Abstract:BackgroundBiocompatible smart materials have ushered in revolutionary applications, conferring significant benefits upon humanity. Among these materials, DNA hydrogels stand as a prime representative, which has increasingly piqued the curiosity of researchers within food technology recently. Amidst this rapid advancement, it is imperative to undertake a comprehensive summary and analysis of their applications in this domain, providing a foundational reference for further.Scope and approachThis review initially delves into the classification based on its crosslinking methods and summarizes their distinctive characteristics. Drawing from these attributes, the article elucidates and analyzes instances of applications in the realm of food in detail. Based on the present development, opportunities and challenges concerning industrial applications are deliberated upon, alongside a compilation of potential cutting-edge technologies that could integrate with DNA hydrogels.Findings and conclusionsIn the field of food safety testing, DNA hydrogels are rapidly expanding into multiple detection domains, encompassing food pathogens, heavy metals, and other small molecular contaminants. Leveraging the diverse functional DNA structures, these detection platforms can effortlessly extend to a wide array of detection targets. The responsive sol-gel transition offers convenient detection signal transduction methods for portable rapid detection. Moreover, due to DNA's exceptional biocompatibility and responsiveness, DNA hydrogels play a pivotal role in intelligent control release within oral delivery systems. However, considerable efforts are still required for industrial applications, particularly in addressing the high costs of DNA synthesis, the relatively lower mechanical strength, and the thorough exploration of storage methodologies.
Responsive Polypeptide-Dna Hydrogel Crosslinked by G-Quadruplex

Abstract:In the past thirty years, DNA molecule has received considerable attention as a promising building material due to its precise base-paring recognition, the programmable sequences, the predictable secondary structures, etc. Based on DNA self-assembly technique, DNA supramolecular hydrogels can be fabricated which are crosslinked networks swollen in an aqueous phase by the supramolecular interaction and also have widely applications in the biomedical field. In this paper, a new type of DNA-based supramolecular hydrogel was prepared by simply mixing two components: Polypeptide-DNA and G-quadruplex. We have successfully synthesized single-stranded DNA (ssDNA) grafted polypeptide via the copper-catalyzed click reaction. By introducing the intermolecular G-quadruplex structure, which is rich in guanine and is capable of forming a four-stranded structure in the presence of potassium, the hydrogel can be formed by DNA hybridization in a fast and in-situ manner and also obtain thermal and ionic responsive gel-sol transition, respectively. In this experiment, we firstly used polyacrylamide gel electrophoresis (PAGE), circular dichroism spectra (CD), UV-Vis spectroscopy to demonstrate the G-quadruplex structure formation, and the assembling process was efficient which are formed the parallel structure and the melting temperature (T-m) was about 43 degrees C. Then by the rheological tests, the shear-storage modulus (G') was obviously higher (up to 2.4 kPa) than the shear-loss modulus (G '') over the entire time range, and a minimum concentration of 0.5 wt% polypeptide-DNA was required to form a gel-like state, and the mechanical strength of the DNA hydrogels increased with increasing concentration of the DNA building blocks. What's more, the hydrogels can switch between the gel and the sol state when changing the temperature from 25 to 50 degrees C many times, indicating that the DNA hydrogels respond to thermal in a reversible way. At last, by using the properties of crown ether complexing potassium ion, the DNA hydrogel can obtain the ion responsiveness properties. Due to its designable responsiveness and good biocompatibility, this supramolecular hydrogel would have great potential applications in tissue engineering and 3D bioprinting field.

Abstract:Deoxyribonucleotide (DNA) is uniquely programmable and biocompatible, and exhibits unique appeal as a biomaterial as it can be precisely designed and programmed to construct arbitrary shapes. DNA hydrogels are polymer networks comprising cross-linked DNA strands. As DNA hydrogels present programmability, biocompatibility, and stimulus responsiveness, they are extensively explored in the field of biomedicine. In this study, we provide an overview of recent advancements in DNA hydrogel technology. We outline the different design philosophies and methods of DNA hydrogel preparation, discuss its special physicochemical characteristics, and highlight the various uses of DNA hydrogels in biomedical domains, such as drug delivery, biosensing, tissue engineering, and cell culture. Finally, we discuss the current difficulties facing DNA hydrogels and their potential future development.

Abstract:As emerging biofunctional materials, DNA hydrogels have made great progresses in many engineering fields related to biomedicine in recent years. This review systematically summarizes synthetic strategies of DNA hydrogels and highlights their latest applicaitons in the filelds concerning therapeutic systems, cell culture platforms, tissue engineering materials, and biosensors. Also, future perspectives and challenges of the DNA hydrogels are presented. With the remarkable development of DNA nanotechnology, interest in DNA molecules has expanded beyond its biological role to building blocks in materials science. As a unique branch of DNA‐based materials, DNA hydrogels have exhibited many fascinating characteristics, including broad biocompatibility, precise programmability, convenient modification, and tunable mechanical properties, which make DNA hydrogels ideal biomaterials. Moreover, by combining with functional nucleic acids, such as aptamers, i‐motif nanostructures, CpG oligodeoxynucleotides, and DNAzymes, DNA hydrogels can be further tailored to provide additional target recognition, therapeutic potential, and catalytic activities, allowing them to play important roles in biosensing and medical applications. This review, aims to provide readers with an up‐to‐date overview of the important developments of biomedical DNA hydrogels. First, it introduces different synthetic strategies of hydrogels that utilize DNA as building materials and functional units within the hydrogel networks and discuss their advantages in biomedical applications. Subsequently, new approaches and applications of biomedical DNA hydrogels in the recent years are highlighted, such as therapeutic systems, cell culture platforms, tissue engineering materials, and biosensors. Finally, future perspectives and remaining challenges of DNA hydrogels in biomedicine are presented.

Abstract:In recent years, DNA supramolecular hydrogels have attracted much attention due to their injectability, biocompatibility, responsiveness and self-healing properties. In this work, we designed a linear DNA brick containing one duplex with two cytosine (C)-rich sequence on both ends. This brick can first assemble to form duplex under pH 8 condition. After adjusting the pH to 5, the C-rich sequence tends to form intermolecular i-motif structure, which joins the linear DNA molecules together to form interlocked cyclic structures and yield the DNA hydrogel. By adjusting the length and bending curvature of the duplex part of the molecule, one can change the basic unit of the hydrogel structure to tune the properties of the DNA hydrogel.
Impact of comorbid panic and posttraumatic stress disorder on outcomes of collaborative care for late-life depression in primary care.

Abstract:OBJECTIVE Comorbid anxiety disorders may result in worse depression treatment outcomes. The authors evaluated the effect of comorbid panic disorder and posttraumatic stress disorder (PTSD) on response to a collaborative-care intervention for late-life depression in primary care. METHODS A total of 1,801 older adults with depression were randomized to a collaborative-care depression treatment model versus usual care and assessed at baseline, 3, 6, and 12 months, comparing differences among participants with comorbid panic disorder (N=262) and PTSD (N=191) and those without such comorbid anxiety disorders. RESULTS At baseline, patients with comorbid anxiety reported higher levels of psychiatric and medical illness, greater functional impairment, and lower quality of life. Participants without comorbid anxiety who received collaborative care had early and lasting improvements in depression compared with those in usual care. Participants with comorbid panic disorder showed similar outcomes, whereas those with comorbid PTSD showed a more delayed response, requiring 12 months of intervention to show a significant effect. At 12 months, however, outcomes were comparable. Interactions of intervention status by comorbid PTSD or panic disorder were not statistically significant, suggesting that the collaborative-care model performed significantly better than usual care in depressed older adults both with and without comorbid anxiety. CONCLUSIONS Collaborative care is more effective than usual care for depressed older adults with and without comorbid panic disorder and PTSD, although a sustained treatment response was slower to emerge for participants with PTSD. Intensive and prolonged follow-up may be needed for depressed older adults with comorbid PTSD.

Abstract:Due to the similarity to extracellular matrix, hydrogels have been widely used in biomedical field such as drug delivery, tissue engineering and in vitro cell culture. Deoxyribonucleic acid (DNA) is known for the clear chemical composition, designable sequence and unique structural rigidity, which makes DNA an ideal material for construction of hydrogels. Based on our research, this review summarizes recent developments in DNA supramolecular hydrogels. Firstly, the construction strategies including the pure and hybrid DNA supramolecular hydrogels are introduced. While the construction of pure DNA supramolecular hydrogels is mainly based on base pairing or physical entanglement, covalent or non-covalent doping is also demonstrated to play important roles in preparing the hybrid hydrogels. Then, the physical properties of the DNA supramolecular hydrogels are discussed, such as the excellent permeability, tunable mechanical strength and dynamic properties. Next, the functionalization strategies are also summarized, including sequence design, chemical modification and network interpenetration. Furthermore, the applications of DNA supramolecular hydrogels in biomedicine are introduced, focusing on three-dimensional cell culture and printing, tissue engineering, immunotherapy and drug delivery. Finally, some challenges and future perspectives in the development of DNA supramolecular hydrogels are discussed. It is expected that with the flourishing development, DNA supramolecular hydrogels will have more applications. [GRAPHICS] .

Abstract:DNA-based supramolecular hydrogels are important and promising biomaterials for various applications due to their inherent biocompatibility and tunable physicochemical properties. The three-dimensional supramolecular matrix of DNA formed by non-covalently dynamic cross-linking provides exceptional adaptability, self-healing, injectable and responsive properties for hydrogels. In addition, DNA hydrogels are also ideal bio-scaffold materials owing to their tissue-like mechanics and intrinsic biological functions. Technically, DNA can assemble into supramolecular networks by pure complementary base pairing; it can also be combined with other building blocks to construct hybrid hydrogels. This review focuses on the development and construction strategies of DNA hydrogels. Assembly and synthesis methods, diverse responsiveness and biomedical applications are summarized. Finally, the challenges and prospects of DNA-based supramolecular hydrogels are discussed.

Abstract:In recent years, DNA has emerged as a fascinating building material to engineer hydrogels due to its excellent programmability, which has gained considerable attention in biomedical applications. Understanding the structure‐property relationship and underlying molecular determinants of DNA hydrogel is essential to precisely tailor its macroscopic properties at molecular level. In this review, we highlight the rational design principles of DNA molecular networks based on molecular dynamics of polymers on the temporal scale, which can be engineered via the backbone rigidity and crosslinking kinetics. By elucidating the underlying molecular mechanisms and theories, we aim to provide a comprehensive overview of how the tunable DNA backbone rigidity and the crosslinking kinetics lead to desirable macroscopic properties of DNA hydrogels, including mechanical properties, diffusive permeability, swelling behaviors, and dynamic features. Furthermore, we also discuss how the tunable macroscopic properties make DNA hydrogels promising candidates for biomedical applications, such as cell culture, tissue engineering, bio‐sensing, and drug delivery. Abstract:DNA hydrogels play an increasingly important role in biomedicine and bioanalysis applications. Due to their high programmability, multifunctionality and biocompatibility, they are often used as effective carriers for packing drugs, cells, or other bioactive cargoes in vitro and in vivo. However, the stability of the DNA hydrogels prevents their in-demand rapid release of cargoes to achieve a full therapeutic effect in time. For bioanalysis, the generation of signals sometimes needs the DNA hydrogel to be rapidly degraded when sensing target molecules. To meet these requirements, stimulus-responsive DNA hydrogels are designed. By responding to different stimuli, self-degradable DNA hydrogels can switch from gel to solution for quantitative bioanalysis and precision cargo delivery. This review summarizes the recently developed innovative methods for designing stimuli-responsive self-degradable DNA hydrogels and showed their applications in the bioanalysis and biomedicines fields. Challenges, as well as prospects, are also discussed.

Abstract:DNA-based hydrogels are exceptional materials for biological applications because of their numerous advantages such as biodegradability, biocompatibility, hydrophilicity, super absorbency, porosity, and swelling. Among these advantages, the ability of DNA-based hydrogels to respond to specific physical and chemical triggers and undergo reversible phase transitions has garnered significant attention in the fields of disease diagnosis (biosensors) and treatment (drug delivery). This article focuses on the recent advancements in the research of DNA-based hydrogels and discusses the different types of these hydrogels, the synthetic methods, their unique properties, and their applications in biosensors and drug delivery. The types of DNA hydrogels are categorized based on their building blocks, and the process of synthesis as well as the unique characteristics of DNA-based hydrogels are described. Then, DNA-based responsive hydrogels utilized as intelligent materials for the development of biosensors are reviewed. Furthermore, this article also presents the current status of DNA-based responsive hydrogels in drug delivery for cancer treatment, wound healing, and other therapeutic applications. Ultimately, this paper discusses the current challenges in expanding the practical application of DNA-based hydrogels.

Abstract:Extracellular matrix (ECM) provides essential supports three dimensionally to the cells in living organs, including mechanical support and signal, nutrition, oxygen, and waste transportation. Thus, using hydrogels to mimic its function has attracted much attention in recent years, especially in tissue engineering, cell biology, and drug screening. However, a hydrogel system that can merit all parameters of the natural ECM is still a challenge. In the past decade, deoxyribonucleic acid (DNA) has arisen as an outstanding building material for the hydrogels, as it has unique properties compared to most synthetic or natural polymers, such as sequence designability, precise recognition, structural rigidity, and minimal toxicity. By simple attachment to polymers as a side chain, DNA has been widely used as cross-links in hydrogel preparation. The formed secondary structures could confer on the hydrogel designable responsiveness, such as response to temperature, pH, metal ions, proteins, DNA, RNA, and small signal molecules like ATP. Moreover, single or multiple DNA restriction enzyme sites could be incorporated into the hydrogels by sequence design and greatly expand the latitude of their responses. Compared with most supramolecular hydrogels, these DNA cross-linked hydrogels could be relatively strong and easily adjustable via sequence variation, but it is noteworthy that these hydrogels still have excellent thixotropic properties and could be easily injected through a needle. In addition, the quick formation of duplex has also enabled the multilayer three-dimensional injection printing of living cells with the hydrogel as matrix. When the matrix is built purely by DNA assembly structures, the hydrogel inherits all the previously described characteristics; however, the long persistence length of DNA structures excluded the small size meshes of the network and made the hydrogel permeable to nutrition for cell proliferation. This unique property greatly expands the cell viability in the three-dimensional matrix to several weeks and also provides an easy way to prepare interpenetrating double network materials. In this Account, we outline the stream of hydrogels based on DNA self-assembly and discuss the mechanism that brings outstanding properties to the materials. Unlike most reported hydrogel systems, the all-in-one character of the DNA hydrogel avoids the "cask effect" in the properties. We believe the hydrogel will greatly benefit cell behavior studies especially in the following aspects: (1) stem cell differentiation can be studied with solely tunable mechanical strength of the matrix; (2) the dynamic nature of the network can allow cell migration through the hydrogel, which will help to build a more realistic model to observe the migration of cancer cells in vivo; (3) combination with rapidly developing three-dimension printing technology, the hydrogel will boost the construction of three-dimensional tissues and artificial organs.

Abstract:Deoxyribonucleic acid (DNA) hydrogels combine the properties of DNAs and hydrogels, and adding functionalized DNAs is key to the wide application of DNA hydrogels. In stimuli-responsive DNA hydrogels, the DNA transcends its application in genetics and bridges the gap between different fields. Specifically, the DNA acts as both an information carrier and a bridge in constructing DNA hydrogels. The programmability and biocompatibility of DNA hydrogel make it change macroscopically in response to a variety of stimuli. In order to meet the needs of different scenarios, DNA hydrogels were also designed into microcapsules, beads, membranes, microneedle patches, and other forms. In this study, the stimuli were classified into single biological and non-biological stimuli and composite stimuli. Stimuli-responsive DNA hydrogels from the past five years were summarized, including but not limited to their design and application, in particular logic gate pathways and signal amplification mechanisms. Stimuli-responsive DNA hydrogels have been applied to fields such as sensing, nanorobots, information carriers, controlled drug release, and disease treatment. Different potential applications and the developmental pro-spects of stimuli-responsive DNA hydrogels were discussed.

Abstract:It is the first time that the properties of functional DNA hydrogels based on nanomaterials are systematically overviewed. And their recent applications in biosensing, drug delivery, cell culture, and environmental protection are summarized and highlighted. With the extensive attention of DNA hydrogels in biomedicine, biomaterial, and other research fields, more and more functional DNA hydrogels have emerged to match the various needs. Incorporating nanomaterials into the hydrogel network is an emerging strategy for functional DNA hydrogel construction. Surprisingly, nanomaterials‐based DNA hydrogels can be engineered to possess favorable properties, such as dynamic mechanical properties, excellent optical properties, particular electrical properties, perfect encapsulation properties, improved magnetic properties, and enhanced antibacterial properties. Herein, the preparation strategies of nanomaterials‐based DNA hydrogels are first highlighted and then different nanomaterial designs are used to demonstrate the functional regulation of DNA hydrogels to achieve specific properties. Subsequently, representative applications in biosensing, drug delivery, cell culture, and environmental protection are introduced with some selected examples. Finally, the current challenges and prospects are elaborated. The study envisions that this review will provide an insightful perspective for the further development of functional DNA hydrogels.

Abstract:Soft hydrogel materials have attracted increasing attention from scientists due to the programmability of nucleic acids. DNA hydrogels are increasingly used in biomedicine due to their natural biocompatibiity and degradation properties. Moreover, the addition of various molecules to these hydrogels can enhance the superiority and utility of DNA hydrogels. Therefore, the present study aimed to provide a timely overview of historical perspectives and recent research advances in DNA hydrogels and their applications in bioengineering and stimulus-responsive systems. The state-of-the-art problems and future prospects of DNA hydrogels were also discussed.

Abstract:The cell microenvironment plays a crucial role in regulating cell behavior and fate in physiological and pathological processes. As the fundamental component of the cell microenvironment, extracellular matrix (ECM) typically possesses complex ordered structures and provides essential physical and chemical cues to the cells. Hydrogels have attracted much attention in recapitulating the ECM. Compared to natural and synthetic polymer hydrogels, DNA hydrogels have unique programmable capability, which endows the material precise structural customization and tunable properties. This review focuses on recent advances in programmable DNA hydrogels as artificial extracellular matrix, particularly the pure DNA hydrogels. It introduces the classification, design, and assembly of DNA hydrogels, and then summarizes the state-of-the-art achievements in cell encapsulation, cell culture, and tissue engineering with DNA hydrogels. Ultimately, the challenges and prospects for cellular applications of DNA hydrogels are delivered.
