Abstract:Over the past few decades, South American Rivers, including the Paraná River system, have been intensely impacted by dam constructions. To mitigate adverse impacts, fish passages were incorporated into two large dams on the Paraná River (Itaipu and Porto Primavera) to reestablish longitudinal connectivity, which is very important for conserving populations of migratory fish species. The aim of this study was to estimate genetic diversity, population differentiation and gene flow, using microsatellites markers and mitochondrial haplotypes (d-loop), in populations of Prochilodus lineatus, a long distance migratory species commonly found in Paraná River basin drainages. Samples were obtained from six locations in the Upper and Middle Paraná River: Porto Primavera Reservoir, downstream of the Porto Primavera Reservoir, four tributaries of this reservoir (Pardo, Peixe, Verde and Aguapeí Rivers) and in the Iguaçu River, downstream of the Itaipu dam. High genetic diversity was observed in all samples, as expected for species with large population sizes and high migration rates. However, the influence of other factors, such as secondary contacts, fish passages and restocking were also suggested. In general, evidence of spatial population structuring was not found, even though there were two large dams in the study area. This could also be attributed to the brief period that has elapsed since the dams were built, the influence of migratory and reproductive patterns and the possible contributory effects of the fish passages. Furthermore, the absence of population structuring also corroborates existing information that emphasizes the importance of unimpeded tributaries for conserving migratory fish populations in fragmented watersheds.

Abstract:Assessing genetic diversity within species is key for conservation strategies in the context of human-induced biotic changes. This is important in marine systems, where many species remain undescribed while being overfished, and conflicts between resource-users and conservation agencies are common. Combining niche modelling with population genomics can contribute to resolving those conflicts by identifying management units and understanding how past climatic cycles resulted in current patterns of genetic diversity. We addressed these issues on an undescribed but already overexploited species of sardine of the genus Harengula . We find that the species distribution is determined by salinity and depth, with a continuous distribution along the Brazilian mainland and two disconnected oceanic archipelagos. Genomic data indicate that such biogeographic barriers are associated with two divergent intraspecific lineages. Changes in habitat availability during the last glacial cycle led to different demographic histories among stocks. One coastal population experienced a 3.6-fold expansion, whereas an island-associated population contracted 3-fold, relative to the size of the ancestral population. Our results indicate that the island population should be managed separately from the coastal population, and that a Marine Protected Area covering part of the island population distribution can support the viability of this lineage.

Abstract:The arapaima (<i>Arapaima gigas</i>) is one of the most emblematic species of the Amazon region, and has a long history of exploitation. The present study investigated the connectivity and genetic variability of the arapaima in the region of the confluence of the Amazon and Araguaia-Tocantins basins, with emphasis on the population from the Lago Piratuba Biological Reserve, in the Brazilian state of Amapá. The study was based on the analysis of 11 microsatellite loci and the mtDNA control region sequences of 114 arapaima collected at six localities in the region of the eastern Amazon. The genetic analysis found significant differentiation, which indicated the presence of three distinct populations, the Lower Amazon-Marajó, Lago Piratuba, and Lower Tocantins populations, which lacked any significant level of gene flow. All three populations presented evidence of recent bottlenecks and distinct levels of genetic diversity, which was high only in the Lower Amazon-Marajó population. The conservation strategies for this region should consider the specific characteristics of these three populations and the different patterns of diversity found. The genetic characterization of the Lago Piratuba reserve as a unique population reinforces the importance of the implementation of adequate management strategies in the region.

Abstract:Traditionally, the apparent paucity of biogeographic barriers in marine environments when compared to terrestrial and freshwater habitats has been associated with high gene flow rates among geographically distant populations. However, physical traits such as tide currents, temperature, and salinity levels may serve as ecological boundaries thus leading to restricted-range phylogeographic patterns (e.g., the outflow plume from the Amazonas-Orinoco rivers between the Caribbean and the Brazilian Province) according to adaptive features of coastal organisms. To assess the degree of cohesiveness among populations and species of marine and estuarine fishes along a latitudinal gradient from Western South Atlantic, we carried out comparative phylogenetic and species delimitation analyses based on Cytochrome C Oxidase I (COI) sequences of 34 fish taxa from the Caribbean and Brazilian coasts. Distinct values of genetic diversity were revealed for both Provinces, ranging from moderate (1 to 2%) to high (≥ 2%) in 11.76% and 20.59% of the analyzed taxa, respectively. Furthermore, a significant genetic differentiation was observed within the nominal taxa Diapterus auratus , Citharichthys spilopterus , and Scorpaena plumieri from the Caribbean, as well as for Haemulon plumierii between the Caribbean and Brazilian Provinces. Such divergence is likely to result from temporal isolation among local populations during sea-level fluctuations during the Pliocene-Pleistocene period. The present findings demonstrate that similar biogeographic boundaries may result in species-specific patterns of genetic connectivity, possibly associated with ecological constraints. Since molecular operational taxonomic units (MOTUs) were identified in certain formal taxa from both Provinces, a systematic revision of these groups is highly recommended. At last, multispecies COI data proved to be helpful to phylogeographic inferences and to support appropriate policies for the conservation of natural resources.

Abstract:Neglected cryptic diversity can lead to the permanent loss of locally adapted alleles, which can reduce resilience to rapid environmental change. It can also result in overestimation of fisheries stock sizes that can result from treating different species as if they belonged to one. Bluefish (Pomatomus saltatrix) is considered a circumtropical and subtropical species and an important fishery resource all over the world. Differences in ecologically relevant traits are observed among isolated populations. Also, in the Southwestern Atlantic, molecular data suggest multiple populations, but these are treated as a single fish stock by regulatory agencies due to a lack of definitive information. We used whole mitogenome sequences, nuclear (rho) and mitochondrial (coxI and cytb) genes, as well as microsatellites to investigate historical and current genetic population structure and parameters of bluefish in the Western Atlantic. A total of 263 samples were collected along the Brazilian coast and in the USA (New Jersey, Northwest Atlantic). Data revealed the existence of two evolutionarily significant units (ESU) of bluefish along the South American coast, later confirmed by whole mitogenome sequencing of both haplogroups. These two ESUs have a mostly parapatric distribution, with some areas of overlap, which vary along the year. We also conducted seasonal sampling in Brazil to investigate migration patterns. ESUs occur mostly north and south of parallel 23° 40′ S, with an overlap area that varied seasonally. The level of differentiation between those two ESUs in the SW Atlantic, even in sympatry, is as high as that found between them and those from the NW Atlantic and Europe. Parapatric distribution and restricted gene flow suggest the existence of ecological barriers and local adaptation. The splitting of an ancient population from the Southwestern Atlantic into two putative species is important to understand bluefish evolutionary diversification and has implications for fishery regulatory measures in Brazil.

Abstract:Introduction: Semaprochilodus laticeps is a freshwater fish species native to the Orinoco Basin that is exploited in Colombia at two different stages of its life cycle: fry (for the ornamental trade) and adults (for the consumer market). This double exploitation of juveniles and adults raises concerns about the population stability of the species. Objective: To evaluate the sustainability of the exploitation of the species in these two life stages. Methods: 1 277 specimens of S. laticeps were sampled between June and December 2017. Population parameters were estimated from length-frequency data using ELEFAN 1 routine of the FISAT II package. Length at first maturity (Lm) and length at maximum yield (Lopt) were determined using the Froese and Binohlan equations. Virtual population analysis was performed based on Jones length cohort analysis. Results: Population parameters were as follows: K = 0.75, L∞ = 53.1 cm LE, Z = 2.92 yr-1, M = 1.24 yr-1 and F = 1.68. Lm was estimated to be 29.6 cm SL and Lopt at 33.4 cm SL. Of note, 96.3 % of the fish caught in the consumer fishery exceeded Lm and 80 % exceeded Lopt. The results of the virtual population analysis suggested that a very low proportion (0.85 %) of the population was caught as fry, possibly due to natural mortality. Conclusions: Based on the results, as no indicators of overexploitation were identified, it is recommended to continue with the dual use of S. laticeps. It is recommended to maintain the existing conservation measures, such as the closed season during the reproductive period, to ensure sustainable fishery exploitation. The results obtained can serve as a reference to analyze the situation of other species currently fished for both the ornamental (fry) and food (adult) markets.

Abstract:Small-sized fishes play a crucial ecological role in the dynamics of riverine systems, but their low dispersal capacity and high endemism make them highly vulnerable to environmental change. In the Neotropical region, each watershed typically supports unique populations of small-sized fishes, and in this study, we used Psalidodon paranae (Characidae) as a model to understand the relationships between its population patterns (including age, growth, and reproductive biology) and environmental characteristics. Standardised monthly sampling of P. paranae was carried out at ten sampling sites from June 2010 to May 2011, and environmental variables (including water quality and river characteristics) were measured to characterise these sites. Multivariate techniques and generalised linear models were used to analyse the longitudinal distribution of P. paranae , which was significantly influenced by river depth and width, and suspended solid concentrations. Linear and non-linear models were used to describe the length–weight relationship (resulting in an isometric pattern, b = 3.0256) and the growth patterns ( L ∞ = 21.8 cm, k = 0.51 year −1 , and Φ′ = 2.3989). The analysis of gonadal maturity was combined with somatic and reproductive indices, allowing the identification of spawning periods (occurring mainly during the austral summer) and areas (sites with headwater characteristics). Our results indicate that P. paranae has a good adaptability to the different environmental conditions, and this first assessment of age and growth parameters constitutes a reference for monitoring and further evaluation of long-term changes.

Abstract:ABSTRACT In Brazil, the use and diversity of the common names of fish species, coupled with taxonomic uncertainties, hinder the reliability of fishing statistical data. In this scenario, there are the so-called pilombetas of the São Francisco River, an important fishing resource in region. Despite its importance, the real diversity of species identified in the area remains obscure. In order to properly identify and delimit the species popularly known as pilombetas, an integrative approach involving traditional taxonomy, geometric morphometrics and molecular systematics was applied. Results from geometric morphometrics and molecular analyses were consistent with the results of the traditional morphological analysis, also indicating the delimitation of six taxa belonging to Engraulidae in the lower São Francisco River. In addition, species delimitation methods revealed an intrapopulation genetic divergence of 1.7% for Lycengraulis grossidens. The results revealed that the currently known richness species of Engraulidae in the studied area has been underestimated. Thus, an updated taxonomic key is herein proposed for the Engraulidae species from the lower São Francisco River and estuary. The integrative analysis approach revealed to be effective to address taxonomic questions and help the management of stocks, ensuring the maintenance of local diversity of fishes in the Neotropical region.

Abstract:Long‐term systematic records of cetacean strandings are valuable indicators of temporal responses of specific cetacean populations and communities to natural or anthropogenic pressures. They also help guide management objectives and conservation actions. Generalized additive models (GAMs) were used to analyse 10 years of systematic and opportunistic surveys of Guiana dolphin strandings in the Amazon Estuary region, Northern Brazil. The spatial distribution of strandings was assessed through kernel density maps, and age‐at‐death structure of this population was described. The model results revealed marked seasonality in the strandings and, by extension, the mortality of Guiana dolphins in the Amazon Estuary. This seasonality varied between the inner estuarine regions and Atlantic coastal area, reflecting the environment's characteristics. Long‐term decline of strandings in the Amazon Estuary inner region and unusual high mortality of young sexually mature individuals were detected. These results, besides the high stranding rate detected for the species, are mostly attributed to the species' interaction with intense fishing activity in the region. Conservation efforts should consider the seasonality of Guiana dolphin strandings in the Amazon Estuary. The systematic monitoring of strandings, especially in high‐frequency areas, is recommended for assessing long‐term species mortality. Additionally, implementing management actions related to the species' interaction with fishing activities, such as regulatory enhancements, gillnet modifications and seasonal no‐fishing zones, is advised. Educational campaigns targeting fishermen and riverine communities are essential to reduce bycatch and the folkloric use of the species. This study underscores the effectiveness of stranding data as a source of biological and ecological information, and as a tool for understanding human actions affecting coastal populations of aquatic mammals. It emphasizes the need for urgent studies to characterize this population, clarify the influence of bycatch as an anthropogenic pressure and implement effective conservation actions to preserve the Guiana dolphin population.

Abstract:Pleistocene climate changes were major historical events that impacted South American biodiversity. Although the effects of such changes are well-documented for several biomes, it is poorly known how these climate shifts affected the biodiversity of the Pantanal floodplain. Fish are one of the most diverse groups in the Pantanal floodplains and can be taken as a suitable biological model for reconstructing paleoenvironmental scenarios. To identify the effects of Pleistocene climate changes on Pantanal's ichthyofauna, we used genetic data from multiple populations of a top-predator long-distance migratory fish, Salminus brasiliensis. We specifically investigated whether Pleistocene climate changes affected the demography of this species. If this was the case, we expected to find changes in population size over time. Thus, we assessed the genetic diversity of S. brasiliensis to trace the demographic history of nine populations from the Upper Paraguay basin, which includes the Pantanal floodplain, that form a single genetic group, employing approximate Bayesian computation (ABC) to test five scenarios: constant population, old expansion, old decline, old bottleneck following by recent expansion, and old expansion following by recent decline. Based on two mitochondrial DNA markers, our inferences from ABC analysis, the results of Bayesian skyline plot, the implications of star-like networks, and the patterns of genetic diversity (high haplotype diversity and low-to-moderate nucleotide diversity) indicated a sudden population expansion. ABC allowed us to make strong quantitative inferences about the demographic history of S. brasiliensis. We estimated a small ancestral population size that underwent a drastic fivefold expansion, probably associated with the colonization of newly formed habitats. The estimated time of this expansion was consistent with a humid and warm phase as inferred by speleothem growth phases and travertine records during Pleistocene interglacial periods. The strong concordance between our genetic inferences and this historical data could represent the first genetic record of a humid and warm phase in the Pantanal in the period since the Last Interglacial to 40 ka.

Abstract:Queiroz-Brito, Maria Clara Gonçalves ; Defavari, Gabriela Rocha ; Rosa, Ierecê De Lucena ; Torres, Rodrigo Augusto ; Hippocampus reidi represents the most abundant species of the genus Hippocampus along the Brazilian coast. Despite being charismatic, the species is globally threatened due to habitat degradation and commercial exploration, especially in Brazil, which is the leader in exportation and consumption of the species. Through mitochondrial (cytochrome b and control region) and nuclear (1st intron S7) data, the current study investigates the variation and genetic structure of H. reidi along the Brazilian coast, from Pará to Santa Catarina states. The mitochondrial data indicate the presence of two lineages: (1) North/Northeast and (2) South/Southeast, which was partially recovered by nuclear data. This scenario could be related to temperature differences and circulation patterns of the Brazil and North-Brazil currents, which define these groups into biogeographic sub-provinces. The lineages occur in sympatry in Bahia state, which can be explained by the occurrence of secondary contact during the last glacial maximum. Despite presenting two lineages, for management and conservation, three units are indicated: (1) North/Northeast, (2) Bahia, and (3) South/Southeast. The North/Northeast unit proved to be more vulnerable, presenting the lowest genetic diversity indices, representing a priority for future conservation actions.

Abstract:Genetic connectivity and historical population expansion are characteristics that have largely been studied and observed in several members of the Lutjanidae family throughout the Western South Atlantic, particularly in Brazilian waters. In this study, the population genetic structure of Lutjanus jocu was investigated using four genomic regions: two from mitochondrial DNA (Control Region and Cytochrome B) and two nuclear genes (Delta 6 desaturase—intron 8 and Ribosomal Protein S7—intron 1), in 144 specimens distributed in six locations on the Atlantic coast of Brazil. The results revealed high levels of genetic diversity for both mtDNA and nuDNA markers, and intense gene flow, indicating, consistent with other studies, that this species is represented by a single genetic stock along the Brazilian coast. This is likely due to the high dispersal capacity of L. jocu larvae, driven by the main equatorial currents, as well as biological and ecological factors, contributing to a genetically homogeneous population. Historical demographic analysis and neutrality tests suggest that the species experienced population expansion, possibly during the Pleistocene. Information regarding the genetic structure of populations is extremely important, especially for species of high economic and fishing importance, and can be very useful in guiding stock management and management measures.

Abstract:Sustainable management of transboundary fish stocks hinges on accurate delineation of population structure. Genetic analysis offers a powerful tool to identify potential subpopulations within a seemingly homogenous stock, facilitating the development of effective, coordinated management strategies across international borders. Along the West African coast, the Atlantic chub mackerel (Scomber colias) is a commercially important and ecologically significant species, yet little is known about its genetic population structure and connectivity. Currently, the stock is managed as a single unit in West African waters despite new research suggesting morphological and adaptive differences. Here, eight microsatellite loci were genotyped on 1,169 individuals distributed across 33 sampling sites from Morocco (27.39°N) to Namibia (22.21°S). Bayesian clustering analysis depicts one homogeneous population across the studied area with null overall differentiation (F ST = 0.0001ns), which suggests panmixia and aligns with the migratory potential of this species. This finding has significant implications for the effective conservation and management of S. colias within a wide scope of its distribution across West African waters from the South of Morocco to the North-Centre of Namibia and underscores the need for increased regional cooperation in fisheries management and conservation.

Abstract:Recent estimates of the size at first maturity (L 50 ) of Sardinella brasiliensis showed contradictory results with a decreasing in the fish stock biomass encompassed by increasing values of L 50 . The methodological approach used hereby allowed to separate sardines classified in the virginal maturity stage from those categorized in the recovery stage, and ready for one next spawning event. This study evaluated the hypothesis of the existence of separated stocks experiencing distinct environmental conditions and fishing pressures which may have altered L 50 estimates using a robust dataset based on biological samples collected along the entire species distribution area in the southeast-south Brazilian coast [Rio de Janeiro (RJ), São Paulo (SP), Paraná (PR), Santa Catarina (SC) and Rio Grande do Sul (RS)] between 2000 and 2018. A reclassification of the gonadal maturity stages provided a more realistic estimate of L 50. Combining biological, reproductive, fishing data and the mean temperature of the catch (MTC), the leave-one-out classification correctly re-assigned individuals with an overall accuracy of 85% [100% (RJ), 45% (SP), 99% (PR), 99% (SC) and 82% (RS)]. The connectivity between the local populations of S. brasiliensis off RJ (23°S) and the southern populations is limited, contrasting to spatial structured semi-discrete population-units found between SP and RS (24°S-30°S). The northern extreme population-unit (RJ, 22°S-23°S) showed an expressive reduction of L 50 , and a negative correlation was detected between the increasing MTC values and the abundance of early maturing individuals and recruits of the species. Stock specific L 50 estimates seemed to act as indicators of long term environmental fluctuations.

Abstract:Abstract Habitat fragmentation is among the main threats to freshwater fish biodiversity, with expected effects including genetic impacts due to disturbance of migration and declining population size. Major concern falls on highly exploited species such as Prochilodus magdalenae , a migratory characiform fish endemic to the Magdalena River basin supporting much of the Colombian artisanal fishery, whose migration route was interrupted by the Ituango hydroelectric project in the Cauca River. To determine the potential effects of fragmentation, this study evaluated the population genetics of this species on both geographical (upstream and downstream of the dam) and temporal (before and after the construction) scales by using 11 species-specific microsatellite markers. Contrary to expectation, genetic diversity and structure remained relatively stable. This study provided no evidence of genetic impacts on P. magdalenae associated with fragmentation over the short term (4–10 years, 5–12 generations) despite persisting threats, genetic evidence of bottleneck, and a high degree of inbreeding, showing the ability of this species to withstand disturbance of its habitat.

Abstract:The Critically Endangered seventy-four seabream Polysteganus undulosus , a slow-growing sparid that forms spawning aggregations off South Africa, faced heavy exploitation from 1910 until a fishery moratorium was put in place in 1998. Utilising temporal samples from 1962/1963 (mid-collapse) and 2005/2006 (post-collapse), we assessed genetic diversity at six microsatellite loci. Amplification success for archived samples was low (43%), necessitating a rarefaction approach, revealing a 40% decrease in allelic diversity. Significant genetic differences between recent and archived samples confirmed the impact of overfishing. Simulation studies indicated that missing genotypes did not affect these tests, validating the genetic differences. Using a coalescent-based approach, a 10-fold decrease in effective population size ( N e) was estimated over this 43-year period ( N e = 43.88 to 4.76). Simulations provided corrected N e of 36.86, accounting for missing genotypes that were responsible for inflated values. Assuming 25% of pristine levels in the 1960s, pristine N e values ranged from 147 to 176 in 1910, suggesting a 96.8–98.3% genetic decline over 88 years of exploitation. This study emphasises severe genetic consequences of overfishing on P. undulosus diversity and effective population size. The results provide vital genetic baseline data for recovery assessment and conservation efforts, as well as an analytical framework to evaluate the stock decline using partial genotyping data gathered from degraded archived samples.

Abstract:Depending on the time of addition, prostaglandin I2 (PGI2; greater than or equal to 10(-9) M) either inhibits or reverses platelet agglutination mediated by human factor VIII-related von Willebrand factor activity (FVIIIvWF) and ristocetin, or bovine FVIIIvWF alone. 6-Keto-PGF1 alpha, the inactive metabolite of PGI2, is without effect, PGI2 inhibition is potentiated by the phosphodiesterase inhibitor, theophylline, and is not the result of PGI2 suppression of ADP release. PGI2 (+/- theophylline) does not inhibit ristocetin-induced binding of purified human 125I-FVIIIvWF multimers to washed platelets or to platelets treated with PGI2 and then formalin fixed (although subsequent agglutination of these platelets is impaired). Washed platelets treated previously with 2-aminoethylisothiouronium bromide (AET), an agent that reduces disulfide bonds and alters platelet membranes, also bind human 125I-FVIIIvWF multimers without agglutinating. We conclude that FVIIIvWF-mediated agglutination requires both functional platelet FVIIIvWF binding sites and platelet-platelet cohesion sites, and that platelet surface cohesion sites are altered by AET and PGI2. PGI2 from adjacent intact endothelial cells may prevent excessive platelet accumulation on exposed subendothelium without suppressing an essential hemostatic process--the binding of platelets to subendothelial FVIIIvWF.
