Abstract:The aqueous two-phase extraction process (APTE) for polysaccharides from the fruits of Ilex cornuta (Lindl. et Paxt.) (PFIC) was studied, and its monosaccharide composition and antioxidant activities were analyzed. After screening and selection of isopropanol/ammonium sulfate as the aqueous two-phase system, various parameters of crude extract amount, ultrasonic time, extraction temperature and extraction time on the extraction process was investigated in detail and optimized by response surface methodology (RSM). The obtained PFIC was characterized by FT-IR and its monosaccharide composition was analyzed by GC-MS. Furthermore, the antioxidant activities of PFIC were evaluated by measuring the scavenging rates of DPPH radical, hydroxyl radical and superoxide anion radical. Under the optimal conditions in which crude extract amount was 2.23 g, ultrasonic time was 17.5 min, extraction temperature was 69 â„ƒ and the extraction time was 1.55 h, the yield of polysaccharides reached 50.87% which was close to the predicted value of 50.78%. FT-IR analysis confirmed that the extract was polysaccharide, and GC-MS analysis indicated that the monosaccharide components of the polysaccharides were rhamnose, arabinose, xylose, mannose, glucose and galactose. Antioxidant results showed that PFIC showed strong scavenging ability to DPPH radical, fine scavenging ability to hydroxyl radical and certain scavenging ability to superoxide anion radical. The ultrasonic-assisted APTE process for PFIC optimized by RSM is reasonable and reliable, and the obtained PFIC has the potential to be utilized as natural antioxidants.
Ultrasonic-assisted Modifications of Macroporous Resin to Improve Anthocyanin Purification from a Pyrus Communis Var. Starkrimson Extract

Abstract:The present study presents an attempt to modify the surface properties of macroporous resins (MRs) in order to improve anthocyanin adsorption and desorption from Pyrus communis var Starkrimson fruit peel extract. A number of MRs were tested to optimise the ultrasonic-assisted adsorption (UAA) conditions; including ultrasonic power (100-400 W), resin-to-extract ratio (1-3 g/50 mL) and temperature (20-40 degrees C). Similarly, varying ultrasonic-assisted desorption (UAD) conditions were optimised; including ultrasonic power (200-600 W), resin-to-solvent ratio (1-4 g/50 mL), ethanol concentration (60-90% v/v) and temperature (20-40 degrees C). The Amberlyst 15 (H) cationic resin was found to be superior to the other tested resins. The maximum adsorption capacity (659 mu g/g) of cyanidin 3-galactoside (Cy 3-gal) was achieved under the optimised UAA conditions (400 W, 20 degrees C and 1 g/50 mL), while 616 mu g/g of Cy 3-gal was recovered under the optimised UAD conditions (582 W, 1 g/50 mL, 60% and 20 degrees C). Moreover, titratable-acid and total-sugar contents were found to be significantly lower under UAA than under conventional-assisted adsorption (CAA). ANOVA revealed that process factors had significant effects on the Cy 3-gal purification, as depicted by their linear, quadratic and interactive effects. While anthocyanin adsorption was found to be significantly improved at lower ultrasonic power, higher power promoted the desorption process. Adsorption under optimized UAA conditions followed pseudo second-order kinetics and multilayer adsorption (Freundlich isotherm) onto the Amberlyst 15 (H) resin surface was observed. The particle-size distribution curve and scanning electron microscopic images also revealed higher resin-surface roughness, peeling and the appearance of pores on the surface under ultrasonication. This is the first study to use ultrasonication to modify a cationic exchange resin for the improvement of Cy 3-gal purification from a fruit extract. This study can recommend the use of ultrasonication as a low-cost green technique that can improve macroporous resin characteristics for better purification of compounds from an extract

Abstract:Abstract Ultrasonic extraction was a new development method to achieve high-efficiency extraction of Lycium barbarum polysaccharides instead of hot water extraction. In this paper, the single factor method combined with the box Behnken design of response surface method was used to study the influence of extraction time, extraction temperature, material liquid ratio and ultrasonic power on the extraction rate of L . barbarum polysaccharide. The results indicated that the best extraction rate of L . barbarum polysaccharide was 12.54 ± 0.12% under the conditions of 80 min for extraction time, 73 °C for extraction temperature, 1 g:38 mL for material to liquid ratio, and 185 W for ultrasonic power. Under the same operating conditions, the yield of L . barbarum polysaccharide using ultrasonic extraction was 83.3%, which was higher than that hot water extraction. Moreover, the extraction time of ultrasonic extraction was only 47% of that using hot water extraction. This suggested that there was great potentials of using ultrasonic extraction in the realization of high-efficiency extraction of L. barbarum polysaccharide. The results of this study could also provide a theoretical basis for the coupling of ultrasonic extraction and ultrasonic concentration process to develop the integrated equipment of both ultrasonic extraction and ultrasonic concentration.
Optimization of ultrasonic extraction of polysaccharides from dried longan pulp using response surface methodology

Abstract:Ultrasonic technology was applied for polysaccharides extraction from the dried longan pulp and response surface methodology (RSM) was used to optimize the effects of processing parameters on polysaccharides yields. Three independent variables were ultrasonic power (X1), extraction time (X2) and ratio of water to raw material (X3), respectively. The statistical analysis indicated that three variables and the quadratic of X1 and X2 had significant effects on the yields, and followed by the significant interaction effects between the variables of X2 and X3 (p<.05). A mathematical model with high determination coefficient was gained and could be employed to optimize polysaccharides extraction. The optimal extraction conditions of polysaccharides were determined as follows: Ultrasonic power 680W, extraction time 4.5min, ratio of water to raw material 25mL/g. Under these conditions, the experimental yield of polysaccharides was 4.455±0.093%, which was agreed closely with the predicted value (4.469%).

Abstract:The aim of this study was to optimize the conditions for combined ultrasonication and enzymatic treatment of whole fruit Lycium barbarum. The synergistic effects of ultrasonication and enzymatic treatment on the fermentation process, bioactive compounds, and antioxidant activity were investigated. Optimal combination of sonication time, enzyme ratio (cellulase: pectinase: protease), and enzyme concentration was determined by optimization experiments to be 10 min, 1:1:1, and 0.75%, respectively. Under these conditions, the fermentation process greatly accelerated, the viable cell count of the lactic acid bacteria after fermentation for 36 h reached (5.57 ± 0.38) × 108 CFU g−1 compared with (8.63 ± 0.29) × 107 CFU g−1 for samples without pretreatment. The total phenol and flavonoid contents of Lycium barbarum with pretreatment after fermentation for 36 h increased by 10.69% and 19.62%, respectively, compared with samples without pretreatment. The highest antioxidant activities, as measured by a 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, hydroxyl radical scavenging assay, and superoxide anion scavenging assay, were obtained for the treated Lycium barbarum.

Abstract:Due to the large molecular weight and complex structure of Laminaria japonica polysaccharides (LJP), which limit their absorption and utilization by the body, methods to effectively degrade polysaccharides had received more and more attention. In the present research, hot water extraction coupled with three-phase partitioning (TPP) was developed to extract and isolate LJP. Ultrasonic L. japonica polysaccharides (ULJP) were obtained by ultrasonic degradation. In addition, their physicochemical characteristics and in vitro biological activities were investigated. Results indicated that ULJP had lower weight-average molecular weight (153 kDa) and looser surface morphology than the LJP. The primary structures of LJP and ULJP were basically unchanged, both contained α-hexo-pyranoses and were mainly connected by 1,4-glycosidic bonds. Compared with LJP, ULJP had stronger antioxidant activity, α-amylase inhibitory effect and anti-inflammatory effect on RAW264.7 macrophages. The scavenging rate of DPPH free radicals by ULJP is 35.85%. Therefore, ultrasonic degradation could effectively degrade LJP and significantly improve the biological activity of LJP, which provided a theoretical basis for the in-depth utilization and research and development of L. japonica in the fields of medicine and food.

Abstract:The effects of a synergized two-step subcritical water extraction (SWE) and simultaneous multi-frequency ultrasound-assisted alcohol/salt aqueous two-phase extraction (MFU-AATPE) on the yield, physicochemical, structural characteristics, and antioxidant activities of Lentinus edodes polysaccharides (LEPs) was investigated. Crude LEP extracted under subcritical extraction (31.14 %) was purified using dual and triple frequency ultrasound-assisted aqueous two-phase systems giving a yield of 94.57 % and 97 % respectively. Compared with dual-frequency sonication (20/40 kHz), triple frequency sonication (20/40/60 kHz) gave maximum yield and improved the desalination rate by 33.6 %. Congo red analysis revealed triple helix structures. GC analysis showed the existence of mannose, glucose, arabinose, xylose, and galactose whilst spectral similarities were observed through Fourier Transform Infrared Spectroscopy. The average molecular weight (Mw) was 83.6170 Da, 2384 Da and 2387 Da for control, dual and triple frequency samples respectively. Ultrasound treated polysaccharides displayed stronger antioxidant activities against the DPPH, ABTS and hydroxyl radicals, thus demonstrating their potency in reducing oxidation. However, the microstructure of the triple frequency treated sample was altered compared to the dual-frequency sample. Therefore, this study demonstrated the efficacy of SWE in crude LEPs extraction, illustrated MFU-AATPE as an efficient technique that selectively isolates polysaccharides and enhances extraction efficiencies.

Abstract:Sugarcane bagasse (SCB), a widely available lignocellulosic by-product, could be potentially used in macrofungal bioconversion for high-value bioactive compounds. For macrofungus with poor lignin degradation capacity, its recalcitrant lignin structure blocked the release of fermentable sugars for cell growth. Herein, the main objective was to construct an effective delignification pretreatment strategy for SCB biotransformation and synthesis of bioactive substances by Agaricus sinodeliciosus var. Chaidam. Different pretreatments including chemical (alkali and Fenton oxidation) and ultrasound methods were used to intensify the delignification process. Physico-chemical characteristics of native and pretreated SCB were examined via XRD, XPS, FT-IR and C-13 ssNMR to characterize crystallinity index, surface lignin and chemical structure. Lignocellulose-degrading enzymes were also used to assess the ability of the macrofungus to degrade SCB. By comparing morphological structure, chemical composition and fermentation characteristics, a suitable multiple sequential pretreatment method was finally identified. Our findings revealed that Fenton-ultrasound-alkali pre-treatment has an optimum delignification effect on SCB with never-uncovered excellent ability to detoxify harmful substances produced by Fenton oxidation. Under this pretreatment condition, the cellulose content was increased by 106.45%, whereas hemicelluloses and lignin were decreased by 48.34% and 74.15%, respectively. The macrofungal growth, intracellular polysaccharides, total phenols and terpenoids formation were 2.4, 1.46, 1.36 and 1.66 times higher than those of the untreated, respectively. Fermentation kinetic results showed outstanding fit and prediction of macrofungal growth and enzymatic activity secretion on above pretreated SCB. This study shed light on developing a novel pretreatment strategy for low-cost SCB lignocellulose transformation into high-value products.
Effect of high-intensity ultrasound on the physicochemical properties and nanostructure of citrus pectin.

Abstract:Background Modified pectin has been found to have various biological activities. The preparation of modified pectin is generally accomplished by either chemical or enzymatic depolymerisation processes, but both methods have several disadvantages. Ultrasound treatment is simple and requires shorter times and lower temperatures than conventional techniques used for processing plant materials. In recent years the application of ultrasound to modify polysaccharides has received increasing attention. The objective of this study was to use ultrasound to modify citrus pectin. Results The average molecular weight of citrus pectin decreased under different ultrasonic conditions. The average molecular weight decreased from 464 to 296 kDa after 30 min of sonication. The degree of methylation of citrus pectin changed slightly and its monosaccharide component remained unchanged when high-intensity ultrasound was applied. The reduced (Gal+Ara)/Rha ratio after ultrasonication suggested degradation in the neutral sugar side chains of citrus pectin. Atomic force microscopy results confirmed the degradation of citrus pectin chains by ultrasound at nanolevel. Conclusion Ultrasound is an effective way to pretreat or modify pectin. The degradation of citrus pectin is due to the cavitational effects of ultrasound. Thus ultrasound may be useful in establishing environmentally friendly extraction and modification technologies for pectin. (c) 2013 Society of Chemical Industry

Abstract:Ultrasound–microwave combined extraction (UMCE), gradient ethanol precipitation, chemical characterization, and antioxidant and hypoglycemic activities of Lycium barbarum leaf polysaccharides (LLP) were systematically studied. The optimal conditions for UMCE of LLP achieved by response surface method (RSM) were as follows: microwave time of 16 min, ultrasonic time of 20 min, particle size of 100 mesh, and ratio of liquid to solid of 55:1. Three novel polysaccharide fractions (LLP30, LLP50, LLP70) with different molecular weights were obtained by gradient ethanol precipitation. Polysaccharide samples exhibited scavenging capacities against ABTS and DPPH radicals and inhibitory activities against α-glucosidase and α-amylase. Among the three fractions, LLP30 possessed relatively high antioxidant and hypoglycemic activities in vitro, which showed a potential for becoming a nutraceutical or a phytopharmaceutical for prevention and treatment of hyperglycemia or diabetes.

The ultrasonic-assisted enzymatic extraction, characteristics and antioxidant activities of lychee nuclear polysaccharide

Abstract:A response surface methodology (RSM) and one-factor method were established to investigate the optimum conditions for the extraction of lychee nuclear polysaccharides (LSP) by ultrasonic-assisted pectinase. The content of basic components in polysaccharides was determined. The antioxidant activity was well determined and compared the differences in the activities of the polysaccharides extracted by water extraction (LSP-HW) and those extracted at ultrasound-assisted enzyme (LSP-UAE). The activity of lychee nuclear polysaccharide increased with the increase of concentration. The anti-hydroxyl radical and anti-lipid peroxidation abilities of lychee nuclear polysaccharide were more excellent, and LSP-HW was slightly better than LSP-UAE in terms of activity, but the difference was not significant. In terms of solubility; LSP-HW without deproteinization was the best, followed by LSP-UAE, and the worst was LSP-UAE after deproteinization. The higher the glycoprotein content, the better the solubility of its polysaccharide. Compared with the existing extraction methods, this experiment greatly improved the extraction rate of lychee nuclear polysaccharides and further enriched the antioxidant activities of polysaccharides.

Abstract:Litchi polysaccharides are a kind of macromolecular polymers with various biological activities and a wide range of molecular weights. In this study, two separate fractions, with average molecular weights of 378.67 kDa (67.33%) and 16.96 kDa (6.95%), which were referred to as LP1 and LP2, respectively, were separated using an ultrafiltration membrane. Their physicochemical properties, and immunomodulatory and prebiotic activity were compared. The results revealed that LP2 contained more neutral sugar, arabinose, galactose and rhamnose, but less uronic acid, protein, mannose and glucose than LP1. Compared with LP1, LP2 possessed higher solubility and lower apparent viscosity. LP2 exhibited stronger stimulation on macrophage secretion of NO, TNF-α and IL-6, as well as better proliferation of Lactobacillus plantarum, Leuconostoc mesenteroides, Lactobacillus casei and Bifidobacterium adolescentis. These results suggest that an ultrafiltration membrane might be used to prepare a highly-active polysaccharide fraction from litchi pulp that may be used for food or drug development.
Separation of polysaccharides from Lycium barbarum L. by high-speed countercurrent chromatography with aqueous two-phase system

Abstract:The traditional method for isolation and purification of polysaccharides is time-consuming. It often involves toxic solvents that destroy the function and structure of the polysaccharides, thus limiting in-depth research on the essential active ingredient of Lycium barbarum L. Therefore, in this study, high-speed countercurrent chromatography (HSCCC) and aqueous two-phase system (ATPS) were combined for the separation of crude polysaccharides of Lycium barbarum L (LBPs). Under the optimized HSCCC conditions of PEG1000-K 2 HPO 4 -KH 2 PO 4 -H 2 O (12:10:10:68, w / w ), 1.0 g of LBPs-ILs was successfully divided into three fractions (126.0 mg of LBPs-ILs-1, 109.9 mg of LBPs-ILs-2, and 65.4 mg of LBPs-ILs-3). Moreover, ATPS was confirmed as an efficient alternative method of pigment removal for LBPs purification, with significantly better decolorization (97.1 %) than the traditional H 2 O 2 method (88.5 %). Then, the different partitioning behavior of LBPs-ILs in the two-phase system of HSCCC was preliminarily explored, which may be related to the difference in monosaccharide composition of polysaccharides. LBPs-ILs-1 exhibited better hypoglycemic activities than LBPs-ILs-2 and LBPs-ILs-3 in vitro . Therefore, HSCCC, combined with aqueous two-phase system, was an efficient separation and purification method with great potential for separating and purifying active polysaccharides in biological samples.

Abstract:Interaksi di antara sistem dan fenomena ketidakstabilan adalah masalah biasa di dalam sistem kuasa teragih. Fenomena ini wujud kerana penukar bersifat beban berkuasa tetap, iaitu sistem kawalan dalamannya yang berfungsi menetapkan voltan keluaran penukar. Galangan masukan bagi penukar adalah bergalangan negatif menaik yang menjurus kepada ketidakstabilan. Dalam kertas kerja ini, analisa dan kawalan terhadap penukar AT–AT yang berfungsi sebagai pemampas aktif telah dilakukan. Penukar AT–AT disambungkan ke sebuah sistem mudah yang terdiri dari penapis L–C dan sebuah beban berkuasa tetap. Penukar AT–AT telah dimodelkan menggunakan teknik purata, dan analisa isyarat kecil telah dilakukan bagi mereka bentuk pengawal bagi mengawal arus masukan penukar. Reka bentuk parameter gelung kawalan adalah berpandukan kepada domain frekuensi. Keberkesanan pemampas aktif dibuktikan menerusi simulasi MATLAB Simulink. The interaction arises because an individual converter known as constant power load (CPL), has internal control function such as to regulate the converter output voltage, which results in the converter tends to draw a constant power. The input impedance of CPL has a negative incremental input impedance, which tends to create instability as it is connected to a system. This paper presents the analysis and control of the DC–DC converter used to perform an active damping function in a DPS, comprising L–C filters and CPLs. The DC–DC converter was modelled using averaged technique. The small–signal analysis is performed and the controller was designed on such a way to regulate the converter input current. The design of the control loop parameters based on the frequency domain is demonstrated. The effectiveness of the active damping method was verified by MATLAB Simulink simulation. [Effects of different extraction methods on chemical compositions and biological activities of polysaccharides from Lycium barbarum]

Abstract:In this study, five polysaccharides from Lycium barbarum(LBPs)(LBP-1-LBP-5) were selectively extracted by different extraction methods, and the chemical composition, structural characteristics, and biological activities of LBPs were explored. The results of chemical composition analysis showed that alkaloids were not detected in the five LBPs. The total polysaccharide content was(81.95%±1.6%)-(92.96%±0.76%), the uronic acid content was(8.26%±0.46%)-(24.81%±0.46%), and the protein content was(0.06%±0.03%)-(1.35%±0.13%). The monosaccharide compositions of the five LBPs were basically same, mainly including glucose, xylose, and galactose. However, there was significant difference in the content ratio of different monosaccharide. The results of infrared spectra analysis indicated that the five LBPs had typical infrared spectral characteristics of polysaccharides. The results of nuclear magnetic resonance characteristic spectrum analysis revealed that the five LBPs had two configurations of α and β. Meanwhile, there were triple helix structures in LBP-2, LBP-3, and LBP-4, which enhanced the activities of polysaccharides. The results of activities screening suggested that the biological activities of the five LBPs were significantly different. LBP-3 showed the highest lipid oxidation clearance rate, and its antioxidant activity was equivalent to that of the positive control group. The inhibitory rate of LBP-4 on α-amylase and its activation rate of alcohol dehydrogenase were better than those of other fractions, and the inhibitory rate of LBP-4 on α-amylase was slightly higher than that of the positive control group when the mass concentration was 10 g·L~(-1). LBP-2 showed stronger inhibitory activity against α-glucosidase and hyaluronidase. This study provides references for the precise development and utilization of LBPs.

Abstract:Polysaccharides of wolfberry (LBPs) with complex structures are most commonly used in the food and pharmaceutical industries as biologically active phytochemicals. However, the structure and yield of LBPs vary according to the extraction method used, and selecting their extraction media remains difficult. A novel type of temperature-switchable deep eutectic solvent (TS-DES), consisting of L -menthol and phenyl salicylate, was designed to extract LBPs from wolfberry fruits. TS-DES was a recyclable extractant above the maximum critical solution temperature (UCST). The increased dispersibility of extraction solvents was imparted by combining them with a high-speed shear dispersion emulsification (HSDE) machine. TS-DES was in full contact with the cell walls of wolfberries, leading to the complete opening of the cell walls, resulting in the efficient extraction of LBPs. Compared with traditional extraction, the yield of LBPs-D reached 4.35%, which was higher than that of LBPs-W (1.78%). This study showed that LBPs-D were high molecular weight, highly branched polysaccharides with antioxidant activity. TS-DES was separated at a lower level than UCST for regeneration and reuse. After 5 cycles, TS-DES still had excellent recovery and extraction performance. This study provides valuable information for efficiently extracting structurally rich bioactive polysaccharides from plants using TS-DES.

Abstract:The aim of this study was to investigate the effects of enzymatic treatments (pectinase, pectin lyase, and cellulase) on the in vitro digestion and fermentation characteristics of whole mulberry fruit juice. The analysis focused on changes in carbohydrate properties within the black mulberry fruit matrix during simulated digestion and fermentation. Human fecal microbiota were collected and introduced to the fruit matrix to monitor the fate of both soluble and insoluble polysaccharides during fermentation. The results revealed that enzymatic treatments enhanced the solubilization of carbohydrates from mulberry fruits, with pectinase showing the most significant effect. Throughout the process of in vitro digestion, there was a gradual increase in the percentage of solubilized carbohydrates from the mulberry juice substrate. The digested suspensions underwent dialysis to remove degradation fragments, and a lower quantity of carbohydrate in the enzyme-treated groups compared to the control. Polysaccharide populations with varying molecular weights (Mw) were obtained from the soluble fractions of mulberry residues for subsequent fermentation. An increase in Mw of soluble polysaccharides was detected by HPSEC during fermentation in certain cases. The gut microbiota demonstrated the ability to convert specific insoluble fractions into soluble components, which were subsequently subjected to microbial utilization. Enzymatic treatments during mulberry juice preparation can potentially positively impact health by influencing gut microbiota and short-chain fatty acid (SCFA) modulations. Enzymes could serve as valuable tools for producing functional fruit and vegetable juices, with the need to specify processing conditions for specific raw materials remaining a subject of further investigation.

Abstract:The effects of ultrasound pretreatment (20 kHz, 30 W/L) on mulberries' texture, microstructure, characteristics of cell-wall polysaccharides, moisture migration, and drying quality were investigated over exposure times ranging from 15 to 45 min. Ultrasound induced softening of mulberry tissue, accompanied by an increase in water-soluble pectin and a decrease in chelate-soluble pectin and Na2CO3-soluble pectin concentrations. Noticeable depolymerization of the pectin nanostructure was observed in the pretreated mulberries, along with a decrease in molecular weight, attributed to side-chain structure cleavage. Ultrasound loosened the cell wall structure, increased free water content and freedom, thereby reducing water diffusion resistance. Ultrasound pretreatment reduced drying time by 11.2 % to 23.3 % at various processing times compared to controls. Due to significantly enhanced drying efficiency, the optimal pretreatment time (30 min) yielded dried mulberries with higher levels of total phenolics and total anthocyanins, along with an increased antioxidant capacity. The results of this study provide insights into the mechanisms by which ultrasound pretreatment can effectively enhance the mulberry drying process.
Ultrasound-assisted deep eutectic solvents extraction of polysaccharides from Loquat leaf: Process optimization and bioactivity study

Abstract:Loquat leaves are the by-product of loquat fruit production. Polysaccharides are one of the main active ingredients in loquat leaves. In this study, polysaccharides were extracted from loquat leaves by ultrasonic-assisted deep eutectic solvents (DESs) extraction method. Further, the extracted crude loquat leaf polysaccharides (CLLP) were purified and separated via S-8 resin and DEAE-52 cellulose column chromatography, respectively. Additionally, the effects of polysaccharides on activity of sperm in boar semen preserved in medium at 17 °C, were evaluated preliminarily. DES, composed of choline chloride/ethylene glycol (1:6, molar ratio), was proved to be the suitable solvent for LLP extraction. The optimized extraction conditions were water content 44 %, liquid-solid ratio 1:29 (g/g), extraction temperature 61 °C and extraction time 98 min. Under these conditions, the LLP yield was 57.82 ± 1.50 mg/g. A homogeneous polysaccharide (LLP1-2, Mw: 2.17 × 104 Da) was isolated from CLLP. Its total sugar, uronic acid and protein contents were 76.31 ± 1.25 %, 14.19 ± 0.67 % and 3.28 ± 0.42 %, respectively. Further, 800 μg/mL LLP1-2 could effectively enhance the antioxidant activity of sperm. This study laid a foundation for DESs and column chromatography in the field of polysaccharide extraction and separation, proving that LLP can be used as a natural antioxidant for sperm preservation.
Synergism of Sweeping Frequency Ultrasound and Deep Eutectic Solvents Pretreatment for Fractionation of Sugarcane Bagasse and Enhancing Enzymatic Hydrolysis

Abstract:Sugarcane bagasse (SCB) is an abundant agricultural waste in China and the conversion of the waste into plethora of useful resources is very vital. To achieve this, fractionation of the waste is highly important in the biomass biorefinery. The present study aims at investigating the synergistic role of deep eutectic solvents (DES) with sweeping frequency ultrasound (SFUS) and fixed frequency ultrasound (FFUS) in the fractionation of SCB to enhance the enzymatic saccharification process. Therefore, the effects of ultrasound (US) and DES conditions on the pretreatment efficiency were investigated. Under optimum SCB pretreatment conditions, FFUS (40 kHz, 60 min) + DES (choline chloride (ChCl)-lactic acid (LA), 120 °C, 3 h) and SFUS (40 kHz, 60 min) + DES (ChCl-LA, 120 °C, 3 h), the lignin removal rates were 80.13 and 85.62%, respectively. The hemicellulose removal rates were 78.08 and 90.46%, respectively; and the contents of glucose, xylose and arabinose in the liquid fractions after FFUS + DES pretreatment were 7.07, 17.95 and 3.01%, respectively. However, the yield of glucose, xylose, and cellobiose after enzymatic hydrolysis of the SFUS + DES pretreated SCB were 86.76, 38.68, and 20.76%. Analytical studies revealed that the SFUS + DES pretreatment can effectively destroy the ultrastructure of SCB and reduce the crystallinity of cellulose. Furthermore, the mechanism of pretreatment with SFUS + DES was proposed, which confirmed the excellent performance of SFUS + DES. Thus, the application of SFUS + DES pretreatment was able to improve the removal of lignin and hemicellulose from SCBs.
