Abstract:Background: Pistacia atlantica Desf., also known as Atlantic pistachio or Mt. Atlas mastic tree, has been used in traditional medicine for its anti-inflammatory properties, helping to reduce inflammation in conditions like arthritis. Additionally, the tree's resin possesses antioxidant properties that can help protect against oxidative stress. The anti-arthritic efficacy of Pistacia atlantica oleo-gum resin extract (PAG) remains unvalidated, as its potential therapeutic benefits have not been substantiated. In the current investigation, we have substantiated the inhibitory properties of the PAG in mitigating the development of rheumatoid arthritis induced by Complete Freund's Adjuvant in rats. Methods: Male Wistar rats weighing 160±10 g were subjected to immunization through intradermal injection of 0.1 mL of Complete Freund's Adjuvant (CFA) into the left hind metatarsal footpad. Following the induction of adjuvant arthritis, oral administration of PAG was initiated at doses of 10, 400, and 800 mg/kg once a day. Prednisolone (10 mg/kg, daily) was employed as a positive control in this study. Various parameters, including paw swelling, arthritic index, body weight loss, spleen index, thymus index, serum cytokines, inflammatory mediators, and histological alterations, as well as nitrite glutathione and catalase level, were measured. Furthermore, the expression and activity of matrix metalloproteinase (MMP)-2 and MMP-9 were evaluated. Results: PAG exhibited noteworthy efficacy in attenuating paw swelling and reducing the arthritic index. Moreover, it resulted in enhanced body weight loss and a consequential reduction in the thymus index. PAG (400mg/kg) significantly decreased the level of serum nitrite in the intervention group, as well as the levels of serum glutathione. On the other hand, catalase activity increased, the activity of MMP-9 decreased, and the level of MMP-2 increased compared to the intervention group. The histopathological results revealed that PAG (at doses of 400 mg/kg) had the best therapeutic effects. Conclusion: In conclusion, the present study confirms the anti-arthritic activity of PAG in rats. The extract of PAG was found to prevent rheumatoid arthritis induced by Complete Freund's Adjuvant, as evidenced by the significant suppression of paw swelling and arthritic index.

Abstract:INTRODUCTION: Gastrointestinal neuropathies are frequently found in diabetic patients. AIM: The aim of this study was to find out the safety, adverse reactions, and long-term effectiveness of Pistacia lentiscus plant extract (mastic gum) in diabetic gastroparesis (DG) with respect to sustainable improvement in gastroparesis symptoms (Gastrointestinal Cardinal Symptom Index [GCSI] score) by observational follow-up study of a single-centric double-blind noninferiority randomized control trial. MATERIALS AND METHODS: Thirty-eight individuals were recruited and equally randomized in two study groups based on GCSI score and TC 99 radionuclide gastric emptying scintigraphy (GES), i.e. the mastic gum group and the levosulpiride group. After 24 weeks, the GCSI score was recalculated in both the groups, and patients were evaluated for the safety, adverse reactions, and long-term effectiveness of mastic gum and the standard drug levosulpiride. RESULTS: In the extended study, mean GCSI score changes at 24 weeks were statistically significant ( P < 0.001) ( t -test) between the two groups. In the mastic gum arm, the change in mean GCSI score at 24 weeks was statistically nonsignificant mean ± (standard deviation [SD]) 16.7± (3.81) compared to the GCSI score at 2-month postintervention mean (SD) 16.35± (2.27) (intragroup P = 0.89) (repeated measures ANOVA). It strongly indicates that mastic gum provided a sustainable improvement in DG symptoms in comparison to levosulpiride, with excellent subjective well-being postintervention, without any obvious significant adverse effects. CONCLUSION: Six-month (24-week) interim analysis of patients suggests that mastic gum gives a sustainable improvement in DG symptoms without any obvious adverse effects as compared to levosulpiride.

Abstract:The current study was conducted to investigate the protective effects of Pistacia Atlantica (mastic gum) hydro-alcoholic extract and Punica Granatum (pomegranate) peel hydro-alcoholic extract treatment against alcohol-induced peptic ulcer in Wister rat and to compare this protective effects between each other and with omeprazole as the most common drug in treatment of peptic ulcer. Thirty five male Wister rats were assigned into five groups (seven rats each group). C- group rats were received normal saline for one week. C+ group rats were received normal saline for one week before receiving one dose of absolute ethanol. OME group rats were received normal saline for one week and one dose of omeprazole one hour before receiving one dose of absolute ethanol. MG group rats were received Pistacia Atlantica hydro-alcoholic extract for one week before receiving one dose of absolute ethanol. PG group rats were received Punica Granatum hydro-alcoholic extract daily for one week before receiving one dose of absolute ethanol. The gross and histopathology lesion of gastric ulcer were assessed. Ulcer index results showed a significant (p<0.05) differences between C+ group and other groups, However, the ulcer index of PG group was significant (p<0.05) compared with OME group.

Abstract:A 75-year-old male patient underwent stenting of a mid-right coronary artery (RCA) lesion because of exertional chest pain. Abstract:OBJECTIVE: This study aimed to investigate the effects of systemic administration of P. eurycarpa Yalt. plant extract on alveolar bone loss and oxidative stress biomarkers in gingival tissue in a rat model of experimental periodontitis.METHODOLOGY: 32 male Wistar albino rats, weighing 200-250 g, were divided into four groups (n=8): Healthy control (HC), Experimental periodontitis control (EPC), Experimental periodontitis 400 mg/kg (EP400), Experimental periodontitis 800 mg/kg (EP800). Experimental periodontitis was induced using the ligating method. Distilled water was administered to the HC and EPC groups and the plant extract was administered to the EP400 and EP800 groups by oral gavage at doses of 400 mg/kg and 800 mg/kg, respectively. The rats were sacrificed on the 15th day. The values of glutathione peroxidase GSH-Px, malondialdehyde (MDA), superoxide dismustase (SOD), interleukin-1β (IL-1β), interleukin-10 (IL-10), total antioxidant status (TAS), total oxidant status (TOS), oxidative stress index (OSI) in the gingival tissues were analyzed by ELISA tests. Alveolar bone loss was assessed using micro-CT images of the maxilla.RESULTS: Although the IL-1β, TOS, OSI results of the healthy control group were lower than those of the other groups, the TAS values were higher (p<0.05). No significant difference was found in the biochemical parameters among the EPC, EP400, and EP800 groups (p>0.05). Alveolar bone loss was significantly reduced in the extract groups compared to the EPC group (p<0.001).CONCLUSION: Within the limitations of this study, it was observed that the systemic P. eurycarpa extract application reduced alveolar bone loss in a rat model of experimental periodontitis. Further studies are needed to elucidate the beneficial effects of P. eurycarpa.

In Vivo Evaluation of the Anti-Inflammatory Effect of Pistacia lentiscus Fruit Oil and Its Effects on Oxidative Stress

Abstract:In order to find new topical anti-inflammatory agents, we had recourse to a medicinal plant. This work was designed to determine the topical anti-inflammatory effect of Pistacia lentiscus fruit oil (PLFO), using carrageenan-induced paw edema rat model, and to evaluate its effects on oxidative stress. The topical anti-inflammatory activity of PLFO was compared to Inflocine® and estimated by measuring the diameter of paw edema, for 5 hours at a 1-hour interval. After that the rats were scarified and the inflamed paw tissue was removed for the exploration of some parameters of oxidative stress and histopathology. PLFO showed a significant anti-inflammatory activity in comparison with the Inflocine. The percentages of edema inhibition were 70% and % 51.5% ( p < 0.01 ), respectively, after five hours. The treatment with PLFO and Inflocine led to significant increases ( p ≤ 0.05 ) in the activities of CAT, SOD, and GPX and significant decreases in the MDA level and AOPP activity in the paw tissue after Carr injection, in comparison with the Carr group. Therefore, our findings demonstrate that PLFO might accelerate the development of new drugs which could be used scientifically as a source for natural health products in the treatment of topical inflammation.

Abstract:Pistachio (Pistacia vera L.) is an important cultivated plant in the Southeastern Anatolia region and its fruits are widely used in the food industry. The previous studies mostly focused on the food minerals and components. However, there is no study on chemical components and biological activity of the soft hulls of the pistachio fruits. In this study, it was focused on the acetone and water extracts of the soft hulls of pistachio fruits. Hence, the acetone extract was subjected to sephadex LH-20 column chromatography and fractioned into five fractions (Frs. A-E) according to molecular weight of components. A triterpen compound isolated from the Fr. B by crystallation method and its chemical structure was characterized as masticadienonic acid by spectroscopic methods. The fractions contain a mixture of the anacardic acids 13:1, 15:2, 15:1 and 17:1 and their derivatives according to their spectroscopic data and GC-MS analysis. Shikimic acid used in the synthesis of the oseltamivir, antiviral agent was also purified at high yield (14.36%) from the water extract of the soft hulls of pistachio fruits. In order to explore the antidiabetic properties of the pure compounds, the extracts and the fractions, their inhibition effects were determined on the activities of digestive enzymes (α-glucosidase and α-amylase). All of the extracts, masticadienonic acid and the fractions containing the anacardic acids strongly inhibited the α-glucosidase activity much more than acarbose. However, all applications exhibited much weaker inhibitory properties againts α-amylase, as compaired to the inhibition effect of the acarbose. Furthermore, the treatments of mesocarps of Pistachio fruits acted as much weaker inhibitors against AChE, whereas the acetone extract and its fractions containing different rates of anacardic acids were powerful agents against BChE as strong as neostigmine and galantamine. The cytotoxic activities of the extracts and the compounds against HUVEC, A549 and H1299 cells were also determined using the MTT analysis method and it was determined that the acetone extract and the compounds, masticadienonic and shikimic acids showed cytotoxic effects on the cells

Abstract:Ethnopharmacological relevance: Pistacia integerrima J.L. Stewart ex Brandis (Family Anacardiaceae) galls are used in Indian ethnomedicine for their antiasthmatic, antioxidant anti-inflammatory, sedative, and spasmolytic properties. However, no scientific studies demonstrate its nephroprotective potential against Cisplatin-induced nephrotoxicity. Materials and Methods: The effect of the essential oil of Pistacia integerrima (7.5, 15 and 30 mg/kg/day) and standard drug Vitamin E (250 mg/kg/day) on the kidney was examined 5 days after treatment with Cisplatin (12 mg/kg body weight, i.p.) induced oxidative renal damage in mice. Renal injury was assessed by assessment of serum creatinine and blood urea nitrogen. Renal oxidative stress was estimated by renal malondialdehyde levels, reduced glutathione levels and enzymatic activities of superoxide dismutase and catalase. Results: Essential oil of Pistacia integerrima (7.5, 15 and 30 mg/kg/day) significantly (p < 0.001) protected the nephrotoxicity induced by Cisplatin. The Cisplatin-induced increase in serum urea and creatinine concentrations was reduced in the essential oil of Pistacia integerrima treated groups. Essential oil of Pistacia integerrima treatment also increased the Cisplatin-induced decline of renal antioxidant enzymes such as superoxide dismutase, catalase, and Glutathione activities. Essential oil of Pistacia integerrima increased the concentration of reduced glutathione and protected the increase of Cisplatin-induced lipid peroxidation. Histopathological investigations of the kidney showed a protective effect of the essential oil of Pistacia integerrima in mice. The essential oil of Pistacia integerrima was found to be effective in protecting against oxidative renal damage. The protection is mediated by preventing the decline of renal antioxidant status. Conclusion: The present investigation demonstrates that the essential oil of Pistacia integerrima Stewart ex Brandis has a protective effect on cisplatin-induced experimental nephrotoxicity, and this effect is attributed to its direct anti-inflammatory and strong antioxidant profile. Hence, the essential oil of Pistacia integerrima Stewart ex Brandis has a solid potential to be used as a therapeutic adjuvant in cisplatin nephrotoxicity.
Efficacy of Pistacia lentiscus Plant (Rumi Mastagi) in Comparison to Levosulpiride in Patients with Diabetic Gastroparesis: A Double-Blind Non-Inferior Randomised Control Trial Study

Abstract:Introduction: Gastrointestinal neuropathies are frequently found in diabetic patients. The pathogenesis of diabetic gastroparesis (DG) is multifactorial. The usual treatment for DG includes dietary modifications, prokinetic and antiemetic agents. There is increasing demand for more effective medicines to treat DG. The current study was conducted on the Pistacia lentiscus stem extract to add to the armamentarium of DG treatment and to find the efficacy of P. lentiscus plant extract (mastic gum) in comparison to levosulpiride in DG for improvement in gastroparesis symptoms and gastric emptying scintigraphy (GES) in a single centric double-blind non-inferiority randomised control trial. Methods: Thirty-eight individuals were recruited and equally randomised into two study groups based on Gastroparesis Cardinal Symptom Index (GCSI) score and TC99 Radionuclide GES, mastic gum group and levosulpiride group. Both pre and post-intervention (8 weeks) GCSI scores were calculated, GES was performed to quantify the improvement in gastric emptying. Power analysis was performed using G*POWER software version 3.1.9.7 and data analysis using SPSS 23.0, variables measured in mean ± standard deviation (SD). Various statistical tests were used such as independent t-test, Chi-square test or Fisher's exact test, Wilcox Mann-Whitney test, analysis of variance (ANOVA) test, and posthoc pairwise tests. Results: The mastic gum is found effective in the improvement of 4 h gastric emptying percentage from the mean (SD) 76.60 (± 9.96) to mean (SD) 97.20 (2.17)% (P < 0.001). Mastic gum has the property of HbA1c reduction, which is more significant than that of levosulpiride (P = 0.044). Mastic gum also had significant Low density lipoprotein (LDL) (mg/dL) levels reduction, (P < 0.001), compared to levosupiride. An absolute increase was observed in haemoglobin (HB) level in mastic gum at a 2-month mean (SD) of 1.03 (0.77) (g/dL) (P-value <0.001). Conclusions: To our knowledge, this is the first study to compare the effect of levosulpiride with mastic gum concerning improvement in diabetic gastroparesis (DG) using GES. In the study, mastic gum was found to have great properties to improve DG with many important pleiotropic effects.
Accumulation of the Major Components from Polygoni Multiflori Radix in Liver and Kidney after Its Long-Term Oral Administrations in Rats

Abstract:Abstract Although Polygoni Multiflori Radix (PMR) has been widely used as a tonic and an anti-aging remedy for centuries, the extensively reported hepatotoxicity and potential kidney toxicity hindered its safe use in clinical practice. To better understand its toxicokinetics, the current study was proposed, aiming to evaluate the biodistributions of the major PMR components including 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucopyranoside (TSG), emodin, emodin-8-O-β-D-glucopyranoside (EMG) and physcion as well as their corresponding glucuronides following bolus and multiple oral administrations of PMR to rats. Male Sprague-Dawley rats received a bolus dose or 21 days of oral administrations of PMR concentrated granules at 4.12 g/kg (equivalent to 20.6 g/kg raw material). Fifteen minutes after bolus dose or the last dose on day 21, rats were sacrificed and the blood, liver, and kidney were collected for the concentration determination of both parent form and glucuronides of TSG, emodin, EMG, and physcion by HPLC-MS/MS. Among all the tested analytes, TSG, EMG, EMG glucuronides in liver and TSG, EMG, as well as all the glucuronides of these analytes in the kidney demonstrated the most significant accumulation after multiple doses. Moreover, the levels of the parent analytes were all significantly higher in liver and kidney in comparison to their plasma levels. Strong tissue binding of all four analytes and accumulation of TSG, EMG, and EMG glucuronides in the liver and TSG, EMG, as well as the glucuronides of all four analytes in the kidney after multiple dosing of PMR were considered to be associated with its toxicity.

Abstract:Gums are polysaccharides, proteins, and minerals that occur naturally in seed coverings and as exudative resinous substance from woody plants. It is reported to have antibacterial, anticancer, blood sugar regulation, and immune system boosting properties. However, the presence of toxic metals in gum is caused for caution as these metals can be harmful if taken in high quantities. The purpose of this study was to determine the amounts of toxic metals in gums collected from the local market, as many consumers tend to use them daily for incense or food ingredients. Gum samples were extracted from several parts of 10 selected medicinal plants (bark, sap, root, latex, leaf glue, and gum). Two fractions of each sample were produced using nitric acid (NHO3), followed by hydrochloric acid (HCl) at first and then hydrogen peroxide (H2O2). The presence of toxic metals in the solutions was determined using an Inductively Coupled Plasma Atomic Emission Spectrometer (ICP OES). The results showed that most of the elements were detected in high concentrations in Commiphora myrrha (Cd, Cu, Fe, K, Mn, Ni, Pb, and Zn) followed by Benzoin resin (Jawi Oud) and Paeonia officinalis. The most prevalent elements detected in all of the herbal gums were potassium (K) and iron (Fe). Fortunately, the sampled herbal gums were found to be within the WHO/FAO permitted range. This study may provide insights about the safety of the selected gums to be used for food applications. Further in vitro and in vivo toxicity studies should be performed to identify the safe dose.

Abstract:The widespread presence of microplastics (MPs) in aquatic ecosystems has raised growing concerns among ecotoxicologists regarding their potential toxicity. This study explored the impacts of polylactic acid (PLA) MPs on the physiology and health of freshwater fish, Cirrhinus mrigala , by dietary exposure for 90 days. The experiment consisted of six groups: five treatment groups (0.5%, 1%, 1.5%, 2%, and 2.5% PLA-MP) and a control group (0% PLA-MP). Each group was comprised of fifteen fish, and the experiment was replicated three times. The exposure severity of PLA-MPs varied from low to high, with treatment levels ranging from 0.5% to 2.5% PLA-MPs, relative to the control group. This exposure significantly affected their growth performance. Additionally, the apparent digestibility of the SFM-based diet decreased with increasing PLA-MPs concentration. Exposure to PLA-MPs induced considerable changes in body composition, characterized by increased moisture and crude fat content and decreased ash content and crude protein. The blood profile, including MCHC, RBCs, Hb, PLT and PCV exhibited significant declines in the high treatment group (2.5% PLA-MPs), while MCH, WBCs and MCV showed notable increases. Furthermore, histopathological examination of the intestine revealed an increase in abnormalities in the intestine at 2.5% PLA-MPs level. The high treatment group (2.5% PLA-MPs) showed the lowest mineral content in the fish muscles. In summary, dietary exposure to PLA-MPs led to alterations in overall body performance across the treatment groups, ranging from low to high severity levels.

Abstract:Background: Dhatryadi ghrita consists of dhatri, is known to have number of curative properties since ages. It does not causes any toxic or adverse effect, but there is no scientific evidence available. Objective: The present piece of research is aimed to study the toxic effects in order to lay down the safety parameters of methanolic extract obtained from Dhatryadi Ghrita in wistar rats (180-200g) as per the standards set by The Organization for Economic Cooperation and Development or OECD. Materials and Methods: Group 1 was used as a control for comparing the behavior of rats from all groups which were administered extracts of different concentrations. The animals in Group 2 were administered a dose of 1000, Group 3, 2000, and Group 4, 3000, respectively in the units of mg/kg and Group 5 were given a dose of 4000 mg/kg accordingly. Results: The acute toxicity studies of the experiment dealing with different doses as varying from 1000-4000 mg/kg, which did not resulted in any death of any animal till 14 days of observation in the experimentation period. Conclusion: Dhatryadi Ghrita is safe in rodent and mice. Hence, the extract is safer for therapeutic use in pharmaceutical formulations. Ghrita in different concentrations were found to be completely safe and non-toxic under acute toxicity studies.
Moricizine Pharmacokinetics in Renal Insufficiency: Reevaluation of Elimination Half‐Life

Abstract:M oricizine HC1 (USAN; also known as moracizine HC1, INN; or Ethmozine, the proprietary name found in existing literature) is a phenothiazine derivative newly marketed in the United States for the clinical management of life-threatening ventricular arrhythmias. Moricizine pharmacokinetics have not been reported in renal insufficiency; however, several early reviews3 relying solely on the published account of a single arrhythmia patient with diminished renal function,4 report an unusually long elimination t112. In this patient, the t112 for moricizine was 47.5 hours. A recent review5 questioned the validity of this long terminal t112 in this patient in light of the extensive hepatic elimination of moricizine and the fact that enterohepatic recycling does occur with the drug. We have examined the data from the single patient with diminished renal function to reevaluate the reported t,2. Preliminary results are also reported from an on-going study in healthy subjects and subjects who are undergoing hemodialysis. The patient was a 65-year-old black man who weighed 85.9 kg and measured 166.4 cm in height at the start of the study. Although the patient was 41% over the ideal body weight (60.9 kg) and under treatment for hypertension, Parkinson’s disease, and adult-onset diabetes, he did not have evidence of cardiac insufficiency or overt signs of renal failure. The creatinine clearance of the patient of 55.7 mL/min was 31% less than the age-adjusted value for a 65year-old healthy man (80.4 mL/min).6 At the time of study, the patient was taking hydrochlorothiazide,

Abstract:Introduction: Lablab purpureus, under the family of Fabaceae, is a plant with various pharmacological activities. The present study was aimed to investigate the phytoconstituents, membrane stabilizing activity, central nervous system (CNS) depressant potential, and gastrointestinal (GI) motility of the methanol extract of L. purpureus seeds (MELPS).Methods: The methanol plant extract was screened for different phytochemical groups. Mice were classified into four groups for in vivo activities. Group-I was designated as negative control and received distilled water (10 mL/kg body weight); group-II served as positive control and received diazepam (1 mg/kg body weight). Group-III and group-IV both were experimental groups and received plant extract at 200 and 400 mg/kg body weight, respectively.Results: Alkaloids, carbohydrates, saponins, glycosides, tannins, phenols, flavonoids, and proteins were found after phytochemical analysis. On hypotonic solution-induced hemolysis of erythrocyte membrane, MELPS9 (9 mg/mL) resulted in the highest percentage of inhibition (60.51 ± 0.889), and on heat-induced hemolysis, MELPS9 (9 mg/mL) resulted in the highest percentage of inhibition (33.97 ± 0.21). In the case of the CNS depressant potential experiment, mice that received a sample at a dose of 400 mg/kg body weight showed the highest result (54.40 ± 4.51) compared with the positive control (14.2 ± 3.70) (P < 0.001). Similarly, 400 mg/kg dose sample exhibited the highest percentage of inhibition (60.51 ± 0.889) of hemolysis and GI motility (22.26%).Conclusion: It can be concluded that the MELPS has potential membrane stability, CNS depressant, and antimotility effects.

Abstract:Safety evaluation of a newly designed polymeric drug delivery system (DDS), with/without the addition of active pharmaceutical ingredients (APIs), is now mandatory for their regulatory approval for human use. Hence, Salvia spinosa seed mucilage/hydrogel (SSH) was treated with methacrylic acid (MAA) to synthesize a composite hydrogel (SSH-co-MAA). Acute oral and acute dermal toxicity studies of the SSH-co-MAA for API delivery were ascertained following OECD guidelines 420 and 402, respectively. Moreover, an ocular toxicity study was also performed and analyzed through Draize scale. Animals of two species, rodent (rat) and non-rodent (rabbit), were divided into four groups. Group A of both rats and rabbits was assigned as control and remained untreated. Meanwhile, groups B, C, and D were labelled as treated groups and received a single dose of SSH-co-MAA, i.e., 0.05, 0.3 and 2 g/kg body weight of the animal. During 14 days after the treatment, animal monitoring was done for behavioral changes, food and water intake, adverse effects, and mortality. All animals remained alive, with no statistically significant abnormality. Hematological and biochemical parameters of control and treated animals were analyzed after the completion of 14 days and found in harmonization. The vital organs of rats were removed to determine absolute organ weights. Histopathology of the vital organs of rats revealed normal cellular architecture, without any lesions. SSH-co-MAA was also free from dermal and ocular toxicity. The overall results of acute oral and dermal toxicity studies prove that SSH-co-MAA is safe, especially after oral administration. Hence, SSH-co-MAA can be used as a non-toxic excipient for drug delivery systems.

Abstract:Objectives: Some species of Prunus L. are popularly used to treat gastric ulcers. However, the possible healing mechanisms of the anti-ulcer activity of P. spinosa, which has proven antioxidant, anti-inflammatory, and wound-healing properties, are unclear. Materials and methods: Ethanol extracts of P. spinosa fruits were administered orally at 100 mg/kg and 200 mg/kg to Wistar albino rats, with an indomethacin-induced gastric ulcer model. The ulcerous areas on the stomach surface were examined macroscopically. Tissues were examined histopathologically and biochemically. LC-HRMS revealed the phytochemical content. Results: TNF-α, IL-6, IL-1β, IL-8, and NF-kB levels were higher in the gastric ulcer group than in the extract groups. The VEGF values did not differ in each group. A significant difference was found between the lansoprazole group and the high-dose P. spinosa group regarding PGE2 levels. A histopathologically significant difference was observed between the healthy group and the indomethacin-applied groups in terms of neutrophilic infiltration of the gastric mucosa. Ascorbic acid (1547.521 µg/g), homoprotocatechuic acid (1268.217 µg/g), and genistein (1014.462 µg/g) were found as the main compounds in the P. spinosa extract by LC-HRMS. Conclusion: Our results demonstrated that P. spinosa protected the gastric mucosa from inflammation and also modulated the PGE2 pathway. When considered in terms of TNF-α, IL-1β, IL-8, IL-6, PGE2, and NF-kB values, it can be concluded that it has a similar or even more positive effect than the reference substance. P. spinosa showed its effects in a dose-dependent manner.

Abstract:Background. Pedalium murex (P. murex) is used in folk medicine for treatment of male infertility. However, scientific data on its safety are limited. Objective. This study was carried out to assess the acute and repeated dose 28-day oral toxicity of the aqueous extracts from P. murex leafy stem and fruit in Wistar rats. Methods. The acute toxicity test was performed according to the line 423 of the Organization for Economic Cooperation and Development (OECD) guidelines. The rats were randomly divided into three groups (n = 3). The control group received distilled water, while the experimental groups were given at a single dose, 5000 mg/kg of each extract. The repeated dose 28-day oral toxicity was performed according to the line 407 of the OECD guidelines. 35 rats divided into 7 groups of 5 male rats each were daily treated for 28 days with each extract at 200 mg/kg, 400 mg/kg, and 800 mg/kg, respectively. The in-life parameters were recorded during the follow-up. At the end of this study, organ weights, hematology, biochemistry, and histology parameters were analyzed. Results. In the acute oral toxicity test, there was no morbidity or mortality related to the treatments. Both extracts belong therefore to category 5 of the globally harmonized system (GHS) of classification. In the repeated dose 28-day oral toxicity test, both extracts did not alter animal’s behavior. However, both extract administration led to proteinuria and renal damages. Conclusion. P. murex leafy stem and fruit aqueous extracts exhibited potential nephrotoxicity. Therefore, care should be taken when they are used over an extended period.
