Abstract:Background: Malaria is a public health menace. Resistance to therapeutic armamentarium is impeding its control. Therefore, research targeting the discovery of novel antimalarial drug arsenals is a priority. The important point to begin the search for such drugs is the folkloric medicinal plants. Ripe fruit of Lagenaria siceraria is bored, rinsed with cold water, and one glass is used as a drink early in the morning for the treatment of malaria in Ethiopian folk medicine. In vivo antimalarial efficacy of the plant was not affirmed scientifically, though. Consequently, the present study was conducted to assess the in vivo antiplasmodial effect of Lagenaria siceraria in P. berghei infected mice. Methods: The fruits were extracted using 80% methanol in water. Acute toxicity test was conducted on the extract. Secondary phytochemicals were assessed. The four day suppressive test was employed in mice infected with P. berghei. Thirty mice were grouped in to five and inoculated with P. berghei. After 3 h, three of the groups received the extract at doses 100, 200 and 400 mg/kg. The remaining groups served as negative (2% Tween80) and positive control (chloroquine). Parasitemia, packed cell volume, weight, temperature and survival time were monitored. SPSS version 22 was used for data analysis. Results: No toxicity was seen in mice. The crude extract elicited significant suppression (p < 0.05) of the parasite compared to the negative control. The highest parasite suppression (77.37%) was measured at the upper dose. Furthermore, the crude extract significantly (p < 0.05) prevented body weight loss, anemia, reduction in temperature and prolonged the survival time compared to the negative control. Conclusion: This study asserted that the fruit of Lagenaria siceraria is enriched with in vivo antimalarial activity. Hence, further in depth antimalarial investigations on the plant is strongly recommended.

Abstract:Background: Malaria continues to wreak havoc on the well-being of the community. Resistant parasites are jeopardizing the treatment. This is a wake-up call for better medications. Folk plants are the key starting point for antimalarial drug discovery. After crushing and mixing the leaves of Coriandrum sativum with water, one cup of tea is drunk daily for a duration of three to five days as a remedy for malaria by local folks in Ethiopia. Additionally, in vitro experiments conducted on the plant leaf extract elsewhere have also demonstrated the plant's malaria parasite inhibitory effect. There has been no pharmacologic research to assert this endowment in animals, though. This experiment was aimed at evaluating the antimalarial efficacy of C. sativum in Plasmodium berghei infected mice. Methods: The plant's leaf was extracted using maceration with distilled water. The extract was examined for potential acute toxicity. An evaluation of secondary phytoconstituents was done. Standard antimalarial screening models (prophylactic, chemosuppressive, curative tests) were utilized to assess the antiplasmodial effect. In each test, thirty mice were organized into groups of five. To the three categories, the test substance was given at doses of 100, 200 and 400 mg/kg/day before or after the commencement of P. berghei infection. Positive and negative control mice were provided Chloroquine and distilled water, respectively. Rectal temperature, parasitemia, body weight, survival time and packed cell volume were ultimately assessed. Analysis of the data was performed using Statistical Package for Social Sciences. Results: No toxicity was manifested in mice. The extract demonstrated a significant inhibition of parasitemia (p < 0.05) in all the models. The inhibition of parasite load was highest with the upper dose in the suppressive test (82.74%) followed by the curative procedure (78.49%). Likewise, inhibition of hypothermia, weight loss hampering, improved survival and protection against hemolysis were elicited by the extract. Conclusions: The results of our experimental study revealed that the aqueous crude leaf extract of C. sativum exhibits significant antimalarial efficacy in multiple in vivo models involving mice infected with P. berghei. Given this promising therapeutic attribute, in depth investigation on the plant is recommended.

Abstract:Abstract— Crude synaptosomal (P2) preparations were obtained from the cerebella of rats in which the granule cell population had been selectively reduced by X‐irradiation treatment and from the cerebella of control animals. In the P2 fraction from control cerebella, the level of glutamate was greater than any other of the 5 amino acids measured and was 2‐fold higher than taurine, which was present at the next highest level. The content of taurine was slightly higher than that found for aspartate and was 3‐fold greater than that observed for GABA. Alanine and glycine were present in the lowest amounts. The levels of glutamate and aspartate were significantly (P < 0.05) lower by 25 and 15%, respectively, in the P2 fraction isolated from the X‐irradiated cerebella in comparison to control values. The content of taurine, GABA, glycine, and alanine were not changed by the X‐irradiation treatment. The uptake of 1.0 μm‐l‐[3H]glutamate and l‐[3H]aspartate was reduced approx 20% by X‐irradiation treatment, whereas the uptake of 1.0 μm‐[3H]GABA and [3H]taurine was unchanged. A more detailed kinetic analysis of l‐[3H]glutamate uptake revealed there was a 20% decrease in the Vmax value with X‐irradiation treatment and no change in the apparent Km value.
Impact of health maintenance organizations and fee-for-service on health care utilization among people with systemic lupus erythematosus.

Abstract:OBJECTIVE To compare health care utilization in people with systemic lupus erythematosus (SLE) in health maintenance organizations (HMOs) and fee-for-service (FFS). METHODS A structured survey was administered to a cohort of 982 people with SLE who were assembled between 2002 and early 2005. A total of 2,656 person-years of observation were completed by the end of 2005. In each year, respondents reported their health care utilization and whether they had HMO or FFS coverage. We compared health care utilization of those in HMOs and FFS, with and without adjustment for socioeconomic, demographic, and health characteristics using repeated-measures regression techniques. RESULTS Compared with people with SLE who were in FFS, those in HMOs were younger (3.3 years), received a diagnosis at an earlier age (3.6 years), had slightly less disease activity (0.4 on a 10-point scale), were more likely to be nonwhite (8.8%), were less likely to be below the poverty line (7.8%), and were less likely to have public insurance (29.7%). The 2 groups did not differ in other characteristics. On an unadjusted basis, subjects with SLE in HMOs had significantly fewer physician visits (3.1; 95% confidence interval [95% CI] 1.7, 4.5) and were less likely to report one or more outpatient surgical visits (6.3%; 95% CI 2.5, 10.0), and hospital admissions (5.5%; 95% CI 1.7, 9.3) than those in FFS. Adjustment reduced the differences in physician visits (2.3; 95% CI 1.1, 3.5), outpatient surgical rates (4.4%; 95% CI 0.6, 8.1), and hospital admission rates (4.0%, 95% CI 0.4, 7.7). CONCLUSION Subjects with SLE in HMOs utilized substantially less ambulatory care and were less likely to have outpatient surgery and hospital admissions than those in FFS; the effects were not completely explained by socioeconomic, demographic, and health characteristics.

Abstract:Malaria, caused by Plasmodium parasites, is a global health concern. Natural products, such as plant extracts, are investigated for new antimalarial agents. Spondias mombin, traditionally used for medicinal purposes, was examined for its effects on parasitemia, haematological parameters, antioxidant activities, liver function biomarkers, and serum electrolytes in Plasmodium berghei‐infected mice. Mice received varying doses of the ethylacetate extract. Various dosages of the extract and the prescribed medication dramatically slowed the parasite's growth. The extract significantly improved red blood cell (RBC) and platelet counts, particularly at higher doses. Hemoglobin levels were decreased dose‐dependently. Antioxidant analysis showed increased superoxide dismutase (SOD), glutathione peroxidase (GPx), and reduced catalase (CAT) activities and malondialdehyde (MDA) levels, indicating robust antioxidant properties. Liver function tests showed mixed effects, with elevated AST, ALP, and TB levels at some doses, yet decreased ALT at the highest dose, suggesting possible hepatoprotective effects. Serum electrolyte levels remained stable. Seventy‐eight (78) bioactive components were identified in the extract by GC‐MS analysis. The extract demonstrated substantial antimalarial and antioxidant capacities, with minimal liver stress and stable electrolyte levels, suggesting safety as an adjunct therapy for malaria. Further research is needed to ascertain the specific antimalarial bioactive constituents and their mechanisms of action.

The antimalarial profiling of Mangifera indica's herbal formulation attenuated biochemical alterations and improved hepatocytes' histoarchitecture in Plasmodium berghei-infected mice

Abstract:Plasmodium parasite causes malaria and affects the biochemical, physiological, and histoarchitecture of the hepatocytes and blood. The resultant effect leads to alterations in the metabolic activities of the liver, erythrocytes, as well as the buffer system. Therefore, we investigated the antiplasmodial activity, histomorphological studies of the hepatocytes and alterations in biochemical parameters in Plasmodium berghei-infected mice administered with the herbal formulation of aqueous extracts of Mangifera indica stem bark and leaves. The plant coarse leaves (250.71 g) and stem bark (509.34 g) were weighed to obtain their ratios, macerated in boiled distilled water (5 L) for 72 h, filtered, and concentrated to obtain the various extracts whereas LD50 calculation gave 5500.19 mg/kg. The extracts were administered to eleven groups of mice at a dosage of 300 mg/kg whereas artesunate and ACT served as the positive control drugs; the antiplasmodial profiling, biochemical, and histological evaluations followed standard protocols. The schizonticidal activity of the extracts were remarkable; moreover, the histological section of the liver (negative control) had increased deposition of hemozoin, sinusoidal congestions, activation of kupffer cells, and portal tract inflammations; however, the other treatment groups in the study drastically reduced inflammation. The biochemical parameters' results revealed metabolic acidosis mitigation; hypocholesterolemia induction; enhanced hyperproteinemia, as well as hypoglycemia mitigation. The antiplasmodial therapeutic response, and biochemical derangements reversal corroborated with improved hepatocytes histoarchitecture of mice highlights the plant's pharmacological efficacy. Abstract:BackgroundMedicinal plant research may contribute to develop new pharmacological control tools for vector borne diseases, such as malaria.MethodsThe effects of methanol extracts (ME) obtained from seed kernel of ripe and unripe Azadirachta indica fruits were studied on erythrocytic proliferation of the rodent malaria parasite Plasmodium berghei strain ANKA and on mice pro-inflammatory response, as evaluated by measuring the matrix-metalloproteinase-9 (MMP-9) and tumour necrosis factor (TNF) plasma levels, in two mouse strains (C57BL/6 and BALB/c) which are considered as prototypical of Th1 and Th2 immune response, respectively.ResultsME obtained from seed kernel of unripe Azadirachta indica fruits decreased by about 30% the proportion of erythrocytes infected with the malaria parasite in C57BL/6 mice in the 4 days suppressive test. In this treatment group, MMP-9 and TNF levels were notably higher than those measured in the same mouse strain treated with the anti-malarial drug artesunate, Azadirachta indica kernel extracts from ripe fruits or solvent. In BALB/c mice, treatment with kernel extracts did not influence parasitaemia. MMP-9 and TNF levels measured in this mouse strain were notably lower than those recorded in C57BL/6 mice and did not vary among treatment groups.ConclusionsThe effects of the ME on the parasite-host interactions appeared to be mouse strain-dependent, but also related to the ripening stage of the neem fruits, as only the unripe fruit seed kernel extracts displayed appreciable bioactivity.

Abstract:Purpose: To investigate the effect of Baphia nitida ethanol leaf extract (BNELE) on heamatological and biochemical indices in Plasmodium berghei-infected mice. Methods: A total of thirty (30) male mice were divided into 6 groups comprising of 5 mice each. Group 1 was normal control, group 2 positive control, group 3 standard group while groups 4 - 6 received 200, 400 and 600 mg/kg of BNELE, respectively. Phytochemical analysis and antimalaria effect of BNELE were investigated using standard methods. Biochemical and haematological parameters of Plasmodium berghei - infected mice given various BNELE dosages were measured using standard techniques. Results: At increasing doses (200, 400, and 600 mg/kg), BNELE produced 63.16, 39.50 and 52.86 % inhibition of parasitemia in P. berghei-passaged mice, respectively, comparable to 43.75 % by 80 mg/kg artemether, a first-line anti-malarial medication. There was significant increase in red blood cell (RBC), packed cell volume (PCV), white blood cell (WBC) and hemoglobulin (HB) in mice administered varying doses of BNELE compared with those in group 2 (p < 0.05). Conclusion: Baphia nitida ethanol leaf extract possesses antiplasmodial properties and aids the restoration of normal haematological and biochemical states in P. berghei-infected mice. However, further and more extensive in vivo studies to ascertain it full antimalarial potentials.

Abstract:Malaria remains a critical global health issue, particularly in tropical and subtropical regions. The disease, caused by Plasmodium parasites, is transmitted by Anopheles mosquitoes and can lead to severe complications and death if untreated. The emergence of drug‐resistant strains highlights the urgent need for new antimalarial agents. Gnetum gnemon, a plant native to Southeast Asia, has shown promise due to its rich bioactive compounds. This study aims to evaluate the suppressive, curative, and prophylactic antimalarial potential of Gnetum gnemon leaf extract (GGE) against Plasmodium berghei in mice. GGE was prepared using a combination of hot water extraction and microwave‐assisted heating. Acute toxicity tests revealed no significant adverse effects at a dose of 3000 mg/kg. The doses of 100, 200, and 400 mg/kg were selected based on preliminary toxicity assessments to systematically investigate the dose‐dependent antimalarial efficacy of the extract. Suppressive tests showed that GGE at doses of 100, 200, and 400 mg/kg significantly reduced parasitemia levels, with the highest dose achieving a 63.97% inhibition. In these tests, GGE also increased the mean survival time (MST) of treated mice compared to untreated controls. However, GGE did not exhibit significant curative effects, as parasitemia levels in the treated groups were similar to the untreated control group. Prophylactic tests indicated that GGE pretreatment did not significantly reduce parasitemia levels or improve MST compared to controls, unlike chloroquine (CQ), which demonstrated potent prophylactic efficacy with a significant increase in MST. These findings suggest that while GGE has notable suppressive antimalarial activity, it does not exhibit strong curative or prophylactic effects at the tested doses. This study contributes to the understanding of plant‐based antimalarial agents and underscores the importance of continued exploration of natural products for malaria treatment.

Abstract:Ethnopharmacological relevance Solanum incanum L. is commonly used in traditional herbal medicine (THM) in Kenya for treating various ailments. Recent developments in disease treatment have introduced the concept of host-directed therapy (HDT). This approach involves targeting factors within the host cell that can impede the growth or replication of a pathogen. One such host factor is delta aminolevulinate dehydratase (δ-ALAD), the second enzyme in the heme biosynthesis pathway utilized by Plasmodium for growth. Studies using mice models have shown an increase in δ-ALAD expression during Plasmodium berghei infection. Another plant in the Solanum genus, S. guaranticum , has been found to inhibit δ-ALAD in red blood cells in vitro and in the brain in vivo . Is it possible that the bioactive compounds in S. incanum extracts could also be effective in HDT for malaria treatment? Aim of study To better assess the effectiveness of S. incanum leaf extracts as a curative and prophylaxis in malaria parasite infection, and to test the plant's ability to decrease δ-ALAD expression. Materials and methods The leaves of S. incanum were collected, dried, and pulverized before being subjected to a successive extraction protocol to obtain crude, hexane, ethyl acetate, and aqueous extract fractions. Phytochemical analysis was conducted on all extract fractions, followed by GC-MS analysis of the fraction with the most potent antimalarial activity. An acute toxicity study was also performed on the extracted fractions. The potency of the extract fractions as curative and prophylactic antimalarial was then evaluated in THM using Plasmodium berghei -infected mice at a dose of 100 mg/kg. The extract fraction with the highest activity was further evaluated at varying doses and its effect on δ-ALAD was measured using RT-qPCR. The percentage of parasitemia and chemosuppression, and mean survival time were used as indices of activity. Results Phytochemical analysis revealed that the ethyl acetate and aqueous extract fractions contained high terpenoids, flavonoids, and phenols levels. However, alkaloids were only present in moderate quantities in the aqueous extract, and quinones were found in high levels only in the crude extract. Additionally, all extract fractions contained saponins in high levels but lacked tannins. While the plant extracts were found to be non-toxic, they did not exhibit curative antimalarial activity. However, all extract fractions showed prophylactic antimalarial activity, with the ethyl acetate extract having the highest percentage of chemosuppression even at doses of 250 and 1000 mg/kg. In the negative control, the expression of δ-ALAD was 5.4-fold, but this was significantly reduced to 2.3-fold when mice were treated with 250 mg/kg of the ethyl acetate fraction. GC-MS analysis of the ethyl acetate fraction revealed high percentages of 2-methyloctacosane, tetracosane, and decane. Conclusion The fractions extracted from S. incanum leaves have been found to possess only antimalarial prophylactic properties, with the ethyl acetate extract fraction showing the most effective results. The activity of this fraction may be attributed to its ability to decrease the expression of δ-ALAD, as it contains an alkane compound implicated with enzyme-inhibitory activity.

Abstract:Ethnopharmacological relevance Phyllanthus muellerianus (Kunze) Exell, a member of the Phyllanthaceae family, is a medicinal plant widely distributed in Africa. Decoctions from the leaves are used in Nigeria to treat fevers, convulsions, some neurological disorders and malaria. Aim of the study This study is to evaluate the anti-malarial properties of methanol extract of Phyllanthus muellerianus (MEPM) leaves and its ethyl acetate fraction using a murine malaria model infected with Plasmodium berghei . Additionally, we seek to investigate the potential modulatory effects of this extract and fraction on CD4 + T-cell populations in the context of malaria infection. Materials and methods The anti-malarial effects of the leaf methanol extract of Phyllanthus muellerianus (MEPM) were screened using three established in vivo models of anti-plasmodial screening namely the curative, suppressive and prophylactic models. The methanol extract (MEPM) was afterwards fractionated into hexane (HFPM), ethyl acetate (EAFPM), and methanol (MFPM) fractions. In the pilot anti-malarial screening of the fractions, EAFPM exhibited the best antiparasitic activity. Subsequently, EAFPM was screened for anti-malarial activity using the three models above. The effects of the MEPM and EAFPM on haematological indices (Hb and PCV) of the inoculated animals were further screened and the mean survival time (MST) of the animals was monitored. CD4 + T cells of various groups were counted before and after treatment using a flow cytometer. The EAFPM was further subjected to HPLC analysis for identification of its major compounds. Results The EAFPM (100 and 200 mg/kg) elicited 88% and 93% cure respectively in the curative model, while artesunate (5 mg/kg,- the positive control) gave 87% protection. The MEPM and EAFPM also gave significant suppression of parasitemia in the suppressive model. The treated groups survived beyond 28 days as against 11 days by the control group (infected but not treated). The treated groups also prevented anaemia seen in the negative control. The EAFPM group significantly modulated the CD4 + T cell. Compounds identified were Gallocatechin, Quercetin -3-O-gallate, Ellagic acid, and Methylellagic acid rhamnoside). Conclusion The study established that the leaf of Phyllanthus muellerianus possesses antimalarial activity, thus lending support to its use in the folkloric treatment of malaria.

Antimalarial activity of borrelidin and fumagilin in Plasmodium berghei-infected mice

Abstract:Background: Malaria remains a significant global health burden, with drug resistance posing a major challenge to its control. The emergence of resistance to antimalarial drugs represents a critical issue in malaria management, as it heightens the likelihood of morbidity and mortality associated with the disease. There is an urgent requirement for a novel candidate drug with a distinct mechanism of action. Aim: In light of the ongoing challenges in malaria management, particularly the emergence of drug resistance, this study aimed to investigate the efficacy of a novel combination therapy of borrelidin and fumagilin against Plasmodium berghei infection on Swiss Webster mice. The findings of this study could contribute to developing new and effective antimalarial treatments. Methods: This study employed a unique approach, using Swiss Webster mice aged 6-8 weeks and dividing them into five groups, each with five mice. The therapeutic efficacy of the combination treatment was evaluated through a comprehensive assessment of parasitemia levels, survival rates, and histological changes in the liver and spleen. This rigorous methodology ensures the reliability and validity of our findings. Results: The combination of borrelidin and fumagilin led to the lowest parasitemia at 5%, contrasting with the control group reaching 15%. Moreover, the combination group exhibited the highest inhibition rate of 69.6% on day nine post-infection. Histopathological alterations were limited to sinusoid dilation, hepatocyte ballooning, and the presence of hemozoin. Conclusion: These findings suggest that the combination of borrelidin and fumagilin holds promise as a potential antimalarial therapy.
Antiplasmodial activity of aqueous extract of Berberis aristata roots against Plasmodium berghei-infected BALB/c mice

Abstract:Context: The rising problem of resistance to present antimalarial drugs stresses the need to look for newer antiplasmodial components with effective modes of action. The roots of Berberis aristata DC. (Berberidaceae) are used in the traditional medicine for malaria in various parts of India. Objective: The objective of this study was to evaluate antiplasmodial activity of B. aristata roots extract for the validation of its traditional medicinal use. Material and methods: Aqueous root extract of Berberis aristata (AREBA) was screened for its in vitro as well as in vivo antiplasmodial activity against lethal rodent malaria parasite Plasmodium berghei NK65. In vitro activity was evaluated against schizont maturation of P. berghei using various concentrations ranging from 1 to 100 µg/mL. For in vivo studies, AREBA at the doses of 150, 250, 350, and 650 mg/kg/d was administered to P. berghei infected BALB/c mice orally for 4 consecutive days (D0-D3). Results: AREBA showed in vitro antiplasmodial activity with an IC50 value of 40 µg/mL. In vivo studies demonstrated a variable dose-dependent chemosuppression with higher efficacy at lower doses. At a dose of 350 mg/kg/d, the suppressive and preventive activities were found to be 67.1% and 53.9%, respectively, followed by enhancing mean survival period up to 12.8 d for the curative assay versus 7.5 d for the untreated mice. Discussion and conclusion: These results provide relevant scientific evidences for the traditional medicinal use of this plant as malaria remedy and further advocates the isolation and characterization of active antiplasmodial principle from this plant.

Abstract:This paper exploits two new alternative approaches to estimate the child health-parental income gradient, using both a threshold model and a more parsimonious random parameters model, applied to the Health Survey for England data 2008-2012. We build on previous research and test the appropriateness of the usual standard age categories (0-3, 4-8, 9-12 and 13-17) exploited in the literature and for policy intervention. Our threshold method estimates different age categories and higher income gradient for children aged between 6 and 8 years old. We further extend our analysis to allow for cohort effects. We find that a higher income is required to improve young children’s health aged 0-2 post 2010. We discuss the relevant reasons and policy implications —most notably that there are socioeconomic child health inequalities exasperated by the recent recession and inequity in the distribution of health interventions towards those most in financial need. Abstract:ETHNOPHARMACOLOGICAL RELEVANCE: Malaria is among the most prevalent and devastating parasitic diseases globally with most cases reported in Sub-Saharan Africa. One of the major reasons for the high malaria prevalence is the ever-increasing emergence of resistant strains of malaria-causing parasites to the currently used antimalarial drugs. This, therefore, calls for the search for antimalarial compounds with alternative modes of action. Plants used in traditional medicine for the treatment of malaria offer possible sources of such compounds. Caesalpinia decapetala has been used traditionally for the treatment of various diseases including malaria. However, the antiplasmodial activity of the plant has never been reported.AIM OF THE STUDY: To determine the ex vivo and in vitro antiplasmodial activities of the extracts of the roots, stem bark and leaves of Caesalpinia decapetala.METHODOLOGY: The roots, stem bark and leaves of Caesalpinia decapetala (Roth) Alston (Caesalpiniaceae) were collected and air-dried under a shade then extracted consecutively with dichloromethane and methanol (1:1 (v/v) (4 × 0.8 L). The extracts were tested for antiplasmodial activities against four strains of Plasmodium falciparum (W2, DD2, 3D7, and D6) and fresh P. falciparum field isolates using the SYBR green I assay. The mean fifty percent inhibition concentration (IC50) was determined for each assay. An acute oral toxicity test was done based on the Organization for Economic Cooperation and Development (OECD 425) guidelines using Swiss albino mice.RESULTS: The leaves and stem bark extracts showed good antiplasmodial activities with IC50 values of 4.54 and 4.86 μg/mL, respectively, when tested against the fresh field isolates ex vivo. Similarly, the roots extract showed an IC50 value of 6.49 μg/mL when tested against field isolates ex vivo. The roots extract showed the highest antiplasmodial activities among the samples when tested against W2 (IC50 = 6.12 μg/mL), DD2 (IC50 = 8.17 μg/mL), and D6 (IC50 = 16.02 μg/mL) strains of P. falciparum whereas the leaves showed the highest activity (IC50 = 9.3 μg/mL) when tested against the 3D7 strain of P. falciparum. No mortality was observed for the mice treated with 2000 mg/kg of the leaves and stem bark extracts. The mouse treated with 2000 mg/kg of the roots extracts regained weight by day 12 of the observation period.CONCLUSION: Caesalpinia decapetala has the potential to suppress the growth of P. falciparum thereby contributing to combating the recurrent emergence of antimalarial drug resistance.

Abstract:Antiplasmodial and analgesic effects of crude ethanol extract of Piper guineense was investigated in mice. The antiplasmodial and analgesic efficacy of the extract was judged on its ability to reduce parasitemia and writhing, respectively, in mice. The antiplasmodial screening involved treating infected mice with 200, 400, and 600 mg/kg body weight of extract while the positive control group was given standard artesunate drug. The analgesic test was carried out by administering 1000, 1500, and 2000 mg/kg body weight of extract to three groups of healthy mice, respectively, after induction of pain with 0.75% acetic acid. The positive control group was given aspirin drug. Parasitemia was reduced by 28.36%, 43.28%, and 62.69% in a dose-dependent pattern in the curative test which was significantly different ( P < 0.05 ) from 96.03% of the standard drug. The reduction of writhing by mice given the extract was also dose-dependent (36.29, 45.43, and 59.07%). Aspirin drug was however more effective (86.36%). The extract was safe at 2000 mg/kg body weight. Phytochemical screening revealed the presence of flavonoids, tannins, phlobatannins, terpenoids, and coumarins. Result obtained in this study demonstrated the efficacy of ethanol extract of Piper guineense as an antiplasmodial and analgesic agent.

Abstract:In Africa, medicinal plants are commonly used to treat malaria and other diseased conditions. The ethanolic leaf and twig extract of Faurea speciosa has been shown to possess promising antiplasmodial properties. This present study was aimed at investigating its antiplasmodial effect in vivo. Qualitative phytochemical screening was carried out on the plant samples using standard methods. The antiplasmodial effect against early infection, curative effect against established infection, and prophylactic effect against residual infection were studied in vivo in Plasmodium berghei-infected mice while the carrageenan-induced edema model in chicks was used for anti-inflammatory tests. The phosphomolybdenum and DPPH radical scavenging assays were used in the evaluation of antioxidant potential. Acute toxicity of the extract was evaluated using the Organization for Economic Cooperation and Development (OECD) guidelines. Phytochemical screening of plant samples revealed the presence of flavonoids, coumarins, tannins, saponins, and glycosides. Faurea speciosa leaf and twig extract exhibited significant antiplasmodial activities in the mouse model with parasite suppression rates of 66.63%, 71.70%, and 56.93% in the suppressive, curative, and prophylactic tests, respectively. A 55.50% reduction of edema in the anti-inflammatory test indicated moderate success in reducing inflammation. The total antioxidant capacity of the extract was determined to be 65.4 mg AAE/g of extract, while in the DPPH radical scavenging assay, the IC50 value was found to be 499.4 μg/mL. With the exception of an inconsistent rise in urea level, there was no significant difference in the other biochemistry parameters in the acute toxicity studied. The median lethal dose (LD50) of the extract was over 2000 mg/kg. The results of this study show that Faurea speciosa leaf and twig extract has promising antimalarial capabilities and is fairly safe at low concentrations.
