Abstract:Plants create secondary metabolites which may or may not have biological activities and are not always benign. Therefore, it is essential to assess both their bioactivities and safety. The objective of this research is to assess the in vitro toxicity and phytochemistry of extracts from Bauhinia galpinii, Combretum caffrum , and Maytenus undata extracts. In the past, it has been demonstrated that these plants possess potent antifungal activities against mycotoxigenic fungal strains. The MTT assay was used to evaluate the cytotoxicity of methanolic leaf extracts of selected plants on the viability of the African Green monkey kidney and the human colorectal adenocarcinoma cells. Furthermore, the ability of these plants to cause genotoxicity in Vero cells was investigated using the micronucleus test. The identity and quantity of the phytocompounds present in the plant extracts were analyzed using liquid-chromatography time-of-flight mass spectrometry (LC-ToF-MS) analysis. B. galpinii, C. caffrum , and M. undata extracts were cytotoxic to Vero cells at 500 μg/ml. All the selected medicinal plant extracts demonstrated dose-dependent cytotoxicity against both Caco-2 and Vero cells. B. galpinii, M. obtusifolia , and M. undata extracts were all genotoxic towards the Vero cells at 500 μg/ml, while Combretum caffrum showed genotoxicity at 250 μg/ml. Phenolic glycosides, coumarins, flavonoids, and hydrolysable tannins were identified as major phytochemicals in the secondary metabolite profiling. Given the results, caution in terms of quantities consumed should be exercised when these plants are used in traditional medicine.

Abstract:The emergence of drug-resistant Mycobacterium tuberculosis strains is a threat to global health necessitating the discovery of novel chemotherapeutic agents. Natural products drug discovery, which previously led to the discovery of rifamycins, is a valuable approach in this endeavor. Against this backdrop, we set out to investigate the in vitro antimycobacterial properties of medicinal plants from Ghana and South Africa, evaluating 36 extracts and their 252 corresponding solid phase extraction (SPE) generated fractions primarily against the non-pathogenic Mycobacterium smegmatis and Mycobacterium aurum species. The most potent fraction was further evaluated in vitro against infectious M. tuberculosis strain. Crinum asiaticum (bulb) (Amaryllidaceae) emerged as the most potent plant species with specific fractions showing exceptional, near equipotent activity against the non-pathogenic Mycobacterium species (0.39 μg/ml ≤ MIC ≤ 25 μg/ml) with one fraction being moderately active (MIC = 32.6 μg/ml) against M. tuberculosis . Metabolomic analysis led to the identification of eight compounds predicted to be active against M. smegmatis and M. aurum . In conclusion, from our comprehensive study, we generated data which provided an insight into the antimycobacterial properties of Ghanaian and South African plants. Future work will be focused on the isolation and evaluation of the compounds predicted to be active.

Assessment of Anti-Proliferative Activities of Selected Medicinal Plant Extracts Used for Management of Diseases around Lake Victoria Basin

Abstract:The devastating effect of cancer is a worldwide concern. Unfortunately current chemotherapeutic and radio therapeutic modalities have been found to possess side effects coupled with the emergence of drug resistance. This has necessitated the search for novel therapeutic products with better efficacy, safety and affordability through identification of anti-tumor agents from natural products. In this study the antineoplastic activity and phytochemical screening of methanol extracts of four medicinal plants that are native to Lake Victoria Basin, including Piptadiniastrum africanum, Kigelia africana, Centella asciatica and Chaemocrista nigricans was investigated. Extraction and concentration of the collected and dried plant samples to obtain crude extracts as well as phytochemical screening of the crude extracts was done following standard procedures. Lung adenocarcinoma cell line, from American type cell culture (ATCC), was exposed to the extracts and antiproliferative analysis using 3-(4, 5-Dimethyl-2-thiazolyl)-2, 5-diphenyl-2-tetrazolium bromide (MTT) colorimetric assay was performed. Phytochemical analysis of each of the four plants showed presence of terpenoids, alkaloids, saponins, tannins and steroids. Steroids were higher in the bark of P. africanum and the fruits of K. africana. Flavonoid was absent only in extracts of C. nigricans and coumarins present only in the extracts of K. africana. The methanolic extracts of the fruits of K. africana, bark of P. africanum, leaves of C. asciatica and leaves C. nigricans had inhibitory effects with IC50 28.86 ug/ml, 26.57 ug/ml, 15.69 ug/ml and 8.07 ug/ml (p values of 0.079, 0.069, and 0.042 and 0.055, ANOVA) respectively. Statistical differences among fractions of the extract were determined by one way ANOVA and considered significant at P<0.05. The MTT assay results indicated that all of the extracts had the capacity to inhibit the proliferation of tumor cells. The Phytochemical compounds which were tested are antioxidant and shown to trigger morphological changes, such as blebbing pattern and cell shrinkage, associated with apoptosis in the lung adenocarcinoma cell lines. The varying inhibitory activities against tested human lung adenocarcinoma cell lines justify the traditional use of these plants in the management of cancers and other illnesses. These extracts have a potential to be further developed into anti-tumor agents. However, further phytochemical characterization of the extracts is necessary for more extensive biological evaluation of the active ingredients and to exhaustively describe the mechanisms of cancer cell apoptosis of these four plant extracts.

Abstract:Annona senegalensis pers, Sutherlandia frutescens (L.), and Withania somnifera (L.) are abundant plants and widely distributed in the Limpopo, Gauteng, Kwazulu-Natal, North West and Mpumalanga provinces in South Africa. The three plants are among those used by traditional healers and herbalists in South Africa for the treatment of a variety of diseases, including cancer. The current study aimed at the phytochemical screening and characterization of volatile compounds from the three medicinal plants using GC-MS. The methanol leaf extracts were subjected to phytochemical screening using standard chemical tests to detect the presence of different classes of compounds. Volatile compounds were detected by GC-MS analysis, and detected compounds were identified by comparing the MS spectral data with those of compounds deposited in the NIST Library (NIST08). Phytochemical screening indicated the presence of different secondary metabolites such as alkaloids, quinones, steroids, cardiac glycosides, coumarins, and terpenoids in all plants. GC-MS chromatograms allowed the detection and identification of 19 volatile compounds among the three plants with known bioactivities that are important in the management of life-threatening diseases such as cancer and diabetes. The results confirm the leaves of Annona senegalensis, Sutherlandia frutescens, and Withania somnifera as sources of important phytochemicals and therefore justify their use for the treatment of various diseases by traditional healers.

Abstract:Abstract Approximately 20% of all breast cancer cases are Triple negative breast cancer (TNBC), whichis more difficult to treat and has a higher mortality rate than other types of breast cancer. Thereis an active practice of traditional medicine in in South Africa, in the form of infusions madeusing plant material. Previously we demonstrated that water soluble extracts from the leaves ofone of these plants, African garlic (Tulbaghia violacea), is able to induce apoptosis in TNBCcell lines, while also inhibiting the ability of these cells to migrate or penetrate a basementmembrane. In an attempt to isolate the molecular compounds that are present in this extract,water soluble leaf extracts were subjected to Fourier transform infrared spectroscopy (FITR),Gas chromatography–mass spectrometry (GC-MS) and Nuclear magnetic resonance (NMR)spectroscopy. Identified compounds of interest were then used in computational dockingstudies with the anti-apoptotic protein Cox2. The FITR analysis indicated that the extractconsisted of multiple compounds, belonging to a variety of classes. GC-NS and NMRspectroscopy identified 36 and 61 separate compounds respectively. Many of these compoundswere similar in structure to compounds with known anti-cancer properties. Many compoundswith established medical uses were also identified in the extract. Five compounds identified inthe extract, d-Glycero-d-galacto-heptose, H-Pyran-4-one, 2,3-dihydro-3,5-dihydroxy-6-methyl, and Benzaldehyde and 4-(1-methylethyl)- demonstrated high binding affinity towardsCOX2. With d-Glycero-d-galacto-heptose having a higher docking score than manycommercially available COX2 inhibitors. Based on our findings, the compounds within theextract could be used as the basis for the development of safe and effective drugs for thetreatment of TNBC and other cancers. In order to fulfil this potential, future work shouldexamine the molecular pathways affected by the treatment of TNBC cells with this extract orindividual compounds. Abstract:Abstract Background Tuberculosis (TB) is one of the leading causes of death globally, and the rise in drug-resistant forms of TB has become a significant threat. Subsequently, it is crucial to explore new, effective and safe anti-TB agents. This study aimed at conducting phytochemical screening, antimycobacterial activity, and acute toxicity of the selected plant species’ crude extracts to assess their toxicological potentials and efficacies against TB. Methods The aqueous and methanol/dichloromethane (DCM) (1:1) extracts of each selected plant species were subjected to phytochemical screening and antimycobacterial activity using microplate alamar blue assay. For acute toxicity, a single dose (2000 mg/kg) of the aqueous extracts was orally administered to each animal following the Organization for Economic Cooperation and Development (OECD) guidelines No. 425 and then observed for 14 days. The animals were closely observed on the general behavior and clinical signs of toxicity, and body weights were recorded. After the termination of the experiment, hematological, biochemical, and histopathological analyses were performed. Results The extracts contained alkaloids, flavonoids, tannins, saponins, steroids, terpenoids, resins, cardiac glycosides, phenolic compounds, and coumarins. Aqueous extracts showed moderate to weak activity against the susceptible (H 37 Rv) M. tuberculosis strain and weak activity against the MDR-TB strain with Minimum Inhibitory Concentrations (MIC μg/mL) ranging from 293.0–2344.0 and 1172.0–4688.0, respectively. Methanol/DCM extracts showed significant to moderate activity against the susceptible TB strain and moderate to weak activity against the MDR-TB strain with MIC (μg/mL) ranging from 98.0–586.0 and 293.0–781.0, respectively. One mortality was recorded from the A. coriaria treated group following the acute toxicity tests, but the LD 50 of all the extracts was estimated to be above 2000 mg/kg. Histopathological analyses did not show any significant lesions in the examined organs except those from the A. coriaria treated group. Conclusion Phytochemical screening of the extracts revealed the presence of alkaloids, tannins, saponins, flavonoids, steroids, terpenoids, resins, cardiac glycosides, phenolic compounds, and coumarins. All the methanol/DCM extracts of the plant species studied have promising antimycobacterial activity. The selected plant extracts studied exhibited low acute toxicity levels except for A. coriaria and could be safe for formulations into herbal products.
Exploring Water-Soluble South African Tulbaghia violacea Harv Extract as a Therapeutic Approach for Triple-Negative Breast Cancer Metastasis

Abstract:Triple-negative breast cancer (TNBC) accounts for approximately 20% of all breast cancer cases and is characterized by a lack of estrogen, progesterone, and human epidermal growth factor 2 receptors. Current targeted medicines have been unsuccessful due to this absence of hormone receptors. This study explored the efficacy of Tulbaghia violacea, a South African medicinal plant, for the treatment of TNBC metastasis. Extracts from T. violacea leaves were prepared using water and methanol. However, only the water-soluble extract showed anti-cancer activity and the effects of this water-soluble extract on cell adhesion, invasion, and migration, and its antioxidant activity were assessed using MCF-10A and MDA-MB-231 cells. The T. violacea extract that was soluble in water effectively decreased the movement and penetration of MDA-MB-231 cells through the basement membrane in scratch and invasion tests, while enhancing their attachment to a substance resembling an extracellular matrix. The sample showed mild-to-low antioxidant activity in the antioxidant assy. Nuclear magnetic resonance spectroscopy revealed 61 chemical components in the water-soluble extract, including DDMP, 1,2,4-triazine-3,5(2H,4H)-dione, vanillin, schisandrin, taurolidine, and α-pinene, which are known to have anti-cancer properties. An in-depth examination of the transcriptome showed alterations in genes linked to angiogenesis, metastasis, and proliferation post-treatment, with reduced activity in growth receptor signaling, angiogenesis, and cancer-related pathways, such as the Wnt, Notch, and PI3K pathways. These results indicate that T. violacea may be a beneficial source of lead chemicals for the development of potential therapeutic medicines that target TNBC metastasis. Additional studies are required to identify the precise bioactive chemical components responsible for the observed anti-cancer effects.
Toxicity and antimicrobial interactions with conventional antimicrobials of commonly traded medicinal plants from Faraday muthi market (Johannesburg, South Africa)

Abstract:In South Africa, medicinal plants are frequently used to treat infections, with the Faraday muthi market in Johannesburg ensuring accessibility to these medicines. This study aimed to determine the in vitro interactive antimicrobial and toxicity profiles of commonly traded traditional medicinal plants at the Faraday muthi market, with a range of conventional antimicrobials. Minimum inhibitory concentration (MIC) assays were conducted to determine the in vitro interactive profiles and tested against the ESKAPE pathogens ( Enterococcus faecium ATCC 27270, Staphylococcus aureus ATCC 29212, Klebsiella pneumoniae ATCC 13883, Acinetobacter baumannii ATCC 19606, Pseudomonas aeruginosa ATCC 27853, Enterobacter cloacae ATCC 13460) and yeasts, Candida albicans ATCC 10231 and Candida glabrata ATCC 90030. The fractional inhibitory concentration index (ΣFIC) was determined to classify interactions as synergistic, additive, non-interactive or antagonistic. The brine-shrimp lethality assay (BSLA) was conducted to determine the toxicity of synergistic interactions. A total of 816 plant: antibiotic combinations were tested against the ESKAPE pathogens, with 392 combinations exhibiting a definitive ΣFIC. Synergy (6.63 %), antagonism (47.45 %), additive (8.67 %) and non-interactive (37.24 %) profiles were observed. Medicinal plants were most widely synergistic with azithromycin against S. aureus. Hypoxis hemerocallidea with azithromycin against S. aureus (ΣFIC 0.06) was the most synergistic, with no toxicity evident in the BLSA (6.06 % mortality). The most antagonistic interaction was with Eucalyptus sideroxylon and amoxicillin against K. pneumoniae (ΣFIC 138.12). Furthermore, a total of 136 plant: antifungal combinations were tested, where a definitive ΣFIC was obtained for 84 of the combinations. No synergy was observed, but 61.91 % of the combinations were antagonistic. The most antagonistic plant: antifungal combination was observed with Senecio serratuoloides and nystatin against C. albicans (ΣFIC 34.6). The vast antagonism against the ESKAPE pathogens (47.49 %) and yeasts (61.90 %) is cause for concern, particularly with antimicrobial resistance development. The most toxic combinations in the BSLA were when azithromycin was combined with Synaptolepis oliveriana (54.50 % average mortality after 48 h) and with Warburgia salutaris (100 % average mortality after 48 h). The synergy and antagonism observed in this study warrants further investigation for clinical translation, to determine potentially beneficial or harmful combinations where applicable.

Abstract:Development of cheap and reliable diagnostics is pivotal in the control of infectious diseases. Currently, one of the best strategies to tackle infectious disease is the use of natural products from plant origin, due to the perception that long term use of western medicine induces severe complications and also to stop the prevalence of antibacterial infection. This study aims at substantiating the traditional use of selected medicinal plants with antibacterial claim for possible lead for the development of more potent drugs in the treatment of bacterial infections. The preliminary phytochemical content, antioxidant and antibacterial potential of the selected medicinal plants were evaluated using standard procedures. Thirtheen plants were obtained through questionnaire administration: Garcinia kola, Costus afer, Vitellaria paradoxa, Pycnanthus angolensis, Cola acuminata Acanthospermum hipida, Aloe vera, Euadenia trifoliate, Microglossa abzelii, Moringa oleifera, Bryophyllum pinnatum, Baphia nitida and Afromonium melegueta. Thin layer chromatography (TLC) profiling of the various selected plants were carried out using two solvent system consisting of n-Hexane and ethyl acetate in varying ratio (8:2 and 7:3). Eight (8) plants, which showed good chromatographical profiling with the used solvent, were selected for further experiment (Garcinia kola, Vitellaria paradoxa, Acanthospermum hispidum, Aloe vera, Microglossa abzelii, Baphia nitida, Moringa oleifera and Bryophyllum pinnatum). These plants were subjected to qualitative phytochemistry and five plants (Moringa oleifera, Vitellaria paradoxa, Baphia nitida, Bryophyllum pinnatum and Garcinia kola) selected for quantitative phyto-screening for alkaloids, flavonoids, saponins and tannins. The antibacterial screening was carried out using Agar well diffusion and minimum inhibitory concentration (MIC) against the following isolate Staphylococcus aureus, Escherichia coli, Salmonella typhi and Bacillus subtilis. The antioxidant potential of the selected five plants were evaluated using in vitro (DPPH and FRAP) and the best plant was used for in vivo (SOD and MDA) analysis. The phytochemistry of the 13 plants revealed the presence or absence of Alkaloids, saponins, tannins, flavonoids, terpenoids, cardiac glycosides, carbohydrates and protein in the selected plants, and the best five were selected for quantitative evaluation showed they all have good quantity of alkaloids, flavonoids, saponins and tannins. The plants exhibited mild to strong antibacterial activities, having an MIC value ranging from 2.5 to >20 μg/mL when compared to ciprofloxacin (control) that is 5μg/mL. The plants showed good free radical scavenging activities and efficient ferric reducing power in a concentration dependent manner and a good enzyme activity of the superoxide dismutase (SOD) at a concentration of 100 μ/mL with a reduction in malondialdehyde (MDA) production as a marker of lipid peroxidation. The various plants have potentials for development of drugs for the treatment of infectious diseases; and a potential for further evaluation of their antibacterial claim.
Contribution of Sub-Saharan African medicinal plants to cancer research: Scientific basis 2013–2023

Abstract:Cancer incidence and mortality rates are increasing worldwide. Cancer treatment remains a real challenge for African countries, especially in sub-Saharan Africa where funding and resources are very limited. High costs, side effects and drug resistance associated with cancer treatment have encouraged scientists to invest in research into new herbal cancer drugs. In order to identify potential anticancer plants for drug development, this review aims to collect and summarize anticancer activities (in vitro/in vivo) and molecular mechanisms of sub-Saharan African medicinal plant extracts against cancer cell lines. Scientific databases such as ScienceDirect, Google Scholar and PubMed were used to search for research articles published from January 2013 to May 2023 on anticancer medicinal plants in sub-Saharan Africa. The data were analyzed to highlight the cytotoxicity and molecular mechanisms of action of these listed plants. A total of 85 research papers covering 204 medicinal plant species were selected for this review. These plants come from 57 families, the most dominant being the plants of the family Amaryllidaceae (16), Fabaceae (14), Annonaceae (10), Asteraceae (10). Plant extracts exert their anticancer activity mainly by inducing apoptosis and stopping the cell cycle of cancer cells. Several plant extracts from sub-Saharan Africa therefore have strong potential for the search for original anticancer phytochemicals. Chemoproteomics, multi-omics, genetic editing technology (CRISPR/Cas9), combined therapies and artificial intelligence tools are cutting edge emerging technologies that facilitate the discovery and structural understanding of anticancer molecules of medicinal plants, reveal their direct targets, explore their therapeutic uses and molecular bases.

Abstract:Cancer is one of the leading causes of death worldwide. In recent years, African countries have been faced with a rapid increase in morbidity and mortality due to this pathology. Management is often complicated by the high treatment costs, side effects and the increasing occurrence of resistance to treatments. The identification of new active ingredients extracted from endemic medicinal plants is definitively an interesting approach for the implementation of new therapeutic strategies: their extraction is often lower cost; their identification is based on an ethnobotanical history and a tradipratic approach; their use by low-income populations is simpler; this can help in the development of new synthetic molecules that are more active, more effective and with fewer side effects. The objective of this review is to document the molecules derived from African medicinal plants whose in vitro anti-cancer activities and the mechanisms of molecular actions have been identified. From the scientific databases Science Direct, PubMed and Google Scholar, we searched for publications on compounds isolated from African medicinal plants and having activity on cancer cells in culture. The data were analyzed in particular with regard to the cytotoxicity of the compounds and their mode of action. A total of 90 compounds of these African medicinal plants were selected. They come from nine chemical groups: alkaloids, flavonoids, polyphenols, quinones, saponins, steroids, terpenoids, xanthones and organic sulfides. These compounds have been associated with several cellular effects: i) Cytotoxicity, including caspase activation, alteration of mitochondrial membrane potential, and/or induction of reactive oxygen species (ROS); ii) Anti-angiogenesis; iii) Anti-metastatic properties. This review points out that the cited African plants are rich in active ingredients with anticancer properties. It also stresses that screening of these anti-tumor active ingredients should be continued at the continental scale. Altogether, this work provides a rational basis for the selection of phytochemical compounds for use in clinical trials.
Molecular docking, molecular dynamics simulations and binding free energy studies of interactions between Mycobacterium tuberculosis Pks13, PknG and bioactive constituents of extremophilic bacteria

Abstract:Mycobacterial pathogens present a significant challenge to disease control efforts globally due to their inherent resistance to multiple antibiotics. The rise of drug-resistant strains of Mycobacterium tuberculosis has prompted an urgent need for innovative therapeutic solutions. One promising way to discover new tuberculosis drugs is by utilizing natural products from the vast biochemical space. Multidisciplinary methods can used to harness the bioactivity of these natural products. This study aimed to evaluate the antimycobacterial efficacy of functional crude extracts from bacteria isolated from gold mine tailings in South Africa. Bacterial strains were identified using 16S rRNA sequencing. The crude extracts obtained from the bacteria were tested against Mycobacterium tuberculosis H37Rv, Mycobacterium smegmatis mc 2 155, and Mycobacterium aurum A+. Untargeted HPLC-qTOF and molecular networking were used to identify the functional constituents present in extracts that exhibited inhibitory activity. A virtual screening workflow (VSW) was used to filter compounds that were strong binders to Mycobacterium tuberculosis Pks13 and PknG. The ligands returned from the VSW were subjected to optimization using density functional theory (DFT) at M06-2X/6-311++ ( d,p ) level of theory and basis set implemented in Gaussian16 Rev.C01. The optimized ligands were re-docked against Mycobacterium tuberculosis Pks13 and PknG. Molecular dynamics simulation and molecular mechanics generalized born surface area were used to evaluate the stability of the protein–ligand complexes formed by the identified hits. The hit that showed promising binding characteristics was virtually modified through multiple synthetic routes using reaction-driven enumeration. Three bacterial isolates showed significant activity against the two strains of Mycobacterium , while only two, Bacillus subtilis and Bacillus licheniformis , exhibited activity against both Mycobacterium tuberculosis H37Rv, Mycobacterium smegmatis mc 2 155, and Mycobacterium aurum A+. The tentatively identified compounds from the bacterial crude extracts belonged to various classes of natural compounds associated with antimicrobial activity. Two compounds, cyclo-(L-Pro-4-OH-L-Leu) and vazabitide A, showed strong binding against PknG and Pks13, with pre-MD MM-GBSA values of − 42.8 kcal/mol and − 47.6 kcal/mol, respectively. The DFT-optimized compounds exhibited the same docking scores as the ligands optimized using the OPSL-4 force field. After modifying vazabitide A, its affinity to the Pks13 binding site increased to − 85.8 kcal/mol, as revealed by the post-MD MM-GBSA analysis. This study highlights the potential of bacteria isolates from gold mine tailings as a source of new scaffolds for designing and optimizing anti- Mycobacterium agents. These agents synthesized in-silico can be further tested in-vitro to evaluate their efficacy.

Abstract:According to the World Health Organization, women's breast cancer is among the most common cancers with 7.8 million diagnosed cases during 2016–2020 and encompasses 15 % of all female cancer-related mortalities. These mortality events from triple-negative breast cancer are a significant health issue worldwide calling for a continuous search of bioactive compounds for better cancer treatments. Historically, plants are important sources for identifying such new bioactive chemicals for treatments. Here we use high-throughput screening and mass spectrometry analyses of extracts from 100 plant species collected in Chinese ancient forests to detect novel bioactive breast cancer phytochemicals. First, to study the effects on viability of the plant extracts, we used a MTT and CCK-8 cytotoxicity assay employing triple-negative breast cancer (TNBC) MDA-MB-231 and normal epithelial MCF-10A cell lines and cell cycle arrest to estimate apoptosis using flow cytometry for the most potent three speices. Based on these analyses, the final most potent extracts were from the Amur honeysuckle ( Lonicera maackii ) wood/root bark and Nigaki ( Picrasma quassioides ) wood/root bark. Then, 5 × 10 6 MDA-MB-231 cells were injected subcutaneously into the right hind leg of nude mice and a tumour was allowed to grow before treatment for seven days. Subsequently, the four exposed groups received gavage extracts from Amur honeysuckle and Nigaki (Amur honeysuckle wood distilled water, Amur honeysuckle root bark ethanol, Nigaki wood ethanol or Nigaki root bark distilled water/ethanol (1:1) extracts) in phosphate-buffered saline (PBS), while the control group received only PBS. The tumour weight of treated nude mice was reduced significantly by 60.5 % within 2 weeks, while on average killing 70 % of the MDA-MB-231 breast cancer cells after 48 h treatment (MTT test). In addition, screening of target genes using the Swiss Target Prediction, STITCH, STRING and NCBI-gene database showed that the four plant extracts possess desirable activity towards several known breast cancer genes. This reflects that the extracts may kill MBD-MB-231 breast cancer cells. This is the first screening of plant extracts with high efficiency in 2 decades, showing promising results for future development of novel cancer treatments.

Abstract:Salvia africana-lutea L., S. lanceolata L., and S. chamelaeagnea L. are used in South Africa as traditional medicines to treat infections. This paper describes an in-depth investigation into their antibacterial activities to identify bioactive compounds. Methanol extracts from 81 samples were screened against seven bacterial pathogens, using the microdilution assay. Biochemometric models were constructed using data derived from minimum inhibitory concentration (MIC) and ultra-performance liquid chromatography-mass spectrometry data. Active molecules in selected extracts were tentatively identified using high-performance thin layer chromatography (HPTLC), combined with bioautography, and finally, by analysis of active zone eluates by mass spectrometry (MS) via a dedicated interface. Salvia chamelaeagnea displayed notable activity towards all seven pathogens, and the activity, reflected by MICs, was superior to that of the other two species, as confirmed through ANOVA. Biochemometric models highlighted potentially bioactive compounds, including rosmanol methyl ether, epiisorosmanol methyl ether and carnosic acid. Bioautography assays revealed inhibition zones against A. baumannii, an increasingly multidrug-resistant pathogen. Mass spectral data of the eluted zones correlated to those revealed through biochemometric analysis. The study demonstrates the application of a biochemometric approach, bioautography, and direct MS analysis as useful tools for the rapid identification of bioactive constituents in plant extracts.

Abstract:Respiratory tract infections (RTIs) are frequent ailments among humans and are a high burden on public health. This study aimed to determine the in vitro antibacterial, anti-inflammatory, and cytotoxic effects of indigenous medicinal plants used in the treatment of RTIs, namely, Senna petersiana, Gardenia volkensii, Acacia senegal, and Clerodendrum glabrum. Dried leaves were extracted using various organic solvents. Antibacterial activity was quantified using the microbroth dilution assay. Protein denaturation assays were used to evaluate anti-inflammatory activity. The cytotoxicity of the extracts towards THP-1 macrophages was evaluated using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Antioxidant activity was determined using free radical scavenging activity and ferric-reducing power. Total polyphenolics were quantified. Liquid chromatography mass spectrometry was used to evaluate the acetone plant extracts. Nonpolar extracts had noteworthy antibacterial activity against Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, and Mycobacterium smegmatis where MIC values ranged between 0.16 and 0.63 mg/mL. At 100 μg/mL, A. senegal, G. volkensii, and S. petersiana had a nonsignificant effect on the viability of the THP-1 macrophages. The LC-MS analysis of the leaf extracts of S. petersiana detected Columnidin, Hercynine, L-Lysine citrate, and Gamma-Linolenate. A pentacyclic triterpenoid, cochalate, was detected in G. volkensii. Two flavonoids 7-hydroxy-2-(4-methoxyphenyl)-4-oxo-chroman-5-olate and (3R)-3-(2,4-dimethoxyphenyl)-7-hydroxy-4-oxo-chroman-5-olate were detected in the C. glabrum extract. The findings from this study indicated that the leaves of the selected plant extracts possess antioxidant, anti-inflammatory, and antibacterial activity. Therefore, they may serve as good candidates for further pharmaceutical investigations.
Cytotoxic properties, glycolytic effects and high-resolution respirometry mitochondrial activities of Eriocephalus racemosus against MDA-MB 231 triple-negative breast cancer

Abstract:Introduction: Triple-negative breast cancer (TNBC) represents a significant global health crisis due to its resistance to conventional therapies and lack of specific molecular targets. This study explored the potential of Eriocephalus racemosus (E. racemosus) as an alternative treatment for TNBC. The cytotoxic properties and high-resolution respirometry mitochondrial activities of E. racemosus against the MDA-MB 231 TNBC cell line were evaluated. Methods: Hexane solvent and bioactive fraction extractions of E. racemosus were performed, while mass spectrometry-based metabolite profiling was used to identify the phytochemical constituents of the extracts. The extracts were further tested against MDA-MB 231 TNBC cells to determine their cytotoxicity. The mode of cell death was determined using flow cytometry. The activities of caspases 3, 8, and 9 were assessed using a multiplex activity assay kit. Glycolytic activity and High-resolution respirometry measurements of mitochondrial function in the MDA-MB 231 cell line were conducted using the Seahorse XFp and Oroboros O2K. Results: Metabolite profiling of E. racemosus plant crude extracts identified the presence of coumarins, flavonoids, sesquiterpenoids, triterpenoids, and unknown compounds. The extracts demonstrated promising cytotoxic activities, with a half maximal inhibitory concentration (IC50) of 12.84 µg/mL for the crude hexane extract and 15.49 µg/mL for the bioactive fraction. Further, the crude hexane and bioactive fraction extracts induced apoptosis in the MDA-MB-231 TNBC cells, like the reference drug cisplatin (17.44%, 17.26% and 20.25%, respectively) compared to untreated cells. Caspase 3 activities confirmed the induction of apoptosis in both cisplatin and the plant crude extracts, while caspase 8 and 9 activities confirmed the activation of both the intrinsic and extrinsic apoptosis pathways. Increased levels of glycolytic activity were observed in the hexane crude extract. High-resolution respiratory measurements showed elevated mitochondrial activities in all mitochondrial states except for complex-IV activity. Conclusion: These findings support further exploration of E. racemosus as a potential therapeutic agent for TNBC, offering a promising avenue for the development of targeted treatments with minimal adverse effects.
Utilizing Indigenous Flora in East Africa for Breast Cancer Treatment: An Overview

Abstract:Background: Breast cancer is a significant global health challenge, contributing substantially to cancer- related deaths. Conventional treatment methods, including hormone therapy, chemotherapy, surgical interventions, and radiation, have long been utilized. However, these traditional treatments are often associated with serious side effects and drug resistance, limiting their efficacy. Aim: This review aims to explore the potential of medicinal plants used in breast cancer management in East Africa, focusing on their bioactive compounds and anticancer properties. Methods: A comprehensive literature search was conducted to examine the effectiveness of medicinal plants in treating breast cancer across Kenya, Ethiopia, Uganda, Tanzania, and Rwanda. Relevant studies published between 2003 and 2023 were identified using keywords related to breast cancer and medicinal plants. The search was performed across multiple databases, including Google Scholar, PubMed, Scopus, Web of Science Core Collection, and Science Direct. Results: Numerous natural compounds found in East African medicinal plants including Cymbopogon citratus (Lemongrass,) Tabebuia avellanedae, Prunus africana (African Cherry), Euclea divinorum, Berberis holstii, Withania somnifera (Ashwagandha, Curcuma longa (Turmeric), Garcinia mangostana (Mangosteen, Vitis vinifera (Grapevine), Eugenia jambolana (Java Plum), Moringa oleifera (Drumstick Tree), Camellia sinensis (Tea), Glycine max (Soybean), Catharanthus roseus, Madagascar Periwinkle), Rhus vulgaris (Wild Currant) exhibit significant anticancer properties. These compounds have demonstrated the ability to reduce breast cancer aggressiveness, inhibit cancer cell proliferation, and modulate cancer-related pathways. Current research focuses on these natural and dietary compounds to develop more effective strategies for treating breast cancer. Conclusion: The findings suggested that East African medicinal plants hold promise as complementary treatments for breast cancer, offering potential benefits such as affordability, cultural appropriateness, and sustainability. Further research into these plants and their bioactive compounds could revolutionize breast cancer treatment, improving survival rates and addressing the rising incidence of breast cancer-related fatalities. Other: The review underscores the importance of continued research, conservation, and the integration of ancient healing methods to fully harness the potential of East African flora in breast cancer management.
Antimicrobial Activity, in silico Molecular Docking, ADMET and DFT Analysis of Secondary Metabolites from Roots of Three Ethiopian Medicinal Plants

Abstract:Background: Uvaria scheffleri (Annonaceae), Clematis burgensis (Ranunculaceae), and Euphorbia schimperiana (Euphorbiaceae) are medicinal plants traditionally used to treat cough, tuberculosis, asthma, sore throat and skin infections. Methods: Silica gel column chromatographic separation was used to isolate compounds. Crude extract and isolated compounds were evaluated for antimicrobial activity against Staphylococcus aureus, Escherichia coli, and Candida albicans via the broth dilution method. Docking studies were performed with E. coli DNA-Gyrase B and human DNA topoisomerase IIα by using AutoDock Vina. ADMET were predicted by SwissADME, PreADMET, and OSIRIS Property predictions. The optimized structures and molecular electrostatic potential surface of the isolated compounds were predicted by DFT analysis using B3LYP/6-31G basis levels. Results: Silica gel column chromatographic separation afforded five compounds 1-5 of which N-methyl-2,3-bis(2-hydroxybenzyl)-1Н-indol (1) is reported herein for the first time, along with known C-benzylated dihydrochalcone uvaretin (2), bis(2-ethylheptyl) phthalate (3), lupeol (4) and suberosin derivative (5). Dichloromethane roots extract of U. scheffleri showed potent antibacterial activity against S. aureus (MIC = 6.25 µg/mL) compared to gentamicin (MIC=5 µg/mL). In silico, molecular docking analysis of compounds (1and 3-5) showed strong interaction with E. coli DNA gyrase B with a binding energy value ranging from -6.9 to -6.0 kcal/mol compared to ciprofloxacin -7.2 kcal/mol, whereas analysis against human topoisomerase IIα showed binding energy value ranging from -5.9 to -5.3 kcal/mol compared to vosaroxin (-6.2 kcal/mol). Conclusion: The results obtained suggest that N-methyl-2,3-bis(2-hydroxybenzyl)-1Н-indol (1) and coumarin (5) are potential topoisomerase II α inhibitors and might be used as anticancer agents. The ADMET studies showed the highest drug-likeness properties for studied compounds other than bis(2-ethylheptyl) phthalate (3). DFT calculations suggested that studied compounds showed the lowest gap energy and were chemically reactive, and isolated compounds may serve as potential drug candidates that corroborate with the traditional uses of studied plants.
In vitro and computational investigation of antioxidant and anticancer properties of Streptomyces coeruleofuscus SCJ extract on MDA-MB-468 triple-negative breast cancer cells

Abstract:This study aimed to explore the antioxidant potential of the ethyl acetate extract of Streptomyces coeruleofuscus SCJ strain, along with its inhibitory effects on the triple-negative human breast carcinoma cell line (MDA-MB-468). The ethyl acetate extract's total phenolic and flavonoid contents were quantified, and its antioxidant activity was investigated using DPPH (1,1-Diphenyl-2-picrylhydrazyl), ABTS (2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid), and FRAP (Ferric Reducing Antioxidant Power) assays. Furthermore, the cytotoxic effect of the organic extract from Streptomyces coeruleofuscus SCJ on MDA-MB-468 cancer cells was assessed via the crystal violet assay. In tandem, a thorough computational investigation was conducted to explore the pharmacokinetic properties of the identified components of the extract, utilizing the SwissADME and pKCSM web servers. Additionally, the molecular interactions between these components and Estrogen Receptor Beta, identified as a potential target, were probed through molecular docking studies. The results revealed that ethyl acetate extract of SCJ strain exhibited remarkable antioxidant activity, with 39.899 ± 1.56% and 35.798 ± 0.082% scavenging activities against DPPH and ABTS, respectively, at 1 mg/mL. The extract also displayed significant ferric reducing power, with a concentration of 1.087 ± 0.026 mg ascorbic acid equivalents per mg of dry extract. Furthermore, a strong positive correlation ( p < 0.0001) between the antioxidant activity, the polyphenol and the flavonoid contents. Regarding anticancer activity, the SCJ strain extract demonstrated significant anticancer activity against TNBC MDA-MB-468 cancer cells, with an inhibition percentage of 62.76 ± 0.62%, 62.67 ± 0.93%, and 58.07 ± 4.82% at 25, 50, and 100 μg/mL of the extract, respectively. The HPLC-UV/vis analysis revealed nine phenolic compounds: gallic acid, sinapic acid, p- coumaric acid, cinnamic acid, trans-fereulic acid, syringic acid, chloroqenic acid, ellagic acid, epicatechin. Streptomyces coeruleofuscus SCJ showed promise for drug discovery, exhibiting antioxidant and anticancer effects.

Abstract:Cancer is one of the leading causes of death globally. Conventional drugs are expensive and have been reported to have side effects. This directs efforts in cancer research to search for inexpensive solutions with less or no side effects. This study aimed at screening for phytochemicals and the antiproliferative effects of Sclerocarya birrea fruit exocarp (SBFE) and Terminalia prunioides pods extracts (TPPE) on human cervical cancer cell line (HeLa). Extracts were qualitatively evaluated for their phytochemicals using HPLC and LC-MS/MS, and the antiproliferative effects by 4-[-3(4-Iodophenyl)-2-(4-nitro-phenyl)-2H-5-tetrazolio]-1,3-benzene sulfonate (WST-1) assay. HPLC and LC-MS/MS analysis led to identification and quantification of 24 polyphenolic metabolites amongst which shikimic acid (3.6014 mg g -1 ), gallic acid (40.8283 mg g -1 ), and quercetin-3-d-xyloside (3.4677 mg g -1 ) as the major metabolites. Results from antiproliferative effects of extracts were used to make 3 potential anticancer formulations. Furthermore, effects of extracts and formulations on the expressions of cervical cancer markers: Epidermal Growth Factor Receptor (EGFR), Metastasis-Associated in Colon Cancer 1 (MACC1), Vascular Endothelial Growth Factor (VEGF), Cytokeratin Fragment (CYFRA 21-1), and Cluster differentiation 95 (CD95) were evaluated by Reverse transcription quantitative Polymerase Chain Reaction (RT-qPCR). Methanol and ethyl acetate extracts of both plants tested positive for saponins, tannins, flavonoids, phenols, terpenoids, cardiac glycoside and steroids. Methanol extracts were the most effective with IC 50 values of 75 μg/mL and 190 μg/mL for SBFE and TPPE respectively. The formulations: M1E1M2E2 and M1M2 had IC 50 values of 77 μg/mL and 83 μg/mL respectively. All treatments downregulated mRNA expression of EGFR, VEGF, MACC1, CYFRA 21-1, and upregulated CD 95 mRNA expression. Formulations were more effective than individual extracts against HeLa cells. However, there is need for further testing for other possible mechanisms of action and isolation of phytocompounds.
