Abstract:The aim of this study was to analyze soybean proteins as sources of peptides likely to be bitter using fragmentomic and hybrid approaches involving in silico and in vitro studies. The bitterness of peptides (called parent peptides) was theoretically estimated based on the presence of bitter-tasting motifs, particularly those defined as bitter-tasting indicators. They were selected based on previously published multilinear stepwise regression results. Bioinformatic-assisted analyses covered the hydrolysis of five major soybean-originating protein sequences using bromelain, ficin, papain, and proteinase K. Verification of the results in experimental conditions included soy protein concentrate (SPC) hydrolysis, RP-HPLC (for monitoring the proteolysis), and identification of peptides using RP-HPLC-MS/MS. Discrepancies between in silico and in vitro results were observed when identifying parent peptide SPC hydrolysate samples. However, both analyses revealed that conglycinins were the most abundant sources of parent peptides likely to taste bitter. The compatibility percentage of the in silico and in vitro results was 3%. Nine parent peptides with the following sequences were identified in SPC hydrolysates: LSVISPK, DVLVIPLG, LIVILNG, NPFLFG, ISSTIV, PQMIIV, PFPSIL, DDFFL, and FFEITPEK (indicators are in bold). The fragmentomic idea of research might provide a supportive method for predicting the bitterness of hydrolysates. However, this statement needs to be confirmed experimentally.

Abstract:The bitterness of soy protein isolate hydrolysates prepared using five proteases at varying degree of hydrolysis (DH) and its relation to physicochemical properties, i.e., surface hydrophobicity (H0), relative hydrophobicity (RH), and molecular weight (MW), were studied and developed for predictive modelling using machine learning. Bitter scores were collected from sensory analysis and assigned as the target, while the physicochemical properties were assigned as the features. The modelling involved data pre-processing with local outlier factor; model development with support vector machine, linear regression, adaptive boosting, and K-nearest neighbors algorithms; and performance evaluation by 10-fold stratified cross-validation. The results indicated that alcalase hydrolysates were the most bitter, followed by protamex, flavorzyme, papain, and bromelain. Distinctive correlation results were found among the physicochemical properties, influenced by the disparity of each protease. Among the features, the combination of RH-MW fitted various classification models and resulted in the best prediction performance.
Mastering the art of taming: Reducing bitterness in fish by-products derived peptides

Abstract:Processed fish by-products are valuable sources of peptides due to their high protein content. However, the bitterness of these peptides can limit their use. This review outlines the most recent advancements and information regarding the reduction of bitterness in fish by-products derived peptides. The sources and factors influencing bitterness, the transduction mechanisms involved, and strategies for reducing bitterness are highlighted. Bitterness in peptides is mainly influenced by the source, preparation method, presence of hydrophobic amino acid groups, binding to bitter receptors, and amino acid sequence. The most widely utilized techniques for eliminating bitterness or enhancing taste include the Maillard reaction, encapsulation, seperating undesirable components, and bitter-blockers. Finally, a summary of the current challenges and future prospects in the domain of fish by-products derived peptides is given. Despite some limitations, such as residual bitterness and limited industrial application, there is a need for further research to reduce the bitterness of fish by-products derived peptides. To achieve this goal, future studies should focus on the technology of fish by-products derived peptide bitterness diminishment, with the aim of producing high-quality products that meet consumer expectations.

Abstract:Biogenic amines (BAs) are a class of bioactive organics produced during the fermentation of soy sauce. A high concentration of BAs may bring about serious physiological and toxicological effects on the human body. In this study, we reported an optimized process to produce soy sauce with lower BA concentration and found the contents of putrescine, cadaverine and histamine increased with the increase of fermentation temperature but decreased with the increase of NaCl concentration. The final content of total BAs with improved fermentation was 105.56 +/- 0.13 mg/L, which was reduced by 89.11% compared to traditional brewing. Besides, the pilot production test was performed to verify the optimized conditions and physicochemical indexes were measured to better understand the change principle of the chemical compounds. Taken together, we present an effective process to inhibit the formation of BAs while ensuring that characteristic nutrients are not lost.

Abstract:Bitter peptides are a structurally diverse group of oligopeptides often generated in fermented, aged, and hydrolyzed food products that make them unfavorable for consumption. Humans perceive bitterness by a repertoire of 25 human bitter receptors, termed T2Rs. Knowledge of the structural features of bitter receptors and of the factors that stimulate bitter receptors will aid in understanding the mechanism responsible for bitter taste perception. This article reviews the current knowledge regarding structural features of bitter peptides and bitter taste receptors.

Abstract:Bitter peptides are small molecular peptides produced by the hydrolysis of proteins under acidic, alkaline, or enzymatic conditions. These peptides can enhance food flavor and offer various health benefits, with attributes such as antihypertensive, antidiabetic, antioxidant, antibacterial, and immune-regulating properties. They show significant potential in the development of functional foods and the prevention and treatment of diseases. This review introduces the diverse sources of bitter peptides and discusses the mechanisms of bitterness generation and their physiological functions in the taste system. Additionally, it emphasizes the application of bioinformatics in bitter peptide research, including the establishment and improvement of bitter peptide databases, the use of quantitative structure-activity relationship (QSAR) models to predict bitterness thresholds, and the latest advancements in classification prediction models built using machine learning and deep learning algorithms for bitter peptide identification. Future research directions include enhancing databases, diversifying models, and applying generative models to advance bitter peptide research towards deepening and discovering more practical applications.

Abstract:Bitter taste is an aversive taste because it is unconsciously associated with toxic compounds. However, a considerable variability in bitter sensitivity exists in those who have the genetic polymorphism for bitter taste receptors (TAS2Rs). Besides the oral cavity, TAS2Rs are present in many body tissues, including the gastrointestinal tract; therefore, they are crucial players both in the gustatory/hedonic system and in the homeostatic system, triggering numerous biological responses, including adipogenesis, carcinogenesis, or immunity. Bitter-tasting compounds are widely distributed in plant and animal foods and belong to many chemical classes. In this study, the evidence was reviewed on bitter peptides, considering the food sources, their formation in food under different processing and storage conditions and in the gastrointestinal tract during digestion, as well as their biological activities. Bitterness associated with peptides is due to the presence of hydrophobic amino acids in the C-terminus. The current literature mainly explores the enzymes and hydrolysis conditions, with the aim of reducing the formation of bitter peptides in hydrolysate preparation or food. Few studies highlight the bioactivity (namely, antihypertensive, antidiabetic, antioxidant, or immunity boosting), besides the bitterness. However, encapsulation of bitter peptides has been tentatively used to develop antihypertensive and antidiabetic supplements. In the era of personalized nutrition and precision medicine, the evidence available suggests the opportunity to use bitter bioactive peptides as functional ingredients in food. Such types of food may modulate a plethora of physiological mechanisms by targeting TAS2Rs in the gastrointestinal tract, thus modulating appetite sensations or gastrointestinal motility and discomfort according to individual nutritional needs and goals. More studies are needed to optimize the technological strategies to target TAS2Rs by bitter bioactive peptides, improve their stability in food, and validate the biological efficacy through well-designed in vivo studies.

Abstract:Soybean functional peptides (SFPs) are obtained via the hydrolysis of soybean protein into polypeptides, oligopeptides, and a small amount of amino acids. They have nutritional value and a variety of functional properties, including regulating blood lipids, lowering blood pressure, anti-diabetes, anti-oxidant, preventing COVID-19, etc. SFPs have potential application prospects in food processing, functional food development, clinical medicine, infant milk powder, special medical formulations, among others. However, bitter peptides containing relatively more hydrophobic amino acids can be formed during the production of SFPs, seriously restricting the application of SFPs. High-quality confirmatory human trials are needed to determine effective doses, potential risks, and mechanisms of action, especially as dietary supplements and special medical formulations. Therefore, the physiological activities and potential risks of soybean polypeptides are summarized, and the existing debitterness technologies and their applicability are reviewed. The technical challenges and research areas to be addressed in optimizing debittering process parameters and improving the applicability of SFPs are discussed, including integrating various technologies to obtain higher quality functional peptides, which will facilitate further exploration of physiological mechanism, metabolic pathway, tolerance, bioavailability, and potential hazards of SFPs. This review can help promote the value of SFPs and the development of the soybean industry.

Abstract:Over the past few decades, there has been a noticeable surge in the market of plant-based meat analogs (PBMA). Such popularity stems from their environmentally friendly production procedures as well as their positive health effects. In order to meet the market demand, it is necessary to look for plant protein processing techniques that can help them match the quality of conventional meat protein from the aspects of sensory, quality and functionality. Bean proteins are ideal options for PBMA with their easy accessibility, high nutrient-density and reasonable price. However, the high polyunsaturated lipids content of beans inevitably leads to the unpleasant beany flavor of soy protein products, which severely affects the promotion of soy protein-based PBMA. In order to solve this issue, various methods including bleaching, enzyme and fermentation etc. are developed. Among these, fermentation is widely investigated due to its high efficiency, less harm to the protein matrix, targeted performance and low budget. In addition, proper utilization of microbiome during the fermentation process not only reduces the unpleasant beany flavors, but also enhances the aroma profile of the final product. In this review, we provide a thorough and succinct overview of the mechanism underlying the formation and elimination of beany flavor with associated fermentation process. The pros and cons of typical fermentation technologies for removing beany flavors are discussed in alongside with their application scenarios. Additionally, the variations among different methods are compared in terms of the strains, fermentation condition, target functionality, matrix for application, sensory perception etc.

Abstract:Compounds that confer a bitter taste on fruits and vegetables (FAVs) play crucial roles in both plant defense and health promotion. This review details the current knowledge of the distribution, properties (toxicity, pharmacological effects and receptors) and environmental plant responses relating to the biosynthesis, catabolism and transcriptional regulation of 53 bitter plant metabolites in diverse species of FAVs. Some bitter compounds, such as flavonoids, are common in all plant species and make a minor contribution to bitter flavor, but many are synthesized only in specific taxa. They make major contributions to the bitter taste of the corresponding species and some also have significant pharmacological effects. Levels of bitter metabolites are genetically determined, but various environmental cues can affect their final concentration during preharvest development and postharvest storage processes. Molecular approaches are helping to unravel the mechanisms of biosynthesis and regulation of bitter compounds in diverse crop species. This review not only discusses the theoretical basis for utilizing breeding programs and other agricultural technologies to produce FAVs with improved safety, favorable taste and healthier profiles, but also suggests new directions for the utilization of bitter compounds in FAVs for the development of natural pesticides and health-promoting medicines.

Abstract:Hexose-modified peptides, products of the enzymatic hydrolysis of glycated proteins, could be used as markers of diabetes mellitus, the aging process and other diseases. The main difficulty in this approach is the detection of glycated peptides in the complex mixtures of compounds. In this study we investigated the formation of borate complexes of the peptide-derived Amadori products by high-resolution mass spectrometry (HRMS) and tandem mass spectrometry (MS/MS) experiments. It was found that the formation of a complex with the borate ion stabilizes the sugar moiety, resulting in the simplification of the fragmentation patterns of peptide-derived Amadori products. The level of dehydration, as well as the elimination of formaldehyde from the precursor ions of borate complexes, was lower as compared to the free peptide. On the other hand the intensity of the b- and y-type ions for borate complexes is significantly higher in comparison to the free peptide-derived Amadori product. Moreover, the elimination of a whole hexose moiety was not detected in the examined peptides.

Abstract:Background: The bitter and astringent taste of tea and tea extract is still not favored by many consumers, which limits its applications to bakery foods and functional foods. Many bitter and astringent compounds in tea leaves are susceptible, which transform during tea processing or can be modified by food production technologies. Besides, the accumulations of the major bitter and astringent compounds in fresh tea leaves, such as alkaloids and flavonoids, can be regulated by agronomic practices. Many attempts have been made to reduce the bitter and astringent compounds, in order to improve the sensory quality of tea and tea products. Scope and approach: The aim of this review is to describe the characteristics of major bitter and astringent compounds in different types of teas as well as the bitterness and astringency reducing strategies from the aspects of preharvest, processing and post-processing.Key findings and conclusions: Alkaloids, catechins, anthocyanins, phenolic acids, flavonol glycosides, and thea-flavins importantly contribute to the bitter and astringent taste of tea infusion, the contents of which can be modulated during preharvest, processing, and post-processing. Shade treatment is a conventional agronomic practice to reduce the bitterness and astringency of tea. Processing methods like oxidation, roasting/baking and microbial fermentation effectively reduce the bitter and astringent intensities of teas, but have restricted ap-plications to special types of tea. There is a need of new compounds or strategies to decrease the bitterness and astringency of tea and tea products. Employment of these bitterness and astringency reducing strategies should consider the appearance or application requirements of final products.
Microbiology, flavor formation, and bioactivity of fermented soybean curd (furu): A review

Abstract:Soybeans are an important plant-based food but its beany flavor and anti-nutritional factors limit its consumption. Fermentation is an effective way to improve its flavor and nutrition. Furu is a popular fermented soybean curd and mainly manufactured in Asia, which has been consumed for thousands of years as an appetizer because of its attractive flavors. This review first classifies furu products on the basis of various factors; then, the microorganisms involved in its fermentation and their various functions are discussed. The mechanisms for the formation of aroma and taste compounds during fermentation are also discussed; and the microbial metabolites and their bioactivities are analyzed. Finally, future prospects and challenges are introduced and further research is proposed. This information is needed to protect the regional characteristics of furu and to regulate its consistent quality. The current information suggests that more in vivo experiments and further clinical trials are needed to confirm its safety and the microbial community needs to be optimized and standardized for each type of furu to improve the production process.

Abstract:Bitterness, one of the 5 “basic tastes”, is usually undesired by humans. However, abundant literature reported that bitter fruits and vegetables have beneficial health effects due to their bitter contributors. This review provided an updated overview of the main bitter contributors of typical bitter fruits and vegetables and their health benefits. The main bitter contributors, including phenolics, terpenoids, alkaloids, amino acids, nucleosides and purines, were summarized. The bioactivities and wide range of beneficial effects of them on anti-cancers, anti-inflammations, anti-microbes, neuroprotection, inhibiting chronic and acute injury in organs, as well as regulating behavior performance and metabolism were reported. Furthermore, not only did the bitter taste receptors（taste receptor type 2 family, T2Rs） show taste effects, but extra-oral T2Rs could also be activated by binding with bitter components, regulating physiological activities via modulating hormone secretion, immunity, metabolism, and cell proliferation. This review provided a new perspective on exploring and explaining the nutrition of bitter foods, revealing the relationship between the functions of bitter contributors from food and T2Rs. Future trends may focus on revealing the possibility of T2Rs being targets for the treatment of diseases, exploring the mechanism of T2Rs mediating the bioactivities, and making bitter foods more acceptable without getting rid of bitter contributors.

Abstract:Pea protein is an emerging functional ingredient in structuring plant-based meat analogs. Due to its hypoallergenic, non-GMO, and high nutritional value , pea protein has succeeded in capturing the interests of scientists around the globe. Nevertheless, the beany flavor is a significant impediment to consumer acceptance of pea protein products, creating a gap for further research on the formation mechanism and removal methods of beany flavor is essential for the robust development of pea protein food and nutritional products. Herein, this review summarizes the key beany flavor compounds, their formation mechanisms, and interactions between pea protein and off-flavor compounds, along with the extrapolation of the extraction methods, detection methods, sensory evaluation, and the application of omics techniques for flavor analyses. The possible metabolic pathways of related microorganisms to reduce beany flavor in pea protein are also discussed. The formation of beany flavor is a complex process; the lipoxygenase-hydroperoxide lyase mechanism is to be held responsible for the main formation pathway, interaction between flavor compounds and pea protein, as well as the interaction between flavor compounds, collectively impact the overall flavor profiles. Modern food flavor analysis techniques combined with sensory evaluation provide more accurate support for the identification and analyzing beany flavor. Microbial fermentation is to be considered a thought-provoking strategy for decreasing off-flavor. At the same time, its metabolic mechanism requires further exploration, still proving to be an allusion for the development and delivery of plant-based foods with a lessened beany flavor.

Abstract:Bioactive peptides (BAPs) with potential health benefits are gaining popularity as people become more health-conscious. Fermented foods are produced through the vigorous metabolic activities of numerous microorganisms, which convert macromolecules in the food matrix into bioactive components, including BAPs. Access to endogenous BAPs in fermented foods reduces costs while simplifying isolation steps and provides a high-quality direct source of BAPs. Fermented foods-derived BAPs have been shown to have antihypertensive, antioxidant, and antibacterial activities; however, there is a lack of information on the production of BAPs through fermented foods, and a large number of BAPs resources in fermented foods have not been effectively explored. Therefore, this review introduces the current research status, preparation methods, biological functions, and digestive stability of BAPs in fermented foods, followed by a discussion of the shortcomings in current research and future directions to provide a foundation for applications of BAPs in fermented foods in the food and pharmaceutical fields.

Abstract:A new bittern soy sauce was developed; the overall aim of this study was to characterize the quality of the soy sauce. Biochemical changes occurring during experimental soy sauce fermentations were described. Results revealed that, the contents of amino nitrogen, reducing sugars and enzyme activity were slightly lower in soy sauce prepared with bittern than with traditional one. No marked difference in pH was noted between the two types of soy sauce.

Abstract:With a shift toward plant-based foods, pulses and grains belonging to the Fabaceae family, specifically pea, faba, or soybean varieties, have received attention recently as alternative protein sources due to their high protein content, nutritional profile, and emulsifying properties. Maillard reaction, caramelization of carbohydrates, or oxidation of lipids lead to generation and retention of bitter or beany off-flavors in these pulses and grains. Based on protein type, concentration, and functional groups of off-flavor compounds and their positioning, reversible or irreversible interactions between proteins and off-flavors occur that influence the taste of food products. The generation, retention, or interaction of off-flavors with these plant-based proteins can be controlled through different approaches. Bitter taste can be reduced during enzymatic hydrolysis or germination/sprouting due to the release of amino acids or masked during fermentation as a result of the generation of fruity or floral flavors. The pH variation or salt addition during protein extraction can modulate flavor-protein binding. Moreover, modification of proteins alters their surface hydrophobicity and charge, contributing to the reduction of off-flavors. Heating, exposure to electromagnetic field, or addition of biotic elicitors can decrease lipid oxidation and content of off-flavors with beany or grassy notes. The selected storage conditions and packaging materials also impact off-flavor retention and generation during storage. In light of the foregoing, this review article discusses recently followed processing approaches for controlling off-flavors in pea, faba, and soybeans as protein sources while providing an overview of recent studies performed on improving the sensory properties of food products produced from these alternative proteins.

Production and Functionality of Food-derived bioactive peptides: A review.

Abstract:Proteins are present in a wide range of our everyday foods and complex macromolecules made up of one or more polypeptide chains. From these common foods, a large number of bioactive peptides can be produced. An array of biological health effects has been attributed to these peptides, including antioxidative, antihypertensive, antidiabetic, anticoagulant, antithrombotic, anti-inflammatory, hypocholesterolemic and immunomodulating. The chemical structure of biologically active peptides is closely related to its specific biological activity. Furthermore, the amino acid composition and sequence information of peptide molecular chain have an important influence on its physiological activity. Food-derived bioactive peptides are typically released from animal and plant proteins via enzymatic hydrolysis under in vitro or in vivo conditions using different kinds of protease. However, the production, separation and purification of bioactive peptides with desired specific functions have become a focus and challenge. In addition, further studies are needed on the mechanism of action and multiple biological functionalities. Theresent review focuses on the recent biotechnological advances in the production, purification and identification of bioactive peptides were. Structure-activity relationship and methods to evaluate several common food-derived bioactive peptides were reviewed as well.

Abstract:Fermentation is a critical technological process for flavor development in fermented foods. The combination of odor and taste, known as flavor, is crucial in enhancing people's perception and psychology toward fermented foods, thereby increasing their acceptance among consumers. This review summarized the determination and key flavor compound screening methods in fermented foods and analyzed the flavor perception, perceptual interactions, and evaluation methods. The flavor compounds in fermented foods could be separated, purified, and identified by instrument techniques, and a molecular sensory science approach could identify the key flavor compounds. How flavor compounds bind to their respective receptors determines flavor perception, which is influenced by their perceptual interactions, including odor-odor, taste-taste, and odor-taste. Evaluation methods of flavor perception mainly include human sensory evaluation, electronic sensors and biosensors, and neuroimaging techniques. Among them, the biosensor-based evaluation methods could facilitate the investigation of the flavor transduction mechanism and the neuroimaging technique could explain the brain's signals that relate to the perception of flavor and how they compare to signals from other senses. This review aims to elucidate the flavor profile of fermented foods and highlight the significance of comprehending the interactions between various flavor compounds, thus improving the healthiness and sensory attributes.
