Abstract:BACKGROUND: Halophytes such as Salicornia europaea have evolved to exhibit unique mechanisms controlled by complex networks and regulated by numerous genes and interactions to adapt to habitats with high salinity. However, these mechanisms remain unknown. METHODS: To investigate the mechanism by which halophytes tolerate salt based on changes in the whole transcriptome, we performed transcriptome sequencing and functional annotation by database search. Using the unigene database, we conducted digital gene expression analysis of S. europaea at various time points after these materials were treated with NaCl. We also quantified ion uptakes. Gene functional enrichment analysis was performed to determine the important pathways involved in this process. RESULTS: A total of 57,151 unigenes with lengths of >300 bp were assembled, in which 57.5% of these unigenes were functionally annotated. Differentially expressed genes indicated that cell wall metabolism and lignin biosynthetic pathways were significantly enriched in S. europaea to promote the development of the xylem under saline conditions. This result is consistent with the increase in sodium uptake as ions pass through the xylem. Given that PSII efficiency remained unaltered, salt treatment activated the expression of electron transfer-related genes encoded by the chloroplast chromosome. Chlorophyll biosynthesis was also inhibited, indicating the energy-efficient state of the electron transfer system of S. europaea. CONCLUSIONS: The key function of adjusting important primary metabolic pathways in salt adaption was identified by analyzing the changes in the transcriptome of S. europaea. These pathways could involve unique salt tolerance mechanisms in halophytes. This study also provided information as the basis of future investigations on salt response genes in S. europaea. Ample gene resources were also provided to improve the genes responsible for the salt tolerance ability of crops.

Abstract:Sesuvium portulacastrum, a promising facultative halophyte species, has a remarkable ability to survival at severe salt of ~1000 mM NaCl. However, genes encoding proteins involved in the process of salt tolerance in S. portulacastrum are limited. In this study, suppression subtractive hybridization together with reverse northern blot analysis and RT-PCR was used to identify up-regulated genes in roots after treatment with seawater for 2–24 h. We identified a total of 148 unigenes, and most of these genes were predominantly associated with ‘signal transduction and transcription’, ‘stress/defense response’, ‘transport’ and ‘metabolism’. Comparative analysis between physiological response and differentially expressed genes highlighted that sodium (Na+) transport, compartmentation, reactive oxygen species (ROS) scavenging, and sugar-metabolizing regulation play important roles in salt tolerance of S. portulacastrum, whereas inhibition of root elongation induced by high salinity may be related to lignin deposition and disruption of calcium homeostasis.
Coordination of Carbon Fixation and Nitrogen Metabolism in Salicornia Europaea under Salinity: Comparative Proteomic Analysis on Chloroplast Proteins

Abstract:Halophyte, like Salicornia europaea, could make full use of marginal saline land for carbon fixation. How the photosynthesis of S. europaea is regulated under high salinity implicates a significant aspect to exploit this pioneer plant in future. Measurement of photosynthesis parameters demonstrated the reduction of photosynthesis for the 0 and 800mM NaCl treated plants are more likely due to non-stomatal limitation, which might be caused by changes in the enzymes associated with photosynthesis. Different salinity induced ultrastructure changes other than photosynthetic apparatus damage, suggesting the photosynthesis of S. europaea might be affected via biochemical regulation. Comparative proteomics analysis of chloroplast proteins by 2-D gel electrophoresis reproducibly detected 90 differentially expressed proteins, among which 66 proteins were identified by nanoLC MS/MS. Further study of thylakoid membrane proteins by Blue-Native PAGE proved the increase in abundance of light reaction proteins under salinity. Analysis of gene expression patterns of 12 selected proteins provides evidence for the correlations between transcription and proteomics data. Based on our results, a putative model of photosynthesis regulatory network figured out proper coordination of carbon fixation and nitrogen metabolism in chloroplast of S. europaea under salinity, which provided subcellular level insight into salt tolerance mechanism in S. europaea.
Response of Purslane Plants Grown under Salinity Stress and Biostimulant Formulations

Abstract:Purslane has been suggested as an alternative crop suitable for human consumption due to its high content of minerals, omega-3 fatty acids, and several health-beneficial compounds. In this study, we aimed to evaluate the effect of salinity stress (tap water (control), 2000, 4000, 6000, 8000, and 10,000 mg L−1), biostimulant application (putrescine and salicylic acid at 200 mg L−1), and the combination of the tested factors (i.e., salinity × biostimulant application) on the growth and chemical composition of purslane plants (Portulaca oleracea L.) over two growing seasons (2022 and 2023). Irrigation with tap water and putrescine application resulted in the highest plant height, weight of aboveground and underground parts, and number of shoots per plant. In contrast, the lowest values of growing parameters were recorded under severe saline stress (10,000 mg L−1), especially for the plants that were not treated with biostimulants. The same trends were observed for macronutrients (N, P, K), total carbohydrates, total chlorophylls, and vitamin C content in leaves. Moreover, nitrate and proline content was higher in plants grown under salinity stress, especially under severe stress (8000–10,000 mg L−1) without biostimulant application. In general, the application of biostimulants mitigated the negative impact of salinity on plant growth and leaf chemical composition, while the effect of putrescine on the tested parameters was more beneficial than that of salicylic acid. In conclusion, this study provides useful information regarding the use of putrescine and salicylic acid as biostimulatory agents with the aim of increasing purslane growth under salinity conditions.

Abstract:Sesuvium portulacastrum L. is a flowering succulent halophyte in the ice plant family Aizoaceae. There are various ecotypes distributed in sandy coastlines and salty marshlands in tropical and subtropical regions with the common name of sea purslane. These plants are tolerant to salt, drought, and flooding stresses and have been used for the stabilization of sand dunes and the restoration of coastal areas. With the increased salinization of agricultural soils and the widespread pollution of toxic metals in the environment, as well as excessive nutrients in waterbodies, S. portulacastrum has been explored for the desalination of saline soils and the phytoremediation of metals from contaminated soils and nitrogen and phosphorus from eutrophic water. In addition, sea purslane has nutraceutical and pharmaceutical value. Tissue analysis indicates that many ecotypes are rich in carbohydrates, proteins, vitamins, and mineral nutrients. Native Americans in Florida eat it raw, pickled, or cooked. In the Philippines, it is known as atchara after being pickled. S. portulacastrum contains high levels of ecdysteroids, which possess antidiabetic, anticancer, and anti-inflammatory activities in mammals. In this review article, we present the botanical information, the physiological and molecular mechanisms underlying the tolerance of sea purslane to different stresses, its nutritional and pharmaceutical value, and the methods for its propagation and production in saline soils and waterbodies. Its adaptability to a wide range of stressful environments and its role in the production of valuable bioactive compounds suggest that S. portulacastrum can be produced in saline soils as a leafy vegetable and is a valuable genetic resource that can be used for the bioremediation of soil salinity and eutrophic water.

Abstract:Sesuvium portulacastrum has a strong salt tolerance and can grow in saline and alkaline coastal and inland habitats. This study investigated the physiological and molecular responses of S. portulacastrum to high salinity by analyzing the changes in plant phytohormones and antioxidant activity, including their differentially expressed genes (DEGs) under similar high-salinity conditions. High salinity significantly affected proline (Pro) and hydrogen peroxide (H 2 O 2 ) in S. portulacastrum seedlings, increasing Pro and H 2 O 2 contents by 290.56 and 83.36%, respectively, compared to the control. Antioxidant activities, including superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT), significantly increased by 83.05, 205.14, and 751.87%, respectively, under high salinity. Meanwhile, abscisic acid (ABA) and gibberellic acid (GA 3 ) contents showed the reverse trend of high salt treatment. De novo transcriptome analysis showed that 36,676 unigenes were matched, and 3,622 salt stress-induced DEGs were identified as being associated with the metabolic and biological regulation processes of antioxidant activity and plant phytohormones. POD and SOD were upregulated under high-salinity conditions. In addition, the transcription levels of genes involved in auxin ( SAURs and GH3 ), ethylene ( ERF1 , ERF3 , ERF114 , and ABR1 ), ABA ( PP2C ), and GA 3 ( PIF3 ) transport or signaling were altered. This study identified key metabolic and biological processes and putative genes involved in the high salt tolerance of S. portulacastrum and it is of great significance for identifying new salt-tolerant genes to promote ecological restoration of the coastal strand.

Foliar Application of Cerium Oxide-Salicylic Acid Nanoparticles (CeO2:SA Nanoparticles) Influences the Growth and Physiological Responses of Portulaca oleracea L. under Salinity

Abstract:In the present study, the effects of foliar application of salicylic acid (100 μM), cerium oxide (50 mg L-1), and cerium oxide:salicylic acid nanoparticles (CeO2: SA-nanoparticles, 50 mg L-1 + 100 μM) on the growth and physiological responses of purslane (Portulaca oleracea L.) were examined in non-saline and saline conditions (50 and 100 mM NaCl salinity). Foliar applications mitigated salinity-induced adverse effects, and the highest plant height and N, P, Mg, and Mn content were recorded in the variant with non-saline × foliar use of CeO2: SA-nanoparticles. The highest values of fresh and dry weight were noted in the treatment with no-salinity × foliar use of CeO2:SA-nanoparticles. The highest number of sub-branches was observed in the foliar treatments with CeO2-nanoparticles and CeO2:SA-nanoparticles without salinity stress, while the lowest number was noted in the 100 mM NaCl treatment. Moreover, the foliar application of CeO2:SA-nanoparticles and cerium-oxide nanoparticles improved the total soluble solid content, K, Fe, Zn, Ca, chlorophyll a, and oil yield in the plants. The salinity of 0 and 50 mM increased the K content, 1000-seed weight, total soluble solid content, and chlorophyll b content. The use of 100 mM NaCl with no-foliar spray increased the malondialdehyde, Na, and H2O2 content and the Na+/K+ ratio. No-salinity and 50 mM NaCl × CeO2: SA-nanoparticle interactions improved the anthocyanin content in plants. The phenolic content was influenced by NaCl100 and the foliar use of CeO2:SA-nanoparticles. The study revealed that the foliar treatment with CeO2:SA-nanoparticles alleviated the side effects of salinity by improving the physiological responses and growth-related traits of purslane plants.

Abstract:Purslane ( Portulaca oleracea L.) is a popular orphan crop used for its nutritional properties in various parts of the world. It is considered one of the richest terrestrial sources of omega-3 and omega-6-fatty acids (ω-3 and 6-FAs) suggesting its importance for human health. This ethnomedicinal plant is also an important part of traditional healing systems among the indigenous people. Many studies have indicated its tolerance against multiple stresses and found that it easily grows in a range of environmental gradients. It has also been considered one of the important biosaline crops for the future. Despite its huge nutritional, economic, and medicinal importance, it remains neglected to date. Most of the studies on purslane were focused on its ethnomedicinal, phytochemical, pharmacological, and stress-tolerance properties. Only a few studies have attempted genetic dissection of the traits governing these traits. Purslane being an important traditional food crop across the globe can be valorized for a sustainable food security in the future. Therefore, this review is an attempt to highlight the distribution, domestication, and cultivation of purslane and its importance as an important stress-tolerant food and a biosaline crop. Furthermore, identification of genes and their functions governing important traits and its potential for improvement using genomics tools for smart and biosaline agriculture has been discussed.
Thriving under Salinity: Growth, Ecophysiology and Proteomic Insights into the Tolerance Mechanisms of Obligate Halophyte Suaeda fruticosa

Abstract:Studies on obligate halophytes combining eco-physiological techniques and proteomic analysis are crucial for understanding salinity tolerance mechanisms but are limited. We thus examined growth, water relations, ion homeostasis, photosynthesis, oxidative stress mitigation and proteomic responses of an obligate halophyte Suaeda fruticosa to increasing salinity under semi-hydroponic culture. Most biomass parameters increased under moderate (300 mmol L−1 of NaCl) salinity, while high (900 mmol L−1 of NaCl) salinity caused some reduction in biomass parameters. Under moderate salinity, plants showed effective osmotic adjustment with concomitant accumulation of Na+ in both roots and leaves. Accumulation of Na+ did not accompany nutrient deficiency, damage to photosynthetic machinery and oxidative damage in plants treated with 300 mmol L−1 of NaCl. Under high salinity, plants showed further decline in sap osmotic potential with higher Na+ accumulation that did not coincide with a decline in relative water content, Fv/Fm, and oxidative damage markers (H2O2 and MDA). There were 22, 54 and 7 proteins in optimal salinity and 29, 46 and 8 proteins in high salinity treatment that were up-regulated, down-regulated or exhibited no change, respectively, as compared to control plants. These data indicate that biomass reduction in S. fruticosa at high salinity might result primarily from increased energetic cost rather than ionic toxicity.

Abstract:Sesuvium portulacastrum (L.) is a facultative halophyte that grows in subtropical coastal sides of the eastern and western regions of India. In the present study, the activation of antioxidant machinery was observed at 8 and 24 h after 250 mM NaCl stress treatment. Further, GC–MS/MS based metabolomics was performed, independently in root (NR8 and NR24) and shoot (NS8 and NS24). A total of 53 and 62 metabolites were detected in roots and shoots, respectively, that mainly included sugars, amino acids, organic acids and polyols. On the basis of statistical significance, sucrose and ethanolamine were identified as differentially altered metabolites (DAMs) shared under all the treatment conditions. Heat-map based clustering analysis substantiated the role of sugars and amino acids in NR8; while, sugars in NS8/NS24 treatments. For most of the DAMs, extent of change at NR24 was significantly dampened than NR8, which also coincided with reduced correlation strength between different metabolite groups, indicating partial metabolite restoration. The enrichment analysis identified sugars, amino acid and TCA cycle metabolism as major pathways altered under salt stress conditions. A total of 30 metabolomic datasets from 7 different halophytes, including the present study, were utilized to perform meta-analysis. On the basis of cumulative pathway score (CPS) of both root and shoot, aspartate–alanine–glutamate (290.62) was identified as the most significant pathway followed by arginine biosynthesis (0.62) and glyoxylate–dicarboxylate metabolism (0.46) that significantly contributed to salt-adaptation in halophytes. Thus, the holistic analysis of salt-responsive metabolites/pathways from halophytes will pave the way for enhancing salt-tolerance in crop plants.

Abstract:Panicum antidotale , a C4 monocot, has the potential to reclaim saline and drylands and to be utilized as fodder and forage. Its adaptability to survive saline stress has been proven with eco-physiological and biochemical studies. However, little is known about its molecular mechanisms of salt tolerance. In this study, an integrated transcriptome and proteome analysis approach, based on RNA sequencing and liquid chromatography tandem mass spectrometry (LC-MS/MS), was used to identify the said mechanisms. Plants were treated with control (0 mM), low (100 mM), and high (300 mM) sodium chloride (NaCl) treatments to distinguish beneficial and toxic pathways influencing plant biomass. The results indicated differential expression of 3,179 (1,126 upregulated/2,053 downregulated) and 2,172 (898 upregulated/1,274 downregulated) genes (DEGs), and 514 (269 upregulated/245 downregulated) and 836 (494 upregulated/392 downregulated) proteins (DEPs) at 100 and 300 mM NaCl, respectively. Among these, most upregulated genes and proteins were involved in salt resistance strategies such as proline biosynthesis, the antioxidant defense system, ion homeostasis, and sugar accumulation at low salinity levels. On the other hand, the expression of several genes and proteins involved in the respiratory process were downregulated, indicating the inability of plants to meet their energy demands at high salinity levels. Moreover, the impairments in photosynthesis were also evident with the reduced expression of genes regulating the structure of photosystems and increased expression of abscisic acid (ABA) mediated pathways which limits stomatal gas exchange. Similarly, the disturbance in fatty acid metabolism and activation of essential ion transport blockers damaged the integrity of the cell membrane, which was also evident with enhanced malondialdehyde (MDA). Overall, the analysis of pathways revealed that the plant optimal performance at low salinity was related to enhanced metabolism, antioxidative defense, cell growth, and signaling pathways, whereas high salinity inhibited biomass accumulation by altered expression of numerous genes involved in carbon metabolism, signaling, transcription, and translation. The data provided the first global analysis of the mechanisms imparting salt stress tolerance of any halophyte at transcriptome and proteome levels.

Abstract:Seawater desalination by reverse osmosis is growing exponentially due to water scarcity. Byproducts of this process (e.g. brines), are generally discharged directly into the coastal ecosystem, causing detrimental effects, on benthic organisms. Understanding the cellular stress response of these organisms (biomarkers), could be crucial for establishing appropriate salinity thresholds for discharged brines. Early stress biomarkers can serve as valuable tools for monitoring the health status of brine-impacted organisms, enabling the prediction of long-term irreversible damage caused by the desalination industry. In this study, we conducted laboratory-controlled experiments to assess cellular and molecular biomarkers against brine exposure in two salinity-sensitive Mediterranean seagrasses: Posidonia oceanica and Cymodocea nodosa . Treatments involved exposure to 39, 41, and 43 psu, for 6 h and 7 days. Results indicated that photosynthetic performance remained unaffected across all treatments. However, under 43 psu, P. oceanica and C. nodosa exhibited lipid oxidative damage, which occurred earlier in P. oceanica . Additionally, P. oceanica displayed an antioxidant response at higher salinities by accumulating phenolic compounds within 6 h and ascorbate within 7 d; whereas for C. nodosa the predominant antioxidant mechanisms were phenolic compounds accumulation and total radical scavenging activity, which was evident after 7 d of brines exposure. Finally, transcriptomic analyses in P. oceanica exposed to 43 psu for 7 days revealed a poor up-regulation of genes associated with brassinosteroid response and abiotic stress response, while a high down-regulation of genes related to primary metabolism was detected. In C. nodosa , up-regulated genes were involved in DNA repair, cell cycle regulation, and reproduction, while down-regulated genes were mainly associated with photosynthesis and ribosome assembly. Overall, these findings suggest that 43 psu is a critical salinity-damage threshold for both seagrasses; and despite the moderate overexpression of several transcripts that could confer salt tolerance, genes involved in essential biological processes were severely downregulated.

Abstract:Drought and salinity are two of the most severe abiotic stresses affecting agriculture worldwide and bear some similarities regarding the responses of plants to them. The first is also known as osmotic stress and shows similarities mainly with the osmotic effect, the first phase of salinity stress. Multi-Omics Integration (MOI) offers a new opportunity for the non-trivial challenge of unraveling the mechanisms behind multigenic traits, such as drought and salinity resistance. The current study carried out a comprehensive, large-scale, single-omics analysis (SOA) and MOI studies on the leaves of young oil palm plants submitted to water deprivation. After performing SOA, 1955 DE enzymes from transcriptomics analysis, 131 DE enzymes from proteomics analysis, and 269 DE metabolites underwent MOI analysis, revealing several pathways affected by this stress, with at least one DE molecule in all three omics platforms used. Moreover, the similarities and dissimilarities in the molecular response of those plants to those two abiotic stresses underwent mapping. Cysteine and methionine metabolism (map00270) was the most affected pathway in all scenarios evaluated. The correlation analysis revealed that 91.55% of those enzymes expressed under both stresses had similar qualitative profiles, corroborating the already known fact that plant responses to drought and salinity show several similarities. At last, the results shed light on some candidate genes for engineering crop species resilient to both abiotic stresses.

Abstract:In recent years, soil salinization has become increasingly severe, and the ecological functions of saline–alkali soils have deteriorated because of the lack of plants. Therefore, understanding the tolerance mechanisms of saline–alkali-tolerant plants has become crucial to restore the ecological functions of saline–alkali soils. In this study, we evaluated the molecular mechanism underlying the tolerance of Kosteletzkya pentacarpos L. (seashore mallow) seedlings treated with 0.05 or 0.5% saline–alkali solution (NaCl: NaHCO 3 = 4:1 mass ratio) for 1 and 7 days. We identified the key genes involved in tolerance to saline–alkali stress using orthogonal partial least squares regression analysis (OPLS-RA) based on both chlorophyll fluorescence indexes and stress-responsive genes using transcriptome analysis, and, finally, validated their expression using qRT-PCR. We observed minor changes in the maximum photochemical efficiency of the stressed seedlings, whose photosynthetic performance remained stable. Moreover, compared to the control, other indicators varied more evidently on day 7 of 0.5% saline–alkali treatment, but no variations were observed in other treatments. Transcriptome analysis revealed a total of 54,601 full-length sequences, with predominantly downregulated differentially expressed gene (DEG) expression. In the high concentration treatment, the expression of 89.11 and 88.38% of DEGs was downregulated on days 1 and 7, respectively. Furthermore, nine key genes, including KpAGO4 , KpLARP1C , and KpPUB33 , were involved in negative regulatory pathways, such as siRNA-mediated DNA methylation, inhibition of 5′-terminal oligopyrimidine mRNA translation, ubiquitin/proteasome degradation, and other pathways, including programmed cell death. Finally, quantitative analysis suggested that the expression of key genes was essentially downregulated. Thus, these genes can be used in plant molecular breeding in the future to generate efficient saline–alkali–tolerant plant germplasm resources to improve the ecological functions of saline–alkali landscapes.

Abstract:Plant biostimulants are widely applied in agriculture for their ability to improve plant fitness. In the present work, the impact of Graminaceae -derived protein hydrolysate (P) and its lighter molecular fraction F3 (< 1 kDa) on lettuce plants, subjected to either no salt or high salt conditions, was investigated through the combination of metabolomics and transcriptomics. The results showed that both treatments significantly modulated the transcriptome and metabolome of plants under salinity stress, highlighting an induction of the hormonal response. Nevertheless, P and F3 also displayed several peculiarities. F3 specifically modulated the response to ethylene and MAPK signaling pathway, whereas P treatment induced a down-accumulation of secondary metabolites, albeit genes controlling the biosynthesis of osmoprotectants and antioxidants were up-regulated. Moreover, according with the auxin response modulation, P promoted cell wall biogenesis and plasticity in salt-stressed plants. Notably, our data also outlined an epigenetic control of gene expression induced by P treatment. Contrarily, experimental data are just partially in agreement when not stressed plants, treated with P or F3, were considered. Indeed, the reduced accumulation of secondary metabolites and the analyses of hormone pathways modulation would suggest a preferential allocation of resources towards growth, that is not coherent with the down-regulation of the photosynthetic machinery, the CO 2 assimilation rate and leaves biomass. In conclusion, our data demonstrate that, although they might activate different mechanisms, both the P and F3 can result in similar benefits, as far as the accumulation of protective osmolytes and the enhanced tolerance to oxidative stress are concerned. Notably, the F3 fraction exhibits slightly greater growth promotion effects under high salt conditions. Most importantly, this research further corroborates that biostimulants' mode of action is dependent on plants' physiological status and their composition, underscoring the importance of investigating the bioactivity of the different molecular components to design tailored applications for the agricultural practice.

The Mitigating Effects of Biostimulant Amendments on the Response of Purslane Plants Grown under Drought Stress Conditions

Abstract:Portulaca oleracea L. is a wild edible plant with high potential for exploitation in commercial cropping systems due to its nutritional value and great adaptability to abiotic stress conditions. The present study aimed to investigate the response of purslane plants grown under drought stress conditions (100%, 80%, and 60% of field capacity (FC)) and the implementation of biostimulant amendments (control without amendment, plant growth-promoting rhizobacteria (PGPR), mycorrhiza, and effective microorganisms (EMs)) for two consecutive years. In the two-year experiment, the greatest height was recorded in plants grown under no-stress conditions and inoculated with PGPR. The highest branch number, and fresh and dry weight of aboveground and underground parts were observed under no-stress conditions at the mycorrhiza treatment. Moreover, mycorrhiza application in plants growing under 100% FC resulted in the highest N, P, total carbohydrates, and vitamin C and the lowest nitrate and proline contents in leaves. Purslane plants grown under 100% FC and inoculated with PGPR treatment resulted in the highest K and total chlorophyll leaf contents. Additionally, growing plants under mild drought stress (80% FC) combined with biostimulant application (e.g., inoculation with mycorrhiza, PGPR, and EM) may improve plant growth characteristics and mitigate negative stress effects. In general, the applied biostimulant amendments alleviated the adverse effects of drought on plant growth and leaf chemical composition indicating the importance of sustainable strategies to achieve high yield and sufficient quality within the climate change scenario.

Abstract:Salt stress is the major abiotic stress worldwide, adversely affecting crop yield and quality. Utilizing salt tolerance genes for the genetic breeding of crops is one of the most effective measures to withstand salinization. Sophora alopecuroides is a well-known saline-alkaline and drought-tolerant medicinal plant. Understanding the underlying molecular mechanism for Sophora alopecuroides salt tolerance is crucial to identifying the salt-tolerant genes. In this study, we performed tandem mass tag (TMT) based proteomic profiling of S. alopecuroides leaves under 150 mM NaCl induced salt stress condition for 3 d and 7 d. Data are available on ProteomeXchange (PXD027627). Furthermore, the proteomic findings were validated through parallel reaction monitoring (PRM). We observed that the expression levels of several transporter proteins related to the secondary messenger signaling pathway were altered under salt stress conditions induced for 3 d. However, the expression of the certain transferase, oxidoreductase, dehydrogenase, which are involved in the biosynthesis of flavonoids, alkaloids, phenylpropanoids, and amino acid metabolism, were mainly alerted after 7 d post-salt-stress induction. Several potential genes that might be involved in salt stress conditions were identified; however, it demands further investigation. Although salt stress affects the level of secondary metabolites, their correlation needs to be investigated further. SIGNIFICANCE: Salinization is the most severe abiotic adversity, which has had a significant negative effect on world food security over the time. Excavating salt-tolerant genes from halophytes or medicinal plants is one of the important measures to cope with salt stress. S. alopecuroides is a well-known medicinal plant with anti-tumor, anti-inflammatory, and antibacterial effects, anti-saline properties, and resistance to drought stress. Currently, only a few studies have explored the S. alopecuroides' gene function, and regulation and these studies are mostly related to the unpublished genome sequence information of S. alopecuroides. Recently, transcriptomics and metabolomics studies have been carried on the abiotic stress in S. alopecuroides roots. Multiple studies have shown that altered gene expression at the transcript level and altered metabolite levels do not correspond to the altered protein levels. In this study, TMT and PRM based proteomic analyses of S. alopecuroides leaves under salt stress condition induced using 150 mM NaCl for 3 d and 7 d was performed. These analyses elucidated the activation of different mechanisms in response to salt stress. A total of 434 differentially abundant proteins (DAPs) in salt stress conditions were identified and analyzed. For the first time, this study utilized proteomics technology to dig out plentiful underlying salt-tolerant genes from the medicinal plant, S. alopecuroides. We believe that this study will be of great significance to crop genetics and breeding.

Abstract:<p>Salinity is one of the major threats to sustainable agriculture that globally decrease plant production by impairing various physiological, biochemical, and molecular function. In particular, salinity hampers germination, growth, photosynthesis, transpiration, and stomatal conductance. Salinity decreases leaf water potential and turgor pressure and generates osmotic stress. Salinity enhances reactive oxygen species (ROS) content in the plant cell as a result of ion toxicity and disturbs ion homeostasis. Thus, it imbalances nutrient uptake disintegrates membrane and various ultrastructure. Consequently, salinity leads to osmotic and ionic stress. Plants respond to salinity by modulating various morpho-physiological, anatomical, and biochemical traits by regulating ion homeostasis and compartmentalization, antioxidant machinery, and biosynthesis of osmoprotectants and phytohormones, i. e, auxins, abscisic acid, brassinosteroids, cytokinins, ethylene, gibberellins, salicylic acid, jasmonic acid, and polyamines. Thus, this further modulates plant osmoticum, decreases ion toxicity, and scavenges ROS. Plants upregulate various genes and proteins that participate in salinity tolerance. They also promote the production of various phytohormones and metabolites that mitigate the toxic effect of salinity. Based on recent papers, the deleterious effect of salinity on plant physiology is discussed. Furthermore, it evaluates the physiological and biochemical responses of the plant to salinity along with phytohormone response. This review paper also highlights omics (genomics, transcriptomics, proteomics, and metabolomics) approach to understand salt stress tolerance.</p>

Abstract:Abstract Background Salinity severely inhibit crop growth, yield, and quality worldwide. Allotetraploid rapeseed ( Brassica napus L.), a major glycophyte oil crop, is susceptible to salinity. Understanding the physiological and molecular strategies of rapeseed salinity resistance is a promising and cost-effective strategy for developing highly resistant cultivars. Results First, early leaf senescence was identified and root system growth was inhibited in rapeseed plants under severe salinity conditions. Electron microscopic analysis revealed that 200 mM NaCl induced fewer leaf trichomes and stoma, cell plasmolysis, and chloroplast degradation. Primary and secondary metabolite assays showed that salinity led to an obviously increased anthocyanin, osmoregulatory substances, abscisic acid, jasmonic acid, pectin, cellulose, reactive oxygen species, and antioxidant activity, and resulted in markedly decreased photosynthetic pigments, indoleacetic acid, cytokinin, gibberellin, and lignin. ICP-MS assisted ionomics showed that salinity significantly constrained the absorption of essential elements, including the nitrogen, phosphorus, potassium, calcium, magnesium, iron, mangnese, copper, zinc, and boron nutrients, and induced the increase in the sodium/potassium ratio. Genome-wide transcriptomics revealed that the differentially expressed genes were involved mainly in photosynthesis, stimulus response, hormone signal biosynthesis/transduction, and nutrient transport under salinity. Conclusions The high-resolution salt-responsive gene expression profiling helped the efficient characterization of central members regulating plant salinity resistance. These findings might enhance integrated comprehensive understanding of the morpho-physiologic and molecular responses to salinity and provide elite genetic resources for the genetic modification of salinity-resistant crop species.

Abstract:Purslane (Portulaca oleraceae L.) is a widespread weed, which is highly appreciated for its high nutritional value with particular reference to the content in omega-3 fatty acids. In the present study, the nutritional value and chemical composition of purslane plants in relation to plant part and harvesting stage were evaluated. Plants were harvested at three growth stages (29, 43 and 52 days after sowing (DAS)), while the edible aerial parts were separated into stems and leaves. Leaves contained higher amounts of macronutrients than stems, especially at 52 DAS. α-tocopherol was the main isoform, which increased at 52 DAS, as well total tocopherols (values were in the ranges of 197–327 μg/100 g fresh weight (fw) and 302–481 μg/100 g fw, for α-tocopherol and total tocopherols, respectively). Glucose and fructose were the main free sugars in stems and leaves, respectively, whereas stems contained higher amounts of total sugars (values were ranged between 0.83 g and 1.28 g/100 g fw). Oxalic and total organic acid content was higher in leaves, especially at the last harvesting stage (52 DAS; 8.6 g and 30.3 g/100 g fw for oxalic acid and total organic acids, respectively). Regarding the fatty acid content, stems contained mainly palmitic (20.2–21.8%) and linoleic acid (23.02–27.11%), while leaves were abundant in α-linolenic acid (35.4–54.92%). Oleracein A and C were the major oleracein derivatives in leaves, regardless of the harvesting stage (values were in the ranges of 8.2–103.0 mg and 21.2–143 mg/100 g dried weight (dw) for oleraceins A and C, respectively). Cytotoxicity assays showed no hepatotoxicity, with GI50 values being higher than 400 μg/mL for all the harvesting stages and plant parts. In conclusion, early harvesting and the separation of plant parts could increase the nutritional value of the final product through increasing the content of valuable compounds, such as omega-3 fatty acids, phenolic compounds and oleracein derivatives, while at the same time, the contents of anti-nutritional compounds such as oxalic acid are reduced.
