Abstract:In the last years, surface-enhanced Raman scattering based immunochromatographic assay (SERS-ICA) has been getting great attention and been applied for the detection of different target analytes. In this study, a novel SERS-ICA using Ag-MBA@SiO2-Ab as an immunoprobe has been developed for rapid, quantitative and ultrasensitive detection of a pharmaceutical diclofenac (DCF) residue in water samples. Ag, Ag@SiO2 and Ag-MBA@SiO2 nanoparticles (NPs) were synthesized and characterized. The immunoprobe was prepared by sandwiching the Raman reporter mercaptobenzoic acid (MBA) between the core-shell layers and immobilizing the monoclonal antibody (mAb) against DCF on the surface of the Ag@SiO2 NPs. Under optimal conditions, the sensitivity and limit of detection (LOD) of the SERS-ICA for DCF were found to be 9 pg mL(-1) and 0.07 pg mL(-1), respectively. The detection of the SERS-ICA for DCF was completed within 15 min. Ag-MBA@SiO2-Ab was stable within four months without significant loss of SERS intensity. The key to success of the SERS-ICA was the employment of the Ag@SiO2 NPs as the SERS substrate, which possessed gorgeous SERS enhancement capability, predominant biocompatibility and excellent stability. This SERS-ICA is potential as an ultrasensitive analytical method in point-of-care testing, clinical diagnosis, food inspecting and environmental monitoring.

Abstract:Salbutamol (SAL) is known as quick-acting β-2 receptor agonist and its use in humans for pulmonary diseases and/or in animal feed is limited because of associated potential hazardous effects on health. Several techniques are available for the detection of SAL, but are expensive and time consuming. Here for the first time, a novel pre-assembled DNA immunoprobe-based immuno-PCR assay was developed to investigate the levels of SAL in human urine samples and compared the proposed immuno-PCR assay for the detection of SAL with that of direct competitive ELISA. Under the optimized conditions, the assay showed a linear range over seven orders of magnitude, whereas the limit of detection of SAL in buffer was found to be 21 fg/mL. Current immuno-PCR assay exhibited a 300-fold better detection limit for SAL as compared to one achieved with direct competitive ELISA using the same antibody. In conclusion, it has been found that the developed method is highly sensitive and simple method that can significantly enhance the detection sensitivity for small molecules. Moreover it can be modified and used for detecting other small molecules and/or chemicals that exist in the environment and are responsible for the environmental pollution.
Ultrasensitive and Specific Detection of Salbutamol in Swine Feed, Meat, and Urine Samples by a Competitive Immunochromatographic Test Integrated with Surface-Enhanced Raman Scattering

Abstract:Salbutamol (SAL), a kind of β-agonist which can enhance the lean meat-to-fat ratio, has been inhibited as an additive used in animal feeds for livestock production in many countries due to its harmful effect to the consumers. In this study, an ultrasensitive and specific competitive immunochromatographic test (ICT) integrated with surface-enhanced Raman scattering (SERS) for the detection of SAL was described. The immunoprobe was prepared by immobilizing polyclonal antibody against SAL on the surface of Au@Ag nanoparticles in which the Raman reporter (4-mercaptobenzoic acid, MBA) had been sandwiched. After ICT procedures, the specific SERS signals generated from MBA on the test line of the ICT strip were measured for the quantitative determination of SAL. The assay was completed in 15 min. The IC50 and the limit of detection (LOD) values of the assay for SAL were 0.028 ng mL−1 and 3.0 pg mL−1, respectively. There was no cross-reactivity (CR) of the assay with other three β-agonists (clenbuterol, phenylethanolamine A, and ractopamine), showing high specificity of the assay. Spiking experiments indicated that the average recoveries (n = 3) of SAL from swine feed, meat, and urine samples were in ranges of 98.4–105.2 % with the relative standard deviations (RSDs) of 1.7–7.8 %. The results demonstrated that the proposed ICT was a feasible method for ultrasensitive and specific detection SAL in swine feed, meat and urine samples, and could be extended for the detection of other target analytes.
Optical Detection of Fe3+ Ions in Aqueous Solution with High Selectivity and Sensitivity by Using Sulfasalazine Functionalized Microgels

Abstract:A highly selective and sensitive optical sensor was developed to colorimetric detect trace Fe3+ ions in aqueous solution. The sensor was the sulfasalazine (SSZ) functionalized microgels (SSZ-MGs), which were fabricated via in-situ quaternization reaction. The obtained SSZ-MGs had hydrodynamic radius of about 259 ± 24 nm with uniform size distribution at 25 °C. The SSZ-MG aqueous suspensions can selectively and sensitively response to Fe3+ ions in aqueous solution at 25 °C and pH of 5.6, which can be quantified by UV-visible spectroscopy and also easily distinguished by the naked eye. Job’s plot indicated that the molar binding ratio of SSZ moiety in SSZ-MGs to Fe3+ was close to 1:1 with an apparent association constant of 1.72 × 104 M−1. A linear range of 0–12 μM with the detection limit of 0.110 μM (0.006 mg/L) was found. The obtained detection limit was much lower than the maximum allowance level of Fe3+ ions in drinking water (0.3 mg/L) regulated by the Environmental Protection Agency (EPA) of the United States. The existence of 19 other species of metal ions, namely, Ag+, Li+, Na+, K+, Ca2+, Ba2+, Cu2+, Ni2+, Mn2+, Pb2+, Zn2+, Cd2+, Co2+, Cr3+, Yb3+, La3+, Gd3+, Ce3+, and Bi3+, did not interfere with the detection of Fe3+ ions.

Abstract:Surface-assisted laser desorption/ionization mass spectrometry (SALDI-MS) emerges as an effective method for analyzing small molecules. At the current stage, its application is still limited by its detection sensitivity. According to the mechanism of SALDI-MS, the sensitivity highly depends on the laser desorption/ionization (LDI) efficiency. In this study, we adjust the localized surface plasmon resonance (LSPR) between particles by changing the gap, thereby improving the efficiency of LDI. The gaps of nanoparticles can be adjusted by the ionic strength in the solution. With the decrease of the gaps, the thermal effect of the nanoparticles is gradually enhanced and the number of excited electrons is also increased, which improves the sensitivity of SALDI-MS. The prepared substrate with optimized gaps has wide applicability for the detection of small biomolecules and dye molecules. Moreover, the substrate can successfully detect trace small molecules in complex samples. The analysis of sulfonamide in lake water shows the commendable performance of the substrate. The relative standard deviations (RSD) of spot-to-spot and batch-to-batch are 3.73% and 7.22% respectively, confirming the high reproducibility and the detection limit is as low as 100 pM, indicating the high sensitivity of the substrate. The results demonstrate that adjusting the LSPR of metal nanoparticles is a promising method for improving the performance of SALDI-MS.

Abstract:A combination of SiO2@AuNPs@PDA molecularly imprinted and surface-assisted laser desorption/ionization-time-of-flight mass spectrometry (SALDI-TOF MS) was devised as a method for highly specific and ultrasensitive detection of two biogenic amines-histamine (HIS) and tryptamine (TRP)-in real samples. In this strategy, AuNPs modified amino-abundant silica nanospheres (SiO2@AuNPs). The prepared SiO2@AuNPs were used as a substrate to synthesize a molecularly imprinted polymer (MIP) through in situ dopamine self-polymerization with HIS and TRP as the template molecules (SiO2@AuNP@PDA-MIP). The as-prepared MIP structure, properties, and target-analyte identification conditions were characterized and optimized and it was used as the matrix for MS. Compared to the case of nonimprinted materials, the imprinting function endowed the matrix with a higher selectivity for capturing the target molecules. The enriched analytes were directly and rapidly identified using SALDI-TOF MS without elution. Meanwhile, the proposed method has low background interference, good reproducibility and stability, high salt tolerance, and satisfactory linearity (R2 > 0.99), and it enables ultrasensitive detection of HIS and TRP (limits of detection for HIS and TRP were 0.2 and 0.1 ng mL-1, respectively). Moreover, the proposed method was applied to analyze samples of real beer, sausage, and chicken, and the results agreed with those obtained via liquid chromatography-MS, suggesting that the method has excellent practical applications in the field of food safety.

Abstract:Surface-enhanced Raman spectroscopy (SERS) has been widely used in the field of therapeutic drug monitoring (TDM) because of its powerful fingerprinting capability. In this paper, we used an in situ synthesis method to anchor Ag nanoparticles (AgNPs) on the surface of MIL-101(Cr) to obtain MIL-101(Cr)@Ag. Owing to the large specific surface area and ultra-high porosity of MIL-101(Cr)@Ag, we developed a method for the determination of chlorpromazine hydrochloride (CPZ) and aminophylline (AMP) in human serum by using it as a solid-phase extraction sorbent and SERS substrate. The label-free TDM-SERS method was able to evaluate the levels of CPZ and AMP in serum samples with detection limits as low as 8.91 × 10 –2 μg/mL and 3.4 × 10 –2 μg/mL, respectively. In addition, influencing factors including sample solution pH, AgNO 3 concentration, drug adsorption time, and the amount of sample solution were optimized. This protocol provides a new method with good selectivity, stability, reproducibility, homogeneity, and sensitivity for the determination of small-molecule drug content in serum samples. This label-free TDM-SERS method will help to achieve rapid individualized dosing regimens in clinical practice and has potential applications in the field of TDM.

Abstract:A thin-film based on 3-aminopropyl triethoxysilane surface-modified Ce-doped zinc-aluminum layered double hydroxide was synthesized on the inner surface of an aluminum tube. It has been applied to in-tube stir bar sorptive extraction of nonsteroidal anti-inflammatory drugs in saliva samples followed by high-performance liquid chromatography. The sorbent was characterized by scanning electron microscopy, X-ray diffraction, Fourier-transform infrared spectroscopy, energy-dispersive X-ray spectroscopy, and elemental mapping. The extraction parameters including sample pH (4.2), extraction time (10 min), stirring speed (800 rpm), type of eluent (acidified tetrahydrofuran), eluent volume (100 μL), and desorption time (6 min) were thoroughly optimized. Under the optimum conditions, limits of detection were found to be less than 5.0 ng mL-1. Calibration plots were linear within the range 10-1000 ng mL-1 (R2 > 0.9982). Absolute recoveries were calculated in the range 63.5 to 72.4%. The repeatability (intra- and inter-day precision) and reproducibility (tube-to-tube precision) at concentrations of 50, 250, and 500 ng mL-1 were less than 7.6% and 9.4%, respectively. The method accuracy based on the relative error was calculated at these concentrations and ranged from - 4.9 to - 9.3% for intra-day relative error (%) and - 6.8 to - 11% for inter-day relative error (%). Finally, the method applicability was examined for the determination of nonsteroidal anti-inflammatory drugs in saliva samples, and good relative recoveries were obtained within the range 86.5 to 95.2%. As a result, the introduced method can be applied as a suitable alternative to measuring nonsteroidal anti-inflammatory drugs in biological fluids. Abstract:The current research proposed a highly sensitive and selective spectrofluorometric approach for the assay of gastrointestinal medications omeprazole (OMZ) and domperidone (DOM). Green synthesis of metal oxide nanoparticles such as zinc oxide (ZnONPs) and cerium oxide (CeO2NPs) using Pimpinella anisum and Syzygium aromaticum extract was used as fluorescence emission catalysts for the determination of OMZ and DOM. Due to their unique optical properties, nanoparticles (NPs) form the basis for spectrofluorimetric quantification of the selected drugs. The detection studies were performed under λex/λem 350/450 nm and 284/392 nm for OMZ and DOM in the presence of ZnONPs and CeO2NPs, respectively. Under ideal conditions, fluorescence intensities (FI) were linearly with correlation coefficient (r = 0.999) over concentration ranges of 0.1-100 and 0.01-200 μg mL-1 for OMZ, 0.01-100 and 0.01-300 n g mL-1 for DOM in the presence of ZnONPs and CeO2NPs, respectively. Method validation was carried out to guarantee the accuracy, suitability, and precision of the proposed fluorescence (FL) systems for the determination of OMZ and DOM. Analytical method guidelines and requirements were followed. The designed procedure was used effectively to identify the determined drugs in both their pure and commercial versions.

Abstract:Medicine safety has become a large concern and prompts an urgent need to develop a rapid, simple and sensitive analytical method, which can monitor excessive preservatives in medicine. In this work, dispersive liquid-liquid microextraction (DLLME) was combined with surface enhanced Raman scattering (SERS) for a quick analysis of a kind of preservatives, sodium benzoate, in ibuprofen oral solution. The experimental parameters affecting DLLME were systematically investigated. Under the optimal conditions, the whole procedure, including DLLME and the SERS analysis, could be carried out within 10 min. A good linearity between the concentration of sodium benzoate ranging from 10 to 500 mg L-1 and the SERS signal intensity could be obtained, and the correlation coefficient (R2) 0.9986. The method detection limit was 0.56 mg L-1. The relative standard deviation was less than 6.33% for ten replicates at the same sample concentrations. The analytical results prove that the method is suitable for rapid determination of sodium benzoate in ibuprofen oral samples.
A Portable Kit for Rapid Detection of Bromadiolone in Human Blood and Urine Via Surface-Enhanced Raman Scattering Coupled with Salt-Induced Liquid–Liquid Phase Separation

Abstract:Bromadiolone is the most frequently detected anticoagulant rodenticide in human biofluids, resulting in increased cases of poisoning. Here, we present a portable kit that could realize rapid fingerprint detection of DQ in human biofluids. This strategy was based on surface-enhanced Raman scattering (SERS) and salt-induced liquid–liquid phase separation (SILLPS) using a gold/silver core-shell nanoparticle (Au@Ag NPs) substrate. The kit included a portable Raman spectrometer and five tubes that contained Au@Ag NPs colloid, KI solution, saturated NaCl solution, acetonitrile, and Mg(NO3)2 solution. This strategy did not require pretreatment for urine samples. For blood samples, a 2-min SILLPS method was proposed with satisfactory recovery, providing unique insights into efficient pretreatment of complex biomatrix. The detection limits for urine and blood were calculated to be 3.21 ppb and 6.86 ppb, respectively. A clear trend could be observed between signal intensity and logarithmic concentration of bromadiolone ranging from 0 to 2 ppm with high reproducibility. Such a portable kit based on SILLPS-SERS can rapidly provide fingerprint information of poison. This simple-yet-practical “Pretreat & Detect” SERS kit may open up new opportunities for point-of-care detection of poisons and has great potential as a tool in field applications.
Comparative Study for Spectrofluorimetric Determination of Ambroxol Hydrochloride Using Aluminum Metal Transfer Chelation Complex and Biogenic Synthesis of Aluminum Oxide Nanoparticles Using Lavandula spica Flowers Extract

Abstract:The existing study pronounces two newly developed spectrofluorimetric probes for the assay of ambroxol hydrochloride in its authentic and commercial formulations using an aluminum chelating complex and a biogenically mediated and synthesized aluminum oxide nanoparticles (Al2O3NPs) from Lavandula spica flower extract. The first probe is based on the formation of an aluminum charge transfer complex. However, the second probe is based on the effect of the unique optical characteristics of Al2O3NPs in the enhancement of fluorescence detection. The biogenically synthesized Al2O3NPs were confirmed using various spectroscopic and microscopic investigations. The fluorescence detections in the two probes were measured at a λex of 260 and 244 and a λem of 460 and 369 nm for the two suggested probes, respectively. The findings showed that the fluorescence intensity (FI) covered linear concentration ranges of 0.1-200 ng mL-1 and 1.0-100 ng mL-1 with a regression of ˃0.999 for AMH-Al2O3NPs-SDS and AMH-Al(NO3)3-SDS, respectively. The lower detection and quantification limits were evaluated and found to be 0.04 and 0.1 ng mL-1 and 0.7 and 0.1 ng/mL-1 for the abovementioned fluorescence probes, respectively. The two suggested probes were successfully applied for the assay of ambroxol hydrochloride (AMH) with excellent percentage recoveries of 99.65% and 99.85%, respectively. Excipients such as glycerol and benzoic acid used as additives in pharmaceutical preparations, several common cations, and amino acids, as well as sugars, were all found to have no interference with the approach.
Integrated Sensing Platform Validated for the Efficient and On-Site Screening of Amine-Containing Illicit Drugs

Abstract:Efficient and reliable technologies for the on-site detection of illicit drugs are important in drug-facilitated crime investigations. However, the development of such technologies is challenging. Based on the synthetic optimization, introducing a boron ester functional group to the two furanic indicators endows the stimulus-responsive properties synergistically. The ring-opening reaction of the indicators in the presence of amine-containing illicit drugs generated well-known donor-acceptor Stenhouse adducts, accompanied by strong color changes. A small-size and lightweight laminated sensor was integrated based on the outstanding ratiometric variations of the two active furanic indicators. A prototype platform was fabricated equipped with a circuit control, a mini pump, and a signal processing system. A user-friendly detection and efficient screening of amine-containing illicit drugs, including phenethylamines, amphetamines, cathinones, and tryptamines in the liquid states were conducted. The ratiometric response of the sensor was linear in the concentration range of 2.1-10.6 μg·mL-1 for methamphetamine·HCl and methcathinone ·HCl. The detection limits for the two illicit drugs at the sublevel (ng·mL-1) were found to be 8.4 and 9.0 ng·mL-1, respectively. Double-blind field tests and different illicit drugs were evaluated with good screening capability. Successful trials showed the potential applications of the developed prototype platform for efficient and on-site analytical determination.

Abstract:Salbutamol (SAL) residues in edible animal tissues have various adverse effects on human health after digestion. Herein, we constructed an innovative functionalized microorganism nanozyme-mediated immunochromatographic assay for sensitive and accurate dual-colorimetric detection of SAL by introducing the Yeast@IrO2 as bifunctional signal tag. The Yeast@IrO2 was synthesized by self-assembling iridium (IV) oxide nanoparticles (IrO2 NPs) on the surface of inactivated yeast cells, which showed high monodispersity, uniform shapes, preeminent biocompatibility and rapid antibodies labeling capacity. After reaction for 15 min, through further employing [3,3′,5,5’ Tetramethylbenzidine (TMB) and hydrogen peroxide (H2O2)] as substrate to trigger the color reaction, the developed biosensor performed with outstanding performance, including limit of detection (0.012 ng/mL), linearity (R2 > 0.99) and selectivity. Concurrently, the visual limit of detection (0.2 μg/kg) and satisfactory recoveries (85.6%–103.9%) were obtained by monitoring SAL in pig liver and beef samples. Briefly, this work provided a universal platform for sensitive, precise and rapid detection of toxic and hazardous substances in food safety fields.
Combination of in-situ electro synthesized Zn-Al-LDH@ pencil graphite fiber and three phase hollow fiber LPME for microextraction of some antibiotics in urine samples and quantification via HPLC-UV

Abstract:In the present study, a combination of two useful extraction techniques including fiber solid phase microextraction based on the new synthesized sorbent and the porous hollow fiber liquid phase microextraction is reported. The selective anion-exchangeable Zn-Al-layered double hydroxide as a fiber was electrodeposited on the surface of pencil graphite substrate through simple and cost-effective method. The prepared fiber was characterized and put into the lumen of the porous hollow fiber, which was filled by alkaline solution. The obtained optimum condition for the method was as follows: donor phase HCl concentration of 0.10 mM, acceptor phase NaOH concentration of 7.60 mM and extraction time of 16 min. It is necessary to adjust the concentration of the donor phase in order to extract acidic analytes from an aqueous sample into an organic phase. The method was used for the quantification of three antibiotics including amoxicillin, ciprofloxacin and cefixime. The extracted antibiotics were analyzed by high performance liquid chromatography-ultraviolet detection (HPLC-UV). In the optimal conditions, the linearity of the proposed method was in the range of 0.50-500.00 μg L-1 for selected analytes in water and urine matrices. All calibration curves showed determination of coefficient higher than 0.9958. The limits of detection and limits of quantification were calculated to be between 0.08 and 0.16 μg L-1 and 0.28-0.56 μg L-1, respectively. In order to define the precision of the method, the inter-day and intra-day relative standard deviations were determined between 3.25 and 5.73% in the three selected concentrations. The enrichment factors (EFs) were between 46.20 and 55.81 for selected drugs. Moreover, the calculated absolute recoveries were between 69.30 and 83.71%. The relative recoveries were obtained in the range of 93.00-105.00%. The proposed method was also employed for analysis of various urine samples containing the selected drugs. The obtained recoveries indicated that the method was useful and applicable in complicated biological samples.
Novel disposable screen-printed sensors based on copper oxide nanoparticles for sensitive voltammetric assay of antazoline

Abstract:The fabrication and electroanalytical validation of a novel homemade screen-printed carbon electrode enriched with copper oxide nanoparticles (CuONPs/SPCEs) were demonstrated for sensitive and selective voltammetric determination antazoline (ANT) in eye drop solution and spiked serum samples. With a reported electrocatalytic activity, CuONPs catalyzed the oxidation of ANT at the electrode surface showing an irreversible anodic oxidation peak at 0.9 V under a diffusion-controlled electrode reaction. Based on the electroanalytical and molecular orbital calculation studies, oxidation of ANT molecule undergoes via oxidation of the aliphatic nitrogen atom within the ANT moiety. Linear calibration curves were illustrated covering the ANT concentration ranged from 0.01 to 3.819 μg mL −1 with and limit of detection 3 ng mL −1 . The fabricated sensors enriched with CuONPs exhibited high fabrication and measurement reproducibility with a prolonged shelf lifetime (6 months). The electroanalytical approach presented adequate sensitivity and selectivity with a short analysis time, applying low-cost instrumentation and simple pretreatment protocol avoiding handling hazardous organic solvents. Trace ANT was monitored in the presence of different interfering species, degradation products, and xylometazoline hydrochloride (ZYL) in the co-formulated ophthalmic pharmaceutical samples. The CuONPs based voltammetric sensors were introduced as a reliable eco-friendly analytical tool for monitoring ANT in ophthalmic solutions and biological fluids with acceptable recovery values.

Abstract:We have described a sulfonamide-selective ambient ion source coupled with electrospray ionization mass spectrometry (ESI-MS) for selective extraction and determination of trace sulfonamide antibiotics. It is obtained by modifying an iron sheet with a sulfadiazine-templated hydrophilic molecularly imprinted polymer (SF-HMIP). It behaves as both an online extractor and a MS ion source. Five sulfonamide antibiotics, including sulfamethoxazole (SMZ), sulfamerazine (SMR), sulfisoxazole (SIZ), sulfathiazole (ST), and sulfameter (SMD), were chosen to evaluate SF-HMIP coupled with ESI-MS, which showed good linearity in the range of 0.2-1000 ng/mL with correlation coefficient values (R2) over 0.9946. The limits of detection (LODs) for analysis of pure water and honey were in the range of 0.1-0.2 and 0.2-1.5 ng/mL, respectively. Limits of quantitation (LOQs) for analysis of pure water and honey were in the range of 0.3-0.5 and 1.0-5.0 ng/mL, respectively. The results demonstrated that SF-HMIP combined with ESI-MS could be applied for the direct analysis of five trace sulfonamide compounds in honey and pure water with recoveries ranging from 76 to 129%.
Ag/N-doped Reduced Graphene Oxide Incorporated with Molecularly Imprinted Polymer: an Advanced Electrochemical Sensing Platform for Salbutamol Determination.

Abstract:In this work, the metallic silver and non-metallic nitrogen co-doped reduced graphene oxide (Ag-N-RGO) was first synthesized by a simple and cost-effective strategy, and then a molecularly imprinted polymer (MIP) was formed in situ at the surface of the prepared composite via electropolymerization of o-phenylenediamine in the presence of salbutamol as the template molecule. The electrochemical characterizations demonstrate that the bifunctional graphene-based composite shows improved catalytic performance than that of pristine graphene doped with one-component or none. The MIP sensor based on Ag-N-RGO owns high porous surface structure, resulting in the increased current response and enhanced recognition capacity than that of non-imprinted sensor. The outstanding performance of the developed sensor derives from the combined advantages of Ag-N-RGO with effective catalytic property and MIP with excellent selectivity. Under the optimal conditions, the electrochemical response of the developed sensor is linearly proportional to the concentration of salbutamol in the range of 0.03–20.00µmolL−1 with a low detection limit of 7 nmol L−1. The designed sensor has exhibited the multiple advantages such as low cost, simple manufacture, convenient use, excellent selectivity and good reproducibility. Finally, the proposed method has been extended for the determinations of salbutamol in human urine and pork samples, and the satisfactory recoveries between 98.9–105.3% are achieved.
Development of bismuth sulfide nanorods and polyamidoamine dendrimer on reduced graphene oxide as electrode nanomaterials for electrochemical determination of salbutamol

Abstract:Dendrimers, a new class of nanomaterials, are receiving more attention in various fields. In this study, by combining the advantages of polyamidoamine (PAMAM) dendrimer with reduced graphene oxide (rGO) and bismuth sulfide (Bi2S3), we came to design a new composite and its application for electrochemical sensors was investigated for the first time. As a new approach in the preparation of the composite, PAMAM was used for the first time to increase the surface of Bi2S3 with rGO, which ultimately led to an increase in the active surface area of the sensor (5 times compared to the bare electrode). For the first time, we used the sonochemical method for interaction between PAMAM with Bi2S3 and rGO, which was a simpler and faster method to prepare the composite. The purposeful design of the composite was done by using the experimental design method to obtain the optimum composition of components. The new nanocomposite was successfully applied for simple and sensitive electrochemical sensing of salbutamol for controlling the health of food. Salbutamol is used as a prohibited additive in animal and poultry feed. The sensor has good sensitivity (35 times increase compared to the bare electrode) and a low detection limit (1.62 nmol/L). Moreover, it has acceptable selectivity, good repeatability (1.52-3.50%), good reproducibility (1.88%), and satisfactory accuracy (recoveries: 84.6-97.8%). An outstanding feature of the sensor is its broad linear range (5.00-6.00 × 102 nmol/L). This sensor is well suited for the determination of salbutamol in milk, sausage, and livestock and poultry feed samples.
Application of Aluminum Hydroxide for Improvement of Label-Free SERS Detection of Some Cephalosporin Antibiotics in Urine

Abstract:This report is dedicated to development of surface-enhanced Raman spectroscopy (SERS) based analysis protocol for detection of antibiotics in urine. The key step of the protocol is the pretreatment of urine before the detection to minimize background signal. The pretreatment includes extraction of intrinsic urine components using aluminum hydroxide gel (AHG) and further pH adjusting of the purified sample. The protocol was tested by detection of a single antibiotic in artificially spiked samples of real urine. Five antibiotics of cephalosporin class (cefazolin, cefoperazone, cefotaxime, ceftriaxone, and cefuroxime) were used for testing. SERS measurements were performed using a portable Raman spectrometer with 638 nm excitation wavelength and silver nanoparticles as SERS substrate. The calibration curves of four antibiotics (cefuroxime is the exception) cover the concentrations required for detection in patient's urine during therapy (25/100‒500 μg/mL). Random error of the analysis (RSD < 20%) and limits of quantification (20‒90 μg/mL) for these antibiotics demonstrate the applicability of the protocol for reliable quantitative detection during therapeutic drug monitoring. The detection of cefuroxime using the protocol is not sensitive enough, allowing only for qualitative detection. Additionally, time stability and batch-to-batch reproducibility of AHG were studied and negative influence of the pretreatment protocol and its limitations were estimated and discussed.
