Abstract:Excessive fat deposition due to impaired fat metabolism in chickens is a major problem in the poultry industry. Nutritional interventions are effective solutions, but current options are limited. A safe phytochemical, rutin, has shown positive effects in animals, but its effect on lipid metabolism in poultry remains unknown. Hence, this study is to investigate the effects of rutin on egg quality, serum biochemistry, fat deposition, lipid peroxidation and hepatic lipid metabolism in post-peak laying hens. A total of 360 Taihang laying hens (49-week-old) were randomly divided into five groups and fed a basal diet (control group, 0%) and a basal diet supplemented with 300 (0.03%), 600 (0.06%), 900 (0.09%), and 1,200 (0.12%) mg rutin/kg feed, respectively. The results showed that eggshell strength was significantly (p < 0.05) higher in the dietary rutin groups, whereas yolk percentage (p < 0.05), total cholesterol (TC) (p < 0.01) and yolk fat ratio (p < 0.01) decreased linearly (p < 0.05) in the dietary rutin groups. Importantly, dietary rutin reduced serum triglyceride (TG) and TC levels, decreased abdominal lipid deposition and liver index (p < 0.05), and which concomitantly decreased hepatic lipid (TG, TC, and free fatty acid) accumulation (p < 0.05). An increase (p < 0.05) in total antioxidant capacity and superoxide dismutase activity and a decrease (p < 0.05) in malondialdehyde levels were also found. At the same time, the activities of hepatic lipase, acetyl-CoA carboxylase and malic enzyme in the liver were decreased (p < 0.05). Dietary rutin also increased (p < 0.05) the expression of fatty acid oxidation-related genes (carnitine palmitoyl transferase 1, peroxisome proliferator-activated receptor α, farnesoid X receptor). Additionally, it decreased fatty acid synthesis genes (sterol regulatory element binding protein-1c, acetyl-CoA carboxylase α, stearoyl-CoA desaturase 1) (p < 0.05). In conclusion, the addition of rutin (0.06-0.12%) to the diet improved the fat metabolism and increased liver antioxidant capacity in post-peak laying hens, and these positive changes improved egg quality to some extent.
Evaluation of Dietary Metabolizable Energy Concentrations on Meat Quality and Lipid Metabolism-Related Gene Expression in Yellow-Feathered Chickens.

Abstract:This study evaluated the effects of different dietary metabolizable energy (ME) concentrations on the meat quality, carcass traits, volatile flavour and lipid metabolism-related gene expression levels in yellow-feathered chickens. In total, 600 Huxu female chickens aged 90 days were randomly assigned to six dietary treatments, each with 10 replicates of 10 birds. During the finisher phase, the birds were fed diets containing 2880 (low), 2940, 3000, 3060, 3120 and 3180 (high) kcal ME/kg. The results showed that the average daily gain of chickens increased as the dietary ME concentration increased, while the feed to gain improved (p < 0.05), and the intramuscular fat content of breast muscle increased (p < 0.05). The energy concentration had no effect on the breast muscle pH (45 min and 24 h), colour parameter (L*) or percentage of drip loss (p > 0.05), but the shear force values decreased significantly (p < 0.05). The diameter and area of the breast muscle fiber decreased and the muscle fibre density increased as the dietary ME concentration increased (p < 0.05). The highest ME concentration (3180 kcal) increased the percentages of aldehydes (hexanal, heptanal, 2,4-nonadienal, octanal, nonanal and 2-decenal), alcohols (2-nonen-1-ol, trans-2-undecen-1-ol, 7-hexadecenal, 2-hexyl-1-decanoal and n-nonadecanol-1,3,7,11-trimethyl-1-dodecanol), alkanes (2,6-dimethyl-heptadecane) and carboxylic acids (9-hexadecenoic acid), but reduced the percentages of octadecanal, octadecane, heneicosane and tetradecanal (p < 0.05). In addition, the mRNA gene expression levels of fatty acid-binding protein 3 and apolipoprotein B were significantly upregulated in the liver, whereas that of cholesteryl ester transfer protein was significantly downregulated. In conclusion, increasing the ME diet to 3180 kcal/kg significantly improved the quality and flavour of the meat from yellow-feathered broilers. our finding may help poultry producers to improve the taste of meat by regulating genes related to lipid metabolism, thereby achieving the flavour and taste characteristics preferred by consumers.
Effects of Dietary Perilla Seed Oil Supplementation on Lipid Metabolism, Meat Quality, and Fatty Acid Profiles in Yellow-feathered Chickens

Abstract:This study evaluated the effect of the dietary replacement of 1% lard (CT) with 1% perilla oil (PO), 0.9% perilla oil + 0.1% anise oil (PA), or 0.9% perilla oil + 0.1% ginger oil (PG) on indices of lipid metabolism, antioxidant capacity, meat quality, and fatty acid profiles from Yellow-feathered chickens at day 63. Compared with the CT chickens, those given perilla oil had decreased (P < 0.05) plasma lipid levels including triglycerides (TG), total cholesterol (TCH), and low-density lipoprotein cholesterol (LDL-C). Hepatic TG, TCH levels, and fatty acid synthase activity were also decreased (P < 0.05) in chickens fed diets containing perilla oil. Abdominal fat percentage was significantly decreased in birds fed the PG compared to CT diets. Birds fed the PA or PG diets had increased (P < 0.05) hepatic total SOD, glutathione peroxidase, and glutathione-S-transferase than in chickens given PO alone. In addition, the content of reduced glutathione (GSH) in breast muscle was lower (P < 0.05) in birds fed PO compared with those given PG, and the reverse was true for content of malondialdehyde. Compared with the CT diet, the PO diet decreased breast muscle shear values and increased yellowness (b*) of breast muscle (P < 0.05). Birds fed the PA or PG diets had meat with better overall acceptability than those fed the CT diet. Chickens fed perilla oil diets exhibited higher contents of α-linolenic acid (C18:3n-3), DHA (22:6n-3), polyunsaturated fatty acids, and n-3 fatty acids, together with a lower content of myristic acid (C14:0), palmitic acid (C16:0), stearic acid (C18:0), total saturated fatty acids, and n-6/n-3 ratio compared to controls (P < 0.05). These findings indicate that perilla oil has the potential to decrease lipid-related indices and improve fatty acid profiles of breast meat in chickens without adverse effect on antioxidant status or meat quality; this was even better when perilla oil was given together with anise oil or ginger oil.

Abstract:This research investigated the effects of dietary rutin supplementation on the intestinal morphology, antioxidant capacity, immunity, and microbiota of aged laying hens. The results showed that 500 mg/kg rutin supplementation increased the villus height of jejunum (P < 0.05). Rutin affected the immune system of the ileum and the jejunum. In the jejunum, a diet with 500 mg/kg rutin supplementation enhanced secretory immunoglobulin A (sIgA) and reduced tumor necrosis factor-α (TNF-α) levels (P < 0.05). A diet with 1000 mg/kg rutin supplementation increased jejunal sIgA, immunologlobulin M (IgM), and interleukin-4 (IL-4) levels while decreasing interleukin-1β (IL-1β), TNF-α, and interferon-γ (IFN-γ) levels (P < 0.05). Meanwhile, a diet with 500 mg/kg rutin increased sIgA, immunologlobulin G (IgG), IgM, and interleukin-10 (IL-10) levels and reduced TNF-α and IFN-γ levels in the ileum (P < 0.05). In the ileum, a diet with 1000 mg/kg rutin supplementation raised sIgA, IgG, IgM, IL-4, and IL-10 levels while decreasing IL-1β, TNF-α, and IFN-γ levels (P < 0.05). At the family level, a diet with 500 mg/kg rutin supplementation raised the relative abundance of Monoglobaceae and decreased the relative abundance of Eubacteriaceae (P < 0.05) compared to the control group. In the 1000 mg/kg rutin group, the relative abundance of Lactobacillaceae and Unclassified Coriobacteriale was considerably lower and the relative abundance of Monoglobaceae was higher than the control group (P < 0.05). This study showed that a diet with rutin supplementation can improve the intestinal health of aged laying hens, and the mechanism is related to improving the intestinal morphology and intestinal immune status, and regulating the intestinal microbes.

Analysis of the Physical Meat Quality in Partridge (alectoris Chukar) and Its Relationship with Intramuscular Fat

Abstract:This study was undertaken to assess the effect of various levels of intramuscular fat (IMF: <0.5%, 0.5-0.99%, 1.0-1.49%, 1.5-1.99%, 2.0-2.49%, and >2.5%) on the physical meat quality of partridge. Physical characteristics such as moisture, pH, shear force, water-holding capacity (WHC), and color, along with IMF, were measured on 414 partridges (pectoral muscle). In this study, partridge meat was described as a kind of tender and moderately juicy meat with a nice color. Additionally, tenderness was significantly different between IMF < 0.5% and IMF > 0.5% levels (P < 0.01). IMF >0.5% also obtained higher value of WHC than IMF <0.5% although the difference was statistically nonsignificant. IMF levels did not affect pH or color. The results showed that IMF content was not significantly correlated with physical meat quality. However, in terms of eating experience, the minimum IMF level for better meat quality for consumer was 0.5%. Pearson correlation coefficients indicated that IMF content had significant negative correlations with moisture content but not with pH (r = -0.066), shear force (r = -0.072), WHC (r = 0.085), or color (L*(r = -0.049), a*(r = -0.028) and b*(r = 0.045)). Besides, meat pH had significant negative correlations with WHC (r = -0.036; P < 0.01) and lightness (L*) (r = -0.292; P < 0.01). Consequently, we consider pH to be one of the most important factors in evaluating meat quality of partridge.
Feed Restriction Improves Lipid Metabolism by Changing the Structure of the Cecal Microbial Community and Enhances the Meat Quality and Flavor of Bearded Chickens.

Abstract:Excessive fat deposition in full-fed Bearded chickens does not only reduce carcass yield but also causes consumer rejection of meat. Feed restriction (FR) is an effective method to save on feed cost, reduce carcass fat deposition, and improve meat quality. A total of 560 150-d Bearded chickens were randomly divided into seven groups (each with eight replicates of ten birds) for 40 days. The control group was fed with the basal diet ad libitum (CON), and the other six groups were fed with 90% of the feed intake (90% FI), 80% FI, 70% FI, 90% metabolizable energy (90% ME), 80% ME, and 70% ME of the CON, respectively. Compared to the CON group, FR increased meat yield, but the total weight of the Bearded chickens was slighter; 80% FI and 70% ME improved the relative lipid metabolism indices of chickens, especially the levels of triglycerides and total cholesterol in the plasma and liver (p < 0.05), and decreased calpastatin activity in the breast muscle (p < 0.05). Additionally, 16S rRNA sequencing of cecal microbial community indicated that an increase in the abundance of Hydrogenoanaerobacterium and Bacteroides plebeius was observed in the 80% FI group (p < 0.05), and an enrichment in Olsenella, Catabacter, and Lachnospiraceae were observed in the 70% ME group (p < 0.05) compared to the CON group. Moreover, compared to the CON group, the L * value of the breast muscle significantly decreased, and a * value significantly increased in the 80% FI group (p < 0.05). Notably, the concentrations of threonine, lysine, aspartic acid, glutamic acid, proline, and arginine and the activity of calpain in breast muscle increased in the 80% FI group more than in the CON group (p < 0.05), while valine, isoleucine, leucine, phenylalanine, lysine, alanine, tyrosine and proline decreased in ME restriction groups (p < 0.05). Taken together, our results indicated that 80% FI could improve lipid metabolism by changing the structure of the cecal microbial community, and the meat quality and flavor of the Bearded chickens in 80% FI group was improved with a promoted meat color score, flavor substances, and the calproteinase system.
Effects of Duzhong (eucommia Ulmoides Oliv.) on Growth Performance and Meat Quality in Broiler Chicks

Abstract:To investigate the effects of Eucommia ulmoides Oliv. (EUO) on growth performance and meat quality in broilers, two hundred and forty Avian commercial broilers were randomly allotted to 4 treatments with 4 replicates, 15 birds per replicate in earlier stage (1-21 days) and 11 birds per replicate in later stage (22-49 days). Birds were fed the same basal diet supplemented with 0 (control), 0.5, 1.5 and 2.5% EUO, respectively for a period of 7 weeks ad libitum. The results on growth performance showed that dietary supplementation with 2.5% EUO increased average daily feed intake by 3.15% (p<0.01) and survival percentage by 9.21% (p<0.05). Compared with the control group, leg muscle percentage in 2.5% EUO added group was increased by 7.67% (p<0.05). The contents of soluble collagen in breast muscle were increased by 13.25% (p<0.1), 24.10/0 (p<0.01), 18.07% (p<0.05) and total collagen by 3.47% (p>0.05), 17.03% (p<0.05), 45.42% (p<0.01), respectively with the supplementation of different level of EUO. Birds treated with 0.5, 1.5 and 2.5% EUO also resulted in an increase of Fe in breast muscle by 14.93%(p<0.11, 82.55% (p<0.05), 68.48% (p<0.05) and Mn by 50.00% (p<0.05), 63.64% (p<0.01), 72.73% (p<0.01), respectively. In addition, supplementation with EUO in the diet increased contents of serine, glutamic acid and lysine in breast muscle. It was indicated that incorporation of moderate levels of EUO in broiler diets is beneficial to improve growth performance and meat quality.

Abstract:The cases of two siblings affected with bird-breeder's lung are reported. The analysis of their HLA group demonstrated that both belonged to the haplotype HLA Bw15. The finding of a familial incidence of this type of alveolitis favors the presence of a genetic susceptibility for the disease. The same HLA group has been found in the rare families heretofore reported with bird-breeder's lung. Given that the pathogenesis of extrinsic allergic alveolitis has recently been linked to cellular immunity (T cells), and that the presence of a common HLA haplotype in patients suffering from bird-breeder's lung is related to the capacity for an immune response in general, and to the cellular immune response in particular, the presence of a genetic susceptibility in this disease appears as very likely, although the influence of other factors can not be ruled out.
Effects of Supplementation of Microalgae (aurantiochytrium Sp.) to Laying Hen Diets on Fatty Acid Content, Health Lipid Indices, Oxidative Stability, and Quality Attributes of Meat

Abstract:The present study is conducted to investigate the effects of dietary docosahexaenoic acid (DHA)-rich microalgae (MA, Aurantiochytrium sp.) on health lipid indices, stability, and quality properties of meat from laying hens. A total of 450 healthy 50-wk-old Hy-Line Brown layers were randomly allotted to 5 groups (6 replicates of 15 birds each), which received diets supplemented with 0, 0.5, 1.0, 1.5, and 2.0% MA for 15 weeks. Fatty acid contents and quality properties of breast and thigh muscles from two randomly selected birds per replicate (n = 12) were measured. The oxidative stability of fresh, refrigerated, frozen, and cooked meat was also determined. Results indicated that supplemental MA produced dose-dependent enrichments of long-chain n-3 polyunsaturated fatty acids (n-3 LC-PUFA), predominantly DHA, in breast and thigh muscles, with more health-promoting n-6/n-3 ratios (1.87–5.27) and favorable lipid health indices (p < 0.05). MA supplementation did not affect tenderness (shear force) and color (L*, a*, and b* values) of hen meat nor muscle endogenous antioxidant enzymes and fresh meat oxidation (p > 0.05). However, the n-3 LC-PUFA deposition slightly increased lipid oxidation in cooked and stored (4 °C) meat (p < 0.05). In conclusion, MA supplementation improves the nutritional quality of hen meat in terms of lipid profile without compromising meat quality attributes. Appropriate antioxidants are required to mitigate oxidation when such DHA-enriched meat is subjected to cooking and storage.

Abstract:This study aimed to investigate the effects of rutin on laying performance, egg quality, serum biochemical parameters, serum immunoglobulin, and antioxidant status in laying hens fed a diet containing stored soybean meal (SBM). The dietary treatments used were arranged in a 2 × 2 factorial arrangement with 2 types of SBM (FSM: SBM was stored in the cold storage warehouses at −20 °C for 45 days and was considered as fresh SBM; RTSM: SBM was stored in room temperature warehouse (15 °C to 25 °C), average temperature was 20 °C for 45 days) and 2 levels of rutin (0 and 500 mg/kg). In total, 384 Hy-Line Brown laying hens (220 days of age) were randomly divided into 4 groups with eight replicates. Pre-feeding period was 1 week, and the experiment lasted for 8 weeks. The results showed that the RTSM diet had no effect on laying performance ( p > 0.05). Compared with the diet without rutin, diet with 500 mg/kg rutin exhibited a tendency to increase egg mass compared with the diet without rutin ( p = 0.096), and there was a trend of interaction effect on average egg weight ( p = 0.061). The RTSM diet decreased the egg Haugh unit significantly compared with the FSM diet at the end of 8th week ( p < 0.05). Compared with the diet without rutin, dietary rutin supplementation could affect egg quality as evidenced by increased eggshell thickness, albumen height, egg yolk colour at the end of 4th week and increased eggshell strength and eggshell thickness at the end of 8th week ( p < 0.05). Furthermore, dietary rutin supplementation increased urea nitrogen, decreased the immunoglobulin M (IgM) content in serum compared with the diet without rutin ( p < 0.05). Compared with the diet without rutin, dietary supplementation with rutin improved the total antioxidant capacity in the serum and liver ( p < 0.05). The interactions of SBM × Rutin were observed on egg yolk ratio at the end of 8th week, and the urea nitrogen and IgM content in serum. These findings suggested that diet with RTSM had adverse effects on egg quality in laying hens. The RTSM diet negatively impacted egg quality of laying hens over time when compared to the FSM diet. Dietary rutin supplementation could improve the egg quality, and antioxidant capacity of laying hens. Dietary with soybean meal stored in room temperature warehouse reduced egg quality of laying hens. Dietary rutin supplementation improved egg quality and antioxidant capacity of laying hens independent of stored SBM. There was a significant interaction between stored soybean meal and rutin on egg yolk ratio, serum UN and IgM levels.

Cellular Antioxidant Mechanism of Selenium-Enriched Yeast Diets in the Protection of Meat Quality of Heat-Stressed Hens

Abstract:The effects and mechanisms of dietary selenium-enriched yeast (SeY, at Se 0, 0.25, and 0.50 mg/kg) on breast meat quality and oxidative stability of heat-stressed laying hens were studied. Two hundred and forty healthy Jingbai cross-bred hens at late laying period were randomly assigned to 4 experimental groups: one control group reared in a controlled thermal environment, and three treatment groups reared with heat stress (HS). The birds assigned to the controlled thermal environment were fed the basal diet, and the birds in the HS groups were fed the basal diet supplemented with 0, 0.25, or 0.50 mg Se (as SeY)/kg diet for 28 days. Chronic HS decreased pH(45)(min), increased lightness and drip loss, and impaired postmortem oxidative stability of breast muscle. The inclusion of SeY effectively reduced these stress-induced negative impacts, particularly on paleness, drip loss, and pH(45min) decline. The SeY significantly enhanced the cellular antioxidant defense system, suppressed the reactive oxygen species (ROS) overproduction, and mitigated the mitochondrial damage and apoptosis of stressed muscle. In conclusion, dietary modification with SeY reduced quality loss and oxidative instability of meat from heat-stressed hens by maintaining a balanced mitochondrial redox status and inhibiting ROS induced myocyte apoptosis.

Abstract:This study aimed at examining the effects of curcumin supplementation on growth performance, antioxidant capacity, and meat quality of ducks. To investigate these effects, 600 healthy ducks were randomly assigned to four treatment groups with 10 replicates pens, and each pen contained 15 ducks. Ducks were fed a diet containing curcumin at levels of 0, 300, 400, and 500 mg kg−1 in different groups. The results demonstrated that curcumin supplementation is beneficial to the growth performance (p < 0.05) of ducks and antioxidant capacity (p < 0.05) of duck meat. In addition, dietary curcumin raised the meat quality of ducks, improving the meat color, increasing water-holding capacity, and inhibiting lipid and protein oxidation. In conclusion, the present study provides important insights into both the nutrient and qualities of ducks, finding that a dietary inclusion of 400–500 mg/kg of curcumin (kg−1) has the greatest effect.

Each diet was fed randomly into three tanks, each tank containing 30 fish with an initial body mass of (10.27 ± 0.62) g. The feeding trial was conducted in an indoor recirculating aquiculture system at 28 °C for 56 days. According to the findings, the inclusion of 100 mg/kg rutin significantly improved the growth performance of yellow catfish and reduced the feed conversion ratio; however, the growth promotion effect was diminished when the diet was supplemented with 500 mg/kg of rutin. The inclusion of 500 mg/kg rutin in the diet significantly reduced the level of crude lipid and protein of the whole fish. Serum activities of alkaline phosphatase, albumin, and total protein were all significantly increased when fish were fed the diet supplemented with 500 mg/kg rutin, while serum glucose was significantly lower compared to the control group. Meanwhile, dietary rutin at a concentration of 500 mg/kg significantly induced the hepatic mRNA expressions of antioxidant-related genes (including Cu/Zn-SOD, Mn-SOD, CAT, GPx) and inflammatory-associated genes (including TNFα, IL-10, LYZ). Incorporating rutin at doses of 100 mg/kg and 500 mg/kg into the diets resulted in a notable increase in superoxide dismutase (SOD) activity, while simultaneously reducing malondiadehyde (MDA) content in the liver and intestine. Intestinal villus height, villus width, muscular thickness, and lumen diameter were significantly increased with the administration of 500 mg/kg of dietary rutin. Gut microbial diversity analysis indicated that supplementing diets with 100 mg/kg and 500 mg/kg rutin significantly enhanced the abundance of Cetobacterium while decreasing Plesiomonas richness. In conclusion, dietary rutin levels at 100 mg/kg could enhance the growth, antioxidant capability, and intestinal health of yellow catfish under present experimental conditions.

Abstract:The aim of this study was to determine the effects of curcumin on meat quality and antioxidant profile of breast muscle in broilers. In experiment 1, birds were fed basal,diet with an additional 0, 50, 100, or 200 mg/kg curcumin, respectively. The results showed that dietary curcumin significantly increased the redness values of meat, catalase activity, and ABTS radical scavenging activity and decreased drip loss at 48 h. In experiment 2, birds reared under heat stress were assigned to similar treatments as experiment 1. Significant differences the redox status of breast muscle were observed between the control and heat stress groups. The various levels of curcumin significantly prevented reactive oxygen species overproduction, enhanced the antioxidant defense system, and alleviated the abnormal change of antioxidant-related gene expression of muscle in heat-stressed birds. It was concluded that curcumin, as a potential antioxidant, improved meat quality and oxidant stability of muscle in broilers, whereas the inclusion of 50 and 100 mg/kg would be more efficient.

Abstract:This study was conducted to investigate the effects of selenomethionine (Se-Met) and sodium selenite (SS) supplementation on meat quality, selenium (Se) distribution, and antioxidant status in broilers. A total of 360 seven-days-old Ross 308 broilers of an average body weight 162 +/- 0.59 g were randomly allotted to three treatments, each of which included three replications of 40 birds. The treatments included a control diet containing 0.04 mg Se/kg and other two diets that contained 0.15 mg Se/kg supplemented by SS or Se-Met. The experiment lasted for 42 days. Selenium supplementation improved (P < 0.05) the Hunter a value of breast muscle in 8 and 16 h and decreased (P < 0.05) the drip loss of breast muscle in 24 and 48 h. Both Se sources and exposed time significantly influenced (P < 0.01) the drip loss of breast muscle. Selenium and glutathione concentrations in serum and in the studied organs were significantly higher (P < 0.05) after dietary Se supplementation while the Se-Met group showed the highest value (P < 0.05). Glutathione peroxidase activity in serum and in the studied organs was also significantly elevated (P < 0.05) by dietary Se supplementation while SS increased the glutathione peroxidase activities in pancreas and breast muscle to a larger extent (P < 0.05) than did Se-Met. The addition of Se from either source caused a significant increase (P < 0.05) in superoxide dismutase activities in tissues (except for kidney) whereas the Se-Met group was more effective (P < 0.05) than the SS group in breast muscle. Selenium supplementation increased (P < 0.05) the total antioxidant capability in serum, liver, kidney and breast muscle while the Se-Met group proved to be more effective (P < 0.05) than the SS group except for kidney. The Se-supplemented diets had a lower (P < 0.05) malondialdehyde concentration in serum and in the studied organs but the effect was more pronounced (P < 0.05) when Se-Met was used except for the serum. These results indicated that Se-Met supplementation was more effective than SS supplementation for depositing Se in serum and tissues, enhancing the antioxidant status and reducing the drip loss of breast muscle.
Transcriptome Analysis Reveals the Differentially Expressed Genes Associated with Growth in Guangxi Partridge Chickens

Abstract:The Guangxi Partridge chicken is a well-known chicken breed in southern China with good meat quality, which has been bred as a meat breed to satisfy the increased demand of consumers. Compared with line D whose body weight is maintained at the average of the unselected group, the growth rate and weight of the selected chicken group (line S) increased significantly after breeding for four generations. Herein, transcriptome analysis was performed to identify pivotal genes and signal pathways of selective breeding that contributed to potential mechanisms of growth and development under artificial selection pressure. The average body weight of line S chickens was 1.724 kg at 90 d of age, which showed a significant increase at 90 d of age than line D chickens (1.509 kg), although only the internal organ ratios of lung and kidney changed after standardizing by body weight. The myofiber area and myofiber density of thigh muscles were affected by selection to a greater extent than that of breast muscle. We identified 51, 210, 31, 388, and 100 differentially expressed genes (DEGs) in the hypothalamus, pituitary, breast muscle, thigh muscle, and liver between the two lines, respectively. Several key genes were identified in the hypothalamus-pituitary-muscle axis, such as FST, THSB, PTPRJ, CD36, PITX1, PITX2, AMPD1, PRKAB1, PRKAB2, and related genes for muscle development, which were attached to the cytokine-cytokine receptor interaction signaling pathway, the PPAR signaling pathway, and lipid metabolism. However, signaling molecular pathways and the cell community showed that elevated activity in the liver of line S fowl was mainly involved in focal adhesion, ECM-receptor interaction, cell adhesion molecules, and signal transduction. Collectively, muscle development, lipid metabolism, and several signaling pathways played crucial roles in the improving growth performance of Guangxi Partridge chickens under artificial selection for growth rate. These results support further study of the adaptation of birds under selective pressure.

Abstract:IntroductionIt is of great importance to seek agro-industrial byproducts that can serve as unconventional or alternative feedstuffs for poultry, especially those that are substitutive sources of n-3 polyunsaturated fatty acids (PUFAs), as this will contribute to alleviating feed shortage pressure and improving poultry performance and product quality. In this study, the effects of dietary fermented peony seed dreg (FPSD) on the production performance of hens and fatty acid composition of meat and egg yolk were evaluated.MethodsA total of 480 54-week-old Xinyang chickens were divided into 5 groups: control (basal diet); 5% peony seed dreg (PSD); and 5%, 7% and 9% FPSD, and each group consisted of 6 replicates with 16 birds per replicate. Production performances were recorded daily, and egg quality, serum parameters, antibody titers and fatty acid profile in the muscle and yolk were measured at 64 weeks of age.Results and discussionEgg production, feed conversion ratio and egg albumen quality were improved (p < 0.05) by 5% and 7% FPSD groups compared to 5% PSD and the control diet. Immune status was not influenced by PSD, but antibody titres against H7N9 and ND were improved by FPSD diets during most of the experimental periods. Dietary 5% and 7% FPSD increased (p < 0.05) serum high density lipoprotein concentrations and glutathione peroxidase actives. Total n-3 polyunsaturated fatty acids (PUFA) in meat and yolk increased gradually, while the total saturated fatty acids (SFA) and the n-6/n-3 PUFA ratio decreased gradually with increasing dietary FPSD levels. In conclusion, up to 7% dietary FPSD has the potential use to be utilized as a supplement in Xinyang laying hen diets to produce n-3 PUFA-enriched meat and eggs and positively affect production performance and health status.
