Abstract:We explored how genetic variability of black spruce populations, shaped by their postglacial history and adaptation to local climate, affects the species' response to future climate change (CC). By analyzing tree rings and genetic data from 62 populations planted in four common gardens across Canada, we found that populations with varied postglacial histories will respond differently to CC, impacting their future growth and carbon sequestration potential. Understanding these genetic differences is crucial for accurately projecting the species' response to CC. Climate change (CC) necessitates reforestation/afforestation programs to mitigate its impacts and maximize carbon sequestration. But comprehending how tree growth, a proxy for fitness and resilience, responds to CC is critical to maximize these programs' effectiveness. Variability in tree response to CC across populations can notably be influenced by the standing genetic variation encompassing both neutral and adaptive genetic diversity. Here, a framework is proposed to assess tree growth potential at the population scale while accounting for standing genetic variation. We applied this framework to black spruce (BS, Picea mariana [Mill] B.S.P.), with the objectives to (1) determine the key climate variables having impacted BS growth response from 1974 to 2019, (2) examine the relative roles of local adaptation and the phylogeographic structure in this response, and (3) project BS growth under two Shared Socioeconomic Pathways while taking standing genetic variation into account. We modeled growth using a machine learning algorithm trained with dendroecological and genetic data obtained from over 2600 trees (62 populations divided in three genetic clusters) in four 48‐year‐old common gardens, and simulated growth until year 2100 at the common garden locations. Our study revealed that high summer and autumn temperatures negatively impacted BS growth. As a consequence of warming, this species is projected to experience a decline in growth by the end of the century, suggesting maladaptation to anticipated CC and a potential threat to its carbon sequestration capacity. This being said, we observed a clear difference in response to CC within and among genetic clusters, with the western cluster being more impacted than the central and eastern clusters. Our results show that intraspecific genetic variation, notably associated with the phylogeographic structure, must be considered when estimating the response of widespread species to CC.

Abstract:The most common tool to predict future changes in species range are species distribution models. These models do, however, often underestimate potential future habitat, as they do not account for phenotypic plasticity and local adaptation, although being the most important processes in the response of tree populations to rapid climate change. Here, we quantify the difference in the predictions of future range for Norway spruce, by (i) deriving a classic, occurrence-based species distribution model (OccurrenceSDM), and (ii) analysing the variation in juvenile tree height and translating this to species occurrence (TraitSDM). Making use of 32 site locations of the most comprehensive European trial series that includes 1,100 provenances of Norway spruce originating from its natural and further beyond from its largely extended, artificial distribution, we fit a universal response function to quantify growth as a function of site and provenance climate. Both the OccurrenceSDM and TraitSDM show a substantial retreat towards the northern latitudes and higher elevations (−55 and −43%, respectively, by the 2080s). However, thanks to the species’ particularly high phenotypic plasticity in juvenile height growth, the decline is delayed. The TraitSDM identifies increasing summer heat paired with decreasing water availability as the main climatic variable that restricts growth, while a prolonged frost-free period enables a longer period of active growth and therefore increasing growth potential within the restricted, remaining area. Clear signals of local adaptation to climatic clines spanning the entire range are barely detectable, as they are disguised by a latitudinal cline. This cline strongly reflects population differentiation for the Baltic domain, but fails to capture the high phenotypic variation associated to the geographic heterogeneity in the Central European mountain ranges paired with the species history of postglacial migration. Still the model is used to provide recommendations of optimal provenance choice for future climate conditions. In essence, assisted migration may not decrease the predicted range decline of Norway spruce, but may help to capitalize on potential opportunities for increased growth associated with warmer climates.
Heritable Variation in Tree Growth and Needle Vegetation Indices of Slash Pine (pinus Elliottii) Using Unmanned Aerial Vehicles (Uavs)

Abstract:Vegetation indices (VIs) are mainly associated with plant yield, and yield-related physiological traits can assist in breeding selection. Tree breeding programs require a rapid assessment of a large number of individual trees across multiple regions. Unmanned aerial vehicle (UAV) multispectral platforms combined with high spatial, temporal and spectral resolutions can readily assess the VIs at various plant growth phases, which has been deeply detailed for evaluating the physiological status of agricultural crops and forest stands. However, the underlying genetics of foliar spectral bands, VIs and growth traits from a large scale of samples, especially in the case of forest trees, have been less investigated. In this paper, two sites of slash pine breeding populations were completely investigated to monitor the spectral bands, VIs, and tree growth traits. This allows us to determine the genotypic variation of these traits and to estimate the heritability (h(2)). The results show that the estimated h(2) ranges from 0.04 to 0.36, and the site may affect tree growth because the traits between two sites come to different h(2) values. At the two sites, the h(2) values of the green red difference vegetation index (GNDVI) and anthocyanin reflectance index (ARI) at Site 1 and the red edge chlorophyll index (RECI), leaf chlorophyll index (LCI), red green blue vegetation index (RGBVI), normalized difference red edge index (NDRE), NIR and red edge at Site 2 were less than 0.1. However, breeding selection is still possible for most VIs and growth traits at both sites based on different breeding targets and families (families 3, 5, 10, 11, 14 and 17 at Site 1 and 4, 6, 8, 19 and 13 at Site 2 for optimal spectral bands; families 2, 8, 7, 13, 16, and 18 at Site 1 and 6, 7, 11, 14, 15, 18, and 20 at Site 2 for optimal VIs; families 8 and 16 at Site 1 and families 7 and 18 at Site 2 for optimal growth traits) with a variety of optimal traits are selected. This paper demonstrates the potential use of UAV-based imagery technologies in the breeding selection of slash pine trees.

Abstract:Tree improvement programs often focus on improving productivity-related traits; however, under present climate change scenarios, climate change-related (adaptive) traits should also be incorporated into such programs. Therefore, quantifying the genetic variation and correlations among productivity and adaptability traits, and the importance of genotype by environment interactions, including defense compounds involved in biotic and abiotic resistance, is essential for selecting parents for the production of resilient and sustainable forests. Here, we estimated quantitative genetic parameters for 15 growth, wood quality, drought resilience, and monoterpene traits for Picea glauca (Moench) Voss (white spruce). We sampled 1,540 trees from three open-pollinated progeny trials, genotyped with 467,224 SNP markers using genotyping-by-sequencing (GBS). We used the pedigree and SNP information to calculate, respectively, the average numerator and genomic relationship matrices, and univariate and multivariate individual-tree models to obtain estimates of (co)variance components. With few site-specific exceptions, all traits examined were under genetic control. Overall, higher heritability estimates were derived from the genomic- than their counterpart pedigree-based relationship matrix. Selection for height, generally, improved diameter and water use efficiency, but decreased wood density, microfibril angle, and drought resistance. Genome-based correlations between traits reaffirmed the pedigree-based correlations for most trait pairs. High and positive genetic correlations between sites were observed (average 0.68), except for those pairs involving the highest elevation, warmer, and moister site, specifically for growth and microfibril angle. These results illustrate the advantage of using genomic information jointly with productivity and adaptability traits, and defense compounds to enhance tree breeding selection for changing climate.

Abstract:Phenology is an important indicator of environmental variation and climate change impacts. In conifers monitoring phenology of photosynthesis through remote sensing has been unreliable, because needle foliage varies little throughout the year. This is challenging for modelling ecosystem carbon uptake and monitoring phenology for enhanced breeding (genomic selection) and forest health. Here we demonstrate that drone-based carotenoid-sensitive spectral indices, such as the chlorophyll carotenoid index (CCI), can be used to track phenology in conifers by taking advantage of the close relationship between seasonally changing carotenoid levels and the variation of photosynthetic activity. Physiological ground measurements, including photosynthetic pigments and maximum quantum yield of chlorophyll fluorescence, indicated that CCI tracked the variation of photosynthetic activity better than other vegetation indices for 30 white spruce seedlings measured over one year. A machine learning approach, using CCI derived from drone-based multispectral imagery, was used to model phenology of photosynthesis for the entire pedigree population (6000 seedlings). This high-throughput drone-based phenotyping approach is suitable for studying climate change impacts and environmental variation on the physiological status of thousands of field-grown conifers at unprecedented speed and scale.

Abstract:Local adaptation in tree species has been documented through a long history of common garden experiments where functional traits (height, bud phenology) are used as proxies for fitness. However, the ability to identify genes or genomic regions related to adaptation to climate requires the evaluation of traits that precisely reflect how and when climate exerts selective constraints. We combine dendroecology with association genetics to establish a link between genotypes, phenotypes and interannual climatic fluctuations. We illustrate this approach by examining individual tree responses embedded in the annual rings of 233 Pinus strobus trees growing in a common garden experiment representing 38 populations from the majority of its range. We found that interannual variability in growth was affected by low temperatures during spring and autumn, and by summer heat and drought. Among-population variation in climatic sensitivity was significantly correlated with the mean annual temperature of the provenance, suggesting local adaptation. Genotype-phenotype associations using these new tree-ring phenotypes validated nine candidate genes identified in a previous genetic-environment association study. Combining dendroecology with association genetics allowed us to assess tree vulnerability to past climate at fine temporal scales and provides avenues for future genomic studies on functional adaptation in forest trees.

Abstract:Selection of climate-change adapted ecotypes of commercially valuable species to date relies on DNA-assisted screening followed by growth trials. For trees, such trials can take decades, hence any approach that supports focussing on a likely set of candidates may save time and money. We use a non-stationary statistical analysis with spatially varying coefficients to identify ecotypes that indicate first regions of similarly adapted varieties of Douglas-fir ( Pseudotsuga menziesii (Mirbel) Franco) in North America. For over 70,000 plot-level presence-absences, spatial differences in the survival response to climatic conditions are identified.

Abstract:Predicting likely species responses to an alteration of their local environment is key to decision-making in resource management, ecosystem restoration and biodiversity conservation practice in the face of global human-induced habitat disturbance. This is especially true for forest trees which are a dominant life form on Earth and play a central role in supporting diverse communities and structuring a wide range of ecosystems. In Europe, it is expected that most forest tree species will not be able to migrate North fast enough to follow the estimated temperature isocline shift given current predictions for rapid climate warming. In this context, a topical question for forest genetics research is to quantify the ability for tree species to adapt locally to strongly altered environmental conditions (Kremer et al. ). Identifying environmental factors driving local adaptation is, however, a major challenge for evolutionary biology and ecology in general but is particularly difficult in trees given their large individual and population size and long generation time. Empirical evaluation of local adaptation in trees has traditionally relied on fastidious long-term common garden experiments (provenance trials) now supplemented by reference genome sequence analysis for a handful of economically valuable species. However, such resources have been lacking for most tree species despite their ecological importance in supporting whole ecosystems. In this issue of Molecular Ecology, De Kort et al. () provide original and convincing empirical evidence of local adaptation to temperature in black alder, Alnus glutinosa L. Gaertn, a surprisingly understudied keystone species supporting riparian ecosystems. Here, De Kort et al. () use an innovative empirical approach complementing state-of-the-art landscape genomics analysis of A. glutinosa populations sampled in natura across a regional climate gradient with phenotypic trait assessment in a common garden experiment (Fig. ). By combining the two methods, De Kort et al. () were able to detect unequivocal association between temperature and phenotypic traits such as leaf size as well as with genetic loci putatively under divergent selection for temperature. The research by De Kort et al. () provides valuable insight into adaptive response to temperature variation for an ecologically important species and demonstrates the usefulness of an integrated approach for empirical evaluation of local adaptation in nonmodel species (Sork et al. ).

Adaptation to local climate in multi-trait space: evidence from silver fir (Abies alba Mill.) populations across a heterogeneous environment

Abstract:Heterogeneous environments, such as mountainous landscapes, create spatially varying selection pressure that potentially affects several traits simultaneously across different life stages, yet little is known about the general patterns and drivers of adaptation in such complex settings. We studied silver fir (Abies alba Mill.) populations across Switzerland and characterized its mountainous landscape using downscaled historical climate data. We sampled 387 trees from 19 populations and genotyped them at 374 single-nucleotide polymorphisms (SNPs) to estimate their demographic distances. Seedling morphology, growth and phenology traits were recorded in a common garden, and a proxy for water use efficiency was estimated for adult trees. We tested whether populations have more strongly diverged at quantitative traits than expected based on genetic drift alone in a multi-trait framework, and identified potential environmental drivers of selection. We found two main responses to selection: (i) populations from warmer and more thermally stable locations have evolved towards a taller stature, and (ii) the growth timing of populations evolved towards two extreme strategies, 'start early and grow slowly' or 'start late and grow fast', driven by precipitation seasonality. Populations following the 'start early and grow slowly' strategy had higher water use efficiency and came from inner Alpine valleys characterized by pronounced summer droughts. Our results suggest that contrasting adaptive life-history strategies exist in silver fir across different life stages (seedling to adult), and that some of the characterized populations may provide suitable seed sources for tree growth under future climatic conditions.

Abstract:Plants respond to rapid environmental change in ways that depend on both their genetic identity and their phenotypic plasticity, impacting their survival as well as associated ecosystems. However, genetic and environmental effects on phenotype are difficult to quantify across large spatial scales and through time. Leaf hyperspectral reflectance offers a potentially robust approach to map these effects from local to landscape levels. Using a handheld field spectrometer, we analyzed leaf‐level hyperspectral reflectance of the foundation tree species Populus fremontii in wild populations and in three 6‐year‐old experimental common gardens spanning a steep climatic gradient. First, we show that genetic variation among populations and among clonal genotypes is detectable with leaf spectra, using both multivariate and univariate approaches. Spectra predicted population identity with 100% accuracy among trees in the wild, 87%–98% accuracy within a common garden, and 86% accuracy across different environments. Multiple spectral indices of plant health had significant heritability, with genotype accounting for 10%–23% of spectral variation within populations and 14%–48% of the variation across all populations. Second, we found gene by environment interactions leading to population‐specific shifts in the spectral phenotype across common garden environments. Spectral indices indicate that genetically divergent populations made unique adjustments to their chlorophyll and water content in response to the same environmental stresses, so that detecting genetic identity is critical to predicting tree response to change. Third, spectral indicators of greenness and photosynthetic efficiency decreased when populations were transferred to growing environments with higher mean annual maximum temperatures relative to home conditions. This result suggests altered physiological strategies further from the conditions to which plants are locally adapted. Transfers to cooler environments had fewer negative effects, demonstrating that plant spectra show directionality in plant performance adjustments. Thus, leaf reflectance data can detect both local adaptation and plastic shifts in plant physiology, informing strategic restoration and conservation decisions by enabling high resolution tracking of genetic and phenotypic changes in response to climate change.

Mapping canopy foliar functional traits in a mixed temperate forest using imaging spectroscopy

Abstract:Foliar functional traits are key drivers of ecological processes in forests. Despite progress in forest foliar trait mapping from imaging spectroscopy, there is a need to build environment-specific, spectra-trait models trained from tree-level measurements to improve the accuracy of local trait maps. We mapped 12 foliar functional traits in a mixed temperate forest using airborne imaging spectroscopy. Top-of-canopy foliar samples from tree crowns (n = 166), representing a total of 16 species, were collected using a drone platform to measure foliar traits for individual trees, from which tree-level crown spectra were also determined. Partial least squares regression (PLSR) models were used to predict foliar traits from tree-level reflectance spectra (400-2400 nm). These models predicted leaf mass per area (LMA), specific leaf area (SLA) and equivalent water thickness (EWT) with high accuracy (R2 > 0.8, %RMSE < 15). Models for pigment, nitrogen and cellulose concentrations showed a moderate performance (R2 = 0.53-0.68, %RMSE = 17.24-21.31). Poorest performance was observed for lignin, carbon, leaf dry mass content (LDMC) and hemicellulose (R2 = 0.24-0.44, %RMSE = 20.67-26.13). High-resolution (1.25 m pixel-1) foliar trait maps were produced for the entire 16-km2 study area. Our study adds to the extensive research aiming to use remote sensing to monitor forest functional trait biodiversity at larger scales and provides models that capture intraspecific variation across many tree species from a mixed temperate forest in eastern Canada.

Abstract:Remote sensing is increasingly used in forest inventories. However, its application to assess genetic variation in forest trees is still rare, particularly in conifers. Here we evaluate the potential of LiDAR and RGB imagery obtained through unmanned aerial vehicles (UAVs) as high-throughput phenotyping tools for the characterization of tree growth and crown structure in two representative Mediterranean pine species. To this end, we investigated the suitability of these tools to evaluate intraspecific differentiation in a wide array of morphometric traits for Pinus nigra (European black pine) and Pinus halepensis (Aleppo pine). Morphometric traits related to crown architecture and volume, primary growth, and biomass were retrieved at the tree level in two genetic trials located in Central Spain and compared with ground-truth data. Both UAV-based methods were then tested for their accuracy to detect genotypic differentiation among black pine and Aleppo pine populations and their subspecies (black pine) or ecotypes (Aleppo pine). The possible relation between intraspecific variation of morphometric traits and life-history strategies of populations was also tested by correlating traits to climate factors at origin of populations. Finally, we investigated which traits distinguished better among black pine subspecies or Aleppo pine ecotypes. Overall, the results demonstrate the usefulness of UAV-based LiDAR and RGB records to disclose tree architectural intraspecific differences in pine species potentially related to adaptive divergence among populations. In particular, three LiDAR-derived traits related to crown volume, crown architecture, and main trunk—or, alternatively, the latter (RGB-derived) two traits—discriminated the most among black pine subspecies. In turn, Aleppo pine ecotypes were partly distinguishable by using two LiDAR-derived traits related to crown architecture and crown volume, or three RGB-derived traits related to tree biomass and main trunk. Remote-sensing-derived-traits related to main trunk, tree biomass, crown architecture, and crown volume were associated with environmental characteristics at the origin of populations of black pine and Aleppo pine, thus hinting at divergent environmental stress-induced local adaptation to drought, wildfire, and snowfall in both species.

Abstract:Populus species with wide geographic ranges display strong adaptation to local environments. We studied the clinal patterns in phenology and ecophysiology in allopatric Populus species adapted to similar environments on different continents under common garden settings. As a result of climatic adaptation, both Populus tremula L. and Populus balsamifera L. display latitudinal clines in photosynthetic rates (A), whereby high-latitude trees of P. tremula had higher A compared to low-latitude trees and nearly so in P. balsamifera (p = 0.06). Stomatal conductance (g s) and chlorophyll content index (CCI) follow similar latitudinal trends. However, foliar nitrogen was positively correlated with latitude in P. balsamifera and negatively correlated in P. tremula. No significant trends in carbon isotope composition of the leaf tissue (δ(13)C) were observed for both species; but, intrinsic water-use efficiency (WUEi) was negatively correlated with the latitude of origin in P. balsamifera. In spite of intrinsically higher A, high-latitude trees in both common gardens accomplished less height gain as a result of early bud set. Thus, shoot biomass was determined by height elongation duration (HED), which was well approximated by the number of days available for free growth between bud flush and bud set. We highlight the shortcoming of unreplicated outdoor common gardens for tree improvement and the crucial role of photoperiod in limiting height growth, further complicating interpretation of other secondary effects.

Abstract:1.Climate warming is modifying the movement of air masses over Northern latitudes, producing warming and cooling events across the boreal regions. These new conditions changes may mismatch plant phenology from weather conditions, and affect the growing period of trees. Understanding the processes of local adaptation in bud phenology can help to predict the response of plants to these rapid and unexpected environmental changes.2.Our study monitored bud burst and bud set weekly during four growing seasons in black spruce [Picea mariana (Mill.) B.S.P.] saplings planted in a common garden and originating from five provenances representing the whole latitudinal distribution of the closed boreal forest in Quebec, Canada. We compared the variance in bud phenology among populations and years, and analyzed the relationships with temperatures at the origin sites.3.Bud burst and bud set occurred in mid‐May and mid‐July, respectively, with a large variability among provenances and between the study years. A delayed bud phenology was observed in the provenances from warmer sites, with bud burst and bud set being 1.1 and 1.4 days later for every additional degree in mean annual temperature at the origin site, respectively. Populations with earlier bud bursts also showed earlier bud sets, thus the growing season was similar among provenances. The heritability of bud set was higher than that of bud burst, with estimates of 0.26 and 0.21, respectively. On average, variance in bud phenology among provenances reached 5.3%, which was higher than that within provenances (2.6%). The factor year explained 37.7‐69.7% of the variance in bud phenology.4.Synthesis. The findings demonstrate the evolutionary response to climate variations in the boreal biome. Endogenous and environmental components have different effects on the phases of bud burst and bud set. This complex interaction among drivers of bud phenology may prevent reliable predictions of changes in the growing season under changing climate. An earlier growth reactivation due to higher spring temperatures could result in higher frost risks to the developing buds. Despite a longer period for photosynthesis expected for the evergreen black spruce under warmer conditions, the period spent for bud development could remain unchanged.

Abstract:Understanding the genetic basis of local adaptation is challenging due to the subtle balance among conflicting evolutionary forces that are involved in its establishment and maintenance. One system with which to tease apart these difficulties is clines in adaptive characters. Here we analyzed genetic and phenotypic variation in bud set, a highly heritable and adaptive trait, among 18 populations of Norway spruce (Picea abies), arrayed along a latitudinal gradient ranging from 47°N to 68°N. We confirmed that variation in bud set is strongly clinal, using a subset of five populations. Genotypes for 137 single-nucleotide polymorphisms (SNPs) chosen from 18 candidate genes putatively affecting bud set and 308 control SNPs chosen from 264 random genes were analyzed for patterns of genetic structure and correlation to environment. Population genetic structure was low (F(ST) = 0.05), but latitudinal patterns were apparent among Scandinavian populations. Hence, part of the observed clinal variation should be attributable to population demography. Conditional on patterns of genetic structure, there was enrichment of SNPs within candidate genes for correlations with latitude. Twenty-nine SNPs were also outliers with respect to F(ST). The enrichment for clinal variation at SNPs within candidate genes (i.e., SNPs in PaGI, PaPhyP, PaPhyN, PaPRR7, and PaFTL2) indicated that local selection in the 18 populations, and/or selection in the ancestral populations from which they were recently derived, shaped the observed cline. Validation of these genes using expression studies also revealed that PaFTL2 expression is significantly associated with latitude, thereby confirming the central role played by this gene in the control of phenology in plants.
Quantifying vertical profiles of biochemical traits for forest plantation species using advanced remote sensing approaches

Abstract:Biochemical traits in forest vegetation are key indicators of leaf physiological processes, specifically photosynthetic and other photochemical light pathways, and are critical to the quantification of the terrestrial carbon cycle. Advances in remote sensing sensors and platforms are allowing multi-dimensional and continuous-spatial information to be acquired in a fast and non-destructive way to quantify forest biochemical traits at multiple spatial scales. Here we demonstrate the use of high spectral resolution, hyperspectral data combined with high density three-dimensional information from Light Detection and Ranging (LiDAR) both acquired from an unmanned aerial system (UAS) platform, to quantify and assess the three-dimensional distribution of biochemical pigments on individual tree canopy surfaces. To do so, a DSM based fusion method was developed to integrate the 3D LiDAR point cloud with hyperspectral reflectance data. Regression-based models were then developed to predict a number of biochemical traits (i.e., chlorophyll (Chl) a, b, total Chl and total carotenoids (Cars) content) from a suite of common spectral indices at three vertical canopy levels, and were evaluated using a leave-one-out cross-validation approach. One-way ANOVA and Duncan's multiple comparison post hoc tests were used to investigate the vertical distribution of biochemical pigments on individual tree canopy surfaces, and in response to age and species. Our results demonstrated that a number of vegetation indices, derived from the hyperspectral data, were strongly correlated with a number of biochemical traits (Adj-R2 = 0.85–0.91; rRMSE = 5.19–6.38%). In general, models fitted using leaf samples from the upper, middle and lower canopies separately (Adj-R2 = 0.85–0.91; rRMSE = 5.19–6.38%) had similar accuracy to the models developed with pooled data (Adj-R2 = 0.87–0.90; rRMSE = 5.21–6.11%). The differences between separate models and global models were not statistically significant (P > 0.05). However, the distribution of biochemical pigments across vertical layers varied significantly. For dawn redwood (Metasequoia glyptostroboides) and poplar (Populus deltoides), the results were consistent in that the lower component of the canopy (least light impacted) had the highest chlorophyll and carotenoids biochemical traits. Moreover, the vertical distribution of biochemical traits on individual tree canopy surfaces changed with age likely due to the growth variation from the photosynthetic activity of the canopy. This study indicates the potential of using fused 3D point cloud information with spectral data to monitor physiological activities of forest canopy for carbon accumulation estimation as well as precision forestry applications such as nutrition diagnosis, water regulation and subsequent productivity enhancement of these planted forest systems.

Abstract:Cycles of glaciation have induced phases of species expansion and contraction, occasionally occurring at continental scales. Our study, employing both ecological and genetic approaches, indicates that hybridization between Norway and Siberian spruce aided both species in adapting to climate change. Current global climate change is expected to affect biodiversity negatively at all scales leading to mass biodiversity loss. Many studies have shown that the distribution of allele frequencies across a species' range is often influenced by specific genetic loci associated with local environmental variables. This association reflects local adaptation and allele changes at those loci could thereby contribute to the evolutionary response to climate change. However, predicting how species will adapt to climate change from this type of data alone remains challenging. In the present study, we combined exome capture sequences and environmental niche reconstruction, to test multiple methods for assessing local adaptation and climate resilience in two widely distributed conifers, Norway spruce and Siberian spruce. Both species are keystone species of the boreal forest and share a vast hybrid zone. We show that local adaptation in conifers can be detected through allele frequency variation, population‐level ecological preferences, and historical niche movement. Moreover, we integrated genetic and ecological information into genetic offset predictive models to show that hybridization plays a central role in expanding the niche breadth of the two conifer species and may help both species to cope better with future changing climates. This joint genetic and ecological analysis also identified spruce populations that are at risk under current climate change.

Abstract:The health of coniferous forests in the western U.S. is under threat from mega-drought events, increasing vulnerability to insects, disease, and mortality. Forest densification resulting from fire exclusion increases these susceptibilities. Silvicultural treatments to reduce stand density and promote resilience to both fire and drought have been used to reduce these threats but there are few quantitative evaluations of treatment effectiveness. This proof-of-concept study focused on such an evaluation, using field and remote sensing metrics of mature ponderosa pine (Pinus ponderosa Doug. Laws) in central Oregon. Ground metrics included direct measures of transpiration (sapflow), branch and needle measures and chlorosis; drone imagery included thermal (TIR) and five-band spectra (R, G, B, Re, NIR). Thermal satellite imagery was derived from ECOSTRESS, a space-borne thermal sensor that is on-board the International Space Station (ISS). All metrics were compared over 2 days at a time of maximum seasonal drought stress (August). Tree water status in unthinned, light, and heavy thinning from below density reduction treatments was evaluated. Tree crowns in the heavy thin site had greater transpiration and were cooler than those in the unthinned site, while the light thin site was not significantly cooler than either unthinned or the heavy thin site. There was a poor correlation (Adj. R2 0.10–0.13) between remotely sensed stand temperature and stand-averaged transpiration, and tree level temperature and transpiration (Adj. R2 0.04–0.19). Morphological attributes such as greater needle chlorosis and reduced elongation growth supported transpirational indicators of tree drought stress. The multispectral indices CCI and NDRE, along with the NIR and B bands, show promise as proxies for crown temperature and transpiration, and may serve as a proof of concept for an approach to evaluate forest treatment effectiveness in reducing tree drought stress.

Abstract:Abstract The environment could alter growth and resistance tradeoffs in plants by affecting the ratio of resource allocation to various competing traits. Yet, how and why functional tradeoffs change over time and space is poorly understood particularly in long-lived conifer species. By establishing four common-garden test sites for five lodgepole pine populations in western Canada, combined with genomic sequencing, we revealed the decoupling pattern and genetic underpinnings of tradeoffs between height growth, drought resistance based on δ13C and dendrochronology, and metrics of pest resistance based on pest suitability ratings. Height and δ13C correlation displayed a gradient change in magnitude and/or direction along warm-to-cold test sites. All cold test sites across populations showed a positive height and δ13C relationship. However, we did not observe such a clinal correlation pattern between height or δ13C and pest suitability. Further, we found that the study populations exhibiting functional tradeoffs or synergies to various degrees in test sites were driven by non-adaptive evolutionary processes rather than adaptive evolution or plasticity. Finally, we found positive genetic relationships between height and drought or pest resistance metrics and probed five loci showing potential genetic tradeoffs between northernmost and the other populations. Our findings have implications for deciphering the ecological, evolutionary, and genetic bases of the decoupling of functional tradeoffs due to environmental change.
