Abstract:Background: Food grade Streptococcus thermophilus produces biological exopolysaccharides (EPSs) with great potential with respect to catering for higher health-promoting demands; however, how S. thermophilus regulates the biosynthesis of EPS is not completely understood, decelerating the application of these polymers. In our previous study, maltose, soy peptone and initial pH were three key factors of enhancing EPS yield in S. thermophilus CS6. Therefore, we aimed to investigate the regulating mechanisms of EPS biosynthesis in S. thermophilus CS6 via the method of comparative transcriptome and differential carbohydrate metabolism. Results: Soy peptone addition (58.6 g L-1 ) and a moderate pH (6.5) contributed to a high bacterial biomass and a high EPS yield (407 mg L-1 ). Maltose, soy peptone and initial pH greatly influenced lactose utilization in CS6. Soy peptone addition induced a high accumulation of mannose and arabinose in intracellular CS6, differential monosaccharide composition (mannose, glucose and arabinose) in EPS and high radical [2,2-diphenyl-1-picrylhydrazyl, superoxide and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)] scavenging activities. Carbohydrate transportation, sugar activation and eps cluster-associated genes were differentially expressed to regulate EPS biosynthesis. Correlation analysis indicated high production of EPSs depended on high expression of lacS, galPMKUTE, pgm, gt2-5&4-1 and epsLM. Conclusion: The production of antioxidant EPS in S. thermophilus CS6 depended on the regulation of galactose metabolism cluster and eps cluster. The present study recommends a new approach for enhancing EPS production by transcriptomic regulation for further food and health application of EPS. © 2022 Society of Chemical Industry.

Abstract:Our results highlighted a greater production of EPS from the natural colonies of Microcystis than the unicellular strain under nutrient and combined salinity stress conditions, dominated by the attached form. EPS produced by the colonial form were characterized by more diverse monosaccharides enriched in uronic acids and hexosamines. This complex extracellular matrix gives the cells the ability to aggregate and allows the colonial cyanobacterial population to better cope with an osmotic shock. The transfer of toxic cyanobacterial Microcystis blooms from freshwater to estuaries constitutes a serious environmental problem worldwide that is expected to expand in scale and intensity with anthropogenic and climate change. The formation and maintenance of Microcystis in colonial form is conditioned to the presence of extracellular polymeric substances (EPS). In this study, we attempted to better understand how the mucilaginous colonial form of Microcystis evolves under environmental stress conditions. In particular, we studied and compared the production and the composition of EPS fractions (attached and free) from natural colonies of a Microcystis bloom and from a unicellular M. aeruginosa strain under salinity and nutrient stress (representing a land‐sea continuum). Our results highlighted a greater production of EPS from the natural colonies of Microcystis than the unicellular one under nutrient and combined stress conditions dominated by the attached form. In comparison to the unicellular Microcystis, EPS produced by the colonial form were characterized by high molecular weight polysaccharides which were enriched in uronic acids and hexosamines, notably for the free fraction in response to increased salinities. This complex extracellular matrix gives the cells the ability to aggregate and allows the colonial cyanobacterial population to cope with osmotic shock.

Abstract:Freshwater cyanobacteria can produce large amount of mucilage, particularly during large blooms. The production of these carbon-rich exopolymers (EPS) should influence the carbon-to-nutrient ratios of the organic matter (OM), which are regularly used as a proxy for the herbivorous food quality. However, little is known about the consequences of EPS production on the carbon-to-nutrient ratio of the OM. Two EPS forms can be distinguished: the free fraction composed of soluble extracellular polymeric substances (S-EPS) and the particulate fraction corresponding to the transparent exopolymer particles (TEP). The aim of the study was to determine whether the TEP and S-EPS productions by cyanobacteria influence the carbon-to-nutrient ratios of the particulate OM (POM). Five cyanobacteria species were grown in batch culture and characterized in terms of photosynthetic activity, EPS production, and C, N, P contents. The variability in EPS production was compared with the variability in stoichiometry of the POM. Most of cyanobacteria live in association with heterotrophic bacteria (HB) within the mucilage. The effect of the presence/absence of HB on EPS production and the carbon-to-nutrient ratios of the POM was also characterized for the cyanobacteria Microcystis aeruginosa. We showed that TEP production increased the carbon-to-nutrient ratios of the POM in the absence of HB, while the stoichiometry did not significantly change when HB were present. The C:N ratio of the POM decreased with production of S-EPS by the five species. Lastly, the three colonial species (Chroococcales) tend to produce more TEP than the two filamentous species (Oscillatoriales), with the two picocyanobacteria being the most productive of both TEP and S-EPS.

Abstract:The cyanobacterium Microcystis mainly exists in colonies under natural conditions but as single cells in typical laboratory cultures. Understanding the mechanism by which single cells form small and large colonies can provide a deeper insight into the life history of Microcystis and the mechanisms of Microcystis bloom formation. In this paper, Microcystis aeruginosa cultured under varying light intensities and temperatures exhibited different specific growth rates. Correlations were found between the specific growth rate, extracellular polysaccharide (EPS) content, and morphology of M. aeruginosa. Under low light intensities and temperatures, M. aeruginosa formed small colonies (maximum colony size approximately 100 μm) and exhibited low specific growth rates. By contrast, standard culture conditions yielded single or paired cells with high specific growth rates. Moreover, the EPS content decreased dramatically with increasing specific growth rate. A significant positive linear relationship was observed between the EPS content per cell and colony size. High EPS content and colony formation were associated with low specific growth rates. The specific growth rate in laboratory cultures was higher than the in situ growth rate under natural conditions. This result may explain why Microcystis normally exists as single cells or (more rarely) as paired cells in axenic laboratory cultures after long-term cultivation, but forms colonies under natural conditions.

Abstract:As an important carbon (C) storage in biological soil crusts (BSCs), exopolysaccharides (EPSs) are not only a part of the desert C cycle, but also the key materials for cyanobacteria to resist desert stress. In this study, the influence of initial N concentrations (10, 25 and 50 mg L-1 designated as N10, N25 and N50 respectively) on Microcoleus vaginatusis growth and the excretion of EPSs including RPS (released exopolysaccharides) and CPS (capsule exopolysaccharides) were evaluated at different growth periods. In logarithmic period, higher ratio of biomass to EPSs indicated by (DW-CPS)/EPSs was observed in the N50 group with the highest N concentration (about 40 mg L-1) in the medium, while no difference was observed among the three groups in stationary period when the N concentrations of medium were lower than 25 mg L-1. The CPS/RPS showed similar results with (DW-CPS)/EPSs, and stayed higher than 1 in each group. Notably, obvious difference displayed in the monosaccharidic composition and morphologies between CPS and RPS, but not the N levels. The changes of C/N in cells at different growth period indicate that the excretion of EPSs, a mechanism that maintains the balance of cell C/N ratio, only works when the N in the environment is sufficient. Our results showed that, as the raw material and environmental signal, environmental N concentration regulates the elements (C and N) percentage of cyanobacterial cells and its EPSs excretion pattern, but not the monosaccharidic composition or the morphologies. These results also implied that, as the essential self-protecting materials, more EPSs with higher proportion of CPS would be excreted to response the low N environment.

Abstract:The effects of cell growth phase, substrate type, NaCl, CaCl2 and C/N ratio on the anaerobic production of water-soluble extracellular polymeric substances (EPS) by the photosynthetic bacterium, Rhodopseudomonas acidophila, were examined. The protein content of the EPS decreased from 96.7 to 39.0mgg−1 dry cell weight with increase in cultivation time from 20 to 100h, but remained almost unchanged thereafter. EPS production was inversely proportional to bacterial dry weight. The amount of EPS produced by R. acidophila decreased in order of the following carbon sources: benzoate>acetate>butyrate>propionate>succinate>malonate. High NaCl concentrations resulted in more EPS, but CaCl2 concentration had little or no influence on the EPS production. The magnitude of carbon and nitrogen sources influenced the total amount of EPS, and also the ratio of carbohydrate to protein in its composition.

Abstract:Exopolysaccharides (EPSs), which are produced by lactic acid bacteria, have been found to improve the texture and functionality of fermented dairy products. In a previous study, four nitrogen sources were identified as affecting the yield, molecular weight and structure of EPSs produced by Streptococcus thermophilus IMAU20561 in M17 medium. In this genomic and transcriptomics study, a novel eps gene cluster responsible for assembly of repeating units of EPS is reported. This eps cluster (22.3 kb), consisting of 24 open reading frames, is located in the chromosomal DNA. To explore the biosynthetic mechanisms in EPS, we completed RNA-seq analysis of S. thermophilus IMAU20561 grown in four different nitrogen sources for 5 h (log phase) or 10 h (stationary phase). GO functional annotation showed that there was a significant enrichment of differentially expressed genes (DEGs) involved in: amino acid biosynthesis and metabolism; ribonucleotide biosynthesis and metabolism; IMP biosynthesis and metabolism; and phosphorus metabolism. KEGG functional annotation also indicated enrichment of DEGs involved in amino acid biosynthesis, glycolysis, phosphotransferase system, fructose, and mannose metabolism. Our findings provide a better understanding the genetic traits of S. thermophilus , the biosynthetic pathways needed for the production of EPS, and a theoretical basis for screening dairy starter cultures.

Abstract:This work aimed to develop the submerged cultivation conditions for improved exopolysaccharides (EPS) production by Armillaria luteo-virens Sacc. The effects of culture temperature, aeration rate, inoculum level, initial pH, and additives on EPS formation and mycelial growth are investigated. The aeration rate, initial pH, and inoculum level significantly affected EPS production under the submerged cultivation. The developed conditions were as follows: cultivation temperature 23 °C, initial pH 5.0, aeration rate 0.5 vvm, 0.5% Tween 80, inoculum level 5% (v/v), and shaking speed 120 r/min. Under the developed conditions, the highest EPS production was 13.01 g/L at 5 days culture time. EPS production was examined in a 5 L bioreactor, and an unstructured kinetic model for EPS formation was well developed. The verified investigations in the large-scale cultivation system showed that the developed models are able to predict the submerged cultivation process of EPS formation. Current results revealed that the submerged cultivation conditions can be utilized to control EPS production, and the unstructured models developed are suitable for explaining EPS production by A. luteo-virens Sacc QH in a large-scale cultivation bioreactor.

Abstract:Exopolysaccharides (EPS) produced by microorganisms play a vital role in physiological and ecological processes. However, the mechanisms of EPS synthesis and release in anaerobic environments remain poorly understood. Here, we provide the first evidence of anaerobic EPS synthesis by the fungus Schizophyllum commune 20R-7-F01, isolated from coal-bearing sediments ~2.0 km below the seafloor. Under anaerobic conditions, the fungus exhibited significantly higher specific EPS production (1.57 times) than under aerobic conditions. Transcriptomic analysis revealed 2057 differentially expressed genes (DEGs) in the strain cultured anaerobically for 7 days compared to aerobically. Among these genes, 642 were significantly upregulated, while 1415 were significantly downregulated, mainly associated with carbon metabolism pathways. Genes involved in glycolysis and EPS synthesis, including hexokinase (HK), phosphoglucomutase (PGM), and (1 → 3)-β-glucan synthase (GLS), were significantly upregulated, while those related to the TCA cycle, respiratory chain, and pentose phosphate pathway were downregulated under anaerobic conditions. These findings highlight the oxygen-dependent regulation of EPS synthesis and suggest that EPS may serve as a key mechanism for fungal adaptation to anaerobic environments.
Effects of nutrients on exopolysaccharide production and surface properties of Aeromonas salmonicida

Abstract:Extracellular polysaccharide (EPS) and capsular polysaccharide (CPS) production by Aeromonas salmonicida A450 and the influence of the capsule on cell surface properties were studied. A. salmonicida did not produce CPS or EPS when glucose, phosphate, magnesium chloride, or trace mineral components were absent from the medium. The addition of yeast extract improved capsule production. Neither EPS nor CPS formation depended on the C/N ratio, although it appeared to be influenced by the level of carbon and nitrogen in the culture. Both EPS and CPS production started at the end of the logarithmic growth phase. The amounts of EPS and CPS produced were not influenced by temperature changes between 15 and 20 degrees C and was maximal from pH 7 to 7.5. Cell surface properties were strongly influenced by capsule production; high CPS production was associated with enhanced cell hydrophilicity and autoagglutination. The effect of CPS on cell surface properties was independent of the presence of the surface protein array (A-layer).

Abstract:Extracellular polysaccharides (EPSs) produced by microalgae and cyanobacteria are molecules with a great ecological significance for the producing organisms, serving in a wide array of biological processes and increasing the organism tolerance to environmental stresses. In addition, due to their distinctive chemical, rheological properties and the biological activity of some of them, these macromolecules could find application in industrial, pharmaceutical and medical fields. In this chapter, the current knowledge regarding the structure and composition of EPSs, the factors eliciting their synthesis, and the related codifying genes involved is reviewed. It is also underlined and discussed how, notwithstanding the huge number of studies available, different aspects still need to be clarified. For example, there are limited informations concerning EPS synthetic pathways, and how these processes are influenced by environmental factors. Expand the knowledge on these aspects could help elaborating biotechnological strategies to increase the yields and direct the synthesis to polymers with desired characteristics.
Proteome trade-off between primary and secondary metabolism shapes acid stress induced bacterial exopolysaccharide production

Abstract:The exopolysaccharide (EPS) produced by is a high-value bioproduct in food and health industries, and its biosynthesis has been found as a secondary metabolic pathway to mediate acid stress. To quantitatively investigate acid stress response in and model EPS production, this study measured metabolomics, proteomics and growth data for HMX2 cultured at 4 different pH values. The growth and metabolomics data showed that under acid stress, the EPS production flux was evidently enhanced while the glycolysis and cellular growth were inhibited. The following proteomic analysis found that EPS biosynthetic proteins were significantly up-regulated under acid stress and pinpointed Fur as the most probable transcriptional factor controlling EPS biosynthesis in . Furthermore, we identified a proteome trade-off between primary metabolism and EPS biosynthesis, which were then mechanistically depicted by a regulatory proteome constrained flux balance analysis (RPCFBA) model. As the first metabolic model that can simulate secondary metabolism, the RPCFBA model demonstrated good accuracy in predicting growth rates and EPS production fluxes of HMX2, validated by experimental data. The perturbation on carbon sources further showed the potential of applying the presented modeling framework to the design and control of microbial secondary metabolism.

Abstract:Microalgal-bacterial symbioses are prevalent in aquatic ecosystems and play a pivotal role in carbon sequestration, significantly contributing to global carbon cycling. The understanding of the contribution of exopolysaccharides (EPSs), a crucial carbon-based component, to the structural integrity of microalgal-bacterial symbioses remains insufficiently elucidated. To address this gap, our study aims to enhance our comprehension of the composition and primary structure of EPSs within a specific type of granular microalgal-bacterial symbiosis named microalgal-bacterial granular sludge (MBGS). Our investigation reveals that the acidic EPSs characteristic of this symbiosis have molecular weights ranging from several hundred thousand to over one million Daltons, including components like glucopyranose, galactopyranose, mannose, and rhamnose. Our elucidation of the backbone linkage of a representative exopolysaccharide revealed a →3)-β-D-Galp-(1→4)-β-D-Glcp-(1→ glycosidic linkage. This linear structure closely resembles bacterial xanthan, while the branched chain structure bears similarities to algal EPSs. Our findings highlight the collaborative synthesis of acidic EPSs by both microalgae and bacteria, emphasizing their joint contribution in the production of macromolecules within microalgal-bacterial symbiosis. This collaborative synthesis underscores the intricate molecular interactions contributing to the stability and function of these symbiotic relationships.

Abstract:Microcystis possesses the capacity to form colonies and blooms in lakes and reservoirs worldwide, causing significant ecological challenges in aquatic ecosystems. However, little is known about the determining factors of physico-chemical surface properties that govern the competitive advantage of Microcystis. Here, The physico-chemical surface properties of Microcystis wesenbergii and Microcystis aeruginosa, including specific surface area (SSA), hydrophobicity, zeta potential, and functional groups were investigated. Additionally, the extracellular polysaccharide (EPS) were analyzed. Laboratory-cultured Microcystis exhibited hydrophilic, a negative zeta potential and negatively charged. Furthermore, no significant relationship was shown between these properties and the cultivation stage. Microcystis wesenbergii exhibited low free energy of cohesion, high surface free energy, high growth rate, and high EPS content during the logarithmic phase. On the other hand, M. aeruginosa displayed lower free energy of cohesion, high surface free energy, high EPS content, and high growth rate during the stationary phase. These characteristics contribute to their respective competitive advantage. Furthermore, the relationship between EPS and surface properties was investigated. The polysaccharide component of EPS primarily influenced the SSA and total surface energy of Microcystis. Likewise, the protein component of EPS influenced hydrophobicity and surface tension. The polysaccharide composition, including glucuronic acid, xylose, and fructose, mainly influenced surface properties. Additionally, hydrophilic groups such as O-H and P-O-P played a crucial role in determining hydrophobicity in Microcystis. This study elucidates that EPS influenced the SSA, hydrophobicity, and surface free energy of Microcystis cells, which in turn impact the formation of Microcystis blooms and the collection.
Improvement of Exo-Polysaccharides Production and Modeling Kinetics by Armillaria Luteo-Virens Sacc. in Submerged Cultivation

Abstract:Armillaria luteo-virens Sacc. is a special medicinal and edible mushroom in China, and its wild type has been investigated that it contains some functional components. The optimization of submerged culture conditions for mycelium growth and exo-polysaccharide (EPS) production in an edible mushroom A. luteo-virens Sacc. was studied in shake flasks and bioreactors with response surface methodology (RSM). A fractional factorial design (FFD) and central composite design (CCD) were applied to optimize the main factors that affect the fungal polysaccharides production in a submerged culture. Glucose is very important for EPS production by A. luteo-virens Sacc., and yeast extract powder also significantly affects fungal EPS production through fractional factorial experiment. The optimized cultivation medium constituents were derived from the predicted model using central composite design as follows: 31.26g/l glucose, 1.06g/l yeast extract powder, 1.00g/l K2HPO4, 0.50g/l KH2PO4, and 0.45g/l MgSO4·7H2O, pH unadjusted. The predicted maximal EPS concentration was 5.40g/l using the optimized culture medium compositions at 23°C for 5 days. Additionally, the submerged cultivation process was explained using the optimized cultivation medium. The cultivation kinetics model constructed was suitably depicting the culture process in bioreactor cultivation scale.

Abstract:Exopolysaccharides (EPSs) produced by diverse group of microbial systems are rapidly emerging as new and industrially important biomaterials. Due to their unique and complex chemical structures and many interesting physicochemical and rheological properties with novel functionality, the microbial EPSs find wide range of commercial applications in various fields of the economy such as food, feed, packaging, chemical, textile, cosmetics and pharmaceutical industry, agriculture, and medicine. EPSs are mainly associated with high-value applications, and they have received considerable research attention over recent decades with their biocompatibility, biodegradability, and both environmental and human compatibility. However, only a few microbial EPSs have achieved to be used commercially due to their high production costs. The emerging need to overcome economic hurdles and the increasing significance of microbial EPSs in industrial and medical biotechnology call for the elucidation of the interrelations between metabolic pathways and EPS biosynthesis mechanism in order to control and hence enhance its microbial productivity. Moreover, a better understanding of biosynthesis mechanism is a significant issue for improvement of product quality and properties and also for the design of novel strains. Therefore, a systems-based approach constitutes an important step toward understanding the interplay between metabolism and EPS biosynthesis and further enhances its metabolic performance for industrial application. In this review, primarily the microbial EPSs, their biosynthesis mechanism, and important factors for their production will be discussed. After this brief introduction, recent literature on the application of omics technologies and systems biology tools for the improvement of production yields will be critically evaluated. Special focus will be given to EPSs with high market value such as xanthan, levan, pullulan, and dextran.

Ecological significance and industrial application of extracellular polysaccharides from cyanobacteria: A review

Abstract:Cyanobacteria can produce extracellular polysaccharides (EPS) and release them to the environment, which is one of the adaptive mechanisms of cyanobacteria evolved to face complicated and variable environment. As a protective barrier between cell and environment, the EPS play a protective role against desiccation, ultraviolet radiation, bio-mineralization, or predation by protozoans. The EPS produced by cyanobacteria are a kind of complex anionic heteropolymer polysaccharides, more than 75% of which are composed of six or more monosaccharaides. Glucose is the most frequent kind of monosaccharaides. The characteristics of the EPS vary with cyanobacterial species. Most of the EPS show an anionic feature due to the charged groups, such as uronic acid and sulfate groups. Sulfate group is the basis of the anti-viral properties of polysaccharides, while hydrophobic groups such as ester-linked acetyl groups, peptides moieties, and deoxy sugars contribute to emulsifying properties. Therefore, the EPS produced by cyanobacteria have a broad application prospect in many industries, such as food additives, cosmetology, pharmacology, and wastewater treatment. The industrial application of the EPS can not only reuse the cyanobacteria bloom, bringing considerable economic benefits, but also provide a solution to the salvaging cyanobacteria distribution problem, avoiding its secondary pollution. Until now, there is no commodity from cyanobacterial EPS, and many technical problems are needed to be solved between the theoretical study and large batch industrial production.

Abstract:Abstract Streptococcus thermophilus ASCC 1275 has two chain length determining genes - epsC and epsD - in its eps gene cluster, and produces two times more EPS in sucrose medium than that in glucose and lactose. Hence, we investigated the influence of sugars (glucose, sucrose and lactose), at log phase (5 h) and stationary phase (10 h), on the global proteomics of S. thermophilus 1275 to understand the differentially expressed proteins (DEPs) during EPS production using isobaric tags for relative and absolute quantitation (iTRAQ)-based proteomic analysis. Among 98 DEPs in sucrose medium, most of them were mapped into EPS biosynthesis pathway and other related metabolisms. There was an upregulation of several proteins involved in sugar transport (phosphoenolpyruvate (PEP) phosphotransferase system), EPS assembly ( epsG1D ) and amino acid metabolism (methionine, cysteine/arginine metabolism) in sucrose medium. This study showed that increased EPS production in S. thermophilus 1275 requires a well-co-ordinated regulation of pathway involved in both EPS assembly and amino acid metabolism along with the availability of sugars. Thus, it provided valuable insights into the biosynthesis and regulation of EPS in S. thermophilus 1275, and potential gene targets for understanding high-EPS strains.

Towards Understanding the Role of Extracellular Polymeric Substances in Cyanobacterial Microcystis Aggregation and Mucilaginous Bloom Formation

Abstract:The development of mucilaginous cyanobacterial Microcystis blooms is a serious environmental and ecological problem, and information on the bloom-formation mechanism has been lacking until now. The aggregation of microbial cells was attributed to the matrix of extracellular polymeric substances (EPS). In this study, the quantitative role of EPS matrix in Microcystis aggregation and mucilaginous bloom formation was investigated. The results showed that when EPS matrix was extracted, the aggregation abilities decreased by 27.6% and 57.4% for the lab-cultured Microcystis suspension and the field-sampled Microcystis aggregates, respectively. The extended DLVO theory revealed that EPS extraction increased the energy barrier and the values of the second energy minimum, which accounted for the deteriorated aggregation. Further analysis showed an increasing attraction energy of EPS matrix during the Microcystis bloom development, whereas the predominant contribution originated from tightly bound EPS (TB-EPS) and loosely bound EPS (LB-EPS) for the lab-cultured and field-sampled Microcystis samples. The heterogeneous energy contribution of EPS subfractions was found to be associated with the variations in organic contents. Specifically, Microcystis aggregates exhibited a higher organic content of TB-EPS than of LB-EPS compared with the lab-cultured Microcystis suspension, whereas organic content in only the LB-EPS fraction for the bloom samples was significantly higher (p < 0.01) than that of the Microcystis aggregates. Based on these results, a conceptual model of EPS function was proposed in which TB-EPS plays an important role in the formation of Microcystis aggregates, after which LB-EPS contributed to the subsequent development from Microcystis aggregates to mucilaginous bloom formation.
Effects of Nutrients on the Growth of and Free Exopolysaccharide Biosynthesis by Streptococcus thermophilus 937 in a Chemically Defined Medium

Abstract:The free exopolysaccharide (f-EPS) produced by Streptococcus thermophilus is a natural texture modifier with health-promoting properties and has thus become one of the most interesting metabolites for researchers. The present work aimed to further understand the nutritional requirements for the growth of and the f-EPS production by S. thermophilus. The types and concentrations of compounds in the complete chemically defined medium were changed in turn to evaluate the effects of single nutrients on the growth of and f-EPS production by S. thermophilus 937. The results showed that cysteine, glutamine, histidine, methionine, tryptophan, tyrosine, leucine, isoleucine, and valine played an important role in maintaining the rapid and stable growth of S. thermophilus 937. S. thermophilus 937 also required calcium pantothenate, niacin, pyridoxine, riboflavin, and thiamine hydrochloride as essential nutrients for growth. Increases in the concentrations of lactose, glutamate, histidine, or isoleucine significantly increased the production of free exopolysaccharide by S. thermophilus 937, and when the lactose concentration increased to 20 g·L-1 and the concentration of the three-amino-acid combination increased to 15 mM, the f-EPS yield increased to a maximum of 35.34 μg·mL-1. This finding indicated that lactose and the 3 amino acids exert synergistic effects on the promotion of f-EPS production. In addition, lactose and the three amino acids have strain specific promoting effects on f-EPS production by S. thermophilus. This study provides a further understanding of the effects of nutrients on the biosynthesis of f-EPS by S. thermophilus.
