Abstract:The present study was carried out to investigate the beneficial impact of seed priming with polyethylene glycol (PEG) under different concentrations of zinc oxide nanoparticles (nano-ZnO), i.e., 0, 250, 500, and 750 mg L −1 in two cultivars of Oryza sativa (Zhu Liang You 06 and Qian You No. 1). Physiological parameters were improved by priming with 30 % PEG in both cultivars under stress treatments. Seed priming with 30 % PEG improved α-amylase activities and total soluble sugar contents of both cultivars under nano-ZnO stress. In addition, glutathione reductase (GR) activity, reactive oxygen species (ROS) accumulation, and proline contents decreased after the priming treatment in both cultivars under different nano-ZnO concentrations. Expression of GR1 , GR2 , Amy2A , and Amy3A genes in shoots and roots of both cultivars increased and had higher transcription levels under the nano-ZnO stress condition. Fourier transform infrared spectroscopy (FTIR) analysis did not show any significant effects of the priming treatment on the band observed at 3400, 900, 1600, and 1000 cm −1 corresponding to alkenyl stretch (C = C), carboxyl acid (O-H), nitrile (C = N), and aromatic (C-H), respectively, in both cultivars under nano-ZnO stress.

Seed Priming with Chitosan Improves Maize Germination and Seedling Growth in Relation to Physiological Changes under Low Temperature Stress

Abstract:Low temperature stress during germination and early seedling growth is an important constraint of global production of maize. The effects of seed priming with 0.25%, 0.50%, and 0.75% (w/v) chitosan solutions at 15 °C on the growth and physiological changes were investigated using two maize (Zea mays L.) inbred lines, HuangC (chilling-tolerant) and Mo17 (chilling-sensitive). While seed priming with chitosan had no significant effect on germination percentage under low temperature stress, it enhanced germination index, reduced the mean germination time (MGT), and increased shoot height, root length, and shoot and root dry weights in both maize lines. The decline of malondialdehyde (MDA) content and relative permeability of the plasma membrane and the increase of the concentrations of soluble sugars and proline, peroxidase (POD) activity, and catalase (CAT) activity were detected both in the chilling-sensitive and chilling-tolerant maize seedlings after priming with the three concentrations of chitosan. HuangC was less sensitive to responding to different concentrations of chitosan. Priming with 0.50% chitosan for about 60∼64 h seemed to have the best effects. Thus, it suggests that seed priming with chitosan may improve the speed of germination of maize seed and benefit for seedling growth under low temperature stress.

Abstract:Polyamines (PAs) have been demonstrated to be involved in plant in response to abiotic stresses including chilling stress. The present study was designed to investigate the effects of seed priming with 5 mM of spermidine (Spd) and 8.5 mM of 5-Aminolevulinic acid (ALA) on seed polyamines metabolism associated with the improvement of chilling tolerance in two rice cultivars, Zhu Liang You 06 (ZY) and Qian You No.1 (QY). Germination percentage, seedling growth and seedling vigor index was decreased under chilling stress, but this physiological parameters was improved by Spd and ALA priming in both studied cultivars as compared with unprimed seeds. As well, total phenolics, flavonoids and glycine-betaine were improved by priming treatment. Contrarily, significant decrease of ca-amylase activity, soluble sugars and soluble protein contents of both cultivars was observed in chilling stressed plants as compared with normal growth condition (25 degrees C). However, priming with Spd and ALA significantly increased a-amylase activity, soluble sugars and soluble protein contents with more prominent increase in QY cultivar. Results showed that chilling stress significantly improved superoxide dismutase (SOD), peroxidase (POD), ascorbate peroxidase (APX) and glutathione peroxidase (GPX), and further enhancement was observed by Spd and ALA-primed seeds. Spd and putrescine (Put) were decreased under chilling stress, while a reverse tendency was observed in case of spermine (Spm) content. The enzymes involved in the PAs biosynthesis, arginine decarboxylase (ADC), ornithine decarboxylase (ODC) and S-adenosylmethionine decarboxylase (SAMDC) was improved by priming treatment. The relative expressions of genes encoding enzymes involved in PAs biosynthesis increased by Spd and ALA priming. Effects of Seed Priming with Chitosan Solutions of Different Acidity on Seed Germination and Physiological Characteristics of Maize Seedling

Abstract:The effects of seed priming with acidic(pH5.1) and near neutral(pH6.4) chitosan solutions on seed germination and physiological characteristics of seedling were investigated using four maize materials.The results showed that priming treatments with two acidic chitosan solutions significantly enhanced the energy of germination,reduced mean germination time and increased shoot height and chlorophyll content.There were no significant difference in germination percentage between primed new seeds of three materials and un-treated control.The germination percentage of old seeds of Chaotian No.3 was,however,lower than that of the control.The POD activity and MDA content of the shoots of Suyu No.1 and Chaotian No.3 were obviously increased and decreased respectively after priming with pH5.1 chitosan solution.The POD activity was significantly decreased,and MDA content had no significant difference in the shoots of Chaotian No.3 old-aged seeds compared to the control.Concerning the CAT activity in the shoots of the three maize materials,including old-aged seeds,there were no markedly differences between the treatments and the control.Thus,it seems that chitosan is a new promising priming material for maize seed treatment,and no significant difference was found between the two acidic chitosan solutions and both are efficient in improving vigor of maize seeds.
Cold Plasma Treatment and Exogenous Salicylic Acid Priming Enhances Salinity Tolerance of Oryza Sativa Seedlings

Abstract:The present study was designed to highlight the effects of cold plasma (10 kV) treatment and priming with 2 mM salicylic acid (SA) and their combination (10 kV of plasma + 2 mM SA) on the physiological parameters and metabolism of two cultivars of Oryza sativa, i.e., Zhu Liang You 06 (ZY) and Qian You No. 1 (QY), under salinity stress (150 mM NaCl) and normal growth condition (0 mM NaCl). Seed germination and seedling growth were enhanced by SA priming and cold plasma treatment either alone or in combination under salinity stress. Photosynthetic pigments, photosynthetic gas exchange, and chlorophyll fluorescence were improved by cold plasma treatment and SA priming under salinity stress as compared to the untreated seeds. The activities of antioxidant enzymes were significantly improved by the combination of SA priming and cold plasma treatment in both cultivars under salinity stress. There were rapid changes in the cellular content of sodium (Na+) and calcium (Ca+), where the plants grown under saline conditions accumulate more Na+ and less Ca+ contents resulting in ionic imbalances. Interestingly, cold plasma and SA treatments diminished this action by reducing Na+ accumulation and increasing K+ and Ca+ contents in the plant cell under salinity stress. The activities of enzymes involved in secondary metabolism assimilation were up-regulated with cold plasma and SA priming either alone or combination under salinity stress. An increase in reactive oxygen species (ROS) accumulation and malondialdehyde (MDA) content was also observed under salinity stress condition. On contrast, seed treated with SA and plasma alone or combined resulted in a significant decrease in ROS and MDA contents under salinity stress. Our results indicated that SA priming and cold plasma treatment either alone or combined improved plant uptake of nutrients in both cultivars under stress conditions. The ultrastructural changes were observed to be more prominent in ZY than QY cultivar. Plants without SA priming or cold plasma treatments have a big vacuole due to the movement of ions into the vacuole directly from the apoplast into the vacuole through membrane vesiculation leading to membrane destabilization. However, SA priming and cold plasma treatment alone or combined helped the plants to recover their cell turgidity under salinity stress.

Abstract:Osmopriming with PEG has potential to improve seed germination, seedling emergence, and establishment, especially under stress conditions. This research investigated germination performance, seedling establishment, and effects of osmopriming with PEG on physiology in sorghum seedlings and their association with post-priming stress tolerance under various soil moisture stress conditions. Results showed that seed priming increased the environmental range suitable for sorghum germination and has potential to provide more uniform and synchronous emergence. Physiologically, seed priming strengthened the antioxidant activities of APX, CAT, POD, and SOD, as well as compatible solutes including free amino acid, reducing sugar, proline, soluble sugar, and soluble protein contents. As a result, seed priming reduced lipid peroxidation and stabilized the cell membrane, resulting in increased stress tolerance under drought or excessive soil moisture environments. Overall, results suggested that seed priming with PEG was effective in improving seed germination and seedling establishment of sorghum under adverse soil moisture conditions. Osmopriming effectively strengthened the antioxidant system and increased osmotic adjustment, likely resulting in increased stress tolerance.

Abstract:Abiotic stresses such as drought, extreme temperature, and salinity can negatively impact seed germination and plant growth and have become major limitations to crop production. Most crops are vulnerable to abiotic stress factors during their early growth phase, especially during seed germination and seedling emergence. Rapid crop seed germination and seedling establishment is known to provide competitive advantages over weeds and improve yields. Seed osmopriming is defined as a pre-sowing treatment in which seeds are soaked in osmotic solutions to undergo the first stage of germination, but radicle protrusion has not occurred. The process of osmopriming involves prior exposure of seeds in low-water-potential solutions. Osmopriming can generate a series of pre-germination metabolic activities, increase the antioxidant system activities, and prepare the seed for radicle protrusion. Polyethylene glycol (PEG) is a popular osmopriming agent that can alleviate the negative impacts of abiotic stresses. This review summarizes research findings on crop responses to seed priming with PEG under abiotic stresses. The challenges, limitations, and opportunities of using PEG for crop seed priming are discussed with the goal of providing insights into future research towards effective application of seed priming in crop production.

Abstract:Water stress can adversely affect seed germination and plant growth. Seed osmopriming is a pre-sowing treatment in which seeds are soaked in osmotic solutions to undergo the first stage of germination prior to radicle protrusion. Seed osmopriming enhances germination performance under stressful environmental conditions, making it an effective method to improve plant resistance and yield. This study analyzed the effect of seed osmopriming with polyethylene glycol (PEG) on seed germination and physiological parameters of Coronilla varia L. Priming treatments using 10% to 30% PEG enhanced germination percentage, germination vigor, germination index, vitality index, and seedling mass and reduced the time to reach 50% germination (T50). The PEG concentration that led to better results was 10%. The content of soluble proteins (SP), proline (Pro), soluble sugars (SS), and malondialdehyde (MDA) in Coronilla varia L. seedlings increased with the severity of water stress. In addition, under water stress, electrolyte leakage rose, and peroxidase (POD) and superoxide dismutase (SOD) activities intensified, while catalase (CAT) activity increased at mild-to-moderate water stress but declined with more severe deficiency. The 10% PEG priming significantly improved germination percentage, germination vigor, germination index, vitality index, and time to 50% germination (T50) under water stress. Across the water stress gradient here tested (8 to 12% PEG), seed priming enhanced SP content, Pro content, and SOD activity in Coronilla varia L. seedlings compared to the unprimed treatments. Under 10% PEG-induced water stress, primed seedlings displayed a significantly lower MDA content and electrolyte leakage than their unprimed counterparts and exhibited significantly higher CAT and POD activities. However, under 12% PEG-induced water stress, differences in electrolyte leakage, CAT activity, and POD activity between primed and unprimed treatments were not significant. These findings suggest that PEG priming enhances the osmotic regulation and antioxidant capacity of Coronilla varia seedlings, facilitating seed germination and seedling growth and alleviating drought stress damage, albeit with reduced efficacy under severe water deficiency.

Abstract:AbstractThe present study was designed to highlight the impact of seed priming with polyethylene glycol on physiological and molecular mechanism of two cultivars of Oryza sativa L. under different levels of zinc oxide nanorods (0, 250, 500 and 750 mg L−1). Plant growth parameters were significantly increased in seed priming with 30% PEG under nano-ZnO stress in both cultivars. Whereas, this increase was more prominent in cultivar Qian You No. 1 as compared to cultivar Zhu Liang You 06. Significant increase in photosynthetic pigment with PEG priming under stress. Antioxidant enzymes activities of superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) as well as malondialdehyde (MDA) contents were significantly reduced with PEG priming under nano-ZnO stress. Gene expression analysis also suggested that expression of APXa, APXb, CATa, CATb, CATc, SOD1, SOD2 and SOD3 genes were down regulated with PEG priming as compared to non-primed seeds under stress. The ultrastructural analysis showed that leaf mesophyll and root cells were significantly damaged under nano-ZnO stress in both cultivars but the damage was prominent in Zhu Liang You 06. However, seed priming with PEG significantly alleviate the toxic effects of nano-ZnO stress and improved the cell structures of leaf and roots in both cultivars.

Abstract:Saline stress is one of the most critical abiotic stress factors that can lessen crops' productivity. However, emerging nanotechnology, nano-fertilizers, and developing knowledge of phytochromes can potentially mitigate the negative effects of saline stress on seed germination. Therefore, the aim of this study was to investigate the effects of seed priming either with zinc oxide nanoparticles (ZnO-NPs; 50 and 100 mg L−1) or 24-epibrassinolide (EBL; 0.2 and 0.4 μM) and their combinations on maize (Zea mays L.) grains sown in salt-stressed soil (50 and 100 mM NaCl). Saline stress treatments significantly affected all germination traits and chemical analysis of seeds as well as α-amylase activity. Compared to un-primed seeds, seed priming with ZnO-NPs or EBL and their combinations significantly increased the cumulative germination percentage, germination energy, imbibition rate, increase in grain weight, K+ content, and α-amylase activity, and significantly reduced germination time, days to 50% emergence, Na+ uptake, and Na+/K+ ratio of maize sown in salt-stressed-soil (50 or 100 mM NaCl). The combination of 100 mg ZnO-NPs L−1 + 0.2 μM EBL resulted in the highest improvements for most of the studied traits of maize seeds sown in salt-stressed soil in comparison to all other individual and combined treatments.

Abstract:Background: Drought stress poses a significant challenge to agricultural productivity, particularly in regions with limited water availability. Maize, a staple crop worldwide, is highly susceptible to drought stress, leading to reduced seed germination, stunted growth and ultimately diminished yields. Addressing this issue, in this study various phytohormones and polyamines were used as priming agents for seeds to evaluate its performance to mitigate drought stress in maize seed germination and vigour. It plays crucial roles in regulating plant growth and development, as well as responses to various environmental stresses, including drought. These hormones modulate physiological and biochemical processes, leading to adaptive changes that helps maize plants to cope with water scarcity. Methods: The experiment was conducted by exposing maize seeds primed with different phytohormones and polyamines in optimum along with non primed seeds (control) to various levels of PEG concentration ranging from -0.2 MPa to -1.0 MPa for creating drought stress. Primed seeds subjected to a germination test. The seedling growth parameters and seed vigour were evaluated to analyse better performing seed priming treatment to mitigate drought stress. Result: From the experiment it is concluded that increase in concentration of PEG (from -0.2 Mpa to -1.0 MPa) decreased the seed germination and seed vigour traits in maize. Among seed priming treatments, seeds primed with melatonin 150 μM showed greater performance seedling vigour and its related traits even under water deficit stress condition. In conclusion, seed priming with melatonin exhibited superior efficacy in mitigating drought stress in maize. This finding highlights the potential of melatonin as a promising seed priming agent for enhancing drought tolerance in maize seeds. As seed priming is simple, easy and cost effective method, farmers can use this technology as one of their practice to eliminate the effect of drought stress in maize crop.

Abstract:Different techniques, including exogenous application of different chemicals as seed priming, are in practice to improve seed germination or seedling emergence for better crop production, especially under water-deficit conditions. The present study was conducted to compare the effects of seed priming with glutathione alone or in combination with Zn on seed germination behavior, seedling growth, photosynthetic pigments, and lipid peroxidation in relation to alterations in seed metabolites, seedling antioxidative potential, and biochemicals under polyethylene glycol (PEG)-induced water stress. Three levels of glutathione (1.5, 3.0, and 4.5 mM) were applied separately as well as in combination with 0.5% Zn as seed priming. Water deficit significantly adversely affected seed germination potential, seedling growth, and biosynthesis of photosynthetic pigments in association with disturbed metabolic activities in germinating seeds, and increased lipid peroxidation in terms of the contents of H 2 O 2 and malondialdehyde (MDA). Seed priming with glutathione alone or in combination with Zn significantly reduced the adverse impacts of water stress on seed germination and seedling growth in association with positive boosting influences on seed metabolic activities and increased biosynthesis of photosynthetic pigments, activities of antioxidant enzymes, and contents of non-enzymatic antioxidants, with more accumulation of reducing sugars (RS), non-reducing sugars (NRS), total soluble sugars (TSS), and total soluble proteins (TSP). Overall, combined application of glutathione and Zn for seed priming was found to be better than separate applications. The 3 mM level of glutathione in combination with Zn was most effective in ameliorating the adverse impacts of water stress.

Abstract:Salinity is one of the most devastating environmental factors limiting crop productivity worldwide. Therefore, our study investigates the effect of seed priming with zinc oxide nanoparticles (ZnO NPs: 0, 50, and 100 mg L−1), 24-epibrassinolide (EBL: 0.0, 0.2, and 0.4 μM), and their combined treatments on maize (Zea mays L.) grown with different levels of saline stress (i.e., control, 5, 10 dS m−1) under semi-controlled conditions. Higher saline stress (10 dS m−1) negatively influenced the growth traits, physiological attributes, and elemental (i.e., Zn and K) uptake for both roots and shoots of maize, whereas it increased Na+ accumulation and Na+/K+ ratio in comparison to other treatments. However, seed priming with ZnO NPs and EBL as well as their combinations showed amelioration of the detrimental effects of saline stress on the growth and physiological and biochemical performance of maize. In general, seed priming with combined treatments of ZnO NPs and EBL were significantly more effective than either ZnO NPs or EBL as individual treatments. A combination of 100 mg L−1 ZnO NPS + 0.2 μM EBL resulted in the highest values of root length, root surface area, stem diameter, relative leaf water contents, total chlorophyll, net rate of photosynthesis, zinc accumulation, and K+ uptake, while it resulted in the lowest Na+ and Na+/K+ ratio, especially under the highest saline-stress treatment. Thus, we concluded that seed priming with combined ZnO NPs and EBL can effectively mitigate the saline-stress-mediated decline in the morphological, physiological, and biochemical traits of maize.

Abstract:Soil alkalinity is a severe threat to crop production globally as it markedly retards plant growth. Different techniques are used to mitigate alkaline stress, but priming techniques are considered the most appropriate. The current study was carried out in complete randomized design (CRD) to evaluate the effect of different priming techniques on maize crop grown under different levels of alkalinity stress. The experiment was comprised of different treatments of alkalinity stress (AS) including, control, 6 dS m-1 and 12 dS m-1 and different priming techniques including control, hydro-priming (HP), osmo-priming (OP) with potassium nitrate: KNO3) and redox-priming (RP) with hydrogen peroxide (H2O2). Results indicated that alkalinity stress significantly reduced plant growth and biomass production and induced severe alterations in physiological attributes and antioxidant activities. Soil alkalinity significantly reduced the root and shoot growth and subsequent biomass production by increasing electrolyte leakage (70.60%), hydrogen peroxide (H2O2: 31.65%), malondialdehyde (MDA: 46.23%) and sodium (Na+) accumulation (22.76%) and reduction in photosynthetic pigments, relative water contents (RWC), total soluble proteins (TSP) and free amino acids, potassium (K+) accumulation. However, priming treatments significantly alleviated the alkalinity-induced toxic effects and improved plant growth. OP (KNO3) remained the top performing. It appreciably improved plant growth owing to the improved synthesis of photosynthetic pigments, better RWC (16.42%), TSP (138.28%), FAA (178.37%), and K+ accumulation (31.385) and improved antioxidant activities (APX and CAT) by favoring the Na+ exclusion and maintenance of optimum Na+/K+. In conclusion, KNO3 priming is an imperative seed priming practice to improve maize growth and biomass production under alkalinity stress.

Abstract:Seeds of three waxy maize (Zea mays L.) cultivars, cv. Suyunuo 1, Huyunuo 1 and Zhenuoyu 1, were used to investigate the effects of sand priming on germination and physiological changes under a high-salt concentration (1% NaCl) stress. Waxy maize seeds were mixed with sand containing 4% (v/w) water, sealed in plastic box, and then were primed at 15 degrees C for 48 h. While sand priming significantly affected the speed of germination in two cultivars, and contributed to shorten mean germination time for the three cultivars studied. It also significantly enhanced shoot height, seedling fresh and dry weight in the three cultivars compared to unprimed control. Sand priming also increased the content of soluble sugar and the activities of peroxidase (POD) and catalase (CAT). At the same time, the malondialdehyde (MDA) and proline (Pro) accumulation decreased under a high-salt stress condition. Thus, it indicated that sand priming could improve speed of germination and seedling growth, suggesting beneficial effects on salt tolerance of waxy maize seedlings.
The Synergistic Priming Effect of Exogenous Salicylic Acid and H2O2 on Chilling Tolerance Enhancement during Maize (Zea mays L.) Seed Germination

Abstract:Chilling stress is an important constraint for maize seedling establishment in the field. To examine the role of salicylic acid (SA) and hydrogen peroxide (H2O2) in response to chilling stress, we investigated the effects of seed priming with SA, H2O2, and SA+H2O2 combination on maize resistance under chilling stress (13°C). Priming with SA, H2O2, and especially SA+H2O2 shortened seed germination time and enhanced seed vigor and seedling growth as compared with hydropriming and non-priming treatments under low temperature. Meanwhile, SA+H2O2 priming notably increased the endogenous H2O2 and SA content, antioxidant enzymes activities and their corresponding genes ZmPAL, ZmSOD4, ZmAPX2, ZmCAT2, and ZmGR expression levels. The α-amylase activity was enhanced to mobilize starch to supply metabolites such as soluble sugar and energy for seed germination under chilling stress. In addition, the SA+H2O2 combination positively up-regulated expressions of gibberellic acid (GA) biosynthesis genes ZmGA20ox1 and ZmGA3ox2, and down-regulated GA catabolism gene ZmGA2ox1 expression; while it promoted GA signaling transduction genes expressions of ZmGID1 and ZmGID2 and decreased the level of seed germination inhibitor gene ZmRGL2. The abscisic acid (ABA) catabolism gene ZmCYP707A2 and the expressions of ZmCPK11 and ZmSnRK2.1 encoding response receptors in ABA signaling pathway were all up-regulated. These results strongly suggested that priming with SA and H2O2 synergistically promoted hormones metabolism and signal transduction, and enhanced energy supply and antioxidant enzymes activities under chilling stress, which were closely relevant with chilling injury alleviation and chilling-tolerance improvement in maize seed. Highlights:Seed germination and seedling growth were significantly improved under chilling stress by priming with SA+H2O2 combination, which was closely relevant with the change of reactive oxygen species, metabolites and energy supply, hormones metabolism and regulation.

Abstract:The potential of wheat (Triticum aestivum) seed osmo-priming for post-germinative osmo-stress resistance was evaluated. Further, for comparative evaluation of seed osmo-priming and plant stress-acclimation, a set of plants was stress-acclimated prior to osmo-stress treatment. Under osmo-stress at the post-germination stage, plants from osmo-primed seeds produced 2.3% higher root biomass, 21.6% longer roots, accumulated 15.1% higher soluble sugar content and respectively, 10.4 and 12.8% higher activity of SOD and POD in the roots compared with plants from unprimed seeds. At the post-seedling establishment stage, seed-osmo-primed plants produced 29.9% higher root biomass, 15.2% longer roots, accumulated 32.7% higher soluble sugar content and 31.2% higher proline content in the roots, while the activity of SOD and POD was respectively, 19.6 and 13.1% higher than the unprimed plants. Plant stress-acclimation induced osmo-stress tolerance was exemplified by 6.9% higher root biomass, 4.6% longer roots, 52.7% higher soluble sugar content in roots, 23.9% higher proline content in the roots and 27.0% higher activity of SOD in the roots under osmo-stress when compared with the unprimed plants. The data of seedling morphology and physiological activity under control conditions showed adverse effects of stress acclimation whereas seed osmo-priming-induced stress resistance resulted in advanced germination and higher osmolyte accumulation.

Abstract:Water stress is one of the most important yield limiting factors in maize (Zea mays L.) around the world, particularly in developing countries which have limited access to reliable irrigation. Water stress can be overcome either by developing additional water resources or improving plant resistance. The use of different organic and inorganic substances might be important to improve plant resistance to water stress. Present study investigated the protective role of silicon (Si) and chitosan in maize under water stress. Experimental plan comprised of two soil moisture regimes i.e., 100% field capacity (FC) as control and 50% FC (water stress), two Si levels (0 and 0.5%), two chitosan levels (0 and 0.75%) with three replications. Silicon and chitosan were applied through seed priming. Water stress limited the plant growth and yield by inducing oxidative damage, chlorophyll and photosynthesis degradation, membrane destabilization and stomatal closure. Seed priming with Si and chitosan reversed the oxidative damage as evidenced by marked reduction in malondialdehyde (MDA) by activating catalase, peroxidase and superoxide dismutase. Application of Si and chitosan also improved soluble protein, chlorophyll, photosynthesis and potassium uptake, leading to enhanced plant growth and yield. Grain yield improved by 17.9, 9.86 and 26.3% at 100% FC while 54.2, 34.0 and 56.9% at 50% FC with Si, chitosan and Si + chitosan, respectively compared to respective soil moisture regime without Si and chitosan. It was suggested that integrated use of Si and chitosan via seed priming could be an important strategy for inducing resistance to water stress in maize.

Abstract:Maize ( Zea mays ) is moderately sensitive to salt stress. Therefore, increasing salinity in soil causes the arrestment of physiological processes and retention of growth and development, consequently leading to yield loss. Although many strategies have been launched to improve salt stress tolerance, plant growth-promoting rhizobacteria (PGPR) are considered the most promising approach due to being more environmentally friendly and agronomically sustainable than chemicals. Therefore, this study aims to investigate the potential of Bacillus spp. and the role of microRNA-mediated genetic regulation in maize subjected to seed bio-priming application to mitigate salt stress effects. To this end, maize seeds were bio-primed with the vegetative form of B. pumilus , B. licheniformis, and B. coagulans both individually or combined, subsequently treated to NaCl, and the seedlings were screened morphologically, physiologically, and transcriptionally. The study revealed that seed bio-priming with B. licheniformis reduced the stress effects of maize seedlings by increasing catalase (CAT) and ascorbate peroxidase (APX) activities by 2.5- and 3-fold, respectively, tolerating the decrease in chlorophyll content (CC), upregulating miR160d expression which led to a 36% increase in root fresh weight (RFW) and a 39% increase in shoot fresh weight (SFW). In conclusion, Bacillus spp. successfully alleviated salt stress effects on maize by modulating antioxidant enzymes and miRNA expression.
