Abstract:Polyamines are low molecular weight amines that serve numerous pivotal functions in eukaryotic cells, such as contributing to processes like embryogenesis, organogenesis in plants, and host interactions in fungi. Over the years, there has been significant interest in these compounds concerning the enhancement of plant resistance to pathogenic infections. As polyamines play indispensable roles in both plant and fungal functionalities, elucidating their precise significance in the interplay between these two organisms remains intricate. Furthermore, it is becoming evident that polyamines can yield disparate effects contingent upon factors such as prevailing conditions, concentrations, and the specific species of both plants and fungi.

Abstract:Agrobacterium tumefaciens causes crown gall disease on various plant species by introducing its T-DNA into the genome. Therefore, Agrobacterium has been extensively studied both as a pathogen and an important biotechnological tool. The infection process involves the transfer of T-DNA and virulence proteins into the plant cell. At that time the gene expression patterns of host plants differ depending on the Agrobacterium strain, plant species and cell-type used. Later on, integration of the T-DNA into the plant host genome, expression of the encoded oncogenes, and increase in phytohormone levels induce a fundamental reprogramming of the transformed cells. This results in their proliferation and finally formation of plant tumors. The process of reprogramming is accompanied by altered gene expression, morphology and metabolism. In addition to changes in the transcriptome and metabolome, further genome-wide ("omic") approaches have recently deepened our understanding of the genetic and epigenetic basis of crown gall tumor formation. This review summarizes the current knowledge about plant responses in the course of tumor development. Special emphasis is placed on the connection between epigenetic, transcriptomic, metabolomic, and morphological changes in the developing tumor. These changes not only result in abnormally proliferating host cells with a heterotrophic and transport-dependent metabolism, but also cause differentiation and serve as mechanisms to balance pathogen defense and adapt to abiotic stress conditions, thereby allowing the coexistence of the crown gall and host plant.

Abstract:In the present review, we concentrated on primary signaling and regulatory events in plant cells that are rapidly evoked in response to extracellular polyamines in vivo and induce cellular and biochemical responses. Downstream pathways that allow polyamines to regulate plant growth, development, and stress tolerance are analyzed. The results of the recent studies on polyamine metabolism and its regulatory mechanisms are also discussed. Taken together, the data suggest that endogenously and exogenously regulated polyamines by activating ion transport, calcium dynamics, lipid, protein kinase, protein conjugation, and nucleic acid regulation mechanisms modulate downstream transcriptomic, proteomic, metabolomic, and hormonal pathways affecting growth, development, and stress tolerance.

Abstract:Polyamines (PAs), including Spermidine (Spd), Spermine (Spm) and Putrescine (Put) are small physiologically active alkli compound that are ubiquitous in all plant cells. Polyamines play a critical role in the somatic embryogensis, development of flowers and fruit, anti-senescence and stress challenges. Many results in recent years showed that Polyaminese have positive effect on fruit setting, fruit growth and fruit quality in many fruit crops. The present paper reviews the physiological action of polyamines on pollen growth, fruit setting and fruit growth and especially discusses the important roles of polyamines in fruit development in various stress. The mechanism of physiological action of polyamines on fruit development and potential application of polyamines for increasing blossoming and fruit setting and the prospect of research on molecular mechanism are also discussed.

Abstract:This review focuses on the intricate relationship between plant polyamines and the genetic circuits and signaling pathways that regulate various developmental programs and the defense responses of plants when faced with biotic and abiotic aggressions. Particular emphasis is placed on genetic evidence supporting the involvement of polyamines in specific processes, such as the pivotal role of thermospermine in regulating xylem cell differentiation and the significant contribution of polyamine metabolism in enhancing plant resilience to drought. Based on the numerous studies describing effects of the manipulation of plant polyamine levels, two conceptually different mechanisms for polyamine activity are discussed: direct participation of polyamines in translational regulation and the indirect production of hydrogen peroxide as a defensive mechanism against pathogens. By describing the multifaceted functions of polyamines, this review underscores the profound significance of these compounds in enabling plants to adapt and thrive in challenging environments. Abstract:Spermine, spermidine and putrescine polyamines are naturally occurring ubiquitous positively charged amines and are essential metabolites for biological functions in our life. These compounds play a crucial role in many cell processes, including cellular proliferation, growth, and differentiation. Intracellular levels of polyamines depend on their biosynthesis, transport and degradation. Polyamine levels are high in cancer cells, which leads to the promotion of tumor growth, invasion and metastasis. Targeting polyamine metabolism as an anticancer strategy is considerably rational. Due to compensatory mechanisms, a single strategy does not achieve satisfactory clinical effects when using a single agent. Combination regimens are more clinically promising for cancer chemoprevention because they work synergistically with causing little or no adverse effects due to each individual agent being used at lower doses. Moreover, bioactive substances have advantages over single chemical agents because they can affect multiple targets. In this review, we discuss anticancer strategies targeting polyamine metabolism and describe how combination treatments and effective natural active ingredients are promising therapies. The existing research suggests that polyamine metabolic enzymes are important therapeutic targets and that combination therapies can be more effective than monotherapies based on polyamine depletion.

Abstract:Polyamines play an important role in plant response to abiotic stress. S-adenosyl-l-methionine decarboxylase (SAMDC) is one of the key regulatory enzymes in the biosynthesis of polyamines. In order to better understand the effect of regulation of polyamine biosynthesis on the tolerance of high-temperature stress in tomato, SAMDC cDNA isolated from Saccharomyces cerevisiae was introduced into tomato genome by means of Agrobacterium tumefaciens through leaf disc transformation. Transgene and expression was confirmed by Southern and Northern blot analyses, respectively. Transgenic plants expressing yeast SAMDC produced 1.7- to 2.4-fold higher levels of spermidine and spermine than wild-type plants under high temperature stress, and enhanced antioxidant enzyme activity and the protection of membrane lipid peroxidation was also observed. This subsequently improved the efficiency of CO2 assimilation and protected the plants from high temperature stress, which indicated that the transgenic tomato presented an enhanced tolerance to high temperature stress (38 degrees C) compared with wild-type plants. Our results demonstrated clearly that increasing polyamine biosynthesis in plants may be a means of creating high temperature-tolerant germplasm.

Abstract:Polyamine biosynthesis is frequently dysregulated in cancers, and enhanced flux increases intracellular polyamines necessary for promoting cell growth, proliferation, and function. Polyamine depletion strategies demonstrate efficacy in reducing tumor growth and increasing survival in animal models of cancer; however, mechanistically, the cell-intrinsic and cell-extrinsic alterations within the tumor microenvironment underlying positive treatment outcomes are not well understood. Recently, investigators have demonstrated that co-targeting polyamine biosynthesis and transport alters the immune landscape. Although the polyamine synthesis-targeting drug 2-difluoromethylornithine (DFMO) is well tolerated in humans and is FDA-approved for African trypanosomiasis, its clinical benefit in treating established cancers has not yet been fully realized; however, combination therapies targeting compensatory mechanisms have shown tolerability and efficacy in animal models and are currently being tested in clinical trials. As demonstrated in pre-clinical models, polyamine blocking therapy (PBT) reduces immunosuppression in the tumor microenvironment and enhances the therapeutic efficacy of immune checkpoint blockade (ICB). Thus, DFMO may sensitize tumors to other therapeutics, including immunotherapies and chemotherapies.

Abstract:A precise apprehension of natural products and their role in the growth and protection of plants is the key to predicate the development of novel products for enhanced plant production, both in vivo as well as in vitro. Over the years, polyamines (PAs) constitute the prime members of ubiquitously appearing natural plant products, tangled with a variety of plant functions ranging from inducing organogenesis to mitigating stress. Various pathways for PA biosynthesis have been elucidated in plants, animals, and micro-organisms stipulating the presence of some PA representatives in both eukaryotes and prokaryotes systems. PAs are low molecular weight diamines that mediate the basal metabolism of plants by regulating various cellular processes including molecular signaling, cell division and differentiation, totipotency, oxidative damage, and various stress responses. Conjugation of PAs with other small molecules like phenolic acids and other cellular components often leads to the accumulation of incongruous interaction between plants and pathogens. PAs thwarts the various plant stress responses by diminishing the toxicity symptoms, modulating the stomatal closure and antioxidant capacity, increasing the germination efficiency, imputing tolerance against the bacterial pathogens, and reducing the viral multiplication. This study explored and summarized the significance of the correlation between different PAs and diverse physiological parameters in plants and supplemented the functioning of PAs under harsh environmental conditions in plants.

Abstract:Trichosanthin (TCS) from Trichosanthes kirilowii Maximowicz (T. kirilowii) can be used to treat choriocarcinoma. In this work, we established a novel system to produce TCS in crown gall tissues of T. kirilowii infected by Agrobacterium tumefaciens C58 (A. tumefaciens). In the crown gall tissues, a nopaline synthase (NOS) gene of A. tumefaciens was identified by polymerase chain reaction (PCR), and nopaline accumulation was confirmed by a high-voltage filter paper electrophoresis. Furthermore, we optimized conditions to culture the crown gall tissues able to grow fast and produce TCS in an auxin-free medium, and found that a fungal elicitor of Armillaria mellea was capable of stimulation of TCS secretion into the medium. Moreover, we identified that the TCS purified from the crown gall tissues could induce gastric cancer cell death. These data underscore the usefulness of our system as an inexpensive and virtually unlimited source of TCS.
Unregulated GmAGL82 due to Phosphorus Deficiency Positively Regulates Root Nodule Growth in Soybean

Abstract:Nitrogen fixation, occurring through the symbiotic relationship between legumes and rhizobia in root nodules, is crucial in sustainable agriculture. Nodulation and soybean production are influenced by low levels of phosphorus stress. In this study, we discovered a MADS transcription factor, GmAGL82, which is preferentially expressed in nodules and displays significantly increased expression under conditions of phosphate (Pi) deficiency. The overexpression of GmAGL82 in composite transgenic plants resulted in an increased number of nodules, higher fresh weight, and enhanced soluble Pi concentration, which subsequently increased the nitrogen content, phosphorus content, and overall growth of soybean plants. Additionally, transcriptome analysis revealed that the overexpression of GmAGL82 significantly upregulated the expression of genes associated with nodule growth, such as GmENOD100, GmHSP17.1, GmHSP17.9, GmSPX5, and GmPIN9d. Based on these findings, we concluded that GmAGL82 likely participates in the phosphorus signaling pathway and positively regulates nodulation in soybeans. The findings of this research may lay the theoretical groundwork for further studies and candidate gene resources for the genetic improvement of nutrient-efficient soybean varieties in acidic soils.

Abstract:Since the era of plant tissue culture bloomed, we have started approaching from a biotechnological perspective to overcome the massive challenges like inducing embryogenesis and organogenesis, initiating rooting, increasing the number of plantlets, establishing a callus from various organs of plants and also enhancing the metabolite content which was a mind-boggling thought once upon a time. Use of various elicitors, altering the media components, the strength of media, pH, precursor feeding etc. have all contributed tremendously in the in-vitro techniques used for culturing rare, endemic and medicinal plants for the commercial purposes. Owing to the demand for the plant products and drugs, the search for the other superior novel methods to increase its quantity and quality has not been stopped. Thus, one such method is the use of chemical compounds with many amino groups which serves as an additional source of nitrogen in the media and these organic compounds are called polyamines. Polyamines are known to play a wide role in plant physiological processes helping them in differentiation, inducing totipotency, increasing cell division and also in molecular signaling. Polyamines have a versatile application in this field ranging from establishing a callus to the elicitation of secondary metabolites. Thus, polyamines can be considered as a boon to the plant tissue culture field. In this review article, we have mainly focused on the in-depth applications of major polyamines like putrescine, spermidine and spermine in the field of plant tissue culture.

Role of polyamines in plant growth regulation of Rht wheat mutants

Abstract:Besides their protective role, polyamines also serve as signalling molecules. However, further studies are needed to elucidate the polyamine signalling pathways, especially to identify polyamine-regulated mechanisms and their connections with other regulatory molecules. Reduced height (Rht) genes in wheat are often used in breeding programs to increase harvest index. Some of these genes are encoding DELLA proteins playing role in gibberellic acid signalling. The aim of the present paper was to reveal how the mutations in Rht gene modify the polyamine-regulated processes in wheat. Wild type and two Rht mutant genotypes (Rht 1: semi-dwarf; Rht 3: dwarf mutants) were treated with polyamines. Polyamine treatments differently influenced the polyamine metabolism, the plant growth parameters and certain hormone levels (salicylic acid and abscisic acid) in these genotypes. The observed distinct metabolism of Rht 3 may more likely reflect more intensive polyamine exodus from putrescine to spermidine and spermine, and the catabolism of the higher polyamines. The lower root to shoot translocation of putrescine can contribute to the regulation of polyamine pool, which in turn may be responsible for the observed lack of growth inhibition in Rht 3 after spermidine and spermine treatments. Lower accumulation of salicylic acid and abscisic acid, plant hormones usually linked with growth inhibition, in leaves may also be responsible for the diminished negative effect of higher polyamines on the shoot growth parameters observed in Rht 3. These results provide an insight into the role of polyamines in plant growth regulation based on the investigation of gibberellin-insensitive Rht mutants.

Abstract:The biogenic aliphatic polyamines (spermine, spermidine, and putrescine) are responsible for numerous cell functions, including cell proliferation, the stabilization of nucleic acid conformations, cell division, homeostasis, gene expression, and protein synthesis in living organisms. The change of polyamine concentrations in the urine or blood is usually related to the presence of malignant tumors and is regarded as a biomarker for the early diagnosis of cancer. Therefore, the detection of polyamine levels in physiological fluids can provide valuable information in terms of cancer diagnosis and in monitoring therapeutic effects. In this review, we summarize the recent advances in fluorescent methods for polyamine detection (supramolecular fluorescent sensing systems, fluorescent probes based on the chromophore reaction, fluorescent small molecules, and fluorescent nanoparticles). In addition, tumor polyamine-suppressing strategies (such as polyamine conjugate, polyamine analogs, combinations that target multiple components, spermine-responsive supramolecular chemotherapy, a combination of polyamine consumption and photodynamic therapy, etc.) are highlighted. We hope that this review promotes the development of more efficient polyamine detection methods and provides a comprehensive understanding of polyamine-based tumor suppressor strategies.
Comparative study on the effects of putrescine and spermidine pre-treatment on cadmium stress in wheat

Abstract:In several cases a correlation was found between polyamines and abiotic stress tolerance. However, the individual polyamines may have different effects, which also vary depending on the type of treatment. When applied as seed soaking or added hydroponically 0.5mM putrescine and spermidine, different changes were induced during 50µM cadmium stress in wheat plants. Seed-soaked plants were exposed to cadmium immediately after germination for 5 days, while plants pre-treated with polyamines hydroponically were stressed at age of 14 days for 7 days. Putrescine pre-treatment was beneficial both as seed soaking and applied hydroponically, while spermidine only had a protective effect in the case of seed soaking, enhancing the Cd-induced oxidative stress when were pre-treated hydroponically. The differences observed were related to the polyamine metabolism. The accumulation of endogenous putrescine beyond a certain amount may be in relation with the negative effect of hydroponic spermidine pre-treatment during Cd stress. The increased putrescine content was also correlated with the highest accumulation of Cd, salicylic acid and proline contents in plants treated with a combination of spermidine and Cd. However, the expression level of the gene encoding phytochelatin synthase was only influenced by hydroponically applied spermidine, which decreased it under cadmium stress. Changes in the activities of antioxidant enzymes, diamine and polyamine oxidases were also discussed.

Liquid-liquid coexistence in NaCl aqueous solutions: a simulation study of concentration effects.

Abstract:In this paper we investigate by means of molecular dynamics computer simulations how the hypothesized liquid-liquid critical point of water shifts in supercooled aqueous solutions of salt as a function of concentration. We study sodium chloride solutions in TIP4P water, NaCl(aq), for concentrations c = 1.36 mol/kg and c = 2.10 mol/kg. The liquid-liquid critical point is found up to the highest concentration investigated, and its position in the P-T plane shifts to higher temperatures and lower pressures upon increasing concentration. For c = 2.10 mol/kg it is also located very close to the temperature of maximum density line of the system. The results are discussed and compared with previous results for bulk TIP4P water and for c = 0.67 mol/kg NaCl(aq) and with experimental findings. We observe a progressive shrinkage of the low-density liquid region when the concentration of salt increases; this suggests an eventual disappearance of the liquid-liquid coexistence upon further increase of NaCl concentration.

Abstract:Polyamine(s) (PA, PAs), a sort of N-containing and polycationic compound synthesized in almost all organisms, has been recently paid considerable attention due to its multifarious actions in the potent modulation of plant growth, development, and response to abiotic/biotic stresses. PAs in cells/tissues occur mainly in free or (non- or) conjugated forms by binding to various molecules including DNA/RNA, proteins, and (membrane-)phospholipids, thus regulating diverse molecular and cellular processes as shown mostly in animals. Although many studies have reported that an increase in internal PA may be beneficial to plant growth under abiotic conditions, leading to a suggestion of improving plant stress adaption by the elevation of endogenous PA via supply or molecular engineering of its biosynthesis, such achievements focus mainly on PA homeostasis/metabolism rather than PA-mediated molecular/cellular signaling cascades. In this study, to advance our understanding of PA biological actions important for plant stress acclimation, we gathered some significant research data to succinctly describe and discuss, in general, PA synthesis/catabolism, as well as PA as an internal ameliorator to regulate stress adaptions. Particularly, for the recently uncovered phenomenon of urea-antagonized NH 4 + -stress, from a molecular and physiological perspective, we rationally proposed the possibility of the existence of PA-facilitated signal transduction pathways in plant tolerance to NH 4 + -stress. This may be a more interesting issue for in-depth understanding of PA-involved growth acclimation to miscellaneous stresses in future studies.
