Abstract:Concentration of air pollutants, particularly ozone (O3), has dramatically increased since pre-industrial times in the troposphere. Due to the strong oxidative potential of O3, negative effects on both emission and lifetime in the atmosphere of plant volatile organic compounds (VOCs) have already been highlighted. VOCs alteration by O3 may potentially affect the attraction of pollinators that rely on these chemical signals. Surprisingly, direct effects of O3 on the olfaction and the behavioral response of pollinators have not been investigated so far. We developed a comprehensive experiment under controlled conditions to assess O3 physiological and behavioral effects on two pollinator species, differing in their ecological traits. Using several realistic concentrations of O3 and various exposure times, we investigated the odor antennal detection and the attraction to VOCs present in the floral scents of their associated plants. Our results showed, in both species, a clear effect of exposure to high O3 concentrations on the ability to detect and react to the floral VOCs. These effects depend on the VOC tested and its concentration, and the O3 exposure (concentration and duration) on the pollinator species. Pollination systems may, therefore, be impaired in different ways by increased levels of O3, the effects of which will likely depend on whether the exposure is chronic or, as in this study, punctual, likely causing some pollination systems to be more vulnerable than others. While several studies have already shown the negative impact of O3 on VOCs emission and lifetime in the atmosphere, this study reveals, for the first time, that this impact alters the pollinator detection and behavior. These findings highlight the urgent need to consider air pollution when evaluating threats to pollinators.

Abstract:Fine needle aspiration (FNA) is a diagnostic tool whose inexpensiveness, simplicity and innocuousness have led it to be increasingly accepted in daily medical practice. This method, which is useful for the study of lesions that are accessible by radiological exploration or palpation, provides information about the cytological aspects of punctured lesions. Sometimes, such information can be extended to the histological area through study of cell blocks, true microbiopsies, which are of great diagnostic use. This was the technique used in the case described in the present report.

Abstract:Tropospheric ozone (O3) mixing ratios have increased substantially since preindustrial times and high O3 peaks are increasingly common. Plant-pollinator interactions are central to natural ecosystem functioning and food production systems but could be negatively affected by unfavourable environmental conditions such as elevated O3. Ecosystem functioning is threatened by O3, which can degrade floral volatile organic compounds (VOCs) used by pollinators as olfactory cues during foraging. It can also exert oxidative stress on VOC-emitting plants and receiving organisms, potentially disturbing the sending and receiving of VOC signals. The aim of this study was to determine the effects of elevated ozone on the foraging behaviour of Bombus terrestris on three species of the Brassicaceae, with a particular focus on bumblebee choices and the mechanisms underpinning differences observed. Moreover, the study was designed to fill a gap between observations in small-scale laboratory experiments and large-scale modelling through empirical observations in polytunnels that represent a medium-large-scale artificial environment. Using 10 × 3 × 2 m polytunnels the effects of O3 on pollinator foraging parameters on Sinapis alba, Sinapis arvensis and Raphanus raphanistrum were assessed. Significant effects of elevated O3 (100 ± 10 ppb) on the time taken for the first bee to alight on a flower and the cumulative amount of time spent on flowers was observed. To further investigate the underlying mechanisms, a laboratory test was conducted to determine the effects of ozone on the VOC blend composition of S. alba flowers. Synthetic VOC blends representing O3-altered and unaltered profiles were reconstituted and utilized in polytunnel and olfactometry experiments. The results indicated that a reduction of olfaction-mediated orientation, probably via VOC-degradation or direct effects of O3 on bees, was responsible for the altered foraging parameters of B. terrestris, suggesting that the presence of elevated O3 could have negative effects on the foraging efficiency of important pollinator species.

Abstract:Cytokines, particularly interferons, may participate in the development of type I diabetes. This involvement could be from direct cytotoxic actions of the interferons on the pancreatic beta-cells or from an indirect influence on the number, activity, or type of inflammatory cells that invade the islets in type I diabetes. To examine directly the role of interferon (IFN)-gamma in a mouse model of type I diabetes, we have introduced an inactivating mutation in the IFN-gamma gene (ifg) into NOD mice. The genetic absence of IFN-gamma does not prevent either insulitis or diabetes in the NOD mice, but it does increase the time to onset. Although it might have been predicted that the absence of IFN-gamma in these mice would lead to an increase in expression of Th2 T-helper cell-related cytokines, we found instead a profound decrease in the expression of two of the characteristic Th2 cytokines, interleukin (IL)-4 and IL-10. We also demonstrate that the splenocytes taken from IFN-gamma-deficient diabetic mice are fully capable of transferring diabetes to naive recipients.
Atmospheric transformation of plant volatiles disrupts host plant finding

Abstract:Plant-emitted volatile organic compounds (VOCs) play important roles in plant-insect interactions. Atmospheric pollutants such as ozone (O3) can react with VOCs and affect the dynamics and fidelity of these interactions. However, the effects of atmospheric degradation of plant VOCs on plant-insect interactions remains understudied. We used a system comprising Brassica oleracea subsp. capitata (cabbage) and the specialist herbivore Plutella xylostella to test whether O3-triggered VOC degradation disturbs larval host orientation and to investigate the underlying mechanisms. Larvae oriented towards both constitutive and larva-induced cabbage VOC blends, the latter being the more attractive. Such behaviour was, however, dramatically reduced in O3-polluted environments. Mechanistically, O3 rapidly degraded VOCs with the magnitude of degradation increasing with O3 levels. Furthermore, we used Teflon filters to collect VOCs and their reaction products, which were used as odour sources in behavioural tests. Larvae avoided filters exposed to O3-transformed VOCs and spent less time searching on them compared to filters exposed to original VOCs, which suggests that some degradation products may have repellent properties. Our study clearly demonstrates that oxidizing pollutants in the atmosphere can interfere with insect host location and highlights the need to address their broader impacts when evaluating the ecological significance of VOC-mediated interactions.

Abstract:Human activities, urbanization, and industrialization contribute to pollution that affects climate and air quality. A main atmospheric pollutant, the tropospheric ozone (O3), can damage living organisms by generating oxidative radicals, causing respiratory problems in humans and reducing yields and growth in plants. Exposure to high concentrations of O3 can result in oxidative stress in plants and animals, eventually leading to substantial ecological consequences. Plants produce volatile organic compounds (VOCs) emitted in the environment and detected by pollinators (mainly by their antennae), foraging for nutritious resources. Several pollinators, including honey bees, recognize and discriminate flowers through olfactory cues and memory. Exposure to different concentrations of O3 was shown to alter the emission of floral VOCs by plants as well as their lifetime in the atmosphere, potentially impacting plant-pollinator interactions. In this report, we assessed the impacts of exposure to field-realistic concentrations of O3 on honey bees' antennal response to floral VOCs, on their olfactory recall and discriminative capacity and on their antioxidant responses. Antennal activity is altered depending on VOCs structure and O3 concentrations. During the behavioral tests, we first check consistency between olfactory learning rates and memory scores after 15 min. Then bees exposed to 120 and 200 ppb of ozone do not exert specific recall responses with rewarded VOCs 90 min after learning, compared to controls whose specific recall responses were consistent between time points. We also report for the first time in honey bees how the superoxide dismutase enzyme, an antioxidant defense against oxidative stress, saw its enzymatic activity rate decreases after exposure to 80 ppb of ozone. This work tends to demonstrate how hurtful can be the impact of air pollutants upon pollinators themselves and how this type of pollution needs to be addressed in future studies aiming at characterizing plant-insect interactions more accurately.

Abstract:There is growing concern about sensory pollutants affecting ecological communities. Anthropogenically enhanced oxidants [ozone (O 3 ) and nitrate radicals (NO 3 )] rapidly degrade floral scents, potentially reducing pollinator attraction to flowers. However, the physiological and behavioral impacts on pollinators and plant fitness are unknown. Using a nocturnal flower-moth system, we found that atmospherically relevant concentrations of NO 3 eliminate flower visitation by moths, and the reaction of NO 3 with a subset of monoterpenes is what reduces the scent's attractiveness. Global atmospheric models of floral scent oxidation reveal that pollinators in certain urban areas may have a reduced ability to perceive and navigate to flowers. These results illustrate the impact of anthropogenic pollutants on an animal's olfactory ability and indicate that such pollutants may be critical regulators of global pollination.

Abstract:Plants produce distinct blends of volatile compounds that attract pollinators (floral odors) or natural enemies of insect herbivores (herbivore-induced plant volatiles). The admixture of these blends in the atmosphere may alter the attraction of insect mutualists and ultimately affect plant fitness. Here, using synthetic blends of Brassica rapa floral volatiles and real B. rapa flowers, we investigated how floral odors impact the foraging behavior of parasitoids. In an olfactometer setting, floral odors reduced the attractiveness of plants infested by herbivores to parasitoids by 43.5% and affected four out of five parasitoid species tested. Additionally, experiments with the parasitoid Cotesia glomerata revealed that the effects of floral odors are dose-dependent and that floral odors were less disruptive under wind tunnel conditions than under olfactometer conditions. Electroantennogram recordings showed that C. glomerata antennae do respond to floral compounds, but that floral compounds do not inhibit antennal responses to herbivore-induced leaf volatiles. In conclusion, floral odors can act as background pollutants decreasing the attractiveness of chemical blends used by natural enemies to locate their hosts. Under natural conditions, such interferences could affect the outcome of tritrophic interactions and may play an important role in the evolution of plant volatile signaling.

Abstract:Among air pollutants, tropospheric ozone (O 3 ) is one of the most stressful for organisms due to its strong oxidative potential. For instance, high ozone concentration ([O 3 ]) has the potential to affect (i) the emission of volatile organic compounds (VOCs) by plants and (ii) the lifetime of these VOCs in the atmosphere, and consequently disturb crucial signals in the interactions between plants and other organisms. However, despite the determinant role of VOCs emitted by flowers for pollinator attraction, a very limited number of studies have investigated the impact of O 3 on floral VOCs. In this study, we investigated the effect of high [O 3 ] episodes on the VOCs emitted by a flowering Mediterranean plant: the true lavender ( Lavandula angustifolia Mill., Lamiaceae). To do so, in controlled conditions, we exposed (i) the entire plant to high but realistic [O 3 ] (200 ppb for 5 h) and (ii) only the VOCs emitted by lavender to increasing [O 3 ] (0, 40, 80, 120, and 200 ppb). We sampled VOCs of lavender in both conditions and analyzed them by Gas Chromatography-Mass Spectrometry in order to qualify and quantify the flowering lavender’s emissions and the reaction of VOCs with O 3 in the atmosphere. Our results showed that exposure to high [O 3 ] during a short period (5 h) did not affect the emission of VOCs by flowering lavender. Incidentally, we also showed that the chemical signal varied in quantities and proportions over the day. Moreover, we showed that after their emission by the plant, composition of the VOCs changed quantitatively and qualitatively in an atmosphere containing [O 3 ] naturally observed nowadays. Quantities of several of the major terpenes emitted by lavender decreased drastically during O 3 exposure, whereas concentrations of some VOCs increased, such as carbonyls and carboxylic acids, which are probably reaction products of terpenes with O 3 . Exposure to high [O 3 ] thus directly affected the proportions of VOCs in the atmosphere. Because pollinators generally use a blend of VOCs in particular proportions as a signal to localize flowers, the numerous pollinators of lavender may experience difficulty in recognizing specific floral odors during frequent and moderate [O 3 ] episodes in the Mediterranean region.

Abstract:Biogenic volatile organic compounds (bVOCs), synthesised by plants, are important mediators of ecological interactions that can also undergo a series of reactions in the atmosphere. Ground-level ozone is a secondary pollutant generated through sunlight-driven reactions between nitrogen oxides (NO x ) and VOCs. Its levels have increased since the industrial revolution and reactions involving ozone drive many chemical processes in the troposphere. While ozone precursors often originate in urban areas, winds may carry these hundreds of kilometres, causing ozone formation to also occur in less populated rural regions. Under elevated ozone conditions, ozonolysis of bVOCs can result in quantitative and qualitative changes in the gas phase, reducing the concentrations of certain bVOCs and resulting in the formation of other compounds. Such changes can result in disruption of bVOC-mediated behavioural or ecological interactions. Through a series of gas-phase experiments using Gas Chromatography Mass Spectrometry (GC-MS) and Proton Transfer Reaction Mass Spectrometry (PTR-MS), we investigated the products and their yields from the ozonolysis of a range of ubiquitous bVOCs, which were selected because of their importance in mediating ecological interactions such as pollinator and natural enemy attraction and plant-to-plant communication, namely: ( E )-β-ocimene, isomers of α and β-farnesene, α-terpinene and 6-methyl-5-hepten-2-one. New products from the ozonolysis of these compounds were identified, and the formation of these compounds is consistent with terpene-ozone oxidation mechanisms. We present the degradation mechanism of our model bVOCs and identify their reaction products. We discuss the potential ecological implications of the degradation of each bVOC and of the formation of reaction products.

Tissue-Specific Emission of (E)-Α-bergamotene Helps Resolve the Dilemma when Pollinators Are Also Herbivores

Abstract:More than 87% of flowering plant species are animal-pollinated [1] and produce floral scents and other signals to attract pollinators. These floral cues may however also attract antagonistic visitors, including herbivores [2]. The dilemma is exacerbated when adult insects pollinate the same plant that their larvae consume. It remains largely unclear how plants maximize their fitness under these circumstances. Here we show that in the night-flowering wild tobacco Nicotiana attenuata, the emission of a sesquiterpene, (E)-α-bergamotene, in flowers increases adult Manduca sexta moth-mediated pollination success, while the same compound in leaves is known to mediate indirect defense against M. sexta larvae [3, 4]. Forward and reverse genetic analyses demonstrated that both herbivory-induced and floral (E)-α-bergamotene are regulated by the expression of a monoterpene-synthase-derived sesquiterpene synthase (NaTPS38). The expression pattern of NaTPS38 also accounts for variation in (E)-α-bergamotene emission among natural accessions. These results highlight that differential expression of a single gene that results in tissue-specific emission of one compound contributes to resolving the dilemma for plants when their pollinators are also herbivores. Furthermore, this study provides genetic evidence that pollinators and herbivores interactively shape the evolution of floral signals and plant defense.

Abstract:Currently, a global analysis of the information available on the relative composition of the floral scents of a very diverse variety of plant species is missing. Such analysis may reveal general patterns on the distribution and dominance of the volatile compounds that form these mixtures, and may also allow measuring the effects of factors such as the phylogeny, pollination vectors, and climatic conditions on the floral scents of the species. To fill this gap, we compiled published data on the relative compositions and emission rates of volatile organic compounds (VOCs) in the floral scents of 305 plant species from 66 families. We also gathered information on the groups of pollinators that visited the flowers and the climatic conditions in the areas of distribution of these species. This information allowed us to characterize the occurrence and relative abundances of individual volatiles in floral scents and the effects of biotic and climatic factors on floral scent. The monoterpenes trans-β-ocimene and linalool and the benzenoid benzaldehyde were the most abundant floral VOCs, in both ubiquity and predominance in the floral blends. Floral VOC richness and relative composition were moderately preserved traits across the phylogeny. The reliance on different pollinator groups and the climate also had important effects on floral VOC richness, composition, and emission rates of the species. Our results support the hypothesis that key compounds or compounds originating from specific biosynthetic pathways mediate the attraction of the main pollinators. Our results also indicate a prevalence of monoterpenes in the floral blends of plants that grow in drier conditions, which could link with the fact that monoterpene emissions protect plants against oxidative stresses throughout drought periods and their emissions are enhanced under moderate drought stress. Sesquiterpenes, in turn, were positively correlated with mean annual temperature, supporting that sesquiterpene emissions are dominated mainly by ambient temperature. This study is the first to quantitatively summarise data on floral-scent emissions and provides new insights into the biotic and climatic factors that influence floral scents.

Abstract:Tropospheric ozone (O 3 ) is likely to affect the chemical signal emitted by flowers to attract their pollinators through its effects on the emission of volatile organic compounds (VOCs) and its high reactivity with these compounds in the atmosphere. We investigated these possible effects using a plant-pollinator interaction where the VOCs responsible for pollinator attraction are known and which is commonly exposed to high O 3 concentration episodes: the Mediterranean fig tree ( Ficus carica ) and its unique pollinator, the fig wasp ( Blastophaga psenes ). In controlled conditions, we exposed fig trees bearing receptive figs to a high-O 3 episode (5 h) of 200 ppb and analyzed VOC emission. In addition, we investigated the chemical reactions occurring in the atmosphere between O 3 and pollinator-attractive VOCs using real-time monitoring. Finally, we tested the response of fig wasps to the chemical signal when exposed to increasing O 3 mixing ratios (0, 40, 80, 120 and 200 ppb). The exposure of the fig tree to high O 3 levels induced a significant decrease in leaf stomatal conductance, a limited change in the emission by receptive figs of VOCs not involved in pollinator attraction, but a major change in the relative abundances of the compounds among pollinator-attractive VOCs in O 3 -enriched atmosphere. Fig VOCs reacted with O 3 in the atmosphere even at the lowest level tested (40 ppb) and the resulting changes in VOC composition significantly disrupted the attraction of the specific pollinator. These results strongly suggest that current O 3 episodes are probably already affecting the interaction between the fig tree and its specific pollinator.

Fuel and Fluid Homeostasis During Long-Term Exercise in Healthy Subjects and Type I Diabetic Patients

Abstract:OBJECTIVE This study was designed to examine metabolic and hormonal effects of long-term exercise in healthy subjects and insulin-dependent (type I) diabetic patients. RESEARCH DESIGN AND METHODS Two studies were performed. First, 16 healthy males (32 ± 3 yr) were studied during a semitriathlon competition (2 km swimming, 90 km biking, and 21 km running). Second, 9 type I diabetic males (41 ± 2 yr) and 17 healthy matched control subjects were studied during a 75 km cross-country skiing race. Blood samples were taken before and immediately after exercise, and also during the ski race. RESULTS During the semitriathlon race, serum insulin, C-peptide, glucagon cortisol, growth hormone ACTH, prolactin, and plasma renin activity increased two- to ninefold, whereas serum testosterone fell. Apart from a fall in magnesium, serum electrolyte concentrations remained unchanged. Before long-term skiing, patients reduced their insulin dose by 30–40%. They were hyperglycemic during the initial part of the race, but near normoglycemic thereafter. There were large interindividual variations in the increments of counterregulatory hormones, whereas serum testosterone and luteinizing hormone fell quite uniformly. Plasma renin activity and aldosterone concentrations rose similarly in diabetic and healthy subjects, whereas the rise in antidiuretic hormone was slightly greater in diabetic patients. During the initial part of the race, serum atrial natriuretic peptide fell in both groups. CONCLUSIONS Severalfold increments in hormone concentrations contribute to the maintenance of fuel and fluid homeostasis during long-term exercise. With an appropriate adjustment of insulin dose and diet, also type I diabetic patients can participate in competitive long-term exercise.
Elevated ozone and carbon dioxide affects the composition of volatile organic compounds emitted by Vicia faba (L.) and visitation by European orchard bee (Osmia cornuta)

Abstract:Recent studies link increased ozone (O 3 ) and carbon dioxide (CO 2 ) levels to alteration of plant performance and plant-herbivore interactions, but their interactive effects on plant-pollinator interactions are little understood. Extra floral nectaries (EFNs) are essential organs used by some plants for stimulating defense against herbivory and for the attraction of insect pollinators, e.g., bees. The factors driving the interactions between bees and plants regarding the visitation of bees to EFNs are poorly understood, especially in the face of global change driven by greenhouse gases. Here, we experimentally tested whether elevated levels of O 3 and CO 2 individually and interactively alter the emission of Volatile Organic Compound (VOC) profiles in the field bean plant ( Vicia faba , L., Fabaceae), EFN nectar production and EFN visitation by the European orchard bee ( Osmia cornuta , Latreille, Megachilidae). Our results showed that O 3 alone had significant negative effects on the blends of VOCs emitted while the treatment with elevated CO 2 alone did not differ from the control. Furthermore, as with O 3 alone, the mixture of O 3 and CO 2 also had a significant difference in the VOCs’ profile. O 3 exposure was also linked to reduced nectar volume and had a negative impact on EFN visitation by bees. Increased CO 2 level, on the other hand, had a positive impact on bee visits. Our results add to the knowledge of the interactive effects of O 3 and CO 2 on plant volatiles emitted by Vicia faba and bee responses. As greenhouse gas levels continue to rise globally, it is important to take these findings into consideration to better prepare for changes in plant-insect interactions.

Abstract:Insects are a diverse group of organisms that provide important ecosystem services like pollination, pest control, and decomposition and rely on olfaction to perform these services. In the Anthropocene, increasing concentrations of oxidant pollutants such as ozone have been shown to corrupt odor-driven behavior in insects by chemically degrading e.g. flower signals or insect pheromones. The degradation, however, does not only result in a loss of signals, but also in a potential enrichment of oxidation products, predominantly small carbonyls. Whether and how these oxidation products affect insect olfactory perception remains unclear. We examined the effects of ozone-generated small carbonyls on the olfactory behavior of the vinegar fly Drosophila melanogaster. We compiled a broad collection of neurophysiologically relevant odorants for the fly from databases and literature and predicted the formation of the types of stable small carbonyl products resulting from the odorant's oxidation by ozone. Based on these predictions, we evaluated the olfactory detection and behavioral impact of the ten most frequently predicted carbonyl products in the fly using single sensillum recordings (SSRs) and behavioral tests. Our results demonstrate that the fly's olfactory system can detect the oxidation products, which then elicit either attractive or neutral behavioral responses, rather than repulsion. However, certain products alter behavioral choices to an attractive odor source of balsamic vinegar. Our findings suggest that the enrichment of small carbonyl oxidation products due to increased ozone levels can affect olfactory guided insect behavior. Our study underscores the implications for odor-guided foraging in insects and the essential ecosystem services they offer under carbonyl enriched environments.

Abstract:Plant volatiles, particularly biogenic volatile organic compounds (BVOCs), emitted in urban areas have attracted attention as olfactory signals between plants and other organisms, including insects. However, in urban areas, elevated ozone (O 3 ) levels inhibit plant growth and degrade olfactory signals, including both insect pheromones and BVOCs, resulting in disrupted biological communication. In this article, we review recent findings on how O 3 modifies olfactory interactions, focusing on both the emitters and receivers of these signals. The influence of O 3 on herbivorous insects and their enemies alters the pressure caused by herbivorous damage in the field, which can affect the development of the defensive capacities of plants at the hereditary level. To address the challenges posed by O 3 in biological interactions, BVOC characteristics (e.g., emission rate and species, blend composition, O 3 reactivity, and oxidative products) and O 3 effects on insects (e.g., preference and antennal detectivity) should be clarified. At the same time, BVOC emissions are expected to increase with rising temperatures, which will likely increase the impact of BVOCs on O 3 formation in the future. Therefore, it is necessary to devise strategies, such as selecting non- or low BVOC emitters, to regulate BVOC emissions from urban greening plants and mitigate O 3 risks to olfactory interactions and plant health. Abstract:Pollinators seek high quality nectar and pollen rewards critical for their growth and reproduction. Volatile emissions from inflorescences may signal plant health and floral resource quality to these flower-visiting insects. To understand the relationship between floral volatile emission and pollinator preference in a field setting, we conducted a replicated garden experiment consisting of 18 native perennial species from three different plant families with varying nutritional resource quality. We collected flower-visiting insects and floral volatile emissions over two field seasons, detecting over 60 volatile compounds and over 150 insect species. We collated trait data of visiting insects (bee or non-bee, body size, degree of sociality, nesting behavior, and whether the visitor was non-native) and evaluated interactions between these traits and volatile composition. Among the insect traits, bees and larger visitors had negative associations with sesquiterpenes, while in contrast visitors with complex nesting behaviors and social insects had positive associations. When comparing plant traits with bee traits, bee visitors had negative associations with purple inflorescences and larger insects and social insects both had positive associations with yellow inflorescences. We tested for direct associations between volatile class and insect taxa and found that sesquiterpenes had a strong positive relationship with hoverfly presence. This work demonstrates the complexity of floral volatiles as signals for pollinating insects and how floral scent composition allows individual compounds to act synergistically or antagonistically. These results can help us to better understand pollinator preferences and visitation patterns in the broad context of chemical ecology.
