Abstract:Methotrexate successful therapy encounters various challenges in chemotherapy, such as poor oral bioavailability, low specificity, side effects and the development of drug resistances. In this study, it is proposed a dual-targeted nanocarrier comprising magnetite/chitosan nanoparticles for an efficient Methotrexate delivery. The formation of the particles was confirmed through morphological analysis using electron microscopy and elemental mappings via energy dispersive X-ray spectroscopy. These nanoparticles exhibited a size of ≈ 270 nm, a zeta potential of ≈ 24 mV, and magnetic responsiveness, as demonstrated by hysteresis cycle analysis and visual observations under a magnetic field. In addition, these particles displayed high stability, as evidenced by size and surface electric charge measurements, during storage at both 4 ºC and 25 ºC for at least 30 days. Electrophoretic properties were examined in relation to pH and ionic strength, confirming these core/shell nanostructure. The nanoparticles demonstrated a pH-responsive drug release as observed by a sustained Methotrexate release over the next 90 h under pH ≈ 7.4, while complete release occurred within 3 h under acidic conditions (pH ≈ 5.5). In the biocompatibility assessment, the magnetite/chitosan particles showed excellent hemocompatibility ex vivo and no cytotoxic effects on normal MCF-10 A and cancer MCF-7 cells. Furthermore, the Methotrexate-loaded nanoparticles significantly enhanced the antitumor activity reducing the half-maximal inhibitory concentration by ≈ 2.7-fold less compared to the free chemotherapeutic.
Magnetic Hybrid Wax Nanocomposites as Externally Controlled Theranostic Vehicles: High MRI Enhancement and Synergistic Magnetically Assisted Thermo/Chemo Therapy

Abstract:To fight against cancer, smarter drugs and drug delivery systems are required both to boost the efficiency of current treatments while reducing deleterious side effects, and combine diagnosis/monitoring with therapy (theranosis) in the search for the final goal of personalized medicine. This work presents the design, preparation, and proof-of-principle validation of a novel hybrid organic-inorganic nanocomposite joining together non-invasive imaging capabilities through magnetic resonance imaging and externally actuated therapeutic properties through a combination of chemo- and thermotherapy. The lipidic matrix of the nanocomposite was composed of carnauba wax, which was simultaneously dual loaded with magnetite nanoparticles and the anticancer drug Oncocalyxone A. Obtained formulations were fully characterized and showed outstanding performances as T2 -contrast agents in magnetic resonance imaging (r2 >800 mm-1 s-1 ), heat generating sources in magnetic hyperthermia (specific absorption rate, SAR>200 W g-1 Fe ), and magnetically responsive drug delivery vehicles. The potential of the designed formulations as theranostic agents was validated in vitro and results indicated a synergistic thermo/chemotherapeutic effect derived from heat generation and controlled drug delivery to cancer growth. Thereby, this external control over the drug delivery profile and the integrated imaging capability open the door to personalized cancer medicine and real-time monitoring of tumor progression.
Magnetic and Ph Dual-Responsive Nanoparticles for Synergistic Drug-Resistant Breast Cancer Chemo/Photodynamic Therapy

Abstract:Background Drug resistance is one of the prime reasons of chemotherapy failure in breast cancer and is also an important factor affecting prognosis. Purpose In this study, we constructed a functional magnetic mesoporous silica-based nanocomposite (MMSN) for breast cancer chemotherapy/photodynamic therapy. Methods MMSN was characterized by scanning electron microscopy and transmission electron microscopy to observe the morphology. The size distribution and zeta potential of the MSNs were determined using Malvern Particle Size Analyzer. Anti-tumor activity in vitro was investigated by CCK-8 assay, flow cytometry and transwell experiment, and the anti-tumor activity in vivo was probed into by magnetic targeting, toxicity, and antitumor effects in breast cancer-bearing BABL/c nude mice. Results The results showed that the release of doxorubicin in the nanocomposites was pH sensitive, and the cumulative release rate reached 80.53% at 60 h under acidic conditions. The nanocomposites had a high cellular uptake ability in MCF-7/ADR cells, and the IC50 value of the nanocomposites on MCF-7/ADR cells was 4.23 μg/mL, much smaller than that of free DOX (363.2 μg/mL). The nanocomposites could effectively reverse resistance and induce apoptosis of MCF-7/ADR cells. The blood biochemistry parameters and H&E staining results showed no serious adverse effects after treatment with the nanocomposites. Prussian blue staining showed that the nanocomposites were able to target tumor tissues in tumor-bearing mice under a magnetic field. The combined chemical/photodynamic therapy significantly inhibited tumor growth in vivo. Conclusion Nanocomposites with magnetic and pH dual-responsive performance has shown a promising platform for enhanced drug-resistant breast cancer treatment.

Abstract:In the present work, Fe3O4 nanoparticles with superparamagnetic properties were prepared and capped by using Chitosan. The synthesized NPs were studied by using transmission electron microscopy (TEM) and Fourier transform infrared (FTIR) spectroscopy. Average particle size and surface charge of the synthesized NPs were characterized by using Malvern Zetasizer instrument. TEM images showed the morphology and size distribution of uncoated Fe3O4 NPs, exhibiting the uniform sized NPS with an average particle size of about 10 nm. Vibrating Scanning Magnetometry (VSM) experiments, showed the superparamagnetic nature of the prepared nanoparticles. Fe3O4 NPs showed ferromagnetic magnetization which is very sensitive towards the sample's nanostructure. The results of paramagnetic studies exhibited the substantial reduction in paramagnetic behavior after Chitosan coating but sufficient for responding in magnetic field. Further, the in-vitro ability of the Chitosan coated Fe3O4 NPs as contrast agents in efficient Ultra sound/Magnetic resonance (US/MR) imaging was investigated. These findings demonstrated that the Chitosan coated super para magnetic iron oxide nanoparticles (SPION) have reported significant contrast-enhanced imaging potential for dual-mode US/MR imaging. Hence, the prepared Chitosan coated SPION composites administration serve as potential guide in the diagnosis and treatment of cancers.
Multi-Stimuli-Responsive DOX Released from Magnetosome for Tumor Synergistic Theranostics

Abstract:BACKGROUND: To improve responses to tumor microenvironments for achieving a better therapeutic outcome in combination cancer therapy, poly(ε-caprolactone)-SS-poly(methacrylic acid) diblock copolymer (PCL-SS-PMAA) with a disulfide linkage between the hydrophobic and hydrophilic junctions was synthesized.MATERIALS AND METHODS: Repeating units of PCL and PMAA in PCL-SS-PMAA were controlled and formulated into polymersomes (PSPps). Truncated octahedral Fe3O4 nanoparticles (IONPs) were synthesized and encapsulated to produce IONPs-PSPps NPs and doxorubicin (DOX) was further loaded to produce IONPs-PSPps@DOX NPs for theranostic applications.RESULTS: IONPs-PSPps NPs remained a superparamagnetic property with a saturation magnetization value of 85 emu⋅gFe3O4 -1 and a relaxivity value of 180 mM-1⋅s-1. Upon exposure to an alternating magnetic field (AMF), IONPs-PSPps NPs increased temperature from 25°C to 54°C within 15 min. Among test groups, the cell apoptosis was greatest in the group exposed to IONPs-PSPps@DOX NPs with AMF and magnet assistance. In vivo T2-weighted magnetic resonance images of A549 tumor-bearing mice also showed highest contrast and greatest tumor suppression in the tumor with AMF and magnet assistance.CONCLUSION: IONPs-PSPps@DOX NPs are a potential theranostic agent having multifaceted applications involving magnetic targeting, MRI diagnosis, hyperthermia and chemotherapy.

Abstract:Multi‐chambered core/shell magnetic nanoparticles are developed as personalized and precise nanocarriers to provide real‐time, full‐time diagnosis and treatment integration of tumors. The supraparticles possess unique properties of hierarchical structure and full‐time release ability, showing in vitro precise uptake against cancer cells and in vivo multimodal imaging capability. To a certain extent, theranostic nanoplatforms promote tumor treatment efficiency. However, timely monitoring of the critical stages and signal sustainability of the entire process is challenging. In this study, multi‐chambered core/shell magnetic nanoparticles (MC‐MNPs) as drug and imaging agent multi‐loaded nanocarriers with a synergistic release function are reported. Supraparticles with stable chambers are formed by the supercooling self‐assembly of several core/shell magnetic nanoparticles composed of amphiphilic copolymers as the core and hydrophilic magnetic iron oxide nanoparticles as the shell. Desalinized doxorubicin and coumarin 6 are stored in different cavities of nanocarriers, and chitosan is used as an outer encapsulation layer. Based on their construction properties, MC‐MNPs can exhibit gradient‐degraded and steady‐released controllability in the tumor environment. Furthermore, real‐time accumulation situations and full‐time diagnostic signals of nanocarriers are thoroughly demonstrated using fluorescence imaging and T2‐weighted magnetic resonance imaging before and after magnetic hyperthermia in targeted tumors under an alternating magnetic field. Thus, MC‐MNPs as theranostic nanocarriers exhibit great potential for the timely monitoring and full‐time guidance of tumor treatment.

Magnetic mesoporous silica nanoparticles mediated redox and pH dual-responsive target drug delivery for combined magnetothermal therapy and chemotherapy

Abstract:A combination of multiple therapies is conducive to the treatment efficacy of cancer. Chemotherapy combined with magnetothermal therapy provides an outstanding synergy for cancer treatment. In this study, magnetic mesoporous silica nanoparticles (MMSN) were developed as a multipurpose nanoplatform for doxorubicin (DOX) delivery and magnetothermal treatment of cancer simultaneously. The MMSN were composed of Fe 3 O 4 and mesoporous silica as core and shell. The folic acid (FA) linked chitosan (CS) was conjugated at the outlets of MMSN (MMSN-SS-FA) using disulfide bonds to avert the premature release of the drug in mesopores of MMSN, target folate receptor-overexpressed cancer cells and increase the safety and physiological stability of MMSN. The temperature of the MMSN suspension could be elevated from 37 °C to 45 °C at the concentration of 1 mg/mL under an alternating magnetic field (AMF), which exceeded the tumor ablating temperature. The in vitro drug release proved that the DOX/MMSN-SS-FA displayed the redox/pH/AMF multiple-responsive release. The cellular uptake results indicated that the MMSN-SS-FA could target folate receptor-overexpressed HeLa cells. The DOX/MSNS-SS-FA with AMF had the highest cytotoxicity at the equivalent drug concentration towards HeLa cells with a combination index (CI) of 0.276, demonstrating the synergistic treatment effect of chemotherapy and magnetic-thermal therapy. Meanwhile, the DOX/MMSN-SS-FA+AMF showed the strongest tumor inhibitory effect of 84 % on tumor growth. Therefore, the constructed MMSN-SS-FA with good biocompatibility and biosafety can act as multifunctional nano-platforms for tumor treatment.

Abstract:Achieving accumulation of a target drug in tumor tissue, optimal release behavior and concurrent diagnostic imaging with therapy (theranostic) requires an ideal chemotherapy protocol that involves the responsive control of drug delivery. In the current study, we report the construction of a novel theranostic drug delivery system based on radial mesoporous silica, which is hybridized with multiscale magnetic nanoparticles for MRI-guided and alternative magnetic field (AMF) responsive chemotherapy for breast cancer. Superparamagnetic iron oxide nanoparticles (IONPs) with multiscale sizes were prepared via a hydrothermal method (for larger IONPs, lIO NPs) and a thermal decomposition method (for ultra-small IONPs, uIO NPs). lIO NPs act as the heat mediator that is responsive to the AMF, while uIO NPs act as the T-2 contrast agent for MRI. Mesoporous silica nanoparticles with radially oriented mesochannels were further in situ grown onto the surfaces of the lIO NPs, and both the uIO NPs and doxorubicin, an anti-cancer drug, can be readily incorporated within the mesochannels. To endow the as-prepared doxorubicin-loaded nano-hybrids with a good targeting ability, the surfaces of the particles were modified with folic acid (FA). This prepared drug delivery system based on magnetic/silica nanocomposites showed high drug loading efficiency and AMF stimuli responsive release properties. Meanwhile, in vitro MRI measurements revealed an excellent MRI contrast effect with a high relaxation value of 308 mM(-1) s(-1). Furthermore, both in vitro cytotoxicity analysis and in vivo anti-tumor effect evaluation demonstrated that the drug delivery system possesses great potential as an MRI-guided stimuli-responsive theranostic platform for effective active targeting chemotherapy of cancer.

Abstract:In recent years, the novel minimally invasive combination therapy for tumors has garnered widespread attention due to its controllable therapeutic target, effective combination therapy outcome, and minimal toxic side effects. This study integrated inorganic nanospheres of ferric oxide (Fe 3 O 4 ) with mesoporous silica nanoparticles (MSN), along with polymeric organic polymers—polyaniline (PANI) and hyaluronic acid (HA)—to create a multifunctional tumor treatment platform (Fe 3 O 4 @MSN@PANI@HA), aiming to enhance the efficacy of therapies that have monotherapy limitations. The Fe 2+ within the Fe 3 O 4 @MSN@PANI@HA nanoparticles can react with the unique hydrogen peroxide present in the tumor microenvironment to generate hydroxyl radicals (·OH), which possess cytotoxic properties that can selectively eradicate tumor cells, thereby facilitating chemodynamic therapy (CDT). Moreover, Fe 3 O 4 @MSN@PANI@HA nanoparticles, when loaded with the chemotherapy drug doxorubicin hydrochloride (DOX), yield Fe 3 O 4 @MSN@PANI@HA-DOX nanoparticles. It has been demonstrated that these nanoparticles exhibit commendable drug-loading efficiency and controlled drug-release capabilities, contributing to the chemotherapy of tumors. The inclusion of PANI in the Fe 3 O 4 @MSN@PANI@HA nanoparticles endows them with an exceptional photothermal conversion capability. Upon exposure to an 808-nm near-infrared laser (NIR), the photothermal conversion efficiency of the composite nanomaterials reaches 42.76%, thereby enabling photothermal therapy (PTT) through the thermal ablation of tumor cells. The HA component of the composite nanoparticles provides excellent biocompatibility and a sustained drug-release effect. Simultaneously, it could target tumor cells by recognizing specific receptors that are overexpressed on the cell surfaces, directing the transport of the composite nanoparticles to the tumor site, and selectively destroying tumor cells. This paper confirms the potent therapeutic impact of composite nanoparticles on tumors and offers a novel perspective for designing multifunctional composite nanomaterials.

Abstract:Novel multifunctional core-shell nanoparticles (NPs) have attracted widespread attention due to their easy-to-modify surface properties and abundant functional groups. This study introduces a facile approach to synthesize Ag@ iron oxide (Fe3O4) @C NPs, and modify with amino-poly (ethylene glycol) (PEG)-carboxyl and folate (FA) on the exposed carbon surface to produce high contrast for excellent stability, good biocompatibility, cancer cell targeting, and synergistic treatment. The multi-armed PEG at the edge of Ag@Fe3O4@C NPs provides the materials an excellent capacity for doxorubicin (DOX) loading. The carbon layer could be used as a photothermal reagent due to its excellent near-infrared (NIR) absorbance capacity, and Fe3O4 was used as a reagent for magnetic resonance (MR) imaging. In vivo combination therapy with this agent was administered in a mouse tumor model, and a remarkable synergistic antitumor effect that is superior to that obtained by monotherapy was achieved. Concerning these features together, these unique multifunctional Ag@Fe3O4@C-PEG-FA/DOX NPs could be regarded as an attractive nanoplatforms for chemo-photothermal synergistic tumor therapy with dual-modal fluorescence and MR imaging-guided targeting.
Superparamagnetic Hybrid Nanospheres Based on Chitosan Obtained by Double Crosslinking in a Reverse Emulsion for Cancer Treatment

Abstract:Nowadays, the Magnetically Targeted Drug Delivery System (MTDDS) is among the most attractive and promising strategies for delivering drugs to the target site. The present study aimed to obtain a biopolymer–magnetite–drug nanosystem via a double crosslinking (ionic and covalent) technique in reverse emulsion, which ensures the mechanical stability of the polymer support in the form of original hybrid nanospheres (NSMs) loaded with biologically active principles (the 5-Fluorouracil (5-FU)) as a potential treatment for cancer. Obtained NSMs were characterized in terms of structure (FT-IR), size (DLS), morphology (SEM), swelling, and 5-FU entrapment/release properties, which were dependent on the synthesis parameters (polymer concentration, dispersion speed, and amount of ionic crosslinking agent). SEM analysis results revealed that NSMs presented a spherical shape and are homogeneous and separated. Moreover, NSMs’ ability to load/release 5-FU was tested in vitro, the results confirming, as expected, their dependence on the varied synthesis process and NSM swelling ability in physiological liquids. The drug transport mechanism through the polymer matrix of its release is the Fickian type. The morphological, bio-material characteristics and the ability to include and release an antitumor drug highlight the utility of the NSMs obtained for targeting and treating some tumor diseases.

Abstract:Developing intelligent nanomedicines for cancer diagnosis and treatment (theranostics) is pivotal for improving treatment outcomes. In this study, we synthesized and evaluated dual-mode MRI contrast agents utilizing superparamagnetic iron oxide nanoparticles (SPIONs) as the core and samarium ions as the coating. Samarium ions were grafted onto the polymeric coating layer of the nanoparticles via electrostatic deposition. Characterization of the Cis/Fe3O4/PEG-co-PMMA/SmO2 core/shell structures involved size, morphology, drug loading, encapsulation efficiency, and their potential as T1-T2 dual-model contrast agents. Longitudinal relaxivity (r1) and transverse relaxivity (r2) were estimated using NMR relaxation times. In vivo investigations assessed the effectiveness of Cisplatin, particularly in human breast cancer (MCF-7) cells, using wound healing assays and molecular analysis. Remarkably, the Cis/Fe3O4/PEG-co-PMMA/SmO2 formulation exhibited enhanced cytotoxicity against MCF-7 cells compared to Cisplatin alone, indicating its promising role as an inhibitory agent against breast cancer cell proliferation.

Abstract:Although chemotherapy with magnetic nanocarriers has received a significant advancement in the field of cancer treatment, multimodal diagnosis and combitorial therapy using a single nanoplatform will have much better efficacy in achieving greater heights. Here, we have constructed a smart theranostic system by combining pH sensitive tartaric acid stabilizedFe3O4magnetic nanocarriers (TMNCs) with SPECT imaging and chemotherapeutic agents for image-guided chemo-hyperthermia therapy. The carboxyl enriched exteriors of TMNCs provide sites for conjugation of chemotherapeutic drug (doxorubicin hydrochloride, DOX) and radiolabeling (Cerium-141, 141Ce). The usage of 145.4 keV gamma made it an ideal choice for in-vivo SPECT-CT imaging which has shown retention of nanoformulation in tumor site even after 28 days. Further, the TMNCs show very high transverse relaxation rate (r2) of 171 mM-1s-1 which is higher than the clinically approved magnetic resonance imaging (MRI) contrast agents like ferumoxtran (65 mM−1 s−1) and ferumoxides (120 mM−1 s−1). Further, the developed drug loaded hybrid platform showed significantly higher cytotoxicity towards breast cancer cells and the same is augmented by in-vitro magnetic hyperthermia. Bright field microscopy and cell cycle analysis suggests that the cell death occurred through induction of G2-M arrest and subsequent apoptosis. These findings clearly suggested the potential ability of developed hybrid nanoplatform for image guided combination therapy.

Surface design and preparation of multi-functional magnetic nanoparticles for cancer cell targeting, therapy, and imaging

Abstract:Recently, theranostic candidates based on superparamagnetic iron oxide nanoparticles (SPIONs) providing the combination of therapy and diagnosis have become one of the most promising system in cancer research. However, poor stability, premature drug release, lack of specific tumor cell targeting, and complicated multi-step synthesis processes still hinder them for potential clinical applications. In this research, the multi-functional magnetic nanoparticles (MNPs-DOX) were prepared via a simple assembly process for targeted delivery of doxorubicin (DOX) and enhanced magnetic resonance (MR) imaging detection. Firstly, the multi-functional copolymer coating, polyamidoamine (PAMAM), was designed and synthesized by Michael addition reaction, where N,N-bis(acryloyl)cystamine served as backbone linker, and DOX, dopamine (DA), and polyethylene glycol (PEG) acted as comonomers. The PAMAM was then directly assembled to the surface of SPIONs by the ligand exchange reaction with SPIONs forming the MNPs-DOX. The hydrophilic PEG moieties provide the nanoparticles with colloidal stability and good-dispersity in aqueous solution. Comparing with the quick release of free DOX, the drug release behavior of MNPs-DOX exhibited a sustained drug release. Because the chemical cleavage of disulfide in the polymer backbone, a high cumulative drug release up to 60% in GSH within 48 h was observed, rather than only 26% in PBS (pH 7.4) without GSH. The MR imaging detection experiment showed that the MNPs-DOX had an enhanced T-2 relaxivity of 126 mM(-1) S-1 for MR imaging. The results of the cytotoxicity assays showed a remarkable killing effect of cancer cells by MNPs-DOX due to the FA tumor-targeting ligand, comparing with non-targeted drug molecules. All the results showed that the as prepared multi-functional magnetic nanoparticles may serve as a promising theranostic candidate for targeted anticancer drug delivery and efficient detection through MR imaging in medical application.

Abstract:Theragnostics polymer nanoparticles (NPs) loaded simultaneously with anticancer drug docetaxel (Dtxl) and superparamagnetic iron oxide (SPIO) nanocrystals were developed for both cancer therapy and ultrasensitive MRI. These multifunctional polymer vesicles were formed by carboxy-terminated poly(lactic-co-glycolic) acid using a single emulsion evaporation method. The active tumor-targeting single chain prostate stem cell antigen antibodies (scAbPSCA) were conjugated on the surface of polymer vesicles by using functional poly(ethylene glycol). The diameter of NPs was about 147 nm and the SPIO and drug encapsulation efficacy was 23% and 6.02%, respectively. Vibration simple magnetometer and X-ray diffraction proved that the superparamagnetic behavior of SPIO was not changed during NPs formation and modification. The NPs exhibited a triphasic drug release pattern in vitro over 30 days. Enhanced cellular uptake ability and antiproliferative effect of the targeted NPs in prostate cancer PC3 cell line by using the confocal laser scanning microscopy and cytotoxicity assay were observed. Moreover, the Prussian blue staining and the MRI assay in vitro demonstrated that the NPs have a high SPIO clustering effect. Therefore, these stable and tumor-targeting polymer NPs could be promising multifunctional vesicles for simultaneous targeting imaging, drug delivery and real time monitoring of therapeutic effect.
An integrated nanoplatform for theranostics via multifunctional core–shell ferrite nanocubes

Abstract:Magnetic core-shell ferrite nanocubes (MNCs) were prepared by a two-step pyrolysis. The MNCs not only exhibit an excellent magnetothermal effect, but also can be used as T-2 magnetic resonance (MR) imaging agents. To obtain their good biocompatibility and targeting ability, MNCs were modified with poly(ethylene glycol) (PEG) and hyaluronic acid (HA). To further construct an integrated nanoplatform for theranostics, doxorubicin (DOX) was loaded onto the surface of MNCs by pH and heat sensitive chemical bonding. Notably, the MNCs showed a great stability and magnetothermal effect. Moreover, they showed negligible toxicity and synergistic therapy in vitro. Meanwhile, their MR imaging in vivo was further verified. A novel integrated nanoplatform facilitates excellent targeted MR imaging guided synergism of magnetothermal and chemotherapy. The multifunctional nanocubes will be capable of playing a vital role in future cancer therapy.

Abstract:Construction of stimuli-responsive theranostic nanoagents that can increase the accuracy of imaging diagnosis and boost the therapeutic efficacy has been demonstrated for a promising approach for diagnosis and treatment of cancer. Herein, we constructed a novel theranostic agent with Co-P nanocomposites as core, mesoporous silica as shell, and manganese dioxide (MnO2) nanosheets as gatekeeper, which have been employed for pH-activatable T1/T2 dual-modality magnetic resonance imaging (MRI)-guided chemotherapeutical and photothermal combination anticancer therapy in vitro and in vivo. Co-P core-enabled theranostic platform could be applied for both photothermal therapy and T2-weighted MRI in the normal circulation owing to its strong near-infrared absorbance and intrinsic magnetic properties. In the acidic environment of tumors, MnO2 cap could be dissolved into Mn2+ ions to not only realize pH-responsive on-demand drug release but also activate T1-weighted MRI contrast enhancement. Such T1/T2 dual-mode MR imaging provides further comprehensive details and accurate information for tumor diagnosis, and the on-demand chemo-photothermal synergetic therapy greatly improved the therapeutic effectiveness and effectively mitigated side effects. These findings demonstrate that Co-P@mSiO2@DOX-MnO2 are promising as pH-responsive theranostic agents for tumor diagnosis and treatment, and stimulate interest in exploration of novel stimuli-responsive theranostic nanoagents which posssess good potential for clinical application in the future.

Abstract:In this review, we discussed the establishment of a so-called "theranostic" system by instituting the basic principles including the use of: [1] magnetic iron oxide nanoparticles (MION)-based drug carrier; [2] intra-arterial (I.A.) magnetic targeting; [3] macromolecular drugs with unmatched therapeutic potency and a repetitive reaction mechanism; [4] cell-penetrating peptide-mediated cellular drug uptake; and [5] heparin/protamine-regulated prodrug protection and tumor-specific drug re-activation into one single drug delivery system to overcome all possible obstacles, thereby achieving a potentially non-invasive, magnetic resonance imaging-guided, clinically enabled yet minimally toxic brain tumor drug therapy. By applying a topography-optimized I.A. magnetic targeting to dodge rapid organ clearance of the carrier during its first passage into the circulation, tumor capture of MION was enriched by >350 folds over that by conventional passive enhanced permeability and retention targeting. By adopting the prodrug strategy, we observed by far the first experimental success in a rat model of delivering micro-gram quantity of the large β-galactosidase model protein selectively into a brain tumor but not to the ipsi- or contra-lateral normal brain regions. With the therapeutic regimens of most toxin/siRNA drugs to fully (>99.9%) eradicate a tumor being in the nano-molar range, the prospects of reaching this threshold become practically accomplishable.

Abstract:Superparamagnetic iron oxide nanoparticles have recently been investigated for their potential to kill cancer cells with promising results, owing to their ability to be targeted and heated by magnetic fields. In this study, novel hydrogel, chitosan Fe3O4 magnetic nanoparticles were synthesized to induce magnetic hyperthermia, and targeted delivering of chemotherapeutics in the cancer microenvironment. The characteristic properties of synthesized bare and CS-MNPs were analyzed by various analytical methods: X-ray diffraction, Fourier transformed infrared spectroscopy, Scanning electron microscopy and Thermo-gravimetric analysis/differential thermal analysis. Magnetic nanoparticles were successfully synthesized using the co-precipitation method. This synthesis technique resulted in nanoparticles with an average particle size of 16 nm. The pure obtained nanoparticles were then successfully encapsulated with 4-nm-thick chitosan coating. The formation of chitosan on the surface of nanoparticles was confirmed by physicochemical analyses. Heating experiments at safe magnetic field (f = 100 kHz, H =10-20 kA m-1) revealed that the maximum achieved temperature of water stable chitosan-coated nanoparticles (50 mg ml-1) is fully in agreement with cancer therapy and biomedical applications.

Abstract:The magnetic resonance imaging diagnosis and high-efficiency tumor targeting treatment are of notable importance for cancer therapy. In this study, starch-octanoic acid (ST-OA) micelles were successfully prepared to co-encapsulate the anti-tumor drug doxorubicin (DOX) and superparamagnetic iron oxide nanoparticles (SPIONs), taking advantage of the hydrophobic core of the core-shell micellar structure. Size distribution, morphology and in vitro release behaviors of micelles were investigated. In addition, magnetic properties and the anti-tumor theranostic performance were confirmed by conducting the cellular uptake, in vivo imaging and anti-tumor activity experiments. Therefore, co-encapsulation and specific delivery of the chemotherapeutic drug and contrast agent into tumor cells can realize the diagnosis and treatment of malignant tumor at the same time.
