Abstract:The uncoupling of osteoblastic and osteoclastic activity is central to disorders such as osteoporosis, osteolytic malignancies, and periodontitis. Numerous studies have shown explicit functions for bone morphogenetic proteins (BMPs) in skeletogenesis. Their signaling activity has been shown in various contexts to be regulated by extracellular proteins, including Twisted gastrulation (TWSG1). However, experimental paradigms determining the effects of BMP regulators on bone remodeling are limited. In this study, we assessed the role of TWSG1 in postnatal bone homeostasis. Twsg1‐deficient (Twsg1−/−) mice developed osteopenia that could not be explained by defective osteoblast function, because mineral apposition rate and differentiation markers were not significantly different compared with wildtype (WT) mice. Instead, we discovered a striking enhancement of osteoclastogenesis in Twsg1−/− mice, leading to increased bone resorption with resultant osteopenia. Enhanced osteoclastogenesis in Twsg1−/− mice was caused by increased cell fusion, differentiation, and function of osteoclasts. Furthermore, RANKL‐mediated osteoclastogenesis and phosphorylated Smad1/5/8 levels were enhanced when WT osteoclasts were treated with recombinant BMP2, suggesting direct regulation of osteoclast differentiation by BMPs. Increase in detectable levels of phosphorylated Smad 1/5/8 was noted in osteoclasts from Twsg1−/− mice compared with WT mice. Furthermore, the enhanced osteoclastogenesis in Twsg1−/− mice was reversed in vitro in a dose‐dependent manner with exposure to Noggin, a BMP antagonist, strongly suggesting that the enhanced osteoclastogenesis in Twsg1 mutants is attributable to increased BMP signaling. Thus, we present a novel and previously uncharacterized role for TWSG1 in inhibiting osteoclastogenesis through regulation of BMP activity.
Attentional disengagement from negative natural sounds for high-anxious individuals

Abstract:Background and objectives: Previous studies have not consistently concluded whether high-anxious persons exhibit attentional bias towards negative natural auditory stimuli. The present study explores whether auditory negative stimuli could induce attentional bias to negative sounds in real life and investigates the exact nature of these biases using an emotional spatial cueing task. Design: Experimental study with a mixed factorial design. Method: We created two groups according to the state-trait anxiety scale, namely high and low trait anxiety. Participants (N = 68 undergraduate students) were required to respond to an auditory target after receiving a negative (aversive sounds from natural life) or neutral auditory stimuli. Results: A 2 (Validity: valid/invalid) × 2 (Cue Valence: negative/neutral) × 2 (Anxiety Group: LA/HA) repeated-measures ANOVA on reaction times revealed that participants with high trait anxiety exhibited slower reaction times in invalid trials following negative cues than following neutral cues. Higher levels of trait anxiety were associated with more difficult attentional disengagement from negative auditory information. Conclusions: The results demonstrate that impaired attentional disengagement was one of the mechanisms by which high-anxious participants exhibited auditory attentional bias to natural negative information.
Selective attention to sound features mediates cross-modal activation of visual cortices

Abstract:Abstract Contemporary schemas of brain organization now include multisensory processes both in low-level cortices as well as at early stages of stimulus processing. Evidence has also accumulated showing that unisensory stimulus processing can result in cross-modal effects. For example, task-irrelevant and lateralized sounds can activate visual cortices; a phenomenon referred to as the auditory-evoked contralateral occipital positivity (ACOP). Some claim this is an example of automatic attentional capture in visual cortices. Other results, however, indicate that context may play a determinant role. Here, we investigated whether selective attention to spatial features of sounds is a determining factor in eliciting the ACOP. We recorded high-density auditory evoked potentials (AEPs) while participants selectively attended and discriminated sounds according to four possible stimulus attributes: location, pitch, speaker identity or syllable. Sound acoustics were held constant, and their location was always equiprobable (50% left, 50% right). The only manipulation was to which sound dimension participants attended. We analysed the AEP data from healthy participants within an electrical neuroimaging framework. The presence of sound-elicited activations of visual cortices depended on the to-be-discriminated, goal-based dimension. The ACOP was elicited only when participants were required to discriminate sound location, but not when they attended to any of the non-spatial features. These results provide a further indication that the ACOP is not automatic. Moreover, our findings showcase the interplay between task-relevance and spatial (un)predictability in determining the presence of the cross-modal activation of visual cortices.
Threat-Related Corticocortical Connectivity Elicited by Rapid Auditory Looms

Abstract:While sounds of approaching objects are generally more salient than those of receding ones, the traditional association of this auditory looming bias with threat perception is subject to debate. Differences between looming and receding sounds may also be learned through non-threatening multisensory information, or influenced by confounding stimulus characteristics. To investigate, we analyzed corticocortical connectivity patterns from electroencephalography, examining the preferential processing of looming sounds under different attentional states. To simulate rapid distance changes we used complementary distance cues, previously studied in the looming bias literature. Notably, despite the absence of conscious threat perception, we observed crucial involvement of frontal cortical regions typically associated with threat and fear responses. Our findings suggest an underlying bias towards the ventral 'what' stream over the dorsal 'where' stream in auditory information processing, even when the participants' task was solely focused on the discrimination of movement direction. These results support the idea, that the perceptual bias towards looming sounds reflects an auditory threat detection mechanism, while offering insights into the neural function involved in processing ecologically relevant environmental cues.

Assessing Behavioral and Neural Correlates of Change Detection in Spatialized Acoustic Scenes

Abstract:The ability to detect changes in complex auditory scenes is crucial for human survival, yet the neural mechanisms underlying this process remain elusive. This study investigates how the presence and location of sound sources impacts active auditory change detection as well as neural correlates of passive change detection. We employed stimuli designed to minimize semantic associations while preserving naturalistic temporal envelopes and broadband spectra, presented in a spatial loudspeaker array. Behavioral change detection experiments tasked participants with detecting new sources added to spatialized and non-spatialized multi-source auditory scenes. In a passive listening experiment, participants were given a visual decoy task while neural data were collected via electroencephalography (EEG) during exposure to unattended spatialized scenes and added sources. Our behavioral experiments (N = 21 and 21) demonstrated that spatializing sounds facilitated change detection compared to non-spatialized presentation, but that performance declined with increasing number of sound sources and higher hearing thresholds at high frequencies, exclusively in spatialized conditions. Slower reaction times were also observed when changes occurred from above or behind the listener, exacerbated by a higher number of sources. EEG experiments (N = 32 and 30), using the same stimuli, showed robust change-evoked responses. However, no significant differences were detected in our analysis as a function of spatial location of the appearing source.

Auditory Spatial Bisection of Blind and Normally Sighted Individuals in Free Field and Virtual Acoustics

Abstract:Sound localization is an important ability in everyday life. This study investigates the influence of vision and presentation mode on auditory spatial bisection performance. Subjects were asked to identify the smaller perceived distance between three consecutive stimuli that were either presented via loudspeakers (free field) or via headphones after convolution with generic head-related impulse responses (binaural reproduction). Thirteen azimuthal sound incidence angles on a circular arc segment of ±24° at a radius of 3 m were included in three regions of space (front, rear, and laterally left). Twenty normally sighted (measured both sighted and blindfolded) and eight blind persons participated. Results showed no significant differences with respect to visual condition, but strong effects of sound direction and presentation mode. Psychometric functions were steepest in frontal space and indicated median spatial bisection thresholds of 11°–14°. Thresholds increased significantly in rear (11°–17°) and laterally left (20°–28°) space in free field. Individual pinna and torso cues, as available only in free field presentation, improved the performance of all participants compared to binaural reproduction. Especially in rear space, auditory spatial bisection thresholds were three to four times higher (i.e., poorer) using binaural reproduction than in free field. The results underline the importance of individual auditory spatial cues for spatial bisection, irrespective of access to vision, which indicates that vision may not be strictly necessary to calibrate allocentric spatial hearing.

Abstract:Studies on sound perception show a tendency to overestimate the distance of an approaching sound source, leading to a faster reaction time compared to a receding sound source. Nevertheless, it is unclear whether motor preparation and execution change according to the perceived sound direction and distance, particularly when the sound falls inside the individual's peripersonal space. In this study we developed several auditory stimuli by means of two speakers, generating sounds moving toward the perceiver but stopping at different distances from her/him. Participants were asked to raise their arms as soon as the sound stopped, and their premotor and motor movement components were recorded by means of electromyography (EMG). Error in locating the perceived sound distance was also measured by asking participants to walk to the point in space where they believed the sound had stopped. Results showed that action initiation was anticipated as a function of sound distance: the closer the sound, the earlier the movement onset, when the sound entered the subject's peripersonal space. Less error for distance estimation was present when the sound was inside the peripersonal space with a modulation in the order of a few centimeters. Overall, our results reveal a link between perceptual bias in sound distance evaluation and peripersonal space, suggesting the presence of motor plan specificity.

Abstract:BackgroundNumerous studies have investigated the neural underpinnings of passive and active deviance and target detection in the well-known auditory oddball paradigm by means of event-related potentials (ERPs) or functional magnetic resonance imaging (fMRI). The present auditory oddball study investigates the spatio-temporal dynamics of passive versus active deviance and target detection by analyzing amplitude modulations of early and late ERPs while at the same time exploring the neural sources underling this modulation with standardized low-resolution brain electromagnetic tomography (sLORETA).MethodsA 64-channel EEG was recorded from twelve healthy right-handed participants while listening to ‘standards’ and ‘deviants’ (500 vs. 1000 Hz pure tones) during a passive (block 1) and an active (block 2) listening condition. During passive listening, participants had to simply listen to the tones. During active listening they had to attend and press a key in response to the deviant tones.ResultsPassive and active listening elicited an N1 component, a mismatch negativity (MMN) as difference potential (whose amplitudes were temporally overlapping with the N1) and a P3 component. N1/MMN and P3 amplitudes were significantly more pronounced for deviants as compared to standards during both listening conditions. Active listening augmented P3 modulation to deviants significantly compared to passive listening, whereas deviance detection as indexed by N1/MMN modulation was unaffected by the task. During passive listening, sLORETA contrasts (deviants > standards) revealed significant activations in the right superior temporal gyrus (STG) and the lingual gyri bilaterally (N1/MMN) as well as in the left and right insulae (P3). During active listening, significant activations were found for the N1/MMN in the right inferior parietal lobule (IPL) and for the P3 in multiple cortical regions (e.g., precuneus).DiscussionThe results provide evidence for the hypothesis that passive as well as active deviance and target detection elicit cortical activations in spatially distributed brain regions and neural networks including the ventral attention network (VAN), dorsal attention network (DAN) and salience network (SN). Based on the temporal activation of the neural sources underlying ERP modulations, a neurophysiological model of passive and active deviance and target detection is proposed which can be tested in future studies.

Abstract:The ability to detect unexpected or deviant events in natural scenes is critical for survival. In the auditory system, neurons from the midbrain to cortex adapt quickly to repeated stimuli but this adaptation does not fully generalize to other rare stimuli, a phenomenon called stimulus-specific adaptation (SSA). Most studies of SSA were conducted with pure tones of different frequencies, and it is by now well-established that SSA to tone frequency is strong and robust in auditory cortex. Here we tested SSA in the auditory cortex to the ear of stimulation using broadband noise. We show that cortical neurons adapt specifically to the ear of stimulation, and that the contrast between the responses to stimulation of the same ear when rare and when common depends on the binaural interaction class of the neurons.
Mismatch negativity between discriminating and undiscriminating participants on the front-back sound localization

Abstract:Sound localization in the front-back dimension is reported to be challenging, with individual differences. We investigated whether auditory discrimination processing in the brain differs based on front-back sound localization ability. This study conducted an auditory oddball task using speakers in front of and behind the participants. We used event-related brain potentials to examine the deviance detection process between groups that could and could not discriminate front-back sound localization. The results indicated that mismatch negativity (MMN) occurred during the deviance detection process, and P2 amplitude differed between standard and deviant locations in both groups. However, the latency of MMN was shorter in the group that could discriminate front-back sounds than in the group that could not. Additionally, N1 amplitude increased for deviant locations compared to standard ones only in the discriminating group. In conclusion, the sensory memories matching process based on traces of previously presented stimuli (MMN, P2) occurred regardless of discrimination ability. However, the response to changes in the physical properties of sounds (MMN latency, N1 amplitude) differed depending on the ability to discriminate front-back sounds. Our findings suggest that the brain may have different processing strategies for the two directions even without subjective recognition of the front-back direction of incoming sounds.
Vigilance to Spatialized Auditory Displays: Initial Assessment of Performance and Workload

Abstract:Human Factors, Ahead of Print. ObjectiveThe present study was designed to evaluate human performance and workload associated with an auditory vigilance task that required spatial discrimination of auditory stimuli.BackgroundSpatial auditory displays have been increasingly developed and implemented into settings that require vigilance toward auditory spatial discrimination and localization (e.g., collision avoidance warnings). Research has yet to determine whether a vigilance decrement could impede performance in such applications.MethodParticipants completed a 40-minute auditory vigilance task in either a spatial discrimination condition or a temporal discrimination condition. In the spatial discrimination condition, participants differentiated sounds based on differences in spatial location. In the temporal discrimination condition, participants differentiated sounds based on differences in stimulus duration.ResultsCorrect detections and false alarms declined during the vigilance task, and each did so at a similar rate in both conditions. The overall level of correct detections did not differ significantly between conditions, but false alarms occurred more frequently within the spatial discrimination condition than in the temporal discrimination condition. NASA-TLX ratings and pupil diameter measurements indicated no differences in workload.ConclusionResults indicated that tasks requiring auditory spatial discrimination can induce a vigilance decrement; and they may result in inferior vigilance performance, compared to tasks requiring discrimination of auditory duration.ApplicationVigilance decrements may impede performance and safety in settings that depend on sustained attention to spatial auditory displays. Display designers should also be aware that auditory displays that require users to discriminate differences in spatial location may result in poorer discrimination performance than non-spatial displays.

Abstract:Completely ignoring a salient distractor presented concurrently with a target is difficult, and sometimes attention is involuntarily attracted to the distractor's location (attentional capture). Employing the N2ac component as a marker of attention allocation toward sounds, in this study we investigate the spatiotemporal dynamics of auditory attention across two experiments. Human participants (male and female) performed an auditory search task, where the target was accompanied by a distractor in two-third of the trials. For a distractor more salient than the target (Experiment 1), we observe not only a distractor N2ac (indicating attentional capture) but the full chain of attentional dynamics implied by the notion of attentional capture, namely, (1) the distractor captures attention before the target is attended, (2) allocation of attention to the target is delayed by distractor presence, and (3) the target is attended after the distractor. Conversely, for a distractor less salient than the target (Experiment 2), although responses were delayed, no attentional capture was observed. Together, these findings reveal two types of spatial attentional dynamics in the auditory modality (distraction with and without attentional capture).

Abstract:The effects attention has on intensity deviant Mismatch Negativity responses is an unknown factor in basic sensory neuroscience. It would be useful to understand how attention would affect responses to intensity deviants compared to each other (upward vs downward), and compared to other sensory dimensions such as frequency. Overall, previous research indicates that attention may modulate neuronal gain in healthy participants and change the amplitudes of evoked responses, and may mainly affect the responses to regularly repeating (standard) stimuli rather than deviants. Gain may respond differently in participants with tinnitus and/or hyperacusis under the same conditions compared to controls. Overall, results of the passive task condition were consistent with previous research. Auditory attention magnified MMN in response to upward deviants, while visual attention attenuated it in both control and tinnitus groups. However, auditory attention selectively enhanced downward deviant MMN in the tinnitus group (compared to passive attention). Using the auditory attention paradigm may be advantageous in MMN studies on tinnitus/hyperacusis because the observed differences would be particularly large.

Abstract:In two experiments investigating hemispheric asymmetries in auditory distraction, the spatial location of to-be-ignored sound was manipulated. Prior studies indicated a left-ear disadvantage for changing-state sequences during short-term serial recall but lacked a direct measure of the changing-state effect. Experiment 1 compared changing-state with steady-state sequences in a visual-verbal serial recall task, confirming that left-ear disruption resulted from the acoustically varying nature of the sound, emphasizing right hemisphere dominance for processing acoustic variation in unattended stimuli. Experiment 2 replicated these findings and explored participants' metacognitive awareness of auditory distractors' disruptive potential. While participants were aware that changing-state sequences were more disruptive than steady-state sequences, they lacked awareness of the left-ear disadvantage. The study suggests individuals have metacognitive awareness of the disruptive impact of changing-state over steady-state sound but not of the accompanying left-ear disadvantage, raising implications for theoretical accounts of auditory distraction.

Abstract:OBJECTIVE To investigate the relationship of Msp AI polymorphism in the promoter region of cytochrome P450c 17alpha (CYP17) gene with bone mass and bone size in Shanghai men of Han nationality. METHODS The CYP17 Msp AI genotype was determined by polymerase chain reaction-restriction fragment length polymorphism in 397 unrelated men (324 healthy men, 73 osteoporosis patients) aged 46-80 years of Han nationality in Shanghai. Bone mineral density (BMD), bone mineral content (BMC), and bone cross-section area (CSA) at lumber spine 1-4 and at any sites of proximal femur, including femoral neck, trochanter and Ward's triangle were measured by duel-energy X-ray absorptiometry. RESULTS Frequency distributions of CYP17 genotype were TC (51.1%), CC (33.8%), and TT (15.1%). The allele frequencies T and C were 40.7% and 59.3%, respectively. Allele frequencies did not deviate from Hardy-Weinberg equilibrium. The frequencies of CYP17 Msp AI genotype did not show difference between osteoporosis cases and healthy controls. In group of all population, or in subgroups of osteoporosis patients and healthy men, CYP17 Msp AI genotype was not significantly associated with BMD, BMC, and CSA at lumber spine 1-4 and at any sites of proximal femur after having been adjusted for age, weight, and height with analysis of covariance. CONCLUSION Msp AI polymorphism of CYP17 gene is not a genetic factor that influence the variation of bone mass and bone size in Shanghai men of Han nationality.
Attentional and linguistic interactions in speech perception

Abstract:The role of attention in speech comprehension is not well understood. We used fMRI to study the neural correlates of auditory word, pseudoword, and nonspeech (spectrally rotated speech) perception during a bimodal (auditory, visual) selective attention task. In three conditions, Attend Auditory (ignore visual), Ignore Auditory (attend visual), and Visual (no auditory stimulation), 28 subjects performed a one-back matching task in the assigned attended modality. The visual task, attending to rapidly presented Japanese characters, was designed to be highly demanding in order to prevent attention to the simultaneously presented auditory stimuli. Regardless of stimulus type, attention to the auditory channel enhanced activation by the auditory stimuli (Attend Auditory>Ignore Auditory) in bilateral posterior superior temporal regions and left inferior frontal cortex. Across attentional conditions, there were main effects of speech processing (word+pseudoword>rotated speech) in left orbitofrontal cortex and several posterior right hemisphere regions, though these areas also showed strong interactions with attention (larger speech effects in the Attend Auditory than in the Ignore Auditory condition) and no significant speech effects in the Ignore Auditory condition. Several other regions, including the postcentral gyri, left supramarginal gyrus, and temporal lobes bilaterally, showed similar interactions due to the presence of speech effects only in the Attend Auditory condition. Main effects of lexicality (word>pseudoword) were isolated to a small region of the left lateral prefrontal cortex. Examination of this region showed significant word>pseudoword activation only in the Attend Auditory condition. Several other brain regions, including left ventromedial frontal lobe, left dorsal prefrontal cortex, and left middle temporal gyrus, showed Attention x Lexicality interactions due to the presence of lexical activation only in the Attend Auditory condition. These results support a model in which neutral speech presented in an unattended sensory channel undergoes relatively little processing beyond the early perceptual level. Specifically, processing of phonetic and lexical-semantic information appears to be very limited in such circumstances, consistent with prior behavioral studies.

Abstract:The successful integration of visual and auditory stimuli requires information about whether visual and auditory signals originate from corresponding places in the external world. Here we report crossmodal effects of spatially congruent and incongruent audio-visual (AV) stimulation. Visual and auditory stimuli were presented from one of four horizontal locations in external space. Seven healthy human subjects had to assess the spatial fit of a visual stimulus (i.e. a gray-scaled picture of a cartoon dog) and a simultaneously presented auditory stimulus (i.e. a barking sound). Functional magnetic resonance imaging (fMRI) revealed two distinct networks of cortical regions that processed preferentially either spatially congruent or spatially incongruent AV stimuli. Whereas earlier visual areas responded preferentially to incongruent AV stimulation, higher visual areas of the temporal and parietal cortex (left inferior temporal gyrus [ITG], right posterior superior temporal gyrus/sulcus [pSTG/STS], left intra-parietal sulcus [IPS]) and frontal regions (left pre-central gyrus [PreCG], left dorsolateral pre-frontal cortex [DLPFC]) responded preferentially to congruent AV stimulation. A position-resolved analysis revealed three robust cortical representations for each of the four visual stimulus locations in retinotopic visual regions corresponding to the representation of the horizontal meridian in area V1 and at the dorsal and ventral borders between areas V2 and V3. While these regions of interest (ROIs) did not show any significant effect of spatial congruency, we found subregions within ROIs in the right hemisphere that showed an incongruency effect (i.e. an increased fMRI signal during spatially incongruent compared to congruent AV stimulation). We interpret this finding as a correlate of spatially distributed recurrent feedback during mismatch processing: whenever a spatial mismatch is detected in multisensory regions (such as the IPS), processing resources are re-directed to low-level visual areas.
Listener-Position and Orientation Dependency of Auditory Perception in an Enclosed Space: Elicitation of Salient Attributes

Abstract:This paper presents a subjective study conducted on the perception of salient auditory attributes depending on the listener's position and head orientations in an enclosed space. Two elicitation experiments were carried out using the Repertory Grid Technique; in-situ and laboratory experiments aimed to identify perceptual attributes among ten different combinations of the listener's positions and head orientations in a concert hall. Results revealed that, between the in-situ and laboratory experiments, the listening positions and head orientations were clustered identically. Ten salient perceptual attributes were identified from the data obtained from the laboratory experiment. Whilst these included conventional attributes such as ASW (apparent source width) and LEV (listener envelopment), new attributes such as PRL (perceived reverb loudness), ARW (apparent reverb width) and Reverb Direction were identified, and they are hypothesised to be sub-attributes of LEV (listener envelopment). Timbral characteristics such as Reverb Brightness and Echo Brightness were also identified as salient attributes, which are considered to potentially contribute to the overall sound clarity.

Abstract:The vigilance decrement, a temporal decline in detection performance, has been observed across multiple sensory modalities. Spatial uncertainty about the location of task-relevant stimuli has been demonstrated to increase the demands of vigilance and increase the severity of the vigilance decrement when attending to visual displays. The current study investigated whether spatial uncertainty also increases the severity of the vigilance decrement and task demands when an auditory display is used. Individuals monitored an auditory display to detect critical signals that were shorter in duration than non-target stimuli. These auditory stimuli were presented in either a consistent, predictable pattern that alternated sound presentation from left to right (spatial certainty) or an inconsistent, unpredictable pattern that randomly presented sounds from the left or right (spatial uncertainty). Cerebral blood flow velocity (CBFV) was measured to assess the neurophysiological demands of the task. A decline in performance and CBFV was observed in both the spatially certain and spatially uncertain conditions, suggesting that spatial auditory vigilance tasks are demanding and can result in a vigilance decrement. Spatial uncertainty resulted in a more severe vigilance decrement in correct detections compared to spatial certainty. Reduced right-hemispheric CBFV was also observed during spatial uncertainty compared to spatial certainty. Together, these results suggest that auditory spatial uncertainty hindered performance and required greater attentional demands compared to spatial certainty. These results concur with previous research showing the negative impact of spatial uncertainty in visual vigilance tasks, but the current results contrast recent research showing no effect of spatial uncertainty on tactile vigilance.
