Abstract:Agarwood is resin-containing wood produced by plants that have been injured. It is widely used in herbal medicine, incense, decorative items, and so on. In this study, we conducted resin area statistical analysis, determined starch particle and reducing sugar contents, and performed multivariate statistical analysis of chemical composition by GC-MS and UPLC-Q-TOF-MS to explore the different components in sections cut from an agarwood column, designated as A1-A4. The results showed that after stimulation by Agar-Bit inducer, the internal phloem parenchyma cells of the column started to form agarwood, and then starch granules were converted into soluble reducing sugars and agarwood resin. Section A1 showed rapid loss of starch granules, resulting in higher contents of reducing sugars and resin. The resin areas of agarwood in the respective sections were different, gradually decreasing on going from A1 to A4. Total numbers of metabolites of 87 and 63 were identified by GC-MS and UPLC-Q-TOF-MS, respectively. Of these, 10 and 16 metabolites with significant differences (variable importance projection >1) were selected through multivariate statistical analysis. These metabolites included chromones, sesquiterpenes, alkanes, and fatty acids. Among them, 6-methoxy-2-(2-phenylethyl)chromone and 6,7-dimethoxy-2-(2-phenylethyl)chromone were significant markers detected by both GC-MS and UPLC-Q-TOF-MS, which may be essential substances responsible for differences in the agarwood-forming capacities of the cut sections. In conclusion, there has been limited research on the different agarwood-forming capacities of agarwood columns. Here, we explored the differences in various sections of agarwood through chemical analysis to provide a more comprehensive and in-depth understanding of its constitution.
Whole-Tree Agarwood-Inducing Technique: An Efficient Novel Technique For Producing High-Quality Agarwood In Cultivated Aquilaria Sinensis Trees

Abstract:Agarwood is the fragrant resin-infused wood derived from the wounded trees of Aquilaria species. It is a valuable non-timber forest product used in fragrances and as medicine. Reforestation for Aquilaria trees in combination with artificial agarwood-inducing methods serves as a way to supply agarwood and conserve of wild Aquilaria stock. However, the existing agarwood-inducing methods produce poor-quality agarwood at low yield. Our study evaluated a novel technique for producing agarwood in cultivated Aquilaria trees, called the whole-tree agarwood-inducing technique (Agar-Wit). Ten different agarwood inducers were used for comparison of Agar-Wit with three existing agarwood-inducing methods. For Aquilaria trees treated with these ten inducers, agarwood formed and spread throughout the entire tree from the transfusion point in the trunk to the roots and branches of the whole tree. Agarwood yield per tree reached 2,444.83 to 5,860.74 g, which is 4 to 28 times higher than that by the existing agarwood-inducing methods. Furthermore, this agarwood derived from Agar-Wit induction was found to have a higher quality compared with the existing methods, and similar to that of wild agarwood. This indicates Agar-Wit may have commercial potential. Induction of cultivated agarwood using this method could satisfy the significant demand for agarwood, while conserving and protecting the remaining wild Aquilaria trees.
Analysis on the Quality of Agarwood Produced Via the Whole-tree Agarwood-inducing Technique in Different Area

Abstract:Objective: To analyze and evaluate the quality of agarwood produced by the whole-tree agarwood-inducing technique in Aquilaria sinensis( Lour.) Gilg trees from different area. Methods: We have analyzed all the quality standard index of the agarwood defined in ‘Chinese Pharmacopoeia’( Volume I),including characteristic description, microscopic characteristic, TLC, color reaction and extractives content. Results: The description,microscopic feature,TLC chromatogram and color reaction of the four test agarwood sample from Qiongzhong County and Haikou City of Hainan Province and Lianjiang County and Huazhou County of Guangdong Province are same as the standard agarwood sample. The alcohol soluble extractive contents of the four test agarwood are 17. 87%,11. 15%,24. 02% and 16. 5% respectively. Conclusion: These four test agarwood produced by whole-tree agarwood-inducing technique in Aquilaria sinensis( Lour.) Gilg in different area,are measure up to the standard of ‘Chinese Pharmacopoeia’.
Isolation and screening of fungi for enhanced agarwood formation in Aquilaria sinensis trees

Abstract:Agarwood is a resinous heartwood of Aquilaria sinensis that is formed in response to mechanical wounding. In the present study pre-treatment of Aquilaria sinensis was carried out, and then the dominant fungi were isolated and purified from the surface and electroshock holes of trees. The isolated Trichoderma sp . and Neurospora sp . were then screened for resistance against benzyl acetone and then inoculated into healthy Aquilaria sinensis trees. After six months, the agarwood was collected for analysis. The chemical composition of incense was analyzed using gas chromatography-mass spectroscopy, and 82 chemical constituents were identified. Agarwood products formed by using Trichoderma sp . and Neurospora sp . consisted of 50.22% and 48.71% ether extracts, respectively, which surpassed the 10% threshold specified by the Chinese Pharmacopoeia. Similarly, relative aromatic contents in the two agarwood products were 30.1% and 32.86%, while proportions of sesquiterpene constituents were 10.21% and 11.19%, respectively. These two agarwood-specific chemical constituents accounted for a large proportion of the total chemical composition, which showed that the generated agarwood was of good quality. The results of the study revealed that both Trichoderma sp . and Neurospora sp . were able to effectively induce agarwood production in Aquilaria sinensis trees in 6 months. This study expands the library of fungi that promote the production of agarwood from Aquilaria sinensis trees.

Composition variation of agarwood-associated microbial communities from Aquilaria sinensis

Abstract:Agarwood, derived from Aquilaria sinensis and Aquilaria malaccensis , is of medicinal and ecological value and religious importance as incense. The existing imbalance between short supply and increasing demand of this product remains to be solved. Thus, the biologically artificial agarwood-inducing methods commonly called whole-tree agarwood-inducing techniques (agar-wit) have been established to dramatically improve agarwood yield within a short period. However, several studies reported a lower content of ethanol-soluble extractive in the agar-wit agarwood than in the natural agarwood. To further understand the role of microorganisms in agarwood formation, we investigated and contrasted the endophytic bacteria and fungi between different types of agarwood from A. sinensis through high-throughput sequencing. Results showed that the same dominant phyla of bacteria consisting of Proteobacteria, Actinobacteria, and Acidobacteria were shared by the natural agarwood and agar-wit agarwood. Meanwhile, Ascomycota and Basidiomycota constituted the similar dominant fungal phyla of these two kinds of agarwood. However, the principal microbial communities at the genus or order level evidently varied from natural agarwood to agar-wit agarwood. Moreover, the bacterial communities are closely connected with terpenoid and carbohydrate metabolism, which indicated that the bacterial communities also play a vital role in agarwood formation. In conclusion, the higher concentrated abundance of the dominant microbial communities in agar-wit agarwood than in natural agarwood may promote agarwood formation, however, the low evenness of microbial communities also lowers the content of ethanol-soluble extractive.
Salty treatment increased bioactive compounds accumulation during agarwood development in Aquilaria sinensis trees

Abstract:To compare the bioactive compounds in agarwood induced by different methods in Aquilaria sinensis(Lour.) Gilg trees, a two dimensional thin layer chromatograph(2D-TLC) combined with effect directive analysis(EDA) was developed. Three antioxidants were found by 2D-TLC-DPPH and further identified as 2-(2-phenylethyl) chromones(PECs) with LC-MS/MS. The 3 antioxidants decreased along agarwood formation and their compositions in drilling induced agarwood differed with those in microbe culture induced agarwood. Further study showed NaCl treatment promoted antioxidants accumulation in agarwood induced by drilling or hot drilling. Hot drilling combined with salty stimulation was most efficient in some chemicals accumulation, which were identified as PECs with antioxidant, tyrosinase or β-glucosidase inhibiting activities by 2D-TLC-EDA-LC-MS/MS. This study provided a 2D-TLC-EDA-LC-MS/MS method for bioactive compounds screen and qualification of agarwood. Based on this method, non-conventional methods were found to accelerate the accumulation of some bioactive PECs in A. sinensis trees.

Abstract:Recently, "artificial agarwood" manufactured by the artificial treatment on cultivated agarwood trees is popular in agarwood-producing countries. Although there are various treatment methods, they are not standardized. Moreover, factors that may affect the generated chemical compounds have not been investigated. In this research, the effects of different treatment methods and individual differences on the quantities and types of 2-(2-phenylethyl)chromone in agarwood were investigated to experimentally produce artificial agarwood using Aquilaria sinensis. Each solvent-extracted agarwood sample was analyzed using Liquid Chromatography/Tandem Mass Spectrometry (LC–MS/MS), and peaks were identified by comparing ten types of 2-(2-phenylethyl)chromone with reference standards. The composition and 2-(2-phenylethyl)chromone content of each agarwood sample were observed based on the type of chemical compound, and results indicated that when the treatment method was different, the accumulation pattern of the 2-(2-phenylethyl)chromones differed even when the number of resinification years was the same. Furthermore, the findings of this study showed that additional treatment on a single branch produced more 2-(2-phenylethyl)chromones. Moreover, market products composed of artificial agarwood pieces derived from different tree species and collected from different location were analyzed.

Abstract:Background: Agarwood is a highly sought-after resinous wood for uses in medicine, incense, and perfume production. To overcome challenges associated with agarwood production in Aquilaria sinensis, several artificial agarwood-induction treatments have been developed. However, the effects of these techniques on the metabolome of the treated wood samples are unknown. Therefore, the present study was conducted to evaluate the effects of four treatments: fire drill treatment (F), fire drill + brine treatment (FS), cold drill treatment (D) and cold drill + brine treatment (DS)) on ethanol-extracted oil content and metabolome profiles of treated wood samples from A. sinensis. Results: The ethanol-extracted oil content obtained from the four treatments differed significantly (F < D < DS < FS). A total of 712 metabolites composed mostly of alkaloids, amino acids and derivatives, flavonoids, lipids, phenolic acids, organic acids, nucleotides and derivatives, and terpenoids were detected. In pairwise comparisons, 302, 155, 271 and 363 differentially accumulated metabolites (DAM) were detected in F_vs_FS, D_vs_DS, F_vs_D and FS_vs_DS, respectively. The DAMs were enriched in flavonoid/flavone and flavonol biosynthesis, sesquiterpenoid and triterpenoid biosynthesis. Generally, addition of brine to either fire or cold drill treatments reduced the abundance of most of the metabolites. Conclusion: The results from this study offer valuable insights into synthetically-induced agarwood production in A. sinensis.

Abstract:Agarwood is a dark resinous wood, of high quality and in a short period, can be formed after inoculation with the efficient fungus in Aquilaria sinensis . However, the action site and mechanism of the fungus inducing the agarwood formation remain unclear. In this work, the efficient fungal infection was used to induce agarwood formation on A. sinensis , and samples were collected according to inoculation time. Subsequently, the xylem structure, histochemistry, defense enzyme activities, and the accumulation and in-situ distribution of chemical composition were investigated. The transition layer (TL) was the critical layer for the continuous accumulation of agarwood, and began to appear in the injection of fungal inducers for 15 days (15D). The parenchyma cells of the xylem rays and interxylary phloem were the main action site of fungus in the formation and accumulation of agarwood. The sesquiterpenes and 2-(2-phenylethyl) chromones (PECs) were mainly accumulated in the agarwood layer (AL) and decay layer (DL), while their relative contents were highest in the injection of fungal inducers for 30 days and 180 days (30D and 180D), respectively. Among them, PECs appeared earlier than sesquiterpenes, and both were flindersia-type PECs. They were mainly concentrated in the cell corners and intercellular layer of the cell wall. In addition, Redundancy analysis (RDA) and Mantel test revealed that the different layer percentage (DLP), phenylalanine ammonia lyase (PAL), and starch grains (SG) were the drivers of PECs, and catalase (CAT) was the driver of sesquiterpenes. Therefore, this study will provide a theoretical foundation for revealing the mechanism of fungus-induced agarwood formation and inducing high-quality and high-yielding agarwood in the future.

Abstract:Obtained from Aquilaria Lam. and Gyrinops Gaertn., agarwood is a prestigious perfume and medicinal material in the world. Its primary chemical constituents and indicators of agarwood's development are 2-(2-phenylethyl)chromones (PECs). However, how PECs affect its quality, accumulation, and transformation pattern is still unclear. The present study investigated this issue by monitoring resin filling in agarwood generated by the whole-tree agarwood-inducing technique over a span of a year, observing the ethanol extract concentration at different sampling times, and statistically examining PECs in agarwood from each sampling period. In agarwood, the resin accumulated over time, except during the 4th-6th month due to the creation of a barrier layer. The relative content of total PECs demonstrated an overall increase throughout the year but a decrease from the 4th month to the 6th month, and the relative content of 19 PECs that persisted throughout the year was positively correlated with the content of ethanol extracts. In addition, the process of chromone accumulation was accompanied by the production and transformation of different types of chromones, with flindersia type 2-(2-phenylethyl)chromones, epoxy-2-(2-phenylethyl)chromones, and diepoxy-2-(2-phenylethyl)chromones being the major chromone components; in addition, the content of 5,6,7,8-tetrahydro-2-(2-phenylethyl)chromones kept increasing after 6 months of agarwood formation. Three main trends were identified from 58 analogs of PECs, each with notable variation. The first type had the highest content at the beginning of resin formation. The second type had the highest content at 6 months and then started to decrease, and the third type had a slowly increasing content. As a whole, this study systematically investigated the accumulation of PECs during injury-induced agarwood production in A. sinensis, which is of scientific significance in resolving the transformation of PECs and revealing the secret of agarwood formation.

Abstract:The present study is to investigate the chemical constituents and anti-inflammation of agarwood produced via whole-tree agarwood-inducing technique( Agar-Wit) from Aquilaria sinensis by column chromatographic technique and semi-preparation HPLC.Eleven sesquiterpenes were isolated from the agarwood produced by Agar-Wit,and their structures were identified on the basis of physiochemical characteristics and spectroscopic data analysis as baimuxinol( 1),5α,7α( H)-eudesm-11( 13)-en-4α-ol( 2),( 7 S,9 S,10 S)-( +)-9-hydroxy-selina-4,11-dien-14-al( 3),petafolia A( 4),7( 11)-eremophilen-8-one( 5),neopetasane( 6),petafolia B( 7),11-hydroxy-valenc-1( 10)-en-2-one( 8),( 4αβ,7β,8αβ)-3,4,4α,5,6,7,8,8α-octahydro-7-[1-( hydroxymethyl) ethenyl]-4α-methylnaphthalene-1-carboxaldehyde( 9),12-hydroxy-4( 5),11( 13)-eudesmadien-15-al( 10),and( 4 R,5 R,7 S,9 S,10 S)-(-)-eudesma-11( 13)-en-4,9-diol( 11). Among them,compound 1 was a new natural product,and this is the first time to report its13 CNMR spectroscopic data. Compounds 4,9 and 10 were reported from Aquilaria for the first time,and all the compounds are firstly isolated by Agar-Wit from A. sinensis. The anti-inflammatory activity of RAW264. 7 cells with lipopolysaccharide-induced was evaluated.As a result,1,4 and 9 showed potential anti-inflammatory activities with IC50 values( 2. 5±0. 35),( 3. 2±0. 2),( 4. 3±0. 56) μmol·L-1,respectively. This work provided scientific foundation for quality evaluation of the agarwood produced by Agar-Wit.
Analysis of Chemical Constituents of Essential Oil of Agarwood Produced by Whole-Tree Agarwood-Inducing Technique

Abstract:The essential oil was extracted from four batches of agarwood induced by whole-tree agarwoodinducing technique(Agar-Wit)through steam distillation method,and analyzed by gas chromatography-mass spectrometry(GC-MS).38Types of common constituents were identified from four batches of agarwood sample,and the total relative content was 93.59%,97.53%,91.37%and 95.62%,respectively.Moreover,the total relative content of 5types of characteristic constituents was 24.42%,34.77%,19.91%and 24.35%,respectively.The results showed that,while producing agarwood by Agar-Wit,the induced agarwood after 12 months would be close to the contrast agarwood in the total relative contents of 38types of volatile constituents and 5 types of characteristic constituents,and the induced agarwood after 15months would surpass the contrast agarwood.
The Characteristic Fragrant Sesquiterpenes and 2-(2-Phenylethyl)chromones in Wild and Cultivated “Qi-Nan” Agarwood

Abstract:Recently, cultivated “Qi-Nan” (CQN) agarwood has emerged as a new high-quality agarwood in the agarwood market owing to its similar characteristics, such as high content of resin and richness in two 2-(2-phenylethyl)chromone derivatives, 2-(2-phenylethyl)chromone (59) and 2-[2-(4-methoxyphenyl)ethyl]chromone (60), to the wild harvested “Qi-Nan” (WQN) agarwood. In this study, we compared the chemical constituents and fragrant components of two types of WQN agarwood from A. agallocha Roxb. and A. sinensis, respectively, with CQN agarwood and ordinary agarwood varieties. Additionally, we analyzed different samples of WQN agarwood and CQN agarwood by GC-MS, which revealed several noteworthy differences between WQN and CQN agarwood. The chemical diversity of WQN was greater than that of CQN agarwood. The content of (59) and (60) was higher in CQN agarwood than in WQN agarwood. For the sesquiterpenes, the richness and diversity of sesquiterpenes in WQN agarwood, particularly guaiane and agarofuran sesquiterpenes, were higher than those in CQN. Moreover, guaiane-furans sesquiterpenes were only detected by GC-MS in WQN agarwood of A. sinensis and could be a chemical marker for the WQN agarwood of A. sinensis. In addition, we summarized the odor descriptions of the constituents and established the correlation of scents and chemical constituents in the agarwood.

Abstract:As a traditional medicinal herb and valuable natural spice in China, Aquilaria sinensis (Lour.) Gilg has many significant pharmacological effects. Agarwood is the resinous heartwood acquired from wounded A. sinensis trees, and is widely used in pharmaceuticals owing to its excellent medicinal value. In this study, the chemical composition of volatile components and alcohol extracts from different organs of A. sinensis and agarwoods grown in different regions were investigated using GC-MS. The results showed that Vietnam agarwood had the highest moisture content, which was attributed to the local climate, while the fruit and bark of A. sinensis had higher moisture contents than the other organs. The volatile components of A. sinensis organs included 3-ethyl-5-(2-ethylbutyl)-octadecane, oleic acid 3-(octadecyloxy) propyl ester, and docosanoic acid 1,2,3-propanetriyl ester, while the alcohol extracts of A. sinensis organs contained benzoic acid ethyl ester, hexadecanoic acid ethyl ester, oleic acid, and n-hexadecanoic acid. Furthermore, the main active ingredients in agarwood from different habitats were sesquiterpenoids, aromatic species, and chromone compounds. The role of chromone compound 2-phenylethyl-benzopyran as an elicitor and the mechanism of agarwood formation were also investigated. Antioxidant tests showed that essential oils from agarwood and A. sinensis had antioxidant capacities by comparison with butylated hydroxytoluene and vitamin E. An antibacterial activity test showed that the inhibition effect of the essential oil was better against Gram-positive bacteria than against Gram-negative bacteria.

Writing histories of sexuality in the Middle East.

Abstract:The study of histories of sexuality in the region that today we term the Middle East has only recently become a domain populated by historians, and much inno vative work continues to be accomplished by literary scholars, social scientists, and, recently, art historians. This interdisciplinarity results in part from the fact that our primary sources confound modern disciplinary taxonomies—we encounter chroni cles as literature, poetry as political satire, belles lettres as social commentary, painted images more sexually graphic than the written word—so that silences in one genre or field of study may be vocalized in another. Because literary texts, particularly of the medieval and early modern periods, openly engaged sexual matters, literary and philological studies preceded the more purely historical. The buildup to histories of sexuality was gradual. The opening of the rich Ottoman archives following World War II facilitated social history and so cially oriented political history (Iranian studies lacks a comparable archive). In the 1980s, women's and then gender history was a significant stimulus to exploration of social cultures, slavery, gender and politics, spatial configurations of gender, and so on. Important publications of the early 1990s focused specifically on the history of sexualities in the region, and since then the field has made steady if not spectacular progress. Some preliminary observations may help to advance this forum's goal of facil itating comparative and transnational histories. First, the term "Middle East" is not a wholly accurate label for the region covered here, and its narrowness masks the rich potential for transnational history-writing that the actual history of the region invites. Abstract:The volatile compositions of the Agarwoods from different production places were extracted by solid phase microextraction(SPME),and isolated and identified by GC-MS,then the quality of Agarwood was evaluated.The investigation showed that the Agarwoods from different places were all consisted of sesquiterpenoids and aromatic compounds,but their chemical compositions and contents were quite different.The six characteristic components,including 4-phenyl-2-butanone,p-methoxy-benzylacetone,agarospirol,agarofuran,hinesol and baimuxinal,and their contents were used as the evaluation indexes for the quality of the Agarwoods.The results indicated that the contents of characteristic constituents in Laos Agarwood were the highest(about 20％),the contents in the Cambodian Agarwood were the second(17％),while the contents in the Chinese Agarwoods(Hainan and Dongguan) were the lowest.Therefore,the Laos Agarwood had the best quality,the Cambodian Agarwood was slightly poorer,and they both were superior to the Chinese Agarwood.The method showed the advantages of low sample consumption and strong characteristic.

Abstract:The main chemical constituents of agarwood are sesquiterpenes and chromones, which can be divided into different categories depending on their molecular skeletons. Agarwoods are obtained from different plant species: Aquilaria sinensis, A. malaccensis, A. crassna, and A. subintegra. In this review, we systematically summarized the structures of 367 compounds isolated from agarwoods originating from four main species. We structurally classified all the components into 11 different types and summarized the number of compounds in each type. Different and identical components were obtained by enumerating the chemical compositions of the different species. Knowledge regarding the chemical constituents of agarwoods of different species will aid understanding of the chemical compositions of agarwoods and will subsequently identify similar compounds that can serve as standards for quality control to provide a reference for future studies on agarwoods from different species and to increase their usefulness.

Abstract:Agarwood, the resinous wood of the genus Aquilaria Lam. and Gyrinops Gaertn. of the family Thymelaeaceae, is a very valuable traditional medicinal material and spice. Due to the high economic value of agarwood, unscrupulous merchants have led to the prevalence of fake agarwood in the trade market in pursuit of high profits. Therefore, it is crucial to explore the characteristics of agarwood and establish a simple and rapid method to identify authenticity. This study compared the authenticity of the microstructure of agarwood from different producing areas with that of artificially simulated fake agarwood. The included phloem of authentic agarwood contains brown to brownish resin. And these resins were naturally and stably distributed. The fake agarwood is divided into two categories: one itself is non-agarwood wood (without included phloem), and the other is artificially counterfeiting. However, the filling of resin in the included phloem of agarwood did not occur after artificially simulated counterfeiting treatments. Of the 18 commercially available samples, 10 had the same microstructure as agarwood, but six of them were not completely filled with resin in the included phloem. Therefore, the resinous included phloem is a key characteristic structure of agarwood. It can be used as a basis for authenticity identification of agarwood. This will provide a convenient and rapid method for promoting and popularizing agarwood authentication in trade, customs enforcement, CITES management, and other fields.

Abstract:Aquilaria, renowned for its agarwood, and valued for its aromatic wood and rich resin, finds use in cosmetics, fragrances, incense, and medicine. Identifying the agarwood-producing species among 21 species of Aquilaria is challenging. This study analyzes chemical compounds in agarwood oil from 4 Aquilaria species: Aquilaria beccariana, Aquilaria crassna, Aquilaria malaccensis, and Aquilaria subintegra using gas chromatography-flame ionization detector (GC-FID). Statistical analysis explores compound abundance, employing methods like mean and Z-score tests. This analysis summarizes those 14 compounds that are consistently present based on zero and non-zero observations, and the Z-score test highlights five chemical compounds, with three compounds appearing in both analyses. These compounds can serve as a reference for future studies on Aquilaria species and agarwood oil, enhancing classification efforts.
