Abstract:To overcome the long start-up period in cultivating aerobic granular sludge (AGS) under hypersaline environment, mycelial pellets (MPs) of halotolerant fungus Cladosporium tenuissimum NCSL-XY8 were inoculated to try to realize the ultra-rapid development of salt-tolerant AGS by stable transition of 'hollow' MPs into 'solid' AGS without apparent fragmentation. The granules directly met the standard of AGS after inoculating MPs (Day 0), and it basically satisfied relatively strict standards of AGS (SVI30 < 50 mL/g, D50 > 300 μm, D10 > 200 μm and SVI30/SVI5 > 0.9) under anaerobic/aerobic mode during whole cultivation processes. Microstructure of the granular cross section clarified that MPs with hollow/loose inner layer transitioned into solid/dense AGS under anaerobic/aerobic mode within 7 days, while formed skin-like floating pieces and unstable double-layer hollow granules under aerobic mode. Organics removal reached relatively stable within 13 days under anaerobic/aerobic mode, 6 days faster than aerobic mode. This study provided a strategy for ultra-rapid and stable development of AGS, which showed the shortest granulation period in various AGS-cultivation strategies.

Abstract:Aerobic granular sludge (AGS) was rapidly cultivated in an integrated oxidation ditch with two-zone clarifiers by using a novel external sludge treatment and return mode to treat low concentrations of actual domestic sewage. The selective pressure created by the two-zone clarifiers can retain the well-settling granules and discharge light flocs with poor settleability. The granules stayed in the reactor, which induced bacterial attachment to the granules that acted as nuclei, while the discharged flocs can stimulate microorganisms to secrete large amounts of extracellular polymeric substances (EPS) under the external conditioning of CaCl2 and natural air drying. Then, this surplus sludge was returned to the reactor to create more small granules that combined with each other through the action of hydraulic shear forces to achieve rapid granulation. The results showed that AGS was formed successfully in the reactor on day 18, and after 51 days of continuous operation, the biomass concentration and settling ability were further improved (the mixed liquor suspended solids (MLSS) and sludge volume index at 5 min (SVI5) were stable at approximately 3500 mg/L and 40.0 mL/g, respectively). During the whole experimental period, the biological sludge activity was greatly improved, and the EPS and microbial community changed significantly, including an enrichment of microbes with EPS secretion and granule stabilization functions. The study results reveal that the pollutant removal efficiency improved after granulation. Furthermore, this approach required less energy and is eco-friendly for potential full-scale implementation.
Synergistic Effect of Chitosan-Based Sludge Aggregates CS@NGS Inoculum Accelerated the Start-Up of Biofilm Reactor Treating Aquaculture Effluent: Insights into Performance, Microbial Characteristics, and Functional Genes.

Abstract:The moving bed bioreactor (MBBR) process has drawn more attention as a promising biological wastewater treatment process. Nevertheless, achieving quick start-up and microbial biofilm formation remains a significant challenge. Consequently, the present study investigated a novel chitosan-based natural sludge (CS@NGS) seeding strategy for the accelerated start-up of MBBR. Three identical bioreactors were employed; the first bioreactor was without sludge seed as the control (BR1), the second was inoculated only with sludge (BR2), and the third was inoculated with CS@NGS according to the proposed seeding method (BR3). All bioreactors were utilised to treat simulated recirculating aquaculture systems (RAS) effluent. Resultantly, the CS@NGS shortened the start-up period from over twenty to seven days due to the enhanced initial microbial adhesion and biofilm formation. Under optimal conditions, the ammonium removal in BR3 approached 100%, which was relatively higher than BR2 (96.35 +/- 1.12%) and BR1 (92.56 +/- 2.17%). Moreover, a low nitrite accumulation was exhibited in the effluents, approximately <= 0.03 mg L-1. The process performance correlated positively with core bacteria from the genera Nakamurella , Hyphomicrobium , Nitrospira , Paenarthrobacter , Rhodococcus , and Stenotrophobacter. The quantitative polymerase chain reaction (qPCR) results demonstrated that the CS@NGS enhanced the expressions of amoA, nxrB, nirK, nirS, narG, and napA nitrogen metabolism-related functional genes to varying degrees. The present study findings can assist the rapid start-up of aquaculture biofilters utilised to solve high nitrite and ammonia accumulation in recirculated water from industrial RAS.

Abstract:Aerobic granular sludge (AGS) technology showed great potentials in treatment of hypersaline industrial wastewater such as shale gas flowback water (SGFW), while long start-up period was still a major bottleneck. To both strengthen its granulation and pollutant removal efficiency, herein, one multifunctional fungus Aspergillus sydowii NCSL-XY1 was first isolated with the capacities including formation of fungal pellets, salt tolerance, and efficient degradation of organics in SGFW. Subsequently, its appropriate fungal pellets were used to try to shorten the start-up period of AGS in SGFW. The results showed that inoculating fungal pellets evidently accelerated the formation of AGS from 41 to 16 days. Also, it could enhance the TOC removal from ∼30% to ∼45% in the initial start-up period (Day 2-Day 11). Mechanisms analysis showed that the inoculation of fungal pellets increased EPS secretion and microbial community diversity. When the reactor stabilized, both of the AGS in R1 and R2 had similar D 50 (∼1200 μm), SVI 30 (∼30 mL/g), and SVI 30 /SVI 5 (∼0.95). Also, the TOC and PAM removal in R1 and R2 were also similar and maintained at ∼71% and ∼20%. Overall, the inoculated fungal pellets played significant roles of both biomass carrier and synergistic degrader in facilitation of AGS start-up, while it had limited functions after the granules maturation. This study provided a universally effective bioaugmentation mode (screening degrading fungus and inoculating its fungal pellets) for AGS cultivation in treatment of refractory industrial wastewater.

Abstract:Upregulation of programmed death 1 ligand 1 (PD-L1) in cancer cells and its ligation to PD-1 on T cells facilitates cancer cell escape from immune surveillance. Therapies with PD-1 or PD-L1 antibodies have resulted in marked clinical responses in various cancer types. Hence, modulators that inhibit PD-L1 expression in cancer cells may serve as a novel strategy by which to enhance host immune responses. In the present study, we investigated the effects of miR-375 on PD-L1 expression in head and neck squamous cell carcinoma (HNSCC) cells by qRT-PCR and western blot analyses. We confirmed that miR-375 inhibited IFN-γ-induced PD-L1 surface expression in HNSCC cells, and we observed that Janus kinase 2 (JAK2) is a bona fide target of miR-375 and further activated signal transducer and activator of transcription 1 (STAT1). Additionally, miR-375-mediated inhibition of PD-L1 expression was dependent on the JAK2/STAT1 pathway. Therefore, by attenuating PD-1/PD-L1 signaling, miR-375 may also serve as a modulator to increase the cell immune responses to HNSCC.

Abstract:AGS technology has been studied for decades, and many successful experimental research approaches and full-scale processes have been proposed. However, a precise, comprehensive understanding of the granulation mechanism remains lacking. In this study, sequencing batch reactors were partially inoculated with (S1) or without AGS (S2) to explore aerobic granulation performance in simulative low-strength wastewater. The COD, NH4+-N removal efficiency and SND rate were 91.61%, 92.47%, and 84%, respectively, in S1 and higher in this reactor than in S2 (86.76%, 85.21%, and 75%, respectively), indicating the superiority of partial inoculation with AGS. The ratios of P release/NaAc uptake both increased to nearly 0.250, revealing the robust capacity for phosphorus enrichment. Moreover, the increment of EPS content in S2 (31.73 mg/g MLSS) was 47% higher than that in S1 (21.54 mg/g MLSS) and was contributed mainly by PS, which increased to 30.17 mg/g MLSS (S2). These findings indicated active sludge for granulation was more sensitive to the ambient conditions, and secrete greater PS. FTIR and 3D-EEM revealed that numerous hydroxyl groups were assembled within EPS upon granulation, and humic acid-like substances first increased and then decreased over the operation, implying robust cohesion and positive microbial activities. Microbial community analysis demonstrated that Paracoccus and unclassified_Cytophagales dominated in both reactors, accounting for 29.42% and 10.87%, respectively, of the community in S1 and 26.62% and 18.54%, respectively, of the community in S2. Metabolic pathway analysis revealed enhancement of microbial metabolism, cellular processes, and signaling pathways, such as methanol oxidation, methylotrophy, and nitrite denitrification pathways.

Abstract:The adaptation of microbial community is vital for the potential use of aerobic granular sludge (AGS) as a technology for high -salinity wastewater. Although AGS has dense microbial aggregates and excellent settling ability, it requires a long adaptation period and may become unstable when exposed to salt stress. This study investigated the adaptation process of microbial community in salt -tolerant AGS (S-AGS) reactors to identify core microbes and their synergistic action during the AGS adaptation period. Compared to the control groups (R1, municipal sludge; R2, industrial sludge), the S-AGS (R3, mixed sludge) was quickly formed within 30 days and maintained with good structural stability (integrity coefficient > 0.95, particle size > 1200 mu m) and high removal efficiency to COD ( >90 %) and TN ( >78 %). The core community of S-AGS in R3 was dominated by Thauera spp. and Pseudofulvimonas spp., while Rhodobacter spp. and Flavobacterium spp. were significantly inhibited during high salt stress. Co -occurrence network analysis revealed that Thauera spp. and Pseudofulvimonas spp. were the essential modules for S-AGS. The RT-qPCR results indicated that a higher level of nitrogen metabolism -related genes ( nxrB , napA ) ensured the functional potential of simultaneous nitrification-denitrification (SND). Furthermore, the results of microbial community analysis indicated that microbial synergy via enhancing ion transport and tryptophan secretion played a vital role in resisting the salt stress of AGS. This study provided a deeper insight into the stable development of S-AGS through microbial synergy, thereby providing practical implications for the saline wastewater treatment.

Abstract:Aerobic granular sludge (AGS) can achieve simultaneous carbon, nitrogen and phosphorus removal owing to its three-dimensional oxygen gradient structure. However, long start-up period and poor operational stability restrict its application and promotion. A novel rapid granulation strategy, viz., the short-term (7 days) addition of ferric chloride at the commissioning stage, was developed and verified in this study. The granulation period was shortened by 9 days, and the formed granules were compact and dense with an Fe3+ concentration of 250 mg L−1. The addition of flocculant not only maintained a high sludge concentration during the initial stages of granulation (5.3 g L−1), but also stimulated the secretion of TB-EPS and increased protein and polysaccharide contents, thereby expediting granule formation. Additionally, ferric chloride induced a diverse microbial community in granules, resulting in the emergence of new genera, such as Thaurea, Brevundimonas and Kinneretia, which improved pollutant removal performance and flocculent aggregation. The removal efficiencies of COD, PO43--P, and NH4+-N stabilized at 94.2, 62.4, and 71.3%, respectively. Therefore, it has been demonstrated that short-term ferric chloride dosing has a synergistic effect on aerobic granulation.GRAPHICAL

Abstract:This study presents a novel strategy to accelerate the start-up of aerobic granular sludge (AGS) system and ensure the nutrient removal during cultivation. This new method consists of preparing the chitosan-based sludge aggregates outside the reactor and then seeding the reactor with such sludge aggregates. To prepare chitosan-based sludge aggregates, chitosan was dissolved in acetic acid solution acting as a cationic flocculant to bind negatively charged sludge together, and then the dissolved chitosan was in situ precipitated by readjusting pH to form stable sludge aggregates. The chitosan-induced charge neutralization and water-insolubility of chitosan were the two main reasons for the super-rapid formation of chitosan-based sludge aggregates. The as-prepared chitosan-based sludge aggregates had a much lower sludge volume index at 30 min (SVI30) (90.1 mL/g) than the original sludge (SVI30 = 328.0 mL/g). They also had some AGS-like characteristics such as large particle size (1300 μm) and fast settling velocity (23.8 m/h). Consequently, short settling time can be achieved and excessive biomass wash-out can be avoided in the rapid start-up of AGS system with chitosan-based sludge aggregates as inoculant, which was beneficial to accelerating sludge granulation while maintaining nutrient removal. Additionally, the abundances of filamentous bacteria and Candidatus Accumulibacter and the content of extracellular polymeric substances increased during cultivation, which could also contribute to the AGS formation. By seeding chitosan-based sludge aggregates in the anaerobic/oxic sequencing batch reactor, complete granulation was rapidly achieved in 10 days, and good removals of nitrogen and phosphorus was obtained after 14-18 days of cultivation.

Abstract:Aerobic granular sludge (AGS) needs a long start-up time and always shows unstable performance when it is used to treat low-strength wastewater. In this study, a rapid static feeding combined with Fe2+ addition as a novel strategy was employed to improve the formation and stability of AGS in treating low-strength wastewater. Fe-AGS was formed within only 7 days and showed favorable pollutant removal capability and settling performance. The ammonia nitrogen (NH4+-N) and chemical oxygen demand (COD) concentration in the effluent were lower than 5 mg/L and 50 mg/L after day 23, respectively. The sludge volume index (SVI) and mixed liquid suspended solids (MLSS) was 37 mL/g and 2.15 g/L on day 50, respectively. Rapid static feeding can accelerate granules formation by promoting the growth of heterotrophic bacteria, but the granules are unstable due to filamentous bacteria overgrowth. Fe2+ addition can inhibit the growth of filamentous bacteria and promote the aggregation of functional bacteria (eg. Nitrosomonas, Nitrolancea, Paracoccus, Diaphorobacter) by enhancing the secretion of extracellular polymeric substances (EPS). This study provides a new way for AGS application in low-strength wastewater treatment.
Enhanced aerobic granular sludge formation by applying Phanerochaete chrysosporium pellets as induced nucleus

Abstract:The long start-up period is a major challenging issue for the widespread application of aerobic granular sludge (AGS). In this study, a novel rapid start-up strategy was developed by inoculating Phanerochaete chrysosporium (P. chrysosporium) pellets as the induced nucleus in a sequencing batch airlift reactor (SBAR) to enhance activated sludge granulation. The results demonstrated that P. chrysosporium pellets could effectively shorten the aerobic granulation time from 32 to 20 days. The AGS promoted by P. chrysosporium pellets had a larger average diameter (2.60–2.74 mm) than that without P. chrysosporium pellets (1.78–1.88 mm) and had better biomass retention capacity and sedimentation properties; its mixed liquor suspended solids (MLSS) and sludge volume index (SVI30) reached approximately 5.2 g/L and 45 mL/g, respectively. The addition of P. chrysosporium pellets promoted the secretion of extracellular polymeric substances (EPS), especially protein (PN). The removal efficiencies of chemical oxygen demand (COD), ammonia nitrogen (NH4+-N), total nitrogen (TN), and total phosphorus (TP) in P. chrysosporium pellets reactor were 98.91%, 89.17%, 64.73%, and 94.42%, respectively, which were higher than those in the reactor without P. chrysosporium pellets (88.73%, 82.09%, 55.75%, and 88.92%). High throughput sequencing analysis indicated that several functional genera that were responsible for the formation of aerobic granules and the removal of pollutants, such as Acinetobacter, Pseudomonas, Janthinobacterium, and Enterobacter, were found to be predominant in the mature sludge granules promoted by P. chrysosporium pellets.

Abstract:Aerobic granular sludge (AGS) technology has been recognized as a promising alternative to alleviate the osmotic stress of hypersaline wastewater. However, the response of AGS process to composite hypersaline wastewater on removal performance and populations was yet to be understood. In this work, two sequenced batch reactors were operated in parallel in absence (R0) and presence (R1) of high concentration sulfate as proxy for single and mixed salts (30 g salt·L−1) respectively. Results demonstrated that the presence of sulfate in hypersaline wastewater enhanced chemical oxygen demand (COD) and total nitrogen (TN) removals of 95.3% and 65.5% respectively with lower accumulations of nitrite. High-throughput 16 S rRNA gene sequencing technique elucidated that Denitromonas (31.6%) and Xanthomarina (17.0%) were the more dominant genera in AGS response to mixed salts with high sulfate and laid the biological basis for strengthening removal performance. The enrichment of halophilic Luteococcus (23.5%) in the AGS surface indicated the potential role of mixed salts in shaping the physical properties and surface population structure of AGS. Our work could facilitate the potential applications of AGS technology for industrial hypersaline wastewater treatment with complicated compositions.

Abstract:Salt-tolerant aerobic granular sludge (SAGS) technology is currently a topic of significant interest in research focused on the treatment of saline wastewater. However, the structural stability and nitrogen removal efficiency of SAGS cultured from activated sludge has been found to be insufficient. In this study, an efficient strategy for the cultivation of SAGS, seeding anaerobic granular sludge in a SBR, was explored. Performance results showed that at 3% (w/v) salinity, the chemical oxygen demand (COD), NH4+-N and total nitrogen (TN) removal efficiency was 93.3 ± 1.3%, 92 ± 6.5% and 76 ± 8.4%, respectively. The morphology and settling properties of granules did not change significantly. Bacterial community analysis revealed that the dominant AOB and denitrifiers in SAGS were likely to be Nitrosomonas and Defluviimonas, respectively, while the abundance of NOB of Nitrospira was very low. qPCR results showed that the abundance of nirS and nirK decreased significantly under high salinity conditions, which likely explains the relatively low efficiency of TN removal and NO2−-N accumulation. This study describes a new SAGS culture strategy and promotes the development of salt-containing wastewater biological treatment technology.

Abstract:Rapid granulation was realized in a continuous-flow reactor with a two-zone sedimentation tank (CFR-TST), and a novel strategy involving the continuous addition of dewatered sludge during the start-up phase to treat the actual domestic sewage with low concentration. The selection pressure produced by the two-zone sedimentation tank can discharge light flocs with poor settleability and retain the fine particles in the reactor. The continuous addition of dewatered sludge in the start-up phase can increase the small particles in the aeration tank by acting as nuclei to accelerate granulation. The experimental results indicated that aerobic granular sludge (AGS) was formed successfully on day 25 (the sludge volume index at 5 min (SVI5) was 37.7 mL/g, the mean particle size was 0.42 mm, and the average settling velocity was 35.2 m/h); after operated continuously for 45 days, the sedimentation capacity and biomass concentration were further enhanced (SVI5 and the mixed liquor suspended solids (MLSS) were stable in the range of 25.8–31.3 mL/g and 4200−4600 mg/L, respectively). Although the dewatered sludge with low biological activity had a slight negative impact on pollutants degradation in the start-up phase, the biological activity of the sludge was enormously upgraded (compared with those of the inoculated dewatered sludge, the SOUR and dehydrogenase activity of the mature AGS increased 3.72 times and 5.01 times on day 84, respectively), and microbes that promote the secretion of EPS and facilitate the degradation of pollutants were enriched during the whole process of granulation. Moreover, the approach developed in this case was simple and eco-friendly for potential full-scale implementation.

The results showed that the granulation time in MPs-MBR was shortened by at least 65 days, accounting for enhanced mean size of granules (0.68-0.76 mm), increased mixed liquor suspended solids (MLSS) concentration (12.8 g/L) and improved settling ability (78.1 mL/g), in comparison with that of 0.23-0.28 mm, 9.8 g/L and 102.1 mL/g in control MBR. MPs-MBR demonstrated significant advantages in terms of COD reduction (97.0-99.1%), NH4+-N reduction (100%) and TN reduction (32.27-42.33%). MPs, extracellular polymeric substances (EPS) and filamentous bacteria acted as inducible nucleus, crosslinking matter and supporting skeleton, respectively, in favor of promoting the formation and stabilization of AGS with a four-layered structure. The relevant mechanism was underlined by rheological analysis, indicating that MPs addition enhanced non-Newtonian flow characteristics and network structure of sludge.

Abstract:Direct cultivation of aerobic granular sludge (AGS) in membrane bioreactor (MBR) has gained increasing attention. Mycelial pellets (MPs) has been shown capable of promoting rapid granulation of aerobic sludge in MBR, yet mechanisms remain unclear and in-depth insight into cross-scale interactions between MPs and indigenous microbiota as well as the corresponding protein expression functions is necessary. Herein, we found that the addition of MPs in MBR resulted in massive growth of metazoans with 40-400 /mL for rotifers, 20-140 /mL for nematodes and 2-420 /mL for oligochaetes in the initial phase of granulation. This facilitated the MPs to rapidly aggregate with bacteria to form defensive granules for physical protection from predation by metazoans, which inhibited the overgrowth of filamentous bacteria Thiothrix and promoted the reproduction of functional bacteria related to nitrogen removal ( Nitrospira, Trichococcus and Acinetobacter ). Proteomic analysis demonstrated that the upregulation of functional proteins was mainly ascribed to the decrease of Thiothrix and the increase of Nitrospira , resulting in the enhancement of metabolic pathways involved in glycolysis/gluconeogenesis, citrate (TCA) cycle, oxidative phosphorylation, pyruvate metabolism, nitrogen metabolism and biosynthesis of amino acids, which was responsible for MPs-induced AGS with denser structure, more abundant proteins and β-polysaccharides, higher species diversity, significant nitrogen removal (33.12-42.33%) and lower membrane fouling potential. This study provided a novel and comprehensive insight into the enhanced granulation of aerobic sludge by MPs and the functional superiority of MPs-induced AGS in MBR system.

Abstract:Aerobic granular sludge which had good performance to pollutants removal was successfully cultivated within 18 days in a pilot scale sequencing batch reactor, about 25% mature aerobic granular sludge was inoculated when the setting time of activated sludge was reduced to 10 min. Anaerobic biological selector was implemented to inhibit filamentous bacteria overgrowth, where the maximum COD could reach to 1703.74 mg/L. The cultivated aerobic granular sludge was irregular and pale yellow, average particle size, SVI, SV₃₀/SV₅, PN/PS, EPS and water content were 1.58 mm, 67.64 mL/g, 0.91, 2.17, 268.90 mg EPS/g MLVSS and 98.16% on the 18th day. Mechanism of rapid granulation mainly included crystal nucleus hypothesis and selection pressure hypothesis. The inoculated aerobic granules could maintain stable under short setting time environment, making it directly as the crystal nucleus and the carriers for new particles without obvious disintegration, which eventually shortened the granulation time greatly.

Abstract:Abstract In this study, the feasibility of aerobic granular sludge formation without inoculated sludge was detected. After 10 days operation, AGSs were discovered in reactor and the MLSS and settling ability of AGS enhanced gradually. After 21 days operation, the effluent NH 4 + -N and COD keep low than 10 and 100 mg/L, respectively. During the formation of AGS, the content of EPS gradually increased and remained stable. Microbial community showed that EPS secretion genera (such as Paracoccus, Flavobacterium and Thauera ) were main genera in AGS.

Abstract:Starvation and antibiotics pollution are two frequent perturbations during breeding wastewater treatment process. Supplying magnetite into anaerobic system has been proved efficient to accelerate microbial aggregates and alleviate the adverse effect caused by process disturbance. Nevertheless, whether these magnetite-based granules are still superior over normal granules after a long-term starvation period remains unknown, the responsiveness of these granules to antibiotics stress is also ambiguous. In current study, we investigated the resilience of magnetite-based anaerobic granular sludge (AnGS) to starvation and oxytetracycline (OTC) stress, by unravelling the variations of reactor performance, sludge properties, ARGs dissemination and microbial community. Compared with the AnGS formed without magnetite, the magnetite assisted AnGS appeared more robust defense to starvation and OTC stress. With magnetite supplement, the average methane yield after starvation recovery, 50 mg/L and 200 mg/L OTC stress was enhanced by 48.95%, 115.87% and 488.41%, respectively, accompanied with less VFAs accumulation, improved tetracycline removal rate (76.3-86.6% vs. 51.0-53.5%) and higher ARGs reduction. Meanwhile, magnetite supplement effectively ameliorated the potential sludge breakage by triggering more large granules formation. Trichococcus was considered an important impetus in maintaining the stability of magnetite-based AnGS process. By inducing more syntrophic methanogenesis partnerships, especially for hydrogenotrophic methanogenesis, magnetite ensured the improved reactor performance and stronger resilience at stress conditions.
