Abstract:Radix astragali polysaccharide (RAP) is a water-soluble heteropolysaccharide. It is an immune promoter and regulator, and has antivirus, antitumor, anti-aging, anti-radiation, anti-stress, anti-oxidation and other activitys. The extraction, separation, purification, structure, activity and modification of RAP were summarized. Some extraction methods of RAP had been introduced, and the separation and purification methods of RAP were reviewed, and the structure and activity of RAP were highly discussed. Current derivatization of RAP was outlined. Through the above discussion that the yield of crude polysaccharides from Radix astragali by enzyme-assisted extraction was significantly higher than that by other extraction methods, but each extraction method had different extraction effects under certain conditions, and the activity efficiency of RAP was also different. Therefore, it is particularly important to optimize the extraction method with known better yield for the study of RAP. In addition, the purification and separation of RAP are the key factors affecting the yield and activity of RAP. At the same time, there are still few studies on the derivatiration of Radix astragali polysaccharide, but the researches in this area are very important. RAP also has many important pharmacological effects on human body, but its practical application needs further study. Finally, studies on the structure-activity relationship of RAP still need to be carried out by many scholars. This review would provide some help for further researches on various important applications of RAP.

Abstract:Ethnopharmacologic relevance Rehmannia glutinosa (Gaertn.) DC. ( RG ) is a widely used herb for clearing heat and cooling the blood. Polysaccharides from Rehmannia glutinosa (Gaertn.) DC. (RGPs) have a variety of biological activities, including antioxidation, hypoglycemia, immune enhancement, hematopoiesis promotion, and antianxiety. Aim of the study This review provides up-to-date and comprehensive information on the extraction and separation methods, structural characteristics, and pharmacological activities of RGPs. A more in-depth study on the structure and clinical pharmacology of the RGPs was investigated. To further explore the pharmacological effects of RGPS, and lay a foundation for the safe clinical application and expansion of application scope. Materials and Methods Use Google Scholar, Scifinder, PubMed, Springer, Elsevier, Wiley, Web of Science and other online database search to collect the literature on extraction, separation, structural analysis and pharmacological activity of RGPs published before December 2022. The key words are "extraction", "isolation", "purification" and "pharmacological action" and "Rehmanniae polysaccharide". Results Rehmannia glutinosa has been widely used in the treatment of diabetes since ancient times, and is known as one of the "Four Sacred Medicines" for the treatment of diabetes, along with Ginseng, Psidium Guajava and Pueraria Mirifica. The active ingredients of Rehmannia glutinosa that have been studied more in the treatment of diabetes are Rehmannia glutinosa polysaccharide and Rehmannia glutinosa oligosaccharide. The content of polysaccharides varies due to different extraction methods, and separation and purification methods. RGPs have a wide range of pharmacological activities, including antitumor, immunomodulatory, neuroprotective effect, hypoglycemic activity, cardioprotective and antioxidant activities. These pharmacological properties lay a foundation for the treatment of tumors, inflammation, hyperglycemia, myocardial ischemia, oxidative stress and other diseases with RGPs. Conclusion Based on its effects of promoting hematopoiesis, antitumor and enhancing immunity, RGPs have been clinically applied in the treatment of chronic aplastic anemia and esophageal cancer, but other effects of RGPs have not been reflected in the clinical practice. In the future, more in-depth research can be conducted on the molecular structure analysis, toxicity, side effects and clinical pharmacological effects of RGPs to further explore the pharmacological effects of RGPs and to lay the foundation for safe clinical application and expansion of application scope.

Abstract:Ethnopharmacological relevance Saposhnikoviae Radix (SR) is a traditional Chinese medicine, known as "Fangfeng". As one of the main active components, Saposhnikoviae Radix polysaccharides (SP) demonstrated a range of biological activities, especially immunity regulation activity. Aims of the study This study aimed at exploring whether polysaccharides have activity after degradation, then discovering the potential effective fragments of SP. Materials and methods Here we establish the chromatographic fingerprints method for 32 batches of 1-phenyl-3-methyl-5-pyrazolone (PMP) derivatives of oligosaccharides by HPLC, meanwhile evaluating its immunomodulatory activity in vivo . Then, the potential effective fragments of SP were screened out based on the spectrum-effect relationship analysis between fingerprints and the pharmacological results. Besides, liquid chromatography ion trap-time of flight mass spectrometry (LC-IT-TOF MS) coupled with multiple data-mining techniques was used to identify the potential effective oligosaccharides. Results These findings showed that the hydrolysate of SP have significant immunomodulatory, and the immunity regulation activity varies under different hydrolysis conditions. The 4 potential effective peaks of the hydrolysate of SP were mined by spectrum-effect relationship. Finally, the chemical structure of 4 potential effective oligosaccharide fragments of SP was elucidated based on LC-IT-TOF MS. F10 was inferred tentatively to be Hex 1 → 6 Hex 1 → 6 Hex 1 → 6 Hex 1 → 6 Hex 1 → 6 Gal; F18 was confirmed to be Rhamnose; F14 was inferred tentatively to be Hex 1 → 4 Hex 1 → 4 Hex 1 → 4 Gal and F25 was tentatively inferred to be Ara 1 → 6 Gal. Conclusions This study may provide a sound experimental foundation in the exploration of the active fragments from macromolecular components with relatively complex structures such as polysaccharides.

Abstract:Although the increasing studies have corroborated the biological activities and great market utilization value of polysaccharide fractions derived from Polygonati rhizome, a well-known edible and medicinal plant, Polygonati rhizome polysaccharides (PRPs) still lack sufficient attention. Herein, we make attempt to systematically summarize recent advances in the extraction, purification, structural characteristics, biological activities, and commercial products of PRPs. Based on the detailed extraction and structural characteristics, the biological activities of PRPs including immune-regulation, anti-osteoporosis, anti-Alzheimer's disease, anti-diabetes and anti-atherosclerotic, are emphatically summarized, as well as the possible related mechanisms. Most importantly, about 365 kinds of commercial functional foods and over 500 patents related to PRPs as the main raw material were analyzed to explore the status quo and bottleneck for the development and utilization of PRPs. In conclusion, this review will benefit to bridge the gap between basic knowledge and market innovations, and facilitate the in-depth utilization of PRPs.

Abstract:Ethnopharmacological relevance Polygonati Rhizome (PR) is a plant that is extensively widespread in the temperate zones of the Northern Hemisphere. It is a member of the Polygonatum family of Asparagaceae. PR exhibits diverse pharmacological effects and finds applications in ethnopharmacology, serving as a potent tonic for more than two millennia. PR's compounds endow it with various pharmacological properties, including anti-aging, antioxidant, anti-fatigue, anti-inflammatory, and sleep-enhancing effects, as well as therapeutic potential for osteoporosis and age-related diseases. Aim of the study This review seeks to offer a thorough overview of the processing, purification, extraction, structural characterization, and biosynthesis pathways of PR. Furthermore, it delves into the anti-aging mechanism of PR, using organ protection as an entry point. Materials and methods Information on PR was obtained from scientific databases (Google Scholar, Web of Science, ScienceDirect, SciFinder, PubMed, CNKI) and books, doctoral theses, and master's dissertations. Results In this investigation, 49 polysaccharides were extracted from PR, and the impact of various processing, extraction, and purification techniques on the structure and activity of these polysaccharides was evaluated. Additionally, 163 saponins and 46 flavonoids were identified, and three key biosynthesis pathways of secondary metabolites were outlined. Notably, PR and Polygonat Rhizomai polysaccharides (PRP) exhibit remarkable protective effects against age-induced injuries to the brain, liver, kidney, intestine, heart, and vessels, thereby promoting longevity and ameliorating the aging process. Conclusions PR, a culinary and therapeutic herb, is rich in active components and pharmacological activities. Based on this review, PR plays a crucial role in lifespan extension and anti-aging, which can be attributed to PRP. Future research should delve deeper into the structural aspects of PRP that underlie its anti-aging effects and explore potential synergistic interactions with other compounds. Moreover, exploring the potential applications of PR in functional foods and pharmaceutical formulations is recommended to advance the development of industries and resources focused on healthy aging.

Abstract:Fifty patients at risk for postoperative lung dysfunction and undergoing elective coronary revascularization have been randomly assigned to receive normothermic (36°C) perfusion with warm heart protection (NP group) or hypothermic (28°C) perfusion with cold heart protection (HP group). Lung function before and after cardiopulmonary bypass (CPB) was studied through the determination of the intrapulmonary shunt (Qs/Qt), the alveolo-arterial oxygen gradient (A-aΔO2), and the artero-alveolar carbon dioxide gradient (a-AΔCO2). The Q s /Q t after CPB was significantly lower in the NP group (27.1 ± 2.6 vs 35.7 ± 2.3) as well as the A-aΔO2 (50.2 ± 1.5 vs 57.6 ±2.4); both data returned to comparable between the groups after 3 h in the intensive care unit. The a-AΔCO2 was significantly lower after CPB in the NP group (5.2 ± 0.74 vs 8.2 ± 0.8). Hospital stay and mortality were comparable in the two groups; intubation time and rate of early extubation showed a trend in favour of the NP group; the rate of patients suffering hypoxia and/or hypercapnia after extubation was significantly lower in the NP group (12%) versus the HP group (44%). Normothermia seems to exert a protective effect against lung dysfunction after CPB. The absence of a rewarming injury associated with reperfusion, a limitation of the hypothermic-induced vasoconstriction due to local cooling of the lung and a better compliance of the normothermic lung are hypothesized as beneficial effects of the ‘all-warm’ strategy.

Abstract:The genus Rhodiola L., an integral part of traditional Chinese medicine and Tibetan medicine in China, exhibits a broad spectrum of applications. This genus contains key compounds such as ginsenosides, polysaccharides, and flavonoids, which possess anti-inflammatory, antioxidant, hypoglycaemic, immune-enhancing, and anti-hypoxic properties. As a vital raw material, Rhodiola L. contributes to twenty-four kinds of Chinese patent medicines and 481 health food products in China, finding extensive application in the health food sector. Recently, polysaccharides have emerged as a focal point in natural product research, with applications spanning the medicine, food, and materials sectors. Despite this, a comprehensive and systematic review of polysaccharides from the genus Rhodiola L. polysaccharides (TGRPs) is warranted. This study undertakes a systematic review of both domestic and international literature, assessing the research advancements and chemical functional values of polysaccharides derived from Rhodiola rosea. It involves the isolation, purification, and identification of a variety of homogeneous polysaccharides, followed by a detailed analysis of their chemical structures, pharmacological activities, and molecular mechanisms, structure-activity relationship (SAR) of TGRPs. The discussion includes the influence of molecular weight, monosaccharide composition, and glycosidic bonds on their biological activities, such as sulfation and carboxymethylation et al. Such analyses are crucial for deepening the understanding of Rhodiola rosea and for fostering the development and exploitation of TGRPs, offering a reference point for further investigations into TGRPs and their resource utilization.

Abstract:Ethnopharmacological relevance: Salvia miltiorrhiza is a traditional Chinese medicine with the application of more than a two-thousand-year history. It is a common medicine used in the clinical treatment of cardiovascular and cerebrovascular diseases and is listed as the top grade in Shennong's Classic of Materia Medica. Polysaccharide is an important chemical component of Salvia miltiorrhiza and has a variety of biological activities. Aim of the study: In this review, we summarized the preparation methods, structural characteristics, and biological activities of Salvia miltiorrhiza polysaccharides, as well as discussed current research problems, providing support for further research, development, and utilization. Materials and methods: By inputting the search term "Salvia miltiorrhiza polysaccharides", relevant research information was obtained from databases such as Google Scholar, PubMed, VIP, Web of Science, and China Knowledge Network (CNKI). Results: It has been found that the monosaccharide composition of Salvia miltiorrhiza polysaccharides containing glucose (Glc), galactose (Gal), mannose (Man), and arabinose (Ara) has antioxidant, anti-tumor, liver protection, and other activities. Conclusions: We summarized the preparation methods, structural information, and biological activities of Salvia miltiorrhiza polysaccharides in this review and discussed the issues that are currently being researched. Although this product has a wide range of biological activities and has high development and utilization potential, its structure information and structure-activity relationship require further investigation.

Abstract:Fritillaria is a traditional Chinese medicine(TCM) with a history of thousands of years. Fritillaria always contain saponins, alkaloids, amino acids, flavones, and polysaccharides. Among them, Fritillaria polysaccharide has a variety of biological activities. Its anti-inflammatory and antiaging activities are new study hotspots. The extraction, purification, quantitative determination, monosaccharide composition, and biological activity of Fritillaria polysaccharides have been examined for several years in an attempt to identify the active components and understand the pharmacological process. In this review, ample original publications related to the distribution, extraction, purification, quantitative determination, monosaccharide composition and biological activities of Fritillaria until 2023 were searched and collected by using various literature databases. Databases included the China National Knowledge Infrastructure, VIP database, Wan Fang database, PubMed, Elsevier, Springer, Science Direct, Google Scholar and Web of Science database, as well as the classic Chinese medical books and PhD and MSc theses. The properties and outcomes of various extractions, purifications, quantitative determination methods, monosaccharide compositions, and biological activities of Fritillaria polysaccharides are discussed here. Additionally, we summarize the research potential of Fritillaria polysaccharide and identify promising research direction candidates.

Abstract:Bupleuri Radix (BR), as a well-known plant medicine of relieving exterior syndrome, has a long history of usage in China. Bupleuri Radix Polysaccharide (BRP), as the main component and an important bioactive substance of BR, has a variety of pharmacological activities, including immunoregulation, antioxidant, antitumor, anti-diabetic and anti-aging, etc. In this review, the advancements on extraction, purification, structure characteristics, biological activities and pharmaceutical application of BRP from different sources (Bupleurum chinense DC., Bupleurum scorzonerifolium Willd., Bupleurum falcatum L. and Bupleurum smithii Woiff. var. Parvifolium Shan et Y. Li.) are summarized. Meanwhile, this review makes an in-depth discussion on the shortcomings of the research on BRP, and new valuable insights for the future researches of BRP are proposed.

Abstract:The genus Schisandra, a member of the Magnoliaceae family, is a well-known tonic traditional Chinese medicine with a long history of traditional medicinal and functional food used in China. Polysaccharides are one of its main active constituents, which have a wide range of bioactivities, such as anti-inflammatory, anti-tumor, neuroprotection, anti-diabetes, hepatoprotection, immunomodulation, and anti-fatigue. In this paper, we review the extraction, isolation, purification, structural characterization, bioactivities, as well as structure-activity relationship of polysaccharides from the genus Schisandra. In conclusion, we hope that this review could provide reference for the subsequent research on structural, bioactivities, development and application of the genus Schisandra polysaccharides.

Abstract:Ethnopharmacological relevance Polygonum cuspidatum Sieb. et Zucc. is mainly distributed in Shanxi, Gansu, and Sichuan province of China. It is also found in Korea and Japan. Its dried roots and rhizomes are used as medicinal herbs and have been used to treat hyperglycemia and various inflammatory disorders. Aim of the review This paper aims to provide an up-to-date review of the developments in the studies involving the extraction and purification, structure analysis, pharmacological effects, and potential applications of polysaccharides obtained from Polygonum cuspidatum . Additionally, the possible future research directions of this plant are discussed. Materials and methods This article used " Polygonum cuspidatum polysaccharide (PCP)" and " Polygonum cuspidatum " as the keywords and gathered relevant data on Polygonum cuspidatum using electronic databases (Elsevier, PubMed, ACS, CNKI, Google Scholar, Baidu Scholar, Web of Science), relevant books, and classic literature about Chinese herb. Results Excluding irrelevant and repetitive documents, 278 documents were finally included, of which 88 were in Chinese and 190 were in English. The CiteSpace software was used to visualize the trends and keywords in this research field. We concluded that the main extraction methods for Polygonum cuspidatum polysaccharide are water extraction and alcohol precipitation, microwave-assisted extraction, ultrasound-assisted extraction, and microjet extraction. High-performance liquid chromatography and column chromatography are also commonly used in the separation and purification of PCP. PCP has antitumor, immunomodulatory, hypoglycemic, and antioxidant effects. This paper provides an updated and deeper understanding of PCP, serving as a theoretical foundation for the further optimization of polysaccharide structures and the development of PCP as a novel functional material for clinical application.

Abstract:This study aimed to isolate polysaccharides from Penthorum chinense Pursh and evaluate their rheological characteristics, physicochemical properties, and antioxidant activity. The optimal conditions for the maximal extraction yield of Penthorum chinense Pursh polysaccharides (4.05 ± 0.12%) were determined by employing a single-factor test and response surface methodology which included an extraction time of 3 h, a liquid–solid ratio of 20 mL/g, and three separate extraction times. The rheological experiments showcased that the P. chinense polysaccharides exhibited typical shear-thinning behavior, with their apparent viscosity being influenced by various parameters such as concentration, pH, temperature, salt content, and freeze–thaw. The purified polysaccharides (PCP-100), having an average molecular weight of 1.46 × 106 Da, mainly consisted of glucose (18.99%), arabinose (22.87%), galactose (26.72%), and galacturonic acid (21.89%). Furthermore, the PCP-100 exhibited high thermal stability and displayed an irregular sheet-like morphology. Its superior reducing power and free radical scavenging ability implied its significant antioxidant activity in vitro. Collectively, these findings provide important insights for the future application of P. chinense polysaccharides in the food industry.

In this study, information about "Polygala tenuifolia Willd.," "Polygala sibirica L.," and "Yuanzhi" was retrieved from scientific databases, including Google Scholar, Baidu Scholar, Web of Science, PubMed, CNKI, and Wan Fang Data. Information from Chinese herbal medicine classics, Yaozhi Data, and the Gaide Chemical Network was also collected. Information related to botany, phytochemistry, pharmacology, toxicity, industrial applications, and processing is summarized in this paper to tap its potentialities and promote its further development and clinical application. More than 320 metabolites have been isolated from PR; saponins, xanthones, and oligosaccharide esters are the main functional metabolites. Pharmacological research shows that its pharmacological action mainly focuses on resisting nervous system diseases, and it also has the functions of anti-oxidation, anti-inflammation, anti-tumor, anti-pathogenic microorganisms and others. The gastrointestinal irritation of its saponins impeded its application, but this irritation can be reduced by controlling the dosage, compatibility with other herbs, or processing. The future progress of PR faces opportunities and challenges. More attention should be paid to the traditional application and processing methods of PR recorded in ancient books. The lack of safety and clinical studies has limited its application and transformation of achievements. Moreover, it is one-sided to take the content of only a few metabolites as the index of processing optimization and quality control, which cannot reflect the full pharmacological and toxicological activities of PR.

Abstract:Abstract The root of Angelica sinensis (Oliv.) Diels, a well-known Chinese herbal medicine, has been used historically as hematopoietic and anti-inflammatory agents for thousands of years. Recent phytochemistry and pharmacological studies have proved that polysaccharides are one of the major active ingredients in A. sinensis . It has been demonstrated that ASPs ( A. sinensis polysaccharides) had various important biological activities, such as hematopoietic, hepatoprotective, hypoglycemic, anti-inflammatory, antitumor, and antioxidant activities. The purpose of this present review is to appraise previous and current literatures on the extraction, purification, structural characterization and biological activities of ASPs. In addition, the structure–activity relationship will be further explored and discussed. We believe that this review will provide a useful bibliography for the investigation, production, and application of ASPs in functional foods and therapeutic agents. Moreover, this review also highlights the challenges of investigation and future considerations for holistic utilization.

Abstract:Abstract Penthorum chinense Pursh is a well-known traditional Chinese medicine, however, little attention has been paid to the polysaccharides of P. chinense (PCP). Therefore, it is great significance to investigate the characteristics and activities of PCP. In this study, response surface methodology was applied to optimize the extraction parameters of PCP. Then, two polysaccharides fractions (PCP-1 and PCP-2) were purified from PCP by using DEAE-52 chromatography, and there preliminary chemical properties and in vitro antioxidant activities were investigated. Results revealed that the optimum extraction conditions of PCP were identified as follow: ratio of water to raw material 20.6 mL/g; extraction time 3.5 h and extraction temperature 85°C. Based on these conditions, the maximum yield of PCP was 3.12% ± 0.19%. Purified fractions PCP-1 and PCP-2 were all acidic heteropolysaccharides, and PCP-1 mainly consisted of galactose and arabinose while PCP-2 mainly consisted of rhamnose, galacturonic acid, galactose and arabinose. Moreover, PCP-2 exhibited stronger scavenging activities against DPPH radical, hydroxyl radical and superoxide anion radical and chelating activity on Fe 2+ in vitro .

Abstract:Fritillaria ussuriensis Maxim. (F.M.) has been widely used in both food and medication for more than 2000 years. In order to achieve its comprehensive utilization and investigate the structural characterization and biology activity, response surface methodology (RSM) was used to optimize the ultrasound-assisted extraction conditions of F.M. polysaccharides. The optimal extraction conditions were ultrasonic power of 174.2 W, ratio of liquid to material of 40.7 mL/g and ultrasonic time of 82.0 min. In addition, a neutral polysaccharide F-1 was obtained, and its structure characterization, antioxidant and immunological activity were evaluated. The structural properties of the polysaccharide were characterized by UV, IR, GC-MS, NMR and AFM. Monosaccharide composition of F-1 (MW 18.11 kDa) was rhamnose, arabinose, glucosamine hydrochloride, galactose, and glucose which under the ratio of 0.9: 3.8: 0.2: 2.9: 92.2. The fractions of F-1 were mainly linked by → 6)-α-D-Glcp-(1 → with branch chain α-D-Glcp-(1 → 4)-α-D-Glcp-(1 → and 4,6)-α-D-Glcp-(1 → residues. Moreover, F-1 has a significant scavenging activity, which can clear hydroxyl radicals, superoxide anion, DPPH and ABTS. In addition, the immunological activity showed that F-1 had an effect on macrophage phagocytic activity. And it can increase the release of inflammatory factors including TNF-α, IL-1β and IL-6. F-1 is a novel polysaccharide with significant activity in antioxidant and immunological activity, which has great potential for antioxidant and immunizer in food, pharmaceutical and cosmetic industries. The study can provide a methodological basis for polysaccharide research and theoretical basis for the industrialized production and practical application.

Abstract:Penthorum chinense has been used in both food and medication for many years, and polysaccharide of which was considered as one of the bioactive compounds. However, the extraction process of polysaccharide from P. chinense (PCP) was not well optimized. Ultrasound-assisted extractionhas been widely employed in the extraction of natural products for its compliance with the concept of green and economic chemistry. To better investigate the structure and biology activity of PCP, response surface methodology was employed to optimize the ultrasound-assisted extraction conditions of PCP. The optimum extraction for the ultrasound-assisted extraction of PCP were obtained as ratio of solvent to material 40 mL/g, ultrasonic power 380 W, and extraction time of 50 min. The yield of PCP reached 8.71% under these optimized conditions. PCP was further purified by using anion exchange chromatography and gel filtration, an acidic fraction PCP-AP-1 was hereby obtained. The results of structural elucidation indicated that PCP-AP-1 was a typical pectic polysaccharide with a molecular weight of 66360 Da, mainly composed of galacturonic acid (68.5 mol%), followed by arabinose (9.8 mol%), rhamnose (9.4 mol%), glucose (7.7 mol%), with homogalacturonan region and rhamnogalacturonan I regions. In vitro study showed that PCP-AP-1 could improve the inflammation induced by lipopolysaccharide in intestinal epithelial cells, which was probably performed through the inhibition of multiple signaling pathways including the inhibition of TLR4, NOD1/2 and NF-κB pathway, as well as the reduction of NLRP3 inflammasome. This study defined the type of polysaccharide present in P. chinense and revealed a potential of application this plant in the prevention of intestinal inflammatory diseases.
