Abstract:The slaughterhouse is very useful in monitoring the health status of livestock, the profitability of their breeding and the effectiveness of therapeutic and/or prophylactic strategies. Over the years, a number of methods have been developed to quantify lesions - especially those effecting the respiratory tract, observed in slaughtered animals. Among these is the "Slaughterhouse Pleurisy Evaluation System" (SPES), which is widely used to score pleural lesions caused by Actinobacillus pleuropneumoniae. The aim of the present study is to develop and assess an alternative method to score pleurisy in slaughtered pigs, based on the inspection of the parietal pleura. This method has been compared with the SPES grid, which is considered as the "gold standard" in this field of study. Preliminary data indicate that the two methods provide almost overlapping results, showing very high correlation coefficients. Scoring pleurisy on the parietal pleura proved to be fast and easy, it could represent a valuable alternative to the SPES method.

Pathological analysis and etiological assessment of pulmonary lesions and its association with pleurisy in slaughtered pigs

Abstract:The intensification of pig farming has posed significant challenges in managing and preventing sanitary problems, particularly diseases of the respiratory complex. Monitoring at slaughter is an important control tool and cannot be overstated. Hence, this study aimed at characterizing both macroscopical and microscopical lesions and identifying the Actinobacillus pleuropneumoniae (APP), Mycoplasma hyopneumoniae (Mhyo), and Pasteurella multocida (PM) associated with pleurisy in swine. For this, a selected slaughterhouse in São Paulo State underwent a thorough examination of carcasses on the slaughter line, followed by lung sampling. The carcasses and lungs underwent macroscopical examination and were classified according to the score of pleurisy and lung samples were allocated into five groups, being: G0: score 0 - no lesions; G1: score 1; G2: score 2; G3: score 3; and G4: score 4. In total, 217 lung fragments were collected, for the histopathological evaluation and detection of the following respiratory pathogens: APP, Mhyo, and PM by qPCR. The results demonstrated that Mhyo and APP were the most prevalent etiological agents (single and co-identification) in lung samples, in different scores of pleurisies, while bronchopneumonia and bronchus-associated lymphoid tissue (BALT) hyperplasia lesions were the most frequent histopathological findings. Positive correlations were found between the quantification of APP DNA with 1) the score of pleurisy (R=0.254); 2) with the score of lung consolidation in all lung lobes (R=0.181 to R=0.329); and 3) with the score of lung consolidation in the entire lung (R=0.389). The study brings relevant information regarding the main bacterial pathogens associated with pleurisy in pigs and helps with understanding the relationship between the abovementioned pathogens and their impact on the respiratory health of pigs.
Thyroid hormones modulate serum leptin levels: observations in thyrotoxic and hypothyroid women.

Abstract:Thyroid hormones and leptin are both involved in the regulation of energy metabolism. Serum leptin concentrations were measured in women with thyrotoxicosis (n = 21, mean age 45 years) or hypothyroidism (n = 14, mean age 44 years) before and 3 months after restoration of the euthyroid state. Serum leptin concentration tended to increase in both hypothyroid (14.7+/-3.5 vs 17.8+/-3.9 ng/ml, p = 0.06) and thyrotoxic (11.9+/-1.7 vs 14.4+/-2.0, p = 0.08) women after treatment (values given as mean +/- SE in the untreated and the euthyroid state respectively). Body mass index (BMI) was lower in thyrotoxic women than in hypothyroid women in the untreated state (22.1+/-0.7 vs. 26.2+/-1.9, p < 0.05). BMI was not different between both groups after treatment (24.5+/-0.7 vs. 26.3+/-2.1, p = 0.37), due to an increase of BMI in the thyrotoxic women; BMI did not change in the hypothyroid group. After controlling for BMI in a multivariate regression analysis, serum leptin concentrations were lower in hypothyroid women than in thyrotoxic women (p < 0.05), whereas posttreatment values of leptin did not differ (p = 0.44). When leptin concentrations were expressed as standard deviation scores (Z-scores) from the mean value of female controls matched for BMI and age as reported earlier, Z-scores were lower in the hypothyroid than in the thyrotoxic women (-0.63+/-0.21 vs. 0.53+/-0.18, p = 0.001). After treatment, Z-scores did not deviate from the expected values (0.05+/-0.28 vs. 0.08+/-0.16, p = 0.98). Z-scores differed before and after treatment in both hypothyroid (p = 0.01) and thyrotoxic (p = 0.02) patients. In conclusion, these data obtained in thyrotoxic and hypothyroid women indicate that thyroid states modulates serum leptin concentrations independent of BMI, with a small decrease in hypothyroidism and a small increase in thyrotoxicosis.
Prevalence of carcass lesions and their effects on welfare, carcass composition and meat quality in slaughtered pigs

Abstract:Abstract The aims of this study were to determine the prevalence of carcass lesions in slaughtered pigs and to quantify their relationships with different animal characteristics, pre-slaughter factors, blood measurements, performance indices, carcass composition and meat quality traits. Data was recorded for 30 journeys referring to 1080 market-weight pigs that originated from 15 commercial small-scale finishing farms. Carcass lesions were visually assessed on the slaughterline in different parts of the carcass, i.e., anterior, middle and posterior, using a three-point scale. Complete blood count was investigated. The following performance indices and carcass composition traits were measured: average lifetime daily weight gain, live, hot and cold carcass weights, cooler shrinkage, dressing percentage, backfat thickness and meatiness. Meat pH and temperature were measured 45 minutes postmortem. Of the 1080 pigs slaughtered in 30 batches, 70.28% displayed some degree of lesions on the carcass (moderate – 30.00%; severe – 40.28%). The carcass lesions were the most prevalent (50.20%) in the posterior part of the pig carcass. RYR1 genotype, live weight, loading density, lairage time, lairage density and slaughter season affected the carcass lesion prevalence. The presence of carcass lesions, irrespective of severity, was associated with alterations in blood measurements in slaughtered pigs, indicating compromised animal welfare. The presence of severe carcass lesions in slaughtered pigs was significantly associated with increased meat pH45min, which led to the highest occurrence of dark, firm and dry pork. In contrast, there was strong evidence of association between the presence of moderate carcass lesions in slaughtered pigs and both decreased meat pH45min and increased meat T45min, which led to the highest occurrence of pale, soft and exudative pork among the carcass lesion groups. In conclusion, this study showed a high prevalence of carcass lesions in slaughtered pigs, whereby the risk of their occurrence was affected by both animal characteristics and pre-slaughter conditions. Also, the presence of carcass lesions in slaughtered pigs, irrespective of severity, was significantly associated with alterations in the blood measurements and pork quality.

Abstract:Bovine respiratory disease (BRD) is still a serious concern in feedlots, where it exerts a negative effect on farm productivity. There is a shortage of studies focused on the evaluation of BRD-associated lesions at the slaughterhouse in clinically healthy animals. The objective of this work was to investigate the prevalence and type of subclinical pneumonic lesions in slaughtered beef cattle, according to the age range and management system, and its impact on carcass weight. A total of 1101 beef cattle intended for human consumption were examined at slaughter. Information on age, sex, management system and carcass weight was recorded. The presence and type of pneumonia were evaluated according to gross and microscopic findings and etiological agents by PCR. Lung pneumonic lesions appeared in 17.9% of animals and were predominant among veal calves. According to the type, chronic catarrhal pneumonia prevailed in the majority of animals, and mixed and extensively reared cattle were more likely to suffer acute fibrinous pneumonia. The presence of pneumonic lesions was associated with a significant decrease in carcass weight that had more of an impact in veal male calves coming from intensive systems. Bacterial infections were the predominant infectious agent and the only cause of acute fibrinous pneumonia, while viruses were infrequent and only found in lesions with chronic catarrhal pneumonia. This study shows the importance of BRD in beef feedlots upon production values and points out the feasibility of slaughterhouse assessment of pneumonia as a method for the evaluation of BRD significance.

Abstract:Background: Severity of lung lesions quantified by thoracic ultrasonography (TUS) at time of bronchopneumonia (BP) diagnosis predicted death among steers not treated for this condition. Further research is needed to confirm that lung lesions detected by TUS can be associated with negative outcomes in cattle with BP that subsequently were treated. Objective: To quantify the effects on relapse rate and average daily gain (ADG) of lung lesions detected by TUS at first BP diagnosis in feedlot cattle. Animals: Prospective cohort of mixed beef-breed steers (n = 93; 243 ± 36 kg) and heifers (n = 51; 227 ± 42 kg) with BP at 4 feedlots. Methods: Thoracic ultrasonography was performed by the same clinician and 16-second TUS videos were evaluated offline for maximal depth and area of lung consolidation, maximum number of comet tails, and maximal depth of pleural fluid. Individual ADG was calculated between 1 and 120 days after arrival. Effects of lesions on relapse rate and ADG were investigated using mixed regression models. Results: Maximal depth of lung consolidation was associated with a higher risk of relapse (odds ratio [OR], 1.337/cm; 95% confidence interval [CI], 1.042-1.714) and lower ADG (- 34 g/cm; -64 to -4). Maximal area of lung consolidation also was associated with a higher relapse risk (OR, 1.052/cm2 ; 1.009-1.097) but not with ADG. Comet tails and pleural fluid were not associated with risk of relapse or ADG. Conclusions and clinical importance: Quantifying maximal depth and area of lung consolidation by TUS at first BP diagnosis can provide useful prognostic information in feedlot cattle.

Abstract:Medical records of 23 dogs with histologically documented orbital neoplasia and admitted to the New York State College of Veterinary Medicine between 1975 and 1984 were reviewed. Almost all (91%) of the tumors were classified as malignant; 74% of the tumors arose as primary neoplasms within the orbit. Eleven tumor types of connective tissue, bone, epithelial, and hemolymphatic origin were represented. The typically afflicted dog was purebred, female, and middle-aged. Review of this series confirmed the clinical impression that orbital neoplasms in dogs are aggressive malignancies with poor long-term prognosis.

Abstract:Skin lesions are iceberg indicators of animal welfare. The study of the bruises in pig carcasses allows the inference of how and when a traumatic moment that resulted in the injury occurred, as well as its severity. A cross-sectional study to investigate the association between pre-slaughter factors and the type of skin lesions in cross-breed fattening domestic pigs was performed on 300 pigs slaughtered in a slaughterhouse in Northern Portugal. Data on the sex of the animals, animals? origin, transport time and lairage time were collected. Skin lesions were evaluated according to location (region) on the carcass, shape, size, number of lesions per region and lesion age (colour). Descriptive, univariate and logistic regression analyses were performed (considering p?0.01). A total of 468 injuries were recorded, distributed by different carcass regions. Most of these injuries occurred on the back (37.2%) and temporally close to slaughter (94.2%). Castrated males were more predisposed to present bruises probably related to fights between animals. Females and animals subjected to longer transport times were more likely to have skin lesion injuries resulting from poor handling. The study highlights the importance of regular monitoring of welfare indicators in a pre-slaughter context for more efficient surveillance of animal welfare.

Detection and quantification by molecular techniques of early infection by Lawsonia intracellularis in suckling piglets

Abstract:Background: Lawsonia intracellularis is the causative agent of Porcine Proliferative Enteropathy (PPE), one of the most prevalent pig enteric diseases worldwide, but with sparse information about early infections in suckling piglets in the epidemiology of PPE. With that aim, this study evaluates the prevalence of L. intracellularis in 3-week-old piglets by analysing ileal digesta content and mucosal scrapings from 383 pigs from 16 farms (aprox., 25 pigs/batch) by real-time qPCR and droplet digital PCR (ddPCR). Results: Forty-nine samples yielded a qPCR positive result. Eleven samples from eight farms were confirmed as positive with concentrations of L. intracellularis from 3.5 log10 to 4.5 log10 bacteria/g of sample. Another 16 samples, eight farms, were classified as low positive (2.07-2.38 log10 bacteria/g) and 22 provided an uncertain result. Finally, 334 samples tested negative for L. intracellularis. At batch level, half of the farms included in the study had at least one positive sample and in 10 farms (62.5%) there was at least one low positive sample. The ddPCR was run in 50 of the 383 samples based on their PCR output (including low positive, uncertain and negative samples). Correlation analyses revealed a strong association between qPCR and the ddPCR results (ρ = 0.75; p < 0.001). The ddPCR allowed us to detect and confirm a positive result in the 19 samples classified as uncertain by the qPCR and detect L. intracellularis in 8 of 15 negatives by qPCR. Conclusions: The results of the study demonstrate that a number of piglets are already infected with L. intracellularis during the suckling period evidencing early infection in certain animals, adding information of PPE epidemiology and opening new research topics such as sow-piglet transmission. Study results also evidence the usefulness of a combination of qPCR and ddPCR to improve qPCR sensitivity but assuring high specificity.

Abstract:Lean Meat Percentage (LMP) is a key index for assessing pork carcass quality as well as determining carcass classification and price. This framework offers to farmers the incentive to achieve the desired organoleptic features; therefore LMP must be measured objectively online, by means of various measuring instruments. The aim of this work was to assess two different methods for predicting the lean meat percentage of pig carcasses in Greece in comparison to the European reference method. For this reason, the same 130 pig carcasses (65 males and 65 females) were measured using 2 prediction devices (Optiscan-TP and Optigrade-MCP) and subsequently were assessed according to European reference method. A formula corresponding to each device was constructed using the ordinary least squares (OLS)method and excluding the same outliers. Calculated RMSEP for all 130 carcasses was 2.46518 for Optiscan-TP and 2.48489 for Optigrade-MCP. It can be concluded that both methods perform entirely according to EU legislation (Commission Regulation (EC) 1249/2008, European Union, 2008), and results obtained are similar regardless of different measurement points and technologies.

A flexible tool for the assessment of the economic cost of pig disease in growers and finishers at farm level

Abstract:Pigmeat is the most consumed red meat globally and consumption is expected to continue to increase. The sector is faced by the risk of epidemic and endemic disease impacts and other adverse influences. The aim of this study was to develop a dynamic simulation model of pig growing and finishing that can be used to model the financial and economic impacts of a variety of scenarios both related to disease effects and other influences on production. The model consists of a physical performance module and financial performance module. The core of the physical performance module comprises three stocks to model the flow of pigs from purchase to slaughter. Mortality rates, daily live weight gain and feed conversion ratios influence the dynamics of the physical performance. Since contracts between farmers and slaughterhouses often include large price penalties for over- and underweight pigs, carcase weight distribution is an important determinant of revenues. The physical performance module, therefore, simulates slaughter weight variations. The financial performance module calculates revenue, costs and gross margins. The revenue calculations take into account price penalties for over- and underweight pigs. To demonstrate the capabilities of the model, we apply the model to assess the economic consequences of production impacts associated with respiratory disease. We use estimated production impacts associated with respiratory disease from a study of all-in-all out growing and finishing systems based on pig production data and information from slaughterhouse monitoring in the UK. Our model suggests a reduction in the gross margin of nearly 40 % as a consequence of the estimated production impacts associated with a 10% increase in respiratory disease prevalence. Due to the lack of reliable information on slaughter weight variation, we also simulate the model using different assumptions about the slaughter weight distribution. An increase in the standard deviation of carcase weights from 8 kg to 12 kg, holding average weights constant, more than halves gross margins under our scenarios. We suggest that for all-in-all-out systems, carcase weight variation is likely to be a substantial factor in reducing income in the presence of respiratory disease and the economic impact of respiratory disease may be underestimated if the effects of disease on variation in carcase weights are not included in any analysis.
Improvement of accuracy in the estimation of lean meat content in pig carcasses

Abstract:Fat thickness including skin and muscle depth measured on the left carcass side between the second and third from the last rib 70 mm off the dorsal midline were measured in a total of 168 pig hybrid carcasses. The lean meat content was then determined on the basis of simplified dissections of the carcasses. Multiple regressions of the measurements of the fat and muscle thickness on the lean meat content obtained by dissections were used to construct the following basic regression formulae for the ultrasound and probe apparatuses: γ IS-D-05 = 60.69798 – 0.89211S IS-D-05 + 0.10560M IS-D-05 and γ IS-D-15 = 60.92452 – 0.77248S IS-D-15 + 0.11329M IS-D-15 , respectively. To increase the accuracy of the prediction formulae, additional measures were included in the calculation which reduced s e by 0.48 to 0.54 percent points. The relationships between the lean meat content and other indicators of carcass value were also assessed. The highest correlation coefficient was determined in the ratio of the fat cover area above the musculus longissimus lumborum et thoracis (MLLT) to the MLLT area ( r = –0.87). On the contrary, the lean meat content demonstrated the lowest correlation with the cold carcass weight ( r = –0.25). Major carcass cuts (ham, loin, shoulder, belly with bones) from the carcasses classified in different SEUROP classes were evaluated. Significant differences between the classes were found in the proportions of cuts without fat cover, fat thickness measured at point “P 2 ”, and fat thickness measured on the midline plane separating the left and right sides of the carcass.

Bayesian evaluation of the accuracy of a thoracic auscultation scoring system in dairy calves with bronchopneumonia using a standard lung sound nomenclature

Abstract:Background: Although thoracic auscultation (AUSC) in calves is quick and easy to perform, the definition of lung sounds is highly variable and leads to poor to moderate accuracy in diagnosing bronchopneumonia (BP). Hypothesis/objectives: Evaluate the diagnostic accuracy of an AUSC scoring system based on a standard lung sound nomenclature at different cut-off values, accounting for the absence of a gold standard test for BP diagnosis. Animals: Three hundred thirty-one calves. Methods: We considered the following pathological lung sounds: increased breath sounds (score 1), wheezes and crackles (score 2), increased bronchial sounds (score 3), and pleural friction rubs (score 4). Thoracic auscultation was categorized as AUSC1 (positive calves for scores ≥1), AUSC2 (positive calves for scores ≥2), and AUSC3 (positive calves for scores ≥3). The accuracy of AUSC categorizations was determined using 3 imperfect diagnostic tests with a Bayesian latent class model and sensitivity analysis (informative vs weakly informative vs noninformative priors and with vs without covariance between ultrasound and clinical scoring). Results: Based on the priors used, the sensitivity (95% Bayesian confidence interval [BCI]) of AUSC1 ranged from 0.89 (0.80-0.97) to 0.95 (0.86-0.99), with a specificity (95% BCI) of 0.54 (0.45-0.71) to 0.60 (0.47-0.94). Removing increased breath sounds from the categorizations resulted in increased specificity (ranging between 0.97 [0.93-0.99] and 0.98 [0.94-0.99] for AUSC3) at the cost of decreased sensitivity (0.66 [0.54-0.78] to 0.81 [0.65-0.97]). Conclusions and clinical importance: A standardized definition of lung sounds improved AUSC accuracy for BP diagnosis in calves.
Simplified assessment of castration-induced pain in pigs using lower complexity algorithms

Abstract:Pigs are raised on a global scale for commercial or research purposes and often experience pain as a by product of management practices and procedures performed. Therefore, ensuring pain can be effectively identified and monitored in these settings is critical to ensure appropriate pig welfare. The Unesp-Botucatu Pig Composite Acute Pain Scale (UPAPS) was validated to diagnose pain in pre-weaned and weaned pigs using a combination of six behavioral items. To date, statistical weighting of supervised and unsupervised algorithms was not compared in ranking pain-altered behaviors in swine has not been performed. Therefore, the aim of this study was to verify if supervised and unsupervised algorithms with different levels of complexity can improve UPAPS pain diagnosis in pigs undergoing castration. The predictive capacity of the algorithms was evaluated by the area under the curve (AUC). Lower complexity algorithms containing fewer pain-altered behaviors had similar AUC (90.1-90.6) than algorithms containing five (89.18-91.24) and UPAPS (90.58). In conclusion, utilizing a short version of the UPAPS did not influence the predictive capacity of the scale, and therefore it may be easier to apply and be implemented consistently to monitor pain in commercial and experimental settings.

Abstract:Information on carcass quality obtained from dissection and conventional carcass evaluation was studied on the Landrace and Yorkshire breeds. Progeny testing pigs (n = 196) were slaughtered at a weight of c. 90 kg. After conventional carcass evaluation, the carcass half was dissected. The objects of study were the most valuable parts of the half carcass (= ham + carré + back + fore back + shoulder + kidney fat) and its skin+fat and meat+bone components. The possibility of restricting the dissection to the ham and the back was also examined. By means of the least squares methods the following results were obtained: The effect of the slaughter weight on the skin+fat, the meat+bone and the valuable part was very significant. The variation due to age was not significant. The carcass quality on the gilts was better than that of the castrates. By stepwise multiple regression procedures estimations were derived for the skin+fat component, the meat+bone component and the most valuable part of the carcass. By dissection of the ham and the back more information was generally obtained about the slaughter quality of the most valuable part than was obtained by the conventional carcass evaluation. The index (= A/B x A/C x10) was calculated, in which A = weight of the meat+bone component, B = age in days and C = weight of the half carcass. The index correlated with the skin+fat component and its percentage as follows: r = —0.34**– –0.55***, and with the meat+bone component and its percentage: r = 0.77*** —0.78***. Possibilities of developing the index were examined.

Abstract:With the development of lung diseases and the wide application of lung ultrasound (LUS), the independent analysis of various indicators in LUS images are of great significance in clinic. In this paper, we proposed a quantitative and qualitative method for extracting, analyzing, and scoring pleural lines with different lesions in LUS images. The extraction module was composed of the customized cascaded localization and segmentation models based on convolution and multilayer perceptron. The analysis module used eight textural and three morphological parameters to quantitatively analyze the features of two different output images from the localization and segmentation models, respectively. The scoring module adopted four supervised machine learning classifiers including support vector machine, k-nearest neighbor, random forest, and decision tree to qualitatively evaluate pleural lines with different severities in LUS images. The experiments were performed on the 5390 LUS images (i.e., segmentation: 1620, scoring: 3770) acquired from Coronavirus Disease 2019 pneumonia patients with convex ultrasound probes. Experimental results showed that the proposed line extraction method can accurately achieve the localization and segmentation of pleural lines. The support vector machine classifier with combining texture and morphological features as input achieved optimal scoring performance with the accuracy, sensitivity, specificity, F1 score, and AUC being 94.47%, 97.31%, 94.50%, 0.9457, and 0.9822, respectively. Compared with other models, it also proves that the proposed method has better scoring performance. Thus, the proposed method has great application potential for clinical application.

Abstract:The development and eventual fate of unimplanted fertilized ova in rats receiving pregnancy-inhibiting doses of luteinizing hormone-releasing hormone (LRH) were studied during the first 14 days of gestation. The results suggest that LRH accelerates passage of ova through the oviducts. Also, the zona pellucida of treated animals is retained by the majority of ova until day 7; zona-encased ova were observed as late as day 10. Elimination of the ova appears to occur by expulsion with uterine fluids through the vagina upon the animals' return to estrus.

Abstract:BackgroundIn today’s porcine industry, lameness has a major welfare and economic impact, and is often caused by osteochondrosis (OC). The etiological factors of the disease have been studied in depth, however, to this day, little is known about the natural course of the disorder and how it can be detected at an early stage in pigs. The aim of this pilot study was to assess the potential of three non-invasive techniques for the detection and monitoring of early OC processes in piglets. A group of weaned piglets (n = 19) were examined longitudinally using radiographs, a visual lameness scoring scheme and a quantitative pressure-mat based locomotion analysis system to detect OC in the humeroradial, femoropatellar and tarsocrural joints. At several time points, a selection of animals was euthanized for post-mortem examinations, including histology, which was the gold standard.ResultsIn this study, clear signs of subclinical signs of OC were observed, however, we were unsuccessful in producing clinical OC. Lesions were observed to be commonly bilaterally symmetric in the joints examined in 80 % of cases. The radiographic examinations showed a clear correlation with the gold standard, particularly when subclinical lesions were of a high histological score. Moreover, radiography was also able to detect the early repair processes, which appeared to take place at least until 14 weeks of age. Both visual scoring and pressure mat analyses showed good intra-assay reproducibility, with the pressure mat showing intra-class correlation values between 0.44 and 0.6 and the inter-observer agreement of visual scoring method was between 88 and 96 %, however their correlation to OC lesions detected by histology was very weak, with only 2 out of 12 traits for the visual scoring method showing significant and biologically logical relations to a specific joint having histological OC lesions. For the pressure mat, only a maximum of 5 associations for specific joints with histological OC lesions were found out of a possible 8.ConclusionAll tested in-vivo methods showed good reproducibility. Radiography was the most reliable technique to detect and monitor longitudinally the earliest signs of OC in these piglets. It also demonstrated that the “Point of No Return” (PNR) of the disease, when repair processes end, might be later than anticipated, after 13 weeks of age. All in all, our study shows that the timing of the use of these in-vivo methods is critical to detect and monitor OC, especially in the early phases of the disease. It also shows the difficulty in producing OC regardless of the optimization of the experimental settings in relation to the etiological factors known to induce OC.
