Abstract:Suspect screening and untargeted analysis using UHPLC-qToF are two advanced analytical approaches now used to achieve an extensive chemical profile of samples, which are then typically confirmed through targeted analysis. These techniques can detect a large number of chemical features simultaneously and are currently being introduced into the study of contaminants of emerging concern (CECs) and into the study of the extent of human chemical exposure (the exposome). Here is described the use of these techniques to characterize chemical mixtures derived from the OECD 301F ready biodegradability test (RBT) of a chemical and natural formulation currently used to treat reflux disease and functional dyspepsia. Untargeted analysis clearly evidenced a different behavior between formulations containing only natural products with respect to that containing synthetic and non-naturally occurring substances. Suspect screening analysis improved the untargeted analysis of the omeprazole-based medicine, leading to the tentative identification of a number of omeprazole-derived transformation products, thereby enabling their preliminary quali-quantitative evaluation. Targeted analysis was then performed to confirm the preliminary data gained from the suspect screening approach. The validation of the analytical method for the quantitative determination of omeprazole and its major metabolite, omeprazole sulphide, has provided robust data to evaluate the behavior of omeprazole during the OECD 301F test. Using advanced analytical approaches, the RBT performed on the two products under investigation confirmed that omeprazole is not readily biodegradable, while the medical device made of natural substances has proven to be readily biodegradable.

Abstract:DNA vaccines represent a new frontier in vaccine technology. One important application of this technology is in the veterinary arena. DNA vaccines have already gained a foothold in certain fields of veterinary medicine. However, several important questions must be addressed when developing DNA vaccines for animals, including whether or not the vaccine is efficacious and cost effective compared with currently available options. Another important question to consider is how to apply this developing technology in a wide range of different situations, from the domestic pet to individual fish in fisheries with several thousand animals, to wildlife programs for disease control. In some cases, DNA vaccines represent an interesting option for vaccination, while in others, currently available options are sufficient. This review will examine a number of diseases of veterinary importance and the progress being made in DNA vaccine technology relevant to these diseases, and we compare these with the conventional treatment options available.
Primary culture of rat thymic non-lymphoid cells: influence of culture time on the expression of macrophage differentiation antigens defined by monoclonal antibodies.

Abstract:A panel monoclonal antibodies (mAbs) raised to rat thymic non-lymphoid cells has been shown to discriminate between distinct subpopulations of macrophages depending on their anatomic localization in the thymus. These reagents were used in this study to examine the expression of macrophage-associated antigens in primary culture of rat thymic stromal cells. The phenotype of both adherent macrophage (AM) monolayers and non-adherent cells (NAC) released in culture medium was studied at different time points after cultivation. More than 95% AM expressed ED1 and R-MC 38 antigens (pan-macrophage markers), class I MHC antigens (OX-18) and iC3b receptor recognized by OX-42 mAb. Most of them (70-85%) were reactive with ED2, R-MC 40, 41 and 42 mAbs specific for cortical and cortico-medullary zone (CMZ) macrophages. A much smaller percentage was positive with R-MC 43/44 and R-MC 46/47 mAbs staining CMZ/medullary macrophages and a subset of cortical macrophages, respectively. A minor subset of AM expressed class II MHC molecules which progressively decreased during cultivation. NAC were phenotypically heterogeneous. In comparison with adherent cells they contained a lower percentage of cortical/CMZ phenotype macrophages. In addition, NAC were slightly enriched in R-MC 43+ cells and more significantly expressed IA/E antigens (85-95%). ED3, R-MC 39 and 45 mAbs reactive with thymic macrophages in situ were mostly non-reactive with AM and NAC in culture.
Tracing pharmaceuticals in a municipal plant for integrated wastewater and organic solid waste treatment

Abstract:The occurrence and removal of 42 pharmaceuticals, belonging to different therapeutic groups (analgesics and anti-inflammatory drugs, anti-ulcer agent, psychiatric drugs, antiepileptic drug, antibiotics, ß-blockers, diuretics, lipid regulator and cholesterol lowering statin drugs and anti-histamines), were studied in the wastewater and sewage sludge trains of a full scale integrated treatment plant. The plant employs a biological nutrient removal (BNR) process for the treatment of municipal wastewater, and a single-stage mesophilic anaerobic co-digestion for the treatment of wasted activated sludge mixed with the organic fraction of municipal solid waste (OFMSW), followed by a short-cut nitrification-denitrification of the anaerobic supernatant in a sequential batch reactor. Influent and effluent wastewater, as well as thickened, digested and treated sludge were sampled and analyzed for the selected pharmaceuticals in order to study their presence and fate during the treatment. Twenty three compounds were detected in influent and effluent wastewater and eleven in sludge. Infiltration of groundwater in the sewer system led to a dilution of raw sewage, resulting in lower concentrations in wastewater (up to 0.7 μg/L in influent) and sludge (70 ng/g d.w.). Due to the dilution, overall risk quotient for the mixture of pharmaceuticals detected in effluent wastewater was less than one, indicating no direct risk for the aquatic environment. A wide range of removal efficiencies during the treatment was observed, i.e. <20% to 90%. The influent concentrations of the target pharmaceuticals, as polar compounds, were undoubtedly mostly affected by BNR process in the wastewater train, and less by anaerobic-co-digestion. Mass balance calculations showed that less than 2% of the total mass load of the studied pharmaceuticals was removed by sorption. Experimentally estimated distribution coefficients (<500 L/kg) also indicated that the selected pharmaceuticals preferably remain in the aqueous phase, and that biodegradation/transformation is the primary removal mechanism for these compounds during wastewater treatment.

Analyzing microbial communities and their biodegradation of multiple pharmaceuticals in membrane bioreactors

Abstract:Pharmaceuticals are of concern to our planet and health as they can accumulate in the environment. The impact of these biologically active compounds on ecosystems is hard to predict, and information on their biodegradation is necessary to establish sound risk assessment. Microbial communities are promising candidates for the biodegradation of pharmaceuticals such as ibuprofen, but little is known yet about their degradation capacity of multiple micropollutants at higher concentrations (100 mg/L). In this work, microbial communities were cultivated in lab-scale membrane bioreactors (MBRs) exposed to increasing concentrations of a mixture of six micropollutants (ibuprofen, diclofenac, enalapril, caffeine, atenolol, paracetamol). Key players of biodegradation were identified using a combinatorial approach of 16S rRNA sequencing and analytics. Microbial community structure changed with increasing pharmaceutical intake (from 1 to 100 mg/L) and reached a steady-state during incubation for 7 weeks on 100 mg/L. HPLC analysis revealed a fluctuating but significant degradation (30-100%) of five pollutants (caffeine, paracetamol, ibuprofen, atenolol, enalapril) by an established and stable microbial community mainly composed of Achromobacter, Cupriavidus, Pseudomonas and Leucobacter. By using the microbial community from MBR1 as inoculum for further batch culture experiments on single micropollutants (400 mg/L substrate, respectively), different active microbial consortia were obtained for each single micropollutant. Microbial genera potentially responsible for degradation of the respective micropollutant were identified, i.e. Pseudomonas sp. and Sphingobacterium sp. for ibuprofen, caffeine and paracetamol, Sphingomonas sp. for atenolol and Klebsiella sp. for enalapril. Our study demonstrates the feasibility of cultivating stable microbial communities capable of degrading simultaneously a mixture of highly concentrated pharmaceuticals in lab-scale MBRs and the identification of microbial genera potentially responsible for the degradation of specific pollutants. Abstract:The widespread presence of organic micropollutants in the environment reflects the inability of traditional wastewater treatment plants to remove them. In this context, advanced oxidation processes (AOPs) have emerged as promising quaternary wastewater treatment technologies since they efficiently degrade recalcitrant components by generating highly reactive free radicals. Nonetheless, the chemical characterization of potentially harmful byproducts is essential to avoid the contamination of natural water bodies with hazardous substances. Given the complexity of wastewater matrices, the implementation of comprehensive analytical methodologies is required. In this work, the simultaneous photoelectrochemical degradation of seven environmentally relevant pharmaceuticals and one metabolite from the EU Watch List 2020/1161 was examined in ultrapure water and simulated wastewater, achieving excellent removal efficiencies (overall >95%) after 180 min treatment. The reactor unit was linked to an online LC sample manager, allowing for automated sampling every 15 min and near real-time process monitoring. Online comprehensive two-dimensional liquid chromatography (LC × LC) coupled with high resolution mass spectrometry (HRMS) was subsequently used to tentatively identify degradation products after photoelectrochemical degradation. Two reversed-phase liquid chromatography (RPLC) columns were used: an SB-C18 column operated with 5 mM ammonium formate at pH 5.8 ( 1 A) and methanol ( 1 B) as the mobile phases in the first dimension and an SB-Aq column using acidified water at pH 3.1 ( 2 A) and acetonitrile ( 2 B) as the mobile phases in the second dimension. This resulted in a five-fold increase in peak capacity compared to one-dimensional LC while maintaining the same total analysis time of 50 min. The LC x LC method allowed the tentative identification of 12 venlafaxine, 7 trimethoprim and 10 ciprofloxacin intermediates. Subsequent toxicity predictions suggested that some of these byproducts were potentially harmful. This study presents an effective hybrid technology for the simultaneous removal of pharmaceuticals from contaminated wastewater matrices and demonstrates how multidimensional liquid chromatography techniques can be applied to better understand the degradation mechanisms after the treatment of micropollutants with AOPs.

Abstract:This study focused on the Analytical Quality by Design (AQbD) optimization of the chromatographic separation and mass spectrometric detection of a wide group of structurally heterogeneous model pharmaceutical compounds (PhCs) and transformation products (TPs), chosen to cover the challenging issues of the co-presence of compounds characterized by (i) a wide range of physicochemical properties, (ii) the same mass transitions, and (iii) different ionisation modes. Italian consumption of PhCs were also considered as election criteria of target analytes. Octadecyl and pentafluorophenyl stationary phases, acetonitrile/methanol ratios and acidity of the eluents, column temperature, initial organic phase percentage, and elution gradient were investigated by AQbD, aiming at optimizing critical resolutions, sensitivities, and analysis time. Statistically significant models were obtained in most cases with fitting and cross-validation coefficients in the ranges of 0.681-0.998 and 0.514-0.967, respectively. After optimization, the analysis of target analytes was performed in a single chromatographic run, adopting a mixed acquisition mode based on scheduled acquisition windows comprising both single polarity and continuous polarity switching. For most investigated analytes the method provided detection limits in the sub-ng/L to low ng/L range, meeting for macrolides the sensitivity requested by the "Watch List" 2018/840/EU. The optimized method was applied to the direct injection analysis of PhCs and TPs in four wastewater treatment plant (WWTP) effluents and surface water (SW) samples collected in the receiving water bodies. Absolute values of matrix effect were found to be far higher than 20% for most target analytes in most samples. Seventeen PhCs and two TPs were quantified in at least one sample, at the wide concentration range of about 1-3200 ng/L. The most occurring PhCs in both WWTP effluents and SWs were levofloxacin (202-1239 and 100-830 ng/L), furosemide (865-3234 and 230-880 ng/L), ketoprofen (295-1104 and 270-490 ng/L), and ibuprofen (886-3232 and 690-1440 ng/L).

Abstract:New analytical methods are needed to efficiently measure the growing list of priority pharmaceuticals in environmental samples. In this regard, a rapid, sensitive, and robust method was developed for quantitation of 168 pharmaceuticals and pharmaceutical metabolites using solid-phase extraction (SPE) and liquid chromatography with tandem mass spectrometry. The extraction protocol and instrumental efficiency were specifically addressed to increase analytical workload and throughput. The optimized protocols, which are five times more efficient than US EPA Method 1694, enabled analyte recoveries that ranged from 77% to 117% for 162 analytes with method quantitation limits (MQLs) as low as 0.1 ng L-1 . To verify the suitability of the improved analytical method for environmental samples, 24-h composite samples of raw wastewater and wastewater effluent, along with downstream surface water, were analyzed. Overall, 143/168 target compounds were identified in at least one of the samples, and 130/168 analytes were present at concentrations above their MQLs. The total mass concentration of the measured analytes decreased by 93% during wastewater treatment. The analyte concentrations in the wastewater effluent were comparable to those measured in surface water 1 km downstream of the wastewater discharge point. Ultimately, the comprehensive method will serve as an important tool to inform the occurrence, fate, transport, and toxicity of a large suite of priority pharmaceuticals and pharmaceutical metabolites in natural and engineered systems. Unraveling the potential of a combined nitritation-anammox biomass towards the biodegradation of pharmaceutically active compounds

Abstract:In the past few years, anaerobic ammonium oxidation-based processes have attracted a lot of attention for their implementation at the mainstream line of wastewater treatment plants, due to the possibility of leading to energy autarky if combined with anaerobic digestion. However, little is known about the potential degradation of micropollutants by the microbial groups responsible of these processes and the few results available are inconclusive. This study aimed to assess the degradation capability of biomass withdrawn from a combined nitritation/anaerobic ammonium oxidation (combined N/A) pilot plant towards five pharmaceutically active compounds (ibuprofen, sulfamethoxazole, metoprolol, venlafaxine and carbamazepine). Batch experiments were performed under different conditions by selectively activating or inhibiting different microbial groups: i) regular combined N/A operation, ii) aerobic (optimal for nitrifying bacteria), iii) aerobic with allylthiourea (an inhibitor of ammonia monooxygenase, enzyme of ammonia oxidizing bacteria), iv) anoxic (optimal for anaerobic ammonium oxidizing bacteria), v) aerobic with acetate (optimal for heterotrophic bacteria) and vi) anoxic with acetate (optimal for heterotrophic denitrifying bacteria). Ibuprofen was the most biodegradable compound being significantly degraded (49-100%) under any condition except heterotrophic denitrification. Sulfamethoxazole, exhibited the highest removal (70%) under optimal conditions for nitrifying bacteria but in the rest of the experiments anoxic conditions were found to be slightly more favorable (up to 58%). For metoprolol the highest performance was obtained under anoxic conditions favoring anammox bacteria (62%). Finally, carbamazepine and venlafaxine were hardly removed (≤10% in the majority of cases). Taken together, these results suggest the specificity of different microbial groups that in combination with alternating operational parameters can lead to enhanced removal of some micropollutants.

Abstract:Background The introduction of white analytical chemistry encourages the development of methods characterized by a balance among greenness, productivity/feasibility and analytical performances. In the environmental analysis of emerging contaminants (ECs), for which high sensitivity and specificity are mandatory, the use of green and sustainable sample preparation needs to be coupled to a reliable analytical determination. Herein, an extraction method based on the use of a biodegradable polymeric film (Mater-Bi) and coupled to LC-MS/MS analysis was developed for the sensitive determination of ECs in wastewater. Results The interaction among a range of ECs and the Mater-Bi film (a commercially available patented blend of polybutylene-terephthalate, starch and fatty acids) was investigated by two sequential experimental designs, to simultaneously study several factors and optimize extraction efficiency. The final method, resembling a fabric phase sorptive extraction, involved pH and ionic strength modification of the sample, 1h extraction and desorption in ethanol. Satisfactory recoveries from real wastewater were obtained for sixteen analytes (56-116 %), as well as excellent precision (inter-day relative standard deviations below 10% for most compounds). Matrix effect was in the range 88-116% at the lower pre-concentration factor, but also acceptable in most cases at the higher pre-concentration factor. LODs in matrix, from 0.004 to 0.159 μg L -1 , were lower than or comparable to those from recent studies employing green extraction procedures. The method demonstrated its applicability to samples from wastewater treatment plants, allowing quantification of pharmaceuticals and UV filters at the μg L -1 and ng L -1 levels, respectively. Significance For the first time, the synthetic biopolymer Mater-Bi, so far unexplored for the use in analytical chemistry, was exploited for a green, simple and extremely cheap extraction protocol. The optimized method is suitable for several ECs, guaranteeing very good accuracy, precision and specificity, also thanks to the LC-MS/MS analysis. The evaluation by green and white analytical chemistry metrics highlighted its superiority to conventional extraction methods.

Abstract:The wide diffusion of Emerging Organic Micropollutants (EOMs) in the environment is receiving increasing attention due to their potential toxicological effects on living organisms. So far, the Wastewater Treatment Plants (WWTPs) have not been designed with the purpose to remove these contaminants; therefore, they can represent the major source of release into the environment both through the effluent and the wasted sludge. The fate of EOMs in the WWTPs is still not completely known; further investigations are therefore needed to assess if it is possible to exploit the existing treatment units to reduce EOM concentrations or which processes must be implemented to this purpose. Among the wide class of EOMs, the present study focused on the following drugs of abuse: amphetamine (AM), methamphetamine (MET), 11-nor-Δ9-THC-9carboxy (THC-COOH) and benzoylecgonine (BEG). Presence and removal efficiency of these drugs in the activated sludge tank of a WWTP for domestic sewage was investigated through analyses at both full-scale and laboratory scale. Determinations conducted in the full-scale WWTP highlighted that, among the searched drugs, AM was found to be the most abundant in the influent and effluent of the biological oxidation tank, while 11-nor-Δ9-THC-9carboxy was present at the lowest concentration. Some removal took place in the units prior to the oxidation tank, although the main reduction was observed to occur in the biological oxidation reactor. All the drugs showed a wide variability of the measured concentrations during the week and the day. Taking into account results from both full-scale observations and batch tests, removals in the biological reactor were found within the following ranges: 33–84% for AM, 33–97% for MET, 33–57% for BEG and 29–83% for THC-COOH. These removals were due to a combination of adsorption and biodegradation mainly, while volatilization did not play a significant role. Other processes, e.g. hydrolysis, were likely to occur.

Abstract:An innovative floating photocatalyst, based on BiOBr grown on LECA, is fabricated and tested in the degradation of two common non‐steroidal anti‐inflammatory drugs, diclofenac, and ibuprofen, under solar light irradiation in different conditions. The very promising results pave the way for BiOBr/LECA as an alternative to traditional materials for the effective removal of pharmaceutical drugs from water. Freshwater represents one of the most precious resources on the planet, so it is fundamental to preserve it. In this work, an innovative sunlight‐driven device composed of bismuth oxybromide (BiOBr) grown on a material derived from natural sources, i.e., Lightweight Expanded Clay Aggregates (LECA), is developed to clean surface waters under natural solar light irradiation. For this purpose, the photodegradation of two non‐steroidal anti‐inflammatory drugs, ibuprofen, and diclofenac, is investigated under varying operative conditions. Laboratory‐ and real‐scale experiments reveal that the fabricated floating BiOBr/LECA photocatalyst fully degrades diclofenac, whereas limited abatement of ibuprofen is observed. Based on the identification of specific transformation products (TPs) during the degradation, this behavior seems to be strongly related to the different structures of the two drugs. In fact, the main TP produced during diclofenac degradation derives from dechlorination and ring condensation: this type of photocatalytic degradation pathway is generally favored over the C─C bonds's cleavage, which is a unique possibility for IBU abatement. Moreover, the potential partial adsorption of these species on the photocatalyst's active sites can cause their deactivation. Finally, reusability tests demonstrate the high stability of the floating composite.

Abstract:The reuse of the same chromatography resin to purify multiple products is of high economic and ecologic interest in the pharmaceutical industry. An effective cleaning program to ensure no product carryover from one product to the subsequent is key to enable a multi-product use strategy. The methods utilised to assess product carryover during resin lifetime studies for a given product are not specific enough to detect carryover to support multi-product use. Here the proposed use of liquid chromatography coupled to mass spectrometry (LC-MS) is the analytical method of choice due to its high sensitivity and selectivity. A proof-of-concept study using protein A resin and two distinct IgG product streams produced by CHO cell lines was conducted. The experimental design was developed to determine any carryover from previous products after a multiple product Protein A resin reuse process, using LC-MS and capillary electrophoresis (CE). The capability of LC-MS coupled with bioinformatic analysis to detect carryover where CE with UV detection was not able to detect any carryover was demonstrated. The findings from this approach suggest that the acceptance of chromatographic resin reuse for multiple products will be dependent on the development of analytical/bioinformatics tools such as those proposed in this work.

Abstract:Recycling and recovering valuable resources from wastewater treatment plants is an important aspect in circular economy. The safe use of sludge and sludge-related products deriving from wastewater treatment strictly depends on their chemical contamination, especially by emerging pollutants. In this work, an analytical method was developed for the determination of a range of selected compounds, included in a recent European watch-list (macrolides, fluoroquinolones, neonicotinoids, carbamates and estrogens), in recovery materials from innovative pilot systems. Both the instrumental analysis by high-performance liquid chromatography-tandem mass spectrometry and the pre-treatment strategy (quick, easy, cheap, effective, rugged and safe technique) were optimized for the purpose. The final method performance were evaluated, revealing determination coefficients (R2) of 0.993–0.9999 for the matrix-matched calibration curves, good accuracy (recovery 68–104% and matrix effect 70–123%), satisfactory precision (relative standard deviation <20%) and limits of detection and quantitation in the low ng g−1 levels. Ten different recovery material samples were analyzed, showing contamination by few analytes, mainly antibiotics and estrone; ciprofloxacin and azithromycin were the most abundant compounds (up to 500–600 ng g−1). On the contrary, neonicotinoid pesticides were not detected, except for one sample (sample 10, the only compost material).The application of the described method is an essential part of a broader investigation on the suitability and safety of innovative materials coming from waste water treatment plants, in the view of a risk assessment related to their usage.

Abstract:A new analytical method utilizing ultra-performance liquid chromatography (UPLC) tandem mass spectrometry (MS/MS) has been developed to determine 16 pharmaceuticals from 8 therapeutic classes in wastewater: bezafibrate, clofibric acid, carbamazepine, caffeine, chloramphenicol, diclofenac, gemfibrozil, indomethacin, ketoprofen, mefenamic acid, metoprolol, nalidixic acid, N,N-diethyl-meta- toluamide, propranolol, sulpiride and trimethoprim. Key parameters of MS/MS, UPLC and solid phase extraction (SPE) were optimized. In general, recovery of target pharmaceuticals was over 70% for the wastewater effluent samples and 50% for the influent samples. The effects of matrix suppression, loss during the pretreatment as well as instrument variability were successfully corrected by two internal standards, and acceptable relative recovery was obtained. Target compounds were quantitatively analyzed using multiple reaction monitoring (MRM) mode, and the detection limits ranged from 0.3 to 20 ng/L. A detailed study, matrix effect in effluent wastewater was also present. The method was applied to detecting pharmaceuticals in the wastewater from three wastewater treatment plants (WWTPs) in Beijing, China and the results demonstrated that most target compounds were detectable in both the influent and effluent, with the mean concentrations ranging from 20.5 to 5775.6 ng/L and 4.6 to 418.6 ng/L, respectively.

Abstract:Ecological harm and human health risks caused by environmental pollution with active pharmaceutical ingredients (API) nowadays is recognised as issue of growing concern. Widespread presence of human and veterinary API in aquatic environment clearly indicates persistence and low removal efficiency of these compounds at conventional pharmaceutical and municipal wastewater treatment plants (WWTP). Bioaugmentation of activated sludge systems with specialized microorganisms could be a powerful and environmentally friendly tool to enhance the removal efficiency of recalcitrant API. Selection of inoculum strains, that have appropriate enzymatic pathways to metabolise complex molecules of API, belonging to different therapeutic classes, is of great importance. This study evaluated the potential of pure cultures of 10 bacteria, 10 yeasts and 3 filamentous fungi previously isolated from activated sludge of pharmaceutical WWTP to degrade less investigated API – Oxytocin, Zopiclone and Meldonium dihydrate as sole carbon source and in cometabolic manner with presence of skim milk powder as additional nutrient source. Bacteria Sphingobacterium thalpophilum and filamentous fungi Fusarium solani and Fusarium udum showed very high treatment efficiency of all tested API in laboratory-scale bioaugmentation tests and were recognized as culture with high metabolic potential to be used in bioaugmentation for removal of pharmaceutical micropollutants.

Developing the OECD 106 fate testing protocol for active pharmaceuticals in soil

Abstract:The ability to determine accurately the fate of APIs in soil is essential for rigorous risk assessment associated with wastewater reuse or biosolid recycling to land, particularly in lower income countries where water and fertiliser is scarce. Four APIs (naproxen, ofloxacin, propranolol and nevirapine) with wide ranging functionality were used as examples in the development of the OECD 106 soil partitioning and/or degradation study, with naproxen used to illustrate applying the full methodology. The data showed key methodological criteria require careful consideration and testing to generate accurate and consistent results. Only glass fibre membranes were suitable for all APIs, without unduly adsorbing APIs to their surface, thus effectively restricting the minimum practical pore size to 0.7 µm. Polypropylene plastic centrifuge tubes were shown to be suitable, with careful determination of recoveries. Direct injection liquid chromatography-mass spectrometry could reliably resolve all 4 APIs down to less than µg L-1 in soil solutions, although allowance for matrix effects via standard additions was required in some cases. Greatest analytical challenges were found for the highest molecular weight API with the greatest affinity for sorption to surfaces (ofloxacin). Key variables that can impact on partitioning such as solution pH and dissolved organic carbon concentrations were shown to vary within tests over time and should be accounted for.

Abstract:This study reviews the efficiencies of various biological treatment systems for pharmaceutical removal from wastewater, highlighting the need for optimized parameters and tailored approaches to mitigate ecological and health risks, emphasizing ongoing innovation for sustainable solutions. This review presents a comprehensive analysis of current research on biological treatment processes for removing pharmaceutical compounds (PhCs) from wastewater. Unlike previous studies on this topic, our study specifically delves into the effectiveness and drawbacks of various treatment approaches such as traditional wastewater treatment facilities (WWTP), membrane bioreactors (MBRs), constructed wetlands (CW), and moving bed biofilm reactors (MBBR). Through the examination and synthesis of information gathered from more than 200 research studies, we have created a comprehensive database that delves into the effectiveness of eliminating 19 particular PhCs, including commonly studied compounds such as acetaminophen, ibuprofen, diclofenac, naproxen, ketoprofen, indomethacin, salicylic acid, codeine, and fenoprofen, amoxicillin, azithromycin, ciprofloxacin, ofloxacin, tetracycline, atenolol, propranolol, and metoprolol. This resource provides a depth and scope of information that was previously lacking in this area of study. Notably, among these pharmaceuticals, azithromycin demonstrated the highest removal rates across all examined treatment systems, with the exception of WWTPs, while carbamazepine consistently exhibited the lowest removal efficiencies across various systems. The analysis showcases the diverse results in removal efficiency impacted by factors such as system configuration, operation specifics, and environmental circumstances. The findings emphasize the critical need for continued innovation and research, specifically recommending the integration of advanced oxidation processes (AOPs) with existing biological treatment methods to improve the breakdown of recalcitrant compounds like carbamazepine. Enhancing HRT and SRT improves removal but adds complexity and costs. Tailored treatment approaches needed based on contaminants and conditions.

Abstract:The occurrence of emerging pollutants (EPs) such as pharmaceuticals and personal care products (PPCPs) has raised serious concerns about the possible adverse effects on ecosystem integrity and human health. Wastewater treatment facilities appear to be the main sources of PPCPs released in aquatic environments. This research examines the effectiveness of groundwater microbial community activities to remove phenoxyethanol (Phy-Et), currently exploited as a preservative in many cosmetic formulations at a maximum concentration of 1% but which has shown, at higher levels of exposure, adverse systemic effects on animals. Mesocosm experiments were carried out for 28 days using two different concentrations of the substance (5.2 mg/L and 27.4 mg/L). The main results obtained through chemical and microbiological investigations revealed a significant Phy-Et reduction (≈100% when added at a concentration of 5.2 mg/L and ≈84% when added at a concentration of 27.4 mg/L), demonstrating that some autochthonous microorganisms in the analyzed samples played a "key role" in removing this compound, despite its proven antimicrobial activity. Nevertheless, the decrease in the "natural attenuation" efficacy (≈16%) when using higher concentrations of the chemical suggests the existence of a "dose-dependent effect" of Phy-Et on the process of biodegradation. Biomolecular investigations carried out through next-generation sequencing (NGS) revealed (i) the presence of a significant fraction of hidden microbial diversity to unravel, (ii) variations of the composition and species abundance of the groundwater microbial communities induced by Phy-Et, and (iii) a biodiversity reduction trend correlated to the increase of Phy-Et concentrations. Overall, the preliminary information obtained from the experiments carried out at the laboratory scale appears encouraging, although it reflects only partially the complexity of the phenomena that occur in natural environments and influences their "auto-purification capability". Accordingly, this research paves the way for more in-depth investigations to develop appropriate tools and protocols to evaluate the occurrence and fate of Phy-Et in nature and assess the impact of its release and the effects of long-term exposure (even at low concentrations) on ecosystems and health.
