Abstract:Several new 1,3,4-oxadiazole derivatives containing a pyrazole ring were designed and synthesized from ethyl acetoacetate and triethyl orthoformate as starting materials via multi-step reactions. The compound structures were confirmed by melting point, 1 H NMR and HRMS. They were evaluated for fungicidal and herbicidal activities. Four of the compounds exhibited moderate fungicidal activity against Colletotrichum species. Most of the compounds had moderate-to-good activity as a herbicide.
Synthesis, antibacterial and antifungal activity of myricetin derivatives containing piperidine and amide fragments

Abstract:Background: Continuous use of synthetic bactericides and fungicides is causing pathogens to develop resistance, resulting in increased use of pesticides and affecting food security. The green pesticides derived from natural products could reduce or avoid 'pesticide hazards' caused by synthetic pesticides as a result of their unique mechanism of action. Therefore, it is of great significance to create green pesticides with novel structures. Results: Herein, 30 novel myricetin derivatives containing piperidine and amide fragments were designed and synthesized using active group splicing. Among them, compound Z30 had excellent inhibitory effect against Xanthomonas oryzae pv. Oryzae (Xoo) with the half effective concentration (EC50 ) of 2.7 μg mL-1 . Compound Z26 not only exhibited better antibacterial activity against Xaxonopodis pv. Citri (Xac) with EC50 of 3.9 μg mL-1 , but also displayed higher antifungal activity against Rhizoctonia solani (Rs) with EC50 of 8.3 μg mL-1 . In vivo experiments proved that Z30 against bacterial blight of rice and Z26 against rice blast exhibits significant protective and curative effect. Scanning electron microscopy (SEM) and transmission electron microscopy (TEM) showed that Z26 and Z30 could change the integrity of cell wall and membrane of pathogen Xoo, Xac and Rs, resulting in cytoplasmic leakage and eventually death. Enzymatic assay, molecular docking and molecular dynamics simulations (MDs) indicated that Z26 could be used as a potential succinate dehydrogenase inhibitor (SDHI). Conclusion: Z26 and Z30 significantly reduced the pathogenicity of the pathogens, which provided a new idea and direction for the development of green pesticides. © 2023 Society of Chemical Industry.

Abstract:Succinate dehydrogenase inhibitor is a low-toxic and high active fungicide. In order to develop novel and broad-spectrum succinate dehydrogenase inhibitor (SDHI) fungicides, thiophene was introduced into the structure of pyrazole carboxamide fungicides. Twenty four pyrazole-thiophene carboxamides were designed, synthesized and characterized by H NMR, C NMR and HRMS. The crystal structure of N-(4-methoxyphenyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene2-carboxamide (7i) was determined by X-ray diffraction. The antifungal activity of all the synthesized compounds was determined against six plant pathogenic fungi, and preliminary bioassays suggested that some compounds exhibited good antifungal activity against Rhizoctonia solani, Fusarium graminearum and Botrytis cinerea. Among them, N-(4fluorophenethyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene-2-carboxamide (7c) exhibited the best antifungal activities against R. solani in vitro with EC50 value of 11.6 μmol/L, and N-(2-fluorophenyl)-4-(1-methyl-1H-pyrazol-4-yl)thiophene-2-carboxamide (7j) against F. graminearum with EC50 value of 28.9 μmol/L. And N-(4-chlorophenyl)-4-(1-methyl-1H-pyrazol-4yl)thiophene-2-carboxamide (7h) showed similar inhibition abilities with thifluzamide against B. cinerea with EC50 value of 21.3 μmol/L. The molecular docking results showed that the high antifungal activitie compounds form stronger interactions with important amino acid residues of succinate dehydrogenase.

Abstract:19 myricetin derivatives containing thiazole were designed and synthesized. Their fungicidal activities in vitro against ten species of plant pathogenic fungi were evaluated. Bioassay results indicated that some of compounds exhibited remarkable antifungal activities. Among them, Z17 showed the strongest antifungal activity against Phomopsis sp , with in vitro EC 50 of 12.3 μg/mL, which was superior to those of the control drug azoxystrobin (32.2 μg/mL) and fluopyram (77.7 μg/mL). In addition, Z18 also had the inhibitory activity against the Alternaria brassicae , with an EC 50 value of 32.5 μg/mL, which was much higher than azoxystrobin (49.3 μg/mL). Z17 exhibited good protective activity (62.5 %) against Phomopsis sp on kiwifruit in vivo at 200 μg/mL. The results of in vivo experiments revealed that Z18 could effectively defend against the infestation of cabbage by Alternaria brassicae , and improve the protection ability of the crop. Scanning electron microscopy (SEM) and fluorescence microscopy (FM) demonstrated that Z17 could destroy the integrity of cell membrane of pathogen Phomopsis sp , thus affecting the normal growth of mycelia. The results of the mechanism research further confirmed that the action of Z17 changed the mycelial morphology of Phomopsis sp , affected the permeability of cells, increased the leakage of cytoplasm and MDA contents. Molecular docking simulation results revealed that compound Z17 could readily bind with the active site of SDH and could be a potential SDH inhibitor. In summary, this study provides new ideas for effectively controlling plant fungal diseases and developing new green chemical pesticide products. Abstract:A series of new piperidine-4-carbohydrazide derivatives bearing a quinazolinyl moiety were prepared and evaluated for their fungicidal activities against agriculturally important fungi. Among these derivatives, the chemical structure of compound A45 was clearly verified by X-ray crystallographic analysis. The antifungal bioassays revealed that many compounds in this series possessed good to excellent inhibition effects toward the tested fungi. For example, compounds A13 and A41 had EC50 values of 0.83 and 0.88 μg/mL against Rhizoctonia solani in vitro, respectively, superior to those of positive controls Chlorothalonil and Boscalid (1.64 and 0.96 μg/mL, respectively). Additionally, the above two compounds also exhibited notable inhibitory activities against Verticillium dahliae (with EC50 values of 1.12 and 3.20 μg/mL, respectively), far better than the positive controls Carbendazim and Chlorothalonil (19.3 and 11.0 μg/mL, respectively). More importantly, compound A13 could potently inhibit the proliferation of R. solani in the potted rice plants, showing good in vivo curative and protective efficiencies of 76.9% and 76.6% at 200 μg/mL, respectively. Furthermore, compound A13 demonstrated an effective inhibition of succinate dehydrogenase (SDH) activity in vitro with an IC50 value of 6.07 μM. Finally, the molecular docking study revealed that this compound could be well embedded into the active pocket of SDH via multiple noncovalent interactions, involving residues like SER39, ARG43, and GLY46.

Abstract:In this study, a series of novel difluoromethyl pyrazole derivatives were synthesized and their structures were confirmed by 1 H NMR, 13 C NMR, and HRMS techniques. Subsequently, their antifungal activities in vitro against Mucor fragilis ( M. fragilis ), Trichoderma atroviride ( T. atroviride ), and Mucor bainieri ( M. bainieri ) at concentrations of 50 and 100 μg/mL were evaluated using the mycelium growth rate assay. Bioactivity results showed that the target compounds revealed lower antifungal activities against M. fragilis , T. atroviride , and M. bainieri than those of prochloraz and carbendazim. Additionally, the antibacterial activities against Xanthomonas axonopodis pv. citri ( Xac ) and Xanthomonas oryzae pv. oryzicola ( Xoo ) were determined using the turbidimeter test method. The bioassay results demonstrated that some of the target compounds exhibited moderate to good antibacterial activities. Notably, 2-methoxyphenyl 3-(difluoromethyl)-1-methyl-1 H -pyrazole-4-carboxylate displayed superior antibacterial effects against Xoo and Xac with inhibition rates of 62, 86 and 53, 80%, respectively, compared to those of thiodiazole copper.

Abstract:Plant bacteria such as Xanthomonas axonopodis pv. citri (Xac), Pseudomonas syringae pv. actinidiae (Psa), Xanthomonas oryzae pv. oryzae (Xoo), and tobacco mosaic virus (TMV) have created huge obstacles to the global trade of food and economic crops. However, traditional chemical agents used to control these plant diseases have gradually become disadvantageous due to long-term irregular use. Therefore, finding new and efficient antibacterial and antiviral agents is becoming imperative. In this study, a series of myricetin derivatives containing a quinazolinone moiety were designed and synthesized, and the antibacterial and antiviral activities of these compounds were evaluated. The bioassay results showed that some target compounds exhibited good antibacterial activities in vitro and antiviral activities in vivo. Among them, the median effective concentration (EC50) value of compound L18 against Xac was 16.9 μg/mL, which was better than those of the control drugs bismerthiazol (BT) (62.2 μg/mL) and thiodiazole copper (TC) (97.5 μg/mL). Scanning electron microscopy (SEM) results confirmed that compound L18 inhibited the growth of Xac by affecting the morphology of cells. Microscale thermophoresis (MST) test results indicated that the dissociation constant (K d) value of compound L11 against TMV-CP was 0.012 μM, which was better than that of the control agent ningnanmycin (2.726 μM). This study reveals that myricetin derivatives containing a quinazolinone moiety are potential antibacterial and antiviral agents.

Abstract:A series of flavonol derivatives containing piperazine and quinoxaline had been designed and synthesized. The biological activity test results showed that some of the target compounds had good antifungal activity against various fungi. N5 had the best antifungal activity against Phomopsis sp ( P.s. ) and Phytophthora capsica ( P.c. ) The half maximal effective concentration (EC 50 ) was 12.9 and 25.8 μg/mL against P.s. and P.c. , respectively, which were better than azoxystrobin ( Az , 25.4 and 71.1 μg/mL). In addition, the protective and curative activities of N5 against kiwifruit were 85.9 and 67.0% at 200 μg/mL in vivo, which were better than that of Az (65.9 and 57.0%). The protective and curative activities against chili leaves were 80.6 and 66.5% at 200 μg/mL, which were better than that of Az (77.6 and 60.0%). The scanning electron microscopy (SEM) experiment showed that the action of N5 caused the mycelium to bend and fold, changed its morphology and caused damaged to the mycelium. Through the measurement of relative conductivity, leakage of cytoplasmic contents and determination of malondialdehyde (MDA) content indicated that N5 could damage the integrity of pathogenic fungal cell membranes, change the permeability of cell membranes, and affect the normal growth of mycelium.

Abstract:To unceasingly expand the molecular diversity of 1,3,4-oxadiazole-2-carbohydrazides, herein, small fragments (including -CH2-, -OCH2-, and -SCH2-) were incorporated into the target compounds to screen out the potential succinate dehydrogenase inhibitors (SDHIs). The bioassay results showed that the antifungal effects (expressed by EC50) against Sclerotinia sclerotiorum, Botryosphaeria dothidea, Fusarium oxysporum, and Colletotrichun higginsianum could reach 1.29 (10a), 0.63 (8h), 1.50 (10i), and 2.09 (10i) μg/mL, respectively, which were slightly lower than those of carbendazim (EC50 were 0.69, 0.13, 0.55, and 0.80 μg/mL, respectively). Especially, compound 10h was extremely bioactive against Gibberella zeae (G. z.) with an EC50 value of 0.45 μg/mL. This outcome was better than that of fluopyram (3.76 μg/mL) and was similar to prochloraz (0.47 μg/mL). In vivo trials against the corn scab (infected by G. z.) showed that compound 10h had control activity of 86.8% at 200 μg/mL, which was better than that of boscalid (79.6%). Further investigations found that compound 10h could inhibit the enzymatic activity of SDH in the G. z. strain with an IC50 value of 3.67 μM, indicating that potential SDHIs might be developed. Additionally, the other biological activities of these molecules were screened simultaneously. The anti-oomycete activity toward Phytophthora infestans afforded a minimal EC50 value of 3.22 μg/mL (10h); compound 4d could strongly suppress the growth of bacterial strains Xanthomonas axonopodis pv. citri and Xanthomonas oryzae pv. oryzae with EC50 values of 3.79 and 11.4 μg/mL, respectively; and compound 10a displayed some insecticidal activity toward Plutella xylostella. Given their multipurpose features, these frameworks could be actively studied as potential pesticide leads.

Abstract:In this paper, a series of novel 1,2,4-trizaole-substituted 1,3,4-oxadiazole derivatives with a dual thioether moiety were constructed. The synthetic compounds were characterized by 1 H NMR, 13 C NMR, HRMS, and single crystal diffraction. The antimicrobial activities of title compounds against fungi ( Pyricutaria oryzae Cav., Phomopsis sp., Botryosphaeria dothidea , cucumber Botrytis cinerea , tobacco Botrytis cinerea , blueberry Botrytis cinerea ) and bacteria ( Xanthomonas oryzae pv. oryzicola , Xoc ; Xanthomonas axonopodis pv. citri , Xac ) revealed these compounds possessed excellent antibacterial activity through mycelial growth rate method and turbidity method, respectively. Among them, compounds 7a , 7d , 7g , 7k , 7l , and 7n had the antibacterial inhibition rate of 90.68, 97.86, 93.61, 97.70, 97.26, and 92.34%, respectively. The EC 50 values of 7a , 7d , 7g , 7k , 7l , and 7n were 58.31, 48.76, 58.50, 40.11, 38.15, and 46.99 μg/mL, separately, superior to that of positive control pesticide thiodiazole copper (104.26 μg/mL). The molecular docking simulation of compound 7l and glutathione s -transferase also confirmed its good activity. The in vivo bioassay toward Xac infected citrus leaves was also performed to evaluate the potential of compounds as efficient antibacterial reagent. Further study of antibacterial mechanism was also carried out, including extracellular polysaccharide production, permeability of bacterial membrane, and scanning electron microscope observations. The excellent antibacterial activities of these compounds provided a strong support for its application for preventing and control plant diseases.

Abstract:A series of 1,4-benzoxazin-3-one derivatives containing an acylhydrazone moiety were designed, synthesized and evaluated for their in vitro antifungal activities against Gibberella zeae , Pellicularia sasakii , Phytophthora infestans , Capsicum wilt , and Phytophthora capsica . The structures of target compounds were characterized by 1 H NMR, 13 H NMR, 19 F NMR and HRMS. The preliminary antifungal evaluation of all target compounds showed that some target compounds possessed moderate to good activities against G. zeae , P. sasakii , P. infestans and C. wilt . Among them, compounds 5L and 5o exhibited noticeable inhibition effects against G. zeae with the EC 50 values (effective concentration for 50% activity) of 20.06 and 23.17 μ g/ml, respectively, which were even nearly double effective than that of hymexazol (40.51 μ g/ml). Meanwhile, compound 5q displayed a notable inhibitory effect toward P. sasakii , with the EC 50 value of 26.66 μg/ml, which was better than that of hymexazol (32.77 μ g/ml). In addition, compound 5r yielded the EC 50 value of 15.37 μ g/ml against P. infestans , which was less than those of hymexazol (18.35 μ g/ml) and carbendazim (34.41 μ g/ml). Eventually, compound 5p showed higher inhibitory effect against C. wilt , with EC 50 value of 26.76 μ g/ml, which was better than that of hymexazol (>50 μ g/ml).

Abstract:A series of myricetin derivatives containing phenoxypyridine structure were designed and synthesized on the basis of the natural product myricitrin, which were structurally characterized on NMR and HRMS. The results of antiviral activity tests showed that some of the target compounds exhibited better inhibitory effects. Among them, B19 and B21 showed better curative activity with EC 50 values of 195.67 and 173.64 μ g/mL, respectively, which were superior to that of the control agent ningnanmycin(NNM) (238.30 μ g/mL). B21 showed better protective activity with EC 50 value of 236.37 μ g/mL, which was better than that of the control agent NNM (269.89 μ g/mL). The results of microcalorimetry showed that B1 , B19 and B21 had good binding affinity with tobacco mosaic virus capsid protein (TMV-CP), and the K d values were 0.059, 0.093 and 0.069 μ M, which was higher than that of NNM (K d = 2.78 μ M). The molecular docking results showed that the hydrogen bond lengths between B1 , B21 and the key amino acid residues of TMV-CP were shorter and with more tightly bound than those of NNM. In B21 -treated tobacco leaves, chlorophyll content and superoxide dismutase (SOD) activity were significantly elevated, indicating that B21 can participate in regulating plant photosynthesis as well as defense enzymes and inducing plants to improve disease resistance. Abstract:In this study, using the botanical active components of carvacrol, thymol, guaiacol, and sesamol as the lead structures, 19 novel botanical active component derivatives containing carboxamide and 1,3,4-thiadiazole thioether moieties ( 5a − 5s ) were synthesized and structurally characterized by 1 H NMR, 13 C NMR, and HRMS. The antibacterial bioassay results in vitro showed that compound 2-(2-methoxyphenoxy)-N-(5-(methylthio)-1,3,4-thiadiazol-2-yl)acetamide ( 5k ) revealed excellent inhibitory activities against Xanthomonas axonopodis pv. citri (Xac) and Xanthomonas oryzae pv. oryzicolaby (Xoc), with the median effective concentration (EC 50 ) values of 22 and 15 μg/ml, respectively, which were even better than those of thiodiazole copper and bismerthiazol. Meanwhile, all the target compounds revealed lower in vitro inhibitory effects on Mucor bainieri (M. bainieri), Mucor fragilis (M. fragilis), and Trichoderma atroviride (T. atroviride), than carbendazim.

Abstract:In this study, using the botanical active component thiochromanone as the lead compound, a total of 32 new thiochromanone derivatives containing a carboxamide moiety were designed and synthesized and their in vitro antibacterial activities against Xanthomonas oryzae pv. oryzae (Xoo), Xanthomonas oryzae pv. oryzicolaby (Xoc), and Xanthomonas axonopodis pv. citri (Xac) were determined, as well as their in vitro antifungal activities against Botryosphaeria dothidea (B. dothidea), Phomopsis sp., and Botrytis cinerea (B. cinerea). Bioassay results demonstrated that some of the target compounds exhibited moderate to good in vitro antibacterial and antifungal activities. In particular, compound 4e revealed excellent in vitro antibacterial activity against Xoo, Xoc, and Xac, and its EC50 values of 15, 19, and 23 μg/mL, respectively, were superior to those of Bismerthiazol and Thiodiazole copper. Meanwhile, compound 3b revealed moderate in vitro antifungal activity against B. dothidea at 50 μg/mL, and the inhibition rate reached 88%, which was even better than that of Pyrimethanil, however, lower than that of Carbendazim. To the best of our knowledge, this is the first report on the antibacterial and antifungal activities of this series of novel thiochromanone derivatives containing a carboxamide moiety.

Design, synthesis and antifungal activity of indole derivatives containing 1,3,4-oxadiazole

Abstract:In this study, 21 indole derivatives containing 1,3,4-oxadiazole were designed and synthesized, the results of the biological activity test showed that the target compounds had certain antifungal activity against 12 plant pathogenic fungi in vitro, among which. E1 showed excellent bioactivity against Botrytis cinerea (B.c.), Tomato Botrytis cinerea (F.M.) and Phomopsis sp (P.s.), with median effective concentration (EC50) values of 2.8, 5.1 and 5.2 µg/mL, which are higher than those of the control drug azoxystrobin (Az) at 15.2, 31.2 and 15.2 µg/mL. In vivo tests on blueberry leaves, tomato leaves and kiwifruit showed that E1 at 200 µg/mL offered stronger protective effect against B.c. (91.9 %) than Az (83.8 %) in blueberry leaves, F.M. (83.3 %) than Az (72.9 %) in tomato leaves and P.s. (89.3 %) than Az (86.9 %) in kiwifruit. Scanning electron microscope (SEM) experiments showed that B.c. hyphae treated with E1 had abnormal shrinkage and obvious morphological changes. The results of the mechanism study showed that E1 could change the integrity of the cell wall and cell membrane of pathogen B.c., which led to the increase in malondialdehyde (MDA), cell leakage and permeability and the rupture of the cell membrane. Because of their strong antifungal effects on plant fungus, indole derivatives containing 1,3,4-oxadiazole were predicted to develop into novel fungicides.

Abstract:A series of novel 1,5-diaryl-1H-pyrazol-3-oxy derivatives containing oxyacetic acid or oxy(2-thioxothiazolidin-3-yl)ethanone moieties were prepared from methyl 3-arylacrylates via a serial of reactions included addition-cyclization, oxidation, substitution, hydrolysis, and condensation. Their structures were confirmed by 1H-NMR, 13C-NMR, IR, and elemental analysis. In addition, the crystal structure of the compound 2-(1,5-diphenyl-1H-pyrazol-3-yloxy)-1-(2-thioxothiazolidin-3-yl)ethanone was determined by single crystal X-ray diffraction analysis. Bioassay results indicated that the compound 2-(5-(4-chlorophenyl)-1-phenyl-1H-pyrazol-3-yloxy)-1-(2-thioxo-thiazolidin-3-yl)ethanone exhibited moderate inhibitory activity against Gibberella zeae at the dosage of 10 mu g mL-1.
Novel 5-Sulfonyl-1,3,4-thiadiazole-Substituted Flavonoids as Potential Bactericides and Fungicides: Design, Synthesis, Three-Dimensional Quantitative Structure-Activity Relationship Studies.

Abstract:Flavonoids, ubiquitous natural products, provide sources for drug discovery owing to their structural diversity, broad-spectrum pharmacological activity, and excellent environmental compatibility. To develop antibacterial and antifungal agents with novel mechanisms of action and innovative structures, a series of novel 5-sulfonyl-1,3,4-thiadiazole-substituted flavonoids were designed and synthesized, and their biological activities against seven agriculturally common phytopathogenic microorganisms were evaluated. The results of the antimicrobial bioassay showed that most of the target compounds displayed excellent inhibitory effects against Xanthomonas oryzae, Rhizoctonia solani, and Colletotrichum orbiculare. Compounds 1, 3, 7, 9, 13, and 14 exhibited remarkable antibacterial activity against X. oryzae pv. oryzae with EC50 values below 10 μg/mL, which were superior to bismerthiazol (70.89 μg/mL). Compound 2 (EC50 = 0.41 μg/mL) displayed the most effective inhibitory potency against R. solani in vivo, comparable protective effects with the positive control carbendizam. Preliminary mechanistic studies indicated that compound 2 induced disordered entanglement of hyphae, shrinkage of hyphal surfaces, extravasation of cellular contents, and vacuole swelling and rupture, which disrupted normal hyphal growth. Subsequently, compounds 35-53 with good antifungal activity were designed and synthesized based on reliable three-dimensional quantitative structure-activity relationship (3D-QSAR) models. Compound 49 showed high efficacy and superior antifungal activity against R. solani, with an EC50 value of 0.28 μg/mL and a half-maximal effective concentration of 0.46 μg/mL.

Abstract:24 chalcone derivatives containing 1,3,4‐thiadiazole were synthesized. The results of bioactivity tests indicated that some of the target compounds exhibited superior antifungal activities in vitro. Notably, the EC50 value of D4 was 14.4 μg/mL against Phomopsis sp, which was significantly better than that of azoxystrobin (32.2 μg/mL) and fluopyram (54.2 μg/mL). The in vivo protective activity of D4 against Phomopsis sp on kiwifruit (71.2%) was significantly superior to azoxystrobin (62.8%) at 200 μg/mL. The in vivo protective activities of D4 were 74.4 and 57.6% against Rhizoctonia solani on rice leaf sheaths and rice leaves, respectively, which were slightly better than those of azoxystrobin (72.1 and 49.2%) at 200 μg/mL. Scanning electron microscopy (SEM) results showed that the mycelial surface collapsed, contracted and grew abnormally after D4 treatment. Finally, the results were further verified by in vivo antifungal assay, fluorescence microscopy (FM) observation, determination of relative conductivity, membrane lipid peroxidation degree assay, and determination of cytoplasmic content leakage. Molecular docking results suggested that D4 could be a potential SDHI.

Abstract:A series of novel thiochroman-4-one derivatives incorporating carboxamide and 1, 3, 4-thiadiazole thioether moieties were synthesized. Bioassay results indicated that the EC50 values of compound 6-chloro-N-(5-(methylthio)-1, 3, 4-thiadiazol-2-yl)-4-oxothiochromane-2-carboxamide (5a) against Xanthomonas oryzae pv. Oryzae (Xoo) and Xanthomonas axonopodis pv. Citri (Xac) were 24 and 30 μg/mL, respectively, which were even better than those of bismerthiazol and thiadiazole copper. Meanwhile, compound 6-methyl-4-oxo-N-(5-(propylthio)-1, 3, 4-thiadiazol-2-yl)thiochromane-2-carboxamide (5m) showed a better antifungal activity against Botrytis cinerea (B. cinerea), with an inhibition rate of 69%, than carbendazim. As far as we know, this is the first report on the antibacterial and antifungal activities of this series of novel thiochroman-4-one derivatives incorporating carboxamide and 1, 3, 4-thiadiazole thioether moieties.
