction between protein and saccharides. Characterization results indicated that CP treatment improved hydrogen bonding and electrostatic interaction between zein and AOS. The CP zein-AOS NPs underwent dispersion and rearrangement, resulting in smaller aggregates with better dispersibility. Among the various induction conditions tested, the zein-AOS85 NPs (induced at 85 W for 2 min) exhibited superior performance as delivery wall materials, with smaller particle size (234.67 nm), larger specific surface area (9.443 m2/g), and higher surface charge (-35.43 mV). In addition, zein-AOS85 showed high stability when used as delivery wall material, providing more binding sites and self-assembly dynamics for nutrients. Curcumin was used as the nutrient model in this study, and CP was found to enhance hydrogen bonding, electrostatic interaction, and hydrophobic interaction between zein, AOS, and nutrients, resulting in increased encapsulation efficiency (EE) from 63.80 % to 85.17 %. The delivery system also exhibited good pH, ionic strength, storage, and dispersion stability.

Abstract:Nano/micro-carrier systems have shown promising application in the biomedical field as various delivery carriers. The composite material and fabrication method determine their microstructures, properties and thus their potential applications. Since approved as tablet coating material by the U.S. Food and Drug Administration (US-FDA), zein has been widely investigated as one of protein-based materials in the past few decades. Zein is renewable, biodegradable and relatively inexpensive in comparison with animal proteins (e.g., gelatin and albumin). This paper reviews the current landscape of zein-based nano/micro-carrier systems, with particular emphasis on nano/microparticles, nano/microcapsules and their design, fabrication, assembly mechanisms and biomedical applications especially for controlled drug delivery. The benefits, challenges and related solutions of zein-based colloidal carrier systems are also discussed. In addition, investigations on the molecular structure, biocompatibility and immunogenicity of zein are summarized and discussed.
The force of Zein self-assembled nanoparticles and the application of functional materials in food preservation

Abstract:Zein self-assembled nanoparticles (Z-NPs) are an excellent delivery carrier for bioactive components. However, the poor stability of its application in the food industry is the main problem. This paper focused on the self-assembly force of Z-NPs and the factors affecting the stability of Z-NPs. Meanwhile, the modification methods of zein and its interaction with food additives were analyzed. Additionally, its application in the field of food preservation was reviewed. The main interactions between zein and polyphenols encompass hydrogen bonding, non-covalent interactions, and hydrophobic interactions. Besides, the interactions with polysaccharides involve both covalent and non-covalent interactions. Furthermore, the protein interactions entail hydrophobic interactions, electrostatic interactions, hydrogen bonds, and π-π stacking. The primary driving forces governing zein self-assembly encompass electrostatic interactions, hydrogen bonding, van der Waals forces, hydrophobic interactions, and π-π stacking. Meanwhile, functionalized Z-NPs can be used in the food preservation industry to prolong the shelf life of food.
Preparation, characterization, and antioxidant activity of zein nanoparticles stabilized by whey protein nanofibrils

Abstract:The application of natural biopolymers to develop colloidal carriers for delivering hydrophobic bioactive molecules has become one of the hottest topics in food science. In this study, a simple pH-driven method to prepare whey protein nanofibrils stabilized zein nanoparticles was developed. The formation, structure, intermolecular interactions, rheological properties, in vitro digestion, and antioxidant activity of the nanoparticles were investigated. When the mass ratio of zein and whey protein nanofibrils was 1:0.8, the hydrodynamic diameter and zeta potential of the nanoparticles were 413 nm and - 25 mV, respectively. Transmission electron microscope photographs showed that the nanoparticles were a spherical core-shell structure. Fourier transform infrared spectroscopy and surface hydrophobicity measurements indicated that whey protein nanofibrils adsorbed to the surface of zein aggregates through hydrogen bonding and hydrophobic interactions. Differential scanning calorimetry tests confirmed the formation of nanoparticles improved the thermal stability of zein and whey protein nanofibrils. The nanoparticles exhibited less shear-thinning behavior with low apparent viscosity, and it can't be degraded in simulated gastric fluid but can be in simulated intestinal fluid. The antioxidant activity of the nanoparticles was increased by in vitro antioxidant assay when compared to whey protein nanofibrils. This new technology to develop zein-whey protein nanoparticles may be used for preparing nanoparticles of other similar hydrophobic food ingredients. And the findings of this study may provide a theoretical basis for preparation of nanoparticles as a nutrient delivery system.

Abstract:Plant proteins have gained significant attention over animal proteins due to their low carbon footprint, balanced nutrition, and high sustainability. These attributes make plant protein nanocarriers promising for applications in drug delivery, nutraceuticals, functional foods, and other areas. Zein, a major by-product of corn starch processing, is inexpensive and widely available. Its unique self-assembly characteristics have led to its extensive use in various food and drug systems. Zein's functional tunability allows for excellent performance in loading and transporting bioactive substances. Lutein offers numerous bioactive functions, such as antioxidant and vision protection, but suffers from poor chemical stability and low bioavailability. Nano-embedding technology can construct various zein-loaded lutein nanodelivery systems to address these issues. This review provides an overview of recent advances in the construction of zein-loaded lutein nanosystems. It discusses the fundamental properties of these systems; systematically introduces preparation techniques, structural characterization, and functional properties; and analyzes and predicts the target-controlled release and bioaccessibility of zein-loaded lutein nanosystems. The interactions and synergistic effects between Zein and lutein in the nanocomplexes are examined to elucidate the formation mechanism and conformational relationship of zein-lutein nanoparticles. The physical and chemical properties of Zein are closely related to the molecular structure. Zein and its modified products can encapsulate and protect lutein through various methods, creating more stable and efficient zein-loaded lutein nanosystems. Additionally, embedding lutein in Zein and its derivatives enhances lutein's digestive stability, solubility, antioxidant properties, and overall bioavailability.
A novel zein/ Auricularia cornea Ehrenb polysaccharides coated quercetin nanocarrier: Characterization, stability, antioxidant activity, and bioaccessibility

Abstract:This study aimed to develop a novel and stable delivery system based on zein and Auricularia cornea Ehrenb polysaccharides (ACEP) for quercetin (Que). The pH-driven self-assembly combined with electrostatic deposition methods were employed to construct zein-ACEP nanoparticles (ZA NPs). The optimized nanocomposite was prepared at zein: ACEP mass ratios of 3:2, and it had superior ionic strength stability (0–75 mM NaCl), thermal stability (80°C for 30 min), average diameter of 185.6 nm, zeta-potential of −42.8 mV, and Que encapsulation efficiency of 75.8 %. The assembly of composites was facilitated primarily by hydrophobic, hydrogen-bonding, and electrostatic interactions. Que encapsulated with ZA NPs exhibited superior light stability, stronger antioxidant activity, and bioaccessibility than Que. Additionally, ZA NPs showed sustainable release capacity, and Que release from nanoparticles is consistent with an anomalous (non-Fickian) transport dominated by skeleton erosion. These findings demonstrate that ZA NPs have potential applications as an effective and viable delivery system for Que in functional food fields.

Abstract:Genistein is one of major isoflavones derived from soybean products and it is believed to have beneficial effects on human health. However, its low water-solubility and poor oral bioavailability severely hamper its use as a functional food ingredient or for pharmaceutical industry. In this study, zein and zein/carboxymethyl chitosan (CMCS) nanoparticles were prepared to encapsulate genistein using a combined liquid–liquid phase separation method. The physicochemical properties of fabricated nanoparticles were characterized by dynamic light scattering (DLS), atomic force microscopy (AFM), and Fourier transform infrared spectroscopy (FTIR). The results demonstrated that genistein encapsulated with zein nanoparticles significantly improved its water dispersibility, antioxidant activity in the aqueous phase, and photostability against UV light. Moreover, genistein encapsulated in zein nanoparticles showed a sustained release property. Furthermore, it was found that encapsulation efficiency of genistein was significantly enhanced after CMCS coating, and this effect was more pronounced after the complex nanoparticles cross-linked with calcium ions when compared with the use of zein as a single encapsulant. In addition, compared to zein nanoparticles without biopolymer coating, CMCS coating significantly enhanced the thermal and storage stability of the formed nanoparticles, and delayed the release of genistein. A schematic diagram of zein and zein/carboxymethyl chitosan (CMCS) nanoparticles formation mechanism for encapsulation of genistein was proposed. According to the results of the current study, it could be concluded that encapsulation of genistein in zein/CMCS nanoparticles is a promising approach to improve its water dispersibility, antioxidant activity, photostability against UV light and provide controlled release for food/pharmaceutical applications.

Abstract:Herein, we reviewed zein-based potential platforms as therapeutic cues for oral drug delivery.Intensive research efforts are being focused on engineering zein-based micro- and nano-constructs.Zein-based constructs are stable, robust, and recoverable than that of free counterparts.The selection of polymer with biologically tunable functionalities is a key point for its suitable application. Natural polymers (especially proteins) are preferred over synthetic counterparts, in this regard. Zein is a natural protein obtained from maize (corn) having useful characteristics like biocompatibility, biodegradability and hydrophobic in nature. The utilization of polymers with multifunctional characteristics, as an excipient, is a highly requisite strategy for the improvement of polymer-based delivery systems. Preference of oral route of drug administration has always motivated researchers to put their efforts in improvement and development of oral drug delivery systems particularly for drugs that have low oral bioavailability or required targeting. Recently, zein has been utilized by a number of researchers for the improvement of oral drug delivery systems such as enhancement of bioavailability, preparing sustained release dosage form and also targeting/protecting drugs. In this review, we have enlightened applications of zein in the development of different oral drug delivery systems. Novel Fabrication of Zein-Soluble Soybean Polysaccharide Nanocomposites Induced by Multifrequency Ultrasound, and Their Roles on Microstructure, Rheological Properties and Stability of Pickering Emulsions

Abstract:In this work, soluble soybean polysaccharides (SSPS) were employed together with multifrequency ultrasound to fabricate zein nanocomposites which were conducive to enhancing the stability of high internal phase emulsions (HIPEs). Compared with non-ultrasonic treated zein colloidal particle samples (132.23 ± 0.85 nm), the zein nanoparticles samples induced by dual-frequency ultrasound exhibited a smaller particle size (114.54 ± 0.23 nm). Furthermore, the particle size of the zein composite nanoparticles (256.5 ± 4.81) remarkably increased with SPSS coating, consequently leading to larger fluorescence intensity together with lower zeta-potential (−21.90 ± 0.46 mv) and surface hydrophobicity (4992.15 ± 37.28). Meanwhile, zein-SSPS composite nanoparticles induced by DFU showed remarkably enhanced thermal stability. Fourier transform infrared (FTIR) spectroscopy and Circular dichroism (CD) spectroscopy were also used to characterize zein-SSPS composite nanoparticles. The results confirmed that DFU combined with SSPS treatment significantly increased β-sheets (from 12.60% ± 0.25 b to 21.53% ± 0.37 c) and reduced α-helix content (34.83% ± 0.71 b to 23.86% ± 0.66 a) remarkably. Notably, HIPEs prepared from zein-SSPS nanocomposites induced by dual-frequency simultaneous ultrasound (DFU) at 40/60 kHz showed better storage stability. HIPEs stabilized by DFU induced zein-SSPS nanoparticles exhibited higher storage modulus (G′) and loss modulus (G′′), leading to lower fluidity, together with better stability contributing to the water-binding capacity and three-dimensional (3D) network structure of the HIPEs emulsion. The findings of this study indicate that this method can be utilized and integrated to further extend the application of zein and SSPS and explore HIPEs.

Abstract:Zein, as one of the natural and GRAS proteins in plant, is renewable, nontoxic, biocompatible and biodegradable. Over the past decade, many research efforts have been devoted to zein-based biomaterials for several industrial applications. Combining with research experiences in our research group, the preparation methods, characterizations and pharmaceutical applications of zein-based nanoparticles were summarized in this review. Zein NPs with different particle nanostructures have been prepared by chemical crosslinking, desolvating, dispersing and micromixing strategies. The pharmaceutical applications of zein NPs are mainly focus on the drug delivery. Zein NPs can improve the drug stability, increase the oral bioavailability, control the drug release and enhance the drug targeting, thereby improving the pharmaceutical effect effectively. More efforts are required to analyze the relationship among preparation methods, particle nanostructures and pharmaceutical properties in virtue of quality by design approach, and further promote the scale-up production and clinical application of zein NPs.

Abstract:Nanomaterials, due to their unique structural and functional features, are widely investigated for potential applications in a wide range of industrial sectors. In this context, protein-based nanoparticles, given proteins’ abundance, non-toxicity, and stability, offer a promising and sustainable methodology for encapsulation and protection, and can be used in engineered nanocarriers that are capable of releasing active compounds on demand. Zein is a plant-based protein extracted from corn, and it is biocompatible, biodegradable, and amphiphilic. Several approaches and technologies are currently involved in zein-based nanoparticle preparation, such as antisolvent precipitation, spray drying, supercritical processes, coacervation, and emulsion procedures. Thanks to their peculiar characteristics, zein-based nanoparticles are widely used as nanocarriers of active compounds in targeted application fields such as drug delivery, bioimaging, or soft tissue engineering, as reported by others. The main goal of this review is to investigate the use of zein-based nanocarriers for different advanced applications including food/food packaging, cosmetics, and agriculture, which are attracting researchers’ efforts, and to exploit the future potential development of zein NPs in the field of cultural heritage, which is still relatively unexplored. Moreover, the presented overview focuses on several preparation methods (i.e., antisolvent processes, spry drying), correlating the different analyzed methodologies to NPs’ structural and functional properties and their capability to act as carriers of bioactive compounds, both to preserve their activity and to tune their release in specific working conditions.

Abstract:In this study, a combination of whey protein (hydrophilic coating) and polydopamine (crosslinking agent) was used to improve the stability and functionality of quercetin-loaded zein nanoparticles. There are two key benefits of the core-shell nanoparticles formed. First, the ability of the polydopamine to bind to both zein and whey protein facilitates the formation of a stable core-shell structure, thereby protecting quercetin from any pro-oxidants in the aqueous surroundings. Second, neutral and hydrophilic whey proteins were used for the surface coating of the nanoparticles to further enhance the sustained and slow release of quercetin, facilitating its sustained release into the body at a slow and steady rate. The results of this study will promote the innovative development of precise nutritional delivery systems for zein and provide a theoretical basis for the design and development of dietary supplements based on hydrophobic food nutrient molecules.

Abstract:The application of zein in food preservation was discussed from a unique perspective of application types, including coating and film. For the study of coating, edibility is considered because the coating adheres to the surface of food directly. For the study of film, plasticizers improve their mechanical properties, while barrier performance and antibacterial performance are achieved by nanoparticles; the incorporation of polyphenols is mainly due to their antibacterial and antioxidant properties; other biopolymers realize the complementarity between zein and biopolymers within films. In the future, the interaction between the edible coating and food matrix needs to be concerned. The mechanism of various exogenous additives and zein in the film should be noticed. Importantly, food safety and the possibility of large-scale application should be followed. Additionally, the intelligent response is one of the key development directions of zein-based film in the future.

Abstract:Characterization of zein aqueous solutions, as a function of the ethanol content and pH, was performed, giving information on the zein aggregation state for the construction of complexes. The aggregation state and surface charge of zein was found to depend on the mixed solvent composition and pH. Nonstoichiometric complex nanoparticles (NPECs) were prepared by electrostatically self-assembling zein, as the polycation, and sodium alginate or chondroitin sulfate, as the polyanions, at a pH of 4. A wide range of parameters were investigated: the alcohol–water content in the zein solutions, the charge molar ratios, the solution addition order and the addition rate. The resulting nanoparticles were characterized by dynamic and electrophoretic light scattering, circular dichroism and scanning electron microscopy. The smallest size for the NPECs (100 nm) was obtained when the polysaccharides acted as the titrate with an addition rate of 0.03 mL·min−1. The NPECs with the best characteristics were selected for loading with ciprofloxacin and then deposited on a cellulosic material in order to evaluate their antibacterial activity. Substantial drug encapsulation with desired drug release profiles were found together with notable antibacterial efficiency, showing the tunability of the properties for both the zein and its complexes with polysaccharides, together with their application potential in the biomedical field.
