Abstract:This study aimed to evaluate the effects of aging on the quality of breast meat from broilers affected of wooden breast. Samples that were classified as normal (control), moderate (hardness verified only in one region of breast fillet), and severe (hardness verified in all the extension of breast fillet) were evaluated fresh and after three and seven days of aging. Normal samples and samples with a moderate degree of myopathy showed greater water-holding capacity, which may benefit the processing industry of poultry meat. During the aging process, increase was observed in total collagen concentration (from 0.41% in normal samples to 0.56% in samples with severe degree). Samples of chicken breast affected by moderate degree showed higher myofibril fragmentation index (MFI = 115) than normal chicken samples (95.65). Although chicken samples affected with severe degree of wooden breast myopathy are more tender than normal chicken breasts, they produce more exudate, which can be detrimental to the processing of poultry meat. The aging process may improve the reduction of cooking weight loss and protein loss in exudation of broilers’ breasts affected by wooden breast myopathy.

Abstract:Storage temperature and duration plays an important role in meat processing. Observations in poultry processing plants have shown a serious deviation in storage condition compared to the recommended procedures. Furthermore, there is still a paucity of evidence on the effects of storage temperature and duration on meat quality and microbial population. The aim of this study was to determine the effects of different temperature and duration during storage on physico-chemical properties and microbiological quality of broiler chicken Pectoralis major muscle. Eighty birds were slaughtered and processed, following which the packed boneless breast (PBB) (each bird was to provide two breast muscle samples; left breast and right breast) was divided into four groups, each consisted of 40 PBB. Each group was subsequently assigned to storage either at 4 °C, −10 °C, −18 °C or −40 °C, for 24 h before 20 PBB samples from each group were transported to the respective laboratory for meat quality and microbiological analysis. The remaining 20 PBB from each storage temperature were stored for 72 h before being transported for analysis. Results have shown significant increases in drip loss and cooking loss as the storage temperature decreases. Similarly, storage duration significantly affected cooking loss, of which, samples stored for 72 h exhibited higher cooking loss compared to those stored for 24 h. For color, significant differences were only observed in lightness (L*) and redness (a*) values. Longer duration of storage had significantly improved tenderness whereby, samples that have been stored for a shorter duration presented higher pH values. Populations of coliform and Salmonella decreased significantly with decreasing temperature and increasing storage duration.

Abstract:Cylindrical specimens (50 mm diameter and 160 mm length) of fresh pork muscle (boneless rib portions) packed in plastic bags were frozen by pressure shift freezing (PSF) at 100, 150, and 200 MPa, air blast freezing (A.BF), and liquid immersion freezing (LIF). Temperature and phase transformations of the muscle tissue were monitored during the freezing process at three locations: center, midway between the center and the surface, and near the surface. Pork muscle quality changes [color, drip loss (both thawing and cooking), texture (shear force), and protein stability (DSC thermal profiles)] were evaluated after thawing the frozen samples at room temperature (20 degreesC). Employing pressures above 150 MPa caused very significant (P < 0.01) color changes in pork muscle during the PSF process. The PSF process reduced thawing drip loss of pork muscle but did not cause obvious changes in total drip loss following thawing and subsequent cooking. PSF at 150 and 200 MPa resulted in considerable denaturation of myofibrillar proteins of pork muscle. The PSF process also caused an increase in the pork muscle toughness as compared with that of unfrozen, ABF, and LIF samples.

Abstract:This research aimed to investigate the effects of pigskin gelatin hydrolysate (PGH) on the quality changes of longissimus lumborum (LL) muscles during freezing. The samples were firstly assigned into six groups, including control, sucrose and sorbitol group (SUSO) as positive control, 0%, 1%, 2% and 4% PGH group. The thawing loss of frozen meat, microscopic observation of ice crystal formed during freezing, myowater mobility in muscle tissues, and protein structure changes were determined. PGH reduced the thawing loss of frozen meat by 5.32%. Microscopic observation showed that ice crystal area reduced to 15.54% with 4% PGH treatment. The PGH also reduced the loss of immolized water in meat during freezing. The Raman spectra showed that the protein structure remained more intact in the group of 4% PGH. It can be concluded that the addition of PGH effectively diminished the deterioration of muscle qualities, enhanced the cryoprotective of the muscles during freezing, and this enhancement was associated with their increasing amount.

Abstract:This work investigated the effects of different thermal temperatures (84 °C for 35 min, 95 °C for 30 min, and 121 °C for 20 min) on the shelf life and microbial diversity of Dezhou-braised chicken. During refrigerated storage at 4 °C, the increase rate of total viable counts, pH and TVB-N value, was lower in 95 °C-treated and 121 °C-sterilized groups, when compared with 84 °C-treated group. Electronic nose revealed that the fresh odor of Dezhou-braised chicken treated by 84 °C and 95 °C could be maintained during storage. Additionally, 95 °C contributed to the maintenance of good texture of chicken. High throughput sequencing showed that Bacillus and Clostridium were only very active in 84 °C-treated samples, but not in 95 °C-treated and 121 °C-sterilized samples. Taken together, 95 °C can be developed as one potential thermal treatment temperature for Dezhou-braised chicken, due to its positive effects on maintaining fresh odor and texture together with extending shelf life while also protecting food safety.
Mechanisms of chicken processing quality changes during the early postmortem time: the role of the changes in myofibrillar protein

Abstract:The properties of white meat, such as chicken and fish, changed quickly in the early postmortem period. This study focused on the relationship between myofibrillar protein and the eating quality of chicken in this period. The results showed that the eating quality of chicken, such as texture, water holding capacity and shear force value, decreased in the early postmortem period. The western blot analysis indicated that the desmin and troponin T were degraded significantly with postmortem time. Meanwhile, the myofibril protein degeneration in this period was evaluated by Fourier transform infrared and Differential Scanning Calorimetry. The findings demonstrated that the ordered structure of the myofibrillar protein was destroyed, and its thermal stability was reduced with the prolongation of postmortem time. Subsequently, the transmission electron microscope results suggested that the heating processing could destroy the connection between the weakened Z-line and the myofilament. Overall, the decline in thermal stability and degradation of myofibrillar may worsen the structural instability of chicken during heating, which may be a significant contributor to the variations of chicken quality in the early postmortem period.

Abstract:In the past few yr, an emerging muscle abnormality termed wooden breast (WB) was found to affect broilers' Pectoralis major muscles. Although different studies have been performed in order to evaluate the effect of WB on meat quality, there is no evidence concerning its impact on the proteolytic processes taking place during meat aging. Thus, this study aimed at investigating the effect of a 7-day storage of broiler breast fillets on free calcium concentration, calpain activity, and proteolysis. Both the superficial and the deep layers of the Pectoralis major muscles were considered. Although similar electrophoretic profiles were observed by comparing the corresponding sampling positions, an evident lack of a high-molecular weight protein band, ascribed to nebulin, was found in the superficial layer of the WB fillets at 10 h postmortem. Compared to normal fillets (NB), both the superficial and the deep layer of WB exhibited a significantly higher amount of free calcium at 168 h postmortem (96 and 88 vs. 20 and 53 μM; P ≤ 0.001). Casein zymograms evidenced the presence of μ/m-calpain and its autolyzed form migrating as a doublet within the gel. Interestingly, neither the occurrence of WB nor the intra-fillet sampling position exerted any relevant effect on calpain activity. Indeed, a significant reduction (P ≤ 0.05) in the unautolyzed μ/m-calpain activity coupled with a remarkable increase (P ≤ 0.05) in the autolyzed form activity was observed during storage. Concurrently, if compared to NB, a significantly larger (P ≤ 0.05) amount of desmin was detected in both the superficial and the deep layers of the WB samples at 10 h postmortem. Then, a sharp decrease of the intact desmin band coupled with a progressive accumulation of its 39-kDa degradation fragment was observed without any significant difference among groups. In conclusion, the increased hardness that typically affects the WB cases seemed not to be exclusively attributable to differences in the proteolytic processes taking place within the postmortem period.

Impact of frozen storage on some functional properties and sensory evaluation of goose meat

Abstract:The objective of this study was to investigate the changes in the functional properties (pH, water holding capacity [WHC], water binding capacity [WBC], cooking losses [CL], defrosting losses [DL]), color parameters (L*, a*, b*, C, h°, ΔE), and sensory evaluation of breast (BM) and leg (LM) muscles from 17-wk-old female White Kołuda geese packaged in a vacuum and stored in frozen conditions at -20°C. During 17 wk, the geese were fed ad libitum on the same complete feed. The samples (18 BM and 18 LM) from the right part of the carcasses were stored for 30, 90, 80, 270, and 365 d. The changes in functional properties were established using a standard method used in the meat industry, according to Wierbicki et al. (1962), Grau and Hamm (1953), and CIE, (1986). Sensory evaluation was established according to defined parameters in PN-ISO 8586-2:2008. The time of frozen storage affected the decrease in WHC and WBC of BM and LM. Moreover, the LM can be characterized by a higher WHC and WBC compared to the values in the BM. It was established that CL and DL, which are the critical quality indicators, negatively increased in BM and LM during frozen storage. Considering the sensory evaluation and L*, a*, b*, C, it was established that changes in BM and LM during frozen storage were unfavorable. The scores given for smell, taste, consistency, and general appearance, as also L*, a*, and b* parameters decreased significantly during frozen storage. In addition, BM received lower scores for general appearance (at 180th and 270th day), and L* (in all frozen storage), than LM. BM and LM characterized the parameter ΔE in the range of 0.44 to 1.45, which allowed us to conclude that slight color differences were visible in these muscles (<2). Based on the study, it can be suggested that the optimal frozen storage time for BM and LM should not be longer than 180 d.

Abstract:Several attempts have so far been made to change the intracellular concentrations of ions in muscle fibres and axons. This was achieved by soaking tissues in ion-free or hypotonic solutions (Tobias, 1950; Adrian, 1956; Stephenson, 1957; Koketsu & Kimura, 1960), or by injecting solutions intracellularly (Grundfest, Kao & Altamirano, 1954; Gibbs & Johnson, 1962). However, these methods did not allow one to change drastically intracellular ionic compositions at will. An attempt was also made to perfuse the frog muscle fibre internally, but the method was not satisfactory enough to examine any problems concerned (Davies, 1961). Recently successful methods of intracellular perfusion of squid giant axons have been established by two groups (Oikawa, Spyropoulos, Tasaki & Teorell, 1961; Baker, Hodgkin & Shaw, 1961 a, b). The method of the former group depends on removal of axoplasm by inserting a large capillary (300,u in diameter), while that of the latter depends on squeezing out axoplasm. Several recent studies by these methods have demonstrated the importance of sodium, potassium and calcium ions in membrane potential and excitation (Baker et al. 1961 a, b; Tasaki, 1962; Baker, Hodgkin & Shaw, 1962a, b, c; Tasaki, Watanabe & Takenaka, 1962; Tasaki & Shimamura, 1962). For example, the resting potential was decreased by lowering the internal potassium concentration, whereas the depolarized membrane was still able to produce action potentials of large amplitude. Raising the internal sodium concentration decreased the spike height, although the spike could be restored to some extent by adjusting the external calcium and magnesium concentrations. Abstract:This study investigated the effects of deep freezing and storage temperature (-50°C, -60°C, and -80°C) on the quality and freshness of lamb. To compare the qualities of deep frozen and stored lamb, fresh control and normal freezing conditions (-18°C) were adopted. As quality and freshness parameters, drip loss (thawing loss and cooking loss), water-holding capacity (WHC), texture profile analysis (TPA), thiobarbituric acid reactive substances (TBARS), and total volatile basic nitrogen (TVBN) were evaluated during 5 months of storage. Temperature influenced the drip loss and WHC, and deep freezing minimized the moisture loss during frozen storage compared to the normal freezing condition. Lamb frozen and stored at deep freezing temperature showed better tenderness than that stored in normal freezing conditions. In particular, lamb frozen at lower than -60°C exhibited fresh lamb-like tenderness. Regardless of temperature, evidence of lipid oxidation was not found in any frozen lamb after 5 months, while TVBN was dependent on the applied temperature. Therefore, this study demonstrated that deep freezing could potentially be used to maintain freshness of lamb for 5 months. From the quality and economic aspects, the freezing and storage condition of -60°C is estimated as the optimum condition for frozen lamb.
Oxidative damage to poultry, pork, and beef during frozen storage through the analysis of novel protein oxidation markers

Abstract:The susceptibility of meats from different animal species (beef quadriceps femoris, porcine longissimus dorsi, and chicken pectoralis major) to undergo protein oxidation during frozen storage (20 weeks/-18 °C) was studied through the analysis of novel oxidation markers. Frozen storage induced protein carbonylation (α-aminoadipic and γ-glutamic semialdehydes), carboxylation (α-aminoadipic acid), and formation of Schiff bases in meat from the three species. Major rates of protein and lipid oxidation products [thiobarbituric-acid-reactive substances (TBARS) and hexanal] were found in beef patties. Among the endogenous factors having a potential influence on the susceptibility of meat to undergo protein oxidation, heme iron seemed to play a major role. The present study illustrates the severe chemical modifications induced by oxidative stress during frozen storage of ground meat and provides original insight into the underlying mechanisms and factors.

Abstract:The effect of 7-day subfreezing storage on the physicochemical properties, nutritional composition, and microstructure of pork was investigated. After 7 days of chilling at 4°C, the meat exhibited color deterioration and the development of off-flavors. In contrast, the -12°C treatment significantly reduced the deterioration in water-holding capacity and color of samples (p < 0.05) and prevented changes in pH value. Similarly, the treatments at -12 and -18°C effectively preserved the meat's tenderness, thiobarbituric acid-reactive substances, protein solubility, and textural properties, maintaining these qualities close to those of fresh meat (p > 0.05). The nutrient content of samples stored at -12°C was comparable to those stored at -18°C (p > 0.05). Furthermore, subfreezing at -12°C was found to protect muscle integrity, promoting the formation of an elastic gel network and a homogenous muscle fiber structure. Therefore, the study concludes that 7-day subfreezing storage at -12°C can reduce protein denaturation and maintain thequality of pork, a result that is typically achieved under more extreme freezing conditions at -18°C.
Wooden Breast Chicken Fillets: Viability in the Preparation of Hamburgers and Bologna

Fillets (pectoralis major) were collected from a commercial slaughterhouse and classified as normal (no visible alteration) or WB (with characteristic visual alterations) by a trained specialist. The quality parameters and physical, chemical, and technological stability of the resulting meat products during the storage period, at 1, 30, 60, and 90 days from production were evaluated. Hamburgers with WB showed a significant increase of 2.38% in moisture content and a reduction of 17% in protein content, whereas bologna showed an increase of 1.15% in moisture content. The inclusion of WB meat did not influence the yield and stability of these products, but resulted in higher pH and softer hamburgers with greater lipid oxidation. The results suggest that the manufacture of meat products with a lower percentage of WB meat in their formulation is a viable alternative to minimize costs and reduce economic losses generated by the incidence of the anomaly.

Abstract:Abstract Fresh meat is vulnerable to microbial contamination and loss of quality. Hence, freezing is used to mitigate these issues during transport and storage. However, freezing may negatively impact the same quality characteristics it was meant to preserve, resulting in products that are dissatisfying to the consumer. During the freezing process of meat, water forms ice crystals of various shapes and sizes. The degree of ice crystal growth, mainly due to the freezing rate, has been shown to affect several meat quality characteristics, including tenderness, juiciness, and color. Previous research has primarily evaluated the effect of freezing/thawing on the quality of beef retail cuts. To the best of our knowledge, however, minimal research has been devoted to examining the impact of freezing/thawing on the quality of beef primal and sub-primal cuts. Therefore, this study aimed to gain deeper insight into the histological, ultrastructural, and physiochemical changes occurring in whole beef ribs in response to different freezing/thawing treatments. To achieve our objective, we purchased 32 choice beef rib primals from a local harvest facility 24 h postmortem. Ribs were vacuum packaged and randomly divided into four groups (8 ribs/group). The first group was frozen in a −20 °C freezer (slow freezing), stored in the same freezer for 12 weeks, and thawed at 4 °C for 24 h (slow thawing). The second group was frozen in an ultralow temperature freezer (−80 °C; fast freezing), stored in the same freezer for 12 weeks, and thawed at 4 °C for 48 h. The third and fourth groups were frozen and stored in the same manner as groups one and two, respectively, but were thawed in a water bath (10 °C) for 8 h (fast thawing). Two 2.5-cm-steaks from the middle and each end were collected to represent the entire rib. One steak from each of the three areas was used for color, cook loss, and tenderness evaluation, while the other three supplied tissue for drip loss and lipid oxidation analysis. Data were analyzed using the mixed model of SAS with P ≤ 0.05 considered significant. No differences were observed in color or tenderness between the treatments (P > 0.05). However, slow thawing increased drip loss, regardless of the freezing rate (P < 0.05). Greater cook loss was observed in the fast frozen-slow thawed samples compared with fast frozen-fast thawed samples (P < 0.05). The slow frozen-fast thawed steaks had greater lipid oxidation than the slow frozen-slow thawed treatment (P < 0.05). In general, our data show that subjecting whole beef rib sections to fast freezing may reduce water loss at the expense of increased lipid oxidation. This demonstrates that additional research is needed to further optimize beef primal cut storage.

Abstract:Abstract Limited shelf life of chilled poultry meat hinders its transportation over long distances to remote areas of consumption. In this regard, it is necessary to develop a storage technology for chilled poultry meat with prolonged shelf life. The study was carried out on meat samples of broiler chickens produced in accordance with GOST 31962-2013 and cooled by water evaporation. An IS 203.4 temperature recorder was used to measure meat temperature, and a TESTO 205 device was used to measure pH. The microbiological, physicochemical, and sensory characteristics of meat were determined according to standard methods. The shelf life of chilled meat was determined in accordance with MUK 4.2.1847-04 Sanitary and epidemiological assessment of justification for the shelf life and storage conditions of food products . The study showed that meat stored at −2.0 ± 1.0 °C remains fresh for 20 days in terms of microbiological, physicochemical and organoleptic characteristics. It was found that meat can be transferred from a supercooled state to a chilled one in order to increase the shelf life to 15 days at a storage temperature of -2.0 ± 1.0 °C.

Abstract:The aim of present study was to investigate the influence of conventional freezing (CF, −18 °C), low-temperature freezing (LF, −40 °C), and ultra-low-temperature freezing (ULF, −80 °C) on the quality, moisture distribution, microstructure, and flavor profile of hand-grabbed mutton (HGM) during frozen storage (0, 30, 60, 90, 120, 150 and 180 days). The TPC, TVB-N, and TBARS values increased significantly with prolonged storage, while the moisture content decreased ( P < 0.05). Additionally, the concentrations of aldehydes, alcohols, ketones, acids, and alkenes decreased significantly as the storage duration increased. However, the concentrations of esters and heterocyclics increased ( P < 0.05). Notably, at 30–180 days of storage, the TBARS and TVB-N values in ULF samples were significantly lower than those in CF and LF samples, while the moisture content was significantly higher ( P < 0.05). Low field-nuclear magnetic resonance (LF-NMR) analysis showed that ULF decreased water migration and maintained the original texture characteristics of HGM during frozen storage. The ULF and LF groups had significantly higher levels of volatiles than the CF group ( P < 0.05). The findings show that ULF, with its relatively rapid freezing rates, can still maintain the high quality of HGM after 180 days of frozen storage, contributing to quality control.

Abstract:Freezing is an important accepted method of food preservation to keep the safety of meat products. The methodology used in freezing and thawing processes can effect on the quality of frozen foods. The aim of this study was to evaluate the sensory and microbial analysis of about 48 random samples of freshly slaughtered chicken breast and thigh meat (24 for each). Samples were collected from local commercial retail shops in Tanta city submitted to freezing (-18ᴼC) for (1 week). Then, four different thawing methods were evaluated by using microwave oven for one minute, refrigeration at around 3 ᴼC for 22 hrs., under tap water for 4hrs. and finally at room temperature for 4 hrs. The obtained results revealed that the best sensory character for breast and thigh meat samples thawed and rethawed in refrigerator were 8.6,8.6,7 and 8, respectively. Also the best APC (log cfu/g) in breast and thigh meat samples thawed and rethawed in microwave were 3.84±0.06, 00.00,4.78±0.43 and 2.01±1.7, respectively. As well as the best Enterobacteriaceae count was in breast and thigh meat samples thawed and rethawed in microwave were 0.00,0.00,0.00 and 1.86, respectively. Overall, using of the microwave thawing was faster than the traditional one and showed the less bacterial load, while thawing in refrigerator was showed the best sensory quality.

Abstract:This study investigated the protein oxidation properties of lamb following chilled-then-frozen storage. Experimental (n=360) M. longissimus lumborum (LL) were randomly sampled from the boning room of a commercial Australian abattoir, at 24h post mortem, and assigned to five chilled storage periods (0, 2, 4, 6 and 8weeks) and six subsequent frozen storage periods (0, 4, 8, 12, 24 and 52weeks). Upon completion of each storage treatment combination, corresponding LL were sub-sectioned and analysed for carbonyl content, protein solubility, nitrate/nitrite content, particle size analysis and estimated myoglobin fractions. The association between these protein measures and shear force was also explored. During chilled storage, particle size and sarcoplasmic protein solubility decreased which indicated protein degradation, while frozen storage only affected myoglobin oxidation. Tenderness was best explained by decreased particle size, decreased deoxymyoglobin and increased oxymyoglobin. No carbonyl effects were observed. It can be concluded that, according to these analyses, that in chilled-then-frozen lamb carbonyl formation was negligible.

Lipid and protein oxidation of emulsified chicken patties prepared using abdominal fat and skin

Abstract:Skin (SK) and abdominal fat (AF) are by-products that are abundantly produced in poultry slaughterhouses. Skin is used in meat products, especially in sausages, but its use is related to microbiological contamination and susceptibility to oxidation. The aim of the present study was to evaluate the effect of SK replacement with AF on the quality characteristics of emulsified patties stored under freezing conditions (-18°C) for 90 D. The AF showed higher lipid content than did the SK, but the SK had a higher predominance of polyunsaturated fatty acids. Regarding the partial chemical composition, the treatments showed differences in moisture, lipids, and total collagen. The values for para-anisidine and carbonyl compounds at the end of 90 D of storage were not influenced by the incorporation of SK or AF. The judges also did not observe differences in the color and aroma of the emulsified products during the 90 D. Furthermore, volatile compounds considered oxidation markers were not detected at the end of the storage period. Given these results, emulsified patties made with SK or AF undergo, to a similar degree, low levels of lipid and protein oxidation when stored under freezing conditions (-18°C) for 90 D, which allows the use of some of these lipid sources in meat products.

Abstract:This research investigated the combined effects of cooking methods (household griddle (C1), household conventional oven (C2), industrial oven (C3), storage temperature (refrigeration /freezing) and packaging system (aerobic and vacuum), on quality of chicken burgers. The results show that refrigeration storage favours the retention of moisture content and the juiciness of chicken burgers, but the application of vacuum (RV) was the best option to maintain the juiciness and moisture content (p < 0.05) of the samples prepared in an industrial oven with forced-air convection. The highest protein content was observed in the samples that were frozen in vacuum packaging (FV), and the lipid content was highest in the samples chilled in conventional packaging (RC) at 1 atm. Regardless of the cooking method used and the internal pressure of the packages (≤ 1 atm), refrigerated storage contributed to the best scores (p < 0.05) for color and flavor attributes. The treatments that presented the highest sensory acceptance index were the frozen samples in vacuum packaging prepared in the industrial oven, conventional oven, and grill (90%, 82.66%, and 74.33%, respectively).
