Abstract:Biodegradable macroporous hydrogels were prepared using a biodegradable crosslinker based on chitosan derivatives. The chitosan crosslinker was synthesized through the amidating reaction of amino groups in the chitosan oligosaccharide (CSO) chains and carboxyl groups in acrylic acid (AA) under the catalysis of 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimidehydro-chloride (EDC) and N-hydroxysuccinimide (NHS). The amidating reaction is able to provide polymerizable carbon-carbon double bonds for CSO. The chemical structure of the resulting compound chitosan oligosaccharide-graft-acrylic acid (CSO-g-AA) was characterized by Fourier transform infrared spectroscopy (FTIR) and H-1 nuclear magnetic resonance (H-1 NMR) spectroscopy. The content of double bonds was measured by titration analysis. The measurement showed a grafting ratio of AA on CSO of about 3.5% (on average two acryoyl groups per CSO-g-AA chain) when the applied molar ratio of AA to CSO was 1.0. Polyacrylamide (PAM) cryogels were synthesized by the free radical polymerization of acrylamide using a modified freezing polymerization method crosslinked by CSO-g-AA. The obtained cryogels were macroporous hydrogels which could be degraded into solutions of linear polymer chains when treated with appropriate enzymes. Then we used snailase as a model enzyme to study the biodegradation process. The degradation was monitored by morphological studies using a confocal laser scanning microscope (CLSM) and scanning electron microscope (SEM), mechanical strength, and swelling ratio. The duration of the degradation process was adjustable from one month to two months when using different concentrations of snailase solutions.

Abstract:Comprehensive SummaryMacroporous hydrogels are water‐swollen polymer networks with porous structures beyond the mesh size. They provide high specific surface areas and hieratical mass transfer channels which are desired for emerging applications including cell culturing, bio‐separation, and drug delivery. A variety of approaches have been developed to fabricate macroporous hydrogels, including gas foaming, porogen templating, phase separation, 3D printing, etc. Alternatively, ice templating utilizes the crystallization of water as the porogenation mechanism which doesn't need the leaching of porogens. The porous structures can be easily manipulated by controlling the morphology of ice (size, orientation, etc.) upon freezing. In addition, mechanical properties of obtained porous hydrogels are commonly better than those of other macroporous hydrogels since the pore walls are made up of concentrated polymers through the freezing step. These three characteristics, that is, the green process, the highly tunable morphology, and the enhanced mechanical strength make the ice templating a superior porogenation method. This review expects to give an overview on macroporous hydrogels prepared by the ice templating, namely, cryogels. Recent progresses on emerging cryo‐porogenation methods are introduced. Fundamental understanding concerning the behavior of the polymer chains during the formation of ice is proposed. Future opportunities on the application of cryogels are prospected.

Abstract:Structured xylan-based hydrogels, reinforced with cellulose nanocrystals (CNCs), have successfully been prepared from water suspensions by cross-linking during freeze-casting. In order to induce cross-linking during the solidification/sublimation operation, xylan was first oxidized using sodium periodate to introduce dialdehydes. The oxidized xylan was then mixed with CNCs after which the suspension was frozen unidirectionally in order to control the ice crystal formation and by that the pore morphology of the material. Finally the ice crystal templates were removed by freeze-drying. During the freeze-casting process hemiacetal bonds are formed between the aldehyde groups and hydroxyl groups, either on other xylan molecules or on CNCs, which cross-links the system. The proposed cross-linking reaction was confirmed by using cross-polarization magic angle spinning (CP/MAS) nuclear magnetic resonance (NMR) spectroscopy. The pore morphology of the obtained materials was analyzed by scanning electron microscopy (SEM). The materials were also tested for compressive strength properties, both in dry and water swollen state. All together this study describes a novel combined freeze-casting/cross-linking process which enables fabrication of nanoreinforced biopolymer-based hydrogels with controlled porosity and 3-D architecture.
Hydrogels based on gelatin, xanthan gum, and cellulose obtained by reactive extrusion and thermopressing processes

Abstract:Polysaccharides and proteins are compatible macromolecules that can be used to obtain biopolymeric hydrogels through physical interactions. In this study, an environmentally friendly strategy is being proposed to produce gelatin-xanthan gum- cellulose hydrogels, without the addition of chemical synthetic crosslinkers. Xanthan gum was employed as an alternative crosslinking agent, and cellulose was used as a potential reinforcing agent in the polymeric matrix. Firstly, the biopolymers were mixed by the extrusion process, and glycerol was used as a plasticizer. Then, the polymeric mixture was molded by thermopressing to obtain hydrogels as laminated films. All hydrogels formulations resulted in films with smooth surfaces, without pores or cracks, resulting in amorphous polymeric matrices. The obtained hydrogels had a pH-dependent degree of swelling, the highest swelling values were obtained at pH 4 (5.3-7.9 g/g) after 24 h of immersion. Cellulose acted as a reinforcing agent for hydrogels, increasing thermal stability, tensile strength, and Young's modulus of films when employed at the higher level (7%). The strategy employed in this study to obtain nontoxic hydrogels without synthetic crosslinkers was effective, resulting in materials with promising properties to be used as pharmaceutical forms to deliver active compounds in cosmetic or pharmaceutical products.
Investigating the fabrication and functional properties of new composite hydrogels containing gellan/ alginate/ xanthan gum

Abstract:The objectives of this study were to create composite hydrogels from gellan, alginate, and xanthan gums and study their functional properties. The characteristics of gellan (1.25–0.05%), gellan-alginate (1: 0, 0.25: 0.75, 0.5: 0.5, 0.75: 0.25 and 0:1), and gellan (0.5%)-alginate(0.5%)-xanthan (0.1, 0.15 and 0.3%) hydrogels were investigated. The results showed the gellan hydrogel structure had dense mesh and small pore size compared to alginate gel. With the combination of gellan and alginate, the pore size of network and swelling index increased. Tri-polymeric hydrogels created a network with bigger pore size, which improved the swelling capacity of the hydrogel. But, the swelling index decreased with increasing xanthan gum concentration from 0.1% to 0.3% (w/w). The gellan-alginate gel had more hardness and consistency value compared to the mono-polymeric and tri-polymeric hydrogels. Thermal stability of hydrogels increased by the combination of gums and FTIR confirmed the interaction of the polymers. As a result, multi-polymeric hydrogels of alginate-gellan-xanthan can create new structures with more swelling ability, consistency, and thermal stability for use in the nutraceutical delivery and restructuring of foods.

Abstract:Hydrogel features can be designed and optimized using different crosslinking agents to meet specific requirements. In this regard, the present work investigates the physico-chemical features of cellulose-based hydrogels, designed by using different epoxy crosslinkers from the same glycidyl family, namely epichlorohydrin (ECH), 1,4-butanediol diglycidyl ether (BDDE), and trimethylolpropane triglycidyl ether (TMPTGE). The effect of the crosslinker's structure (from simple to branched) and functionality (mono-, bi- and tri-epoxy groups) on the hydrogels' features was studied. The performances of the hydrogels were investigated through the gel fraction, as well as by ATR-FTIR, DVS, SEM, DSC, and TG analyses. Also, the swelling and rheological behaviors of the hydrogels were examined. The advantages and limitations of each approach were discussed and a strong correlation between the crosslinker structure and the hydrogel properties was established. The formation of new ether bonds was evidenced by ATR-FTIR spectroscopy. It was emphasized that the pore size is directly influenced by the crosslinker type, namely, it decreases with the increasing number of epoxy groups from the crosslinker molecule, i.e., from 46 ± 11.1 µm (hydrogel CE, with ECH) to 12.3 ± 2.5 µm (hydrogel CB, with BDDE) and 6.7 ± 1.5 µm (hydrogel CT, with TMPTGE). The rheological behavior is consistent with the swelling data and hydrogel morphology, such as CE with the highest Qmax and the largest pore size being relatively more elastic than CB and CT. Instead, the denser matrices obtained by using crosslinkers with more complex structures have better thermal stability. The experimental results highlight the possibility of using a specific crosslinking agent, with a defined structure and functionality, in order to establish the main characteristics of hydrogels and, implicitly, to design them for a certain field of application.

Abstract:Natural polysaccharides are abundant, inexpensive, renewable, modifiable and have biodegradable and biocompatible characteristics. Polysaccharides offer a very promising source for materials of tomorrow. This review addresses recent progress in polysaccharide-based cryogels, one kind of novel physical hydrogels prepared by freeze–thaw technique under mild conditions and in the absence of organic solvents and toxically crosslinking agents. Polysaccharides used to fabricate physical cryogels here are, hyaluronan, carboxymethylated curdlan, carboxymethylated cellulose, xanthan, β-glucan, locust bean gum, starch (amylose, amylopectin and their mixtures), maltodextrins and agarose. Composite cryogels of based on polysaccharides and polyvinyl alcohol are also introduced. Various physically crosslinked cryogels from polysaccharides with tunable structural, mechanical, biological properties as well as multiple applications are considered and the investigations of the formation mechanism for these cryogels are also addressed.

Abstract:Cryogels offer a promising macroporous platform that can be employed as either a functional ingredient in food composites or a colloidal template for incorporating bioactives, including probiotic living cells. The aim of the present work is to explore the functionality of two plant seed polysaccharides, flaxseed gum (FG) and alfalfa galactomannan (AAG), in individual and combined (1:1 ratio) milk protein-based cryogels, namely sodium caseinate (NaCas) and whey protein isolate (WPI). These cryogels were created by freeze-drying hydrogels formed via L. rhamnosus GG – a human gut-relevant probiotic strain – fermentation. Our findings showed that including gum in the composition limited volume contraction during lyophilisation, reduced macropore size and thickened cryogel skeleton vessels. Furthermore, gum-containing cryogels displayed improved thermal stability and slower water disintegration rates. The AAG-stabilised cryogels specifically showed a notable reduction in monolayer water content compared to FG. From a mechanistic viewpoint, AAG influenced the physicochemical and microstructural properties of the cryogels, most probably via its self-association during cryogenic processing, promoting the development of intertwined protein-gum networks. FG, on the other hand, enhanced these properties through electrostatic complexation with proteins. Cryogels made from protein-polysaccharide blends exhibited promising techno-functional properties for enhancing and diversifying food product innovation.

Abstract:Water-stable macroporous hydrogels, inspired by the structural and chemical characteristics of plant stems are expected to open a wide range of possibilities in soft materials for passive liquid transport. However, obtaining efficient materials for these applications still poses a major challenge due to the complexity of shaping hydrogels at the relevant scale-length. Here, water-stable macroporous hydrogels were fabricated using alginate and TEMPO-oxidized cellulose via a new approach involving ice templating and topotactic ion-crosslinking with Ca2+. This approach allows to fully avoid the energy-intensive lyophilization process and results in composite hydrogels with pore sizes akin to those found in celery xylem, a model we chose for plant stems. Importantly, the pore size could be tailored by adjusting both the ice-growth velocities and the ratios of alginate to oxidized cellulose. The resulting hydrogels displayed remarkable water stability along with viscoelastic properties and wettability that depend on the alginate and oxidized cellulose ratios. Mechanical properties, such as compression stress and toughness, consistently increased with higher alginate contents. In addition, liquid transport measurements on crosslinked hydrogels with varying compositions and ice growth velocities revealed comparable rising speeds to those observed in celery, confirming the ability of polysaccharide-based hydrogels obtained by ice templating and topotactic crosslinking as relevant materials to mimic the function of plant stems.

Abstract:Porous hydrogels have brought more advantages than conventional hydrogels when used as chromatographic materials, controlled release vehicles for drugs and proteins, matrices for immobilization or separation of molecules and cells, or as scaffolds in tissue engineering. Polysaccharide-based porous hydrogels, in particular, can address challenges related to bioavailability, solubility, stability, and targeted delivery of natural antioxidant compounds. Their porous structure enables the facile encapsulation and controlled release of these compounds, enhancing their therapeutic effectiveness. In this context, in the present study, the cryogelation technique has been adopted to prepare novel dextran (Dx)-based porous hydrogels embedding polyphenol-rich natural extract from Picea abies spruce bark (SBE). The entrapment of the SBE within the Dx network was proved by FTIR, SEM, and energy-dispersive X-ray spectroscopy (EDX). SEM analysis showed that entrapment of SBE resulted in denser cryogels with smaller and more uniform pores. Swelling kinetics confirmed that higher concentrations of Dx, EGDGE, and SBE reduced water uptake. The release studies demonstrated the effective stabilization of SBE in the Dx-based cryogels, with minimal release irrespective of the approach selected for SBE incorporation, i.e., during synthesis (~3–4%) or post-synthesis (~15–16%). In addition, the encapsulation of SBE within the Dx network endowed the hydrogels with remarkable antioxidant and antimicrobial properties. These porous biomaterials could have broad applications in areas such as biomedical engineering, food preservation, and environmental protection, where stability, efficacy, and safety are paramount.

Abstract:Thermosensitive hydrogels have received great research interest, but the elaborate experimental design, inferior mechanical and anti-freeze properties have limited the practical application of thermosensitive hydrogels. For address these challenges, here, polyol (glycerol, xylitol, and sorbitol) and deionized water were selected as mixed solvents to fabricate multifunctional natural gelatin UCST hydrogels by simple physical blending. XRD and FT-IR indicate that the polyol assists the gelatin molecular chain to form short-range β-sheets without affecting the triple helix structure. The short-range β-sheets acts as the physical cross-linking point, which leads to a denser three-dimensional network of the hydrogel. The results indicate that the phase transition temperature and mechanical properties of UCST hydrogels can be modulated by simply changing the amount of polyol in the solvent mixture. In addition, the polyol forms extensive hydrogen bonds with water, which gives the hydrogel excellent freeze resistance, anti-drying and adhesive properties. In this paper, natural UCST gelatin hydrogels with long-term stability, recyclability, thermoplasticity, anti-freeze and adhesive properties were prepared by a simple scheme, and the facile process and cheap cost lays the foundation for the application of UCST hydrogels in drug delivery, skincare products and wearable devices.

Abstract:The low cost and environmental advantages of Xanthan gum make its production and application scale exceed that of other polysaccharides. However, the temperature resistance of Xanthan gum limits its application. In this study, polysaccharide supramolecular Xanthan gum network (XG-β-CD/AD) based on β-cyclodextrin and adamantane was prepared for enhanced oil recovery. The structure of Xanthan gum was characterized by Fourier infrared spectroscopy, nuclear magnetic resonance spectroscopy and thermogravimetric analysis. The rheological properties of the modified polysaccharide network in aqueous solution were systematically studied. The results showed that physical cross-linking of host-guest interacion enhanced the thickening ability of the polymer. Shear rheology, extensional rheology and dynamic modulus test proved that XG-β-CD/AD had excellent rheological properties. The micromorphology, dynamic light scattering and circular dichroism clarified the molecular conformation, the host-guest interaction can improve conformational transition temperature (Tm) and inorganic salt tolerance of Xanthan gum. Under harsh environment (90 °C, 30000 mg/L brine), the oil recovery of XG-β-CD/AD is 6 %-11 % higher than that of XG at the same conditions, showing a better ability to improve the recovery rate. This study provides a research idea for the selection, development and application of biomacromolecular materials.

Abstract:This paper discusses the structure morphology and the thermal and swelling behavior of physically crosslinked hydrogels, obtained from applying four successive freezing–thawing cycles to poly (vinyl alcohol) blended with various amounts of κ-carrageenan. The addition of carrageenan in a weight ratio of 0.5 determines a twofold increase in the swelling degree and the early diffusion coefficients of the hydrogels when immersed in distilled water, due to a decrease in the crystallinity of the polymer matrix. The diffusion of water into the polymer matrix could be considered as a relaxation-controlled transport (anomalous diffusion). The presence of the sulfate groups determines an increased affinity of the hydrogels towards crystal violet cationic dye. A maximum physisorption capacity of up to 121.4 mg/g for this dye was attained at equilibrium.

Abstract:Cross-linking of polysaccharides by metal ions provides polymer gels highly required by industrial applications. In this article, we study the rheological properties and microstructure of solutions of a stiff anionic polysaccharide xanthan cross-linked by chromium (III) ions, and we demonstrate that their properties are highly sensitive to the preparation pH. Stable gels are obtained in a wide range of pH from 2.4 to 7.8. The maximum elastic modulus is observed for the gels made at pH 6.3, and by freeze-fracture transmission electron microscopy it is shown that they are characterized by the most dense network structure. However, out of this pH interval, no gelation is observed. At low pH ( 7.8) the absence of gelation is caused by the transformation of reactive chromium ions into insoluble chromium hydroxide. At the same time, for the gels initially formed at pH 6.3, subsequent change of pH to strongly acidic (1.4) or basic (8.9) medium does not affect appreciably their properties, meaning that chromium cross-links are stable once they are formed. These observations open a reliable route to produce polysaccharide gels with required mechanical properties in a wide pH range where they initially cannot be prepared. It is also shown that the increase of pH to 6.3 of the initially ungelled solution prepared at pH 1.5 results in gelation. This effect offers a facile way for delayed gelation of polysaccharides, which is especially required by oil industry.

Abstract:Abstract Hydrogel is a 3D framework of hydrophilic polymeric material that quickly absorbs and retains a huge amount of water (or other fluid) and offers versatile functionality. A series of unique carboxymethyl cellulose/Xanthan gum/polyvinyl alcohol (CXP) blended hydrogels, containing both the natural and synthetic polymers, were prepared by following the blending and casting approach. The polymers were incorporated through chemical crosslinking by tetraethyl orthosilicate (TEOS). The fabricated hydrogels showed all required features: non-toxicity, biocompatibility, and improved mechanical strength. The addition and variation in TEOS (crosslinker) significantly impacted the key characteristics of CXP hydrogel. The scanning electron microscopy (SEM) images showed the porous structure and indicated that the pore's size and intensity were reduced with the surge in TEOS content. Fourier transform infrared spectroscopy (FTIR) results confirmed the successful incorporation of various polymeric strands through crosslinking by TEOS. The thermogravimetric analysis (TGA) highlighted the greater stability of all the hydrogels over high temperatures. The crosslinked hydrogel displayed higher thermal resilience than the uncross-linked one. The differential thermal analysis (DTA) also confirmed that the addition of TEOS content drastically enhanced the thermal endurance of crosslinked hydrogels in comparison with the neat hydrogel. All the specimens exhibited good swelling ability in distilled water during the swelling studies. This study also reflected that the addition of crosslinker in a limited amount (50 μL) has significantly enhanced the swelling but further increase in concentration hindered the water uptake. The swelling response of blends towards pH revealed low swelling of films in acidic and basic pH, but maximal swelling in neutral media. This unique pH response of hydrogels at neutral pH along with the biocompatibility made them suitable for injectable managed drug carrier.

The current work aimed to develop starch/xanthan hydrogels through extrusion and thermopressing processes, using citric acid (CA) as crosslinking agent and sodium hypophosphite (SHP) as catalyst. The hydrogels were produced with different levels of CA (0.00, 0.75, 1.50 and 2.25 g/100 g polymer). Hydration, mechanical, thermal and microstructural properties of the hydrogels were determined. Swelling behavior of the materials with CA was lower than the control. Additionally, CA ensured the preservation of hydrogels integrity after the swelling process. Gel fraction increased with CA 0.75 g/100 g. Starch/xanthan hydrogels crosslinked with CA demonstrated lower strength than non-crosslinked hydrogels, which may be related to acid hydrolysis of the polymer chains. CA-SHP increased the storage modulus of the hydrogels. Reactive extrusion and thermopressing were efficient methods in the production of crosslinked starch/xanthan based hydrogels.</p>

Abstract:The development of fully biobased hydrogels obtained by simple routes and in the absence of toxic or environmentally harmful reagents is a major challenge in meeting new societal demands. In this work, we discuss the development of hydrogels made from cellulose nanocrystals (CNCs) and xyloglucan (XG), two non-toxic, renewable, and biobased components. We present three strategies to fine-tune the functional properties. The first one consists in varying the XG/CNC ratio that leads to the modulation of the mechanical properties of hydrogels as well as a better comprehension of the gel mechanism formation. The second relies on tuning the XG chains' interaction by enzymatic modification to achieve thermoresponsive systems. Finally, the third one is based on the increase in the hydrogel solid content by osmotic concentration. The high-solid-content gels were found to have very high mechanical properties and self-healing properties that can be used for molding materials. Overall, these approaches are a case study of potential modifications and properties offered by biobased nanocolloidal hydrogels.

Abstract:This research aimed to evaluate the gelation process of ferulated pectin (FP) and ferulated arabinoxylan (AXF) in a new mixed hydrogel and determine its microstructural characteristics. FP from sugar beet (Beta vulgaris) and arabinoxylan from maize (Zea mays) bran were gelled via oxidative coupling using laccase as a crosslinking agent. The dynamic oscillatory rheology of the mixed hydrogel revealed a maximum storage modulus of 768 Pa after 60 min. The scanning electron microscopy images showed that mixed hydrogels possess a microstructure of imperfect honeycomb. The ferulic acid content of the mixed hydrogel was 3.73 mg/g, and ferulic acid dimer 8-5′ was the most abundant. The presence of a trimer was also detected. This study reports the distribution and concentration of ferulic acid dimers, and the rheological and microstructural properties of a mixed hydrogel based on FP and AXF, which has promising features as a new covalent biopolymeric material.

Abstract:The application of biopolymers in tissue engineering is of a great interest due to of their inherent properties such as cell adhesion, biodegradation, bioavailability, and viscoelasticity. In this study, we synthesized cryogels based on biopolymers of gelatin, chitosan, and chondroitin sulfate by cryopolymerization and studied the effect of chondroitin sulfate on changing the physicochemical properties of cryogels such as pore size, pore volume, density, gel fraction, and biodegradation. A macroporous surface of the synthesized polymers has been investigated by SEM. The glass transition temperatures of the crosslinked cryogels, determined by the DSC method, were higher compared to that of the non-crosslinked cryogel used as a reference. The results of the MTT test showed that aqueous extracts of the prepared cryogels had no toxic effect on rat adipose-derived mesenchymal stem cells. The research in this area is of great importance and provides new insights into novel, effective methods for obtaining biopolymers that can be used as carriers of cells.
