Abstract:Background: The sterile insect technique (SIT) has proven to be an effective approach in managing the population of major invasive pests. Our previous studies showed that irradiation of Cydia pomonella males at a dosage of 366 Gy X-rays resulted in complete sterility. However, the mating competitiveness of sterilized males is significantly compromised, which can be attributed to a decline in their ability to fly. Results: In this study, we examined the flight patterns of both male and female adults of C. pomonella. The results revealed significant variations in the average flight speed of both genders at different stages of maturity, with females displaying longer flight duration and covering greater distances. Effect of irradiation on the flight performance of 3-day-old male moths was further evaluated, as they demonstrated the longest flight distance. The findings indicated a significant decrease in flight distance, duration, and average speed, due to wing deformities caused by irradiation, which also limited the dispersal distance of moths in orchards, as indicated by the mark-and-recapture assay. Reverse-transcription quantitative polymerase chain reaction analysis revealed a down-regulation of flight-related genes such as Flightin, myosin heavy chain, and Distal-less following radiation exposure. Conclusion: These findings demonstrate that X-ray irradiation at a radiation dose of 366 Gy has a detrimental effect on the flight ability of male C. pomonella adults. These insights not only contribute to a better understanding of how radiation sterilization diminishes the mating competitiveness of male moths, but also aid in the development and improvement of SIT practices for the effective control of C. pomonella. © 2023 Society of Chemical Industry.
Linalool fumigation improves mating competitiveness of males for population suppression of the global fruit pest Cydia pomonella

Abstract:BACKGROUND: The implementation of sterile insect technique (SIT) has proven effective in the area-wide suppression of several significant agricultural and sanitary pests by employing traditional cobalt-60 (60 Co-γ) as a radiation source. Recently, X-ray has been validated as a feasible alternative to 60 Co-γ radiation sources. Nonetheless, higher doses of X-ray irradiation lead to insect sterility but diminish mating competitiveness, thereby impacting the effectiveness of SIT applications. Thus, it is crucial to ascertain the optimal irradiation dose and develop strategies to enhance the mating competitiveness of sterile insects to enhance SIT efficacy. RESULTS: In this study, we determined the effect of various X-ray irradiation doses (ranging from 0 to 366 Gy) on the fecundity, fertility, and mating competitiveness of Cydia pomonella, a globally invasive fruit pest. Results demonstrated that the sterility rate of sterile males increased proportionally with irradiation dose up to 200 Gy, beyond which it plateaued. Notably, exposure to 200 Gy of irradiation notably decreased the mating competitiveness of male, as evidenced by a mating competitiveness index of 0.17 in laboratory and 0.096 in the orchard. This decline in mating competitiveness is likely linked to the down-regulation of genes associated with the recognition of sex pheromones, specifically CpomOR3a, CpomOR3b, and CpomOR5, following X-ray irradiation. Fumigation of the plant volatile, linalool at varying concentrations (70, 83, and 96 μL/m3) resulted in differential enhancements in male mating competitiveness, with the moderate concentration significantly improving the competitiveness of sterilized males, possibly by restoring their ability to recognize sex pheromones. Implementation of repeated releases of sterilized males on a pilot scale led to a notable reduction in the population of C. pomonella in the field. CONCLUSION: These findings indicate that fumigation with plant volatiles has the potential to mitigate male sterility induced by X-ray irradiation, offering a promising approach to enhance the efficacy of SIT applications for the control of C. pomonella.

Experimental autoimmune thyroiditis: modulation of the disease level in high and low responder mice by thymosin.

Abstract:Experimental autoimmune thyroiditis (EAT) in mice is characterized by production of anti-thyroglobulin autoantibodies and lymphoid infiltration of the thyroid gland. The pathogenesis of EAT is genetically controlled, but its mechanism is not yet clear. To investigate the mode of the expression of genetic control in the development of the disease, we attempted to modulate immune responses with thymosin fraction 5 (T-5) in mice which are either high or low responders to thyroglobulin. Severe thyroid lesions in high responder mice appear 2-3 weeks after immunization, while low responders develop very mild or no lesions. High responder mice have also higher antibody titer than the low responder strain. T-5 administered in five daily injections before or simultaneously with immunization, was strongly suppressive to EAT development in the high responder strain while there was no effect on thyroiditis level in low responder mice. T-5 also decreased the severity of thyroiditis in high responder mice when injected 2-4 weeks after immunization. In low responder mice, however, the same treatment increased the thyroiditis level. The effects of late injection of T-5 were dose-dependent. In all experiments, the antibody titres were not affected by T-5 treatment. The results demonstrate the ability of T-5 to modulate development and intensity of EAT. The contrasting effects of T-5 in high and low responder mice illustrate the relationship between the effects of T-5 and host immune status prior to study, suggesting that the level of immunoregulatory cell activity may differ in these two strains.

Abstract:The sterile insect technique (SIT) is widely used to control Lepidopteran pests by inducing inherited sterility. The noctuid moth Xestia c-nigrum is a polyphagous pest whose subterranean larvae severely injure cereals and some vegetables. The goals of this study were to assess the impact of X-ray irradiation on the development and survival of X. c-nigrum and use the data to select suitable sterilizing doses for potential future use in pest management. Batches of male pupae were exposed to 0 (control), 10, 30, 50, 100, 200, 300, or 400 Gy of X-rays, approximately 24 h before adult emergence. Exposure of late-stage pupae to 10–200 Gy of radiation had no significant effect on adult emergence, but all doses (10–400 Gy) reduced adult longevity, the number of spermatophores in mated females, and the number of eggs laid per female in the irradiated parental generation compared with the controls. Exposure to 10 and 30 Gy had no significant effects in the F1 generation on 1) the rate of egg hatch, 2) the duration of larval or pupal development, or 3) adult longevity. However, exposure to 50 Gy reduced the rate of egg hatch in the F1 generation, and when male pupae were exposed to 100 Gy only 1% of the F1 eggs hatched. Also at 100 Gy, the developmental durations of larvae and pupae were significantly prolonged, and longevity of adult moths was reduced. There were no significant differences between the control group and any treatments in 1) the sex ratio of the F1 adults, 2) the duration of F1 pre-oviposition or oviposition periods, or 3) the number of eggs laid per F1 female. Our findings indicate that a dose of 100 Gy can effectively slow pest development and reduce larval survival in the F1 generation. In addition, F1 adults from lines treated with 100 Gy were able to mate and lay eggs, but all F2 eggs failed to hatch. Our results suggest that use of X-ray irradiation has potential to control this polyphagous pest at the regional level.

Abstract:The sterile insect technique (SIT) is widely used for the inundative release of sterile mass-reared males to control lepidopteran pests. SIT based on X-ray irradiation is an eco-friendly alternative to chemical control. However, its use in Ephestia elutella , a stored tobacco pest currently controlled with insecticides, is poorly explored. This study aims to investigate the effects of X-ray irradiation on E. elutella to determine the optimal sterilizing dose and processing developmental stage for improving SIT application. The pupal stage was most suitable for irradiation that was more tolerant than the other insect stages including eggs, larvae, and adults. Subsequently, male pupae were irradiated with X-ray doses of 0, 50, 100, 150, 200, 250, and 300 Gy and mated with unirradiated females. Their emergence, longevity, egg number, egg hatch rate, developmental duration, survival rate, induced sterility, and male mating competitiveness were evaluated. The results suggest that a dose of 200 Gy can be applied to effectively induce sterility in male pupae, after which induced sterility and male mating competitiveness can be balanced by increasing the release ratio (sterile:normal). When the release ratio was 15:1, it was found that 71.91% of the wild population could be suppressed. The results of this study show that the SIT based on X-ray irradiation can be successfully used to manage E. elutella, improves our understanding of the biological effects of the SIT, and expands its future application to the control of other pests.

Abstract:Gamma radiation from isotopic sources has been used in sterile insect technique (SIT) programs worldwide, but it might be difficult to continue using these sources in future SIT programs because of social issues. Therefore, an alternative sterilization source to gamma rays, such as X-rays, needs to be developed. The physical properties of radiation are different between gamma rays and X-rays: for example, X-rays have a shorter penetration depth than gamma rays. Therefore, X-rays may not fully confer male sterility, depending on the target pest insects. The present study investigated whether the West-Indian sweetpotato weevil Euscepes postfasciatus (Fairmaire) and the sweetpotato weevil Cylas formicarius (Fabricius) are sterilized by X-rays generated in a low-energy X-ray irradiator, without deterioration of male mating ability, at the doses currently used in the eradication programs for E. postfasciatus (150 Gy) and C. formicarius (200 Gy) using gamma rays at Okinawa, Japan. The results demonstrated that it is possible to use X-rays in future SIT programs for E. postfasciatus and C. formicarius, because X-ray irradiated males were almost completely sterilized without deterioration of their mating ability.

Abstract:Spodoptera litura is an omnivorous pest that has spread globally. Because irradiation sterilization technology has a great potential for control of S. litura , the effect of 25–150 Gy doses of X-rays on pupal survival, flight and reproductive variables of adult moths were analyzed in this research. The X-ray irradiation with the dose of 25–150 Gy significantly affected the reproductive ability of females. Irradiating male pupae with 25–150 Gy doses of X-rays had no effect on mating, life span, or flight ability of adult moths, but significantly reduced survival and fecundity of their offspring, and the sterility rate of the F 1 generation was 52.65%–99.9%. The results of logistic curve fitting showed that the sterility impact was 84% at the most appropriate irradiation dose (71.26 Gy). The sterility control was 91% in an indoor mating competition experiment when the release ratio of irradiated males (75 Gy) to nonirradiated males reached 12.6:1. The effects of X-ray irradiation doses on biological variables of S. litura and the most effective release ratio determined here provide a theoretical foundation for using radiation sterilization technology to control S. litura .

Abstract:The sterile insect technique (SIT) is considered an environmentally friendly, autocidal control tactic to manage insect pests. This work dealt with the improvement of quality management of the Indian meal moth Plodia interpunctella (Hübner) for enhanced efficacy of the SIT. The results indicated that egg hatching of irradiated mature eggs of P. interpunctella was higher than that of younger eggs, indicating that mature eggs were significantly more tolerant than younger eggs. Moreover, our data revealed that a dose of 500 Gy completely prevented pupal formation in irradiated young and mature larvae. Crosses between irradiated and non-irradiated adults resulted in considerable variations in fecundity. The mating competitiveness index (CI) value was higher for a ratio of 5:1:1 (sterile male, fertile male, and fertile female, respectively) as compared with the ratio 1:1:1 for the irradiated individuals of all life stages. Low temperature (5 °C) maintenance of irradiated pupae significantly affected adult emergence. Using cylinders to assess flight ability, we found that the flight performance of adults that were developed from cold treated irradiated pupae was influenced by cylinder diameter, cylinder height and the number of hours the insects were in the cylinders. The percentage of DNA damage of the reproductive organs of adults developed from cold treated pupae that were irradiated with 100 and 150 Gy varied significantly. The results of this study should be used to implement pilot-scale field tests aiming at a sterile- to-fertile male ratio of 5 to 1.

Development, Reproduction and Sexual Competitiveness Ofconopomorpha Sinensis(lepidoptera: Gracillariidae) Gamma-Irradiated As Pupae and Adults

Abstract:Male and female litchi stem-end borers, Conopomorpha sinensis (Lepidoptera: Gracillariidae) were γ-irradiated in a 60Co source during several periods of their pupal development and as newly emerged adults. When mature pupae, the most suitable stage for irradiation, received increasing γ-radiation doses, the emergence rate, flight ability and adult longevity were increasingly diminished. Females that emerged from mature pupae irradiated with 200, 250, and 300 Gy did not oviposit any eggs when mated with either non-irradiated males (UM) or treated males (TM), indicating that 200 Gy was a sufficient dose for inducing complete sterility in females. The hatch rates of eggs oviposited by non-irradiated females (UF) mated with males irradiated with either 200, 250 or 300 Gy were 31, 13.5 and 0.67%, respectively. However, if the father—parental generation (P)—of the male in the cross, UF × F1 M, had been irradiated with either 200, 250 or 300 Gy, then the percent hatch of the eggs produced was either 9.37 ± 1.68, 0 or 0%, respectively; which indicated that 250 and 300 Gy applied to P generation males resulted in complete sterility in F1 males. The sexual competitiveness (C) of male litchi stem-end borers that had been irradiated with 250 Gy ranged between 0.53 and 0.62 as measured by ratio tests in laboratory cages with UF:UM:TM ratios of 1:1:1, 1:1:3 and 1:1:5. In a field cage experiment, a mean C value of 0.48 was observed when males were treated with 250 Gy. The C values obtained suggest that males treated with 250 Gy could compete adequately with wild litchi stem-end borers both under laboratory and field cage conditions to warrant pilot scale field tests.
Low‐oxygen hormetic conditioning improves field performance of sterile insects by inducing beneficial plasticity

Abstract:<p>As part of Sterile Insect Technique (SIT) programs, irradiation can effectively induce sterility in insects by damaging germline genomic DNA. However, irradiation also induces other off‐target side effects that reduce the quality and performance of sterilized males, including the formation of damaging free radicals that can reduce sterile male performance. Thus, treatments that reduce off‐target effects of irradiation on male performance while maintaining sterility can improve the feasibility and economy of SIT programs. We previously found that inducing a form of rapid, beneficial plasticity with a one‐hour anoxic conditioning period (physiological conditioning hormesis) prior to and during irradiation improves male field performance in the laboratory while maintaining sterility in males of the cactus moth, <i>Cactoblastis cactorum</i>. Here we extend this work by testing the extent to which this beneficial plasticity may improve male field performance and longevity in the field. Based on capture rates after a series of mark release‐recapture experiments, we found that anoxia‐conditioned irradiated moths were active in the field longer than their irradiated counterparts. In addition, anoxia‐conditioned moths were captured in traps that were farther away from the release site than unconditioned moths, suggesting greater dispersal. These data confirmed that beneficial plasticity induced by anoxia hormesis prior to irradiation led to lower post‐irradiation damage and increased flight performance and recapture duration under field conditions. We recommend greater consideration of beneficial plasticity responses in biological control programs, and specifically the implementation of anoxia‐conditioning treatments applied prior to irradiation in area‐wide integrated pest management programs that use SIT.</p>

Abstract:The mosquito Aedes albopictus can transmit various arboviral diseases, posing a severe threat to human health. As an environmentally friendly method, sterile insect technology (SIT) is considered an alternative to traditional methods such as chemical pesticides to control Ae. albopictus. In SIT, the sterility of male mosquitoes can be achieved by γ-ray or X-ray radiation. Compared with γ-rays, X-rays are easier to obtain, cheaper, and less harmful. However, there is a lack of comparative assessment of these two types of radiation for SIT under the same controlled conditions. Here, we compared the effects of X-ray and γ-ray radiation on the sterility of Ae. albopictus males under laboratory-controlled conditions. Neither type of radiation affected the number of eggs but significantly reduced the survival time and hatch rate. The same dose of γ-rays caused a higher sterility effect on males than X-rays but had a more significant impact on survival. However, X-rays could achieve the same sterility effect as γ-rays by increasing the radiation dose. For example, X-rays of 60 Gy induced 99% sterility, similar to γ-rays of 40 Gy. In the test of male mating competitiveness, the induced sterility and the male mating competitiveness index were also identical at the same release ratio (sterile males/fertile males). At a release ratio of 7:1, nearly 80% of eggs failed to hatch. Sterile males produced by X-ray and γ-ray radiation had similar male competitiveness in competition with field males. In conclusion, a higher dose of X-rays is required to achieve the same sterility effect, compared to γ-rays. When γ-rays are not readily available, high-dose X-rays can be used instead. This study provides data supporting the selection of more suitable radiation for the field release of sterile male mosquitoes.

Abstract:The Sterile Insect Technique (SIT) is a target-specific, biologically based method used to control pestiferous tephritids entailing the release of mass-reared, sterilized males of the target species to achieve sterile male x wild female matings. As documented for several tephritid species, sterilizing irradiation may have adverse effects on various biological parameters, including life span, flight ability, and mating competitiveness. To minimize these impacts and ensure sterility, released flies must be irradiated at a precise dose at a specific and uniform age. The objective of the present study was to determine the impact of pupal age at the time of irradiation on flight ability, male survival, fertility, and mating competitiveness for the oriental fruit fly, Bactrocera dorsalis (Hendel) (Diptera: Tephritidae) irradiated at 100 Gy. We concluded that flies irradiated as pupae 4 days before emergence were significantly compromised with respect to flight ability and male survival when compared to flies irradiated 1 or 2 days before emergence. Additionally, we determined that crosses between males irradiated 1 day before emergence with non-irradiated females yielded significantly more pupae than the same cross with males irradiated at 2, 3, and 4 days before emergence. Although rare, the occurrence of pupae and emerged adults suggested low levels of fertility for flies irradiated as pupae 1 day before emergence, but complete sterility at 2, 3, and 4 days before emergence. These findings suggest that SIT programs can potentially irradiate pupae at 2 or 3 days before emergence without compromising the quality of the released flies, which would permit the distribution of flies over greater distances.

Abstract:BACKGROUND The pepper weevil (PW), Anthonomus eugenii, is an economically significant pest of cultivated Capsicum spp. pepper crops in North America where it remains a challenge to manage because of its cryptic immature life stages. The sterile insect technique (SIT) is a genetic pest management tactic that relies on the release of insects that have been sterilized with ionizing radiation to lower the population reproductive rate. Toward developing an effective PW-SIT program, this study has, for the first time, investigated the effects of gamma irradiation on the sterility and survival of this species. RESULTS Among the array of doses tested, we found that pupal PW males and females irradiated at 110 Gy produced 0 adult offspring. Furthermore, females mated with a male irradiated at 110 Gy had high egg sterility (97.3%), and irradiated females nearly completely failed to lay eggs (97.5%). Individuals irradiated at this dose had a shortened lifespan (LT50s of 12 and 11 days for males and females, respectively) and quantitatively reduced spontaneous flight activity. The eclosion rate of PW pupae was not significantly reduced by any radiation treatment. CONCLUSION This study suggests that PWs irradiated at a gamma radiation dose of 110 Gy as pupae could feasibly be used in PW-SIT program, as both males and females were 100% sterile at this dose. These findings will inform the development of a SIT program that could considerably improve the sustainability and effectiveness of PW management in greenhouse and field pepper crops worldwide. Abstract:Abstract The sterile insect technique is a new approach for the integrated management of Drosophila suzukii Matsumura (Diptera: Drosophilidae), an invasive pest in North America. We evaluated, under laboratory conditions, the mating capacities and success of male D. suzukii , irradiated at a dose of 120 Gy, with and without competition. We also explored the tendency of females to remate depending if their first mate was irradiated or not. We observed that irradiated males have the same mating capacity as control males, copulating with, respectively, 6.4 ± 1.9 females versus 6.9 ± 2.0 females in a 24-hour period. Irradiated males won the competition 37.5% of times, which is not significantly different from competiveness of control males. Female remating can be considered infrequent and not significantly influenced by male treatment: 7.4% of the females first mated with control males and 18.8% of the females first mated with irradiated males remated when given the opportunity two days and four days after the first mating. Latency before mating and mating duration were not significantly influenced by male treatment, but by presence of male competition. Overall, irradiated males thus seem to be as performant as control males, which is an important condition for a successful sterile insect technique programme.
Effect of mass rearing and gamma irradiation on the mating behaviour of Eldana saccharina

Abstract:At low doses of radiation, F1 sterility of the sugarcane stalk borer, Eldana saccharina Walker (Lepidoptera: Pyralidae), can be attained. This bodes well for progression of the sterile insect technique (SIT) as part of an area‐wide integrated pest management strategy against this pest of economic importance. However, success of the SIT is dependent on mating ability and performance of the mass‐reared sterile insects once released into the field. Mating behaviour of non‐irradiated laboratory‐reared male E. saccharina moths and those subjected to radiation (150, 200, or 250 Gy) was investigated in still‐air cage laboratory trials. Video technology was used to determine whether mass rearing and irradiation of adult moths altered their performance in lek formation and mating success, compared to their untreated wild counterparts. Mating frequency, mating duration, onset of mating, and calling during the first two scotophase periods following adult eclosion were used to make these assessments. Results indicated that both irradiated and non‐irradiated laboratory‐reared males are able to form leks and mate with wild females, and are therefore as effective as their wild counterparts with regards to mating performance. However, a quantitative departure of male mating behaviour away from that exhibited by wild males was observed in the non‐irradiated and irradiated laboratory males. These commenced calling and mating earlier than wild males in both scotophases, except for those irradiated at 250 Gy in the case of mating onset. The mating duration of treated males was similar to that of the untreated wild males, except for those irradiated at 150 Gy, which was significantly higher. Our study provides evidence that increasing radiation dose negatively impacts on performance of laboratory‐produced moths relative to their wild counterparts, which in turn could affect the efficacy of the SIT. At 150 and 200 Gy, these parameters are not affected significantly when compared to untreated wild males, and thus these radiation doses can be used safely. However, at higher doses negative effects do become evident.

Abstract:The research relevance on Codling moth peculiarities is predefined by it being a main threat to apple culture. The sensitivity of apple cultivars that have been planted so far and continue to be planted to this pest diverges in different cultivars. The research aims to determine the main elements of Cydia Pomonella control using sex pheromones and environmentally friendly insecticides. To achieve the goal, an experiment was conducted in the village of Dvoran in the Korcha region on three apple cultivars: Golden Delicious, Star King, and Granny Smith. On the trees of these varieties, traps with sex pheromones were used and environmentally friendly insecticides were utilised. The study found that using sex pheromone traps to monitor Cydia Pomonella is simple and less expensive for apple growers. The Spinosad medicine is the most effective in terms of protection against the pest, the second being Indoxacarb. Golden Delicious and Granny Smith cultivars had the lowest level of pest infestation, while the Star King cultivar was the most affected by Cydia Pomonella. It has also been established that apple fruits do not contain toxic residues, and the ecosystem remains clean. Additionally, the use of sex pheromones can be part of an integrated pest management approach that combines different control methods to sustainably manage pest populations in gardens. The practical significance of the obtained results is that they provide apple growers with a safe and effective method of controlling Cydia pomonella populations in their orchards. In addition, the scientific basis for the use of sex pheromones as a method of pest control can contribute to the implementation of sustainable control methods in apple production
X-Ray-Based Irradiation of Larvae and Pupae of the Navel Orangeworm (Lepidoptera: Pyralidae)

Abstract:Abstract The suitability of adult male the navel orangeworm, Amyelois transitella (Walker) for Sterile Insect Technique (SIT) has been reported for both high energy gamma (>1 MeV) and low energy x-ray (90 keV) sterilization. However, research regarding sterilization of NOW larvae and pupae by gamma irradiation indicated nonsuitability due to high mortality. Here, NOW larvae and pupae were irradiated to doses up to 50 Gy with 90 keV x-rays, then paired with nonirradiated colony mates. Sterility of surviving insects was determined by the presence or absence of hatched neonates. While presence of offspring does not guarantee viability, the absence does guarantee sterility (as is appropriate for SIT) and was thus the measure used here. Early stage larvae experienced 77% mortality at a dose of 30 Gy, versus 20% for nonirradiated control. At 40 Gy, mortality reached 98%. Of surviving early stage larvae at 30 Gy, 29% of moth pairs produced offspring. For late stage larvae, no offspring were produced at 40 Gy, but mortality was 73%. For pupae, mortality reached 53% at 30 Gy with 13% still producing neonates, while mortality reached 98% at 40 Gy. These results are consistent with reported results for gamma irradiation of NOW larvae where sterility was observed somewhere between the 30 Gy and 60 Gy data points, but mortality was high. This further confirms the lack of suitability of NOW irradiated in the larval stage, whether by gamma or x-ray, and supports the hypothesis that x-ray and gamma treatments are biologically equivalent at equal doses.

Abstract:One of the most important components of a program that has a sterile insect technique (SIT) component is an appropriate irradiation dose. Knowing the organismal dose-response enables the selection of a dose that induces the highest level of sterility while preserving the sexual competitiveness and other desired qualities of the sterile insect. Finding this balance in Lepidoptera is crucial because of the use of inherited (F1) sterility, where the irradiated parent must be competitive enough to mate while its offspring must be sterile. Manipulations of atmospheric oxygen content have been shown to be an effective way of lowering post-irradiation somatic damage while preserving sterility and improving sterile insect performance, particularly in fruit flies. In this study we tested the irradiation dose response of adults of the cactus moth, Cactoblastis cactorum (Berg) (Lepidoptera: Pyralidae), and the effects of those doses on sterility, longevity, and F1 performance, and whether a nitrogen conditioning-treatment (anoxia) prior to and during irradiation affected these metrics. We found that male and female fecundities were not impacted by dose or atmospheric treatment, but females were sterilized at lower doses than males. Eggs of irradiated parents took longer to hatch than those of unirradiated controls, and offspring of moths irradiated in anoxia lived longer in the absence of food and water. Anoxia conditioning rescued female fertility at intermediate doses but had no similar rescue effect on male fertility, which was always greater than female fertility at a given dose. Males generally lived longer than females and anoxia had a strong effect in lowering the male mortality rate and extending lifespan at a given dose. We show evidence that anoxia-conditioning prior to and during irradiation as part of a lepidopteran program with an SIT component could improve parental and larval performance and longevity.

Abstract:Male adult navel orangeworm, Amyelois transitella (Walker), were irradiated using a laboratory scale x-ray irradiation unit to determine the required dose for complete egg sterility of mated female moths and inherited sterility of F1 and F2 generations. Adult male A. transitella were irradiated in two separate experiments at 100-300 Gy and 50-175 Gy. Mating frequency, fecundity, and fertility of normal females crossed with irradiated parental males was compared with the mating of nonirradiated moths. Mating frequency was 100% for females crossed with nonirradiated control males. At male treatment doses of ≥150 Gy the percentage of females found unmated increased, while multiple-mated females decreased. Female fecundity was not affected while fertility was affected in a dose-dependent relationship to exposure of parental males to x-ray irradiation. Embryonic development of eggs to the prehatch stage and egg eclosion did not occur at radiation doses ≥125 Gy. Emergence of F1 adults was low and occurred only for progeny of parental males exposed to doses ≤100 Gy, with no emergence at ≥125 Gy. Though fecundity appeared similar for control and irradiated F1 females, no F2 eggs hatched for the test exposures of 50-100 Gy. Based on our results, a dose of ≥125 Gy had efficacy in inducing both primary parental sterility in treated male moths and inherited sterility in F1 male and female moths. Results suggest that A. transitella might be considered a candidate for the sterile insect technique using adults irradiated at these relatively low x-ray exposure doses.
