Abstract:Summary Sirolimus is a potent immunosuppressive agent with a novel mechanism of action. It inhibits the transduction of cytokine signals necessary for the proliferation and maturation of T cells. Because sirolimus blocks a broad spectrum of cytokine signals, it seems logical to use it as an adjunct to CsA-based immunosuppression. The high degree of synergy between these two agents, as suggested by the rigorous median-effect analysis, has been confirmed by a reduced rate of rejection episodes among human renal allograft recipients. However, Phase I studies document wide interindividual variation in the pharmacokinetic parameters of 26 stable renal transplant patients, thereby suggesting that optimal therapy may require monitoring of drug concentrations, which is a task that has been somewhat simplified by the good correlation of trough level to AUC. Development of a monoclonal antibody assay system may simplify the monitoring of drug concentrations further. Additional studies of sirolimus will be required to determine the therapeutic concentrations and ratios of sirolimus to CsA that provide optimal immunosuppression, and to assess the possibility of a steroid-free regimen.
Construction of electrochemical immunosensor from MXene/multi-walled carbon nanotubes/gold nanoparticles for specific detection of carcinoembryonic antigen

Abstract:With the advancement of nanotechnology, various types of nanomaterials have been integrated into electrochemical immunoelectrodes to enhance their performance. Among these, MXene stands out as a promising candidate due to its high electron transfer capacity and abundant surface chemical groups. However, the improvement in electrode performance is often hindered by the self-restacking and agglomeration of MXene. To address this issue, multi-walled carbon nanotubes (MWCNTs) were selected to form composites with MXene. Subsequently, a label-free immunosensor, BSA/Ab/AuNPs/MXene-MWCNTs-Nafion/ITO, was fabricated for specific detection of carcinoembryonic antigen (CEA), a widely used tumor marker. The results demonstrated that the incorporation of MWCNTs can effectively prevent the self-stacking of MXene. Moreover, the composites enhanced the loading of gold nanoparticles (AuNPs) to connect the antibodies, thereby improving electronic transmission signals and sensitivity. The sensor exhibited excellent analytical performance towards CEA with a wide linear range (0.050 to 200 ng mL-1) and a low limit of detection of 0.015 ng mL-1 (S/N = 3). The possibility of it being applied in clinical trials was verified by using ELISA and differential pulse voltammetry (DPV) assays to detect CEA in serum samples. The recoveries ranged from 95.34 to 102.09% with relative standard deviations (RSDs) below 5.00%. Furthermore, the sensor displayed satisfactory selectivity, repeatability, and stability. We hope the findings highlight promising prospects for advanced immunosensor development and alternative strategies in cancer diagnosis.
Increased electrocatalyzed performance through hairpin oligonucleotide aptamer-functionalized gold nanorods labels and graphene-streptavidin nanomatrix: Highly selective and sensitive electrochemical biosensor of carcinoembryonic antigen

Abstract:We report a triplex signal amplification strategy for sensitive biosensing of cancer biomarker by taking advantage of hairpin-shaped oligonucleotide-functionalized gold nanorods (HO-GNRs), graphene and the avidin-biotin reation. The strategy expands electrochemical detection of carcinoembryonic antigen (CEA) by using an aptamer as biosensor's recognition element and HO-GNRs as signal enhancer. To construct this biosensor, the GNR was used as a carrier of horseradish peroxidase (HRP) and HO aptamer with a biotin at the 3'-end and a thiol at the 5'-end, which amplified the electrochemical response because of a large molar ratio of HRP to HO. In the presence of target CEA, the binding reactions of CEA with the loop portions of the HOs caused HOs' loop-stem structure opened and exposed the biotins, and then HRP-GNRs-HO conjugates were captured on graphene and streptavidin modified electrodes via the reaction between the exposed biotins and preimmobilized streptavidins. The accumulation of HRP effectively catalyzed the hydrogen peroxide-mediated oxidation of o-phenylenediamine to generate an electrochemical reduction current for CEA detection. Under optimal conditions, the electrochemical biosensor exhibited a wide dynamic range of 5pgmL(-1) and 50ngmL(-1) toward CEA standards with a low detection limit of 1.5pgmL(-1) (signal-to-noise ratio of 3). The proposed biosensor accurately detected CEA concentration in 8 human serum samples from patients with lung diseases, showing excellent correlations with standard chemiluminescence immunoassay. Furthermore, these results of target DNA detection made it abundantly clear that the proposed strategy can also be extended for detection of other relative biomarkers using different functional DNA structures, which shows great prospects in single-nucleotide polymorphisms analysis, biomedical sensing and application for accurate clinical diseases diagnostic.
A Novel Label-Free Microfluidic Paper-Based Immunosensor for Highly Sensitive Electrochemical Detection of Carcinoembryonic Antigen.

Abstract:In this work, a highly sensitive label-free paper-based electrochemical immunosensor employing screen-printed working electrode (SPWE) for detection of carcinoembryonic antigen (CEA) was fabricated. In order to raise the detection sensitivity and immobilize anti-CEA, amino functional graphene (NH2-G)/thionine (Thi)/gold nanoparticles (AuNPs) nanocomposites were synthesized and coated on SPWE. The principle of the immunosensor determination was based on the fact that the decreased response currents of Thi were proportional to the concentrations of corresponding antigens due to the formation of antibody–antigen immunocomplex. Experimental results revealed that the immunoassay enabled the determination of standard CEA solutions with linear working ranges of 50pgmL−1 to 500ngmL−1, the limit of detections for CEA is 10pgmL−1 (S/N=3) and its corresponding correlation coefficients were 0.996. Furthermore, the proposed immunosensor could be used for the determination of clinical serum samples. A large number of clinical serum samples were detected and the relative errors between measured values and reference concentrations were calculated. Results showed that this novel paper-based electrochemical immunosensor could provide a new platform for low cost, sensitive, specific, and point-of-care diagnosis in cancer detection.
Gold Nanoparticle/dna/methylene Blue Nanocomposites for the Ultrasensitive Electrochemical Detection of Carcinoembryonic Antigen

Abstract:A new signal amplification strategy employing nanocomposites composed of gold nanoparticles (GNPs) with DNA and methylene blue (MB) as labels has been developed to construct a novel immunosensor for the highly sensitive bioanalysis of carcinoembryonic antigen (CEA). To fabricate the label, DNA was first linked to GNPs through the formation of an Au–S bond. Next, the resulting bioconjugates were sequentially bound with MB via the guanine (G) bases in DNA specifically and then with secondary CEA antibody (Ab2) via the crosslinking of glutaraldehyde (GA). The receptor–analyte complex is formed by the biorecognition of the as-obtained MB labeled Ab2 with the primary CEA antibody (Ab1) immobilized on the surface of a modified GNP/Chitosan (Chits) composite electrode. After the receptor–analyte complex has formed, square wave voltammetry (SWV) was employed to oxidize the labeled MB in order to detect the CEA. This immunosensor shows a linear response from 0.10 to 2.0pgmL−1 CEA with an improved detection limit of 0.05pgmL−1 as compared to the conventional immunoassay. In addition, this new protocol shows acceptable stability, reproducibility and good recovery (97.5–110.0%) for CEA in human serum with great potential for clinical applications.

Abstract:In order to detect carcinoembryonic antigen (CEA) as a tumor marker in lung cancer for early cancer diagnosis, this study aimed to develop a label-free electrochemical immunosensor based on the immobilization of an Anti-CEA antibody on a metal-organic framework (MOF)-graphene oxide nanocomposite modified glassy carbon electrode (Anti-CEA/Ag-MOF/GO/GCE). Ag-MOF/GO nanocomposite was prepared on the GCE surface using the ultrasonic irradiation method, and Anti-CEA antibody was subsequently immobilized on the surface. Analysis of the crystal structure and morphology of the modified electrode using FE-SEM and XRD revealed that the correct combination of GO nanosheets and Ag-MOF nanoparticles produced a high surface area to trap the antibodies. Electrochemical tests utilizing the CV and DPV methods revealed that the immunosensor's sensitivity, stability, and selectivity were improved by Anti-CEA/Ag-MOF/GO/GCE. Results showed that, with a detection limit of 0.005 ng/mL, the change in the reduction peak current was inversely correlated with the logarithm concentration of CEA in the range of 10-3 to 5000 ng/mL. The suggested CEA immunosensor's applicability in a human serum sample was investigated, and findings of analytical studies via standard addition technique for both ELISA and DPV assays revealed that significant agreement existed between the outcomes of the two assays. Additionally, the recoveries ranged from 99.00% to 99.25%, and all relative standard deviations (RSDs) for the sample detections were below 5.01%, indicating satisfactory accuracy in results measured with the proposed CEA immunosensor, indicating that the prepared CEA immunosensor in this study can be used in clinical applications and human fluids.

Abstract:This study involves the fabrication of a sandwich-type electrochemical immunosensor for sensitive and simultaneous detection of alpha-fetoprotein (AFP) and cancer embryo antigen (CEA). Gold nanoparticle (Au NP)–modified ultrathin metal–organic framework nanosheets (UMOFNs) were prepared (denoted as Cu-UMOFNs@Au NPs and Co-UMOFNs@Au NPs). Furthermore, the Cu-UMOFNs@Au NPs and Co-UMOFNs@Au NPs were used as redox probes to label different antibodies. Subsequently, any potential active sites that remained were blocked using glucose oxidase. Dual catalysis amplification was achieved using the prepared MOF-derived materials via in situ catalysis of glucose to produce H 2 O 2 in the presence of glucose oxidase. H 2 O 2 was further catalyzed by Cu-UMOFNs@Au NP- and Co-UMOFNs@Au NP-derived materials to obtain better signal responses. The results revealed that the linear range of the proposed immunosensor for the simultaneous detection of AFP was 0.001–100 ng mL −1 (detection limit, 0.28 pg mL −1 ; S/N = 3) and that for the simultaneous detection of CEA was 0.001–80 ng mL −1 (detection limit, 0.31 pg mL −1 ; S/N = 3). Notably, the proposed method promises rapid, simple, and cost-effective analysis of biological samples.

Electrochemical Stripping Analysis of Nanogold Label-Induced Silver Deposition for Ultrasensitive Multiplexed Detection of Tumor Markers.

Abstract:A multiplexed electrochemical immunoassay method was developed for simultaneous ultrasensitive measurement of tumor markers based on electrochemical stripping analysis of silver nanoparticles (Ag NPs). The Ag NPs were deposited on a disposable immunosensor array with a reduction reaction catalyzed by nanogold labels. The immunosensor array was prepared by covalently immobilizing capture antibodies on chitosan modified screen-printed carbon electrodes. Through a sandwich-type immunoreaction, antibody-functionalized Au NPs were captured onto immunosensor surface to induce the silver deposition from a silver enhancer solution. The deposited Ag NPs could be directly measured by anodic stripping analysis in KCl solution. The catalytic deposition enhanced the analytical sensitivity for detection of protein markers. The interference of dissolved oxygen could be avoided as the detection was performed with positive stripping potential range. Using carcinoembryonic antigen and α-fetoprotein as model analytes, the proposed multiplexed immunoassay method showed wide linear ranges of three orders of magnitude with the detection limits down to 3.5 and 3.9pgmL−1, respectively. The localized silver deposition, as well as the stripping detection process, eliminated completely the electrochemical cross talk between adjacent immunosensors. The immunosensor array exhibited acceptable reproducibility, stability and accuracy, showing a promising potential in multianalyte determination for clinical application.
Ultrasensitive non enzymatic multiple immunosensor for tumor markers detection by coupling DNA hybridization chain reaction with intercalated molecules

Abstract:In this study, we tried coupling the small signal molecules that could intercalate into DNA double helix with hybridization chain reaction (HCR) technique to fabricate a multiple immunosensor. Doxorubicin hydrochloride (DXH) and methylene blue (MB) were used as signal molecules and alpha-fetoprotein (AFP) and carcinoembryonic antigen (CEA) were selected as model biomarkers. The immunosensor mainly consists of three parts as follows: First, two different primary antibodies (Ab1) immobilized on the surface of gold nanoparticles (Au NPs); Second, secondary antibodies (Ab2) conjugated with DNA primer; Third, long DNA concatemers from HCR were used as a carrier to intercalate amounts of signal molecules (DXH or MB). A sandwich immunocomplex was formed among primary antibodies, target biomarkers and secondary antibodies conjugated with DNA primer via specific recognition reaction. Afterwards, DNA concatemers intercalating amounts of DXH or MB were linked to DNA primer via DNA hybridization. Square wave voltammetry (SWV) was employed to record the response signals from electroactive molecules DXH and MB, and two distinguishable signals were obtained, which peak potentials were at about -0.30V (corresponding to MB) and -0.70V (corresponding to DXH, both vs SCE), respectively. The signal intensities of MB and DXH were linearly related to the logarithm of biomarkers concentration in the range of 0.05pgmL-1-25ngmL-1, and the limit of detection were 0.03pgmL-1 for CEA and 0.02pgmL-1 for AFP (at S/N=3), respectively. Furthermore, the immunosensor exhibited a sensitive electrochemical response to biomarkers in human serum samples and the results obtained were in accordance with reference method, indicating the immunosensor can be applied to real sample analysis in clinic diagnosis.

Abstract:An ultrasensitive enzyme-free electrochemical immunoassay was developed for detection of the fg/mL level carcinoembryonic antigen (CEA) by using a double strand DNA@Au nanoparticle (dsDNA@AuNP) tag and hexaammineruthenium(III) chloride (RuHex) as the electroactive indicator. The dsDNA@AuNP was synthesized by one-pot hybrid polymerization of dsDNA on initiator DNA modified AuNPs via hybridization chain reaction. The immunosensor was prepared by covalently cross-linking capture antibody on chitosan/AuNP nanocomposite modified glass carbon electrode. The AuNPs accelerated the electron transfer and led to high detection sensitivity. With a sandwich-type immunoreaction and a biotin-streptavidin affinity reaction, the dsDNA@AuNP tag was conjugated on the immunocomplex to bring a high amount of RuHex to the electrode surface via electrostatic interaction, resulting in an amplified electrochemical signal. Under optimal conditions, the proposed sensing platform showed a wide linear detection range from 10 fg/mL to 10 ng/mL along with a detection limit of 3.2 fg/mL for CEA. The immunosensor exhibited high sensitivity and good stability, showing a promising application in early cancer diagnosis and could be extended to sensitive electrochemical biosensing of other analytes.
Highly Reusable Electrochemical Immunosensor for Ultrasensitive Protein Detection

Abstract:The detection and quantification of protein biomarkers in bodily fluids is important for many clinical applications, including disease diagnosis and health monitoring. Current techniques for ultrasensitive protein detection, such as enzyme-linked immunosorbent assay (ELISA) and electrochemical sensing, involve long incubation times (1.5-3 hr) and rely on single-use sensing electrodes which can be costly and generate excessive waste. This work demonstrates a reusable electrochemical immunosensor employing magnetic nanoparticles (MNPs) and dually labeled gold nanoparticles (AuNPs) for ultrasensitive measurements of protein biomarkers. As proof of concept, this platform was used to detect C-X-C motif chemokine ligand 9 (CXCL9), a biomarker associated with kidney transplant rejection, immune nephritis from checkpoint inhibitor therapy, and drug-associated acute interstitial nephritis, in human urine. The sensor successfully detected CXCL9 at concentrations as low as 27 pg/mL within ~1 hr. This immunosensor was also adapted onto a handheld smartphone-based diagnostic device and used for measurements of CXCL9, which exhibited a lower limit of detection of 65 pg/mL. Lastly, we demonstrate that the sensing electrodes can be reused for at least 100 measurements with a negligible loss in analytical performance, reducing the costs and waste associated with electrochemical sensing.
Colorimetric and photothermal immunosensor for sensitive detection of cancer biomarkers based on enzyme-mediated growth of gold nanostars on polydopamine

Abstract:Background Detecting cancer biomarker levels in body fluids is essential for medical diagnosis. Enzyme-linked immunosorbent assay (ELISA) has been broadly used to detect cancer biomarkers. However, colorimetric ELISA based solely on nanoparticles (NPs) are susceptible to environmental influences, which often results in the detection inaccuracy, being limited in clinical applications. In this regard, the dual-mode approach would add signal diversity to the detection, making the results more reliable. Results We present colorimetric and photothermal immunosensor that enables direct reading of the color and temperature of the solution. A core-satellite nanoprobe constructed by polydopamine (PDA) as the core and gold seeds as satellites is rationally designed as the signal reporter. When ascorbic acid is present in the solution, PDA can cooperate with ascorbic acid to reduce chloroauric acid and mediate the growth of gold seeds on the PDA surface, inducing a redshift of the localized surface plasmon resonance peak of the nanosensor and the change in photothermal conversion efficiency. The method is further combined with the sandwiched immunoassay to construct an alkaline phosphatase based colorimetric and photothermal ELISA for the highly sensitive and accurate evaluation and detection of prostate-specific antigen (PSA). The linear range was from 0.05 to 100 ng mL −1 with a detection limit of 6.71 pg mL −1 for the colorimetric detection, while the linear range was from 0.5 to 90 ng mL −1 with a detection limit of 0.13 ng mL −1 in the photothermal analysis. The accurate detection of PSA levels in serum samples was well demonstrated with the dual-mode approach. Significance The presented immunoassay allows straightforward, sensitive, and selective readout by color and temperature without advanced instrumentation. Particularly, the LOD was much lower than the threshold in clinical trials for PSA. Therefore, this method has a great prospect in the early diagnosis of cancer biomarkers based on a dual-mode multifunctional platform.

Abstract:Recently, electrochemiluminescent (ECL) immunosensors have received much attention in the field of biomarker detection. Here, a highly enhanced ECL immunosensing platform was designed for ultrasensitive detection of carcinoembryonic antigen (CEA). The surface of the glassy carbon electrode was enhanced by applying functional nanostructures such as thiolated graphene oxide (S-GO) and streptavidin-coated gold nanoparticles (SA-AuNPs). The selectivity and sensitivity of the designed immunosensor were improved by entrapping CEA biomolecules using a sandwich approach. Luminol/silver nanoparticles (Lu-SNPs) were applied as the main core of the signaling probe, which were then coated with streptavidin to provide overloading of the secondary antibody. The highly ECL signal enhancement was obtained due to the presence of horseradish peroxidase (HRP) in the signaling probe, in which the presence of H 2 O 2 further amplified the intensity of the signals. The engineered immunosensor presented excellent sensitivity for CEA detection, with limit of detection (LOD) and linear detection range (LDR) values of 58 fg mL −1 and 0.1 pg mL −1 to 5 pg mL −1 ( R 2 = 0.9944), respectively. Besides its sensitivity, the fabricated ECL immunosensor presented outstanding selectivity for the detection of CEA in the presence of various similar agents. Additionally, the developed immunosensor showed an appropriate repeatability (RSD 3.8%) and proper stability (2 weeks). Having indicated a robust performance in the real human serum with stated LOD and LDR, the engineered immunosensor can be considered for the detection and monitoring of CEA in the clinic.

Abstract:The fabrication and signal amplification play important roles in electrochemical biosensors to achieve high sensitivity and low cost. In this study, a sandwich-type electrochemical aptasensor for dual lung cancer biomarkers, neuron-specific enolase (NSE) and epidermal growth factor receptor (EGFR), was constructed based on black phosphorus–gold nanocomposites (BP–AuNCs) and aptamers. BP–AuNCs nanocomposites were synthesized by an in situ reduction process without any reducing agent and decorated on the surface of the detachable spherical working electrodes (WEs). The signal amplification was obtained by the good electrical conductivity of BP–AuNCs, as well as the large number of binding sites provided for the capture aptamers (Apt 1). Moreover, the addition of secondary aptamers (Apt 2) formed the sandwich-type aptasensor to obtain a dual signal amplification. The resulting signal by the differential pulse voltammetry (DPV) analysis in the solution containing ferroferricyanide redox probe (Fe(CN) $_{{6}}{)}^{\text {3-/4-}}$ is proportional to the concentration of the target biomarkers. The electrochemical aptasensor platform was simply fabricated on the cloth substrate by screen printing. Under the optimized conditions, the proposed aptasensor was used in the simultaneous detection of two lung cancer biomarkers, NSE and EGFR. Good linear detection was obtained in a range of 1–400 ng/mL (NSE) and 1–600 ng/mL (EGFR) with ultralow detection limits of 0.1 ng/mL (NSE) and 0.6 ng/mL (EGFR), respectively. Experimental results display good selectivity, stability, and accuracy, thus providing a promising low-cost platform in the preliminary estimates and diagnosis of lung cancer at early stages.

Abstract:Bioanalytical methods have experienced unprecedented growth in recent years, driven in large part by the need for faster, more sensitive, more portable ("point of care") systems to detect protein biomarkers for clinical diagnosis. Electrochemical detection strategies, used in conjunction with immunosensors, offer advantages because they are fast, simple, and low cost. Recent developments in electrochemical immunosensors have significantly improved the sensitivity needed to detect low concentrations of biomarkers present in early stages of cancer. Moreover, the coupling of electrochemical devices with nanomaterials, such as gold nanoparticles, carbon nanotubes, magnetic particles, and quantum dots, offers multiplexing capability for simultaneous measurements of multiple cancer biomarkers. This review will discuss recent advances in the development of electrochemical immunosensors for the next generation of cancer diagnostics, with an emphasis on opportunities for further improvement in cancer diagnostics and treatment monitoring. Details will be given for strategies to increase sensitivity through multilabel amplification, coupled with high densities of capture molecules on sensor surfaces. Such sensors are capable of detecting a wide range of protein quantities, from nanogram to femtogram (depending on the protein biomarkers of interest), in a single sample.

Abstract:The detection of tumor markers has significant importance in the early diagnosis of cancer. It is an important indicator for evaluating the extent of the disease and monitoring the efficacy of tumor treatment. Therefore, it is meaningful to find a convenient, fast, and sensitive method to detect tumor markers. Carbohydrate antigen 19–9 (CA19-9) is the first discovered carbohydrate antigen tumor marker with the highest sensitivity and specificity for pancreatic cancer. Carcinoembryonic antigen (CEA), as the most widely spectrum carbohydrate antigen tumor marker, can show an upward trend in many tumors. In this work, an electrochemical immunosensing platform was proposed for the detection of CEA and CA199. Nano-silicon sphere-cyclodextrin was synthesized for antibody capture. Graphene and gold nanoparticles were used to modify the electrode, improving the electronic conductivity of the electrode. The synthesized sensor can be used for highly sensitive detection of two tumor markers. This method also provides a new platform for clinical immune analysis of other biomolecules.
Multifunctional Gold Nano-Cytosensor With Quick Capture, Electrochemical Detection, and Non-Invasive Release of Circulating Tumor Cells for Early Cancer Treatment

Abstract:Circulating tumor cells (CTCs) are metastatic tumor cells that shed into the blood from solid primary tumors, and their existence significantly increases the risk of metastasis and recurrence. The timely discovery and detection of CTCs are of considerable importance for the early diagnosis and treatment of metastasis. However, the low number of CTCs hinders their detection. In the present study, an ultrasensitive electrochemical cytosensor for specific capture, quantitative detection, and noninvasive release of EpCAM-positive tumor cells was developed. The biosensor was manufactured using gold nanoparticles (AuNPs) to modify the electrode. Three types of AuNPs with controllable sizes and conjugated with a targeting molecule of monoclonal anti-EpCAM antibody were used in this study. Electrochemical impedance spectroscopy (EIS) and differential pulse voltammetry (DPV) of the cytosensors were performed to evaluate the cell capture efficiency and performance. The captured 4T1 cells by the AuNPs hindered electron transport efficiency, resulting in increased EIS responses. The cell capture response recorded using EIS or DPV indicated that the optimal AuNPs size should be 17 nm. The cell capture response changed linearly with the concentration range from 8.0 × 10 to 1 × 10 7 cells/mL, and the limit of detection was 50 cells/mL. After these measurements, glycine-HCl (Gly-HCl) was used as an antibody eluent to destroy the binding between antigen and antibody to release the captured tumor cells without compromising their viability for further clinical research. This protocol realizes rapid detection of CTCs with good stability, acceptable assay precision, significant fabrication reproducibility with a relative standard deviation of 2.09%, and good recovery of cells. Our results indicate that the proposed biosensor is promising for the early monitoring of CTCs and may help customize personalized treatment options.

Abstract:An electrochemical immunosensor was designed to quantify carcinoembryonic antigen (CEA), which is regarded asa tumour marker and prognostic indicator. The glassy carbon electrode (GCE) surface was modified using MoS2, graphene nanoflakes (GNs), fMWCNTs, and electrodeposited PdNPs for the first time to prepare a Pd-MoS2-GNs@fMWCNTs/GCE. The platform was activated with monoclonal anti-CEA. The prepared nanocomposite material helps in improving the surface area and conductivity of the GCE. Moreover, PdNPs act as a linker between nanocomposite materials and anti-CEA, which enhances electron transport. The procedure used to analyse CEA involve sequential addition of CEA into the solution of 5 mM [Fe(CN)6]3-/4- prepared in 0.1 M PBS (pH 7.4) in the presence of fabricated electrode and DPV and EIS methods were used as detection technique. The change in signal response for the [Fe(CN)6]3-/4- redox reaction before and after CEA interaction is considered as the immunosensor response which is directly correlated with CEA concentration. Further, in DPV, the signal inhibition approach was used to measure CEA by monitoring the decrease in oxidation peak current of [Fe(CN)6]3-/4- due to the formation of CEA/anti-CEA immunocomplex. The electrochemical immunosensor demonstrated low LOD of 2 pg/mL and a correlation coefficient of 0.98 within a broad linear range of CEA concentration from 0.002 to 500 ng/mL. The fabricated immunosensor displayed excellent selectivity, repeatability, and stability. Further, the proposed immunosensor effectively measured CEA levels in human blood serum, making it an effective tool for testing CEA in clinical samples.
