Abstract:To date, it is still not possible to obtain exhaustive information about organic materials in cultural heritage without sampling. Nonetheless, when studying unique objects with invaluable artistic or historical significance, preserving their integrity is a priority. In particular, organic dye identification is of significant interest for history and conservation research, but it is still hindered by analytes' low concentration and poor fastness. In this work, a minimally invasive approach for dye identification is presented. The procedure is designed to accompany noninvasive analyses of inorganic substances for comprehensive studies of complex cultural heritage matrices, in compliance with their soundness. Liquid extraction of madder, turmeric, and indigo dyes was performed directly from paint layers and textiles. The extraction was supported by hydrogels, which themselves can undergo multitechnique analyses in the place of samples. After extraction, Ag colloid pastes were applied on the gels for SERS analyses, allowing for the identification of the three dyes. For the HPLC-MS/MS analyses, re-extraction of the dyes was followed by a clean-up step that was successfully applied on madder and turmeric. The colour change perceptivity after extraction was measured with colorimetry. The results showed ΔE values mostly below the upper limit of rigorous colour change, confirming the gentleness of the procedure.
Development of a Combined Protein and Dye Extraction Approach for the Analysis of Keratin-Based Textiles

Abstract:Archaeological textiles represent precious remains from ancient culture; this is because of the historical and cultural importance of the information that can be obtained by such relics. However, the extremely complicated state of preservation of these textiles, which can be charred, partially or totally mineralized, with heavy soil or biological contamination, requires highly specialized and sensitive analytical tools to perform a comprehensive study. Starting from these considerations, the paper presents a combined workflow that provides the extraction of dyes and keratins and keratin-associated proteins in a single step, minimizing sampling while maximizing the amount of information gained. In the first phase, different approaches were tested and two different protocols were found suitable for the purpose of the unique workflow for dyes/keratin-proteins: a slightly modified urea protocol and a recently proposed new TCEP/CAA procedure. In the second step, after the extraction, different methods of cleanup and workflow for proteins and dyes were investigated to develop protocols that did not result in a loss of aliquots of the analytes of interest and to maximize the recovery of both components from the extracting solution. These protocols investigated the application of two types of paramagnetic beads, unmodified and carboxylate-coated hydrophilic magnetic beads, and dialysis and stage-tip protocols. The newly designed protocols have been applied to cochineal, weld, orchil, kermes, and indigo keratin-based dyed samples to evaluate the effectiveness of the protocols on several dye sources. These protocols, based on a single extraction step, show the possibility of investigating dyes and keratins from a unique sample of 1 mg or lesser, with respect to the thresholds of sensitivity and accuracy required in the study of textile artifacts of historical and artistic values.

Abstract:Detection of natural organic dyestuffs in art objects is a challenge for scientists and conservators, as it is limited by severe sampling restrictions. This work is aimed at developing a sampling-free method for the analysis of colorants traditionally used in historical textiles, with a focus on the preservation of the sample integrity to enable its further characterization with other techniques. Thus, in this study, Surface Enhanced Raman Spectroscopy (SERS) was performed directly on laboratory-dyed textiles for the identification of anthraquinone dyes, generally characterized by strong fluorescence signals that hinder conventional Raman spectra. For this purpose, SERS-active silver nanoparticles were produced by Pulsed Laser Ablation in Liquid (PLAL). PLAL presents some advantages when compared to conventional chemical preparation methods, namely the absence of reagents in the suspension and a stronger control over the nanoparticles morphology that can be obtained by changing the ablation conditions. For the analysis, a small piece of fiber was gently rubbed on a microscope glass slide where few drops of Ag nanoparticles were previously deposited and preconcentrated; micro-Raman measurement followed. To ensure the suitability of the analytical strategy for the characterization of historical artworks, the reversibility of the treatment was demonstrated by Laser Induced Breakdown Spectroscopy analysis of a wool fiber treated with nanoparticles and gently cleaned with diluted sodium citrate (<em class="EmphasisTypeItalic">p</em> H 6-7).
Swab-based minimally invasive analysis of synthetic organic dyes using flexible paper-based SERS substrates: A case study of Chinese New Year paintings

Abstract:Surface-enhanced Raman scattering (SERS) has proven to be a powerful diagnostic technique for conservation science. In this work, a novel swab-based detection strategy is proposed for minimally invasive SERS analysis of synthetic organic dyes in the cultural heritage field, employing flexible paper-based SERS probes prepared with a facile, cost-efficient and reproducible recipe. It was demonstrated that Ag nanostructures/paper probes composed of interconnected Ag nanostructures immobilized on unmodified cellulose paper showed excellent SERS enhancement for the investigated synthetic organic dyes. The high flexibility and conformability of paper-based substrate allows to extract minimal amounts of dyes from the work of art by gently swabbing in a dry-state condition, thereby avoiding potential damage to colorants from close contact with solvents such as water. Dyes could be transferred via swab-based extraction method without removing any visible fragments of the paint layer, which facilitates preserving the physical integrity of the artwork. SERS detection was subsequently performed directly on flexible substrate surfaces after micro-extraction of dyes. The minimally invasive nature of this strategy has further been confirmed by the spectrophotometric color measurements and successfully applied to the identification of synthetic organic dyes on real paper artworks-Chinese woodblock New Year paintings from the late Qing Dynasty. These results highlight the great potential of flexible paper-based Ag substrates based on swab-detection method for minimally invasive analysis of organic dyes in cultural heritage.
Developmental and evolutionary origins of the vertebrate dentition: molecular controls for spatio-temporal organisation of tooth sites in osteichthyans.

Abstract:The rainbow trout (Oncorhynchus mykiss) as a developmental model surpasses both zebrafish and mouse for a more widespread distribution of teeth in the oro-pharynx as the basis for general vertebrate odontogenesis, one in which replacement is an essential requirement. Studies on the rainbow trout have led to the identification of the initial sequential appearance of teeth, through differential gene expression as a changing spatio-temporal pattern, to set in place the primary teeth of the first generation, and also to regulate the continuous production of replacement tooth families. Here we reveal gene expression data that address both the field and clone theories for patterning a polyphyodont osteichthyan dentition. These data inform how the initial pattern may be established through up-regulation at tooth loci from a broad odontogenic band. It appears that control and regulation of replacement pattern resides in the already primed dental epithelium at the sides of the predecessor tooth. A case is presented for the developmental changes that might have occurred during vertebrate evolution, for the origin of a separate successional dental lamina, by comparison with an osteichthyan tetrapod dentition (Ambystoma mexicanum). The evolutionary origins of such a permanent dental lamina are proposed to have occurred from the transient one demonstrated here in the trout. This has implications for phylogenies based on the homology of teeth as only those developed from a dental lamina. Utilising the data generated from the rainbow trout model, we propose this as a standard for comparative development and evolutionary theories of the vertebrate dentition.

Abstract:Identifying the fibres in historical textiles presents a complex challenge due to the wide variety of plant, animal and early synthetic materials that have been used. Traditionally, this identification process involves sampling followed by either microscopic examination or ATR-FTIR spectroscopy. However, there are instances when sampling is restricted due to the good condition or significant value of the object under analysis. Additionally, the presence of leather components alongside textiles can further complicate the identification. This paper proposes a novel non-invasive method for fibre identification based on External Reflection (ER) FTIR spectroscopy, which has been rarely applied to textiles or leather. The current research demonstrates that ER-FTIR spectrum is a viable tool for fibre identification on both recent and historical textiles. The non-invasiveness of the analytical approach enables a comprehensive investigation without compromising the number or position of samples. Respect to ATR-FTIR spectra, the ER-FTIR spectra frequently exhibit an amplification of certain diagnostic bands, facilitating the identification of the various fibres examined in this study (cotton, hemp, viscose, silk, wool, leather, polyamide, acrylic, polyester). The extended spectral range (7500-375 cm-1) which is provided by ER-FTIR spectrometry also contains extra bands in the near infrared region, which can provide key information for the discrimination due to the lack of distortion phenomena. The technique was applied to the characterisation of textile materials coming from a collection of 10 traditional Japanese samurai armours spanning from the 16th to the 20th century (Museo delle Culture, Lugano, Switzerland). For the first time, the results provided a comprehensive overview of the textiles utilized in Japanese armours across various historical periods. Overall, the appearance of materials in samurai armours reflects the evolution of armour-making techniques and the influence of socio-cultural factors throughout Japanese history. Synthetic and semi-synthetic materials were easily detected, revealing the occurrence of a past conservation treatment or the early adoption of modern man-made materials in the manufacturing of traditional armours. The approach outlined in this case study can be applied to textile collections of various kinds, offering a reliable mean to discern the yarn composition and detect non-original components. The method also appears as a valuable prescreening tool for designing a less intrusive yet more informative sampling strategy, should additional details about fibre type and dyeing be necessary.

Abstract:A comprehensive analytical protocol was undertaken to elucidate the yellow colorants employed in wall paintings from the Kizil Grottoes, the Taojia Tomb, the Baiyangzhai Tomb, and polychrome figurines from the Astana Tombs, all positioned along the historical Silk Road and dating to the 6th-10th centuries AD. To unequivocally identify these colorants, multi-analytical techniques were deployed, encompassing digital microscopy, scanning electron microscopy coupled with energy-dispersive X-ray spectroscopy (SEM-EDS), X-ray fluorescence spectroscopy (XRF), micro-Raman spectroscopy, and ultra-performance liquid chromatography triple quadrupole time-of-flight mass spectrometry (UPLC-Triple TOF-MS/MS). This comprehensive approach facilitated the unambiguous identification of both inorganic pigments, specifically vanadinite, and plant-derived colorants, including gamboge (Garcinia sp.) and Chinese cork tree (Phellodendron chinense), within these archaeological artifacts. These findings not only significantly enhance our scientific understanding of the materials and techniques utilized in the creation of these wall paintings and polychrome figurines, but also shed light on the vibrant exchange of materials and technological advancements that flourished along the ancient Silk Road.

Abstract:This research focuses on analyzing wool samples dyed with synthetic dyes from the early 20th century. A methodology to identify and distinguish wool fibers dyed with azo, triphenylmethane, and xanthene dyes, which are no longer in use, using the ATR-FTIR spectra, is presented. Firstly, the dataset was subjected to PCA, which revealed the similarities and differences among the samples, illustrating a distribution pattern based on dye classes. MCR-ALS was employed to extract the spectral profiles of the dyed fibers, thereby enhancing the efficacy of the analytical techniques and extracting the comprehensive information from a single instrument. The combination of ATR-FTIR spectroscopy with chemometric methods, such as PCA and MCR-ALS, has proven to be an effective strategy for identifying and differentiating wool fibers dyed with early azo, triphenylmethane, and xanthene dyes. This approach has demonstrated particular effectiveness in enabling rapid analysis without requiring sampling or pretreatment. Moreover, the analysis is supported by thorough bibliographic research on these no longer used colorants. In order to maximize the potential of non-destructive spectroscopic techniques, such as ATR-FTIR, the approach used has proven to be crucial. This study underscores how chemometric techniques expand the capabilities of spectroscopy, extracting extensive information from a single instrument and aligning with the goals of cultural heritage analysis.

Abstract:Developing a noninvasive technique to identify ancient textile dyes is crucial for human civilization. A SERS measurement approach with the alcohol–water droplet is proposed to realize both microextraction and detection of dyes in textile cultural relics. Without any sample pretreatment, dyes in plant extracts and artificial textiles can be identified. Furthermore, different dyes are distinguished in overdyeing through the fingerprint information of SERS. Eventually, this approach is successfully applied to identify dyes of authentic ancient Chinese textiles. Nowadays, surface‐enhanced Raman spectroscopy (SERS) has become a powerful tool for rapidly detecting and analyzing textile cultural relics due to its ability to provide chemical information with single‐molecule sensitivity. However, preserving a high level of detection sensitivity while avoiding sample damage remains a persistent challenge. In this work, we developed a SERS approach with both microextraction and detection functions. The alcohol–water droplet with iodide‐modified Au nanoparticles (AuIMNPs) is directly dropped on the textile, where dyes strongly bound on textiles can be extracted by ethanol (EtOH). As a result, the sample can be well preserved from being damaged. In particular, the volatility of EtOH allows the molecules to be captured in the hot spots through the capillary effect during droplet evaporation, resulting in a dramatic increase in Raman signal intensity. This highly sensitive strategy can be used to measure dyes in plant extracts and mock‐up textiles. Furthermore, the capability of SERS to provide fingerprint information allows us to distinguish different dyes in overdyeing textiles. Eventually, this approach is successfully applied to identify dyes of authentic ancient Chinese textiles. This rapid, universal, and negligibly invasive approach provides a powerful way to study textile cultural relics.

Abstract:Accurate detection of natural dyes in archaeological textiles is challenging due to limited sample availability. In situ detection of fibers by surface-enhanced Raman scattering (SERS) can overcome this challenge. In this study, we present two methods for the preparation of SERS substrates based on the laser photoreduction reaction and the galvanic displacement reaction, respectively. Our results show that the substrates prepared by these methods, including 450 nm-AgNPs, Cu-based Ag, and Zn-based Ag, offer several advantages such as rapid preparation, easy handling, low cost, high sensitivity and excellent reproducibility. Using these substrates, we successfully detected anthraquinones (alizarin, purpurin), indigoids (indigotin, indirubin), and protoberberine-type dyes in archaeological textiles from the Wei and Jin Dynasties. Notably, we were able to detect both indigo and protoberberine-type dyes in one archaeological fiber. The minimally invasive, in situ analytical approach developed provides a valuable tool for analytical applications where sampling is limited to microscopic amounts.

Microanalysis of organic pigments and glazes in polychrome works of art by surface-enhanced resonance Raman scattering

Abstract:Scientific studies of works of art are usually limited by severe sampling restrictions. The identification of organic colorants, a class of compounds relevant for attribution and provenance studies, is further complicated by the low concentrations at which these compounds are used and by the interference of the protein-, gum-, or oil-binding media present in pigment and glaze samples. Surface-enhanced resonance Raman scattering (SERRS) was successfully used to identify natural organic colorants in archaeological objects, polychrome sculptures, and paintings from samples smaller than 25 microm in diameter. The key factors in achieving the necessary sensitivity were a highly active stabilized silver colloid, obtained by the reproducible microwave-supported reduction of silver sulfate with glucose and sodium citrate, and a non-extractive hydrolysis sample treatment procedure that maximizes dye adsorption on the colloid. Among the examples presented are the earliest so far found occurrence of madder lake (in a 4,000 years old Egyptian object dating to the Middle Kingdom period), and the earliest known occurrence in Europe of the South Asian dyestuff lac (in the Morgan Madonna, a 12th century polychrome sculpture from Auvergne, France).

Abstract:Archaeological textile fibres are often preserved in contact with iron or copper alloy objects through mineralisation. Depending on burial conditions these may take the form of positive or negative casts. Often and perhaps surprisingly many mineralised fibres still preserve small amounts of the original organic matter. While in the past mineralised fibres have usually been investigated with instrumental analysis techniques such as scanning electron microscopy and Fourier transform infrared spectrometry, this article describes the use of simple microscopic chemical staining tests for proteins (animal fibres) and cellulose/lignin (plant fibres) to complement the visual assessment of mineralised fibrous remains. It was found that a classification even of highly degraded archaeological fibres was possible with a combination of naphthol blue black for animal fibres and zinc chloride/iodine (Herzberg stain) and safranine O for plant fibres.

Abstract:This paper proposes a method for optimized the Unmanned Surface Vehicle (USV) structure based on a combination of iSIGHT optimization software and the parameterized model of geometry and finite element. The finite element model of the vehicle was established by using the parameterize language (ANSYS Parametric Design Language) of ANSYS based on the study of the material of the USV hull. And then the expected target function, the variables and the boundary condition were selected, the optimized value was obtained by using the multi-island genetic algorithm. Finally, analysis was conducted on the conditions of sagging and hogging of the USV, and the strength of the hull was calibrated. The results proves that it can be reduce the weight of the boat and improve its performance by using this method, under the precondition of meting the strength and the general arrangement requirements.

Abstract:This paper demonstrates that an educated methodology based on both non-invasive and micro invasive techniques in a two-step approach is a powerful tool to characterize the materials and stratigraphies of an Egyptian coffin, which was restored several times. This coffin, belonging to a certain Mesiset, is now located at the Museo Civico Archeologico of Bologna (inventory number MCABo EG 1963). Scholars attributed it to the late 22nd/early 25th dynasty by stylistic comparison. The first step of the diagnostic approach applied imaging techniques on the whole surface in order to select measurements spots and to unveil both original and restored areas. Images and close microscopic examination of the polychrome surface allowed selecting representative areas to be investigated in situ by portable spectroscopic techniques: X-ray Fluorescence (XRF), Fiber Optic Reflectance Spectroscopy (FORS) and Fourier Transform Infrared spectroscopy (FTIR). After the analysis of the results coming from the first step, very few selected samples were taken to clarify the stratigraphy of the polychrome layers. The first step, based on the combination of imaging and spectroscopic techniques in a totally non-invasive modality, is quite unique in the literature on Egyptian coffins and enabled us to reveal many differences in the ground layer's composition and to identify a remarkable number of pigments in the original and restored areas. This work offered also a chance to check the limitations of the non-invasive approach applied on a complex case, namely the right localization of different materials in the stratigraphy and the identification of binding media. Indeed, to dissolve any remaining doubts on superimposed layers belonging to different interventions, it was necessary to sample few micro-fragments in some selected areas and analyze them prepared as cross-sections. The original ground layer is made of calcite, while the restored areas show the presence of either a mixture of calcite and silicates or a gypsum ground, overlapped by lead white. The original pigments were identified as orpiment, cinnabar and red clay, Egyptian blue and green copper based pigments. Some other pigments, such as white lead, Naples yellow, cerulean blue and azurite were only found in the restored areas.

Abstract:The purpose of the present report was the study and identification of an unspecified sample on a work of art by means only of a microscope coupled to a Raman spectrometer. The origin of the fragment was unknown. The Raman spectra on the virgin sample were giving no results because of a deteriorated surface treatment, in spite of the evident blue color identified by microscopic visual inspection. The sample fragmentation and the preparation of a KBr pellet allowed the distribution of the painting layers of the different components on a flat substrate reducing surface effects. Selecting the areas of different color and focusing there it was possible to identify the pigments from their Raman spectra locally acquired by selective excitation. Raman spectra were assigned by comparison with published databases. It was possible to connect Carbon Black and Orange iron oxide, as documented historically, as constituents of Azurite preparatory layer Morellone, according to a technique generally employed to allow the use of Azurite blue pigment on frescos, consequently the identification of typology of work of art was deducted and attributed to a fresco.

The aim of this study was to develop and evaluate techniques that can be applied in the field of culture and art. A selection of 65 synthetic dyestuffs was chosen to investigate the usefulness of the analytical techniques applied. As a case study three embroideries, designed by the well-known 19th century French painter Emile Bernard, were investigated to obtain more information about the use and behaviour of synthetic dyes.

Abstract:Charles T. Currelly, first director of the Royal Ontario Museum, participated in excavations of the tomb of King Nebhepetre, now known as Mentuhotep II, (Dynasty XI) in Deir el-Bahri, Egypt in 1906. He brought to Canada many objects from the excavations, and objects that he purchased while in Egypt; these formed the initial collection of the museum. Among the objects were seven fragments of fine linen cloth with intricate pleat patterns. Recently, the cloths became the subject of a study to learn how they had retained their pleats for 4000 years. Samples were examined and analysed using polarised light microscopy, scanning electron microscopy-electron dispersive X-ray spectrometry, gas chromatography-mass spectrometry, and pyrolysis-gas chromatography-mass spectrometry. Three of the cloths were likely fragments of clothing re-purposed as bandages and were found to be saturated in mummification balms composed of Pinaceae resin, Pistacia resin, and an essential oil characterised by a high abundance of cedrol, possibly originating from a juniper species. All seven of the cloths were found to have traces of polysaccharides from two probable sources: an arabinogalactan gum such as gum arabic or a fruit gum, and a polyglucoside, possibly starch.

Ancient Textile Deterioration and Restoration: Bio-Cleaning of an Egyptian Shroud Held in the Torino Museum

Abstract:Abstract Ancient textiles are fragile and several factors can affect their integrity. In the present chapter, the main agents of deterioration of old and new textiles, namely physical-chemical (light, oxygen, heat, and humidity) and biological factors as well as human erroneous interventions will be explored. As far as the biological deterioration is considered, the effects of microbial growth, primary and secondary metabolites (acids, solvents, surfactants, pigments) and enzymes (lipases, proteases, and glycosidases) on textile strength and cleanliness will be described in details. The main fungal and bacterial species involved in the damage (textile discoloration, black and green spots, cuts) will be reported. Adhesive application during restoration procedures is discussed to highlight the risk of glue thickening giving rise to dull precipitates on the fabric. The main strategies for oil-stain and glue removal (both animal glue, such as fish collagen, and vegetal glue, i.e. starch) will be described in the paragraph devoted to biorestoration. Finally, a case study concerning an ancient Coptic tunic housed in the Egyptian Museum of Torino, Italy, and biocleaned by means of gellan-immobilized alpha-amylase from Bacillus sp. will be largely discussed by reporting historical data, adhesive characterization, methods for artificial aging of simulated sample and glue removal from the artwork.

Abstract:The old fibers that make up heritage textiles displayed in museums are degraded by the aging process, environmental conditions (microclimates, particulate matter, pollutants, sunlight) and the action of microorganisms. In order to counteract these processes and keep the textile exhibits in good condition for as long as possible, both reactive and preventive interventions on them are necessary. Based on these ideas, the present study aims to test a natural and non-invasive method of cleaning historic textiles, which includes the use of a natural substance with a known antifungal effect (being traditionally used in various rural communities)—lye. The design of the study was aimed at examining a traditional women's shirt that is aged between 80–100 years, using artificial intelligence techniques for Scanning Electron Microscopy (SEM) imagery analysis and X-Ray powder diffraction technique in order to achieve a complex and accurate investigation and monitoring of the object's realities. The determinations were performed both before and after washing the material with lye. SEM microscopy investigations of the ecologically washed textile specimens showed that the number of microorganism colonies, as well as the amount of dust, decreased. It was also observed that the surface cellulose fibers lost their integrity, eventually being loosened on cellulose fibers of cotton threads. This could better visualize the presence of microfibrils that connect the cellulose fibers in cotton textiles. The results obtained could be of real value both for the restorers, the textile collections of the different museums, and for the researchers in the field of cultural heritage. By applying such a methodology, cotton tests can be effectively cleaned without compromising the integrity of the material.
