Abstract:Objective:To establish the method of colIloidal gold immunochromatographic assay for detecting tetracyclines residues.Methods:Nanocolloidal gold particles were prepared and labeled to an anti-tetracyclines monoclonal antibody.Tetracyclines was conjugated to bovine serum a1bumin(BSA)and dispersed on a nitrocellulose membrane to be the test 1ine(T).The more analyte present in the sample,the more effectively it would compete with the Tetracyclines-BSA for binding to the limited amount of gold-labeled anti-tetracyclines monoclonal antibody.The presence or absence of a colored band on the test line indicated a negative or positive result.Results:When measuring the meat sample spiked with tetracyclines,the minimum detection concentration can reach 10 ng/ml.The major advantage of the one-step strip test is that the test can be completed within 5 minutes.No cross-reaction occurred between tetracyclines and other veterinary drugs.Conclusion:The method for tetracyclines is of high sensitivity,specificity and good stability and it could be widely used to detect the tetracycline residues.
Milk Residues Caused by Treatment of Endometritis with Antibacterial Drugs and Effect on Yogurt Production

Abstract:The2,3,5-triphenyltetrazolium chloride(TTC) assay was used to investigate the drug's residues in milk collected from 44 dairy cows with endometriris after antibacterial therapy.Of the 44 cases,41 were treated by 1 to 5 intrauterine infusions of oxytetracycline,chlortetracycline hydrochloride or gentamicin,and 3 were treated mainly by intravenous injections of sulfadiazine of cefaloridine.The results indicated that all 31 cases produced TTC positive milk 1 day after intrauterine infusions of oxytetracycline with 6 cases remaining TTC positive at 6 days after the last infusion.8 cases treated with intrauterine infusion of chlortetracycline produced TTC positive milk 2 days after treatment with a drug residue persisting in milk from 2 to 5 days.2 cases treated with gentamicin were detected TTC positive at 1 and 2 days,respectively.Milk residue persisting more than 9 days was found in the cases treated with intravenous injection of antibacterial drugs. In addition,milk samples were collected from 3 cows treated by intrauterine infusion of chlortetracycline hydrochloride, and then inoculated with a yogurt making bacterial culture to test their yogurt making ability.The results indicated that the milk samples showing TTC positive were not fermented normally;fermentation was badly inhibited in milk samples collected 6 hours after the treatment and recovered 13,24 or 48 hours later.
Highly Sensitive Immunochromatographic Identification of Tetracycline Antibiotics in Milk

Abstract:A rapid immunochromatographic assay was developed for the control of tetracycline (TC). The assay is based on the competition between immobilized TC-protein conjugate and TC in a tested sample for binding with polyclonal anti-TC antibodies conjugated to colloidal gold during the flow of the sample along a membrane strip with immobilized reactants. Conjugation of colloidal gold and the total immunoglobulin (IgG) fraction of polyclonal antibodies was used to increase the assay sensitivity to ensure low content of specific antibodies in the conjugate. This allowed effective inhibition of free TC and conjugate binding in the strip test zone. Photometric marker registration allows control of the reduction of binding, thereby enhancing detection sensitivity. The proposed assay allows TC to be detected at concentrations up to 20 ng/mL, exceeding the limit of detection of the known analogues, in a wide working range (more than two orders) of 60 pg/mL to 10 ng/mL, ensured through the use of polyclonal antibodies. The assay time is 10 min. The efficiency of the designed assay is shown to identify TC in milk; the degree of recovery of TC ranges from 90 to 112%. The precision of the concentrations measurements was no more than 10%.
Identification and dietary exposure assessment of tetracycline and penicillin residues in fluid milk, yogurt, and labneh: A cross-sectional study in Lebanon

Abstract:Background and aim: The safety and quality of dairy products are considered to be of significant importance to human health. Although antimicrobial drugs are essential for disease treatment in modern medicine, the use of these drugs can have undesired consequences for human and animal health. This study aimed to investigate the presence of tetracycline and penicillin residues in raw, pasteurized, and UHT cow's milk of different fat contents, as well as in the dairy products yogurt and labneh, a traditional Lebanese product. Materials and methods: A total of 44 samples, 4 raw, 9 UHT, 9 pasteurized milk, 10 yogurt, and 12 labneh samples from common local brands available in the Lebanese market were collected from Keserwan regions in May 2016. Tetracycline and penicillin residues were determined using a competitive enzyme-linked immunosorbent assay (ELISA) technique. Results: The mean values for tetracycline and penicillin were all below the limit of detection (LOD) of the ELISA kit of a maximum standard concentration of 1.80 µg/kg and 4.00 µg/kg, respectively. All samples tested positive for antibiotic residues. The detection rate for tetracycline in milk (n=22) samples was 86.4% with a mean residues value of 1.16±0.70 μg/kg. The detection rate of tetracycline in labneh (n=12) and yogurt (n=10) samples was 50% for each with a mean value of 1.76±0.40 μg/kg and 0.63±0.12 μg/kg, respectively. As for penicillin residues, 90.9% of the milk (n=22) samples tested positive with a mean value of 0.52±0.25 μg/kg. The detection rate in labneh (n=12) and yogurt (n=10) samples was 0% for penicillin residues, where mean values were all below the LOD (<1.25 μg/kg) for these dairy products. None of the samples exceeded the maximum residue levels. The estimated dietary intake (EDI) for tetracycline and penicillin residues for all dairy products is 2.09 ng/kg body weight (BW)/day resulting in 0.007% of the acceptable daily intake (ADI) and 1.83 ng/kg BW/day resulting in 0.006% of the ADI, respectively. Conclusion: All EDI values were below the ADI set for each antibiotic residue and do not exceed relevant toxicological reference values. However, concerns might still be present from consumption of other animal food products containing residues. Moreover, the long-term exposure to such residues is still unknown as a result of bioaccumulation; it is a challenging process to determine the actual dietary consumption of foods containing antibiotic residues; hence, the human health risk cannot be easily predicted.

Abstract:Tetracycline (TC) is a vital class of broad-spectrum antibiotics widely employed. However, its extensive use raises associated concerns. Due to its persistent nature, there is a risk of trace amounts of TC contaminating food. The complex composition of food samples has sparked considerable interest in developing selective and sensitive TC sensors. Here, a novel sensing material, gCDs@ZIF-8, where ZIF-8 is a metal–organic framework composed of Zn 2+ and 2-methylimidazole, was developed by incorporating fluorescent green carbon dots (gCDs) into the ZIF-8 for TC detection. Consequently, the synthesized gCDs exhibited green emission and a specific response to tetracycline. The incorporation of gCDs into ZIF-8 enhances sensitivity, making it a valuable probe for the quantification of TC. The detection limit for gCDs alone was 0.42 μM, and when encapsulated in ZIF-8, the sensitivity increased, resulting in a limit of detection of 157 nM. Quantifying tetracycline in milk samples demonstrated spike recoveries ranging from 95 to 105 percent. The assay displayed excellent precision, with a relative standard deviation (RSD, n = 3) of less than 2% at each concentration point. The robustness and ruggedness of the assay were evident through the probe's stable shelf life, water stability, and a broad working pH range. Validation tests confirmed the gCDs@MOF-based sensing system's capability to detect tetracycline in various sample matrices. The method's environmental friendliness was assessed using three metric tools (AGREE, AGREEprep, and the Complex GAPI), all confirming its superior eco-friendliness.

Abstract:Therapeutic and subtherapeutic use of veterinary drugs has increased the risk of residue contamination in animal food products. Antibiotics such as tetracycline are used for mastitis treatment of lactating cows. Milk expressed from treated cows before the withdrawal period has elapsed may contain tetracycline residue. This study developed a simple surface-enhanced Raman spectroscopic (SERS) method for on-site screening of tetracycline residue in milk and water. Six batches of silver colloid nanoparticles were prepared for surface enhancement measurement. Milk-tetracycline and water-tetracycline solutions were prepared at seven concentration levels (1000, 500, 100, 10, 1, 0.1, and 0.01 ppm) and spiked with silver colloid nanoparticles. A 785 nm Raman spectroscopic system was used for spectral measurement. Tetracycline vibrational modes were observed at 1285, 1317 and 1632 cm-1 in water-tetracycline solutions and 1322 and 1621 cm-1 (shifted from 1317 and 1632 cm-1, respectively) in milk-tetracycline solutions. Tetracycline residue concentration as low as 0.01 ppm was detected in both the solutions. The peak intensities at 1285 and 1322 cm-1 were used to estimate the tetracycline concentrations in water and milk with correlation coefficients of 0.92 for water and 0.88 for milk. Results indicate that this SERS method is a potential tool that can be used on-site at field production for qualitative and quantitative detection of tetracycline residues.

Abstract:The aim of this study was to investigate the transfer of residues of five β-lactam antibiotics (ampicillin, penicillin G, cloxacillin, dicloxacillin and cephalexin) and two tetracyclines (tetracycline and oxytetracycline) in the processing of cheese and whey powder, evaluating the effect of the processes and the final concentration in each product generated. Raw milk was fortified at two concentration levels with the seven antibiotics. The first concentration level (C1) was chosen according to the maximum residue limit (MRL) of each antibiotic (ampicillin and penicillin G: 4 μg kg −1 ; cloxacillin and dicloxacillin: 30 μg kg −1 ; cephalexin, tetracycline and oxytetracycline: 100 μg kg −1 ). The second concentration level (C2) was spiked as follows according to each antibiotic: 0.5 MRL (cloxacillin, dicloxacillin, cephalexin), 0.1 MRL (tetracycline and oxytetracycline) and 3 MRL (ampicillin and penicillin G). The antibiotics were analyzed by LC-MS/MS. No ampicillin or penicillin G residues were found in cheese or whey powder, although they were detected in whey at concentrations similar to those added to raw milk. Cephalexin was mostly distributed in whey between 82% and 96%, being the antibiotic that presented the highest concentration in whey powder (784 ± 98 μg kg −1 ) when milk was spiked at the MRL. The whey distribution of cloxacillin and dicloxacillin ranged from 57% to 59% for cloxacillin and from 46% to 48% for dicloxacillin, and both concentrated in whey powder. Tetracyclines were the antibiotics that concentrated in cheese, with retentions between 75% and 80% for oxytetracycline and between 83% and 87% for tetracycline. The distribution of antibiotics in the dissimilar stages of the cheese and whey powder production processes, as well as their concentration in the final products, depend on each type of antibiotic. Knowledge of the transfer of antibiotic residues during the process and final disposal is an input for the risk assessment of their consumption.

Abstract:giant congenital pigmented naevi is a great reconstructive challenge for the pediatric and plastic surgeons. due to the increased risk of malignant transformation in such lesions, many procedures have been used to remove giant congenital naevi like dermoabrasion, laser treatment or surgical excision combined with reconstruction through skin expansion or skin grafting; among these, only a complete excision can offer an efficacious treatment. in our centre we use the “tissue expansion” technique in order to achieve a sufficient quantity of normal skin to perform a both staged and radical excision of these giant lesions.

Abstract:Environmental contaminant is one of several problems harming people and wildlife. An example of current emerging contaminants are antibiotics residues that can present in water and food. Although antibiotics are intended to treat or prevent human and animal infections, antibiotics have also been used as animal food supplements for their ability to promote growth and feed efficiency. This overuse of antibacterial has resulted in the accumulation of antibiotics residues in food products which are eventually consumed by human. The continuous unnecessary exposure of human to antibiotics through the direct animals meet or milk, or indirectly through plants or soil can increase the chance of the emergence of multi drug resistance bacteria and consequently adversely affecting human health. New regulations have been imposed regarding antibiotics utilization. Due to the scarce of data regarding antibiotics residue conditions in different types of food intended for human consumption in Saudi Arabia, this study proposed an optimized chromatographic method (HPLC-DAD) followed by an immunoassay approach for specifically detecting tetracyclines antibiotics in animal milk samples. The method was carried out using an RP-C18 column with a mobile phase consisting of 0.01 M KH 2 PO 4 : acetonitrile:methanol (70:20:10, v/v/v) adjusted to pH 4. Improvements were observed in the method in terms of resolution and sensitivity. The protein precipitation method used for extraction demonstrated high percent recoveries of 85–101%. The method was validated according to the guidelines of the International Conference for Harmonization (ICH). It is evidently clear from these findings that the presence of tetracycline and oxytetracycline antibiotics residues in milk products from the Saudi market are below the maximum residual limits (MRLs).

Microextraction of Tigecycline Using Deep Eutectic Solvents and Its Determination in Milk by LC-MS/MS Method

Abstract:The occurrence of tigecycline (TGC), a new first glycylcycline antibiotic residues in food products harmfully influences potential human consumers health. Therefore, analysts are forced to develop new microextraction methods connected with modern extractants for effective isolation of this compound. For this purpose, deep eutectic solvents (DES) as the extraction media were used. Liquid-liquid microextraction (LLME) of tigecycline from milk samples with application of the hydrophobic deep eutectic solvents: decanoic acid:thymol (1:1), thymol:camphor (2:1), dodecanoic acid:menthol (2:1), and dodecanoic acid:dodecanol (1:1) was developed. The studied samples were subjected to a deproteinization process using trichloroacetic acid solution and acetonitrile. The optimal microextraction parameters, molar ratio of DES components, amount of extraction solvents, pH of milk sample, shaking, and centrifugation time, were chosen. Tigecycline in the obtained microextracts of deep eutectic solvents was analyzed using a liquid chromatographic technique connected with a tandem mass spectrometry (LC-MS/MS) system. The limits of detection and quantification values for TGC determination followed by DES-LLME-LC-MS/MS method were in the 1.8 × 10-11 mol L-1 (0.01 μg kg-1) to 4.0 × 10-9 mol L-1 (2.28 μg kg-1) and 5.5 × 10-11 mol L-1 (0.03 μg kg-1) to 1.2 × 10-8 mol L-1 (6.84 μg kg-1) ranges, respectively. The RSD values of precision were in the range 1.4-7.8% (intraday) and 5.4-11.7% (interday). The developed procedures were used for the determination of tigecycline in different bovine milk samples.

Abstract:Foreign biologically active substances that can be contained in milk of cows, which having a negative impact on the health and quality of life of people are antibiotics. Intensive use of antibiotics in veterinary practice contributes to the globalization of micro doses in the environment, accelerates the evolution and spread of resistant forms of microorganisms. The principle of prioritizing the safety of dairy products, based on the mandatory control of raw milk for the content of residual amounts of antibiotics, is implemented in accordance with the requirements of regulatory and technical documents. There is a need to use high-tech laboratory methods of detection of antimicrobial drugs in raw materials and livestock products. The purpose of the research was to control the contamination of raw milk by antibiotics. Raw milk of morning milking from cows of Black-and-White breed was used for the research. Milk was obtained at the zoo station of the Federal State Budgetary Educational Institution of Higher Education “Russian State Agrarian University – Moscow Timiryazev Agricultural Academy”. In the samples residual amounts of antibiotics of diff erent pharmacological groups, most widely used in productive animal husbandry were determined. Sampling of raw milk and their preparation for research was carried out in accordance with State Standard T 26809.1-2014. The determination of residual amounts of antibiotics was carried out by the method of an internal standard for the areas of chromatographic peaks of identifi ed compounds using a calibration characteristic obtained by analyzing calibration solutions of known compounds under similar conditions. It was established that monitoring the health of lactating cows, strict compliance with the rules for the use of veterinary medicines, the exclusion of errors in the technology of production of feed and feed additives, contribute to minimizing contamination of raw milk by antibiotics.
Novel enzyme‐based electrochemical and colorimetric biosensors for tetracycline monitoring in milk

Abstract:Recently, there has been a growing demand to develop portable devices for the fast detection of contaminants in food safety, healthcare, and environmental fields. Herein, two biosensing methods were designed by the use of NAD(P)H‐dependent TetX2 enzyme activity and thionine as an excellent electrochemical and colorimetric mediator/probe to monitor tetracycline (TC) in milk. The nano‐porous glassy carbon electrode (NPGCE) modified with polythionine was first prepared by electrochemically and then TetX2 was immobilized onto the NPGCE using polyethyleneimine. The prepared biosensor provided a high electrocatalytic response toward NAD(P)H by significantly reducing its overpotential. The proposed biosensor exhibited a detection limit of 40 nM with a linear range of 0.1 to 0.8 μM for TC determination. Besides, the thionine probe was used to develop a novel colorimetric assay using a simple enzymatic color reaction within a few minutes. The limit of detection for TC was experimentally achieved as 60 nM, which was lower than the safety levels established by the WHO (225 nM). The correlation between change in the color of the solution and the concentration of TC was used for quality control of milk samples, as confirmed by the standard HPLC method. Development and validation of reverse - phase HPLC method for detection of tetracycline residue in fresh milk samples

Abstract:Dairy products are highly required by humans for growth, development and repairs as they are good sources of essential nutrients. Indiscriminate administration of veterinary drugs for prophylactic and therapeutic purposes could lead to the presence of their residues in dairy products such as milk. Reversed-phase high performance liquid chromatographic method with UV detection was developed and validated for detection of tetracycline in milk from five different samples. Mobile phase consisting of a mixture of deionized water, acetonitrile, monobasic potassium and acetic acid was delivered at a flow rate of 1.0 mL/min. The column used was Agilent ZORBAX ODS 4.6 X 150 mm 5μm.. The eluent was monitored by UV detection at 220 nm. Analysis was performed at 35oC and the total run time was 20 minutes. Analytical parameters such as linearity, accuracy, precision and limit of detection were determined by validation procedure and found to be satisfactory. Calibration curve was linear over the concentration range of 0.01- 0.5 mg/mL with an R2 of 0.9978. The limits of detection and limit of quantification were found to be 0.00023 mg/mL and 0.00110 mg/mL, respectively. The % RSD is found to be 3.62, 0.23 and 0.44 for 0.01, 0.1 and 0.5 mg/mL concentration with percentage recovery of 78.7 – 98.6 %. Generally, the developed method was found to be simple, rapid, precise and accurate for quality control of tetracycline in fresh milk samples.
Surface-Enhanced Raman Spectroscopy for Trace Detection of Tetracycline and Dicyandiamide in Milk Using Transparent Substrate of Ag Nanoparticle Arrays

Abstract:Tetracycline (TC) is an antibiotic utilized in dairy livestock, which can leave trace residues in milk and other dairy products. Dicyandiamide (DCD), a grassland fertilizer, but also potentially an adulterant to increase milk apparent protein content, has been found in milk products that were subsequently banned. Both these chemicals can pose human health risks via the consumption of dairy foods, and therefore rapid, simple detection methodologies are necessary. Herein, we fabricated a transparent and homogeneous substrate by fabricating silver nanoparticle (Ag NP) arrays based on the anodic aluminum oxide (AAO) template-assisted electrochemical deposition approach. The periodic nanostructure was optimized to obtain high SERS activity by varying nanopore diameters and Ag NP sputtering duration to have Ag NP nanogaps in the sub 10 nm regime so that the high sensitivity was achieved with detecting R6G analytes at the 1 × 10–10 M level. The substrate was also used to detect TC and DCD in milk at low concentration levels of 1 × 10–9 M and 1 × 10–7 M, respectively, with good linear correlation between concentration and intensity (TC R2 = 0.96; DCD R2 = 0.99). The assignments of Raman bands of tetracycline and dicyandiamide were also given by density-functional-theory (DFT) calculations. The advantage of this transparent SERS substrate is that it is especially useful for measurement of analytes at the interface between Ag NP substrate layer and the opaque liquid containing the analytes without special pretreatment of the sample. Evolution of a natural TetR protein and development of a Fe3O4 assisted semi-homogeneous fluorescent method for determination of tetracyclines in milk

Abstract:Compared with antibody, the recognition spectrum of a receptor is broader, and its recognition ability can be improved using simple mutagenesis technique. Compared with conventional immunoassay, the magnetic bead based immunoassay is simpler and can be recycled. Compared with colorimetric and luminescent immunoassays, fluoroimmunoassay is simpler because it does not require a substrate. So a method combines these merits is desirable. In this study, two amino acids in the binding pocket of a natural Escherichia coli TetR protein were mutated to produce a mutant, and the molecular docking showed the binding energies and the numbers of contact acid for 10 tetracyclines all increased. The mutant was coupled with Fe3O4 to synthesize a magnetic complex, and a fluorescent tracer was synthesized by coupling quantum dot and minocycline with bovine serum albumin. Under the assistance of 96-well bottom magnet, a semi-homogeneous method based on the two materials was developed on conventional microplate for determination of the 10 tetracyclines in milk. Results showed once assay was finished within 20 min, the limits of detection (drug concentration showing 10% inhibition) for the 10 drugs were in the range of 0.32-0.94 ng/mL, and the magnetic complex could be regenerated for 6 times. Furthermore, the sensitivities were improved for 4-6 folds in comparison with the use of natural TetR. Therefore, this method is simple, sensitive, time-saving and recyclable, and it can be used for routine screening of the 10 tetracyclines in milk.

Abstract:Abstract Introduction Multi-class and multi-residue analyses are very complex procedures because of the physico-chemical properties of veterinary drug residues and other contaminants. The purpose of the study was to develop an analytical method for the sensitive determination of 69 analytes in bovine milk by liquid chromatography electrospray ionisation–tandem mass spectrometry. Material and Methods Antimicrobial, anabolic hormone, lactone, β-agonist, mycotoxin and pesticide residues were analysed in 120 raw milk samples from different dairy farms in North Macedonia. Stable isotopically labelled internal standards were used to facilitate effective quantification of the analytes. Results The linear regression coefficients were higher than 0.99, the limits of detection ranged from 0.0036 to 47.94 μg/L, and the limits of quantification ranged from 0.053 to 59.43 μg/L. The decision limit values ranged from 0.062 to 211.32 μg/L and the detection capability from 0.080 to 233.71 μg/L. Average recoveries of the analytes spiked in raw milk were in the range of 70.83% to 109%, intra-day coefficient of variation (CV) values from 2.41% to 22.29%, and inter-day CV values from 3.48% to 23.91%. The method was successfully applied in the testing of bovine milk samples. In five samples residues were detected. They were sulfadimethoxine (in two samples), enrofloxacin, tetracycline and oxytetracycline and were at concentrations below the EU maximum residue limit. Conclusion The method is useful for routine testing for this group of chemical hazards in bovine milk.

Abstract:In this study, the identity of our recently produced natural TetR protein was identified by using the LC-ESI-MS/MS technique, and its recognition mechanisms, including the binding pocket, contact amino acids, intermolecular forces, binding sites, binding energies, and affinities for 10 tetracycline drugs were studied. Then, it was evolved by site-mutagenesis of an amino acid to produce a mutant, and a fluorescence polarization assay was developed to detect the 10 drugs in milk. The sensitivities for the 10 drugs were improved with IC50 values decreasing from 30.8–80.1 ng/mL to 15.5–55.2 ng/mL, and the limits of detection were in the range of 0.4–1.5 ng/mL. Furthermore, it was found that the binding affinity for a drug was the critical factor determining its sensitivity, and the binding energy showed little influence. This is the first study reporting the recognition mechanisms of a natural TetR protein for tetracyclines and the development of a fluorescence polarization assay for the detection of tetracyclines residues in food samples.

Abstract:Exploring materials with the dual functionality of detecting and removing tetracycline (TC) residues is crucial because of the environmental and health risks posed by antibiotic overuse. This study introduces a dual-emissive luminescent probe, CDs@ZSM-5:Eu3+, created through a solvent-free method combined with subsequent Eu3+ion exchange. The nanocomposite's blue emission, originating from carbon dots (CDs), is quenched by TC via an internal filtering effect, while an antenna effect triggers a strong red fluorescence of a TC-Eu3+chelate. The ratiometric fluorescence changes in CDs@ZSM-5:Eu3+ endow a self-calibrated sensing mechanism for TC, offering a low detection limit of 5.04 nM and a broad detection range of 0.01-50 μM. Demonstrated in real milk samples, the probe exhibits high selectivity and accuracy in detecting TC. The nanocomposite also displayed an impressive TC removal capacity of 238.1 mg g-1 in water, ascribing to the enrichment and electrostatic attraction effects of ZSM-5 toward TC molecules. This research offers a facile strategy for constructing multifunctional zeolite-based hybrids for simultaneous TC detection and removal from aqueous solutions.

Abstract:Antibiotic residues in animal-derived foods pose risk to human health including chemical poisoning and antimicrobial resistance, and therefore, constant monitoring throughout the food supply chain is important. In the present study multi-dimensional fluorescence spectroscopy (3D and 2D) coupled with chemometric tools were tested for potential application to detect, discriminate and quantify penicillin G (PG), sulfadiazine (SF) and tetracycline (TC) in milk by direct measurement. Qualitative and quantitative calibration models were developed for prediction of antibiotic residues. Results demonstrated optimal discrimination of milk samples on the basis of antibiotic type and concentration with close to 100% of accuracy. Negative correlation between antibiotic concentration and fluorescence peak height was displayed (r ≥ 0.963 and p ≤ 0.002). A great potential for quantitative antibiotic determination was established with R2 > 0.9 and low standard errors of estimation indicating acceptable precision for the developed technique. Fluorescence spectroscopy demonstrated high specificity and sensitivity with detection limit below the maximum residue limit of PG, SF and TC in milk. Therefore, fluorescence spectroscopy can be used as an alternative method for rapid screening of antibiotic residues in milk at collection centers and processing plants to ensure product quality and safety.

A double-blind study comparing sodium aurothiomalate and auranofin in patients with rheumatoid arthritis previously stabilized on sodium aurothiomalate.

Abstract:Auranofin, an orally active gold preparation, was compared with sodium aurothiomalate in a double-blind trial in patients with rheumatoid arthritis fulfilling the ARA criteria, who had been stabilized on sodium aurothiomalate for at least six months. Twenty-four patients have so far been entered in the trial, of whom fourteen have been randomly allocated to receive auranofin and ten to receive sodium aurothiomalate. After initial stabilization, patients receive either auranofin 6 mg daily and placebo injection, or sodium aurothiomalate 50 mg monthly and placebo tablets. Five patients have completed one year on auranofin. The remaining nine patients were withdrawn because of loss of efficacy (two), side-effects, (five), loss of efficacy and side-effects (one) and default (one). Four patients have completed one year's treatment with sodium aurothiomalate. Of the remaining six patients, two were withdrawn because of side-effects, three because of poor disease control and one because of side-effects and poor disease control. Diarrhoea occurred in eight patients receiving auranofin. Rashes occurred in both groups but otherwise there were no serious side-effects. The efficacy of both drugs appeared similar, there being no significant differences in morning stiffness, fatiguability, visual analogue pain score, grip strength and articular index. There were also no significant differences in laboratory parameters of efficacy. Auranofin appears to control disease activity in rheumatoid arthritis but diarrhoea is a frequent side-effect.
