Abstract:More recent evidences are supportive of air pollution exposure on diabetes risk, including worsening of whole-body insulin sensitivity, enhancement of hepatic lipogenesis and nonalcoholic fatty liver disease after fine particulate matter (PM2.5) exposure. Therefore, we aimed to explore the lipidomics to get a comprehensive insight about ambient real-world PM2.5 exposure on lipid metabolism in blood and liver. After ambient PM2.5 exposure for 6 months, excess triglyceride accumulation in the liver was observed. Remarkable metabolic alterations including neutral lipids, glycerophospholipids and sphingolipids were noticed. Lipidomic signatures in liver is different from plasma in response to PM2.5 exposure. Lipids including species of ceramide, sphingomyeline and triglyceride may become potential biomarkers of lipotoxicity contributing to PM2.5-induced metabolic dysfunction, and the present study may serve as a reference lipid bank for further studies.

Abstract:Emerging evidence has demonstrated that exposure to fine particulate matter (PM2.5) is a risk factor for lipid metabolic disorders in the liver. However, the effects of PM2.5 exposure time duration on hepatic lipid metabolism remain unknown. In this study, C57BL/6 mice were randomly divided into ambient PM2.5 (PM) or filtered air (FA) exposure chamber for short-term (4 weeks) or long-term (24 weeks) exposure via a whole body exposure system. We measured hepatic triglyceride and free fatty acid levels and analyzed the alteration of lipometabolism-related molecules in the liver. We found that triglyceride levels were significantly elevated in both short-term and long-term PM2.5-exposed mice and free fatty acid levels were increased after long-term PM2.5 exposure. Besides, enzymes for lipolysis and fatty acid oxidation in the liver were inhibited after short-term PM2.5 exposure but adaptively enhanced after long-term PM2.5 exposure. Furthermore, molecules for fatty acid uptake were down-regulated in the short-term PM2.5-exposed mice whereas molecules for lipid export were induced after long-term PM2.5 exposure. Therefore, ambient PM2.5 exposure disturbed hepatic lipid metabolism and the effects varied in different exposure duration. These findings in mice provide new insight into the biological basis of PM2.5-induced human metabolic dysfunction and specific strategies may be applied based on different exposure time periods.

Abstract:Background & aims Emerging evidence supports ambient fine particulate matter (PM 2.5 ) exposure is associated with insulin resistance (IR) and hepatic lipid accumulation. In this study, we aimed to evaluate the sex-dependent vulnerability in response to PM 2.5 exposure and investigate the underlying mechanism by which PM 2.5 modulates hepatic lipid metabolism. Methods Both male and female C57BL/6 mice were randomly assigned to ambient PM 2.5 or filtered air for 24 weeks via a whole body exposure system. High-coverage quantitative lipidomics approaches and liquid chromatography-mass spectrometry techniques were performed to measure hepatic metabolites and hormones in plasma. Metabolic studies, histological analyses, as well as gene expression levels and molecular signal transduction analysis were applied to examine the effects and mechanisms by which PM 2.5 exposure-induced metabolic disorder. Results Female mice were more susceptible than their male counterparts to ambient PM 2.5 exposure-induced IR and hepatic lipid accumulation. The hepatic lipid profile was changed in response to ambient PM 2.5 exposure. Levels of hepatic triacylglycerols (TAGs), free fatty acids (FFAs) and cholesterol were only increased in female mice from PM group compared to control group. Plasmalogens were dysregulated in the liver from PM 2.5 -exposed mice as well. In addition, exposure to PM 2.5 led to enhanced hepatic ApoB and microsomal triglyceride transport protein expression in female mice. Finally, PM 2.5 exposure inhibited hypothalamus-pituitary-adrenal (HPA) axis and decreased glucocorticoids levels, which may contribute to the vulnerability in PM 2.5 -induced metabolic dysfunction. Conclusions Ambient PM 2.5 exposure inhibited HPA axis and demonstrated sex-associated differences in its effects on IR and disorder of hepatic lipid metabolism. These findings provide new mechanistic evidence of hormone regulation in air pollution-mediated metabolic abnormalities of lipids and more personalized care should be considered in terms of sex-specific risk factors.

Abstract:Emerging evidence has shown that exposure to ambient fine particulate matter (PM2.5) is associated with hepatic lipid accumulation. However, the underlying mechanism is not fully characterized yet. Autonomous circadian clock in the liver plays a fundamental role in maintaining lipid metabolism homeostasis. In this study, we evaluated the effects of ambient PM2.5 exposure on the expression of hepatic circadian clock genes and expression rhythm of genes associated with lipid metabolism in mice liver. Male C57BL/6 mice were randomly assigned to ambient PM2.5 or filtered air for 10 weeks via a whole body exposure system. We found that the liver mass was reduced significantly at zeitgeber time (ZT) 8 in mice exposed to PM2.5 but not levels or circadian rhythm of hepatic triglycerides or free fatty acid (FFA). In addition, exposure to PM2.5 led to enhanced expression of bmal1 at ZT0/24, cry1 at ZT16 and rev-erbα at ZT4 and ZT8. Furthermore, the expression of pparα was enhanced in mice liver at ZT4 and ZT8 after PM2.5 exposure, with upregulation of pparα-mediated genes responsible for fatty acid transport and oxidation. Finally, the expression of rate-limiting enzymes for lipid synthesis was all significantly increased in the liver of PM2.5 exposed mice at ZT12. Therefore, the present study provides new perspectives for revealing the etiology of hepatic lipid metabolism abnormality from PM2.5-induced circadian rhythm disorder.
Airborne PM2.5 Pollution: A Double-Edged Sword Modulating Hepatic Lipid Metabolism in Middle-Aged Male Mice

Abstract:Emerging evidence suggests that exposure to airborne fine particulate matter (PM2.5) is closely related to disturbances in hepatic lipid metabolism. However, no systematic study assessed the age vulnerability in effects of PM2.5 exposure on metabolism, and the potential mechanisms remain unknown. This study aimed to investigate the metabolic susceptibility of different life stages to PM2.5 exposure, and to evaluate the underlying molecular mechanisms. Male C57BL/6 mice at three life phases (young, adult, and middle-aged) were exposed simultaneously to concentrated ambient PM2.5 or filtered air (FA) for 8 weeks using a whole-body inhalational exposure system. The average daily PM2.5 concentrations to which mice were actually exposed were 90.71 ± 7.99 μg/m3. The body weight, total food utilization, body composition, glucose metabolic homeostasis of the mice were evaluated. At euthanasia, serum and liver samples were collected to measure lipid profiles and hepatic function. H&E and Oil Red O staining were used to assess the liver cellular structure and hepatic lipid deposition. Transcriptomics and lipidomics were performed to determine the differentially expressed genes and lipid metabolites in the liver. Quantitative RT-PCR and immunoblots were performed to verify the transcriptomics and explore the mechanism for metabolic susceptibility. PM2.5 exposure led to reductions in body weight gain, total food utilization, and fat mass in middle-aged mice but not in young or adults. Exposure to PM2.5 reduced hepatic lipid deposition by enhancing lipolysis and inhibiting the glycerol-3-phosphate (G3P) pathway of hepatic lipogenesis. Furthermore, PM2.5 exposure attenuated hepatic fatty acid metabolism and primary bile acid biosynthesis. Finally, PM2.5 exposure dysregulated hepatic phospholipid metabolism, as evidenced by increased glycerophospholipid synthesis and disturbed sphingolipid metabolism. Therefore, middle-aged male mice were more vulnerable to PM2.5 exposure with double-edged effects, improved metabolism and hepatic TG accumulation but inhibited hepatic fatty acid and bile acid metabolism and dysregulated phospholipid metabolism.

Abstract:Microplastics (MPs), the emerging environmental pollutants, have attracted global attention due to the potential public health challenge and ecological security risk. Recent studies suggested liver as a vulnerable organ to MPs exposure, evidenced by abnormal hepatic lipid metabolism upon MPs intake in multiple animal species. How-ever, the specific changes of lipid metabolism in mammalian livers, as well as the underlying mechanisms, remain to be elucidated. In the present study, C57BL/6 mice were randomly assigned to normal drinking water or drinking water containing 100 mu g L-1 or 1000 mu g L-1 polystyrene (PS) MPs for 8 weeks. MPs exposure exerted no significant effect on body weight, serum triglyceride or total cholesteryl esters. However, mice showed impaired glucose tolerance and hepatic lipid deposition in response to high-dose MPs administration. Further lipidomic analysis showed significant alteration in hepatic lipid species particularly with free fatty acids (FFAs) and tri-acylglycerols (TAGs) in mice exposed to MPs. Meanwhile, the liver transcriptional profile indicated MPs exposure-induced differentially expressed genes (DEGs) were enriched in pathways of lipid metabolism and unfolded protein response. Furthermore, most altered lipid species were significantly correlated with DEGs enriched in lipid metabolic signaling. These findings provide lipidomic and transcriptional signatures of liver in response to MPs exposure, which will shed light on further understanding of the metabolic toxicity of MPs.

Abstract:Particulate matter (PM) is a huge environmental threat and is of major public concern. Oxidative stress and systemic inflammation are known factors that contribute to PM- related damage; however, a systematic understanding of the deleterious pulmonary effects of PM using multi-omics analysis is lacking. In this study, we performed transcriptomic, proteomic, and metabolomic analyses in a mouse model exposed to PM for three months to identify molecular changes in lung tissues. We identified 1690 genes, 326 proteins, and 67 metabolites exhibiting significant differences between PM-challenged and control mice (p < 0.05). Differentially expressed genes and proteins regulated in PM-challenged mice were involved in lipid metabolism and in the immune and inflammatory response processes. Moreover, a comprehensive analysis of transcript, protein, and metabolite datasets revealed that the genes, proteins, and metabolites in the PM-treated group were involved in lysosomal function and lipid metabolism. Specifically, Cathepsin D (Ctsd), Ferritin light chain (Ftl), Lactotransferrin (Ltf), Lipocalin 2 (Lcn2), and Prosaposin (Psap) were major proteins/genes associated with PM-induced pulmonary damage, while two lipid molecules PC (18:1(11Z)/16:0) and PA (16:0/18:1(11Z)) were major metabolites related to PM-induced pulmonary injury. In summary, lipid metabolism might be used as successful precautions and therapeutic targets in PM-induced pulmonary injury to maintain the stability of cellular lysosomal function.
Ambient PM2.5 induces brain injury through PI3K/AKT/FoxO1 pathway

Abstract:Background Fine particulate matter (PM 2.5 )-induced neurological and mental diseases, such as cognitive impairment and stroke, tend to cause disability. In this study, we plan to find the appropriate biological indicators reflecting the early effects of PM 2.5 exposure and explore the toxicity of inhaling PM 2.5 along with its pathological mechanism on nervous system. Methods Male C57BL/6 mice were divided into 6 groups and exposed to concentrated PM 2.5 or filtered air for 2, 4 and 6 months, respectively. Their performances of ethology were tested by Morris water maze, elevated plus maze and buried food pellet test. Weight of whole brain and olfactory bulbs were recorded when the mice were sacrificed and the brain structure was observed by HE staining. ROS, SOD, IL-6, TNF-α, IL-1β, BDNF, β-hydroxybutyric, IgG, albumin, fibrinogen and plasminogen were tested. The mRNA expressions of the cortex were detected by RNA sequencing and real-time PCR, and the corresponding proteins were detected by Western blot. Results The spatial learning memory ability were impaired and the mice performed anxiously after PM 2.5 exposure. Relative brain weight decreased along with age and PM 2.5 inhaled exposure exceeded the process. But neuronal morphology, inflammatory cytokines, ROS, β-hydroxybutyric, BDNF, IgG and fibrinogen did not change. Intriguingly, SOD and albumin decreased, while plasminogen increased after PM 2.5 exposure. PI3K-AKT-FoxO1 pathway was activated after 6-month PM 2.5 exposure. Conclusion Albumin, A/G value and plasminogen are main serum indicators of early-stage (2 month) PM 2.5 exposure, and long-term (6 month) PM 2.5 exposure induced brain injury potentially through the activation of PI3K/AKT/FoxO1 pathway.

Abstract:PM 2.5 can cause adverse health effects via several pathways, such as inducing pulmonary and systemic inflammation, penetration into circulation, and activation of the autonomic nervous system. In particular, the impact of PM 2.5 exposure on the liver, which plays an important role in metabolism and detoxification to maintain internal environment homeostasis, is getting more attention in recent years. In the present study, C57BL/6J mice were randomly assigned and treated with PM 2.5 suspension and PBS solution for 8 weeks. Then, hepatic tissue was prepared and identified by metabolomics analysis and transcriptomics analysis. PM 2.5 exposure can cause extensive metabolic disturbances, particularly in lipid and amino acids metabolic dysregulation.128 differential expression metabolites (DEMs) and 502 differently expressed genes (DEGs) between the PM 2.5 exposure group and control group were detected. The Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses showed that DEGs were significantly enriched in two disease pathways, non-alcoholic fatty liver disease (NAFLD) and type II diabetes mellitus (T2DM), and three signaling pathways, which are TGF-beta signaling, AMPK signaling, and mTOR signaling. Besides, further detection of acylcarnitine levels revealed accumulation in liver tissue, which caused restricted lipid consumption. Furthermore, lipid droplet accumulation in the liver was confirmed by Oil Red O staining, suggesting hepatic steatosis. Moreover, the aberrant expression of three key transcription factors revealed the potential regulatory effects in lipid metabolic disorders, the peroxisomal proliferative agent-activated receptors (PPARs) including PPARα and PPARγ is inhibited, and the activated sterol regulator-binding protein 1 (SREBP1) is overexpressed. Our results provide a novel molecular and genetic basis for a better understanding of the mechanisms of PM 2.5 exposure-induced hepatic metabolic diseases, especially in lipid metabolism.

Abstract:PM2.5-related neurological and mental diseases, such as cognitive impairment and stroke, tend to cause disability. Six-week-old male C57BL/6 mice were divided into 6 groups and exposed to concentrated PM2.5 or filtered air for 2, 4, and 6 months, respectively. The neurobehavioral changes of mice were tested. The weight of the whole brain and olfactory bulbs were recorded at the end of exposure, and the brain structure was observed by hematoxylin and eosin (HE) staining. Serum indicators, mRNA, and protein expressions were detected. The spatial learning memory ability was impaired, and the mice were more anxious after PM2.5 exposure. Relative brain weight decreased with age, and PM2.5 exposure exceeded the decrease of relative brain weight. Interestingly, superoxide dismutase (SOD) and albumin decreased in the PM2.5-exposed groups although neuronal morphology and other serum indicators did not show significant difference between PM and FA groups. Moreover, PM2.5 induced the increase of plasminogen at 2 months but recovered at 4 months and then increased at 6 months again. The results from protein expression and transcriptomic test demonstrated that PI3K/AKT/FoxO1 pathway might be activated after 6-month PM2.5 exposure in mice. Indicators albumin, the percentage of albumin over IgG (A/G value), and plasminogen were the main serous changes in mice after early-stage (2 months) and long-term (6 months) PM2.5 exposure. In addition, early-stage injury induced by PM2.5 might recover at later time point and display significant injury again with the exposure time. PM2.5 exposure-induced brain injury might be associated with the activation of PI3K/AKT/FoxO1 pathway.

Abstract:Chronic ambient fine particulate matter (PM2.5) exposure correlates with various adverse health outcomes. Its impact on the circulating metabolome-a comprehensive functional readout of the interaction between an organism's genome and environment-has not however been fully understood. This study thus performed metabolomics analyses using a chronic PM2.5 exposure mouse model. C57Bl/6J mice (female) were subjected to inhalational concentrated ambient PM2.5 (CAP) or filtered air (FA) exposure for 10 months. Their sera were then analyzed by liquid chromatography-mass spectrometry (LC-MS) and gas chromatography-mass spectrometry (GC-MS). These analyses identified 2570 metabolites in total, and 148 of them were significantly different between FA- and CAP-exposed mice. The orthogonal partial least-squares discriminant analysis (OPLS-DA) and heatmap analyses displayed evident clustering of FA- and CAP-exposed samples. Pathway analyses identified 6 perturbed metabolic pathways related to amino acid metabolism. In contrast, biological characterization revealed that 71 differential metabolites were related to lipid metabolism. Furthermore, our results showed that CAP exposure increased stress hormone metabolites, 18-oxocortisol and 5a-tetrahydrocortisol, and altered the levels of circadian rhythm biomarkers including melatonin, retinal and 5-methoxytryptophol.

Abstract:The 13-valent pneumococcal conjugate (PCV13, Prevenar 13, Pfizer Inc., NY, USA) was developed as a successor to the currently registered 7-valent pneumo coccal conjugate vaccine (PCV7, which contains serotypes 4, 6B, 9V, 14, 18C, 19F and 23F; Prevnar/Prevenar, Pfizer Inc.), for use in infants and young children to prevent disease such as invasive pneumococcal disease (IPD), nonbacteremic pneumonia and acute otitis media (AOM), caused by the 13 pneumococcal serotypes contained in the vaccine. In addition to the serotypes in PCV7, PCV13 contains serotypes 1, 3, 5, 6A, 7F and 19A [1]. For PCV13, the global pediatric filings were initiated in late 2008 and, to date, regulatory applications for PCV13 have been submitted in more than 50 countries spanning six continents. The first country to approve PCV13 was Chile on 14th July 2009. PCV13 has been granted priority review in the USA, Canada, Australia and South Africa. In Europe, this vaccine received positive feedback from the Committee for Medicinal Products for Human Use (CHMP) on 24th September 2009 and received market authorization on 11th December 2009. Streptococcus pneumoniae is a major source of morbidity and mortality worldwide. It is estimated by the WHO that approximately one million children die of pneumococcal disease every year, mostly in developing countries [2]. Pneumococcal infections are the leading cause of death from a vaccine-preventable illness in children younger than 5 years of age [3,4]. Invasive diseases caused by pneumococci include meningitis, bacteremia, and pneumonia with bacteremia and/or empyema [5]. PCV7 was licensed in the USA in the year 2000 and in Europe in 2001. By December 2009, PCV7 was licensed in approximately 100 of 193 member states of the WHO. In addition, it had been included in the routine or national immunization programs or was widely used in 42 of these countries. The achievements of the PCV7 vaccination programs were recently reviewed in detail in this journal [6,7], and are therefore only briefly discussed here.
Illuminating a Time-Response Mechanism in Mice Liver after PM2.5 Exposure Using Metabolomics Analysis.

Abstract:PM2.5 is recognized as an atmospheric pollutant that seriously jeopardizes human health. Emerging evidence indicates that PM2.5 exposure is associated with metabolic disorders. Existing epidemiology and toxicology studies on the health effects of PM2.5 usually focused on its different components and doses, the effects on susceptible populations, or the effects of indoor and outdoor pollution. The underlying mechanisms of exposure time are poorly understood. Liver, as the central organ involved in various metabolisms, has special signaling pathways non-existed in lung and cardiovascular systems. Exacerbation in liver by the prolonged exposure of PM2.5 leads to hepatic function disorder. It is therefore essential to elucidate the mechanism underlying hepatotoxicity after PM2.5 exposure from the perspective of time-response relationship. In this study, targeted metabolomics was utilized to explore the hepatic injury in mice after PM2.5 exposure. Our results showed that prolonged exposure of PM2.5 would aggravate liver metabolic disorders. The metabolic process was divided into three phases. In phase I, it was found that PM2.5 exposure disturbed the hepatic urea synthesis. In phase II, oxidative damages and inflammations obviously occurred in liver, which would further cause neurobehavioral disorders and fat deposits. In phase III, the changes of metabolites and metabolic pathways indicated that the liver has been severely damaged, with the accelerated biosynthesis and fat metabolism. Finally, using ROC analysis coupled with their biological functions, 4 potential biomarkers were screened out, with which we established a method to classify and diagnose the progress of liver damage in mice after PM2.5 exposure. In this paper, we not only established the time-response relationship of PM2.5, but also provided new insights for the classification and prediction of the toxic injury stages in mice liver, which provides a ground work for the future drug intervention to prevent oxidative damage of PM2.5.
Atmospheric particulate matter impairs cognition by modulating synaptic function via the nose-to-brain route

Abstract:Atmospheric particulate matter (PM), a ubiquitous air pollutant, is the leading environmental risk factor for mortality worldwide. Experimental and epidemiological studies consistently suggest a strong link between long-term exposure to PM2.5 (<2.5 μm, fine PM) and cognitive impairment. The neuroinflammatory response is presumed to be one of the main mechanisms of PM2.5-induced cognitive impairment, possibly leading to synaptic dysfunction. However, the main route and mechanism underlying the cause of cognitive dysfunction and pathogenic alterations in PM2.5-exposure mice remain poorly understood. Therefore, this study aimed to investigate the main route and mechanism of PM2.5-induced cognitive impairment. Our results showed that PM2.5 directly entered the brain following nasal administration, and both the short-term PM2.5 administration via atomization and nasal drops induced learning and memory impairments and neuronal damage in adult mice. Moreover, astrocytes and microglia were both activated in the two short-term PM2.5 exposure models, while few changes in the inflammatory response were observed in the peripheral circulatory system. Furthermore, a further transcriptional analysis revealed that short-term PM2.5 administration led to cognitive impairment mainly by modulating synaptic functions and that although glia were activated, the glia-related pathological pathways were not significantly activated. Notably, following PM2.5 exposure, PLX3397-induced microglial deletion did not restore the cognitive function of the mice. In conclusion, our results provide evidence that PM2.5 enters the brain via the nose-to-brain route to impair cognitive function, and short-term exposure to PM2.5 directly alters synaptic function rather than the neuroinflammatory response to affect cognition.

Abstract:Abstract Background & aimsSome epidemiological investigations have revealed airborne fine particulate matter (PM 2.5 ) could induce adverse effects on respiratory system of human. However, the experimental evidences of the harmful effect of PM 2.5 from mid-scale city in China and associated molecular mechanisms was still scarce. In this study, we aimed to evaluate the adverse effect on the lung system of PM 2.5 collected from mid-city of China and elucidate the underlying molecular mechanism through mRNA-seq and microRNA-seq integrated analysis.MethodsWe exposed male Balb/C mice to PM 2.5 collected from Baoji city, China for 8 weeks (298.52 ± 25.86 μg/m 3 ) using a whole-body exposure system. Micro-CT and histopathological analyses were performed to determine the morphology and histopathology changes of lung tissues induced by PM 2.5 . Transcriptome (both mRNA and microRNA) sequencing and the immunohistochemistry assay were performed to reveal the associated underlying mechanisms. The contents of PAHs absorbed in the PM 2.5 , as well as the pearson correlation index between it and the target genes and microRNA were examined.ResultsObvious lung injury including pulmonary dysfunction, immunological responses, and fibrosis were observed from the PM 2.5 exposure group. The content of leucocyte of lung tissue was significantly increased. PM 2.5 mainly induced immune pathways including B cell receptor signaling and cell adhesion molecules (CAMs). The expression levels of the associated genes and microRNA were verified using RT-qPCR. The protein expression of CD19, CD81, and PIK3CD involved into B cell receptor signaling significantly increased in exposure group. The concentrations of benzo(a)pyrene (BaP) showed a marked correlationship with the genes and microRNA. ConclusionsPM 2.5 from the mid-scale city of China caused adverse effects on lung system of mice, which was much stronger than the mega city, indicating the health risk of the air pollution in the developing area should be paid more attention. The toxicity of PM 2.5 may associated with immune system response. This work for the first time provided a new sight into the molecular mechanism of PM 2.5 toxicity on lung system using transcriptomics integrated analysis and help further to develop effective measures to prevent air pollution associated lung disease, especially in the developing area in China.

Abstract:Many environmental toxicants can cause systemic effects, such as fine particulate matter (PM2.5), which can penetrate the respiratory barrier and induce effects in multiple tissues. Although metabolomics has been used to identify biomarkers for PM2.5, its multi-tissue toxicology has not yet been explored holistically. Our objective is to explore PM2.5 induced metabolic alterations and unveil the intra-tissue responses along with inter-tissue communicational effects. In this study, following a single intratracheal instillation of multiple doses (0, 25, and 150 μg as the control, low, and high dose), non-targeted metabolomics was employed to evaluate the metabolic impact of PM2.5 across multiple tissues. PM2.5 induced tissue-specific and dose-dependent disturbances of metabolites and their pathways. The remarkable increase of both intra- and inter-tissue correlations was observed, with emphasis on the metabolism connectivity among lung, spleen, and heart; the tissues' functional specificity has marked their toxic modes. Beyond the inter-status comparison of the metabolite fold-changes, the current correlation network built on intra-status can offer additional insights into how the multiple tissues and their metabolites coordinately change in response to external stimuli such as PM2.5 exposure.

Abstract:Ambient particulate matter (PM2.5) is a potential risk factor for metabolic damage to the liver. Epidemiological studies suggest that elevated PM2.5 concentrations cause changes in hepatic metabolism, but there is a lack of laboratory evidence. Here, we aimed to evaluate the effects of PM2.5 exposure on liver metabolism in C57BL/6j female mice (10 months old) and to explore the mechanisms underlying metabolic alterations and differential gene expressions by combining metabolomics and transcriptomics analyses. The metabolomics results showed that PM2.5 exposure notably affected the metabolism of amino acids and organic acids and caused hepatic lipid and bile acid accumulation. The transcriptomic analyses revealed that PM2.5 exposure led to a series of metabolic pathway abnormalities, including steroid biosynthesis, steroid hormone biosynthesis, primary bile acid biosynthesis, etc. Among them, the changes in the bile acid pathway might be one of the causes of liver damage in mice. In conclusion, this study clarified the changes in liver metabolism in mice caused by PM2.5 exposure through combined transcriptomic and metabolomic analyses, revealed that abnormal bile acid metabolism is the key regulatory mechanism leading to metabolic-associated fatty liver disease (MAFLD) in mice, and provided laboratory evidence for further clarifying the effects of PM2.5 on body metabolism.
Prenatal PM2.5 exposure impairs spatial learning and memory in male mice offspring: from transcriptional regulation to neuronal morphogenesis

Abstract:Background: As one of the environmental risk factors for human health, atmospheric fine particulate matter (PM2.5) contributes to cognitive deterioration in addition to respiratory and cardiovascular injuries. Recently, increasing evidence implicates that PM2.5 inhalation can affect neurological functions in offspring, but the sex-specific outcomes and the underlying biological processes are largely unknown. Objectives: To observe the influence of prenatal PM2.5 exposure on cognitive performance in offspring, to elucidate the neuronal morphological alterations and possible transcriptional regulation based on mRNA-sequencing (mRNA-Seq) data after birth, and to determine the key components of PM2.5 contributing to the adverse effects. Methods: Pregnant C57BL/6J mice were exposed to sterile saline or PM2.5 suspension. Morris water maze test was used to assess the cognitive function in weanling offspring. Microscopic observation was applied to detect neuronal morphogenesis in vivo and in vitro. The cortex tissues from male offspring were collected on postnatal days (PNDs) 1, 7, and 21 for mRNA-Seq analysis. The organic and inorganic components of PM2.5 were separated to assess their contributions using primary cultured neurons. Results: Prenatal PM2.5 exposure impaired spatial learning and memory in weanling male mice, but not female mice. The sex-specific outcomes were associated with mRNA expression profiles of the cortex during postnatal critical windows, and the annotations in Gene Ontology (GO) of differentially expressed genes (DEGs) revealed that the exposure persistently disrupted the expression of genes involved in neuronal features in male offspring. Consistently, axonal growth impairment and dendritic complexity reduction were observed. Importantly, Homeobox A5 (Hoxa5), a critical transcription factor regulating all of the neuronal morphogenesis-associated hub genes on PNDs 1, 7, and 21, significantly decreased in the cortex of male offspring following PM2.5 exposure. In addition, both inorganic and organic components were harmful to axonal and dendritic growth, with organic components exhibiting stronger inhibition than inorganic ones. Conclusion: Prenatal PM2.5 exposure affected spatial learning and memory in male mice by disrupting Hoxa5-mediated neuronal morphogenesis, and the organic components, including polycyclic aromatic hydrocarbons (PAHs), posed more adverse effects than the inorganic components.
