Abstract:C-H activation and functionalization are on the forefront of modern synthetic chemistry. Imagine if any C-H bond of a molecule could be converted to a C-X bond, where X is the target functionality-this would alter the synthetic blueprints for complex target molecules since it would provide novel disconnections in retrosynthetic analysis. Collaborations between many experimental and computational groups have led to rapid developments of new C-H functionalization methods. Our groups represent an example of this; we were brought together as part of the NSF-supported Center for Selective C-H Functionalization. Many examples of experimental-computational synergy for selective Pd(II)-catalyzed C-H activation of aryl and alkyl groups are described in this Account. We describe computations by the Houk group made in response to experimental stimuli by the Yu group. The first section discusses the experimental and computational investigations of oxazoline-directed stereoselective Pd(II)-catalyzed C(sp3)-H bond activation that occurs through the concerted metalation-deprotonation (CMD) pathway involving a monomeric Pd(II) complex. The second section involves two types of bidentate ligands, mono-N-protected amino acid (MPAA) and acetyl-protected aminoethyl quinoline (APAQ) ligands that promote the C-H activation reactions with the ligand as the internal base. In the MPAA-assisted remote C-H bond activation, the basic dianionic amidate ligand participates in the deprotonation of a specific C-H bond. This mechanism accounts for the improved reactivity and selectivity in C-H activation reactions with MPAA ligands. The chiral APAQ ligands enable asymmetric palladium insertion into prochiral C-H bonds on a single methylene carbon center. The dianionic amidate of the APAQ ligand acts as an intramolecular base to deprotonate the methylene C-H asymmetrically and facilitate chiral Pd-C bond formation. The origins of the dramatic differences of five-membered (relatively inactive) and six-membered (highly active) chelation in β-methylene C(sp3)-H activation reactions by a Pd(II) catalyst were explained with density functional theory (DFT) calculations. This is mainly due to the steric repulsions between the ArF group of the substrate and the quinoline group of the ligand. The steric repulsion between the ArF group of the substrate and the quinoline group of the APAQ ligand destabilizes the five-membered chelate transition structure, increasing the energy of the transition state. The third section discusses a mechanism involving a Pd-Ag heterodimeric complex intermediate in the template-directed, Pd(II)-catalyzed remote meta functionalization of toluene derivatives and benzoic acid derivatives. The nitrile directing group of the template coordinates with Ag while the Pd is placed adjacent to the meta-C-H bond in the transition state, leading to the observed high meta selectivity. The selective activation of remote meta-C-H bonds at various distances can be achieved by tuning the template. The dual role of AgOAc as both an oxidant and part of the heteronuclear active species in the mechanism involving PdAg(OAc)3 was determined by DFT calculations and is in accord with literature information about complexes. For the systems discussed in these three sections, the similarity is that they all proceed via the CMD mechanism. The differences lie in the proton acceptors and the active Pd species. Common CMD involves a monomeric Pd mechanism with acetate as the proton acceptor. Both MPAA and APAQ ligands react via monomeric Pd mechanisms with a ligand moiety (the amidate oxygen) as the proton acceptor. Nitrile-containing template-mediated meta-C-H activations proceed via a Pd-Ag heterodimeric mechanism, still with acetate as the proton acceptor. The interaction between our two groups, experts in experiment and computation, and the discoveries made possible by that interplay are highlighted in this Account.

Abstract:New carbene complex catalysts based on palladium chloride, including PEPPSI-complexes (PEPPSI = Pyridine-Enhanced Precatalyst Preparation Stabilization and Initiation), have been synthesized. These catalysts show high efficiency in Buchwald–Hartwig C–N cross-coupling between aryl chlorides/bromides and amines, including sterically shielded reagents, and high efficiency in the Suzuki–Miyaura C–C cross-coupling reaction, where their use results in the formation of several tetra-ortho-substituted biaryls at very low amounts of catalyst under mild conditions. Modification of the catalyst structure to the PEPPSI complex leads to an unexpected decrease in activity in the Buchwald–Hartwig amination, in contrast to the Suzuki–Miyaura reaction, in which the PEPPSI complex showed greater activity in some cases.

Abstract:In the past eight years, the selective cross-coupling of amides by N-C(O) bond activation has emerged as a highly attractive manifold for the manipulation of traditionally unreactive amide bonds. In this Special Issue on Next-Generation Cross-Coupling Chemistry, we report the Suzuki-Miyaura and Buchwald-Hartwig cross-coupling of amides by selective N-C(O) cleavage catalyzed by bench-stable, well-defined carboxylate Pd(II)-NHC (NHC = N-heterocyclic carbene) catalysts {[(NHC)Pd(O2CR)2]}. This class of Pd(II)-NHCs promotes cross-coupling under exceedingly mild room-temperature conditions owing to the facile dissociation of the carboxylate ligands to form the active complex. These readily accessible Pd(II)-NHC precatalysts show excellent functional group tolerance and are compatible with a broad range of amide activating groups. Considering the mild conditions for the cross-coupling and the facile access to carboxylate Pd(II)-NHC complexes, we anticipate that this class of bench-stable complexes will find wide application in the activation of amide N-C(O) and related acyl X-C(O) bonds.

Abstract:The development of more reactive, general, easily accessible, and readily available Pd(II)-NHC precatalysts remains a key challenge in homogeneous catalysis. In this study, we establish air-stable NHC-Pd(II) chloro-dimers, [Pd(NHC)(μ-Cl)Cl]2, as the most reactive Pd(II)-NHC catalysts developed to date. Most crucially, compared with [Pd(NHC)(allyl)Cl] complexes, replacement of the allyl throw-away ligand with chloride allows for a more facile activation step, while effectively preventing the formation of off-cycle [Pd2(μ-allyl)(μ-Cl)(NHC)2] products. The utility is demonstrated via broad compatibility with amide cross-coupling, Suzuki cross-coupling, and the direct, late-stage functionalization of pharmaceuticals. Computational studies provide key insight into the NHC-Pd(II) chloro-dimer activation pathway. A facile synthesis of NHC-Pd(II) chloro-dimers in one-pot from NHC salts is reported. Considering the tremendous utility of Pd-catalyzed cross-coupling reactions and the overwhelming success of [Pd(NHC)(allyl)Cl] precatalysts, we believe that NHC-Pd(II) chloro-dimers, [Pd(NHC)(μ-Cl)Cl]2, should be considered as go-to precatalysts of choice in cross-coupling processes.
Palladium complexes containing NNN pincer type ligands and their activities in Suzuki-Miyaura cross coupling reaction

Abstract:Five new NNN pincer-type ligands and their palladium complexes were successfully synthesised and characterised by FT-IR, 1H NMR, 13C NMR, and UV-vis analyses. TEM analysis was used to observe the morphological character of the black residues obtained from the fourth cycle of the reusability test. Furthermore, suitable crystals of the N2,N6-bis(2-tert-butylphenyl)pyridine-2,6-dicarboxamide and its palladium complex were elucidated with the X-ray single crystal diffraction method. Both the ligand and its palladium complex crystallise in a monoclinic system with space group P21/c for the H2L4 and C2/c for the palladium complex. The structure of the pincer ligand and its palladium complex were stabilised by intramolecular and intermolecular C-H⋅⋅⋅O, C-H⋅⋅⋅N, and N-H⋅⋅⋅N contacts. A Suzuki-Miyaura cross-coupling reaction between aryl halides and phenylboronic acid was used to assess the catalytic abilities of the palladium pincer complexes. All of the prepared complexes exhibited considerable catalytic activity. However, complexes 4 (Acetonitrile-N2,N6-bis(2-tert-butylphenyl)pyridine-2,6-dicarboxamidopalladium(II)) and 5 (Acetonitrile-N2,N6-bis(2-nitrophenyl)pyridine-2,6-dicarboxamidopalladium(II)) provided almost 100% conversion with nearly 100% yield in the reaction between 4-bromotoluene and phenylboronic acid. Furthermore, these active complexes catalysed the reaction of the sterically hindered and deactivated substrates (1-Bromo-4-izobutylbenzene and 2-bromo-6-methoxynaphthalene) with phenylboronic acid, and complete conversion and yields up to 100% were achieved in a short time with the 2-bromo-6-methoxynaphthalene.

Abstract:Air-stable palladium (0) precatalysts are advantageous for facilitating a variety of chemical transformations, and are desir-able precursors for high-throughput experimentation studies. We report investigations into air-stable Pd(0) precatalysts stabi-lized by dimethyl fumarate (DMFU) as an electron-deficient alkene. A Pd(0) DMFU complex with a diazabutadiene (DAB) supporting ligand readily undergoes substitution with both monodentate and bidentate phosphines to form phosphine–Pd–DMFU complexes in situ. These complexes undergo oxidative addition with ArBr substrates, and are also effective precata-lysts for Heck coupling, Suzuki-Miyaura coupling, and Miyaura borylation. Catalytic comparisons of the DAB–Pd–DMFU precursor to other Pd sources reveals benefits and limitations of this system, including high activity in Heck coupling, and challenges with in situ catalyst generation

Abstract:We report the synthesis, characterization and reactivity of an air-stable, well-defined acenaphthoimidazolylidene palladium-BIAN-NHC chloro dimer complex, [Pd(BIAN-IPr)(μ-Cl)Cl]2. This rapidly activating catalyst merges the reactive properties of palladium chloro dimers, [Pd(NHC)(μ-Cl)Cl]2, with the attractive structural features of the BIAN framework. [Pd(BIAN-IPr)(μ-Cl)Cl]2 is the most reactive Pd(II)-NHC precatalyst discovered to date undergoing fast activation under both an inert atmosphere and aerobic conditions. The catalyst features bulky-yet-flexible sterics that render the C-H substituents closer to the metal center in combination with rapid dissociation to monomers and strong σ-donor properties. [Pd(BIAN-IPr)(μ-Cl)Cl]2 should be considered as a catalyst for reactions using well-defined Pd(II)-NHCs.

Abstract:We report the Pd-catalyzed Suzuki–Miyaura cross-coupling of aliphatic amides. Although tremendous advances have been made in the cross-coupling of aromatic amides, C–C bond formation from aliphatic amides by selective N–C(O) cleavage has remained a major challenge. This longstanding problem in Pd catalysis has been addressed herein by a combination of (1) the discovery of N,N-pym/Boc amides as a class of readily accessible amide-based reagents for cross-coupling and (2) steric tuning of well-defined Pd(II)-NHC catalysts for cross-coupling. The methodology is effective for the cross-coupling of an array of 3°, 2°, and 1° aliphatic amide derivatives. The catalyst system is user-friendly, since the catalysts are readily available and are air- and bench-stable. Mechanistic studies strongly support an amide bond twist and external nN → π*C═O/Ar delocalization as a unified enabling feature of N,N-pym/Boc amides in selective N–C(O) bond activation. Abstract:We report a combined experimental and computational investigation of the Suzuki–Miyaura cross‐coupling of amides enabled by [Pd(NHC)(allyl)Cl] precatalysts. Most crucially, mechanistic details pertaining to the Pd0/NHC catalytic cycle were elucidated by computational methods. Mechanistic insights shed light on the role of each ligand about the metal. Sterics play a key role in the initial activation of the catalyst. As a key insight, we have shown that water participates in the activation of the Pd‐NHC catalytic system. Easier activation has led to effect room temperature cross‐coupling of a broad range of amides through selective N−C bond scission under the mildest conditions reported to date. The use of sterically hindered [Pd(IPr*)(cin)Cl] reported herein for the first time in the amide cross‐coupling indicates that increasing flexible steric bulk of the isopropyl wingtip groups of the NHC ligand provides a modular scaffold for promoting amide cross‐coupling in high yields. The precatalytic pathway involving both NHC ligands as well as the catalytic cycle beginning from the Pd0 species are discussed. The mechanistic details provide insight into the amide bond twist (distortion) that leads to N−C cross‐coupling reactions and is required for the efficient N−C bond activation. Under the microscope: A mechanistic study of the palladium‐catalyzed Suzuki–Miyaura cross‐coupling of amides is described.

Abstract:A new series of calix[4]arene supported N-heterocyclic carbene palladium complexes were developed and fully characterized. A mono-substituted calix[4]arene was prepared through conventional procedures, following with the attachment of imidazolyl derivative groups to compose the precursors of novel NHCs ligands. Undergoing the alkylation with n-butylbromide and corresponding metallation with palladium and pyridine, original complexes were obtained. After a full characterization in solution and solid state, the evaluation of catalytic activity was taken out in Suzuki–Miyaura cross-coupling reactions, which revealed good performances.

Abstract:An amide functionlized bidentate NHC-Pd complex was successfully prepared by a facile solvothermal methodology from a bridged bis-benzimidazolium chloride and K2PdCl4. The new bidentate NHC-Pd complex has been effectively applied as catalyst in Suzuki-Miyaura cross-coupling reactions of aryl halides with arylboronic acids in distilled water (yields up to 99.0%). The bridged bis-benzimidazolium ligand from the Pd complex, bearing the amide functional groups on the nitrogen atom, showed to distinctly influence the electronic and steric effects on the catalytic process. The potential of application of the new NHC-Pd catalyst in diverse CC bond forming processes, working under environmentally friendly conditions, is further outlined.

Abstract:A series of novel pyridine stabilized Pd–NHC complexes were developed, which revealed high activities and broad substrates tolerance in the amination of various (hetero)-aryl chlorides. Besides various secondary amines, a wide range of primary anilines and aliphatic amines were also well tolerated. The results highlight us a new strategy to increase catalyst activity in the future catalyst design by alternating the σ-donor property and flexibility of NHC ligands.

Abstract:N,N′-Substituted di-isopropyl (NHC-1), benzyl-isopropyl (NHC-2), and adamantyl-isopropyl (NHC-3) benzimidazolium salts react with palladium(II) bromide in pyridine to afford the corresponding trans-dibromidopyridinepalladium(II) complexes Pd-C1, Pd-C2, and Pd-C3 in high yields. A distorted square planar geometry for Pd-C2 and Pd-C3 was confirmed by single-crystal X-ray diffraction. The palladium(II) complexes show a remarkably higher catalytic activity and selectivity, compared to the literature data, in carbonylative Sonogashira coupling reactions of aryl iodides and aryl diiodides with aryl alkynes, alkyl alkynes, and dialkynes. Excellent yields with as low as 0.03 mol % loading of the catalyst were obtained. In the series of benzimidazolium (NHC) precursors, the 1H NMR signals of the α hydrogen show a consistent probing of the N-substituent donor strength. The density functional theory (DFT) quantum mechanical descriptors of the frontier orbitals were calculated. A linear correlation of the calculated absolute softness of the complexes versus the calculated percent buried volume (%Vbur) of their corresponding ligands was obtained. The catalytic activity experimental data are consistent with the hard soft acid base (HSAB)-predicted high affinity of the softest Pd-C3 complex for soft substrates, such as aryl iodides.This article has not yet been cited by other publications.

Abstract:In this report, we have synthesized two NCS pincer ligands by the Schiff base reaction of 3‐((phenylthio)methoxy)benzaldehyde (P) with alkyl amines (tbutylamine (L1) and 1‐adamantylamine (L2)). The palladium pincer complexes (tbutylamine = C1 and 1‐adamantylamine = C2) of these ligands were synthesized by their reaction with PdCl2(CH3CN)2. The newly synthesized ligands and complexes were characterized using various techniques such as 1H, 13C{1H} Nuclear Magnetic Resonance (NMR), Ultraviolet–visible (UV‐Visible), Fourier Transform Infrared (FTIR) Spectroscopy, and High‐Resolution Mass Spectrometry (HRMS). The structure of ligand and its coordination mode with palladium precursor were studied with the help of single‐crystal X‐ray diffraction. The complexes showed distorted square planar geometry around the palladium center. The palladium pincer complexes were used as catalysts for the regioselective cross‐dehydrogenative alkenation of 2‐arylthiophene derivatives. The complex C2, where sterically bulky adamantyl ligand is part of the side arm showed a higher yield of alkenation reaction. Only 2.5 mol% catalyst loading was sufficient to achieve 74‐95% yields of desired products with excellent functional group tolerance under mild reaction conditions. The poisoning experiments (PPh3 and Hg) showed the homogeneous nature of the catalytic process. The plausible mechanism of the reaction was proposed based on the control experiments and time‐dependent HRMS studies.

Efficient Heterogeneous Palladium Catalysts in Oxidative Cascade Reactions

Abstract:Palladium-catalyzed oxidations involving cascade processes provide a versatile platform for streamlined conversion of simple feedstocks into functional molecules with high atom and step economy. However, the achievement of high palladium efficiency and selectivity in Pd-catalyzed oxidative cascade reactions is still challenging in many cases, as a result of the aggregation of active palladium species to Pd black and the possible side reactions during each bond-forming step. The two current solutions for addressing these issues are either to utilize oxidant-stable ligands or to use electron transfer mediators (ETMs). The former solution, which includes the use of amines, pyridines, sulfoxides, and carbene derivatives, inhibits aggregation of Pd0 during the catalytic cycle, while the latter solution facilitates reoxidation of Pd0 to PdII to improve the activity and selectivity. Following our long-standing interest in Pd-catalyzed oxidations, very recently we developed heterogeneous catalysts to resolve the issues mentioned above in oxidative cascade reactions. The heterogeneous palladium catalysts (Pd-AmP-MCF or Pd-AmP-CNC) comprise palladium nanoclusters (1–2 nm) immobilized on amino-functionalized siliceous mesocellular foam (MCF) or on crystalline nanocellulose (CNC), exhibiting high activity, selectivity as well as excellent recycling ability.In this Account, we will discuss the synthesis and characterizations of the heterogeneous palladium catalysts, as well as their catalytic behaviors, and the mechanisms involved in their reactions. An important aspect of these catalysts in oxidation reactions is the generation of active Pd(II) species within the heterogeneous phase. Typical oxidative cascade reactions of our recent research on this topic include oxidative carbocyclization-carbonylation, oxidative carbocyclization-borylation, oxidative alkynylation-cyclization, oxidative carbonylation-cyclization, and oxidative carbocyclization-alkynylation. These reactions provide access to important compounds attractive in medicinal chemistry and functional materials, such as γ-lactone/γ-lactam-based poly rings, cyclobutenols, highly substituted furans, and oxaboroles. During these processes, the heterogeneous catalysts exhibited much higher turnover numbers (TONs) than their homogeneous counterparts (e.g., Pd(OAc)2) as well as unique selectivity that cannot be achieved by homogeneous palladium catalysts. The origin of the high efficiency and unique selectivity of the heterogeneous catalysts was also investigated. Asymmetric syntheses for the construction of optically pure compounds were realized based on the excellent selectivity in these heterogeneous processes. Kinetic studies revealed that the rate and yield of the reactions were essentially maintained during recycling, which demonstrates that Pd-AmP-MCF and Pd-AmP-CNC are robust and highly active in these oxidative cascade reactions. In addition, inductively coupled plasma optical emisson spectroscopy (ICP-OES) analysis and hot filtration test suggest that these processes most likely proceed via a heterogeneous pathway.Recent progress in our group has shown that the activity of Pd-AmP-MCF and Pd-AmP-CNC could be improved even further by the addition of Ag+ to generate cationic Pd(II). Furthermore, intriguing solvent effects were observed in a Pd-AmP-MCF-catalyzed oxidative cascade process, and solvent-controlled chemoselective transformations were developed based on this property of the catalyst. The heterogeneous strategy of this Account provides solutions to palladium deactivation and selectivity issues in Pd(II)-catalyzed oxidative cascade reactions and enables efficient catalyst recycling, which will open up new opportunities in oxidative cascade reactions.This article has not yet been cited by other publications.

Abstract:NHC-based pincers (NHC = N-heterocyclic carbene) have been broadly employed as supporting platforms, and their palladium complexes have found many synthetic applications. However, previous studies mainly focused on the NHC pincers of palladium featuring an oxidation number of +II. In contrast, oxidation of these well-defined Pd( II ) species and the study of their fundamental high-valent Pd chemistry remain largely undeveloped. In addition, from a perspective of Pd II /Pd IV catalysis, the reactivity and degradation of NHC pincers in catalytically relevant reactions have not been well understood. In this work, a series of Pd( II ) complexes supported by a well-known NHC^Aryl^NHC pincer platform have been prepared. Their reactivity towards various oxidizing reagents, including halogen surrogates, electrophilic fluorine reagents, and alkyl/aryl halides, has been examined. In some cases, ambient-characterizable high-valent Pd NHCs, which have been scarcely reported, were obtained. The carbenes incorporated into the pincer framework proved to be effective spectator donors. In contrast, the central aryl moiety exhibits versatile reactivity and collapse pathways, allowing it to function either as a spectator or a non-innocent actor.

"Homeopathic" palladium nanoparticle catalysis of cross carbon-carbon coupling reactions

Abstract:Catalysis by palladium derivatives is now one of the most important tools in organic synthesis. Whether researchers design palladium nanoparticles (NPs) or nanoparticles occur as palladium complexes decompose, these structures can serve as central precatalysts in common carbon-carbon bond formation. Palladium NPs are also valuable alternatives to molecular catalysts because they do not require costly and toxic ligands. In this Account, we review the role of "homeopathic" palladium catalysts in carbon-carbon coupling reactions. Seminal studies from the groups of Beletskaya, Reetz, and de Vries showed that palladium NPs can catalyze Heck and Suzuki-Miyaura reactions with aryl iodides and, in some cases, aryl bromides at part per million levels. As a result, researchers coined the term "homeopathic" palladium catalysis. Industry has developed large-scale applications of these transformations. In addition, chemists have used Crooks' concept of dendrimer encapsulation to set up efficient nanofilters for Suzuki-Miyaura and selective Heck catalysis, although these transformations required high PdNP loading. With arene-centered, ferrocenyl-terminated dendrimers containing triazolyl ligands in the tethers, we designed several generations of dendrimers to compare their catalytic efficiencies, varied the numbers of Pd atoms in the PdNPs, and examined encapsulation vs stabilization. The catalytic efficiencies achieved "homeopathic" (TON = 540 000) behavior no matter the PdNP size and stabilization type. The TON increased with decreasing the Pd/substrate ratio, which suggested a leaching mechanism. Recently, we showed that water-soluble arene-centered dendrimers with tri(ethylene glycol) (TEG) tethers stabilized PdNPs involving supramolecular dendritic assemblies because of the interpenetration of the TEG branches. Such PdNPs are stable and retain their "homeopathic" catalytic activities for Suzuki-Miyaura reactions for months. (TONs can reach 2.7 × 10(6) at 80 °C for aryl bromides and similar values for aryl iodides at 28 °C.) Sonogashira reactions catalyzed by these PdNPs are quantitative with only 0.01% Pd/mol substrate. Kato's group has reported remarkable catalytic efficiencies for mesoporous catalysts formed by polyamidoamine (PAMAM) dendrimer polymerizations. These and other mesoporous structures could allow for catalyst recycling, with efficiencies approaching the "homeopathic" behavior. In recent examples of Suzuki-Miyaura reactions of aryl chlorides, chemists achieved truly "homeopathic" catalysis when a surfactant such as a tetra-n-butylammonium halide or an imidazolium salt was used in stoichiometric quantities with substrate. These results suggest that the reactive halide anion of the salt attacks the neutral Pd species to form a palladate. In the case of aryl chlorides, the reaction may occur through the difficult, rate-limiting oxidative-addition step.
